The ability of diphenyl diselenide [(PhSe)2] to attenuate oxidative damage was evaluated in the liver, gills, brain, and muscle of carp (Cyprinus carpio) and silver catfish (Rhamdia quelen) experimentally exposed to fipronil (FPN). Initially, the fish were fed a diet without (PhSe)2 or a diet containing 3.0 mg/kg of (PhSe)2 for 60 days. After the 60-day period, the fish were exposed to 0.65 µg/L of FPN for 192 h. The results showed that carp exposed to FPN and not fed with (PhSe)2 exhibited acetylcholinesterase (AChE) inhibition in brain and muscle, and increased thiobarbituric acid-reactive substance (TBARS) in liver, gills, and brain. Furthermore, FPN decreased nonprotein thiols (NPSH) and δ-aminolevulinate dehydratase (δ-ALA-D) in carp liver and gills, and increased plasma glucose and protein levels. In silver catfish, FPN inhibited AChE and increased TBARS levels in muscle. In addition, glutathione S-transferase (GST) decreased in liver and muscle, and plasma glucose was increased. (PhSe)2 reversed some of these effects. It prevented the increase in TBARS levels in liver, gills, and brain in carp and in silver catfish muscle, and reversed the increase in plasma glucose levels in both species. Additionally, (PhSe)2 increased the NPSH levels in carp and silver catfish that had decreased in response to FPN exposure. However, (PhSe)2 was not effective in reversing the AChE inhibition in brain and muscle or the δ-ALA-D decrease in carp liver. Thus, (PhSe)2 protects tissues of both species of fish, mainly by preventing or counteracting the effects of FPN, on TBARS levels, antioxidants, and present anti-hyperglycemic property.  相似文献

The size of a fish is an important factor in its physiology, and metal uptake is affected by animal physiology. In this study, small and large tilapias (Oreochromis niloticus) differing approximately twofold in length and fivefold in weight were compared for their antioxidant response. Both groups were exposed to Cu or Cr (1.0 μg/mL) in a freshwater (?80 mg CaCO3/L, conductivity 1.77 mS/cm) using 2 exposure protocols (20 μM for 48 h and 10 μM for 6 days). Following the exposures, the antioxidant enzyme activities (superoxide dismutase, SOD; catalase, CAT; glutathione peroxidase, GPX; glutathione reductase, GR and glutathione S-transferase, GST) and glutathione (GSH) levels were measured in the liver of fish. Results showed that small fish was affected from exposure conditions much more than large ones as their antioxidant parameters significantly decreased even in controls. Metal exposures of small fish caused significant increases in SOD and CAT activity in acute Cu or Cr exposures. Subchronic Cr exposure of small fish also caused significant increases in CAT, GPx and GST activities, while there was no significant change in Cu-exposed ones. Large fish, however, showed different antioxidant responses as their levels mostly decreased. This study demonstrated that the response of antioxidant system in the liver of tilapia varied in relation to fish sizes and emphasized using different size groups in environmental monitoring and also in evaluation of fish biomarkers.  相似文献

Several diets employed in aquaculture are enriched with selenium (Se), as it is a fundamental element to aquatic vertebrates. Diphenyl diselenide [(PhSe)2], which is a synthetic organoselenium compound, has been considered a potential antioxidant agent in different experimental models. Thus, the aim of this study was to evaluate the effects of dietary diphenyl diselenide at concentrations of 1.5, 3.0, and 5.0 mg/kg for 60 days and to determine its optimal supplemental level for carp, Cyprinus carpio. Neither growth retardation nor hepatoxicity was induced by the inclusion of diphenyl diselenide at concentrations ranging from 1.5 to 5.0 mg/kg. In addition, the inclusion of 3.0 mg/kg of diphenyl diselenide stimulated the weight and length of the carp. The supplementation with 1.5 and 3.0 mg/kg of diphenyl diselenide did not produce oxidative damage in the tissues, verified by peroxidation lipid and protein carbonyl assays. However, at 5.0 mg/kg, it caused an increase of the lipid peroxidation in the liver, brain, and muscle, and inhibited the cerebral acetylcholinesterase activity. An increase of the hepatic superoxide dismutase activity and non-protein thiols content in all tissues and ascorbic acid in the liver, gills, and brain was verified in carp fed with the diet containing 3.0 mg/kg of diphenyl diselenide. This diet had advantageous effects for the fish used in experiments. Therefore, this compound could be considered a beneficial dietary supplement for carp nutrition.  相似文献

