Fourteen dogs with known clinical hypersensitivity to soy and corn were maintained on a limited antigen duck and rice diet until cutaneous manifestations of pruritus were minimal (78 days). Sequential oral challenges with cornstarch, corn and soy were then performed. Subsequently, the dogs were fed a diet containing hydrolysed soy protein and cornstarch. Throughout the study period the dogs were examined for cutaneous manifestations of pruritus and, additionally, serum was collected for measurement of allergen-specific and total immunoglobulin (Ig)E concentrations. Intradermal testing with food antigens was performed prior to entry into the study and after 83 days. A statistically significant clinical improvement was measured between days 0 and 83. Significant pruritus was induced after oral challenge with cornstarch, corn and soy (P = 0.04, 0.002, 0.01, respectively) but not with the hydrolysed diet (P = 0.5). The positive predictive value of the skin test for soy and corn allergy was reduced after feeding a soy and corn free diet. Although increases in soy and corn-specific serum IgE concentrations were measured in individual dogs post challenge they were not statistically significant and could not be used to predict clinical hypersensitivity.  相似文献

The owners of 63 pruritic dogs were instructed to feed them a chicken hydrolysate diet exclusively for six weeks as part of diagnostic investigations into non-seasonal pruritus. Ectoparasitism and microbial infections were eliminated during the dietary trial. The dogs' skin lesions, gastrointestinal signs and frequency of defecation were assessed and scores for pruritus were assigned before they started the diet and before and after they resumed their original diet. An adverse food reaction was diagnosed if the pruritus resolved while they were on the diet, but recurred when they resumed their original food regimen. Seventeen of the 63 dogs were withdrawn from the trial, including four which found the diet unpalatable; however, its palatability was reported to be good or excellent in 48 of the dogs. An adverse food reaction alone was diagnosed in nine (19.6 per cent) of the 46 dogs and another nine had an adverse food reaction and atopy. There were gastrointestinal signs in six of the nine dogs with an adverse food reaction, which resolved on the trial diet but recurred when they resumed their original diet.  相似文献

Canine food allergy can be defined as a non-seasonal, pruritic skin disorder of dogs that is associated with the ingestion of a substance found in the dog's diet. This study records the use of a proprietary dried fish, corn and soyabased diet for the investigation and maintenance of food allergic dogs when fed initially as a restricted allergen diet and then as a maintenance diet after challenge. All the dogs showed evidence of pruritic skin disease and in addition demonstrated gastrointestinal signs. These included the presence of faecal mucus and blood, tenesmus and increased faecal frequency; all the signs associated with colitis. Both cutaneous and gastrointestinal signs resolved when an elimination diet was fed and could be reproduced when the animal was appropriately challenged. Ten dogs were trailed on a home cooked diet of fish and potato and 10 dogs on the proprietary complete food. All the dogs were challenged to identify their food allergies. Nineteen of the dogs have subsequently been successfully maintained on the proprietary food.  相似文献

OBJECTIVE: To evaluate 3 commercially available selected-protein-source diets as maintenance diets in dogs with pruritus caused by adverse food reactions. DESIGN: Randomized crossover trial. ANIMALS: 40 dogs > 6 months of age with pruritus caused by adverse reactions to foods. PROCEDURE: Diagnosis was confirmed by use of diet elimination and provocation studies. Subsequently, dogs were fed 3 commercial diets for 3 weeks each in a randomized, blinded, crossover trial. Dogs were evaluated for pruritus, vomiting, diarrhea, and flatulence. RESULTS: Pruritus recurred in 52.5% of dogs fed a chicken-rice diet, 47.5% of dogs fed a catfish-rice diet, and 85% of dogs fed a venison-rice diet. Overall 95% of the dogs could be managed successfully with at least 1 of the 3 diets. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that commercially available limited-allergen diets with selected protein sources may be appropriate for long-term management of pruritus caused by adverse food reactions. Testing of various protein sources is usually required.  相似文献

Food hypersensitivity was diagnosed in 30 dogs. Clinical signs varied, with pruritus (97%), erythema (50%), and papules (37%) being the most common. Diagnosis was based on resolution of clinical signs when the dogs were fed a restricted ("hypoallergenic") diet, and recurrence of signs when fed their original diet or other foods. No age, breed, or sex predilection was noticed. None of the owners could relate the onset of clinical signs with a recent change in diet. Seven dogs had concurrent fleabite, inhalant, intestinal parasite, or insulin hypersensitivity.  相似文献

