The use of whole grain rye products, beneficial to one's health, could be substantially extended if the typical intensively bitter flavour of rye could be modified without losing the characteristic rye-like flavour. The aim of the study was to evaluate the contribution of non-volatile phenolic compounds on the perceived flavour. Rye grain was milled into five milling fractions. The levels of phenolic compounds, i.e. the phenolic acids, alkylresorcinols and lignans, of the fractions were analysed and related statistically to sensory flavour profiles by partial least-squares (PLS) regression. The non-bound (free) phenolic acids are suggested to be most flavour-active. Cereal and intense flavour and aftertaste were related to vanillic and veratric acids, alkylresorcinol C23:0, and other lignans except for pinoresinol. The perceived bitterness of the bran fractions was suggested to result from pinoresinol and syringic acid. Sinapic and ferulic acids, alkylresorcinols, except for alkylresorcinol C23:0, and syringaresinol seemed to cause the germ-like flavour. Phenolic acids, alkylresorcinols and lignans distributed with a similar pattern in the rye grain as the flavour attributes: the phenolic compounds were clearly located in the outer bran fractions being intense in flavour, but not in the mild-tasting inner layers of the grain.  相似文献

The effects of three independent variables: solvent polarity, temperature and extraction time on the antioxidant capacity, total phenolic content and phenolic acid composition in extracts obtained from sunflower shells before and after enzymatic treatment were studied. Response surface methodology based on three-level, three-variable Box-Behnken design was used for optimization of extraction parameters and evaluation of their effect on antioxidant capacity and total phenolic content in shell extracts.The average antioxidant capacities of extracts from sunflower shells without enzymatic treatment (368.1-1574.4 μmol TE/100 g) were higher than those for cellulolytic and pectolytic enzymes-treated shells (222.7-1419.0 and 270.7-1570.7 μmol TE/100 g, respectively). The content of total phenolic compounds ranged between 58.2-341.2 mg CGA/100 g, 26.7-277.3 mg CGA/100 g and 51.4-301.5 mg CGA/100 g for extracts obtained from shells without enzyme and treated with cellulolytic and pectolytic enzymes, respectively. Total phenolic content (TPC) in the studied shell extracts correlated significantly (p < 0.0001) positively with their antioxidant capacity determined by the ferric reducing antioxidant power (FRAP) method (r = 0.9275). Results of FRAP, TPC and phenolic acid composition in the studied shell extracts depend on the extraction conditions (solvent polarity, temperature, time), but they are independent on the addition of enzyme solutions. The antioxidant capacity and total phenolic content in the resulting extracts increased with a line in extraction temperature and solvent polarity.  相似文献

Most analytical studies on polyphenols in cereals refer to compounds determined in aqueous-organic extracts and alkali hydrolysates, but an appreciable amount of polyphenols bound to cell wall constituents may remain insoluble in the residues of extraction and alkali hydrolysis. The main objective of this work was to determine if sulphuric acid hydrolysis may release significant amounts of polyphenols to be considered for analytical and nutritional studies. HPLC/MS analyses of polyphenols were performed in methanol–acetone extracts, alkali and sulphuric acid hydrolysates of wheat flour, bran and a pool of cereals of the diet. The amount of polyphenols found in the acidic hydrolysates (200–1600 mg/100 g) was higher than in alkali hydrolysates (0.2–372 mg/100 g). Lower amount of polyphenols were found in the methanol–acetone extracts (44–160 mg/100 g). Hydroxybenzoic, caffeic, cinammic, ferulic and protocatechuic acids were the main constituents of the hydrolysates. The contribution of cereals to the intake of dietary polyphenols in Spain was estimated around 360 mg/person/day (65 mg of extractable and 295 mg nonextractable polyphenols).  相似文献

Whole grain wheat and rye are important sources of many bioactive compounds and contribute significantly to the total intake of cereals in many countries. Alkylresorcinols, benzoxazinoids, lignans, phenolic acids, phytosterols and tocols are common bioactive compounds present in these cereals. In this review, we report recent findings (mainly from 2010 onwards) regarding their content, composition, effects of food processing and their uptake, elimination and bioactivities with implications for health.  相似文献