This study investigates the protective effect of betanin against liver injury induced by carbon tetrachloride (CCl4) in common carp (Cyprinus carpio L.). The fish were treated with 1, 2, and 4 % betanin in fodder throughout the experiment. After 20 days of treatment, the fish were intraperitoneally injected with 20 % (v/v in peanut oil) CCl4 at a volume of 0.5 mL/kg body weight. The fish were killed 3 days after CCl4 intoxication, and then, histological and biochemical assays were performed. Results showed that CCl4-induced liver CYP2E1 activity, oxidative stress, and injury, as indicated by the depleted glycogen storage, increased serum aspartate aminotransferase (AST)/alanine aminotransferase (ALT) activities and liver histological damage. Compared with the CCl4 control group, the betanin-treated groups exhibited reduced CYP2E1 activity, decreased malondialdehyde level, increased liver antioxidative capacity (increased glutathione level and superoxide dismutase and catalase activities), increased liver glycogen storage, and reduced serum AST/ALT activities, with significant differences in the 2 and 4 % groups (p < 0.05). Histological assay further confirmed the protective effect of betanin. In conclusion, betanin attenuates CCl4-induced liver damage in common carp. Moreover, the inhibition of CYP2E1 activity and oxidative stress may have significant roles in the protective effect of betanin.  相似文献

Six oxidized fish oil contained diets were formulated to investigate the effect of graded levels of vitamin E (VE) (α-tocopherol acetate: 160, 280, and 400 mg kg?1) associated with either 1.2 or 1.8 mg kg?1 selenium (Se) on growth, body composition, and antioxidant defense mechanism of juvenile largemouth bass. Another control diet containing fresh fish oil with 160 mg kg?1 VE and 1.2 mg kg?1 Se was also prepared. Over a 12-week feeding trial, about 5 % of Micropterus salmoide fed diet OxSe1.2/VE160 showed inflammation and hemorrhage symptoms at the base of dorsal, pectoral, and tail fin. Fish in all treatments survived well (above 90 %). Feed intakes (88.42?89.58 g fish?1) of all treatments were comparable. Growth performances (weight gain and specific growth rate) and feed utilization (feed and protein efficiency ratio) were significantly impaired by dietary oil oxidation, and they did not benefit from neither VE nor Se supplementation. Regardless of dietary VE and Se supplementation, oxidized oil ingestion resulted in markedly decreased hepatosomatic index and intraperitoneal fat ratio. Oxidized oil ingestion also induced markedly lower liver and muscle lipid contents, and these effects could be alleviated by dietary Se supplementation. Dietary oil oxidation stimulated hepatic catalase activities relative to the control, and supplementation of VE abrogated this effect. Hepatic reduced glutathione content in the control was markedly higher than that of treatment OxSe1.2/VE160, without any significant differences comparing with the other oxidized oil receiving groups. Hepatic glutathione peroxidase activity and liver Se concentration reflected dietary Se profile, whereas liver VE level reflected dietary VE profile. Compared with the control, fish fed diet OxSe1.2/VE160 obtained markedly higher serum, liver and muscle malondialdehyde contents, which droppe significantly with increasing either VE or Se supplementation. In conclusion, the overall results in this study suggested that both VE and Se inclusion could protect largemouth bass from the oxidative damage challenged by dietary oil oxidation.  相似文献

This experiment was conducted to compare the effects of different sources of selenium (Se) on growth, muscle Se concentration, muscle proximate composition, blood enzymes and antioxidant status of common carp, Cyprinus carpio. Organic Se (selenomethionine, SeMet), inorganic Se (sodium selenite, Na2SeO3), or nanoselenium were each added to the basal diet at 0.7 mg Se kg?1 diet. Four groups of fish with an average weight of 9.69 ± 0.12 g were fed one of the experimental diets for 8 weeks. Nano‐Se diet resulted in better growth performance (P < 0.05). Fish fed with Se‐supplemented diets showed a higher (P < 0.05) Se content in the muscle and liver tissues. Fish fed nano‐Se had the highest liver glutathione peroxidase, superoxide dismutase and catalase activities (P < 0.05). Liver malondialdehyde level was significantly (P < 0.05) lower in fish fed on SeMet and nano‐Se diets as compared to control. The carp fed on nano‐Se diet had the lowest values of aspartate transaminase and alanine transaminase. Lactate dehydrogenase activity was significantly lower in fish fed on SeMet and nano‐Se diets. This study shows that nano‐Se acts more efficiently on growth performance and antioxidant defence system of common carp than organic and inorganic sources of Se.  相似文献

