Repeat unit length and restriction site variation in ribosomal RNA geneclusters (rDNA) was surveyed in 77 Arachis accessions, includingsamples from 39 accessions of cultivated Arachis hypogaea(2n=4x=40), 36 accessions representing 15 related tetraploid and diploidwild species, and two synthetic amphidiploids. Total genomic DNA wasdigested with five restriction enzymes, and probed with three heterologousribosomal clones of wheat and broad bean. Four rDNA repeat unit lengthclasses were recognized in the Arachis species. Restriction site analysisshowed that some SacI, BamHI and TaqI cleavage sites in rDNA unit werehighly conserved. With few exceptions, the variable BamHI and EcoRV siteswere able to differentiate the taxonomic sections and species, respectively.Arachis hypogaea and A. duranensis accessions produced fourrDNA length classes. Among these, three were identical with those of otherArachis species. A SacI restriction site (s) from probe (Ver6-5) cangenerally distinguish the two subspecies A. hypogaea ssp. hypogaea and A. hypogaea ssp. fastigiata. Forty nine per centof bands were polymorphic across the A. hypogaea accessionsanalysed. This study does not support A. batizocoi to be a progenitorof A. hypogaea. For the gene array, the contribution from eachparental genome can be detected in the two synthetic amphidiploids.  相似文献

Bru1 is currently the major gene conferring brown rust resistance in sugarcane, and diagnostic markers are available. A survey for the presence of this gene was conducted on 391 genotypes including Brazilian cultivars, clones and basic germplasm. The efficiency of these markers for identifying resistant cultivars and artificially inoculated basic germplasm was also evaluated. The Bru1 frequency among cultivars (73.5%) suggests this gene is the prevalent source of brown rust resistance in Brazilian sugarcane breeding programmes. Most of the cultivars known to be resistant were positive for Bru1, although other genes for resistance could be present in lines not having Bru1. Only 17.8% of the basic germplasm accessions were positive for the Bru1 gene, and a low correlation between Bru1 diagnostic markers and brown rust severity was observed for basic germplasm accessions. Overall, Bru1 diagnostic markers proved to be efficient identifying resistant cultivars and clones and have potential to be in screening brown rust resistance in Brazilian breeding programmes.  相似文献

Arachis kempff‐mercadoi is a wild species from the section Arachis. All kempff‐mercadoi accessions originate from the Santa Cruz province of Bolivia and they represent Arachis species with the A genome. From molecular analysis it was found that although cultivated A. hypogaea is made up of A and B genomes, A. kempff‐mercadoi may not be as closely related to it as are some of the other A genome species. Arachis kempff‐mercadoi is of interest because it has multiple disease resistance. It was crossed with a Spanish A. hypogaea cultivar which is susceptible to foliar diseases and to the insect pest Spodoptera litura. The success rate of the cross A. hypogaea (2n = 40) ×A. kempff‐mercadoi (2n = 20) was very low, but it could be increased by culturing immature seeds in vitro. Although the hybrids were triploids, a few fertile pollen grains were obtained due to the formation of restitution nuclei in the F1 plants. Interspecific derivatives at the BC2F2 generation were scored for early leaf spot, late leaf spot and to Spodoptera damage. Screening results showed that 29% of the derivatives had both early and late leaf spot resistance and that less than 5% of the derivatives had resistance to both the foliar diseases and to Spodoptera.  相似文献

