ABSTRACT Liquid swine manure added to acidic soils killed microsclerotia of the wilt fungus Verticillium dahliae. We investigated whether volatile fatty acids (VFAs) in the manure were responsible for this toxicity. The survival of microsclerotia was determined after exposure to various dilutions of manure or its VFA components. Acetic, propionic, and isobutyric acids constituted the major VFAs in the manure, while n-butyric, n-valeric, iso-valeric, and n-caproic acids were present in lesser amounts. Formic acid was not detected. The individual VFAs were more toxic to microsclerotia as the solution pH was decreased, indicating that the protonated forms of the VFAs were toxic (e.g., acetic acid and not acetate). The effective concentration reducing germination of microsclerotia by 95% (EC(95)) for formic and n-caproic acids was approximately 4 mM, the most toxic of the acids tested; for n-valeric, the EC(95) was 9.2 mM, isovaleric was 16.1 mM acids, and acetic, propionic, n-butyric, and isobutyric acids were approximately 30 mM. The toxicity of acetic acid, and likely all the others, was directly related to the duration of exposure. Inhibition of microsclerotia germination followed identical trends in solutions of the manure or in a mixture of VFAs with equivalent concentrations of the individual acids found in the manure. Similarly, germination declined to the same extent in the atmosphere above the manure or the VFA mixture, confirming the toxicity of VFAs to microsclerotia. Thus, under acid conditions, VFAs in liquid swine manure can kill microsclerotia of V. dahliae.  相似文献

ABSTRACT This study examined the mechanisms by which nitrogenous amendments such as meat and bone meal kill the soilborne plant pathogen Verticillium dahliae. The effect of nitrogen products from the amendments on the survival of microsclerotia of V. dahliae was examined by solution bioassay and soil microcosm experiments. Ammonia and nitrous acid but not their ionized counterparts, ammonium and nitrite, were toxic to microsclerotia in bioassays. In microcosms, addition of meat and bone meal (2.5%) to an acidic loamy sand resulted in the accumulation of ammonia and death of microsclerotia within 2 weeks. At lower concentrations (0.5 and 1%), microsclerotia were killed after 2 weeks when nitrous acid accumulated (>0.03 mM). In an alkaline loam soil, microsclerotia survived at 3% meat and bone meal and neither ammonia nor nitrous acid accumulated. The toxicity of ammonia to the pathogen was verified by increasing the concentration of meat and bone meal to 4% or addition of urea (1,600 mg of N per kg) to the loam soil resulting in the accumulation of ammonia (>35 mM) and death of microsclerotia. The toxicity of nitrous acid was verified by adding ammonium sulfate fertilizer to an acidic sand soil. Inhibiting nitrification with dicyandiamide revealed that nitrous acid was generated as a result of the accumulation of nitrite and an acidic pH. Thus, levels to which the toxins accumulated and the effective concentration of amendment were dependent upon the soil examined. Of the two mechanisms identified, accumulation of nitrous acid is the more promising strategy to control plant diseases in acidic soil because it is more toxic than ammonia and is formed at lower concentrations of amendments.  相似文献

This study was conducted to evaluate short- and long-term effects of poultry and sheep manures, as well as sunflower, canola, and potato residues, which commonly are being used as organic amendments (OAs) in agricultural soils, on the pH value, buffering capacity, and nitrogen (N) turnover of a sub-alkaline agricultural soil using laboratory incubation. The tested OAs had pH values ranging from alkaline (manures) to acidic (plant residues), and their effects of the short- and log-term on soil pH value were assessed by fractionation of the OAs alkalinity and evaluating the contribution of OA-borne N mineralization to soil acidity. On average, 45% of potential alkalinity in tested OAs was available for acid-base reactions, soluble alkalinity accounted for 58, 80, 32, 50, and 50% of the excess cations for poultry manure, sheep manure, sunflower, canola, and potato residues, respectively. Buffering capacity of the tested OAs was higher for sunflower residues, potato residues, and poultry manure and lower for canola residues and sheep manure. The tested OAs showed different N mineralization potential, and during the 100 days of incubation, soils amended with sheep manure, poultry manure, and potato residues released 107, 118, 129, and 240?mg?N?kg?1 soil, respectively, more than double than that released from unamended soil. Our results demonstrated that long-term impact of OAs on the soil pH value is not related to the initial OAs pH value and soluble alkalinity and that available alkalinity and N mineralization are key factors regulating the soil pH value on the long-term.  相似文献

