In 1998, a total of 424 sows had sera collected in the Mekong delta in Vietnam. Of these, 283 sows were from 151 small-scale family farms in 19 villages, and 141 from seven large-scale state farms. The sera were subjected to the microscopic agglutination test (MAT) for antibodies to 13 Leptospira serovars. The overall leptospiral seroprevalence for titres > or =1:100 and > or =1:400, was 73 and 29%, respectively, and was higher (P=0.001) at small- than at large-scale farms. The highest seroprevalence was recorded for Leptospira interrogans serovar (sv) bratislava (52%). At small-scale farms, higher prevalences were found to serovars L. interrogans sv icterohaemorrhagiae (P=0.04) and L. interrogans sv pomona (P=0.02).Epidemiological information (at the individual-animal and herd-levels) was collected with a questionnaire. The data were analysed using logistic multiple regression. At the animal-level, sows seropositive for L. interrogans sv australis and sv autumnalis had less direct contact with sows in neighbouring pens (odds ratio (OR)=0.3 and 0.4, respectively) and sows seronegative for L. interrogans sv bratislava were of lower age (OR=0.1 for seropositivity). Also, sows seropositive for L. interrogans sv icterohaemorrhagiae had higher odds (OR=5.8) if they had not been born on the farm (had been introduced to it as gilts).Herds seropositive for sv javanica showed association with farms not taking measures to control the local rodent population (OR=7.8). Serovar pomona was also linked to the use of artificial insemination (AI), as opposed to natural-breeding services (OR=11.2).These results indicate that housing and management could affect the seroprevalence of Leptospira infection in pigs.  相似文献

A serological survey was undertaken to determine the prevalence of leptospirosis, and to investigate associations between leptospiral antibody titers, and herd measures of reproduction. Production records and leptospirosis serology were analyzed for 25 slaughter hogs from each of eleven randomly sampled farrow-finish operations on Prince Edward Island. The effect of selected leptospiral serovars on nonproductive sow days per parity (NPSD/P) and the proportion of pigs born dead was evaluated. The four most common serovars to which antibodies were detected were Leptospira icterohaemorrhagiae, L. bratislava, L. autumnalis and L. pomona, with respective prevalences of 57.1%, 35.1%, 3.4% and 1.5% of PEI slaughter hogs. None of these serovars was associated with increased frequency of stillbirths (p greater than 0.05). However, farms with a higher prevalence of L. bratislava antibody titers tended to have more infertility, as measured by NPSD/P (r = 0.738, p = 0.036 with Bonferroni adjustment). Also, farms with L. pomona antibody titers had higher NPSD/P than farms without L. pomona antibody titers (p = 0.0008 with Bonferroni adjustment). There was no association between NPSD/P and antibodies to either L. autumnalis or L. icterohaemorrhagiae (p greater than 0.05).  相似文献

Serology plays an important role in the diagnosis of leptospirosis. Few laboratories have the resources, expertise, or facilities to perform the microscopic agglutination test (MAT). Thus, there is a need for a rapid and simple serological test that could be used in any diagnostic laboratory. In this study, a genus-specific, heat-stable antigenic preparation from Leptospira interrogans serovar pomona was used in an enzyme-linked immunosorbent assay (ELISA) for the detection of leptospiral antibodies in dog sera. This antigenic preparation reacted with rabbit antisera against L. interrogans serovars bratislava, autumnalis, icterohaemorrhagiae and pomona and with rabbit antiserum against L. kirschneri serovar grippotyphosa. The ELISA showed a relative specificity of 95.6% with 158 dog sera which were negative at a dilution of 1:100 in the MAT for serovars pomona, bratislava, icterohaemorrhagiae, autumnalis, hardjo, and grippotyphosa. The relative sensitivity of this assay with 21 dog sera that revealed serovars MAT titres of > or =100 to different serovars was 100%. This assay is easily standardized, technically more advantageous than MAT, and uses an antigenic preparation that can be routinely prepared in large amounts. It was concluded that this ELISA is sufficiently sensitive test to be used as an initial screening test for the detection of leptospiral antibodies in canine sera, with subsequent confirmation of positive test results with the MAT.  相似文献

Sera collected from 511 unvaccinated urban dogs were examined for antibodies to serovars of Leptospira interrogans representing nine serogroups found in the United Kingdom. The point estimates of the seroprevalence of leptospiral titres in Edinburgh and Glasgow at a minimum serum dilution of 1:10 were 23-5 per cent and 27-5 per cent, respectively, and, in Glasgow, the seroprevalence was significantly higher in males than females, for both crude and age-adjusted values. Serovar canicola was the dominant strain identified, followed by serovars icterohaemorrhagiae, bratislava and ballum. Antibodies against serovars javanica and hardjo also were detected. Urine samples were collected from 84 of the male blood-sampled dogs, and cultured. Serovars canicola and bratislava were isolated from three and four samples, respectively.  相似文献

