A disease named locally as churrío or churrido equino (i.e., equine scours) has occurred for at least 100 years in Uruguay and southern Brazil in farms along both shores of the Merín lake. This report describes cases of churrido equino and provides serologic, pathologic, and DNA-based evidence indicating that the disease is in fact equine monocytic ehrlichiosis (Potomac horse fever). Results of an epidemiological investigation conducted on an endemic farm are also presented. Clinical signs in 12 horses were fever, depression, diarrhea, dehydration, and sometimes colic and distal hind limb edema. Postmortem findings of 3 horses were of acute enterocolitis. Inclusion bodies containing ehrlichial organisms were found in the cytoplasm of macrophages of the large colon of 1 horse. Eleven of the 12 horses were serologically positive to Ehrlichia risticii (indirect fluorescent antibody assay) and, of 3 paired samples, 2 showed seroconversion. Ehrlichia risticii DNA was identified by a nested polymerase chain reaction in peripheral blood of an affected horse. A healthy horse inoculated with peripheral blood from an affected horse developed the disease and antibodies to E. risticii. The disease had a peak incidence in March (summer) and was statistically associated with a marshy ecosystem near the Merín lake, where large numbers of Pomacea spp. (Ampullariidae) snails were found. Incidence density was almost 8 times higher in nonnative horses than in native horses. It was concluded that the previous diarrheic disease of horses known in Uruguay and southern Brazil as churrido equino is equine monocytic ehrlichiosis.  相似文献

Leishmania infantum infection has recently been described in horses in Europe. We report the results of a study on the immune response to L. infantum in horses living in an area endemic for leishmaniosis in NE Spain. Two ELISAs using protein A and anti-horse IgG conjugates were adapted to measure specific antibodies to L. infantum in horse sera. A lymphocyte proliferation assay (LPA) of peripheral blood mononuclear cells to L. infantum antigen was also performed to detect specific cellular immune response to Leishmania. Anti-L. infantum antibodies were detected in the serum of 16 of the horses studied (n=112) using the protein A assay but not in the assay using the anti-horse IgG conjugate. Specific lymphocyte proliferation was observed in 20 out of 55 horses. This study shows that horses in the area studied mount specific immune responses to L. infantum, and must therefore be considered among the species exposed to the parasite in this region. The infrequency of leishmaniosis in horses suggests that the immune response in this species is effective in controlling the infection.  相似文献

The immune response in horses following experimental infection with equine herpesvirus type 1 (EHV-1) was assessed by measuring cytotoxicity for EHV-1-infected target cells. A technique was developed, using [125I]5-iodo-2'-deoxyuridine ([125I]IUDR)-labeled equine fetal kidney cells infected with EHV-1 as the target cells. It was shown that peripheral blood leukocytes from a recovered horse were capable of lysing target cells, as measured by the loss of radio-active label. Following the experimental infection of specific-pathogen-free ponies with EHV-1, cytotoxicity was obtained with fresh autologous serum, peripheral blood leukocytes in autologous serum, and washed peripheral blood leukocytes. Cytotoxicity of the serum and peripheral blood leukocytes was detected as early as one day after infection. It is suggested that cytotoxic antibodies or cells could play an important part in restricting virus spread after infection of the horse with EHV-1.  相似文献

Our investigation has shown that multiple vaccinations with inactivated African horse sickness (AHS) vaccines containing all 9 serotypes and produced at the Central Veterinary Research Laboratory in Dubai, UAE, protect horses from AHS. However, the immunization did not prevent African horse sickness fever (AHSF) in approximately 10% of the vaccinated horses despite high enzyme-linked immunosorbent assay and virus neutralizing antibodies. African horse sickness fever is a very mild form of AHS with similar clinical signs. From all 6 horses which had developed AHSF, no virus was isolated from EDTA blood withdrawn during the acute phase of infection. Despite high neutralizing antibodies, serotype 9 was detected by polymerase chain reaction in 4 of them. All 6 horses recovered within 72 hours, after they developed mild clinical signs of AHS.  相似文献