Wheat bread was lyophilised, ground, extracted and centrifuged. The supernatants were analysed for gluten content by RP-HPLC and a commercial sandwich ELISA. Prolamin extraction solvents contained tris(2-carboxyethyl)phosphine (TCEP; 1, 2, 5, 10, 20, 50 mmol/L), guanidine hydrochloride (GUA; 0 or 2 mol/L) and a buffered salt solution. A commercial cocktail solution (250 mmol/L mercaptoethanol (ME), 2 mol/L GUA, buffered salt solution) as well as 60% (v/v) ethanol were used as control solvents. Wheat flour was the control for the extractability of the native prolamins. 60% ethanol only extracted 37% of the prolamins from wheat bread (cocktail = 100%). When ME was replaced by TCEP the protein yield increased from 35% at the lowest TCEP-level to 95% when 20–50 mmol/L TCEP were used. The use of GUA was essential to extract prolamins quantitatively. Comparative protein analysis using RP-HPLC and ELISA showed that both methods provided comparable prolamin (gliadin) concentrations of the wheat flour (40.3–45.7 mg/g), when 60% ethanol was used as extraction solvent. The extraction yields from bread were considerably lower (16.7–24.7 mg/g). Cocktail and TCEP extracted almost the same amount of protein from flour and bread with TCEP showing slightly lower yields. Total extractable protein (gliadin + glutenin) as determined by RP-HPLC was 70.5–75.3 mg/g, and total gliadin as determined by ELISA was 42.7–44.2 mg/g. Thus, the study has shown that TCEP in combination with GUA extracts proteins from heated, gluten-containing foods as effective as the commercial cocktail solution.  相似文献

In order to investigate the effect of trifluralin, pronamide, haloxyfop-p methyl, propaquizafop and isoxaben on weed control and oilseed rape yield, a two year field experiment was conducted at Darab, Iran, during 2004–2005 and 2005–2006 growing seasons. Herbicides reduced weed biomass compared with the weedy check. In 2004–2005, at 16 WAP (weeks after planting), application of trifluralin plus propaquizafop plus isoxaben at 1200 + 200 + 250 g a.i./ha and trifluralin plus haloxyfop-p methyl plus isoxaben at 1200 + 100 + 500 g a.i./ha provided best control (80%) of wild mustard (Sinapis arvensis L.), compared to other treatments. At 8 and 16 WAP, wild mustard control decreased to 47 and 42%, respectively, when trifluralin was applied alone at 1400 g a.i./ha. In 2005–2006 at 16 WAP, maximum reduction in wild mustard biomass (82%) was achieved with trifluralin plus propaquizafop plus isoxaben at 1200 + 200 + 250 g a.i./ha. In both years, all herbicide treatments controlled fumitory (Fumaria officinalis L.) 64–96%. In 2004–2005, at 16 WAP, wild oat (Avena fatua L.) biomass reduction by trifluralin plus propaquizafop plus isoxaben at 1200 + 200 + 250 g a.i./ha was 95%. In 2005–2006, at 8 and 16 WAP, minimum biomass reduction of wild oat (37–53%) was observed with trifluralin at 1200 and 1400 g a.i./ha. In both years, at 8 and 16 WAP, the most effective herbicide treatments were trifluralin plus propaquizafop plus isoxaben and trifluralin plus haloxyfop-p methyl plus isoxaben regardless of the dose applied and provided maximum reduction in total weed biomass (77–89%) compared to other treatments. Results of both years showed that all herbicide treatments increased oilseed rape grain yield as compared with the weedy check but percentage of oil was not affected by various herbicide treatments. Maximum grain yield was obtained with trifluralin plus haloxyfop-p methyl plus isoxaben at 1200 + 100 + 500 g a.i./ha.  相似文献

Two varieties (Centenario and Oscar Blanco) of Andean native grain, kiwicha (Amaranthus caudatus), were evaluated as sources of dietary fiber and of some bioactive compounds. The impact of low-cost extrusion on the content of these components was studied for technological applications. The content of total dietary fiber in Centenario was higher (16.4%) than in Oscar Blanco (13.8%). In both varieties, the content of total and insoluble dietary fiber decreased during the extrusion process. In Centenario, the content of soluble dietary fiber increased, from 2.5 to 3.1% during the extrusion process. The content of phytic acid in raw kiwicha was 0.3% for both varieties, and the content of total phenolic compounds was 98.7 and 112.9 mg GAE/100 g of sample, for Centenario and Oscar Blanco, respectively.  相似文献