One hundred suspected allergic dogs were skin-tested with 9 food extracts, in addition to other inhalant and insect allergens. Forty-eight of the dogs had +2 or greater intradermal reactions to 1 or more food extracts at concentration of 1,000 protein nitrogen units/ml. Thirty of the food extract-positive dogs were fed a restrictive hypoallergenic diet. Of these 30 dogs, 3 improved, then allergy signs worsened when they were challenge-exposed with the original diet. Fifty-two dogs did not have skin test reactivity to any of the 9 food extracts. Of these 52 dogs, 35 were fed a restrictive diet. Six improved, then allergy signs worsened when dogs were challenge-exposed with the original diet. Skin-testing with selected allergenic food extracts was not useful in identifying food-sensitive dogs.  相似文献

A dietary intervention study was assessed to determine if different sources of starch in homemade diets could significantly modify fecal microbiome of dogs. Twenty-seven adult dogs were enrolled and fed a diet based on a mixture of rice and pasta with fresh raw meat (CD). After 90 d, 8 dogs continued to receive CD diet, 10 dogs received a diet made of a raw meat and a complementary food with rice as the main source of starch (B1), and 9 dogs were fed a diet with the same raw meat and a complementary food with potato as the main source of starch (B2). Samples of feces were collected from each dog in the mornings at the beginning of the study and after 15 d and analyzed for pH, ammonia N (N–NH3) and total N, short chain fatty acids (SCFA) and lactic acid. Relative abundance of fecal microbiota was assessed by sequencing and annotating the V3–V4 regions of the 16S rRNA. Total starch intake was similar between diets but differed in the in vitro rate digestion and in the resistant starch, which was higher in B2 than in B1 and CD diets. Dogs fed B2 diet showed lower (P < 0.05) N–NH3 and pH but higher (P < 0.05) molar proportion of lactic acid. Linear discriminant analysis of the genera relative abundances indicated a significant (P < 0.01) increase of SMB53 genus at the end of the study in B1 diet and of Megamonas genus in B1 and B2 diets in comparison to CD diet. These results suggest that changes of starch source in a raw meat-based diet have limited effects on fecal microbiome in healthy dogs, but underline a high variability of microbiota among dogs.  相似文献

The purpose of this retrospective study was to compare home-prepared and chicken hydrolysate diets in the diagnosis of canine adverse food reactions (AFR). Seventy-two dogs were fed home-prepared diets and 109 were fed hydrolysate. Owners chose the type of diet at presentation, and ingredients of home-prepared diets were selected depending on each dog's dietary history. Ectoparasitic infestations and microbial infections were treated during the trials. Cutaneous and gastrointestinal signs and pruritus scores were recorded before starting the diet, 6 weeks into the trials and after provocation with the original diets. AFR was diagnosed if pruritus resolved during the trial and recurred on dietary provocation. The dropout rate was lower for home-prepared diets although not statistically significant (18.1% home prepared; 24.7% hydrolysate, P=0.377). AFR alone was diagnosed in 10 dogs (17%) using home-prepared diets and in 15 (18.3%) fed the hydrolysate. Gastrointestinal problems were more frequent in dogs with AFR than in dogs without AFR (P=0.001). Another 11 dogs (18.6%) in the home-prepared diet group and 20 (24.4%) in the hydrolysate diet group had AFR concurrent with other pruritic diseases, mainly atopy. The similar frequencies of AFR diagnosis in the two groups (P=0.837 AFR; P=0.416 concurrent AFR) indicate that the chicken hydrolysate diet may be a valuable alternative to home-prepared diets in the diagnosis of canine AFR. Prospective cross-over studies are warranted to confirm these findings.  相似文献

Recent studies have shown that immunological aberrations and epidermal barrier defects could be important in the pathogenesis of canine atopic dermatitis (CAD) and that oral polyunsaturated fatty acids (PUFAs) might influence the epidermal barrier. The aim of this study was to evaluate the effects of a spot-on formulation containing PUFAs and essential oils on pruritus and lesions caused by CAD. Forty-eight privately owned dogs of different breeds, ages and genders diagnosed with atopic dermatitis were included in a randomized, double-blinded, placebo-controlled, multicentre clinical trial. Dogs were treated with a spot-on formulation containing PUFAs and essential oils or placebo on the dorsal neck once weekly for 8 weeks. Before and after the study, CAD extent and severity index-03 (CADESI-03) and pruritus scores were determined by veterinarians and owners, respectively.There was significantly more improvement in CADESI-03 and pruritus scores in the treatment group than in the placebo group (P = 0.011 and P = 0.036, respectively). Additionally, more dogs improved by at least 50% in CADESI-03 and pruritus scores in the treatment group than in the placebo group (P = 0.008 and P = 0.070, respectively). No adverse reactions were observed. The topical preparation containing PUFAs and essential oils was a safe treatment and beneficial in ameliorating the clinical signs of CAD.  相似文献

