Background

Blood feeding by free-living insect vectors of disease is rhythmic and can be used to predict when infectious bites will occur. These daily rhythms can also be targeted by control measures, as in insecticide-treated nets. Culex pipiens form pipiens and C.p. f. molestus are two members of the Culex pipiens assemblage and vectors of West Nile Virus throughout North America. Although Culex species vector human pathogens and parasites, the daily blood feeding rhythms of C.p. f. molestus, to our knowledge, have not been studied. We described and compared the daily blood feeding rhythms of three laboratory-reared populations of Culex pipiens, one of which has confirmed molestus ancestry. We also examined the plasticity of blood feeding time for these three populations.

Results

For most (>70%) C.p. f. pipiens and C.p. f. molestus collected from metropolitan Chicago, IL, blood feeding took place during scotophase. Peak blood feeding occurred in mid-scotophase, 3-6 hours after lights off. For C.p. f. pipiens originating from Pennsylvania, most mosquitoes (> 90%) blood fed during late photophase and early scotophase. C.p. f. molestus denied a blood meal during scotophase were less likely to blood feed during early photophase (< 20%) than were C.p. f. pipiens from Chicago (> 50%). C.p. f. pipiens from Pennsylvania were capable of feeding readily at any hour of photo- or scotophase.

Conclusions

Daily blood feeding rhythms of C.p. f. molestus are similar to those of C.p. f. pipiens, particularly when populations originate from the same geographic region. However, the timing of blood feeding is more flexible for C.p. f. pipiens populations relative to C.p. f. molestus.  相似文献

Culex pipiens mosquitoes are vectors to many viruses and can transmit diseases such as filariasis and avian malaria. The present study evaluated the larvicidal activity of marine-derived endophytic fungi Aspergillus nomius and Aspergillus flavus from the soft coral Sarcophyton ehrenbergi along with two known cyclodepsipeptide compounds, scopularide A (1) and B (2), isolated from A. flavus extract, against third-instar larvae of C. pipiens, using distilled water as a negative control and toosenedanin as a positive control. The structures of the isolated compounds were confirmed by various spectroscopic analyses. The lethal concentrations (LC50 and LC90) were calculated by probit analysis. Scopularide A was the most potent after 96 h treatment, with LC50 and LC90 values of 58.96 and 994.31 ppm, respectively, and with 82.66% mortality at a concentration of 300 ppm. To unravel the biochemical mechanism of the tested extracts and compounds, their effects against protease, chitinase, phenoloxidases and lipase enzymes from the whole-body tissue of C. pipiens were evaluated after 72 h treatment at LC50 dose. Superior activity was observed for A. flavus extract against all tested enzymes. A molecular docking study was conducted for scopularide A and B on the four tested enzymes, to further verify the observed activity. Results revealed good binding affinities for both compounds as compared to the docked ligands, mainly via a number of hydrogen bonds. This was the first study to report the isolation of endophytic fungi A. flavus and A. nomius from the marine soft coral S. ehrenbergi. The endophytic fungal extract of A. flavus was found to be a promising source for a natural larvicidal agent against C. pipiens populations.  相似文献

The full whole culture (FWC), containing parasporal protein toxins of Bacillus thuringiensis israelinsis (Bti) and Bacillus sphaericus 2362 (Bs), either singly or in combination with plant oils and commercial insecticides, was tested against larval and adult stages of Culex pipiens mosquitoes under controlled laboratory conditions. In terms of LC50 values recorded after 24, 48, 72 and 96 h, the bacterial toxins showed high potency towards both larvae and adults of mosquitoes in a dose-dependent manner. Generally, the Bti toxin seemed to be more potent than the Bs toxin. For example, the Bti toxin showed a 24 h LC50 of 8.2 ppm against mosquito larvae compared to 13.6 ppm for the Bs toxin. In the adult bioassay, the obtained 24 h LC50 values were 0.064 and 0.085 mg/cm2, respectively for the two bacterial toxins. The bacterial toxins mixed with plant oils or insecticides at equitoxic doses (e.g., LC25 values) mostly showed potentiation effects, either against larvae or adults of the tested insect. Among a total of 14 paired mixtures, only the joint action estimated for the mixture of malathion + Bti or Bs was accounted as additively. Combining Bti or Bs endotoxins at LC0 with different plant oils and insecticides at LC50 concentration levels each, has resulted in considerable synergism against either larvae or adults. In the case of larval bioassays, the maximum synergistic factor (SF) obtained (ca. 2.0) was entitled to the mixture of Bti + spinosad. In the adult bioassays, the mixtures containing Curcuma longa or Melia azedarach oil extracts with Bti or Bs toxins achieved a SF accounted to 2.0. The results of the present study may be considered as an additional contribution to the area of joint toxicity of biocidal agents combining bacterial toxins, plant oils and traditional insecticides. The reached findings may encourage future research to elucidate its performance under practical field conditions.  相似文献