The presence of off-odors, such as “musty” or “sour,” in grain sorghum decreases the value of the product. Sensory analysis and head space-solid phase micro extraction-gas chromatography-mass spectrometry were conducted on raw sorghum to identify chemicals that would be suitable as sensory references for musty odors in this grain. Two groups of musty grain sorghum notes were differentiated: “dry” and “wet” musty sorghum. Musty sorghum with “wet” odor notes had other off-odor notes described as earthy/damp or moldy. The compounds found in musty sorghum grain were studied as “musty” note standards to use for the development of sensory references. Seven potential compounds were chosen, and a shelf life study was completed on the sensory references. Geosmin (0.1 mg/kg), 1,2-dimethoxybenzene (100 mg/kg), 3-octanone (100 mg/kg), and 1,2,4-trimethoxybenzene (100 mg/kg) were identified as potential candidates to help reproduce musty odor notes when added to clean fresh sorghum grain.  相似文献

This study aimed to evaluate the effect of autohydrolysis temperature of corn residues in the antioxidant activity of the phenolic compounds extracted from the liquid phase. The treatments were carried out at 160, 180, 190 and 200 °C for 30 min in a pressurized batch reactor. Two different methods for phenolic compounds extraction from the autohydrolysis liquors were investigated. For that purpose, solvent extraction with ethyl acetate and acidic precipitation were performed for phenolic compounds recovery. These methods have been compared in terms of extraction yield, physicochemical properties of obtained polyphenols (characterization by Fourier Transform Infrared Spectroscopy, Thermogravimetric Analysis and Gel Permeation Chromatography), total phenolic content, total antioxidant capacity and Trolox equivalent antioxidant capacity (TEAC) values, measured in DPPH (2,2-diphenyl-1-picrylhydrazyl) test system. The maximum phenolic contents ranged from 6.04 mg GAE/100 mg extract in acidic precipitated samples to 16.45 mg GAE/100 mg extract in ethyl acetate soluble fractions. The results indicated that the ethyl acetate fractions possessed the highest antioxidant activity, reaching after 30-60 min the same capacity reported for the reference synthetic antioxidants (Trolox).  相似文献

Cereal based products intended for gluten sensitive individuals, particularly to celiac disease patients, tend to have poor organoleptic qualities and they contain low levels of healthy whole grain compounds. Adding whole grain ingredients, such as malt hydrolysates, could compensate these defects provided that the ingredients are adequately free from toxic prolamin epitopes. Here we demonstrate that the level of toxic prolamin epitopes in the malt autolysates (wheat, barley, rye) were substantially lower than in the native malts but too high to allow “very low in gluten” labelling. To further eliminate the residual levels of toxic prolamin epitopes, a proline-specific endoprotease from Aspergillus niger was added to the malt autolysates. In the resulting malt hydrolysates (of wheat and rye but not barley), the prolamins were indeed greatly reduced and were below the very low gluten limit of 100 mg/kg. Malt hydrolysates with adequately low gluten levels may potentially be used as novel ingredients within gluten-free foods.  相似文献

The demand for food is increasing, not only to meet food security for growing populations, but also to provide more nutritious food, rich in good quality proteins and nutraceutical compounds. The amaranth (Amaranthus hypochondriacus) plant, in addition to its high nutritive and nutraceutical characteristics, has excellent agronomic features. The objective of the present study was to analyze some physical and proximal-nutritional properties of amaranth seeds obtained from different varieties grown in arid zones and characterize their phenolic acids and flavonoids. Two commercial (Tulyehualco and Nutrisol) and two new (DGETA and Gabriela) varieties of A. hypochondriacus were grown at the Mexican Highlands zone. Tulyehualco and DGETA varieties had higher seed yield of 1475 and 1422 kg ha−1, respectively, comparable to corn and soybean production in agricultural areas. Gabriela had the highest protein content of 17.3%, but all varieties had an adequate balance of essential amino acids. Polyphenols as rutin (4.0–10.2 μg g−1 flour) and nicotiflorin (7.2–4.8 μg g−1 flour) were detected. Amaranth can be cultivated in arid zones where commercial crops cannot be grown; the seeds besides their well known nutritive characteristics could be a source of phenolic compounds of high antioxidant properties.  相似文献