 棉花品种抗黄萎病鉴定一般在田间病圃中进行,其结果受病圃中病原菌分布均匀程度、气象等因素影响极大,往往导致鉴定结果不准确。为了使鉴定方法简单、科学、可靠,我们在温室条件下比较了3种苗期接种棉花黄萎病的方法,即切根蘸孢子法(接种浓度为106分生孢子/mL);菌培养物土壤接种法(0.5%、1%、2%,w/w);微菌核土壤接种法(103个微菌核/g土)。结果表明,切根蘸孢子法导致棉苗发病均匀、严重、迅速,播种35~45 d后即可得到均匀一致的发病结果。而其它2种接种方法在播种75 d后才得到相对稳定的发病结果。同时,研究还表明接种浓度为104分生孢子/mL所导致的黄萎病显著比105或106的轻。利用切根蘸孢子法在室内鉴定12个棉花品种或品系的抗黄萎病能力,证明该方法是抗黄萎病快速鉴定的有效方法。此外,该鉴定方法还可快速鉴定黄萎病菌不同菌株的致病性,并可应用于作物对其它土传病害的抗病性鉴定上。  相似文献

ABSTRACT Microplot and field experiments were conducted to evaluate the effects of inoculum density on Verticillium wilt and cauliflower growth. Soil containing Verticillium dahliae microsclerotia was mixed with various proportions of fumigated soil to establish different inoculum densities (fumigated soil was used as the noninfested control). Seven inoculum density treatments replicated four times were established, and the treatments were arranged in a randomized complete block design. Soil was collected from each microplot immediately after soil infestation for V. dahliae assay by plating onto sodium polypectate agar (NP-10) selective medium using the Anderson sampler technique. Five-week-old cauliflower was transplanted into two beds within each 1.2- by 1.2-m microplot. At the same time, several extra plants were also transplanted at the edge of each bed for destructive sampling to examine the disease onset (vascular discoloration) after planting. Cauliflower plants were monitored for Verticillium wilt development. Stomatal resistance in two visually healthy upper and two lower, diseased leaves in each microplot was measured three times at weekly intervals after initial wilt symptoms occurred. At maturity, all plants were uprooted, washed free of soil, and wilt incidence and severity, plant height, number of leaves, and dry weights of leaves and roots were determined. The higher the inoculum density, the earlier was disease onset. A density of 4 microsclerotia per g of dry soil caused 16% wilt incidence, but about 10 microsclerotia per g of soil caused 50% wilt incidence. Both wilt incidence and severity increased with increasing inoculum density up to about 20 microsclerotia per g of soil, and additional inoculum did not result in significantly higher disease incidence and severity. A negative exponential model described the disease relationships to inoculum levels under both microplot and field conditions. Stomatal resistance of diseased leaves was significantly higher at higher inoculum densities; in healthy leaves, however, no treatment differences occurred. The height, number of leaves, and dry weights of leaves and roots of plants in the fumigated control were significantly higher than in infested treatments, but the effects of inoculum density treatments were variable between years. Timing of cauliflower infection, crop physiological processes related to hydraulic conductance, and wilt intensity (incidence and severity) were thus affected by the inoculum density. Verticillium wilt management methods used in cauliflower should reduce inoculum density to less than four micro-sclerotia per g of soil to produce crops with the fewest number of infected plants.  相似文献