Summary Forage quality of various alfalfa (Medicago sativa L.) cultivars has been determined under different environmental conditions, and numerous trials with alfalfa have documented an inverse relationship between forage quality and maturity. Little information is available, however, regarding the comparative forage quality of the germplasm sources from which most USA cultivars were developed. We compared forage quality of these germplasm sources at four phenological stages under disease- and insect-free conditions in a greenhouse. Germplasm sources (cultivars) tested were: Indian (Sirsa #9), African (African), Peruvian (Hairy Peruvian), Flemish (DuPuits), Turkistan (Lahontan), Chilean (Kansas Common), M. varia (Grimm), and Ladak (Ladak). Four harvests were taken and forage was separated into four phenological stages: vegetative, early bud (1–3 buds-per-stem), late bud (>3 buds-per-stem), and bloom. The germplasm source X phenological stage interaction was significant for crude protein (CP) and in vitro digestible dry matter (IVDDM) concentrations. M. varia showed the least decline in IVDDM and CP with increasing maturity. M. varia had higher IVDDM than did African and Indian at late bud and bloom stages. Indian and Flemish had higher CP than did Turkistan and Peruvian at late bud and bloom stages. The genus Medicago encompasses many important forage species for both temperate and tropical regions. M. sativa L., commonly known as lucerne, is one of the most important forage species grown worldwide, but its production suffers seriously from weevil (Hypera postica Gyll.) infestation. The aim of this work was to identify species/accessions with tolerance to weevil and their molecular analysis using simple sequence repeat (SSR) markers. After screening 197 global germplasm encompassing 50 Medicago species for weevil tolerance, 22 lines representing 13 species were identified where leaf damage was ≤15% (P ≤ 0.05). In total, 37 accessions of the 22 lines, five Indian lucerne cultivars with leaf damage ≥75% and 10 accessions of the 13 Medicago species with low to high infestation (>25%) were molecularly assessed using 11 SSR markers (5 newly developed) to delineate closest to lucerne lines for breeding. In total, 33 bands were scored. The SAHN clustering using UPGM algorithm resulted into two main clusters supported with high boot strap values and with genetic similarity ranging from 0.33 to 0.96. Two accessions of M. tenoreana were observed closest to Indian lucerne cultivars. The rich variability revealed can be used as potential resource for transferring genes across species. Although the inter-specific hybridization is difficult preposition in genus Medicago largely due to post fertilization barrier, the identified species/accessions can be utilized on priority in breeding programs especially employing biotechnological tools like culturing of fertilized pods, ovule-embryo culture and electroporation.  相似文献

以花生属5个区组的79份野生花生种质为材料,系统鉴定了野生花生对青枯病的抗性反应,从中发掘高抗青枯病的种质15份,含匍匐区组种质3份、直立区组1份、异形花区组1份、花生区组8份、未命名种质2份,抗病材料频率达到19%,高于栽培种花生资源的抗性频率。通过SSR分析表明,在所获得的抗青枯病野生花生材料中,四倍体野生种A.monticola与栽培种花生的亲缘关系最近,其次为花生区组的二倍体野生种A.duranensis和A.chacoense。根据DNA扩增结果,绘制了抗青枯病种质的指纹图谱,明确了其SSR分子特性。  相似文献

Seed weevil (Bruchus spp.) is one of the most damaging pests to lentil crop, and the current chemical treatments to control this insect present numerous disadvantages. In this work, we have searched for resistant or almost resistant lentil cultivars to be used as potential sources of genes for resistance to bruchids as a more sustainable and environmentally friendly approach to diminish harvest loss. Thus, a lentil germplasm collection of 571 accessions from 27 different countries, and including wild species was screened for susceptibility to seed bruchids under natural field conditions in Central Spain. Infestation was evaluated in comparison with a check (lentil cultivar ??Lyda??) according to the number of eggs per pod and the infestation rate in seeds (IR). A large variation of IR (0?C70?%) was observed among accessions although, in general, wild species were consistently less affected by bruchids than the check. A total of 32 accessions, including Lens culinaris Medikus subsp. culinaris, Lens culinaris Medikus subsp. orientalis (Boiss.) Ponert, Lens nigricans (M. Bieb.) Godr., and Lens lamottei Cezfr., showed lower infestation rates than the check and were selected as potential sources of resistance to this pest. Potential mechanisms of resistance of selected accessions and their inclusion in a breeding program are outlined.  相似文献