Mineral deficiencies are prevalent in human populations and the improvement of the mineral content in cereal products represents a possible strategy to increase the human mineral intake. Nevertheless, most of the inorganic phosphorus (Pi) present in mature cereal seeds (40–80%) is stored as phytate, an anti-nutritional factor that forms complexes with minerals such as Ca, Mg, Zn and Fe reducing their bioavailability. The present study was undertaken: (i) to determine the variation in phytate and mineral concentrations in the whole grains of 84 Italian durum wheat (Triticum durum Desf.) cultivars representative of old and modern germplasm; (ii) to estimate the magnitude of genotype × environment interaction effects; and (iii) to examine the interrelationships among mineral concentrations in durum wheat with the final aim to identify superior durum wheat cultivars that possess low phytate content and high concentration of mineral elements in their whole-wheat flour. The cultivars were grown in field trials during 2004–2005 at Foggia, Italy and during 2005–2006 at Foggia and Fiorenzuola d’Arda—Southern and Northern Italy. The phytate content was estimated indirectly by using a microtitre plate assay evaluating the Pi absorbance at 820 nm, while the Cu, Fe, Mn, Ca, K, Mg, Na and Zn mineral contents were determined by ICP/OES. The contents of Zn and Fe across years and locations ranged from 28.5 to 46.3 mg/kg for Zn with an average of 37.4 mg/kg and from 33.6 to 65.6 mg/kg for Fe with an average of 49.6 mg/kg. Pi grain content was between 0.46 and 0.76 mg/g showing a positive correlation with all minerals except Cu and Zn. Although breeding activity for Fe and Zn would be difficult because G × E interaction is prevalent, multi-location evaluation of germplasm collection help to identify superior genotypes to achieve this objective. The results here reported open the possibility of designing a specific breeding program for improving the nutritional value of durum wheat through the identification of parental lines with low-Pi and high minerals concentration in whole grains.  相似文献

Processing is important both for the sensory and nutritional quality of grains. In the case of whole grains, it especially may influence the levels and bioavailability of the bioactive compounds. Yeast, lactic acid and mixed fermentation were shown to vary in their effects on native and germinated wholemeal rye flour. The presence of Saccharomyces cerevisiae was important for enhanced levels of the analysed compounds. The levels of folates, free phenolic acids, total phenolic compounds, lignans and alkylresorcinols were increased especially by fermentation of germinated rye, which also resulted in lower pH when compared with native rye flour, the level of folate was increased up to seven-fold and that of free phenolic acids by up to ten-fold after germination and fermentation. Tailored fermentation thus offers a tool to further increase the bioactive potential of wholemeal rye.  相似文献

Non-phosphorylated thiamin (T), thiamin monophosphate (TMP), and thiamin diphosphate (TDP) were extracted from whole grains of wheat (Triticum aestivum, cv. Dekan), triticale (Triticosecale, cv. SW Talentro), rye (Secale cereale, cv. Agronom), oats (Avena sativa, cv. Dominik), and barley (Hordeum vulgare, cv. Merlot) at different growth stages (from inflorescence emergence to full ripeness of the grains) and determined with high-performance liquid chromatography and fluorescence detection. No thiamin triphosphate or adenosine thiamin triphosphate was found in these cereals. Total thiamin levels (the sum of T and its phosphate esters) determined on a dry matter (DM) basis were found to be quite similar in all grain samples throughout the analysis period, ranging from 5.59 (wheat) to 13.00 nmol/g DM (oats). In spikelets 3–5 days after flowering, the total thiamin content in all cereals was 40–75% T, 5–13% TMP, and 18–46% TDP. Generally, T increased and TDP decreased continuously with grain development, with maximum T (88–99%), and minimum TDP (2–10%) at ripeness. No TMP was detected in mature grains. The results indicate that variation in T, TMP, and TDP levels is probably due above all to the conversion of phosphorylated thiamin phosphate esters into less highly or non-phosphorylated forms.  相似文献