ABSTRACT Broccoli is resistant to Verticillium dahliae infection and does not express wilt symptoms. Incorporation of broccoli residues reduces soil populations of V. dahliae. The effects of broccoli residue were tested on the colonization of roots by V. dahliae, plant growth response, and disease incidence of both broccoli and cauliflower in soils with different levels of V. dahliae inoculum and with or without fresh broccoli residue amendments. The three soils included a low-Verticillium soil, a high-Verticillium soil, and a broccoli-rotation soil (soil from a field after two broccoli crops) with an average of 13, 38, and below-detectable levels of microsclerotia per g of soil, respectively. ABSTRACT Experiments were conducted in field plots to evaluate the effects of broccoli residue on population dynamics of Verticillium dahliae in soil and on Verticillium wilt development on cauliflower under furrow and subsurface-drip irrigation and three irrigation regimes in 1994 and 1995. Treatments were a factorial combination of three main plots (broccoli crop grown, harvested, and residue incorporated in V.dahliae-infested plots; no broccoli crop or residue in infested plots; and fumigated control plots), two subplots (furrow and subsurface-drip irrigation), and three sub-subplots (deficit, moderate, and excessive irrigation regimes) arranged in a split-split-plot design with three replications. Soil samples collected at various times were assayed for V. dahliae propagules using the modified Anderson sampler technique. Incidence and severity of Verticillium wilt on cauliflower were assessed at 7- to 10-day intervals beginning a month after cauliflower transplanting and continuing until harvest. Number of propagules in all broccoli plots declined significantly (P < 0.05) after residue incorporation and continued to decline throughout the cauliflower season. The overall reduction in the number of propagules after two broccoli crops was approximately 94%, in contrast to the fivefold increase in the number of propagules in infested main plots without broccoli after two cauliflower crops. Disease incidence and severity were both reduced approximately 50% (P < 0.05) in broccoli treatments compared with no broccoli treatments. Differences between furrow and subsurface-drip irrigation were not significant, but incidence and severity were significantly (P < 0.05) lower in the deficit irrigation regime compared with the other two regimes. Abundance of microsclerotia of V. dahliae on cauliflower roots about 8 weeks after cauliflower harvest was significantly (P < 0.05) lower in treatments with broccoli compared with treatments without broccoli. Rotating broccoli with cauliflower and incorporating broccoli residues into the soils is a novel means of managing Verticillium wilt on cauliflower and perhaps on other susceptible crops. This practice would be successful regardless of the irrigation methods or regimes followed on the susceptible crops.  相似文献

 试验研究了土壤温度、湿度、pH值和有机质对棉花黄萎病菌微菌核存活的影响。结果表明:40、30和0℃对微菌核土壤存活影响较大,10和20℃影响较小,尤其以10℃影响最小;土壤含水量越高,对微菌核存活影响越大,在含水量低于15%时,对存活影响较小,尤其以5%的含水量影响最小,表明微菌核对土壤干旱逆境具有较强的抵抗能力;pH值低于5.5或高于8.5的土壤条件对微菌核存活影响较大,pH值为6.5~7.5影响较小;土壤有机质含量越高,微菌核存活率越低,含量低,存活率高。本研究结果表明,只要改善和控制棉花黄萎病菌土壤存活的生态条件,就可以有效地减少土壤存活菌量,达到控制病害发生与发展的目的。  相似文献

为了解北疆棉花黄萎病的发生情况及发病与土壤中黄萎病菌微菌核数量的关系和病原菌种群类型, 2021年对北疆石河子?奎屯?博乐等8市(县)棉田棉花黄萎病发病率?土壤中黄萎病菌的微菌核数量?菌株种群类型进行了抽样调查?结果表明, 北疆未发生棉花黄萎病的棉田占49.2%, 0%<发病率<5.0%的棉田占32.7%, 发病率≥5.0%的棉田占18%?与2013年?2015年相比, 2021年无病田率分别增加17.7和12.7百分点, 发病率≥5%的棉田分别减少15.7和21.6百分点?从棉田黄萎病发病率与土壤中微菌核数量的关系来看, 整体上北疆棉田棉花黄萎病发病率与微菌核数量相关性不显著(r=0.119 1); 分区域看, 石河子-沙湾片区?奎屯-乌苏片区?精河-博乐片区棉田黄萎病发病率与微菌核数量的相关系数分别为0.033 2?0.007 6?0.062 3, 均无显著相关性; 而呼图壁-玛纳斯片区棉田黄萎病发病率与微菌核数量的相关系数为0.635 7, 呈中度正相关?土壤中的黄萎病菌菌株以菌核型为主, 占57.9%, 菌丝型占23.2%, 中间型占18.9%?用特异性引物进行PCR检测表明, 土壤中黄萎病菌落叶型菌株占97.6%, 占绝对优势?本研究将为北疆棉区棉花黄萎病的综合防控提供理论依据?  相似文献