Glycoalkaloids are a class of secondary compounds (nitrogenous, steroidal glycosides), ubiquitously distributed throughout the Solanaceae. Numerous studies (in planta) have shown that certain glycoalkaloids, e.g., α-tomatine, solanocardenine, and leptine, have a negative impact on performance of the Colorado potato beetle,Leptinotarsa decemlineata (Say). However, the presence of other secondary compounds, as well as the nutritional status of the plants used, are additional variables in such experiments. By rearing Colorado potato beetle, from egg to prepupal stage, on a synthetic diet supplemented with the glycoalkaloids α-tomatine, α-chaconine, α-solanine, leptine I and the steroidal aglycone solanidine, we have been able to further establish the effects (ex planta) of these compounds on the Colorado potato beetle. Leptine I displayed a dose-dependent negative activity against the Colorado potato beetle (as measured by larval weight gain and tune to molt), when assayed at 0.31, 0.62 and 1.23 mM concentrations; however, by the fourth stadium no effect was found. When Colorado potato beetle were fed a higher concentration of leptine I (2.4 mM), there was a sustained effect in all stadia. At 2.4 mM, leptine I displayed a greater negative impact on Colorado potato beetle growth and development than did α-tomatine. α-Chaconine at 2.4 mM did not impair Colorado potato beetle performance relative to Colorado potato beetle reared on control diets. α-Chaconine plus α-solanine, at concentrations commonly found inSolanum tuberosum L. foliage (0.6 and 0.3 mM respectively), did not impair Colorado potato beetle performance. The steroidal aglycone solanidine (2.4 mM) had a significant negative impact on Colorado potato beetle performance. Results are discussed in the context of host-plant resistance and insect-plant interactions.  相似文献

We investigated color preference, seasonal abundance, spatial distribution and species composition of thrips in northern highbush blueberries, Vaccinium corymbosum L., in New Jersey (USA). White sticky traps were more attractive to thrips compared with yellow or blue traps. Thrips captures using white sticky traps showed that their flight activity begins 20–30 d after the onset of flowering, with 10, 50 and 90% of trap captures observed at 383, 647 and 1231 degree-day accumulations, respectively (10 °C base temperature). Two methods were used to study thrips distribution within a blueberry bush. First, white sticky traps were placed within the bush canopy at three different heights. The highest numbers of thrips were caught on traps in the middle and top one-third of the canopy while the lowest numbers were caught in the bottom one-third. A second method determined the distribution of thrips on the blueberry plant at different heights and phenological stages. The highest numbers of thrips were found on young leaves at lower parts of the canopy, whereas flowers and fruit had fewer thrips and none were found on buds; these thrips were identified as, Scirtothrips ruthveni (88% of adults) and Frankliniella tritici (12%). The distribution of thrips within a blueberry planting was investigated using an evenly-spaced grid of white sticky traps in combination with on bush beating-tray samples. Thrips counts from traps correlated with direct counts on the bush across the entire blueberry field (macro-scale level); however, within the field (micro-scale level), there was no correlation between the number of thrips on traps and on individual bushes near traps. Early in the season, trap counts were higher on bushes closer to the forest, indicative of movement of thrips from wild hosts into blueberry fields. However, this was not the case for direct on bush counts or trap counts for the later part of the season, where there was no clear forest “edge” effect. Percent fruit injury due to thrips feeding was low, and it correlated with thrips counts on bushes but not from counts on traps. Overall, our data show that thrips counts on sticky traps need to be interpreted with care because these numbers weakly correlated with the numbers of thrips on bushes at the micro-scale level and percent fruit injury; however, they can be useful predictors of thrips activity across entire blueberry fields (macro-scale).  相似文献

