Eight lupin (Lupinus angustifolius L.) genotypes grown at four locations in south central Alberta in 2004 were evaluated for variability in phenolic constituents and antioxidant activity measured by a photochemiluminescence assay. Genotype was the main source of variation for content of phenolic compounds and antioxidant activities. Phenolic compounds in genotypes varied minimally from 11.9 to 14.7 mg catechin equivalent and 4.15 to 4.95 mg rutin equivalent g(-1) lupin for total phenolic and flavonoid contents, respectively. Lupin genotypes exhibited weak antioxidant activity based on water-soluble substances (ACW) of 0.54 to 1.07 micromole Trolox equivalent antioxidant capacities (TEAC)/g with lag time ranging from 70 to 153 s and an antioxidant index of 6.7 to 14.5 and 1.9 to 3.3 micromole TEAC/g based on measurements of lipid-soluble substances (ACL). Antioxidant activity of lupin genotypes was not related to phenolic contents of seeds.  相似文献

Prolipid a known mixture of herbs is used as a plasma lipid lowering medicine. No side effects were registered. However, the bioactive substances of Prolipid were not investigated. Therefore in this investigation Prolipids bioactive compounds and antioxidant activity were studied. The contents of polyphenols and flavonoids were 19.87 ± 2.09 and 3.09 ± 0.31 mg gallic acid equivalent GAE/g DW and 2.09 ± 0.24 and 0.57 ± 0.05 mg catechin equivalent CE/g DW in water and methanol fractions, respectively. Anthocyanins (0.02 ± 0.001 mg/g DW) and flavanols (7.58 ± 0.81 μg CE/g DW) were found only in water fraction. The antioxidant activity of Prolipid, as determined by four different antioxidant assays [ferric-reducing/antioxidant power (FRAP); cupric reducing antioxidant capacity (CUPRAC); trolox equivalent antioxidant capacity (TEAC); 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH)], was higher in water than in methanol fraction. The correlation coefficients between polyphenols, flavonoids and antioxidant activities of Prolipid water extracts with TEAC were 0.97 and 0.90, respectively. It can be concluded that the content of polyphenol compounds in Prolipid is very high and they are the main contributors to Prolipids overall antioxidant activity. Prolipid is widely used in human treatment without known side effects on patients and is comparable to other medicinal plants, and as a strong antioxidant mixture could be used as a supplement to known atherosclerosis preventing diets.  相似文献

Three New Zealand grown tomato (Lycopersicon esculentum L.) cultivars, Aranka, Encore, and Flavourine, were analysed for colour (CIELAB L a b values), ascorbic acid, total phenolics, lycopene, and total antioxidant activity (ABTS assay). Colour analyses of the air-dried tomatoes showed that the colour degradation as measured by the L a b values was minimal, and the colour was not significantly different (p < 0.05) between each of the dried cultivars. In general, there was no significant difference (p < 0.01) between the fresh cultivars in terms of antioxidant components or total antioxidant activity. The average values obtained for antioxidant components in the three fresh cultivars were: ascorbic acid, 276 mg/100 g DM, total phenolics, 613 mg gallic acid equivalents (GAE)/100 g dry matter (DM) and lycopene 38 mg/100 g DM. Total antioxidant activity of the fresh cultivars was 2917 μmole Trolox equivalent antioxidant capacity (TEAC)/100 g DM. Following forced air-drying treatment at 42 C for 48 hr, all cultivars showed significant decreases in ascorbic acid, total phenolics and total antioxidant activity, and increases in extractable lycopene contents. This study was undertaken to identify cultivars that would provide the best colour and nutrient contents after commercial air drying.  相似文献