This study was conducted to study the effects of dietary zinc on lipid peroxidation, protein oxidation and antioxidant defence of juvenile Jian carp (Cyprinus carpio var. Jian) by feeding fish with increasing levels of zinc (15.3, 26.9, 40.8, 58.2, 68.9 and 92.5 mg Zn kg?1) for 6 weeks. Results indicated that malondialdehyde (MDA) content and protein carbonyls (PC) in serum were the highest in fish fed diet containing 15.3 mg zinc kg?1 diet (P < 0.05). Serum antisuperoxide anion (ASA), superoxide dismutase (SOD), glutathione peroxidase (GSH‐Px), glutathione reductase (GR) activities and glutathione (GSH) content were improved with increasing dietary zinc levels up to 40.8 mg zinc kg?1 diet (P < 0.05) and levelled off (P > 0.05). Serum antihydroxy radical (AHR), catalase (CAT) and glutathione‐S‐transferase (GST) activities followed the similar pattern to that observed in ASA. The MDA and PC levels, ASA, AHR, SOD, CAT, GPx, GR, GST activities and GSH content in intestine, hepatosomatic and muscle tissue followed the similar pattern to that observed in serum. The present results indicated that zinc decreased lipid peroxidation and protein oxidation and improved antioxidant defence in fish.  相似文献

An 8-week feeding trial was conducted to compare the effects of supplementing (0.7 mg kg?1) different dietary selenium (Se) sources including organic [selenomethionine (SeMet)], inorganic [sodium selenite (Na2SeO3)], and nanoparticulate Se (nano-Se) on physiological responses of common carp, Cyprinus carpio juveniles (9.7?±?0.1 g). Basal diet without Se supplementation used as control. Fish fed nano-Se supplemented diet had the highest weight gain (97.2?±?10.8%) and feed efficiency ratio (42.4?±?0.8%). Intestinal villi height was significantly taller in fish fed nano-Se diet than in the control group in both foregut and midgut sections. Serum glutathione peroxidase and superoxide dismutase activities were significantly higher in nano-Se and SeMet groups than in control and sodium selenite groups. Fish fed Se-supplemented diets had greater red blood cell counts and hematocrit and hemoglobin values than the control group (P?<?0.05). Nano-Se and SeMet groups showed a significant increase in white blood cell counts, neutrophil percentage, and serum lysozyme activity than the other groups. Fish fed nano-Se diet had the highest serum hemolytic activity, total immunoglobulin, and total protein and albumin contents, as well as the lowest serum total cholesterol and low density lipoprotein levels (P?<?0.05). Overall, significant improvements in growth performance, feed utilization, intestinal morphology, and hemato-immunological and serum biochemical parameters of common carp juveniles suggest nano-Se as an efficient source for providing dietary Se in this species.  相似文献

We studied accumulation and biochemical effects of microcystin-LR (MCLR) in Odontesthes hatcheri after dietary administration of the cyanobacteria Microcystis aeruginosa (1.3 μg MCLR/g body mass, incorporated in standard fish food). After 12 h, MCLR content in liver did not differ between fish fed with crushed or intact cells, demonstrating O. hatcheri’s capacity to digest cyanobacteria and absorb MCLR. In the second experiment, fish received toxic cells, non-toxic cells, or control food; MCLR accumulation was monitored for 48 h. Protein phosphatase 1 (PP1), catalase (CAT), glutathione-S-transferase (GST) activities, and lipid peroxidation (as MDA) were measured in liver and intestine. Methanol-extractable MCLR was determined by PP1 inhibition assay (PPIA); extractable and protein-bound MCLR were measured by Lemieux oxidation-gas chromatography/mass spectrometry (GC/MS). MCLR accumulated rapidly up to 22.9 and 9.4 μg MCLR/g in intestine and liver, respectively, followed by a decreasing tendency. Protein-bound MCLR represented 66 to ca. 100 % of total MCLR in both tissues. PP1 activity remained unchanged in intestine but was increased in liver of MCLR treated fish.CAT and GST activities and MDA content were significantly increased by MCLR only in liver. We conclude that O. hatcheri is able to digest cyanobacteria, accumulating MCLR mostly bound to proteins. Our data suggest that this freshwater fish can be adversely affected by cyanobacterial blooms. However, the rapid decrease of the detectable MCLR in both tissues could imply that sublethal toxin accumulation is rapidly reversed.  相似文献