Lymphocyte stimulation was observed in whole equine blood in the presence of phytohaemagglutinin and M protein extracted from a typical strain of Streptococcus equi. Blood samples were collected from several healthy horses and horse and pony foals and cultured in vitro with varying concentrations of phytohaemagglutinin and M protein for several days. Phytohaemagglutinin was found to induce lymphocyte stimulation in these animals. Highest mean stimulation indices in horse foals (49.3 +/- 24.4) and pony foals (54.7 +/- 32.0) were observed with 0.625 and 1.25 micrograms/mL phytohaemagglutinin, respectively, at either 72 or 96 hours of incubation. Significantly higher radioactive counts per minute in horse and pony foals were recorded in blood cultures incubated with 0.625 and 1.25 micrograms/mL phytohaemagglutinin. M protein induced a dose related stimulation response in adult horses. Maximum stimulation indices were observed against 125 micrograms/mL M protein at 96 hours. These stimulation indices were higher in adult horses (40.0 +/- 2.2) than observed in pony foals (14.4 +/- 15.7). Higher stimulation levels in adult horses indicated either nonspecific stimulation against M protein or previous exposure of these animals to S. equi.  相似文献

Bovine papillomavirus (BPV), the causative agent of papillomas in cattle, has been shown to play a major role in the pathogenesis of equine sarcoids in horses. BPV has also been detected occasionally in normal equine skin. In this study, presence and activity of BPV in normal skin and peripheral blood of 4 groups of horses were evaluated: sarcoid-affected horses, horses living in contact with sarcoid-affected horses, horses living in contact with papilloma-affected cattle and control horses. From each horse, 3 samples on 4 locations were collected: a swab of the intact skin surface and both a swab and a biopsy after decontamination. BPV DNA was found in the normal skin of 24 of 42 horses (57%). Mainly sarcoid-affected horses and horses living in contact with cattle were carriers (73%), but BPV DNA was also detected in 50% of the horses living in contact with sarcoid-affected horses and in 30% of the control population. BPV mRNA was detected in 58% of the samples positive for BPV DNA, although in a much lower quantity compared to sarcoids. In most of the BPV DNA positive samples mild acanthosis, slight basophilic cytoplasmic swelling of the epidermal layers and/or thickening of the basal membrane were noticed, but these observations were also present in several BPV DNA negative normal skin samples. BPV DNA could not be detected in peripheral blood. These findings suggest latent infection and a wide-spread occurrence of BPV in the horse population.  相似文献

OBJECTIVE: To determine whether iatrogenic hemarthrosis of the metacarpophalangeal joint could be used as a model for temporary reversible joint pain in horses. ANIMALS: 8 adult horses. PROCEDURE: Each horse was evaluated on a treadmill before and after injection of 1 metacarpophalangeal joint with 10 mL of autogenous blood. Horses were evaluated subjectively and objectively by use of a computerized force measurement system at intervals until lameness abated. The mean force difference between injected and noninjected limbs at all time periods after injection was compared with the difference between limbs at baseline. From each horse, synovial fluid samples collected before and 24 hours and 30 days after injection were analyzed for total protein concentration and cell type and number. Venous blood samples were collected before and 6 and 24 hours after injection for assessment of plasma cortisol concentration. RESULTS: For 24 hours after injection, the mean force difference between injected and noninjected limbs was significantly increased over baseline. The greatest force difference was detected after 2 and 4 hours. Baseline and 24-hour force data were not significantly different. Compared with baseline values, synovial fluid protein concentration and nucleated cell and RBC counts were increased significantly at 24 hours after injection but were not different at 30 days after injection. No significant changes in plasma cortisol concentration were detected at any time point. CONCLUSIONS AND CLINICAL RELEVANCE: In horses, iatrogenic hemarthrosis of the metacarpophalangeal joint appears to induce temporary reversible lameness with a mild to moderate degree of synovitis.  相似文献