Weedy rice, specifically red rice (Oryza sativa L.), is a major weed in rice which causes up to 80% yield loss and reduction of grain quality. Red rice accessions from Arkansas, U.S.A., were characterized to classify red rice accessions into certain phenotypic groups relevant to weedy rice management. The red rice accessions were 70% strawhull, 22% blackhull, 7% brownhull and <1% goldhull. Generally, blackhull red rice was the tallest (139 cm) and strawhull the shortest (133 cm) among all accessions. Blackhull red rice had more tillers (102/plant), smaller flag leaves (13 cm wide, 34 cm long), and flowered later (1225 heat units) than strawhull red rice which had 85 tillers/plant, 15 cm-wide and 34 cm-long flag leaves, and flowered after accumulating 1195 heat units. Morphological differences between accessions within each hull type were highly significant, showing great diversity within a hull color group as indicated by large ranges in traits. For example, blackhulls were 75–190 cm tall with 18–69 cm long flag leaves, 21–188 tillers and produced 40–949 g seed. Strawhulls were 46–189 cm tall with 18–66 cm flag leaf length, 16–172 tillers and produced 100–608 g seed. Some traits, such as seed production, differed widely between accessions within each hull color group such that the average seed production/accession for blackhull did not differ from that of strawhull weedy rice (196 vs. 192 g/plant). The onset of flowering among all accessions ranged from 56 to 126 d after planting. Red rice accessions formed six phenotypic clusters generally segregated by plant size or flowering time. Each morphotype would have different competitive abilities; thus, weedy rice management could be geared toward plant types. Highly competitive plant types would require intensive control measures to minimize yield losses and reduce the soil seed bank. Flowering dates impact stewardship strategies for herbicide-resistant or any genetically modified rice.  相似文献

High-performance liquid chromatography (HPLC) was used to determine the distribution of phenolic acids in wheat flours produced from five milling extraction rates ranging from 60% to 100% in four cultivars sown in two locations in the 2008–2009 season. Considerable variation was observed in free and bound phenolic acids, and their components in flours with different extraction rates. Most phenolic acids, including the component ferulic, were present in the bound form (94.0%). Ferulic (51.0%) was the predominant phenolic acid in wheat grain, and caffeic (22.8%) and p-coumaric (17.6%) acids were abundant. The phenolic acids and their components were all significantly influenced by effects of cultivar, milling, location, and cultivar × milling interaction, with milling effect being the predominant. The proportions of phenolic compounds varied considerably among milling extractions and cultivars, and their levels depended on both initial grain concentrations and on selection of milling extraction that was incorporated into the final product. The grain phenolic acid concentrations determined ranged from 54 μg g−1 in flour produced at 60% extraction rate to 695 μg g−1 in flour produced at 100% extraction rate, indicating their higher concentrations in bran associated with cell wall materials. Therefore, wholemeal wheat products maximize health benefits and are strongly recommended for use in food processing.  相似文献

Caulerpa racemosa (sea grapes) and Ulva lactuca (sea lettuces) are edible green seaweeds and good sources of bioactive compounds for future foods, nutraceuticals and cosmeceutical industries. In the present study, we determined nutritional values and investigated the recovery of bioactive compounds from C. racemosa and U. lactuca using hot water extraction (HWE) and subcritical water extraction (SWE) at different extraction temperatures (110 to 230 °C). Besides significantly higher extraction yield, SWE processes also give higher protein, sugar, total phenolic (TPC), saponin (TSC), flavonoid contents (TFC) and antioxidant activities as compared to the conventional HWE process. When SWE process was applied, the highest TPC, TSC and TFC values were obtained from U. lactuca hydrolyzed at reaction temperature 230 °C with the value of 39.82 ± 0.32 GAE mg/g, 13.22 ± 0.33 DE mg/g and 6.5 ± 0.47 QE mg/g, respectively. In addition, it also showed the highest antioxidant activity with values of 5.45 ± 0.11 ascorbic acid equivalents (AAE) mg/g and 8.03 ± 0.06 trolox equivalents (TE) mg/g for ABTS and total antioxidant, respectively. The highest phenolic acids in U. lactuca were gallic acid and vanillic acid. Cytotoxic assays demonstrated that C. racemosa and U. lactuca hydrolysates obtained by HWE and SWE did not show any toxic effect on RAW 264.7 cells at tested concentrations after 24 h and 48 h of treatment (p < 0.05), suggesting that both hydrolysates were safe and non-toxic for application in foods, cosmeceuticals and nutraceuticals products. In addition, the results of this study demonstrated the potential of SWE for the production of high-quality seaweed hydrolysates. Collectively, this study shows the potential of under-exploited tropical green seaweed resources as potential antioxidants in nutraceutical and cosmeceutical products.  相似文献

