Six donkeys (Equus asinus) were purchased locally. To screen them before and during Trypanosoma vivax infection, thin and thick blood smears, temperature, haematocrit centrifuge technique (HCT), packed cell volume (PCV), white blood cell counts, and indirect immunofluorescent antibody test (IFAT) were done for Babesia equi. For the IFAT, an anti-horse conjugate was used. In spite of patent B. equi or T. vivax parasitaemia, the donkeys' temperatures remained below 38.5 degrees C; PCV was depressed more in B. equi infection than in T. vivax infection. Four out of the 6 donkeys had B. equi antibodies while 2 of them had detectable parasitaemia. Treatment with either Berenil or Imizol cleared the detectable B. equi parasitaemia, and IFAT was negative at 35-45 days post treatment. However, relapses occurred within 60-70 days after the treatment. In 2 circumstances serological titres were below 1:40 (negative) while there was detectable parasitaemia.  相似文献

This study aimed to determine the erythrocytic lipid peroxidation and haemoglobin oxidation as contributory factors causing anaemia in cattle (Friesian × Egyptian native breed) infected with Babesia bigemina. Blood was collected from 32 cows infected with B. bigemina along with 18 healthy cows as controls for determination of erythrocytic malondialdehyde (MDA), blood methaemoglobin (MetHb), plasma free haemoglobin (PHb), corpuscular osmotic fragility (COF), red blood cell count (RBC), total haemoglobin (Hb) and packed cell volume (PCV). Percentage of parasitaemia varied from 14% to 36%. MDA, MetHb, COF and PHb were significantly increased (P < 0.001) in infected cows versus controls. Parasitaemia was positively correlated (P < 0.001) with MDA, MetHb, COF and PHb. MDA was positively correlated (P < 0.001) with COF and PHb and negatively correlated (P < 0.001) with RBC, Hb and PCV. MetHb was negatively correlated (P < 0.001) with RBC, Hb and PCV and positively correlated (P < 0.001) with COF. In conclusion, B. bigemina infection in cattle is associated with a parasitic burden-dependent corpuscular oxidative damage as indicated by membrane lipid peroxidation and methaemoglobin formation, which are contributed to COF and intravascular haemolysis.  相似文献

Intravenous administration of xylazine to beef cattle (10 animals, 0.2 mg/kg of body weight) resulted in rapid onset (less than 15 minutes) of hyperglycemia. Plasma glucose values increased to 195 +/- 15 mg/dl and 305 +/- 10 mg/dl at 15 minutes and 3 hours, respectively. Concomitantly, plasma insulin concentrations dropped from 23 +/- 2 microU/ml before xylazine to 5.8 +/- 0.7 microU/ml and 2.4 +/- 0.3 microU/ml at 15 minutes and 3 hours, respectively. Parallel decreases (20%) were observed for percentage of hemoglobin, red blood cell number, and packed cell volume. Plasma urea nitrogen was significantly (P less than 0.01) incrased within 3 hours of xylazine administration (6.7 +/- 0.9 mg/dl vs 11.4 +/- 0.7 mg/dl). Marked changes in concentrations of plasma-free fatty acids were not observed. Alternative means of anesthesia must be considered in those instances in which biopsy material is to be used for studies of carbohydrate metabolism in vitro.

The aim of the study was to evaluate the effects of varying concentrations of ethylenediaminetetraacetic acid (EDTA) on blood samples from White Fulani breed of cattle and West African Dwarf goat from Nigeria. Sample sizes of 20 animals were used for both species. Different concentrations of EDTA (2, 4, 8 and 16 mg/ml) were used. The packed cell volume (PCV), red blood cell (RBC) and haemoglobin (Hb) concentration of blood samples collected from White Fulani breed of cattle and West African Dwarf goat into bottles containing 16 mg/ml of EDTA were significantly lower (P < 0.05) than those samples collected from the same animals into bottle containing 2 mg/ml (control). Similarly, the PCV, RBC and Hb values of the West African Dwarf goats in bottles containing 8 mg/ml of EDTA were significantly lower than those of the samples in the control (2 mg/ml). This study has shown that high concentration of EDTA as an anticoagulant can lead to a false erythrocytic index especially the PCV. In collecting blood samples for evaluation of haematological parameters, therefore, the blood volume/anticoagulant ratio must be strictly adhered to prevent error in the evaluated parameters in cattle and goats. Taken together, there is tendency for haemolytic anaemia to occur in blood sampled at higher concentration of anticoagulants in West African Dwarf goat than in White Fulani breed of cattle.  相似文献