In order to understand the blood physiological and biochemical reactions after long-distance transport, 23 Akhal-teke horses (male horse 14, mare 9, aged 2 to 6 years old), were selected and transported from Russian to Alashankou of Xinjiang (5 d road transport). 7 and 35 d after reaching the destination, the jugular vein blood were collected for the determination of physiological and biochemical indicators and stress-related hormone levels. The remote control horses without transport (15 local Akhal-teke horses,male horse 10, mare 5, aged 6 to 10 years old) were used for comparative study. The results showed that on the 7th and 35th day after transport, the number and proportion of granulocytes (Gran), hematocrit (HCT), blood platelet (PLT) and alkaline phosphatase (ALKP) were significantly or extremely significantly higher than those of the local Akhal-teke horses (P<0.05; P<0.01), while the number and percentage of intermediate cells (Mid) and the level of blood glucose (GLU) were significantly or extremely significantly decreased (P<0.05; P<0.01); On the 35th day after transport, the mean red blood cell volume (MCV), phosphorus (PHOS) and total protein (TP) were significantly or extremely signifcantly increased (P<0.05; P<0.01), and the percentage of lymphocytes (LY) was significantly decreased (P<0.05); On the 7th day after transport, the levels of antidiuretic hormone (ADH), growth hormone (GH), UREA and insulin (INS) were significantly or extremely signifcantly decreased (P<0.05; P<0.01), and the total bilirubin (TBIL) and triiodothyronine (T3) were signigicantly increased (P<0.05), but there was no significant difference between the two groups (P>0.05) on the 35th day after transport. Based on the above results, after the long-distance transport, there had a corresponding transient stress response in Akhal-teke horses, which required some adjustment after the recovery period (35 d) to let most of the indicators were consistent with those without long-distance transport. So the horses could not be used until they were in a normal state, in order to avoid long-term irreversible lesions.  相似文献

The objective of the present study was to compare changes of blood parameters induced by the same work and performed in the same conditions in two racehorse breeds, Thoroughbred and Purebred Arabian. The effect of moderate-intensity exercise was studied in 20 stallions--ten Thoroughbreds, aged 2-3 years and the same number of Purebred Arabians, 3-4 years old. All the horses were administrated the same effort test consisting in 1200 m gallop at a speed typical of the daily training sessions. Three jugular venous blood samples were collected for each horse: at rest, just after the end of the gallop and after 30-minute rest. In the gathered blood, a hemoglobin (Hb) concentration was determined as well as plasma level of glucose (Glc), triacylglycerols (TG), glycerol, free fatty acids (FFA), total plasma proteins (TP) and the activity of creatine kinase (CK), lactate dehydrogenase (LDH) and aspartate aminotransferase (AST). In the Arabian horses, an increase in levels of TP, glycerol, FFA and CK activity measured just after exercise was higher than that in Thoroughbreds. Similarly, after a 30-minute rest, a post-exercise rise of TP, AST, glycerol and FFA proved to be higher in the Arabian horses compared to that in the Thoroughbreds. Only TG plasma concentration measured 30 minutes following the effort was significantly lower in the Arabian horses than in Thoroughbreds. It can be concluded that the Thoroughbred horses adapted better to the effort test applied in this study as compared to the Purebred Arabian horses. The parameters related to lipid metabolism proved to be the most sensitive indicators of breed differences in relation to moderate-intensity exercise.  相似文献

OBJECTIVE: To quantify changes in endothelium-derived factors and relate those changes to various aspects of digital hemodynamics during the prodromal stages of carbohydrate overload (CHO)-induced laminitis in horses. ANIMALS: 20 adult horses without abnormalities of the digit. PROCEDURES: Digital and jugular venous blood samples were collected at 1-hour intervals (for assessment of endothelin-1 [ET-1] immunoreactivity and measurement of glucose, insulin, and nitric oxide [NO] concentrations) or 4-hour intervals (CBC and platelet-neutrophil aggregate assessment) for 8 hours or 16 hours after induction of CHO-associated laminitis in horses treated with an ET-1 antagonist. Effects of treatment, collection site, and time and the random effects of horse on each variable were analyzed by use of a repeated-measures model. Where treatment and collection site had no significant effect, data were combined. RESULTS: Compared with baseline values, CHO resulted in changes in several variables, including a significant increase from baseline in digital blood ET-like immunoreactivity at 11 hours; digital blood ET-like immunoreactivity was significantly greater than that in jugular venous blood at 8, 9, 11, and 12 hours. Digital and jugular venous blood concentrations of glucose increased from baseline significantly at 3, 4, and 5 hours; insulin concentration increased significantly at 5 hours; and the number of platelet-neutrophil aggregates increased significantly at 12 hours. CONCLUSIONS AND CLINICAL RELEVANCE: In horses, concurrent increases in venous blood ET-1 immunoreactivity, insulin and glucose concentrations, and platelet-neutrophil aggregates support a role of endothelial dysfunction in the pathogenesis of CHO-induced laminitis.  相似文献