Ascophyllum nodosum is a brown seaweed that grows abundantly in the US Northeast coastal region. This study examined the seasonal variation of A. nodosum in phenolic contents and subsequent antioxidant, α-glucosidase and α-amylase inhibitory activities. A. nodosum was harvested monthly and extracted in hot water and the resulting extracts were spray-dried. The results indicate a clear seasonal variation in terms of phenolic content, with June and July being the highest (36.4 and 37 mg/g, respectively) and May the lowest (21.8 mg/g). The antioxidant activities, in terms of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, correlated with the phenolic contents observed (r = 0.81), with the month of July being the highest (58%) and April the lowest (26%). Similarly in terms of Trolox equivalent, July had the highest activity (15.53 μM) and April and May the lowest (8.40 and 8.27 μM, respectively). α-glucosidase inhibitory activity exhibited a pattern similar to the phenolic contents observed with July having the highest inhibitory activity (IC70 2.23 μg) and April the lowest (IC70 26.13 μg), resulting in an inverse correlation between IC70 values and total phenolic content (r = −0.89). Such seasonal variation is believed to be caused by temperature-related stress considering that A. nodosum is a cold water species.  相似文献

This study aimed to evaluate the effect of autohydrolysis temperature of corn residues in the antioxidant activity of the phenolic compounds extracted from the liquid phase. The treatments were carried out at 160, 180, 190 and 200 °C for 30 min in a pressurized batch reactor. Two different methods for phenolic compounds extraction from the autohydrolysis liquors were investigated. For that purpose, solvent extraction with ethyl acetate and acidic precipitation were performed for phenolic compounds recovery. These methods have been compared in terms of extraction yield, physicochemical properties of obtained polyphenols (characterization by Fourier Transform Infrared Spectroscopy, Thermogravimetric Analysis and Gel Permeation Chromatography), total phenolic content, total antioxidant capacity and Trolox equivalent antioxidant capacity (TEAC) values, measured in DPPH (2,2-diphenyl-1-picrylhydrazyl) test system. The maximum phenolic contents ranged from 6.04 mg GAE/100 mg extract in acidic precipitated samples to 16.45 mg GAE/100 mg extract in ethyl acetate soluble fractions. The results indicated that the ethyl acetate fractions possessed the highest antioxidant activity, reaching after 30-60 min the same capacity reported for the reference synthetic antioxidants (Trolox).  相似文献

This work was carried out to investigate the pulp composition of four mango cultivars (Haden, Tommy Atkins and Ubá) at the ripening stage in relation to three components with antioxidant potential (total phenolics, carotenoids and ascorbic acid). Total phenolic compound content was estimated by the Folin-Ciocalteu reagent and total carotenoid content by spectrophotometry at 450 nm. The contents of β-carotene and total vitamin C (ascorbic acid and dehydroascorbic acid) were quantified by high performance liquid chromatography. Differences were found among the four mango cultivars in all the components analyzed. The content of phenolic compounds ranged from 48.40 (Haden) to 208.70 mg/100 g (Ubá); total carotenoid from 1.91 (Haden) to 2.63 mg/100 g (Palmer); β-carotene from 661.27 (Palmer) to 2,220 μg/100 g (Ubá) and total ascorbic acid ranged from 9.79 (Tommy Atkins) to 77.71 mg/100 g (Ubá). These results corroborated previous information that mangoes are a good source of antioxidants in human diet.  相似文献

Avocado seed contains elevated levels of phenolic compounds and exhibits antioxidant properties. We investigated the effect of Avocado Seed Flour (ASF) on the lipid levels in mice on a hyperlipidemic diet. The concentration of phenols was determined by high-performance liquid chromatography, antioxidant activity was evaluated using the Trolox equivalent antioxidant capacity method, and dietary fiber was measured using the Association of Official Analytical Chemists (AOAC) method. The LD50 of ASF was determined using Lorke’s method and hypolipidemic activity was evaluated in a hypercholesterolemic model in mice. Protocatechuic acid was the main phenolic compound found in ASF, followed by kaempferide and vanillic acid. The total phenolic content in the methanolic extract of ASF was 292.00 ± 9.81 mg gallic acid equivalents/g seed dry weight and the antioxidant activity resulted in 173.3 μmol Trolox equivalents/g DW. In addition, a high content of dietary fiber was found (34.8%). The oral LD50 for ASF was 1767 mg/kg body weight, and treatment with ASF significantly reduced the levels of total cholesterol, LDL-C, and prediction of the atherogenic index. Therefore, the antioxidant activity of phenolic compounds and dietary fiber in ASF may be responsible for the hypocholesterolemic activity of ASF in a hyperlipidemic model of mice.  相似文献