Babesiosis is a common tick borne disease of dogs in tropical and subtropical regions of the world caused by different species of Babesia. The present study aimed to examine erythrocyte lipid peroxide and erythrocytic antioxidant levels in dogs with clinical babesiosis, caused by Babesia gibsoni, and impact of the disease on blood iron, zinc and copper levels. The study was conducted on 10 naturally occurring cases of canine babesiosis with the history of tick infestation, erratic pyrexia, and prolonged illness. Microscopic examination of Giemsa stained peripheral blood smears confirmed B. gibsoni infection in the erythrocytes. Six apparently healthy dogs of different age, sex and breeds, brought for either health checkup or vaccination were used for comparison. Levels of erythrocytic antioxidant enzymes were significantly (P<0.01) higher in sick dogs than those of cytologically negative dogs (catalase: 0.192+/-0.024 units/mg Hb vs 0.074+/-0.004 units/mg Hb; superoxide dismutase: 0.014+/-0.0009 units/mg Hb vs 0.006+/-0.0008 units/mg Hb and lipid peroxide: 6.01+/-0.30 nmol MDA/mg Hb vs 1.89+/-0.10 nmol MDA/mg Hb). The levels of blood micronutrients were significantly low in these dogs (iron: 89.87+/-8.12 microg/g vs 126.44+/-14.65 microg/g; zinc: 3.67+/-1.85 microg/g vs 5.62+/-1.83 microg/g and copper: 0.55+/-0.63 microg/g vs 0.65+/-0.04 microg/g). The study demonstrated oxidative damage in dogs naturally infected with B. gibsoni. Low level of blood iron, zinc and copper seems to have an additional role in the genesis of anaemia and oxidative stress.  相似文献

This work examines the effect of treatment of camels with furosemide (1 or 2 mg/kg, intravenously) on blood volume (BV), plasma volume (PV) and the plasma concentrations of total solids (PTS), plasma total protein (PTP) haemaglobin (Hb) and haematocrit (PCV). The cumulative urine produced during the 4 h following furosemide administration (2 mg/kg) averaged 22.2 mL/kg, compared to 2.3 mL/kg in controls. None of the above parameters were significantly changed by furosemide treatment at either dose.
In another experiment, the effect of the two doses of furosemide on the sequential changes on some of the above parameters was investigated at 5, 10, 15, 30, 45, 60, 75, 90, 105, 120 and 240 min after the drug administration.
On the whole, there were no significant changes in any of the measured parameters, except for small but statistically significant increases in the PCV and Hb at 30 and 45 min post treatment.
It is concluded that, despite the marked diuresis following furosemide administration, the camel appears to be able to maintain its bodily fluid haemostasis.  相似文献

The cardiopulmonary, anesthetic, and postanesthetic effects of an IV infusion of the hypnotic agent propofol were assessed in 6 Greyhounds and 7 non-Greyhounds. After IM injection of acetylpromazine and atropine, a bolus injection of propofol sufficient to allow endotracheal intubation (mean +/- SEM = 4.0 +/- 0.3 mg/kg of body weight in Greyhounds; 3.2 +/- 0.1 mg/kg in non-Greyhounds) was administered, followed by continuous infusion at a rate of 0.4 mg/kg/min for 60 minutes, during which time dogs breathed 100% oxygen. In 23% of all dogs (3 of 13), apnea developed after initial bolus administration of propofol. Arterial blood pressure was well maintained in all dogs, but heart and respiratory rates were decreased significantly (P less than 0.05) during the infusion in Greyhounds. In Greyhounds, mild respiratory acidosis developed after 45 minutes, whereas arterial carbon dioxide tension was increased at all times after propofol administration in non-Greyhounds. In all dogs, PCV and total plasma proteins were unaffected by propofol. Rectal temperature decreased during treatment. Muscle tremors were observed in approximately 50% of dogs (in 3 of 6 Greyhounds and 3 of 7 non-Greyhounds) during and after infusion of propofol. Non-Greyhounds lifted their heads, assumed sternal recumbency, and stood 10 +/- 1, 15 +/- 3, and 28 +/- 5 minutes, respectively, after the end of the infusion; in Greyhounds, the corresponding times were 36 +/- 4, 43 +/- 6, and 63 +/- 7 minutes.  相似文献