Phytosterols and steryl ferulates are bioactive compounds accumulating in the bran and germ of wheat. However, little is known regarding their localisation and composition in the bran layers of the kernel. The aim of this study was to determine the distribution of phytosterols and steryl ferulates in the wheat grain and in the different layers of bran. The wheat fractions, produced by conventional debranning, aleurone separation and a novel electrostatic process, were analysed for phytosterol contents using GC–FID and for steryl ferulate contents using HPLC–UV. The compounds were identified by GC– and LC–MS. Phytosterols and steryl ferulates were concentrated in the bran layers. The steryl ferulates were accumulated in the intermediate layers, whereas the phytosterols were more evenly distributed in the intermediate layers and aleurone cell contents. The phytosterol composition varied within the wheat kernel, while the steryl ferulate composition was similar in different fractions. Sitosterol and campestanyl ferulate were the main compounds. The highest levels of phytosterols (up to 2117 μg/g) and steryl ferulates (up to 703 μg/g) were found in the pearling, aleurone and certain bran fractions. The phytosterol-rich fractions could be utilised in cereal foods to enhance the intake of health-promoting compounds from natural sources.  相似文献

In recent years the strawberry tree (Arbutus unedo L.) is being gradually replaced by other species with higher economic value. With the ultimate goal of selecting superior genotypes, the present work was initiated to study the antioxidant and antimicrobial activities, and total phenolic content in 19 different genotypes of A. unedo leaves from the Trás-os-Montes region of Portugal.The genotype Bragança 1 contains higher total phenolic content (215.0 mg GAE/gextract) whereas the Vila Boa 4 genotype shows lower total phenolic content (148.0 mg GAE/gextract). In both methods tested to evaluate the antioxidant activity, Vila Verde and Donai displayed the highest antioxidant capacity (EC50 values of 0.088 and 0.090 mg/mL, respectively, for DPPH; EC50 values of 0.233 and 0.245 mg/mL, respectively, for reducing power assay) while Vila Boa 2 reported the lowest antioxidant potential (EC50 values of 0.142 and 0.378 mg/mL, respectively, in DPPH and reducing power methods). Linear negative correlations were established between the total phenol contents and the EC50 values for both of the antioxidant activity methods tested. Preliminary assays for antimicrobial potential showed that extracts from A. unedo leaves display antibacterial activity, with MIC values of 1 and 5 mg/mL for some Gram-positive and Gram-negative bacteria, respectively. Taken together, the results suggest that A. unedo leaves are a potential source of natural compounds with valuable bioactive properties that could be explored by the pharmaceutical, chemical and food industries.  相似文献

Eight lupin (Lupinus angustifolius L.) genotypes grown at four locations in south central Alberta in 2004 were evaluated for variability in phenolic constituents and antioxidant activity measured by a photochemiluminescence assay. Genotype was the main source of variation for content of phenolic compounds and antioxidant activities. Phenolic compounds in genotypes varied minimally from 11.9 to 14.7 mg catechin equivalent and 4.15 to 4.95 mg rutin equivalent g−1 lupin for total phenolic and flavonoid contents, respectively. Lupin genotypes exhibited weak antioxidant activity based on water–soluble substances (ACW) of 0.54 to 1.07 μmole Trolox equivalent antioxidant capacities (TEAC)/g with lag time ranging from 70 to 153 s and an antioxidant index of 6.7 to 14.5 and 1.9 to 3.3 μmole TEAC/g based on measurements of lipid-soluble substances (ACL). Antioxidant activity of lupin genotypes was not related to phenolic contents of seeds.  相似文献