The impacts of bacterial infection on cultivated fish species, African catfish, were investigated using oxidative stress biomarkers [lipid peroxidation (LPO) and protein carbonylation] and the activities of important antioxidant/detoxifying enzymes [catalase and glutathione S-transferase (GST)]. Fish were inoculated via oral gavage with one of the following treatments: 1 × 105 CFU/ml of Escherichia coli (EC1), 2 × 105 CFU/ml of E. coli (EC2), 1 × 105 CFU/ml of Vibrio fischeri (V1), 2 × 105 CFU/ml of V. fischeri (V2), gavaged with distilled water and not gavaged. Fish were maintained in the laboratory for 7 days after the bacterial inoculation, and the levels of LPO, protein carbonylation, GST, and catalase activities were determined in the muscle, gills, and liver of fish. Fish inoculated with bacteria (either E. coli or V. fischeri) had a significant higher levels of tissue LPO, protein carbonylation, and GST activities in a tissue-specific pattern (liver > muscle > gills). This appears to be related with the levels of bacterial inoculation, with effects more pronounced in fish inoculated with either EC2 or V2. The catalase activity did not differ significantly between the inoculated and fish that were not inoculated. The results of this study indicate that bacterial inoculation could result in oxidative stress in fish, and liver has a higher rate of oxidative stress per mg tissue compared to the gills and the muscle.  相似文献

Glutathione (GSH) and related enzymes, glutathione transferase (GST), glutathione peroxidase (GPx) and glutathione reductase (GR) form an important phase 2 biotransformation enzymes system. The objective of this study was to compare this enzymes system in three fish species from the river Nile, Oreochromis niloticus, Claris lazera and Cyprinus carpio in order to establish the main differences and to purify and characterize GST from the liver of O. niloticus.The level of GSH and the activity of GST, GPx and GR in the liver, kidney and gills of the three fish species were examined. A simple reproducible procedure for the purification of GST from the liver of O. niloticus to homogeneity, which includes chromatography on DEAE- cellulose followed by affinity chromatography on GSH-sepharose was established. The molecular mass was found to be 25,460 Da by SDS-PAGE. The Michaelis-Meneten constants (Km) of the enzyme for GSH and CDNB were 0.35 mM and 0.42 mM, respectively. The affinity purified enzyme exhibited maximum pH at pH 8.0 and increasing pH above 8.0 did not affect the observed maximum. The purified enzyme acts readily on CDNB, less readily on some standard transferase substrates (1,2-dichloro-4-nitrobenzene and p-nitrophenethyl bromide) and not at all on others (bromosulphophthalein and p-nitrobenzyl chloride). Bromosulfophthalein, cibacron blue and hematin inhibited CDNB-conjugating activity of the purified enzyme with IC50 0.079, 3.98 and 0.126 μM, respectively.  相似文献

Complete dietary fish oil replacement with palm or poultry oil in barramundi (Lates calcarifer) had no detrimental effects on growth or hepatosomatic index of juvenile fish up to an average size of ~50 g. However, it significantly decreased the omega-3 (n-3) long-chain polyunsaturated fatty acid content of the fish muscle (fillet) lipids. This was particularly true for eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) which are recognised for their health beneficial effects in the human diet. As a result of their decreased EPA and DHA content, the peroxidation index of the muscle lipids was also decreased. This was associated with increased simulated retail storage shelf life as indicated by decreased thiobarbituric acid reactive substances in muscle samples from fish fed the palm or poultry oil-based diets. Concomitantly, glutathione peroxidase (GPx) activity, but not glutathione S-transferase (GST) activity or reduced glutathione concentration, was significantly reduced in the liver of barramundi fed the palm or poultry oil-based diets as compared with the fish fed the fish oil-based diet. Furthermore, GPx and GST activity were very low in muscle, much lower than in gastrointestinal tract, liver or swim bladder. Therefore, we propose that liver GPx activity may be a good predictor of fillet shelf life in barramundi and other fish species.  相似文献