Fourteen apricot genotypes grown under similar cultural practices in Trans-Himalayan Ladakh region were studied to find out the influence of genotype on antioxidant capacity and total phenolic content (TPC) of apricot kernel. The kernels were found to be rich in TPC ranging from 92.2 to 162.1 mg gallic acid equivalent/100 g. The free radical-scavenging activity in terms of inhibitory concentration (IC50) ranged from 43.8 to 123.4 mg/ml and ferric reducing antioxidant potential (FRAP) from 154.1 to 243.6 FeSO4.7H2O μg/ml. A variation of 1–1.7 fold in total phenolic content, 1–2.8 fold in IC50 by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 1–1.6 fold in ferric reducing antioxidant potential among the examined kernels underlines the important role played by genetic background for determining the phenolic content and antioxidant potential of apricot kernel. A positive significant correlation between TPC and FRAP (r = 0.671) was found. No significant correlation was found between TPC and IC50; FRAP and IC50; TPC and physical properties of kernel. Principal component analysis demonstrated that genotypic effect is more pronounced towards TPC and total antioxidant capacity (TAC) content in apricot kernel while the contribution of seed and kernel physical properties are not highly significant.  相似文献

In this study, we analyzed eight phenolic compounds (tyrosol, hydroxytyrosol, oleuropein, pinoresinol, and caffeic, ferulic, vanillic, and p-coumaric acid) in 55 mono- and multivarietal extra virgin olive oil samples by high performance liquid chromatography (HPLC) coupled to a coulometric electrochemical array detector (ECD). The phenolic profile of olive oil samples differed depending on the geographical origin and olive variety. The total reducing capacity (total phenolics) of olive oils ranged from about 40 to 530 mg gallic acid equivalents/kg oil. Tyrosol, hydroxytyrosol and pinoresinol were the most abundant phenolic compounds in olive oils. The antioxidant capacity of the olive oil extracts was determined by ferric reducing antioxidant power (FRAP) and Trolox equivalent antioxidant capacity (TEAC) assays. Total reducing capacity was significatly correlated with FRAP (R 2 ?=?0.91, p?<?0.001) and TEAC (R 2 ?=?0.92, p?<?0.001) values. Total reducing capacity, TEAC and FRAP values were significantly correlated with tyrosol, hydroxytyrosol as well as oleuropein concentrations. Hydroxytyrosol, comprising over 40 % of total olive oil phenolics, mainly contributed to the antioxidant activity of olive oils. The present study provides a comprehensive database of polyphenols in olive oils from 9 different countries and four continents.  相似文献

This study was designed to examine the in vitro antimicrobial and antioxidant activities and the amount of total phenolics of the methanol extracts of Ballota rotundifolia L. and Teucrium chamaedrys C. Koch. In the case of antimicrobial activity tests, polar sub-fractions of the methanol extracts of plant species exhibited weakest antimicrobial activity when compared with the non-polar ones. While, non-polar sub-fraction of B. rotundifolia showed moderate activity against A. lwoffii, C. perfringens and the yeasts, T. chamaedrys performed excellent activity pattern against all of the tested microorganisms. The sub-fractions were also screened for their possible antioxidant activities by two complementary tests, namely DPPH free radical-scavenging and β-carotene/linoleic acid assays. Non-polar extracts of the plant species remained inactive in both test systems. On the other hand polar extracts showed remarkable antioxidant activities. In DPPH system, free radical scavenging effect of T. chamaedrys was measured as 18.00 ± 1.42 μg.mg−1. It is extremely important to point out that, polar sub-fraction of T. chamaedrys is found as effective as the positive control BHT. Non-polar sub-fraction of T. chamaedrys found to have the highest total phenolic amount (97.12 ± 1.28 μg/mg). Results obtained from this experiment confirm the relationship between the amount of phenolics and biological activities.  相似文献