Summary Two cycles of recurrent selection for yield were previously conducted using a population of Arachis hypogaea L.-like tetraploid hybrid derivatives selected from among the progeny of a cross between A. hypogaea and A. cardenasii Krap. et Greg. nom. nud., a diploid species with resistance to late leafspot (Cercosporidium personatum (Burk. and Curt.) Deighton). Using the 10 highest yielding parents from each of the above cycles of recurrent selection, a study was conducted for 12 morphological traits and susceptibility to leafspot infection in a replicated test at a single location. The two parents used to initiate the interspecific hybrid population (PI 261942-3 and A. cardenasii) as well as two adapted cultivars (Florigiant and NC 7) were included as checks. The objectives of the study were to determine the amount of genetic variation remaining in the population after two cycles of recurrent selection in order to predict whether further progress from selection could be expected and determine the potential for utilizing wild species for the improvement of quantitative traits in peanut. The results indicate that significant levels of genetic variation remain in the population after two cycles of recurrent selection for all traits measured, including several components of yield and leafspot resistance. This suggests that continued progress in population improvement from further cycles of selection should be possible while enhancing the genetic diversity of cultivated peanut germplasm.Paper no. 12347 of the Journal Series of the North Carolina Agric. Summary About 1000 Brassica oleracea accessions were evaluated in glasshouse tests for response to Plasmodiophora brassicae (clubroot). Resistance was confirmed in some north and west European kales and cabbage. A new source of resistance in cabbage, from Eire, is reported. Most other accessions were highly susceptible but lower levels of susceptibility were observed in open pollinated Brussels sprouts and forms of south European cabbage, cauliflower and broccoli. Modern breeding (as in the production of hybrid cultivars) appears to have resulted in increased susceptibility in several crop types. The implications of these results for the exploitation of germplasm are discussed.  相似文献

A species level germplasm collection representing 76% of known taxa in the genus Vigna subgenus Ceratotropis was evaluated for resistance to two species of bruchid beetles, Callosobruchus chinensis and C. maculatus. Seven taxa consisting of 29 accessions were found to be resistant to C. chinensis and 4 taxa consisting of 24 accessions were found to be resistant to C. maculatus. This compared with no resistant accessions being found in several hundred landrace accessions of mungbean, V. radiata var. radiata, in the same subgenus. Sometimes resistance was found in all accessions of a particular taxon, such as complete resistance to both C. chinensis and C. macualtus in V. umbellata. Other taxa showed intra taxon variation for resistance such as V. reflexo-pilosa andV. minima. The levels and patterns of resistance among taxa were diverse. The results suggest that various factors cause resistance to bruchid in the subgenus Ceratotropis. While the number of eggs laid on seeds generally reflected seed size, one small seeded cultivar of V. mungo var. mungo, black gram, had an unusually high number of eggs laid per seed. No correlation was found between seed size and levels of resistance. The species level germplasm collection, which reflects the core collection concept in trying to maximize genetic diversity in a limited number of accessions, has enabled a large number of potentially useful sources of resistance to bruchid beetles to be found efficiently. Bananas cultivated for export all belong to Cavendish cultivars and are all recognized as very susceptible to nematodes, particularly to the burrowing nematode Radopholus similis and the lesion nematode Pratylenchus coffeae. Even if there have been many changes in the management of banana nematodes in large commercial banana plantations, chemical control still remains most often the last resort method to manage the nematodes, although the number of registered products is definitely declining. Therefore, nematode control though genetic improvement is gaining new interest worldwide. In this study, 55 banana accessions mostly diploids from the Musa acuminata genome group (AA) but including some triploid accessions (AAA), some diploids of the Musa balbisiana genome group (BB) and some interspecific hybrids (AAB, AB) were evaluated for resistance to four nematode species R. similis, P. coffeae, Meloidogyne incognita and M. arenaria. These experiments were conducted in a growth chamber under controlled conditions. All banana accessions were susceptible to nematode species, although many different levels of susceptibility were detected. This study confirmed the good resistance status to R. similis of some cultivars from the Pisang jari buaya and Pisang batuau subgroups and the partial resistance of 17 diploid accessions significantly different from the susceptible reference cv. Grande Naine. This study also showed that 12 diploid accessions exhibited a partial resistance to P. coffeae, including some usual or potential genitors belonging to the wild diploids subspecies burmannica (cvs. Long Tavoy 1 and 2) and burmannicoides (cv. Calcutta 4). No source of resistance to Meloidogyne spp. was found. These screening results, combining for the first time four nematode species, are discussed within the scope of banana breeding in order to produce parental diploid lines with single or combined nematode resistances and further develop triploids that can substitute existing susceptible commercial cultivars.  相似文献