The present study explored the effects of thiamin on antioxidant capacity of juvenile Jian carp (Cyprinus carpio var. Jian). In a 60-day feeding trial, a total of 1,050 juvenile Jian carp (8.20 ± 0.02 g) were fed graded levels of thiamin at 0.25, 0.48, 0.79, 1.06, 1.37, 1.63 and 2.65 mg thiamin kg?1 diets. The results showed that malondialdehyde and protein carbonyl contents in serum, hepatopancreas, intestine and muscle were significantly decreased with increasing dietary thiamin levels (P < 0.05). Conversely, the anti-superoxide anion capacity and anti-hydroxyl radical capacity in serum, hepatopancreas, intestine and muscle were the lowest in fish fed the thiamin-unsupplemented diet. Meanwhile, the activities of catalase (CAT), glutathione peroxidase, glutathione S-transferase and glutathione reductase, and the contents of glutathione in serum, hepatopancreas, intestine and muscle were enhanced with increasing dietary thiamin levels (P < 0.05). Superoxide dismutase (SOD) activity in serum, hepatopancreas and intestine followed a similar trend as CAT (P < 0.05). However, SOD activity in muscle was not affected by dietary thiamin level (P > 0.05). The results indicated that thiamin could improve antioxidant defence and inhibit lipid peroxidation and protein oxidation of juvenile Jian carp.  相似文献

Lipid peroxidation, protein oxidation and antioxidant activities of muscle, intestine, hepatopancreas and serum in juvenile Jian carp (Cyprinus carpio var. Jian) were investigated after feeding graded levels of biotin (0.010, 0.028, 0.054, 0.151, 0.330, 1.540 and 2.680 mg kg?1 diet) for 63 days. Both malondialdehyde and protein carbonyl content in all studied tissues and serum were the lowest in fish fed diets containing 0.151–0.330 mg biotin kg?1 diet and then increased in fish fed the diet with 2.680 mg biotin kg?1 diet (P < 0.05). Similarly, glutamate–oxaloacetate transaminase and glutamate–pyruvate transaminase activities in serum significantly decreased with biotin levels up to 0.151 mg kg?1 diet (P < 0.05). Conversely, capacities of anti-hydroxyl radical (AHR) and anti-superoxide anion (ASA) in the detected tissues and serum significantly improved with biotin levels up to 0.054–1.540 mg kg?1 diet and then decreased in 2.680 mg biotin kg?1 diet group for muscle and intestinal AHR as well as hepatopancreas ASA (P < 0.05). Activities of superoxide dismutase in all studied tissues and serum significantly elevated with biotin levels up to 0.330 mg kg?1 diet and then decreased when fish fed the diet with 2.680 mg biotin kg?1 diet, except intestine (P < 0.05). Meanwhile, activities of catalase, glutathione peroxidase, glutathione-S-transferase and glutathione reductase and total thiol content in all studied tissues and serum showed the upward trend with biotin supplementations (P < 0.05). These results indicated that biotin improved antioxidant status and depressed lipid peroxidation and protein oxidation in all studied tissues and serum.  相似文献

In cytosol from liver of pacu, Piaractus mesopotamicus, a hypoxia-tolerant fish that dwells in Pantanal, we found an enzyme activity capable of modulating the alkenal 4-hydroxy-2-nonenal (HNE) by conjugating it with glutathione (GST–HNE activity). HNE is a downstream metabolite from the oxidation of polyunsaturated fatty acids by reactive oxygen species arisen from mitochondria of animal cells. HNE production may increase more intensively under oxidative stress. Harmful effects to cell survival may occur when HNE increases over 10?4 M. Pacus submitted to hypoxia in July (cold season in Pantanal) showed 40 % less of this GST–HNE conjugating activity in their liver cytosol. Injecting pacus subjected to hypoxia during the cold season with a summer physiological dose of melatonin caused their liver cytosolic GST–HNE activity to increase up to the levels found in the warm season. From October to March (warm season in Pantanal), pacus are prone to oxidative stress particularly during potamodromous active oxygen-demanding swimming, when they migrate up rivers to spawn. Thus, our findings point out that the higher levels of melatonin in circulation during the summer are important to avoid the increase of 4-HNE inside liver cells of this fish species.  相似文献