Sera from Thoroughbred and Standardbred horses in southwest Ontario were tested for antibody to seven Leptospira interrogans serovars (autumnalis, bratislava, canicola, grippotyphosa, hardjo, icterohaemorrhagiae, pomona), using the microscopic agglutination test. There was significantly higher seroprevalence of bratislava than of other serovars, in which prevalence was low. Seroprevalence of bratislava increased significantly with age; only 5% of two to three year old horses had titers greater than or equal to 1:80 compared to 52% of horses older than seven years. Eight of 16 foals from two farms seroconverted at low titers to bratislava between four and eight months of age. Leptospires were not detected by immunofluorescence and isolation techniques in 50 kidneys collected from horses at slaughter. Fetal tissues from 52 aborted horse fetuses were also examined by these methods and serovar kennewicki was identified by immunofluorescence and by isolation in one fetus. Serovar bratislava appears to be widespread in horses in Ontario but unimportant in abortion. The clinical significance of this infection in horses in Ontario is unclear.  相似文献

The prevalence of reactors to different serovars of Leptospira interrogans was determined in 117 swine premises in Quebec. A total of 926 sera were tested, using the microscopic agglutination method, against six leptospiral serovars: pomona, icterohaemorrhagiae, grippotyphosa, hardjo, canicola and ballum. Of the sera tested, 30 (3.2%) reacted to one of the three following serovars: pomona (2%), icterohaemorrhagiae (1.1%) and ballum (0.1%).  相似文献

Two dogs with clinical histories suggestive of leptospirosis were examined serologically and culturally for evidence of leptospiral infection. Antibodies to Leptospira interrogans serovar bratislava were detected in serum from one dog, and the organism was isolated from urine of that dog. In a serologic survey of dogs in the state of Illinois, reactor rates to bratislava were higher than those to canicola or icterohaemorrhagiae. In cases of suspect canine leptospirosis, serovars such as bratislava, not contained in canine vaccines, should be considered in a differential diagnosis.  相似文献

Outdoor reared pigs were used as indicators for investigating the effect of weather conditions in the seroprevalence of Leptospira. Over the period February to March 2008, sera from 386 sows on 11 farms in southern Sweden were tested for antibodies to the following Leptospira serovars: L. interrogans serovar (sv) Bratislava, L. kirschneri sv Grippotyphosa, L. interrogans sv Icterohaemorrhagiae, L. interrogans sv Pomona, L. borgpetersenii sv Tarassovi and one domestic strain (mouse 2A) related to L. borgpetersenii sv Sejroe and L. borgpetersenii sv Istrica. The highest seroprevalence was to this strain (8.0%) followed by sv Bratislava (3.9%). Six of the 11 farms had sows which were seropositive to at least one of the Leptospira serovars. Data on rainfall and temperature were retrieved for the respective farms. For each millimetre of extra rainfall, there was an increase in the odds ratio (OR) for seropositivity to sv Bratislava of 4.3 (95% CI 1.9-10), and to strain mouse 2A of 2.5 (95% CI 1.0-6.4). There was no association between seropositivity and temperature. This study indicates that different climate conditions within the northern temperate climate zone may be of importance for the presence of Leptospira-seropositivity in mammals.  相似文献

Fourteen dogs which had died from acute leptospirosis, three dogs which survived an acute attack of leptospirosis and seven healthy dogs living in close contact with the animals which died were examined for the presence of lepto-spiral antibodies by the microscopic agglutination test. Twelve dogs had titres of 1:3200 or greater. The predominant titre was directed against serovar bratislava in seven cases, serovar grippothyphosa in two cases and both these serovars in two cases. In one case the predominant titre was directed against serovars bratislava, grippothyphosa, copenhageni (serogroup icterohaemorrhagiae) and pomona. The present study and other recent reports suggest that the epidemiology of canine leptospirosis is changing with the emergence of serovars differing from those typically infecting dogs, namely canicola and icterohaemorrhagiae.  相似文献

Eighty-seven cats from the Glasgow area were investigated to determine the prevalence of leptospiral infection and possible resultant disease. Eight (9-2 per cent) of the cats reacted serologically with the antigens of three serovars. Five of the cats were seropositive to Leptospira hardjo, two to L autumnalis and one cat serocon-verted to L icterohaemorrhagiae. A paired serum sample indicated a recent infection in one of the cats. The major clinical sign shown by this cat was ascites. Four of the cats were from rural areas. This appears to be the first report of a serological survey of leptospiral infection in cats in the Glasgow area.  相似文献