The phenolic compounds and flavonoids were determined from the extracts of Withania somnifera root (WSREt) and leaf (WSLEt). The WSREt has 28.26 mg/g total phenolic compounds and 17.32 mg/g flavonoids, whereas WSLEt has 5.4 mg/g total phenolic compounds and 5.1 mg/g flavonoids. The WSREt, WSLEt and glibenclamide were orally administered daily to diabetic rats for 8 weeks. After the treatment, the levels of urine sugar, blood glucose, liver glycogen, and antioxidants like vitamin C and E in plasma and superoxide dismutase (SOD), catalase (CAT), thiobarbituric acid reactive substances (TBARS), glutathione peroxidase (GPx), glutathione-S-transferase (GST) and reduced glutathione (GSH) in liver, kidney and heart were determined. Diabetic rats showed a significant (p < 0.05) elevation in glucose and TBARS and a significant (p < 0.05) reduction in glycogen, vitamin C and E, SOD, CAT, GPx, GST, and GSH levels when compared to normal control rats. Administration of WSREt, WSLEt and glibenclamide to diabetic rats restored the levels to normal. In the light of aforesaid facts, it is suggested that the presence of phenolic compounds including flavonoids in W. somnifera root and leaf extracts and their antioxidant activity may play a vital role in reduction of blood glucose level in alloxan-induced diabetic rats.  相似文献

The aqueous extracts of four marine algae, Caulerpa racemosa var. macrophysa, Gracilaria tenuistipitata var. tenuistipitata, Sargassum sp., and Ulva lactuca, from the coastal areas in Southern Thailand, were prepared by boiling dried seaweed powder in water for 3 h, and by autoclaving each sample at 120 °C for 3 h. They were then freeze-dried and evaluated for their antioxidant activities using DPPH (1, 1-diphenyl-2-picrylhydrazyl), hydroxyl radical (OH) and superoxide anion (O2•−) scavenging assays. Boiling extracts of the seaweeds, except C. racemosa, were found to have higher total phenolic contents (TPC) than those obtained from the autoclave method. The antioxidant results also showed that O2•− scavenging activity existed only in the boiling extracts of C. racemosa, G. tenuistipitata, and U. lactuca. In DPPH and OH assays, however, almost all the boiling extracts were less active than the autoclave ones. Among the four alga species, Sargassum sp. was the most active. Both extracts of this seaweed had the highest TPC and also displayed the strongest DPPH and OH inhibitory activities. A strong positive-correlation between the antioxidant potency and TPC of the autoclave extracts was found, while for the boiling extracts such relation was very weak. This result thus reflected that in addition to the phenolic compounds, there might be some other active components present in these extracts involved in the antioxidant activity.  相似文献

Total antioxidant activity of six non-cultivated but traditionally collected fruits from the south of Europe was assessed by measuring their ability to reduce the hydroxyl radical ( OH· ) \left( {{\hbox{O}}{{\hbox{H}}^\bullet }} \right) and hydrogen peroxide (H2O2), and their Trolox equivalent antioxidant capacity (TEAC). This antioxidant activity was compared with that shown by the synthetic antioxidants BHA (E-320), BHT (E-321) and propyl gallate (E-310). Total phenolics, ascorbic acid and the carotenoid content of the fruits were also analyzed. All fruits showed a high ability to scavenge the OH· {\hbox{O}}{{\hbox{H}}^\bullet } radical, ranging from 60.61% to 81.04% inhibition for Rosa canina and Crataegus monogyna, respectively. The H2O2 scavenging capacity and the TEAC value varied widely, ranging between 3.63% and 87.26% inhibition of H2O2 and between 0.47 and 416.64 mM trolox g−1 FW for Sorbus domestica and Rosa canina, respectively. The antioxidant activity of fruits was higher than that of the synthetic additives analyzed, except in the TEAC assay. The phenolic and carotenoid content of R. canina was much higher than that of the other fruits analyzed and its ascorbic acid concentration was also high, reflecting its higher efficacy towards ABTS ·- {\hbox{ABT}}{{\hbox{S}}^{ \bullet - }} (TEAC assay) and H2O2 species. In spite of these associations, the correlation coefficients between total antioxidant activity and the antioxidant compounds analyzed were not very significant; only phenolics and carotenoids showed a marginal correlation with the TEAC assay. The results support the possible use of R. canina as natural antioxidant to replace the synthetic additives, as well as their use in the production of functional foods with a high antioxidant activity.  相似文献