ABSTRACT Factor(s) involved in soil suppressiveness to Thielaviopsis basicola when hairy vetch was used as a green manure were studied in a cotton production system. Soil suppressiveness was assessed in vitro at hairy vetch amendment levels of 0, 0.25, and 0.75% (wt/wt) by observing chlamydospores, using a nylon fabric technique. Chlamydospore germination in all soils was below 5%, and microscopic examination showed no germ tube lysis or visible propagule destruction. Viability (chlamydospore germination on T. basicola-carrot-etridiazol-nystatin [TB-CEN] medium) was reduced by 29% within 48 h after hairy vetch amendment. Viability also was reduced in atmospheres of amended soils, suggesting that the suppressiveness was due to a volatile factor. In a field study, chlamydospore viability in amended soils was reduced by 16%. T. basicola hyphal growth was more sensitive to ammonia than Rhizoctonia solani or Pythium ultimum, and chlamydospore mortality of T. basicola was 100% in petri dish atmospheres with 0.4 ppm of ammonia (50% lethal dose = 0.15 ppm). Soil atmospheric ammonia was 0.08 and 0.10 ppm for 0.25 and 0.75% amendment levels, respectively, both at 3 and 7 days after incorporation. In the field, 0.11 and 0.14 ppm of ammonia were detected in soil atmospheres 3 and 7 days after incorporation, respectively. The levels of ammonia detected were sufficient to account for the loss in T. basicola chlamydospore viability, indicating that ammonia is responsible for the suppressiveness observed.  相似文献

微菌核是丝状真菌产生的一种休眠结构,由菌丝特化形成。实验室前期研究发现,淡紫紫孢菌在特定的培养条件下可以产生微菌核,并具有较强的抗逆性和稳定性。本研究通过实验室生测和温室盆栽试验,研究了淡紫紫孢菌微菌核对根结线虫的寄生作用和对黄瓜根结线虫病的防治效果,以及不同土壤条件对生防菌定殖的影响。结果表明,微菌核对根结线虫卵的寄生率随着接种浓度的提高而增加,当浓度为300 MS/mL时寄生率超过50%。黄瓜移栽时接种淡紫紫孢菌微菌核,用量为300个/100 g土时对根结线虫防效为44.7%,在育苗基质中提前接入微菌核可防效提高36.0%。土壤水分、温度和pH值对淡紫紫孢菌微菌核的定殖均有显著影响(P<0.05),含水量过高或过低不利于生防菌增殖,含水量为10%~15%,20 d时淡紫紫孢菌种群数量提高6~9倍。本研究为淡紫紫孢菌微菌核制剂的研发和应用提供了理论指导。  相似文献

Microsclerotia of Verticillium dahliae are produced in large numbers on senescing parts of host plants and remain viable in the soil for many years. Changes in the population density, i.e. density of microsclerotia, in the soil were measured in micro-plots using two isolates of V. dahtiae , specific to either field bean or potato, several crop sequences comprising potato, field beans and barley, and either the removal of aerial debris of the crops or incorporation into soil.
Potato was more susceptible to the potato isolate and field bean more susceptible to the field bean isolate. Removal of debris of potato and field bean reduced numbers of microsclerotia in the soil in the subsequent years, but removal of barley straw had no effect. Initially non-infested control micro-plots became infested, probably by the growth of potato roots into the naturally infested subsoil. The rate of increase of the microsclerotial population in the non-infested control micro-plots was larger than in the initially infested treatments, because more colonized debris was produced. It is concluded that removal of aerial debris of host crops is important to reduce the soil population of V. dahtiae.  相似文献