Diversity in 28 accessions representing 12 species of the genus, Cajanus arranged in 6 sections including 5 accessions of the cultivated species, C. cajan, and 4 species of the genus Rhyncosia available in the germplasm collection at ICRISAT was assessed using RFLP with maize mtDNA probes. Cluster analysis of the Southern blot hybridization data with 3 restriction enzymes – 3 probe combinations placed the genus Rhyncosia in a major group well separated from all the species belonging to the genus Cajanus. Within the genus Cajanus, the 4 accessions of C. platycarpus belonging to section Rhynchosoides formed a separate group in contrast to those in other sections of pigeonpea. In the section, Cajanus all the 5 accessions of C. cajan were grouped together and C. cajanifolius belonging to the same section was in a subgroup by itself closer to the main group. The four accessions of C. scarabaeoides, were together and the other species belonging to section Cantharospermum were in different subgroups. The intra-specific variation was seen even within accessions of certain pigeonpea wild species such as C. scarabaeoides, C. platycarpus, C. acutifolius, and even the cultivated species of C. cajan. This study suggests that RFLP of mtDNA can be used for the diversity analysis of pigeonpea and it gives some indications on the maternal lineage among the species. The variations in the mitochondrial DNA hybridization patterns also suggest the extensive rearrangement of the organelle genome among the Cajanus species. Summary The existence of structural differentiation between genomes in section Arachis of the genus Arachis has important implications in the utilization of diploid wild species in this section as a germplasm resource. Maximum expression of desirable characters may not be achieved unless tetrasomic dose levels can be achieved. Possible breeding strategies discussed include natural and induced gene exchange between genomes and chromosome substitution which could be brought about by manipulation of ploidy level and where appropriate the use of ionizing radiation. Summary The increasing popularity of organic agriculture and health food products has led to a renewed interest in hulled wheat species such as emmer (Triticum turgidum ssp. dicoccumSchubler). Knowledge on agronomic and quality traits is required for effective and efficient use of germplasm collections in breeding programs. The objective of this study was to estimate agronomic and grain quality traits of emmer wheat cultivated in Italy. A total of 20 emmer accessions consisting of landraces, breeding lines or cultivars selected from landraces and modern cultivars were examined under low input conditions. The study was conducted for three successive years (2002–2004) at one location of Southern Italy (Foggia). The entries were characterized for agronomic and qualitative traits [grain yield (t ha−1), thousand grain weight (g), test weight (kg hl−1), grain protein content (%), HMWG composition, dry gluten content (%), gluten index and yellow index, alveograph indices and Total Organic Matter (TOM) on cooked pasta]. The results showed a large genetic variability for most of the traits measured and, even if most of the accessions showed inferior bread- and pasta-making performance, modern cultivars exhibited improved quality traits with some potential to perform healthy and tasty food.  相似文献