The object of the study was to determine the chemical composition of Diospyros lotus L. extract and their antioxidant and antiproliferative properties. Eight compounds were isolated from D. lotus and identified as gallic acid, methylgallate, ellagic acid, kaempferol, quercetin, myricetin, myricetin 3-O-β-glucuronide, and myricetin-3-O-α-rhamnoside. D. lotus extract tested in different in vitro systems (DPPH, ABTS, FRAP, and Fe2+ chelating activity assay) showed significant antioxidant activity. The potential antiproliferative properties of D. lotus extract and isolated compounds against nine human cancer cell lines such as COR-L23, CaCo-2, C32, ACHN, A375, A549, Huh-7D12, MCF-7, and LNCaP were investigated in vitro by SRB assay. D. lotus extract demonstrated the highest inhibitory activity against COR-L23 with an IC50 value of 12.2 μg/ml. Among identified hydrolysable tannins, ellagic acid evidenced strong antiproliferative activity against both C32 and A375 cells with IC50 values of 0.8 and 4.1 μg/ml, respectively. Interesting results were observed, also, with gallic acid that showed the highest cytotoxic activity against CaCo-2 (IC50 2.6 μg/ml). Overall, the results of this study suggest that D. lotus displays a good antioxidant activity and has antiproliferative effects. Both activities are related to identified phenolic compounds.  相似文献

Chestnut (Castanea sativa) shell and eucalyptus (Eucalyptus globulus) bark, waste products of the food and wood industries, respectively, were analysed as potential sources of antioxidant compounds. The extraction yield, the antioxidant activity and total phenols content of the extracts were greater in chestnut shell than in eucalyptus bark for most of the extraction conditions essayed. Extraction of chestnut shell with a 2.5% Na2SO3 aqueous solution led to the highest extraction yield, 25.6%, total phenols, 13.4 g gallic acid equivalent/100 g oven-dried shell, and FRAP antioxidant activity, 80.7 mmol ascorbic acid equivalent/100 g oven-dried shell. Extraction with methanol:water (50:50, v/v) provided the best results for eucalyptus bark. The antioxidant activity and the total phenols content of the extracts had a positive linear correlation. FTIR spectroscopy confirmed the higher content of phenolic compounds in chestnut shell extracts compared to eucalyptus bark extracts. Chestnut shell extracts were characterized by the presence of high molecular weight species whereas lower molecular weight species were predominant in eucalyptus bark extracts.  相似文献

The hydro-alcoholic extracts of five Equisetum species, E. arvense L., E. sylvaticum L., E. fluviatile L., E. palustre L. and E. telmateia Ehrh., growing-wild in Serbia were evaluated for their genotoxicity, antimicrobial activity, antioxidant capacity and the results related to the total phenol content and HPLC flavonoid profiles. The total phenol content was 92–349 μmol expressed as equivalents of chlorogenic acid per g of dried plant material. Main identified compounds were kaempferol-, quercetin- glycosides and caffeic acid derivatives. E. telmateia extract showed the greatest antioxidant capacity. Almost all tested microorganisms demonstrated some degree of sensitivity to the examined extracts. All tested extracts at 62.5 μg/ml showed higher incidence of micronucleus formation than in the control sample. The obtained data allowed mutual comparison of examined species and their assessment as possible sources of antioxidants, antimicrobials and/or genotoxic substances.  相似文献