Swine herds suspected to be infected with Leptospira interrogans serovar bratislava were vaccinated with bacterins containing 5 or 6 leptospiral serovars in which serovar bratislava was the unique component. The principal diagnostic feature indicating an infection by this organism was demonstration of antibody against serovar bratislava in sera from stillborn pigs. For 1 breeding cycle after vaccination of herds on 3 farms, 255 of 266 (95.9%) sows and gilts given the 6-serovar bacterin farrowed. In contrast, 233 of 311 (74.9%) sows and gilts given the 5-serovar bacterin farrowed. These results, as evaluated by analysis of variance techniques, showed a significant improvement (P less than 0.01) in reproductive performance for groups vaccinated against serovar bratislava.  相似文献

The clinical features of the disease are presented based on retrospective analysis of the records of eleven dogs diagnosed with leptospirosis using clinical signs and results of the microagglutination test (MAT) between 1991 and 1996. Additionally, Leptospira titres were determined in 30 healthy dogs and 20 hospitalised dogs without clinical or laboratory evidence of leptospirosis. A positive titre for L. grippotyphosa, L. pomona, L. bratislava, L. australis, L. icterohaemorrhagiae and/or L. canicola was found in 16 normal dogs and only one hospitalised patient. Eight of these dogs had titres of > or = 1:800. Only one of them had been vaccinated shortly before sampling. These results suggest that many dogs from the surroundings of Bern, Switzerland have contact with various Leptospira interrogans serovars. In ten healthy dogs, the Leptospira titre was determined before and four weeks after vaccination with leptospiral antigen. Only two of the dogs showed a serologically measurable response to the antigen contained in the vaccine. In dogs MAT titers presumably do not reliably reflect the immune status against leptospiral infections.  相似文献

SUMMARY Serum samples from 272 horses, some III, were tested by the microscopic agglutination test for the presence of antibody to 12 serovars of Leptospira interrogans. Serums from 41.5% of horses reacted to one or more of the serovars tested; the most common reactions were to L. interrogans serovar ballum (15.1%), L. interrogans serovar autumnalis (11.8%), L. interrogans serovar icterohaemorrhagiae (9.9%), L. interrogans serovar pomona (8.1%) and L. interrogans serovar hardjo (7.7%).  相似文献

Serum samples obtained from 1.537 cattle in the 14 local government areas (LGAs) of Plateau State of Nigeria were screened for the presence of leptospiral antibodies using 13 serovars in a modified microscopic agglutination test (MAT). Two hundred and twenty-two (14.4 p.100) of the cattle tested had leptospiral antibody titres of 1:100 or higher to one or more of the test antigens. The prevalence rates of antibodies to individual serovars were: hardjo (35.6 p.100), pomona (11.7 p.100), pyrogenes (11.7 p.100), canicola (9.5 p.100), grippotyphosa (7.7 p.100), bratislava (5.9 p.100), icterohaemorrhagiae (5.9 p.100), ballum (4.5 p.100), autumnalis (3.6 p.100), bataviae (2.3 p.100) and tarassovi (1.8 p.100). The serological prevalence of bovine leptospirosis in the various local government areas of Plateau State of Nigeria differed significantly (P less than 0.05; X2).  相似文献

We surveyed a wild population of white-lipped peccaries (Tayassu pecari) in the Brazilian Pantanal for evidence of Leptospira interrogans. Serum samples from 71 free-ranging T. pecari were obtained between 2003 and 2005 in the southern Pantanal of Mato Grosso do Sul state. We used microscopic microagglutination to test for antibodies against 14 L. interrogans serovars (antibody titers ≥1:100 were considered seropositive). Seventy percent of captured animals tested positive for leptospirosis antibodies. Antibodies against icterohaemorrhagiae and autumnalis serovars were the most prevalent. We used log-linear analyses to test for associations among seropositivity, age class, and sex of captured animals. Seropositivity was strongly associated with animal age class, but independent of sex. Forty-six percent of animals less than 2 years old, 63% of adults during peak reproductive years, and 100% of the oldest age class were seropositive. A nonparametric multivariate procedure (MRPP) showed that the composition of serovar antibody types changed with age, and ANOVA models demonstrated that antibody titers increased with age, suggesting long-term exposure to a greater number and variety (i.e., serovar types) of L. interrogans infections. This study presents the first quantitative survey of antibodies against L. interrogans serovars in a T. pecari population of the Pantanal. The high prevalence of leptospirosis antibodies in free-ranging white-lipped peccaries and the potential impacts on reproduction and population dynamics emphasize the need for further studies investigating the roles of Pantanal wildlife and livestock in the transmission and maintenance of L. interrogans in the environment.  相似文献

Background

Leptospiral infections in poikilothermic (cold blooded) animals have received very little attention and the literature concerning natural infections of these animals is limited. The aim of this study was to determine the prevalence of leptospiral antibodies in reptiles, imported into Slovenia and intended to be pets in close contact with humans. A total of 297 reptiles (22 snakes, 210 lizards and 65 turtles) were tested for specific antibodies against serovars of Leptospira interrogans sensu stricto using the microscopic agglutination test (MAT). Live cultures of different serovars were used as antigens. MAT was performed according to standard procedures and the degree of reaction was interpreted by estimating the percentage of agglutinated leptospires. Samples showing titres of ≥ 50 against one or more serovars were considered as positive.