A commercial, lamb and rice, dog food with an omega-6:omega-3 fatty acid ratio of 5.5:1 was fed in a single-blinded, self-controlled clinical trial to 18 atopic dogs. The pruritus in 8 of these dogs (44.4%) was satisfactorily controlled within 7 to 21 d, returned within 3 to 14 d after the diet was withdrawn, and was again controlled when the diet was reinstated. Plasma and skin levels of examined fatty acids changed in all 18 dogs when their diet was switched to the test diet. Dogs responding to the test diet had a different pattern of fatty acid change as compared to the dogs which failed to respond to the diet, suggesting that there are subsets of atopic dogs with different fatty acid metabolism capabilities.  相似文献

Stomach tissues from 222 beagle dogs, which were used for toxicological studies, were examined histopathologically. Out of 190 dogs fed the same diet, 185 had gastric lesions, whereas all of 32 dogs fed another diet were free from them. The gastric lesions were noted as an accumulation of eosinophils, infiltration of inflammatory cells including lymphocytes, plasma cells and histiocytes in the gastric lamina propria, formation of foreign body type multinucleated giant cells, focal desquamation and papillary proliferation of mucosal epithelium. The lesions were confined to the superficial layer of the antral and pyloric mucosa, and neither desmoplasia nor clinical signs were seen. Multinucleated giant cells often contained amphophilic foreign bodies of varying appearance with inconsistent reaction for periodic acid-Schiff (PAS), but no definite causal agents were detected.  相似文献

OBJECTIVE: To evaluate efficacy of cyclosporine A, administered at either of 2 dosages, in dogs with atopic dermatitis (AD). DESIGN: Multicenter randomized controlled trial. ANIMALS: 91 dogs with AD. PROCEDURE: Dogs were assigned to receive placebo (30 dogs), cyclosporine at a low dosage (2.5 mg/kg [1.1 mg/lb], PO, q 24 h for 6 weeks; 30 dogs), or cyclosporine at a high dosage (5.0 mg/kg [2.3 mg/lb], PO, q 24 h for 6 weeks; 31 dogs). RESULTS: After 6 weeks, mean percentage reductions, compared with baseline scores, in scores of lesion severity were 34, 41, and 67% for dogs treated with the placebo, cyclosporine at the low dosage, and cyclosporine at the high dosage, respectively. Similarly, mean percentage reductions in pruritus scores were 15, 31, and 45%, respectively. Percentage reductions in skin lesion and pruritus scores were significantly higher for dogs given cyclosporine at the high dosage than for dogs given the placebo. Treatment efficacy was significantly associated with whether dogs had a history of seasonal AD. Percentage reductions in skin lesion and pruritus scores were high for dogs treated with cyclosporine at the highest dosage that had a history of nonseasonal AD. Dogs in all groups with seasonal AD improved during the study period. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that oral administration of cyclosporine at a dosage of 5.0 mg/kg once daily is effective in reducing severity of pruritus and skin lesions in dogs with AD, especially those with nonseasonal disease.  相似文献

Shampoo therapy is frequently used on pruritic dogs. However, there are few double-blinded, placebo-controlled studies of this form of therapy. This randomised, double-blinded, placebo-controlled study evaluated the efficacy of a commercial medicated shampoo (DermaTopic; Almapharm) containing chlorhexidine, lactoferrin, piroctone olamine, chitosan and essential fatty acids in 27 dogs with mild to moderate allergic pruritus without secondary skin infections. All dogs received shampoo therapy with either DermaTopic or a shampoo vehicle as placebo twice weekly for four weeks. The extent of pruritus was evaluated before the study and then on a daily basis by the owners using a visual analogue scale. Before beginning the treatment and after four weeks, the skin lesions were evaluated by an experienced clinician with a validated lesion score (Canine Atopic Dermatitis Extent and Severity Index - CADESI). The pruritus was reduced significantly by both DermaTopic and placebo. However, there was no significant difference between both groups. There was no statistically significant difference in the CADESI scores pre- and post-treatment in either group or between the two types of treatment. This study provides further evidence of the benefit of shampoo therapy for pruritic dogs.  相似文献