Haematococcus pluvialis is a microalgae actively studied for the production of natural astaxanthin, which is a powerful antioxidant for human application. However, it is economically disadvantageous for commercialization owing to the low productivity of astaxanthin. This study reports an effective screening strategy using the negative phototaxis of the H. pluvialis to attain the mutants having high astaxanthin production. A polydimethylsiloxane (PDMS)-based microfluidic device irradiated with a specific light was developed to efficiently figure out the phototactic response of H. pluvialis. The partial photosynthesis deficient (PP) mutant (negative control) showed a 0.78-fold decreased cellular response to blue light compared to the wild type, demonstrating the positive relationship between the photosynthetic efficiency and the phototaxis. Based on this relationship, the Haematococcus mutants showing photosensitivity to blue light were selected from the 10,000 random mutant libraries. The M1 strain attained from the phototaxis-based screening showed 1.17-fold improved growth rate and 1.26-fold increases in astaxanthin production (55.12 ± 4.12 mg g−1) in the 100 L photo-bioreactor compared to the wild type. This study provides an effective selection tool for industrial application of the H. pluvialis with improved astaxanthin productivity.  相似文献

Understanding collection methodologies and their limitations are essential when targeting specific arthropods for use in habitat restoration, conservation, laboratory colony formation, or when holistically representing local populations using ecological surveys. For dung beetles, the most popular collection methodology is baited traps, followed by light traps and unbaited flight-intercept traps during diversity surveys. A less common collection method, flotation, is assumed to be laborious and messy, and so only a handful of papers exist on its refinement and strengths. Our purpose was threefold: First, we tested the recovery and survival rates of Labarrus (=Aphodius) pseudolividus (Balthasar) and Onthophagus taurus (Schreber) when floating beetle-seeded dung pats to determine potential collection and safety issues. We collected 72.4 and 78% of the seeded L. pseudolividus and O. taurus, respectively, with >95% survival rating. Second, we developed a flotation-sieving technique that enables users to rapidly collect and passively sort dung beetles with less time and effort. Specifically, we often collected 50–100 g of wild dung beetles within a couple of hours of gathering dung and sorted them in a couple more by allowing dung beetles to sort themselves by size within a series of sieves; Third, we reviewed flotation-based advantages and disadvantages in comparison to other methodologies.  相似文献

Continuous monitoring of vector species composition, abundance, dynamics, feeding pattern, and host finding strategy is the base to determine when, what, and how control should be implemented. Thus, this study was conducted to assess entomological parameters of anopheline mosquitoes in nine villages in Seka district, southwestern Ethiopia, from June to December 2012. Mosquito collection was carried out from selected households in each of the nine study villages using light trap catches from June to December 2012. Differences in mean mosquito density, parity rates before, and after indoor residual spraying (IRS) operation were compared. In total, 1,136 adult female anopheline mosquitoes were collected during the study period. All anopheline mosquitoes collected belong to three species. Anopheles gambiae senso lato Giles was the most predominant (69.7%) followed by Anopheles coustani s.l. Laveran (22.7%) and Anopheles pharoensis Theobald (7.6%). There was significant variation in mean mosquito density among An. gambiae s.l., An. coustani s.l., and An. pharoensis. Parity rate of An. gambiae s.l. before spray operation was significantly higher than after spray operation. The highest peak biting activity of An. gambiae s.l. was between 1800 and 2100 hours. The longevity of An. gambiae s.l. ranged from 3.4 to 12.5 d. The highest vector abundance and parity rate were recorded in July and August. In conclusion, the behavioral plasticity and early biting activity of An. gambiae s.l. could affect current vector control tools (IRS and long lasting insecticidal nets). Hence, it is imperative to explore intervention tools for outdoor malaria vector control in addition to the existing IRS and long-lasting insecticidal nets.  相似文献

Controlled environment experiments were used to determine the effect of light intensity on levels of glycoalkaloids and resistance to the Colorado potato beetle (CPB),Leptinotarsa decemlineata Say, inSolanum chacoense Bitter clones that synthesized acetylated glycoalkaloids, the leptines. Plants were grown under two light intensities, 50 or 250 ?mol m-2S-1, and bioassayed for CPB resistance by placing neonate larvae on excised foliage. Larval development stage and mortality were recorded after four days. The leptine-synthesizing clones responded to the increase in light intensity with a 2–4 fold increase in level of foliar leptines. Larval development rate on foliage from plants grown under high light intensity was reduced and larval mortality increased. Results showed a significant (p>0.05) effect of light on glycoalkaloid synthesis and CPB resistance.  相似文献