The current study was conducted to evaluate daily within-horse variation of plasma alpha-tocopherol concentration (TOC) (Experiment 1 [EXPT1]) and to determine the effect of a single meal on plasma TOC (Experiment 2 [EXPT2]). In EXPT1, venous jugular blood samples were collected from four pregnant (6–7 months of gestation), light horse mares (8.8 ± 2.9 years of age, mean ± SD; range, 5–11 years of age) between 7:30 AM and 8:30 AM, followed by eight hourly samples, and analyzed for plasma TOC. All horses were fed 5 kg dry matter (DM) grass hay 18 hours before sample collection and received no additional feed during the sampling period. Mean within-horse coefficient of variation for TOC was 3.5% ± 0.01%. In EXPT2, seven mature light horses, four mares and three geldings (4.9 ± 4.4 years of age, mean ± SD; range, 2–14 years of age), were randomly assigned to one of two treatments (fasted [FST; n = 7] or fed [FD; n = 7]), followed by reversal of treatments 24 hours later. At 7:30 AM, FD horses were fed 2.1 kg DM grain-mix concentrate (187 IU vitamin E/kg DM) plus 3.3 kg DM mixed-grass hay, whereas FST horses received no feed. Neither group received any feed for 18 hours before 7:30 AM. Venous jugular blood samples were collected at 10:30 AM from all horses and analyzed for plasma TOC. The mean (±SD) within-horse paired-treatment difference was not significant (0.05 ± 0.12 μg/mL). The results suggest that variation in within-horse plasma TOC is relatively small over an 8-hour period in fasting horses and is unaffected 3 hours after a hay grain meal.  相似文献

OBJECTIVE: To determine pharmacokinetics of azathioprine (AZA) and clinical, hematologic, and serologic effects of i.v. and oral administration of AZA in horses. ANIMALS: 6 horses. PROCEDURE: In study phase 1, a single dose of AZA was administered i.v. (1.5 mg/kg) or orally (3.0 mg/kg) to 6 horses, with at least 1 week between treatments. Blood samples were collected for AZA and 6-mercaptopurine (6-MP) analysis 1 hour before and at predetermined time points up to 4 hours after AZA administration. In study phase 2, AZA was administered orally (3 mg/kg) every 24 hours for 30 days and then every 48 hours for 30 days. Throughout study phase 2, blood samples were collected for CBC determination and serum biochemical analysis. RESULTS: Plasma concentrations of AZA and its metabolite, 6-MP decreased rapidly from plasma following i.v. administration of AZA, consistent with the short mean elimination half-life of 1.8 minutes. Oral bioavailability of AZA was low, ranging from 1% to 7%. No horses had abnormalities on CBC determination or serum biochemical analysis, other than 1 horse that was lymphopenic on day 5 and 26 of daily treatment. This horse developed facial alopecia from which 1 colony of a Trichophyton sp was cultured; alopecia resolved within 1 month after the study ended. CONCLUSIONS AND CLINICAL RELEVANCE: Overall, no adverse effects were observed with long-term oral administration of AZA to horses, although 1 horse did have possible evidence of immunosuppression with chronic treatment. Further investigation of the clinical efficacy of AZA in the treatment of autoimmune diseases in horses is warranted.  相似文献