Whole rice has been widely studied due to the abundance of bioactive compounds in its pericarp. Some of the beneficial effects of these compounds on human health have been attributed to their antioxidant and other biological activities, such as enzyme inhibition. In this work, we evaluated the contents of total, soluble and insoluble phenolic compounds of 6 red and 10 non-pigmented genotypes of whole rice as well as their inhibitory effect on the activity of angiotensin I-converting enzyme (ACE). The effects of cooking on phenolics and their inhibitory activities were also investigated. Red genotypes showed high content of phenolics, mainly soluble compounds, at an average of 409.7 mg ferulic acid eq./100 g, whereas overall lower average levels (99.4 mg ferulic acid eq./100 g) at an approximate soluble/insoluble compound ratio of 1:1 were observed in non-pigmented rice. Pigmented rice displayed a greater inhibitory effect on ACE than non-pigmented rice. In fact, a significant correlation between the content of soluble phenolics and ACE inhibition was observed (r = 0.8985, p < 0.05). In addition to significantly reducing the levels of total phenolics and ACE inhibition, cooking altered the soluble/insoluble compound ratio, especially among red rice genotypes.  相似文献

Studies on Indian Green Leafy Vegetables for Their Antioxidant Activity   总被引:1,自引:0,他引:1  
To identify the potential of green leafy vegetables (GLV) as antioxidants, methanolic extracts of Amaranthus sp., Centella asiatica, Murraya koenigii and Trigonella foenum graecum were studied for their antioxidant activity in different systems at multiple concentrations. Total antioxidant activity assessed by phosphomolybdenum method, free radical scavenging activity by 1,1-diphenly-2-picryl hydrazyl (DPPH), reducing power and ferrous ion chelating activity were determined. The GLV were analyzed for ascorbic acid, total and β-carotene and total polyphenol contents. The ascorbic acid, total carotene, β-carotene and total phenolic content (tannic acid equivalents) of the GLV ranged between 15.18–101.36, 34.78–64.51, 4.23–8.84 and 150.0–387.50 mg/100 g GLV, respectively. The extracts were found to have significantly different levels of antioxidant activities in the systems tested. The total antioxidant activity was highest in Murraya koenigii (2,691.78 μmol of ascorbic acid/g sample) and least in Centella asiatica (623.78 μmol of ascorbic acid/g sample). The extract concentration causing 50% inhibition of DPPH (IC50) was determined (M. koenigii < C.asiatica < Amaranthus sp. < T. graecum). The maximum DPPH scavenging activity and reducing power was exhibited by Murraya koenigii. Multiple regression analysis showed that the relationship of total antioxidant activity, free radical scavenging activity, and reducing power with polyphenol and total and β-carotene was highly significant. The antioxidant activity in 70% aqueous acetone extracts of the herb of Ambrosia artemisiifolia L., Asteraceae was evaluated with regards to total polyphenol and flavonoid contents. Antioxidant activity has been assessed by two commonly used in vitro tests, based on determination of ferric-reducing antioxidant power (FRAP assay) and DPPH free radical scavenging ability (DPPH assay), against butylated hydroxytoluene (BHT), ascorbic acid and quercetin, which were used as positive control substances. The results of both antioxidant tests showed that the plant material expressed a considerable activity (DPPH IC50 = 27.6 μg/ml; FRAP value = 2.37 mmol Fe2+/g), attributed to both flavonoid aglyca and resembling glycosides, as verified by dot-blot TLC analysis.  相似文献

Free essential oil methanolic extracts from three different geographical populations of Lippia graveolens in México were screened for antioxidant and antimutagenic properties by the DPPH and Kado microsuspension assay, respectively. Total phenolic and flavonoid contents as well as HPLC identification and quantification of naringenin and rosmarinic acid were also carried out. In addition, a taxonomical phenetic analysis was performed. The L. graveolens extracts showed varying content of phenols and flavonoids. Significant concentration of rosmarinic acid was found for the first time in the species. All the extracts were capable of scavenging DPPH radicals in a concentration dependent fashion; the IC50 values correlate with the phenolic content. None of the extracts was toxic to TA100 and TA98 strains at the concentrations tested; moreover, the extracts at a concentration equivalent to 200 μg of gallic acid inhibited a 39 and 30% the mutagenicity induced by 4-nitro-o-phenylenediamine and sodium azide, respectively. The results suggest that the Mexican oregano is a source of polar bioactive ingredients for the food industry.  相似文献
