The anthelmintic efficacy of 6 non-benzimidazole compounds and thiabendazole against standardised, benzimidazole resistant strains of H. contortus and T. colubriformis in sheep was determined using a controlled test. All compounds were administered intraruminally at their recommended therapeutic dose rates and 4 of the compounds were also assessed at half their recommended therapeutic dose rates. Levimisole at a dose of 6.4 mg/kg body weight, morantel at 4.4 mg/kg and 8.8 mg/kg, naphthalophos at 6.25 mg/kg and 12.5 mg/kg, rafoxanide at 7.5 mg/kg and phenothiazine at 530 mg/kg were 98% or more effective in removing adult infections of benzimidazole resistant H. contortus. Rafoxanide at 3.75 mg/kg, phenothiazine at 265 mg/kg and carbon tetrachloride at 0.05ml/kg were less effective, removing 80%, 73% and 72% respectively of the benzimidazole resistant H. contortus worm burdens. Against the benzimidazole resistant T. colubriformis levamisole at 6.4 mg/kg, and morantel at 4.4 mg/kg and 8.8 mg/kg removed 99%, 68% and 86% respectively of the adult infections. All other anthelmintics had little or no efficacy at the dose rates tested. Thiabendazole at 44 mg/kg had no significant effect against these strains of H. contortus and T. colubriformis. Several non-benzimidazole anthelmintics have shown high efficacy in this experiment and should be suitable for treating infections with benzimidazole resistant H. contortus. Of the anthelmintics tested, only levamisole and morantel show high efficacy against benzimidazole resistant T. colubriformis.  相似文献

Morantel could not be detected (<0.05 pg/ml)in the plasma of cattle or goats followingthe oral administration of morantel tartrate at a dose rate of 10 mg/kg
bodyweight. No morantel was detected in the milk of lactating goats except in
one animal where a concentration of 0.092 pg/ml was detected at 8 h after drug
administration. Morantel was highly effective against Cooperia oncophora infec-
tions in calves treated 6, 9 or 18 days after infection; however, was highly
effective against Ostertagia ostertagi only when treated 18 days after infection.
Morantel did not affect the fecundity of adult 0.ostertagi surviving treatment 18
days after infection which had similar average numbers of eggs in their uteri
(range 13.4 f 0.73-16.8 L 0.98) as did parasites from control animals (range
12.0 k 0.70-13.6 2 0.66). Morantel could be detected at a concentration of 96 k
4.5 pgig (dry weight) in the faeces of a calf 24 h after treatment with I0 mgikg
bodyweight of morantel tartrate. The concentration of morantel in replicate
samples of this faeces exposed to natural atmosphere, but not to soil or soil
organisms, declined slowly over the following 322 days. At day 322 after the start
of the experiment 8.8 pg/g of morantel could be measured in the remaining
faecal material. Throughout the faecal degradation study the concentration of
morantel in the crusts of the replicate sample pats was lower than the

Efficacy of ivermectin in a topical formulation was evaluated in cattle against adult gastrointestinal and pulmonary nematode infections (experiment 1, n = 24), the 2nd- and 3rd-stage larvae of Hypoderma spp (experiment 2, n = 12), and the biting louse Damalinia bovis (experiment 3, n = 12). Nematode infections were induced and grubs and lice were naturally acquired. Treatments consisted of a single dose of ivermectin in a topical formulation of 200, 500, or 1,000 micrograms/kg of body weight in experiment 1 or 500 micrograms/kg in experiments 2 and 3. At 1,000 micrograms/kg, ivermectin was 100% effective against Ostertagia ostertagi, Trichostrongylus colubriformis, Oesophagostomum radiatum, Nematodirus helvetianus, Haemonchus placei, and Dictyocaulus viviparus and was greater than 99% effective against Cooperia oncophora, C punctata, and T axei. At 500 micrograms/kg, the efficacy was 100% against C oncophora, C punctata, O ostertagi, T axei, Oes radiatum, N helvetianus, Haem placei, and Dict viviparus and greater than 99% against T colubriformis. At 200 micrograms/kg, the efficacy was 100% against Oes radiatum, Haem placei, and Dict viviparus, greater than 99% for O ostertagi, 96% for C oncophora, 86% for C punctata, 90% for T axei, 85% for T colubriformis, and 71% for N helvetianus. At 500 micrograms/kg, ivermectin was highly effective against the grubs Hypoderma bovis and H lineatum and eliminated the louse Damalinia bovis.  相似文献