Acid-base, serum electrolyte, plasma protein, and packed cell volume (PCV) values were determined in venous blood samples from 30 red deer (Cervus elaphus) of both sexes showing no clinical signs of disease. The animals were 5 months of age and kept on pasture in the Valley of Mexico, at an altitude of 2450 m. Blood samples were collected without sedation. Mean blood values were: pH 7.411 +/- 0.041, pCO2 37.7 +/- 4.4 mmHg, base excess 0.7 +/- 3.2 mmol/L, actual bicarbonate 24.3 +/- 3.1 mmol/L, total CO2 25.3 +/- 3.2 mmol/L and anion gap 23.5 +/- 5.5 mmol/L. Mean serum electrolyte levels were: Na+ 142.3 +/- 2.5 mmol/L, Cl- 100.5 +/- 2.3 mmol/L, and K+ 7.03 +/- 1.03 mmol/L. Plasma protein and PCV values were 60.0 +/- 6.6 g/L and 0.47 +/- 0.05 L/L, respectively. Blood values determined in this study can be considered reference data for health control and disease diagnosis.  相似文献

OBJECTIVE: To determine pharmacokinetics of single and multiple doses of rimantadine hydrochloride in horses and to evaluate prophylactic efficacy of rimantadine in influenza virus-infected horses. ANIMALS: 5 clinically normal horses and 8 horses seronegative to influenza A. PROCEDURE: Horses were given rimantadine (7 mg/kg of body weight, i.v., once; 15 mg/kg, p.o., once; 30 mg/kg, p.o., once; and 30 mg/kg, p.o., q 12 h for 4 days) to determine disposition kinetics. Efficacy in induced infections was determined in horses seronegative to influenza virus A2. Rimantadine was administered (30 mg/kg, p.o., q 12 h for 7 days) beginning 12 hours before challenge-exposure to the virus. RESULTS: Estimated mean peak plasma concentration of rimantadine after i.v. administration was 2.0 micrograms/ml, volume of distribution (mean +/- SD) at steady-state (Vdss) was 7.1 +/- 1.7 L/kg, plasma clearance after i.v. administration was 51 +/- 7 ml/min/kg, and beta-phase half-life was 2.0 +/- 0.4 hours. Oral administration of 15 mg of rimantadine/kg yielded peak plasma concentrations of < 50 ng/ml after 3 hours; a single oral administration of 30 mg/kg yielded mean peak plasma concentrations of 500 ng/ml with mean bioavailability (F) of 25%, beta-phase half-life of 2.2 +/- 0.3 hours, and clearance of 340 +/- 255 ml/min/kg. Multiple doses of rimantadine provided steady-state concentrations in plasma with peak and trough concentrations (mean +/- SEM) of 811 +/- 97 and 161 +/- 12 ng/ml, respectively. Rimantadine used prophylactically for induced influenza virus A2 infection was associated with significant decreases in rectal temperature and lung sounds. CONCLUSIONS AND CLINICAL RELEVANCE: Oral administration of rimantadine to horses can safely ameliorate clinical signs of influenza virus infection.  相似文献

The mean packed cell volume (PCV) for canine blood samples received by post was significantly higher than that for fresh samples, at each haemoglobin value over the range 9 to 19 g/dl. As a consequence the mean corpuscular volume (MCV) and the mean corpuscular haemoglobin concentration (MCHC) also differed. Apart from one Hb value there was no significant difference in the mean red blood cell (rbc) count over the Hb range 6 to 14 g/dl. It is suggested that the normal values for the PCV and MCV should be 3 to 5% and 5 to 7 f 1 higher respectively for postal samples as compared to fresh samples while the MCHC should be about 3 to 4 g/dl lower.  相似文献