OBJECTIVE: To estimate prevalence of fecal shedding of Salmonella spp among horses in the US horse population and prevalence of Salmonella spp in grain or other concentrate used as horse feed on equine operations in the United States. DESIGN: Cross-sectional survey. SAMPLE POPULATION: Horses on 972 operations in 28 states. PROCEDURE: Fecal samples were collected from horses resident at each operation. Only a single sample was collected from any individual horse; number of horses from which samples were collected on each operation was determined on the basis of number of horses on the operation. A single sample of grain or concentrate was also collected from each operation. All samples were tested for Salmonella spp by means of bacterial culture. RESULTS: Overall, 0.8% (SE, 0.5) of resident horses shed Salmonella spp in their feces. The overall prevalence of operations positive for fecal shedding of Salmonella spp (i.e., operations with > or = 1 horse shedding Salmonella spp in its feces) was 1.8% (SE, 0.7). Prevalence of grain or other concentrate samples positive for Salmonella spp was 0.4%. Serotypes of Salmonella spp that were identified in grain or other concentrate were not those typically associated with clinical disease in horses. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the national prevalence of fecal shedding of Salmonella spp by horses in the United States was 0.8%, and that prevalence of Salmonella spp in grain or other concentrate used for horse feed was 0.4%.  相似文献

Three Standardbred horses were given 0.2 mg (1 mCi) of 75selenomethionine intravenously and a second group of three were given 10 mCi of tritiated diisopropylfluorophosphate (0.5 mg) intravenously. Observations on labeled cells were continued for 250 days after radioselenium injection and 160 days after tritium injection.

The lifespan of erythrocytes using 75selenmethionine was 155 ± 10 days and 148 ± 7.8 days using tritiated diisopropylfluorophosphate. There was no significant difference at the 10% level between the lifespans, using these labels. The uptake of radioselenium into erythrocytes reached a mean maximum of 11.5% at 82 ± 18.9 days after injection of the label. There was an elution of from 17.6% of the injected dose of tritium label down to 7.5% eight days after injection of this isotope. From this study both of these labels appear to be satisfactory for determining the erythrocyte lifespan of the horse.

The mean time of the curve of mean radioselenium activity of peripheral blood leukocytes for the three horses was 5.54 days and the lifespan of these cells was seven days. The mean lifespan of peripheral blood leukocytes of the three horses after in vivo labelling with tritiated diisopropylfluorophosphate was 17.3 hours. No specific labelling was found in bone marrow or peripheral blood cells at this level of tritium labelling by dipping emulsion autoradiography.

The mean lifespan of equine platelets for three horses using radioselenomethionine was 9.2 days and the mean lifespan of equine platelets using tritiated diisopropylfluorophosphate was 6.6 days in a group of three horses.

Whole blood containing 59Fe-labeled erythrocytes (RBC) and unlabeled serum was transfused from a donor horse on 2 occasions into each of 6 recipient horses. Survival of transfused cells was monitored in the recipients as a function of time after transfusion by measuring RBC radioactivity in the recipients. After the 1st transfusion, RBC concentration of 59Fe remained at 60% to 100% of the transfused dose for 4 days, after which radioactivity values dropped to less than 10% of the dose by 6 days in 3 horses. In the 3 other horses, RBC radioactivity dropped immediately after transfusion, reaching minimal values in approximately 48 hours. After the 2nd transfusion, 1 horse retained 80% of the dose in circulating RBC for 4 days; 2 horses demonstrated a rapid loss of circulating radiolabeled RBC, reaching minimal values in 48 hours; and 2 horses demonstrated minimal radioactivity in the RBC mass even immediately after the transfusion. One horse died of anaphylactic shock during the 2nd transfusion. Erythrocyte compatibility tests, using the direct agglutination test, the antiglobulin test, and the hemolytic test, were not effective in predicting survival of transfused RBC.  相似文献