The effects of six extruded diets with different starch sources (cassava flour, brewer's rice, corn, sorghum, peas or lentils) on dog total tract apparent digestibility and glycemic and insulinemic response were investigated. The experiment was carried out on thirty-six dogs with six dogs per diet in a completely randomized design. The diets containing brewer's rice and cassava flour presented the greatest digestibility of dry matter, organic matter and gross energy (p < 0.05), followed by corn and sorghum; pea and lentil diets had the lowest. Starch digestibility was greater than 98% in all diets and was greater for brewer's rice and cassava flour than for lentils and peas diets (p < 0.05). Dogs' immediate post-prandial glucose and insulin responses (AUC < or = 30 min) were greater for brewer's rice, corn, and cassava flour diets (p < 0.05), and later meal responses (AUC > or = 30 min) were greater for sorghum, lentil and pea diets (p < 0.05). Variations in diet digestibility and post-prandial response can be explained by differences in chemical composition of each starch source including fibre content and starch granule structure. The nutritional particularities of each starch ingredient can be explored through diet formulations designed to modulate glycemic response. However, more studies are required to support these.  相似文献

Twenty-seven pruritic dogs were used in this study. When a hypoallergenic diet was fed to these 27 dogs for six weeks, none of the dogs showed improvement of the pruritus. These dogs had a history and clinical signs of atopic dermatitis (AD) as defined by Prelaud's diagnostic criteria. Subsequently, the 27 dogs were isolated for observation for two weeks in the hospital. In the isolation room in the veterinary clinic, cages and tableware were all stainless steel, and carpet was not used. A hypoallergenic diet was continuously fed to the 27 dogs for two weeks, during which time they were kept in the isolation room. PVAS (Pruritus Visual Analog Scale) was performed prior to starting the isolation, at the start of the study and 2 weeks after starting the isolation. In 17 dogs (63%) the pruritus improved in the isolation room. A statistically significant reduction (p < 0.01) of PLS (Pruritus liners score) was recorded 2 weeks after isolation. It was hypothesized that the 17 dogs whose pruritus improved in the isolation room had AD caused by an environmental antigen that was not present in the isolation room. Pruritus of the remaining 10 dogs (37%) did not improve. For 6/10 dogs, the intradermal allergy testing was positive for an environmental antigen. For 4/10 dogs, the intradermal allergy testing was negative for all environmental antigens. Dogs for which sensitivity to an environmental antigen was not identified were thought to have atopic-like dermatitis.  相似文献

Corn is a commonly used ingredient in dry pet foods because there is a stable supply and it is a relatively inexpensive source of nutrients. Corn hybrids are available that are higher in CP and amylose and lower in phytate concentration than conventional hybrids. Approximately 500 mg of high-protein (HP), high-protein, low-phytate (HPLP), and high-amylose (HA) corn were compared with conventional (CONV) corn and amylomaize starch (AM) in triplicate and exposed to pepsin/hydrochloric acid and pancreatin to simulate hydrolytic digestion. Substrate remaining after this was used to determine in vitro colonic fermentation. Organic matter disappearances as a result of hydrolytic digestion were >80% for CONV, HP, and HPLP, whereas HA (60.7%) and AM (43.7%) were lower (P < 0.05). Total digestion (TD) values after hydrolytic digestion and 8 h of fermentation using canine fecal inoculum were greater (P < 0.05) for CONV, HP, and HPLP vs. HA and AM. The residue left after hydrolytic digestion of all substrates was poorly fermented. Five ileal-cannulated dogs were fed each corn hybrid at approximately 31% of the diet in a 5 x5 Latin square design. Dogs fed diets containing HP corn had higher (P < 0.05) ileal OM digestibility (70.3%) and tended (P < 0.10) to have higher DM digestibility (64.6%). Ileal starch digestibilities were lower (P < 0.05) for dogs fed HA (64.0%) and AM (63.0%). Ileal digestibilities of essential (71.2%), nonessential (67.4%), and total (69.0%) AA tended to be higher (P < 0.10) for HP diets compared with CONV (66.4, 62.4, and 64.0%, respectively). Total-tract DM, OM, CP, and GE digestibilities (77, 82, 77, and 84%, on average, respectively) were higher (P < 0.05) for dogs fed CONV, HP, and HPLP than for those fed AM (66.9, 71.6, 72.6, and 76.5%) and HA (60.6, 65.7, 69.7, and 71.5%). Total-tract fat digestibilities were lower (P < 0.05) for dogs fed HA diets (86.6%) than for all other treatments (91.0%, on average). Total-tract starch digestibilities were higher (P < 0.05) for dogs fed CONV, HP, and HPLP (98%, on average) compared with HA (72.8%) and AM (76.5%). No differences were detected among treatments in fecal bifidobacteria, lactobacilli, or Clostridium perfringens concentrations. The experiments demonstrated that HP and HPLP corn had hydrolytic digestion and fermentation characteristics similar to those of CONV corn, whereas HA resulted in similar responses to AM, a well-established resistant starch ingredient.  相似文献