An essential component of all mosquito-rearing activities is the act of blood-feeding the mosquitoes (Diptera: Culicidae). Many options exist for this purpose including live host animals and a diverse array of artificial-feeding methods. Most of the published artificial-feeding methods involve expensive materials, custom-built devices, or are labor-intensive. All of the previously published methods utilize blood sources, which are either expensive, or difficult to obtain. Additionally, much of the research into artificial blood-feeding methods for mosquitoes has focused on two species: Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse). This article presents a modified artificial blood-feeding method that uses affordable and easily sourced materials, does not require any technical knowledge to assemble, and requires minimal time and effort. The combination of inexpensive aluminum plates, Parafilm and polytetrafluoroethylene tape membranes, an electric germination mat, and frozen, food-grade blood produces exceptional feeding rates and abundant egg production. The method has been used for 2 yr at the Lee County Mosquito Control District to successfully maintain laboratory colonies of four species of mosquito: Ae. aegypti, Ae. albopictus, Aedes taeniorhynchus (Wiedemann), and Culex quinquefasciatus (Say). Variations of this method are reported, which can be used for wild and laboratory colonies of multiple species. This modified method is highly accessible for any small-scale mosquito rearing facility with labor or budgetary constraints.  相似文献

Aphid parasitoids are common in Brazilian wheat fields, and parasitize aphids at the wheat tillering stage. However, there is little information available about when this natural enemy occurs in wheat crops. This study investigated the initial occurrence of aphid parasitoids in four commercial wheat crops in northern Paraná during the 2009 crop season. We installed two Malaise traps at each wheat farm, and 400 tillers were assessed weekly in each field for aphid abundance. During this study, we captured 4,355 aphid parasitoids and 197 aphids. Three species of braconid parasitoids were identified, including Aphidius colemani (Viereck 1912), Lysiphlebus testaceipes (Cresson 1880), and Diaeretiella rapae (McIntosh 1855). The aphids species identified were Rhopalosiphum padi (Linnaeus 1758) and Sitobion avenae (Fabricius 1775). This study showed that aphid parasitoids are present in wheat crops even when aphid densities are low, and in one farm, occurred before the aphids colonization. These reports can justified the high efficiency of these natural enemies against aphids in wheat fields.  相似文献

The insecticide resistance in Anopheles gambiae mosquitoes has remained the major threat for vector control programs but the fitness effects conferred by these mechanisms are poorly understood. To fill this knowledge gap, the present study aimed at testing the hypothesis that antibiotic oxytetracycline could have an interaction with insecticide resistance genotypes and consequently inhibit the fecundity in An. gambiae. Four strains of An. gambiae: Kisumu (susceptible), KisKdr (kdr (L1014F) resistant), AcerKis (ace-1 (G119S) resistant) and AcerKdrKis (both kdr (L1014F) and ace-1 (G119S) resistant) were used in this study. The different strains were allowed to bloodfeed on a rabbit previously treated with antibiotic oxytetracycline at a concentration of 39·10–5 M. Three days later, ovarian follicles were dissected from individual mosquito ovaries into physiological saline solution (0.9% NaCl) under a stereomicroscope and the eggs were counted. Fecundity was substantially lower in oxytetracycline-exposed KisKdr females when compared to that of the untreated individuals and oxytetracycline-exposed Kisumu females. The exposed AcerKis females displayed an increased fecundity compared to their nontreated counterparts whereas they had reduced fecundity compared to that of oxytetracycline-exposed Kisumu females. There was no substantial difference between the fecundity in the treated and untreated AcerKdrKis females. The oxytetracycline-exposed AcerKdrKis mosquitoes had an increased fecundity compared to that of the exposed Kisumu females. Our data indicate an indirect effect of oxytetracycline in reducing fecundity of An. gambiae mosquitoes carrying kdrR (L1014F) genotype. These findings could be useful for designing new integrated approaches for malaria vector control in endemic countries.  相似文献

Background

Emerging infectious diseases threaten naïve host populations with extinction. Chytridiomycosis, an emerging infectious disease of amphibians, is caused by the pathogenic fungus Batrachochytrium dendrobatidis (Bd) and has been linked to global declines in amphibians.