The present study is aiming at evaluating the hepatoprotective and antioxidant effects of Astragalus polysaccharide (APS) on the carbon tetrachloride (CCl4)-induced hepatocyte and liver injury in common carp in vitro and in vivo. In vitro, APS (200, 400 and 800 μg/ml) was added to the carp primary hepatocytes before (pre-treatment), after (post-treatment) and both before and after (pre- and post-treatment) the incubation of the hepatocytes with CCl4 at 8 mM in the culture medium. APS at concentrations of 200, 400 and 800 μg/ml significantly improved cell viability and inhibited the elevation of glutamate pyruvate transaminase (GPT), glutamate oxalate transaminase (GOT), lactate dehydrogenase (LDH) and malondialdehyde (MDA) and significantly increased the reduced level of superoxide dismutase (SOD). In vivo administration of APS at the doses of 1.5 and 3 g/kg in the diet for 60 days prior to CCl4 intoxication significantly reduced the elevated activities of GPT, GOT and LDH and increased the reduced levels of total protein and albumin in the serum; meanwhile, the reduced levels of SOD, glutathione and total antioxidant capacity (T-AOC) were markedly increased and the MDA formation was significantly inhibited in liver tissue. Overall results proved the hepatoprotective action of APS, which is likely related to its antioxidant activity. The results support the use of APS as a hepatoprotective and antioxidant agent in fish.  相似文献

In the present study, we tested the hypothesis that dietary histidine could improve antioxidant capacity of juvenile Jian carp (Cyprinus carpio var. Jian). A total of 1,200 juvenile Jian carp were fed graded levels of histidine at 2.3 (unsupplemented control), 4.4, 6.3, 8.6, 10.8 and 12.7 g/kg diet for 60 days. Results showed that the content of malondialdehyde (MDA) and protein carbonyl (PC) in serum and all tissues apparently decreased with increasing histidine levels up to an optimal level and increased thereafter. Anti-superoxide anion (ASA) capacity, glutathione peroxidase (GPX) activities and glutathione (GSH) content in serum and all tissues, anti-hydroxyl radical (a-HR) capacity, catalase (CAT) and glutathione-S-transferase (GST) activities in serum, muscle and intestine, superoxide dismutase (SOD) activities in serum and intestine, as well as glutathione reductase (GR) activity in serum, muscle and hepatopancreas were improved by dietary histidine. Fish fed diet with 8.6 g/kg histidine had lower serum glutamate-pyruvate transaminase (GPT) activity than that fed with control diet, whereas pattern of glutamate–oxaloacetate transaminase (GOT) activity was opposite. The present results suggested that histidine could improve antioxidant capacity and inhibit lipid peroxidation and protein oxidation of juvenile Jian carp.  相似文献

One method of increasing the value of aquacultured product is to produce fillets that are fortified with minerals that are beneficial to human health – that is enhance the functionality of an already healthy product. A good candidate mineral in this regard is selenium (Se) which is of vital importance to normal metabolism in humans. In order to evaluate the dose response and tissue accumulation of supplemental dietary Se, a study was undertaken with hybrid striped bass (HSB). Animals were fed diets supplemented with either organic (0–3.2 mg kg?1 as SelPlex®) or inorganic (0.2 and 0.4 mg kg?1 as sodium selenite) Se for 6 weeks. Because basal fishmeal‐based diets contained 1.22 mg Se kg?1, doses of Se delivered equated to 1.22–4.42 mg kg?1. At trial end, greatest weight gain was observed in fish receiving 0.2 mg Se kg?1, irrespective of form (organic/inorganic). Se accumulation in HSB liver and fillet revealed a classical dose‐response once a threshold level of 0.2 mg Se kg?1 was surpassed. Greatest tissue accumulation of Se was observed in fish fed the 3.2 mg Se kg?1 level (P > 0.0001). A 100 g portion of Se‐enhanced HSB fillet would contain between 33 and 109 μg Se, amounting to a dietary intake of between 25 and 80 μg Se; a level that would satisfy present daily intake recommendations. Comparison of tissue Se levels indicated that the muscle provides a more conspicuous gauge of dietary Se dose‐response than does liver. Dietary treatments of between 0.4 and 1.6 mg organic Se kg?1 reduced (P < 0.024) plasma glutathione peroxidase (GSH‐Px) activity. No differences were observed in ceruloplasmin, lysozyme or GSH‐Px activities between organic and inorganic Se when delivered at the 0.2 mg Se kg?1 level. Ceruloplasmin, lysozyme and GSH‐Px levels were elevated (P ≥ 0.025) in fish fed the diet containing 0.4 mg inorganic Se kg?1.  相似文献