Soil solarization, either singly or in combination with a reduced dosage (34 g/m2) of methyl bromide, was effective in controlling verticillium wilt of globe artichokes for three successive cropping seasons. Both treatments resulted in greatly reduced natural populations of Verticillium dahliae microsclerotia, which were positively correlated with significant reductions in diseased plants as well as with the early formation of primary artichoke heads and increased yield. Propagules of Talaromyces flavus increased and survived better in solarized than in solarized and fumigated soils and could be partially involved in the effectiveness of solarization. Propagules of Aspergillus terreus invariably increased in treated plots but their involvement in the longevity of the treatment is questionable.  相似文献

Effects of crop rotation between rice paddy fields and strawberry nurseries on the control of Verticillium wilt of strawberry were studied. For detecting Verticillium dahliae, the causal agent of Verticillium wilt, in soil, eggplant was used as an indicator plant. We were thus able to detect as low as 1 microsclerotium/g dry soil. In field surveys of Chiba and Hokkaido from 2000 to 2003, V. dahliae was detected in 9 of 10 upland fields but in none of 21 paddy-upland fields. In Hokkaido during 2000–2007, strawberry mother plants were planted, and plantlets were produced in upland and paddy-upland fields to assess V. dahliae infestation. Verticillium wilt of strawberry had never occurred in 72 tested paddy-upland fields, compared to 13.2–73.9% of plantlets infected with V. dahliae in upland fields. In a pot experiment in a greenhouse, two flooding treatments or two paddy rice cultivations suppressed Verticillium wilt symptoms on eggplant. In field experiments, one paddy rice cultivation in Chiba and two in Hokkaido prevented development of Verticillium wilt symptoms on eggplant. Verticillium wilt of strawberry was controlled completely with one paddy rice cultivation in infested fields in Chiba. In these field experiments, the number of microsclerotia of V. dahliae decreased under the flooding conditions for paddy rice cultivation. Based on the reduction in microsclerotia, a crop rotation system with paddy rice for 3 years (three times), green manure for 1 year, and strawberry nursery for 1 year was designed for Hokkaido.  相似文献

ABSTRACT Six potato cultivars were grown with or without the addition of Verticillium dahliae inoculum and were watered at 50, 75, or 100% estimated consumptive use. The applied water x cultivar interaction was significant (P = 0.009 and P = 0.001 for 1996 and 1997, respectively) for the relative area under the senescence progress curve (RAUSPC). With a decrease in water, there was an increase in RAUSPC. A significant interaction of inoculum density x cultivar also was found, based on RAUSPC (P = 0.0194 and P = 0.0033 for 1996 and 1997, respectively). In V. dahliae-infested plots, 'Katahdin' and 'Ranger Russet' were resistant to Verticillium wilt. Population size of V. dahliae in stem apices was significantly lower in 'Katahdin' in both 1996 and 1997 (P = 0.0001) and in 'Ranger Russet' in 1997 (P = 0.0001) than in the other cultivars. 'Russet Burbank' and 'Shepody' had large apical stem populations of V. dahliae and higher RAUSPC values associated with both V. dahliae inoculum and decreased amount of applied water. Marketable tuber yield was unaffected by V. dahliae in both years. Cultivar resistance to Verticillium wilt was related to cultivar tolerance to moisture deficit stress. Results suggest that moisture deficit stress response has the potential to be a useful tool in protocols for screening potato for Verticillium resistance.  相似文献