During infection with T. vivax (strain Y 58) four stages can be distinguished: (a) A prepatent period of about 1 week. (b) An acute or fulminating disease characterized by an almost continuous, fluctuating parasitaemia, a decrease of the number of thrombocytes, blood serotonin level, packed cell volume (PCV) and Hb, serum albumin and γ-globulin fractions, a low serum total lipid content and an initial rise with subsequent decrease of the blood bradykinin level. (c) A critical stage of about 4–5 weeks post infection (pi) during which the animal may develop a progressive parasitaemia and may die from disseminated intravascular coagulation. At necropsy microthrombi were a consistent and most significant observation. Accompanying features were oedema and haemorrhages in the various tissues, pulmonary oedema and lobular infarction, patches of necrosis in liver, spleen and adrenals and nephrotic changes in the kidneys. Perivascular mononuclear infiltrates, resulting in local granuloma formation (heart liver), were a constant feature. Related biochemical changes were an increase of the blood urea and serum lactate dehydrogenase levels. (d) A post-critical stage characterized by an ameliorating condition, periods with no or few trypanosomes in the peripheral blood alternating with relatively high peaks of parasitaemia. This was associated with increased blood bradykinin levels, an increase of the PCV and Hb, serum albumin and γ-globulin fractions, number of thrombocytes and blood serotonin level. A return to normal values of the serum total lipids was observed at this stage. Despite this clinical recovery of the animals, at necropsy (on days 90, 98, and 99 pi) there was a moderate to massive histiocyte-lymphocyte-plasma cellular infiltration of the heart, which led to cardiac insufficiency in two heifers. The rise of the d-amino laevulinic acid dehydratase levels of erythrocytes during the decline of the PCV and Hb values is an indication for the production of many young erythrocytes and thereby for the haemolytic nature of the anaemia.  相似文献

In investigations of 51 pregnant sows, blood Hb levels above 7.45 mmol/l (mean value of 8.94 +/- 0.88) were recorded from 43 animals (Group A), while levels below that margin were recorded from eight (Group B) (mean value of 6.73 +/- 0.31). Iron levels in blood plasma of Group A were 36.9 +/- 8.07 and were thus higher (p less than 0.01) than those in Group B (28.9 +/- 5.19 mumol/l). No difference was found to exist between the two groups with regard to iron fixation capacity as well as copper and zinc in blood plasma. Oral uptake of 50, 500 or 2,000 mg of iron in the form of FeSO4.7H2O with cereal shred as morning ration by 17 or 15 sows failed to cause any increase in blood plasma iron levels, within 360 minutes from uptake. Literature data as well as the results of these analyses have supported the recommendation that Hb variations between 4.96 and 9.93 mmol/l (8-16 g/dl) should be rated as physiological in the blood of pregnant sows. The GDR Standard TGL 35,423 on "Monitoring of Metabolism on Pig Breeding and Fattening Units" should be accordingly amended.  相似文献