The application of 99mTc-HMPAO labeled white blood cells to support the diagnosis of right dorsal ulcerative colitis was studied in two horses with a history and clinical signs consistent with phenylbutazone toxicity. These images were compared to a reference horse unaffected by right dorsal ulcerative colitis. Blood was collected aseptically in heparinized syringes from the patients for in vitro white blood cell (WBC) radiolabeling. The buffy coat was separated out and radiolabeled with 99mTc-HMPAO. The radiolabeled blood was re-injected i.v. and four images of the right and left side of the patient's abdomen were acquired at 4 hours and 20 hours post-injection. Results of the nuclear study revealed no abnormal findings in the abdomen at the four-hour post-injection images in any horse. Images obtained 20 hours post-injection revealed a linear uptake of radiolabeled WBCs in the right cranioventral abdomen in the region of the right dorsal colon in both horses with right dorsal ulcerative colitis. The reference horse had no radiopharmaceutical uptake in this region. This nuclear imaging study was a rapid, non-invasive method to identify right dorsal colon inflammation. These findings not only supported the diagnosis of right dorsal ulcerative colitis, but also facilitated appropriate medical management of each horse.  相似文献

Dexamethasone is a potent corticosteroid anti-inflammatory agent, and in most jurisdictions it is not legal to administer to any horse on the day of a race. This study was conducted to determine whether topical combination of dexamethasone and dimethyl sulfoxide (DMSO) solutions would result in detectable dexamethasone in the blood or urine in horses. Five different concentrations of dexamethasone/DMSO were used to replicate the combinations used on race horses. Serum cortisol concentrations were determined to detect an alteration in the hypothalamus-pituitary-adrenal axis. Dexamethasone/DMSO mixtures were applied topically to the distal limbs in five Standardbred mares. Blood and urine samples were collected at 0, 8, 24, and 32 hours. Samples were originally screened by a commercially available enzyme-linked immunosorbent assay (ELISA) test for dexamethasone. Any samples that were deemed suspect were then analyzed by liquid chromatography/mass spectrometry (LC/MS). Blood cortisol was assayed using a solid-phase chemiluminescence enzyme immunoassay. There was no detectable dexamethasone in either the urine or serum of any horse at any time point. The serum cortisol concentrations were within our laboratory's normal range at all time points. It appears doubtful that detectable blood and urine concentrations of dexamethasone are attributable to absorption from a topical application through intact skin.  相似文献

为了解阿哈尔捷金马经长途运输后血液生理生化反应的情况,试验选用23匹阿哈尔捷金马(公马14匹、母马9匹,年龄2~6岁),从俄罗斯出发经长途公路运输5 d到达新疆阿拉山口,在到达目的地后第7和35天,分别进行颈静脉采血,测定生理生化指标及应激相关激素水平,同时采用异地对照法与未经运输的15匹本地阿哈尔捷金马(公马10匹、母马5匹,年龄6~10岁)进行比较研究。结果发现:与本地阿哈尔捷金马相比,经长途运输后第7、35天的阿哈尔捷金马血液中性粒细胞(Gran)数量及占比、红细胞压积(HCT)、血小板(PLT)数量、碱性磷酸酶(ALKP)活性均有提高(P<0.05;P<0.01),而中间细胞(Mid)数量和百分比、血糖(GLU)水平显著或极显著降低(P<0.05;P<0.01);运输后第35天,平均红细胞体积(MCV)、磷(PHOS)、总蛋白(TP)含量均有所提高(P<0.01;P<0.01),淋巴细胞(LY)百分比则显著降低(P<0.05);运输后第7天抗利尿激素(ADH)、生长激素(GH)、尿素氮(UREA)及胰岛素(INS)含量均有下降(P<0.05;P<0.01),总胆红素(TBIL)、三碘甲状腺原氨酸(T3)水平均显著提高(P<0.05),但这些激素水平在运输后第35天均与对照组无显著差异(P>0.05)。综合以上结果,阿哈尔捷金马经长途运输后,出现了相应的一过性应激反应特征,经过一定的调整期(35 d)后大部分指标和未经长途运输马趋于一致,在此期间尚不能立即进入正常使用状态,以免导致长期不可逆性病变。  相似文献