Results

Antibodies against seven pathogenic serovars of L. interrogans sensu stricto were detected in 46 of 297 reptiles. Among 22 snakes, specific antibodies against pathogenic serovars of three Leptospira species (L. interrogans, L. kirschneri and L. borgpetersenii) at titre levels from 1:50 to 1:400 were detected in 6 snakes. In 31 of 210 lizards, specific antibodies were found in titres from 1:50 to 1:1000 and, finally, among 65 turtles (terrapins and tortoises), 9 had specific antibodies at titre levels between 1:50 and 1:1600. Animals imported from non-EU countries showed significantly higher prevalence (25.0%; 95 confidence interval: 16.7–33.3%) than animals from EU member states (10.4%; confidence interval: 6.1–14.7%).

Conclusions

Reptiles may be considered as potential reservoirs of L. interrogans sensu stricto. Origin of the animals is a risk factor for presence of leptospiral antibodies, especially in lizards. Special attention should be focused on animals from non-EU member states.  相似文献

Sera obtained from cattle in the state of Yucatan, Mexico, were screened using the microscopic agglutination test against 13 serovars of Leptospira interrogans. A total of 62.8% (461/734) cows were positive for one or more serovars. This seroprevalence probably reflects infection because vaccination against leptospirosis has not been practised in Yucatan. The most common antibodies detected were those against antigens of serovars hardjo (54.1%) and tarassovi (53.3%). Region was the only risk factor associated with the seroprevalence of leptospirosis (p < 0.05). This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

All leptospirosis microscopic agglutination test titers for the Leptospira serovars icterohaemorrhagiae, canicola, grippotyphosa, bratislava, hardjo, and pomona conducted on 1,260 blood samples from dogs at the University of Illinois Veterinary Diagnostic Laboratory between March 1996 and March 2001 were evaluated. Low titers (1:100 to 1:400) were predominantly L. icterohaemorrhagiae and L. canicola, which represented the predominant serovars (65.4%) among all positive samples with low titers. L. grippotyphosa was the predominant serovar (72.1%) among samples with clinically significant titers (greater than 1:800). The medical records of 87 dogs with a titer greater than 1:800 that were patients at the Veterinary Teaching Hospital of the University of Illinois were reviewed. A clinical diagnosis of leptospirosis was made in 15 cases (17.2%) based on the elevated titer, appropriate clinical signs, lack of recent vaccination, and lack of concurrent disease that could explain the clinical signs present. Renal disease was present in 10 of the cases, concurrent renal and hepatic disease in two, and hepatic disease in three. In 12 cases, the predominant serovar was L. grippotyphosa; titers to L. grippotyphosa and L. bratislava were equal in magnitude in three cases.  相似文献

Out of 571 serologically investigated dogs from Bratislava and all regions of Slovakia and Moravia, 26.1% presented antibodies reacting with leptospiral antigens. The lowing seropositivity in dogs from Bratislava was found in lap dogs--11.1%, the highest one in farm dogs--35.3% (Tab. I). The seropositivity of police dogs from different regions of Slovakia and Moravia varied from 24.0% to 47.6% (Tab. II). The age of dogs progressing, their seropositivity rose from 3.1% in animals younger than one year to 33.3% in those aged seven years and in older ones (Tab. III). The predominant serovars causing infections of dogs were L. grippotyphosa (58.9% of infections) and serovars of both Sejroe (21.0%) and Icterohaemorrhagiae groups (15.3%). The serovars bratislava/jalna, pomond/mozdok and tarassovi were only rarely involved in these infections, canicola and arborea only exceptionally (Tab. IV). At present, in the Czechoslovak conditions the dogs do not play a significant role as reservoirs of human leptospirosis, and the predominant serovars may only cause irregular and short-time lasting leptospiruria. This is why the reasonableness of actual vaccination of dogs, possibly with the exception of the vaccination against Weil's disease, is doubtful. Dog is a good indicator of the distribution of different leptospiral serovars in its environment. It seems reasonable to practice occasionally serological investigations of these animals in order to detect possible changes in infecting leptospiral serovars. Consequently, adequate epizootological measures could be taken (e. g. modification of the composition of a leptospiral vaccine for dogs).  相似文献