Summary The genus Paspalum contains a number of important forage grasses, including P. dilatatum Poir. (dallisgrass) and P. notatum Flugge (bahiagrass). Even though many Paspalum species are adapted to moist, humid areas, they frequently are subjected to extended periods of drought stress which reduces forage productivity. This study was initiated to determine the range of variation for different indicators of drought tolerance in several Paspalum species. Fifteen species representing a broad cross section of germplasm were grown in a replicated field nursery and sampled for leaf epicuticular wax content on four dates over three years. Wax was extracted with chloroform and quantified by the dichromate oxidation technique. Heat tolerance (solute leakage technique) was also determined by calculating the ratio of the conductivity of the external bathing solution of leaves following a heat treatment to the conductivity of the bathing solution after leaves were boiled. Wax values of the entries ranged from 5.7 to 0.88 mg/dm2. Both relative rankings and absolute wax values were very consistent across all sampling dates. The accessions of Paspalum nicorae Parodi and P. quandrifarum Lam. were consistently high in wax content, while the accessions of P. commune Lillo and P. unispicatum (Scribn. et Merr.) Nash were consistently low. The heat tolerance testing demonstrated that the accession of P. notatum var. saurae Parodi was the most heat tolerant of the entries. Results from this investigation demonstrate the wide range of diversity that exists for both epicuticular wax and heat tolerance within the genus Paspalum.  相似文献

This experiment was carried out to investigate whether and how much field resistance to late blight, caused by Phytophthora infestans, is present in the local cultivated potato germplasm. In total 36 entries were compared in a field experiment in an area highly conducive to late blight development. Of the 36 cultivars 32 were local cultivars belonging to five Solanum species, S. tuberosum (1 accession), S. andigena (18), S. juzepczukii (2), S. stenotomum (9) and S. ajanhuiri (2). The other four cultivars were derived from breeding programmes, one being the Dutch cultivar Alpha used as a highly susceptible control. The 36 cultivars were planted according to a simple 6 × 6 lattice design with three replicates. Each replicate was divided in six incomplete blocks each with six cultivars. The disease severity was assessed weekly during 9 weeks starting 48 days after planting. The area under the disease progress curve (AUDPC) was used as a measure of the field resistance. Nine isolates from surrounding potato fields were tested for their virulence to the resistance genes R1–R11 using 22 differential cultivars. The components of the field resistance of 19 of these cultivars were compared in the greenhouse using a local isolate with virulence to all known R-genes, except to R9. The nine isolates represented seven races with a race complexity varying from 7 to 10 virulence factors. All isolates carried virulence against R1, R2, R3, R7, R10 and R11, while virulence against R9 was absent. The AUDPC among the 32 local cultivars ranged from very large, significantly larger than that of ‘Alpha’ to very small. The AUDPC from S. stenotomum accessions ranged from very large to intermediate, those from S. andigena accessions from large to very small. Especially among the S. andigena accessions interesting levels of field resistance were found. Four components of field resistance were assessed, latency period (LP), lesion size (LS), lesion growth rate (LGR) and relative sporulation area (RSA). All four showed a considerable variation among the cultivars. The LP ranged from 3½ to 6 days. The LS ranged from 225 mm2 to 20 mm2. The LGR varied about six-fold, the RSA more than 10-fold. The components tended to vary in association with one another. LP and LGR were well associated with each other and had a significant correlation with the AUDPC.  相似文献

Genetic relationships were examined among thirty germplasm accessions belonging to five Ocimum species using RAPD markers. A very high degree of polymorphism (98.20%) was observed. UPGMA cluster analysis of genetic similarity indices grouped all the accessions into two major clusters corresponding to previously reported botanical sections. Intra-clustering within the two clusters precisely grouped the accessions belonging to one species in one sub-cluster as expected from their genetic background. Our results show that RAPD technique is a sensitive, precise and efficient tool for genomic analysis in Ocimum species, that may be useful in future studies, by assigning new unclassified germplasm accessions to specific taxonomic groups and reclassifying previously classified accessions of other Ocimum species by traditional criteria on a more objective basis.