Ivermectin delivered continuously from a rumino-reticular sustained release device was prophylactically effective in preventing establishment of nine nematode parasite species in multiply-exposed cattle. Ivermectin dosages which permitted less than or equal to 1% of infected control calf worm populations to establish ranged from less than 2.5 micrograms kg-1 day-1, which totally prevented infection with Dictyocaulus viviparus and Oesophagostomum radiatum, to approximately equal to 30 micrograms kg-1 day-1 required to suppress Nematodirus helvetianus to the same extent. Between these extremes, in decreasing order of sensitivity to enterical sustained release ivermectin, were Ostertagia ostertagi, Trichostrongylus axei, Haemonchus placei, Cooperia punctata, C. oncophora and T. colubriformis which were maximally affected at less than or equal to 10 micrograms kg-1 day-1 of ivermectin.  相似文献

OBJECTIVE: To evaluate efficacy of topically administered doramectin against eyeworms, lungworms, and gastrointestinal nematodes of cattle. ANIMALS: 400 cattle (20 cattle in each of 20 trials). PROCEDURE: Trials were conducted in North America; natural and experimentally induced infections were used. In each trial, cattle were allocated randomly to control (placebo [saline [0.9% NaCl] solution at 1 ml/10 kg of body weight] or untreated; n = 10) or doramectin-treated (500 microg/kg of body weight; 10) groups. Treatments were applied in a single passage along the midline of the back, from the withers to the tailhead. Cattle were euthanatized > or =14 days after treatment, and worm burdens were determined by use of standard techniques. RESULTS: Efficacy of doramectin was > or =95.3% against adults of Thelazia gulosa, T skrjabini, Dictyocaulus viviparus, Haemonchus contortus, H placei, Ostertagia lyrata, O ostertagi, Trichostrongylus axei, Bunostomum phlebotomum, Capillaria spp, Cooperia oncophora, C pectinata, C punctata, C spatulata, C surnabada, Nematodirus spathiger, Strongyloides papillosus, T colubriformis, Oesophagostomum radiatum, and Trichuris spp. Efficacy was 95.1% against fourth-stage larvae of D viviparus, H placei, O lyrata, O ostertagi, T axei, C oncophora, C punctata, C spatulata, C surnabada, N helvetianus, T colubriformis, O radiatum, and Trichuris spp. In addition, efficacy against inhibited fourth-stage larvae of O ostertagi and Ostertagia spp was > or =98.1%. CONCLUSIONS AND CLINICAL RELEVANCE: A single topical application of doramectin pour-on was efficacious against a broad range of nematode species in cattle.  相似文献

A controlled trial was conducted to evaluate the efficacy of the intraruminal ivermectin controlled-release capsule (CRC) (IVOMEC Maximizer CR Capsule for Sheep, Merial Ltd.) against induced incoming third-stage larvae and established adult infections with some rarer gastrointestinal nematode parasites of sheep. Twenty-one worm-free lambs were allocated by restricted randomisation based on body weight within sex to one of the following treatments: unmedicated control, ivermectin CRC given on Day 0 prior to induced infection, and ivermectin CRC given on Day 70 after establishment of induced infection. The ivermectin CRC delivers ivermectin at a minimum dose rate of 20 microg/kg/day for 100 days. Infections were induced by daily administration of third-stage larvae for five consecutive days. Nematodes were counted on Day 84, 14 days after treatment of established infection. The treatment with the ivermectin CRC prevented the establishment of Ostertagia leptospicularis, O. ostertagi, Bunostomum trigonocephalum, Cooperia oncophora, C. punctata, C. surnabada, Nematodirus helvetianus, N. roscidus and Strongyloides papillosus by >99% as compared with the untreated controls (p < 0.01). The administration of the ivermectin CRC reduced established adult infections of O. ostertagi, B. trigonocephalum, C. oncophora, C. punctata, C. surnabada, N. roscidus and S. papillosus by >99% (p < 0.01), and reduced established adult infections of O. leptospicularis and N. helvetianus by 96.5 and 98.4% (p < 0.01), respectively.  相似文献

Paraherquamide, an oxindole alkaloid metabolite of Penicillium paraherquei and Penicillium charlesii, was tested against the adult stages of nine common gastrointestinal and lung nematodes of calves at single, oral dosages of 0.5, 1.0, 2.0 or 4.0 mg kg-1. At dosages 1.0-4.0 mg kg-1 there was 95% or more removal of Haemonchus placei, Ostertagia ostertagi, Trichostrongylus axei, Trichostrongylus colubriformis, Cooperia oncophora, Nematodirus helvetianus, Oesophagostomum radiatum, and Dictyocaulus viviparus. Cooperia punctata, the dosage-limiting species, was virtually unaffected by any dosage except the highest, which produced an efficacy of 89%. The 0.5 mg kg-1 dosage was 95% or more efficacious against H. placei, O. ostertagi, C. oncophora, and D. viviparus, but weaknesses were evident against the other five species. No adverse reaction was observe in any calf.  相似文献