The objectives of Exp. 1 were to determine the effects of production system and genotype on pig performance and health. Sows were bred, gestated, farrowed, and lactated in either an intensive indoor or an intensive outdoor production system. The three dam genotypes of pigs used in each environment were PIC Camborough-15 (C-15), PIC Camborough Blue (CB), and Yorkshire x Landrace (YL). All pigs received 100 mg of iron dextran at d 3 of age. Pigs raised in the outdoor unit had higher blood hemoglobin (Hb) concentrations on d 28 of age than pigs raised indoors (11.5 +/- .22 vs 8.16 +/- .26 g/dL, P < .0001). Outdoor-reared pigs had more white blood cells (WBC) on d 3 than indoor-reared pigs (9.7 +/- .38 vs 8.04 +/- .38 cells/microL x 10(3), P < .05), but outdoor pigs had fewer WBC on d 28 of age than indoor-reared pigs (9.8 +/- .5 vs 11.1 +/- .45 cells/microL x 10(3), P < .05). Genetic lines did not differ in plasma immunoglobulin G (IgG) concentrations at 3 or 28 d of age. Environment and age influenced pig Hb levels and WBC numbers. The objectives for Exp. 2 were to determine whether C-15-405 pigs reared outdoors or indoors needed supplemental iron or whether they would receive enough environmental iron, and how the lack of supplemental iron may impact pig Hb and immunity. Indoor and outdoor pigs received either no supplemental iron, 100 mg, or 400 mg of iron dextran on d 3 of age. Blood percentage neutrophils and neutrophil: lymphocyte ratio were lower (P < .05) indoors, and natural killer cell (NK) activity was greater (P < .05) among indoor- than outdoor-reared pigs (NK % cytotoxicity: 15.6 +/- 2.3 vs 9.7 +/- 2.3). Outdoor-reared pigs that received no injected iron had similar Hb at d 28 of age as indoor-reared pigs that received 100 mg of iron dextran (11.1 +/- .36 vs 10.7 +/- .4 g/dL, P = .59). Supplemental iron may not be necessary in an outdoor production system. Outdoor-reared pigs had lower values for some immune measures, but they had similar survival rates as indoor-reared litters.  相似文献

Hematologic and serum biochemical tests were used to monitor the health of 3 groups of bison in an experimental study of tuberculosis. Bison were randomly assigned to Mycobacterium bovis-infected, M. bovis-sensitized, and uninfected control groups. Hematologic measurements included total and differential leukocyte counts, hemoglobin (Hb), packed cell volume (PCV), fibrinogen, and plasma proteins. Biochemical tests included serum urea nitrogen, creatinine, aspartate amino transferase, sorbitol dehydrogenase, serum calcium, and serum phosphorus. There were no significant differences (P = 0.05) in any test values between groups of bison. The bison data were combined and compared to similar data of cattle. The mean values for PCV and Hb were higher than values (PCV 24-46%, Hb 8-15 g/dl) for cattle. Mycobacterium bovis-infected bison had a slight increase in the number of blood monocytes and lymphocytes when compared to the uninfected bison but were within the normal ranges for bison and cattle. Other hematologic parameters were within normal ranges reported for cattle. Creatinine levels in all bison were above the normal range (1.0-1.5 mg/dl) for cattle. Phosphorus levels for M. bovis-infected and M. bovis-sensitized bison exceeded the normal range (5.6-8.0 mg/dl) reported for cattle. The level for uninfected bison was near the upper limit of normal for cattle. Mean values for other serum biochemical tests were within the normal ranges reported for cattle.  相似文献