This study was conducted to compare the effects of dietary supplementation of selenium (Se, both inorganic and organic) and mannan oligosaccharide (MOS) on the growth, immune function, and antioxidant enzyme activities in the haemolymph of the marron Cherax cainii (Austin, 2002); 0.4 mg kg?1 of sodium selenate, 0.2 g kg?1 Sel-Plex®, or 0.4 % Bio-MOS® was added to the basal diet as sources of inorganic selenium (IS), organic selenium (OS), and MOS, respectively. After 90 days, marron fed all 3 supplemented test diets had significantly higher final weight, specific growth rate (SGR), survival, total haemocyte count (THC), percentage of hyaline cells, glutathione-S-transferase (GST), and glutathione peroxidase (GPx) activities than marron fed without any supplements. The marron fed OS showed significantly higher THC and percentage of hyaline cells than those fed IS and MOS. Marron fed MOS showed the highest final weight, SGR, and percentage of granular cells; however, there was no significant difference in the neutral red time retention between marron fed IS and OS, whereas marron fed OS resulted in the highest GPx activity and total Se levels in the haemolymph but the lowest lipid peroxidase activity; therefore, it is suggested that dietary supplementation with any source of Se and MOS is beneficial to improving growth, survival, and antioxidant activity.  相似文献

Diludine is used as an effective agent reducing mutagenic effects of environmental pollutants, increasing productivity and protecting reproductive system of parental individuals and quality of their offspring. A 10‐week study was conducted to evaluate potential effects of diludine on rainbow trout (Oncorhynchus mykiss) juveniles growth performance, feed utilization, survival rate, liver antioxidant enzyme activities (superoxide dismutase, SOD, EC1.15.1.1 ; catalase, CAT, EC1.11.1.16 ; glutathione peroxidase, GPx, EC 1.11.1.9 ; glutathione reductase, GR, EC1.6.4.2 ; glucose‐6‐phosphate dehydrogenase, G6PD, EC1.1.1.49 and glutathione S‐transferase, GST, EC1.5.1.18 ), the levels of malondialdehyde (MDA) as a lipid peroxidation marker and muscular trace elements at a low water temperature (9 °C). Diludine was added at 0.2, 0.5 and 1 g kg?1 to a fish meal–based control diet, and each diet was fed to triplicate groups of rainbow trout juveniles. After 10 weeks, there were no differences in survival rate between fish fed experimental diets (P > 0.05). It was observed that a significant improvability existed for both growth and feed utilization in fish fed diets supplemented with diludine (P < 0.05). Antioxidant enzymes showed significantly increased activity in liver tissues (P < 0.05) and then a decrease to initial activity levels during the experimental time. (GPx activity was not determined in the juvenile livers.) SOD and GST activities in liver of fish fed the diets with diludine tended to be higher and GR activity tended to be lower than that in fish fed the basal diet. However, in general, it was determined that different concentration of diludine did not affect the studied antioxidant enzyme activities except G6PD in the liver of juveniles fed the diet with 1 g kg?1 diludine. On the other hand, the significant reduction was found in MDA levels in the fish fed the diets with diludine compared with basal diet on the 14th and 28th days of experiment. It was also observed that Cu, Fe, Mg, Zn and Se levels were significantly higher in the juveniles fed the diet with 1 g kg?1 diludine than those fed the basal diet. In conclusion, the results of this trial indicate that 1 g kg?1 dietary supplementation with diludine seems to be most positive for rainbow trout production in cold‐water adaptation.  相似文献