Persistent anthelmintic activity of ivermectin in cattle   总被引:1,自引:0,他引:1  
Two studies are described which demonstrate the persistent activity of ivermectin injected subcutaneously into cattle at 200 micrograms/kg in preventing the establishment of induced infections with the gastrointestinal parasites Ostertagia ostertagi and Cooperia oncophora and the lungworm Dictyocaulus viviparus. These results indicated a reduction in mean worm count compared with the control group for O ostertagi of more than 99, 45 and 94 per cent with a seven, 14 or 21 day interval between treatment with ivermectin and the administration of infective larvae, respectively, in trial 1 and more than 99, more than 99 and 99 per cent at seven, 10 or 14 days, respectively, in trial 2. Corresponding values against C oncophora were 99, 0 and 45 per cent at seven, 14 and 21 days in trial 1 and more than 99, 84 and 31 per cent at seven, 10 and 14 days in trial 2. Against D viviparus, reduction in counts were more than 99, 98 and more than 99 per cent at seven, 14 and 21 days, respectively, in trial 1 and 100, 100 and 100 per cent at seven, 10 and 14 days, respectively, in trial 2. The relevance of these results to the build-up of infective larvae on pasture and infection in cattle is discussed.  相似文献

Two formulations of avermectin B1a, C-076 and MK-933, were examined for anthelmintic activity in two trials, each using 20 calves with experimentally produced nematode infections. In one trial, C-076 was given orally, and in the other trial, MK-933 was given by injection at dosages of 50, 100, and 200 micrograms/kg of body weight. Treated calves were held for 7 to 8 days and then, together with nontreated controls, were euthanatized and were examined for nematodes in their gastrointestinal tracts and lungs. Efficacy of C-076 approached 100% for Dictyocaulus viviparus, Ostertagia ostertagi, Trichostrongylus axei, T colubriformis, and Oesophagostomum radiatum at 50 micrograms/kg. A dosage level of 100 micrograms/kg was required to achieve 97% efficacy against Cooperia punctata. Efficacy of MK-933 at 50 micrograms/kg was variable, but at 100 micrograms/kg, it approached 100% for D viviparus, O ostertagi, Haemonchus placei, T axei, and T longispicularis. Activity against C oncophora was less, reaching 80% efficacy at 200 micrograms/kg.  相似文献

Fenbendazole (methyl-5-(phenylthio)-2-benzimidazole carbamate) at dose rates of 5 mg/kg and above was 100 per cent effective in eliminating a naturally acquired Dictyocaulus filaria infection in sheep. The drug was 100 per cent effective in eliminating concurrent infections of adult Trichostrongylus axei, Haemonchus contortus, Haemonchus placei, Ostertagia circumcincta, Ostertagia ostertagii, Cooperia oncophora, Cooperia mcmasterii, Nematodirus spathiger, Neumatodirus filcollis, Oesophagostomum venulosum and Chabertia ovina. Fenbendazole was 93 per cent and 97 per cent effective at doses of 5 and 10 mg/kg respectively in removing infection with adult T colubriformis, and post-treatment worm-egg production was completely suppressed in surviving female worms. No adverse side-effects were observed in treated sheep at either of the two dose rates used.  相似文献

An experiment was conducted in calves to investigate the efficacy of a morantel sustained release trilaminate bolus (MSRT) to control gastrointestinal parasitism and to assess the development of immunity during the use of MSRT. Two groups (M and U) of four calves each were infected three times a week with a mixed Ostertagia ostertagi and Cooperia oncophora infection for 12 weeks. Calves of Group M received an MSRT at the start of the experiment. Twenty weeks after the start of the experiment, all animals, including a previously uninfected control group (C), received a challenge with 100,000 Ostertagia and 100,000 Cooperia. After a further 4 weeks all calves were necropsied for worm counts. During the trial calves were weighed and faecal egg counts, larval differentiation and pepsinogen concentrations were determined. The results demonstrated the high level of efficacy of the MSRT in reducing the faecal egg output and preventing parasitic gastroenteritis under conditions of a continuous high rate of infection. Efficacy of treatment was higher for Cooperia than for Ostertagia. Post-mortem worm counts suggested a partially impaired immunity build-up in Group M, at least for Cooperia.  相似文献