Dietary n-3 polyunsaturated fatty acids are widely used for amelioration of inflammatory skin disease in dogs. In this study, a diet containing two different sources of n-3 polyunsaturated fatty acid–triglyceride (from menhaden oil) and concentrated ethyl esters–was fed to one group of six purpose-bred dogs, while an isocaloric isonitrogenous diet with corn oil (n-6 polyunsaturated fatty acids) was fed to another group of eight purpose-bred dogs for six weeks. Peripheral blood neutrophils, isolated at week–1 (baseline), week 2 and week 6, were stimulated with calcium ionophore A23187 and the amount of leukotriene B4 produced was determined via reversed-phase high performance liquid chromatography. Analysis of variance of log-transformed data revealed a significant effect for diet ( P  = 0.005) at six weeks, with dogs fed the high n-3 polyunsaturated fatty acid diet having significantly less mean ex vivo neutrophil leukotriene B4 production than dogs fed the high n-6 polyunsaturated fatty acid diet. Further studies on the clinical usefulness of n-3 polyunsaturated fatty acid ethyl esters are warranted.  相似文献

The aim of this study was to investigate the effect of the addition of a purified soluble (pectin) and insoluble (lignocellulose) fibre to a starchy meal on post‐prandial glucose and insulin responses in healthy horses. Four horses were fed in a randomized order three different diets: (i) cracked corn, (ii) cracked corn mixed with purified lignocellulose, and (iii) cracked corn mixed with purified pectin. Each diet was adjusted to a starch intake of 2 g/kg bodyweight (BW). Lignocellulose was aligned to an intake of 0.2 g/kg BW, and pectin was fed in a dosage of 0.1 g/kg BW. Each period consisted of a 10‐day acclimatization to the diet (fed once per day); during this time, the horses were fed 1.2 kg hay/100 kg BW/day. Blood was collected after each acclimatization period before and after the test meal was fed, without any hay. The increase in plasma glucose and insulin, peak values, and area under the curves were similar for all diets. The present findings suggest that adding purified soluble or insoluble fibre to a corn meal does not affect post‐prandial glucose and insulin responses in healthy horses. Feeding strategies for horses with a high energy requirement should include a starch reduction per meal, rather than the addition of purified fibre.  相似文献

Several hydrolysate-based diets have been commercialized for helping diagnose or treat dogs with cutaneous adverse food reactions (CAFR). This systematic review was performed to examine the evidence in favour of reduced immunological and clinical allergenicity of hydrolysates in dogs with CAFR. Citation databases, meeting abstracts and article bibliographies were scanned for relevant citations, and companies were contacted to provide unpublished reports. Eleven studies relevant to this study were identified. Some evidence of reduced serum IgE binding to a soy hydrolysate (1 study) and decreased intradermal test reactivity to hydrolysed proteins (three studies) was found. In four reports, the feeding of dogs suspected of having CAFR with hydrolysate-based diets reduced or eliminated clinical signs in a variable proportion of subjects. The percentage of dogs with CAFR that still reacted to these hydrolysate-based diets could not be assessed, however. Importantly, up to 50% of dogs with CAFR enrolled in three controlled studies exhibited increases in clinical signs after ingesting partial hydrolysates derived from foods to which they were hypersensitive. In conclusion, the limited number of studies undertaken point to reduced – but not eliminated – immunological and clinical allergenicity of hydrolysate-based commercial diets. A variable proportion of dogs with CAFR will exhibit a worsening of clinical signs when fed partial hydrolysates. Clinicians must weigh the clinical benefit of these diets versus their high cost and low risk of reduced appetence or gastrointestinal sign development. At this time, hydrolysate-containing diets are probably best used in dogs suspected not to be hypersensitive to their individual components.  相似文献