Cardiovascular responses to sublethal endotoxin infusion (Escherichia coli, 50 micrograms/ml in lactated Ringer solution at 100 ml/h until pulmonary arterial pressure increased by 10 mm of Hg) were measured 2 times in 5 standing horses. In a 2-period crossover experimental design, horses were either administered hypertonic (2,400 mosm/kg of body weight, IV) or isotonic (300 mosm/kg, IV) NaCl solution after endotoxin challenges. Each solution was administered at a dose of 5 ml/kg (infusion rate, 80 ml/min). Complete data sets (mean arterial, central venous, and pulmonary arterial pressures, pulmonary arterial blood temperature, cardiac output, total peripheral vascular resistance, heart rate, plasma osmolality, plasma concentration of Na, K, Cl, and total protein, blood lactate concentration, and PCV) were collected at 0 (baseline, before endotoxin infusion), 0.25, 1, 1.5, 2, 2.5, 3, 3.5, 4, and 4.5 hours after initiation of the endotoxin infusion. Blood constituents alone were measured at 0.5 hour and cardiovascular variables alone were evaluated at 0.75 hour. By 0.25 hour, endotoxin infusion was completed, a data set was collected, and saline infusion was initiated. By 0.75 hour, saline solutions had been completely administered. Mean (+/- SEM) cardiac output decreased (99.76 +/- 3.66 to 72.7 +/- 2.35 ml/min/kg) and total peripheral resistance (1.0 +/- 0.047 to 1.37 +/- 0.049 mm of Hg/ml/min/kg) and pulmonary arterial pressure (33.4 +/- 0.86 to 58.3 +/- 1.18 mm of Hg) increased for both trials by 0.25 hour after initiation of the endotoxin infusion and prior to fluid administration. For the remainder of the protocol, cardiac output was increased and total peripheral resistance was decreased during the hypertonic, compared with the isotonic, saline trial.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Thermoregulation may limit exercise performance under hot and humid conditions. This study compared heart rate (HR), respiratory rate (RR), rectal temperature (Tr), packed cell volume (PCV) and total plasma protein concentration (TPP) during a submaximal incremental field exercise test under high vs. low ambient temperature and relative humidity. Ten horses were tested 3 times in summer (July) and 3 times in autumn (September). Heart rate was measured continuously, the other variables at rest and immediately after 4 min at 3.5, 4.5 and 7.0 m/s, separated by 3 min rest intervals, and after 5 and 10 min recovery. Data for all variables were significantly greater during exercise and recovery in the hot vs. cool conditions, respectively: after 4 min at 7.0 m/s, HR was 135+/-1 and 123+/-1/min (P<0.0001), Tr was 39.0+/-0.06 and 38.0+/-0.05 degrees C (P<0.0001), RR was 99+/-3 and 50+/-3/min (P<0.0001), PCV was 48.8+/-0.06 and 42.1+/-0.3% (P<0.0001) and TPP was 7.7+/-0.14 and 7.6+/-0.12 g/l (P = 0.026). These data reflect the thermal burden during submaximal exercise under hot conditions in the field. The greater relative PCV increase in the heat probably conferred a thermoregulatory advantage and reflected a greater circulating red cell volume increase rather than a decrease of plasma volume. This study illustrates how differences in environmental conditions can affect assessment of exercise responses and how these factors must be considered in monitoring progress during fitness and acclimatisation regimes in the field.  相似文献

Blood constituents and vascular volume indices were determined in 5 standing horses by use of 2-period crossover experimental design. Horses were either administered hypertonic (2,400 mosm/kg of body weight, i.v.) or isotonic (300 mosm/kg, i.v.) saline solution. Each solution was administered at a dosage of 5 ml/kg (infusion rate, 80 ml/min). Samples for determination of PCV, plasma volume, blood volume, plasma osmolality, total amount of plasma protein and plasma concentrations of protein, Na, K, and Cl were collected at 0 hour (baseline, before fluid infusion) and 0.5 hour (at the end of fluid infusion), and subsequently, at 0.25- or 0.5-hour intervals for 4.5 hours. All horses were given the predetermined dose of fluids by 0.5 hour after beginning the saline infusion. Values of P < or = 0.05 were considered significant. Administration of hypertonic saline solution was associated with decreased mean body weight by 4.5 hours, but weight change after isotonic saline administration was not significant. Other than body weight and plasma protein concentration, between-trial difference (treatment effect) was not observed for any measured variable or index. The F values indicated that increasing the number of horses would have not changed these results. A time effect was evident across both trials, so that mean (+/- SD) plasma volume increased (12.3 +/- 1.07%) and mean plasma protein concentration (-12.1 +/- 1.03%) and PCV (-11.9 + 0.67%) decreased proportionately and transiently in association with administration of either fluid at that volume. Other time effects included increased plasma osmolality and Na and Cl concentrations. Blood volume estimates and total amount of plasma protein remained unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Blood was sampled from fattening pigs fixed with the snout loop in the pen as well as during slaughter after control or container transport. Assays were made of haemoglobin (Hb), haematocrit (Hk), glucose concentration (PG), and lactic acid (MS) in the plasma. The mean values and standard deviations of Hb and Hk were 13.97 +/- 1.14 g/100 ml, 48.36 +/- 3.67 per cent, 15.24 +/- 1.12 g/100 ml, 49.83 +/- 4.04 per cent, 15.32 +/- 1.23 g of 100 ml, and 51.08 +/- 3.73 per cent. The means values of PG were between 51 and 101 mg of 100 ml and those of MS between 8 and 16 mMol/l.  相似文献