Results

We monitored the prevalence of Bd for four years in the Northern leopard frog, Rana pipiens, which is critically imperiled in British Columbia (BC), Canada. The prevalence of Bd initially increased and then remained constant over the last three years of the study. Young of the year emerging from breeding ponds in summer were rarely infected with Bd. Some individuals cleared their Bd infections and the return rate between infected and uninfected individuals was not significantly different.

Conclusions

The BC population of R. pipiens appears to have evolved a level of resistance that allows it to co-exist with Bd. However, this small population of R. pipiens remains vulnerable to extinction.

Several studies have considered maize canopy as a homogeneous medium with a random leaf orientation distribution. Recent studies, however, have detected that maize leaf orientation in the horizontal plane (i.e., leaf azimuth distribution) can react filling empty spaces (e.g., intra-row or inter-row) due to plant spatial arrangement. “Rigid” genotypes present a random distribution of leaf azimuth, independent of planting pattern. “Plastic” genotypes have the ability to modify leaf orientation. They generally present a random leaf azimuth distribution under square planting patterns, but tend to align their leaves perpendicularly to the row direction under rectangular planting patterns. We used three-dimensional models to reconstruct canopy architecture, mimicking these two behaviors. We simulated mid-day (maximum sun elevation 79.4° and 65.3°) and daily light interception of fully developed canopies at various plant densities (3, 9 and 12 plants m−2) and row spacings (0.35 and 0.70 m), and compared the results of these simulations with data from a field experiment. Simulations and field measurements showed that canopy behavior (plastic or rigid) has a significant (P<0.05) effect on light interception. Plastic canopies in a rectangular plant spatial distribution were able to increase (6–10% around mid-day), or maintain (during a day) similar light attenuation coefficients (k) to those under square planting patterns. In rigid canopies, on the contrary, there was a reduction in k (10–12%) when grown under rectangular planting patterns, both at mid-day and during a day. Our results suggest that the plasticity of maize canopy, based on leaf re-orientation, results in similar daily light interception values in different inter-row distances.  相似文献

Based on 10 years of trapping with yellow pan traps it was determined that on the average inflights of green peach aphids,Myzus persicae (Sulzer), start July 26 and cumulative catches of more than five aphids August 8. The lack of significant differences in the timing of first and cumulative catches of aphids between the northern and the southern section of the potato production area is additional evidence thatM. persicae does not usually overwinter in New Brunswick. Important yearly variations in the timing of the first cumulative catch of more than 5 aphids can be simulated by the thermal summation of 1188 DD and forecasted by regression on the timing of first inflights. The use of these results to improve the extension program is discussed.  相似文献

Extracellular traps (ETs) released from vertebrate and invertebrate immune cells consist of chromatin and toxic granule contents that are capable of immobilizing and killing microbes. This recently described innate immune response is not well documented in insects. The present study found that ETs were released by hemocytes of Galleria mellonella (Linnaeus) (Lepidoptera: Pyralidae) in vivo and ex vivo after bacterial stimulation. ET release (ETosis), hemolymph coagulation, and melanization likely contributed to the immobilization and killing of the bacteria. The injection of G. mellonella hemocyte deoxyribonucleic acid (DNA) in the presence of bacteria increased bacterial clearance rate and prolonged insect survival. Taken together, these results indicate the presence of insect hemocyte extracellular traps (IHETs) that protect the insect against microbial infection in the hemocoel and represent the first documentation of ETs in insects in vivo.  相似文献

Trapping tests were performed concurrently in several countries of Central and Southern Europe throughout the spring and summer of 2008, to study the selectivity and performance of floral attractant-baited traps developed for catching Epicometis (Tropinota) hirta Poda (CA-baited traps), Cetonia a. aurata L./Potosia (Protaetia) cuprea Fabr. (ME-baited traps) or Oxythyrea funesta Poda (PH-baited traps) (Coleoptera: Scarabaeidae, Cetoniinae). Amongst the species caught, E. hirta showed strongest attraction to the CA-baited and ME-baited traps. O. funesta was mostly caught by PH-baited traps. In capturing C. a. aurata and P. cuprea, the ME-baited trap appeared to be the most efficient. At two sites in Italy, a related scarab, Tropinota squalida Scop. (Coleoptera: Scarabaeidae, Cetoniinae), was attracted in similar numbers to both CA-baited and ME-baited traps. For the scarab Valgus hemipterus L. (Coleoptera: Scarabaeidae, Valginae), ME-baited traps proved to be the most attractive.  相似文献