Observations of erratic anthelmintic activity of fenbendazole against known standardised thiabendazole-resistant strains of Haemonchus contortus and Trichostrongylus colubriformis in sheep were investigated. Fenbendazole at a dose rate of 10 mg/kg body weight was administered by oral, intra-ruminal or intra-abomasal routes, and was most effective against both resistant strains following intra-ruminal administration. In addition thiabendazole, oxibendazole, fenbendazole, parbendazole and mebendazole plus two unrelated compounds, levamisole and morantel tartrate, were used at one and a half times their suggested or recommended therapeutic dose rate against thiabendazole-resistant strains of H contortus and T colubriformis in sheep; each drug being administered by the intra-ruminal or intra-abomasal routes. Fenbendazole was more effective against both strains following intra-ruminal administration. Parbendazole was more effective against the resistant strain of T colubriformis following intra-ruminal administration. At the dose rate chosen for the other benzimidazoles used against these resistant strains, there was no difference in anthelmintic efficacy due to route of administration. Levamisole was highly effective against both resistant strains, irrespective of the route of administration. In the groups treated with morantel tartrate, the results obtained were difficult to interpret due to mortalities and a highly variable response in the surviving sheep. Fenbendazole, thiabendazole and mebendazole when used at their suggested or recommended therapeutic dose rate in sheep, were highly effective against known thiabendazole-susceptible strains of H contortus and T colubriformis following both intra-ruminal or intra-abomasal administration.  相似文献

Twelve Holstein calves were used to determine the prophylactic efficacy of ivermectin against challenge exposure with gastrointestinal and pulmonary nematodes. Two groups of 6 calves (mean body weight, 205 kg) each were formed by restricted randomization according to body weight. Group-1 calves served as nonmedicated controls. Each calf of group 2 was orally given one prototype sustained-release bolus designed to deliver ivermectin at a continuous daily dose of 8 mg. Third-stage nematode infective larvae were given to the calves on posttreatment days 28 and 42. The calves were euthanatized 77 or 78 days after treatment. Ivermectin was 100% effective (P less than 0.05) in preventing the establishment of infection by Haemonchus placei, Ostertagia ostertagi, Cooperia spp (C punctata, C oncophora, C surnabada), Nematodirus helvetianus, Oesophagostomum radiatum, and Dictyocaulus viviparus and was greater than 99% effective against Trichostrongylus axei. Prophylactic (summer) and/or therapeutic (autumn) administrations of an intra-ruminal sustained-release bolus (MSRB) containing morantel tartrate were compared for the control of gastro-intestinal nematodes in grazing dairy calves over an 8-month period (December to August). The MSRB was designed to release morantel tartrate at 150 mg/day for 90 days.

Treated and untreated groups of calves grazed separate replicated pastures. Both groups of calves receiving the MSRB at the beginning of summer showed a significant mean liveweight advantage (17.7 kg and 20.7 kg for the summer and summer/autumn treated groups, respectively) at the end of the trial period, when compared with untreated calves. The liveweight advantage of calves receiving the MSRB in autumn only (6.5 kg) was not significantly different from that of the controls.

Both the summer and autumn administrations of the MSRB prevented the accumulation of Pre-Type II burdens of Ostertagia spp.

Retention rates of the MSRB were high and chemical analysis of faecal samples indicated a fairly constant release of the anthelmintic over the predicted 90-day period.

The results indicate that the MSRB is potentially useful for the control of gastro-intestinal nematodes, and particularly for the production of ’safe’ pasture. The MSRB could provide an effective alternative to integrated control based primarily on grazing management.  相似文献

In the course of studies on the control of bovine parasitic gastroenteritis in south west Scotland using a morantel sustained release bolus the populations of trichostrongyle infective larvae on herbage were monitored over a two-year period. From the results of the herbage analysis and the worm burdens in parasite naive tracer calves introduced at the end of 18 months, it was clear that considerable numbers of infective larvae of Ostertagia ostertagi and Cooperia oncophora survived for at least 18 months on pastures not grazed at all. The results suggest that in south west Scotland after one, or even two, seasons with no further contamination of grazing, permanent cattle pastures cannot be assumed to be helminthologically 'safe' and that this should be taken into consideration when determining prophylactic strategies.  相似文献

Twelve calves were experimentally inoculated with 6 species of gastrointestinal nematodes and the liver fluke, Fasciola hepatica. Six calves were administered albendazole at 10 mg/kg of body weight, and the drug efficacy was determined by critical and controlled tests. Albendazole was only 74% effective against Haemonchus contortus (cambendazole-resistant strain), 99 to 100% effective against Ostertagia ostertagi, Trichostrongylus colubriformis, Cooperia oncophora, and Oesophagostomum radiatum, and 54% effective against the liver fluke. Except for H contortus, the critical test provided as accurate a measurement of drug activity against gastrointestinal nematodes as the controlled test provided.  相似文献