Agricultural soil samples containing survival structures of the fungal crop pathogens Verticillium dahliae, Sclerotinia sclerotiorum, Sclerotium cepivorum, Pythium ultimum, potato cyst nematodes Globodera rostochiensis and G. pallida and weeds Chenopodium album and Agropyron repens [Elymus repens] were treated in the laboratory with aerated steam at temperatures ranging from 40 to 80°C in a specially constructed apparatus. Steaming at 50 or 60°C for 3min, followed by an 8-min resting period in the steamed soil and immediate removal from the soil thereafter, resulted in 100% kill of all weeds, diseases and nematodes. When steamed at 45°C, there was a small but significant reduction in the survival of V. dahliae microsclerotia but no reduction in survival of S. cepivorum.  相似文献

 微菌核是大丽轮枝菌在土壤中的主要存活结构和黄萎病的初侵染来源。对土壤中大丽轮枝菌微菌核进行定量是黄萎病监测和预警的基础。本研究以大丽轮枝菌Internal Transcribed Spacer (ITS)区特异性引物对P1/P2扩增产物的重组质粒为标准品,构建SYBR Green I实时荧光定量PCR反应的标准曲线,结合土样水筛法建立了土壤大丽轮枝菌微菌核定量检测体系。同时,建立了土壤中微菌核数量与棉花黄萎病发病率的关系模型。结果表明,实时定量PCR检测灵敏度比常规PCR高10倍,检测下限为1个微菌核/克土,在5.54×102~5.54×107copies范围内,DNA拷贝数的对数值与Ct值具有良好的线性关系。建立的土壤中微菌核个数n与Ct值之间的关系为n=e7.3-Ct/3.905。温室人工接种微菌核数量与棉花黄萎病发病率间的线性关系为y=2.710n+0.251。  相似文献

The dependence of the behaviour of metsulfuron-methyl on soil pH was confirmed during incubations under controlled laboratory conditions with two French soils used for wheat cropping. The fate of [14C] residues from [triazine-14C]metsulfuron-methyl was studied by combining different experimen-tal conditions: soil pH (8·1 and 5·2), temperature (28 and 10°C), soil moisture (90 and 50% of soil water holding capacity) and microbial activity (sterile and non-sterile conditions). Metsulfuron-methyl degradation was mainly influenced by soil pH and temperature. The metsulfuron-methyl half-life varied from five days in the acidic soil to 69 days in the alkaline soil. Under sterile conditions, the half-life increased in alkaline soil to 139 days but was not changed in the acidic soil. Metsulfuron-methyl degradation mainly resulted in the formation of the amino-triazine. In the acidic soil, degradation was characterised by rapid hydrolysis giving two specific unidentified metabolites, not detected during incubations in the alkaline soil. Bound residues formation and metsulfuron-methyl mineralisation were highly correlated. The extent of bound residue formation increased when soil water content decreased and was maximal [48 (±4)% of the applied metsulfuron-methyl after 98 incubation days] in the acidic soil at 50% of the water holding capacity and 28°C. 采用强制通风静态堆肥装置研究了添加木霉和鸡粪对牛粪高温堆肥过程中氮素形态及其相关微生物的影响。结果表明:加入木霉和鸡粪均使堆体高温期延长,高温期pH增加幅度减小。对照、加木霉和加鸡粪处理中高温期氨气释放量最高分别为413、197和216 mg/(kg.d);加木霉使高温期的氨态氮最高含量比对照低18.0%,加鸡粪使高温期氨态氮最高含量比对照多10.5%;在高温期,加木霉和加鸡粪处理中硝态氮最高含量分别是对照的1.85倍和1.71倍;堆肥结束时,对照中有机氮含量比初始减少19.1%、总氮含量减少21.6%,加木霉和加鸡粪处理中有机氮含量分别比初始增加4.5%和2.2%、总氮含量分别增加1.7%和5.5%,加木霉和鸡粪均有利于氨基糖态氮和氨基酸态氮的积累,有利于有机氮的形成和保留。整个堆肥过程中氨化细菌数量最大,氨化细菌和硝化细菌数量在加木霉和加鸡粪处理中均比对照多;高温阶段,反硝化细菌数量在加木霉处理中较对照多,固氮菌在加鸡粪处理中较对照多;降温阶段,固氮菌数在加木霉处理中较对照多。  相似文献