Six types of sticky card traps differing in color and trapping adhesive were evaluated for monitoring Asian citrus psyllid in citrus in the United States (Florida and Texas). We coded the six traps as ASYellow, GLYellow, RWYellow, ASACP, ASGreen and MGGreen. The ASYellow, GLYellow and RWYellow traps were a bright yellow color to the human eye but the yellow color of these traps varied in intensity. To the human eye, the ASACP traps were a lime-green color, the MGGreen trap was a fluorescent yellow-green color, and the ASGreen trap was a green color. Spectral reflectance measurements were taken to categorize the color (wavelength) spectrum of each trap. Percent reflectance in the green and yellow wavelength regions was similar among the ASYellow, GLYellow, RWYellow and ASACP traps. Three of the six sticky traps (GLYellow, RWYellow and MGGreen) were treated with a traditional gluey adhesive material. The other three traps were treated with No Mess Sticky Card™ adhesive, a hot-melted, pressure-sensitive adhesive material. Trapping studies were conducted in Florida and Texas during April–May comparing four of the traps and during September–October comparing all six traps. Data collected during April–May indicated there were no significant differences in numbers of adults captured on ASGreen, ASYellow, ASACP and GLYellow traps. In seven comparisons of all six traps during September–October, the GLYellow and RWYellow traps each captured significantly fewer adults in one comparison, the MGGreen trap captured fewer adults in two comparisons, and the ASGreen trap captured fewer in three comparisons. There was no evidence that any one of the six traps would be best at detecting psyllids when adult populations are scarce. There was no significant difference between the two adhesive materials with respect to numbers of adults captured on traps.  相似文献

Species diversity, abundance, and dispersion of rice stem borers in framer’s fields were studied in four major rice growing areas of Kahama District. Stem borer larvae were extracted from the damaged tillers in 16 quadrants established in each field. Adult Moths were trapped by light traps and collected in vials for identification. Results indicated the presence of Chilo partellus, Maliarpha separatella, and Sesamia calamistis in all study areas. The most abundant species was C. partellus (48.6%) followed by M. separatella (35.4%) and S. calamistis was least abundant (16.1%). Stem borers dispersion was aggregated along the edges of rice fields in three locations (wards) namely: Bulige, Chela, and Ngaya. The dispersion in the fourth ward, Kashishi was uniform as established from two of the three dispersion indices tested. Further studies would be required to establish the available alternative hosts, the extent of economic losses and the distribution of rice stem borers in the rest of the Lake zone of Tanzania.  相似文献

Potato (Solanum tuberosum L.) cvs. AC Brador and Shepody grownin vitro exhibit plantlets with a short, rosette-type growth habit, and restricts multi plication rates. In contrast, Russet Burbank develops into plantlets with adequate internode lengthin vitro. Cool-white fluorescent lamps provided irradiation for wooden boxes fitted with yellow plexiglass filters. The resultant irradiation lacked wavelengths from 380 to 525 nm. A box fitted with clear plexiglass was the control. Internode length of plantlets was ca.1.0 cm for all three cultivars when grown in boxes with yellow filters, and was 0.5 cm when clear filters were used. The number of nodes, haulm area, stem length, haulm fresh and dry weight of cvs. AC Brador and Shepody were significantly greater in boxes with yellow filters. Potato cvs. AC Belmont, Eramosa, Hunter, Huron, Mirton Pearl, Raritan and York all have a short growth habitin vitro and stem length was increased by the use of a yellow filter. The incidence of intumescences or oedemas on leaves of potato plantletsin vitro was reduced from 60 to seven percent for AC Brador and from 70 to 10% for Shepody with the use of yellow filters. Blue-green light is therefore putatively involved in the occurrence of intumescences on potato tissuesin vitro.  相似文献