Young feral goats harbouring 3, 7 and 36 day old experimental infections of strains of gastrointestinal nematodes, known to be susceptible to anthelmintics in sheep, were dosed orally with commercially available anthelmintics at the manufacturers' recommended rates for sheep. The drugs and overall removals of Ostertagia circumcincta and Trichostrongylus spp. (T. colubriformis and T. vitrinus) respectively were: morantel citrate 69% and 68%; levamisole hydrochloride 55% and >99%; fenbendazole 98.5% and >99%; oxfendazole >99% and >99%. Results for Haemonchus contortus, while not inconsistent with high efficacy for all four drugs, must be interpreted with caution, in view of the erratic establishment of this worm in the controls.  相似文献

The persistent efficacy of doramectin and ivermectin injectable against moderate and high infection levels of Ostertagia ostertagi and Cooperia oncophora were evaluated in cattle. Calves were allocated to six groups of six animals. On Day 0 animals of Groups I1/I2 and D1/D2 were treated with 0.2mg/kg ivermectin and doramectin injectable, respectively. Animals of the C1, I1 and D1 groups received a daily (moderate) infection of 1000 L3 of O. ostertagi and 1000 L3 of C. oncophora, and animals of the C2, I2 and D2 groups received a daily (high) infection of 10,000 L3 of each species. The animals were infected for 21 days with both species, the infections with C. oncophora and O. ostertagi started from Days 8 and 15 post treatment, respectively. Animals were necropsied on Day 40. The calculation of the persistent activity of ivermectin and doramectin was based on the efficacy against the different developmental and adult stages of both parasites. The present study confirmed that infection levels may influence the duration of persistent efficacy of an anthelmintic. Doramectin had at the moderate infection level a persistent efficacy of at least 35 days against O. ostertagi and at least 28 days against C. oncophora; at the high infection dose persistent efficacy was somewhat shorter i.e. up to 33 days and approximately 28 days, respectively. The duration of persistent efficacy of ivermectin against O. ostertagi at the moderate infection level was between 14 and 25 days, at the high dose level up to 25 days. Persistent efficacy of ivermectin against C. oncophora could, at both infection doses, not be measured, with the present experimental design.  相似文献

The efficacy of the morantel sustained release bolus (MSRB) in reducing gastrointestinal parasitism in first season grazing calves was evaluated during the summer--autumn grazing seasons of 1982 and 1983 in western Oregon. Each of 38 calves (1982) and 40 calves (1983) were randomly assigned to either control or treatment groups which were given MSRB on the day of turnout onto pasture. Mean worm burdens from tracer calves grazed with treated animals in 1982 and 1983 showed overall reductions of 86.4% (P greater than 0.05) and 84.3% (P less than 0.01), respectively, compared to tracers grazed with controls. Ostertagia ostertagi, Cooperia oncophora and Nematodirus helvetianus were the primary nematodes collected at necropsy. Twelve full-season 1982 tracer animals (6 treated and 6 control) indicated an 88.1% (P less than 0.05) overall reduction in mean worm burdens. Mean fecal worm egg per gram (EPG) counts of treated animals reflected a reduction of 69% (P less than 0.05) in 1982 and 90% (P less than 0.05) in 1983. Autumn inhibition of O. ostertagi was observed. In the 1982 trial the control animals showed a slight mean weight gain advantage over the treated group from Day 84 until Day 160 (trial termination) when the mean difference was 7.9 kg. The final mean weight gain advantage of treated animals in 1983 was 13.5 kg (P less than 0.05). These trials demonstrated that the MSRB was an effective anthelmintic for reducing gastrointestinal parasitism in grazing calves and for decreasing pasture larval contamination.  相似文献

A controlled slaughter trial was undertaken to compare the efficacies of oxfendazole (5 mg/kg), morantel citrate (10 mg/kg), levamisole (8 mg/kg) and ivermectin (0.2 mg/kg) against experimentally induced infections of adult Haemonchus contortus, Ostertagia spp., Trichostrongylus colubriformis and Cooperiacurticei in sheep and goats. Ivermectin and oxfendazole achieved similar levels of efficacy in both hosts against all four worm genera as did levamisole and mortantel against H. contortus and C. curticei. Against Ostertagia spp. and T. colubriformis, however, the latter two drugs were less effective in goats than sheep. Neither the numbers of Ostertagia spp. removed from goats by levamisole (81% reduction) nor the numbers of T. colubriformis removed from goats by morantel (56% reduction) were statistically significant (P>0.05).  相似文献
