[Objective] The study aimed to introduce a rapid and effective method that is suitable for extracting genomic DNA from animal and plant. [Method] The genomic DNAs were extracted from tender leaves of 24 peanut cultivars and from the liver,lung and kidney of white mouse through the specifically modified CTAB method. The DNAs were run on agarose gel,next detected by DNA/Protein analyzer. Finally PCR amplification was conducted to detect the quality of DNAs extracted using the modified CTAB method. [Result] The clear and orderly bands were observed in gel detection,and the values of OD260/OD280 for DNAs extracted via modified CTAB method were between 1.77-1.83. The DNAs performed well in PCR amplification. [Conclusion] The DNAs extracted by modified CTAB method could satisfy the requirement of PCR amplification.  相似文献

[ObJective] The study aimed to identify Lycium Linn. at molecular level. [Method] The nrDNA ITS sequence of 5 edible Lycium Linn. germ-plasm resources were investigated. [Result] The nrDNA ITS regions of five edible Lycium Linn. germplasm resources were sequenced. The whole se-quences varied from 628 bp to 632 bp, with the average length of 630 bp. Total 79 variation sites were observed in the sequences, which accounts for 12. 5%. [Conclusion] Sequence analysis based on nrDNA sequencing provides a new approach to identify edible Lycium Linn. germplasm resources.  相似文献

Most of water flow in open channel or in river belongs to steady non-uniform flow.The surface profiles are caused by changes of channel section.It is very important to analyze its computation.According to the regularity of its surface cange,the suitable sectional dimensions of open channel or flood control work can be designed.Commonly,computation of non-uniform flow adopts the traditional methods by hand or by graphic method.The speed and precision of computation are restricted.In this paper,a software to calculate water surface profile is introduced.The software is put forward by using C^ .By means of interpolate method and dialogue between user and computer,we can calculate the water surface profile much more quickly and exactly.  相似文献

Based on the major gene and polygene mixed inheritance model for multiple correlated quantitative traits, the authors proposed a new joint segregation analysis method of major gene controlling multiple correlated quantitative traits, which include major gene detection and its effect and variation estimation. The effect and variation of major gene are estimated by the maximum likelihood method implemented via expectation-maximization (EM) algorithm. Major gene is tested with the likelihood ratio (LR) test statistic. Extensive simulation studies showed that joint analysis not only increases the statistical power of major gene detection but also improves the precision and accuracy of major gene effect estimates. An example of the plant height and the number of tiller of F2 population in rice cross Duonieai x Zhonghua 11 was used in the illustration. The results indicated that the genetic difference of these two traits in this cross refers to only one pleiotropic major gene. The additive effect and dominance effect of the major gene are estimated as -21.3 and 40.6 cm on plant height, and 22.7 and -25.3 on number of tiller, respectively. The major gene shows overdominance for plant height and close to complete dominance for number of tillers.  相似文献

[Objective] This study presents a method for mass rearing of Aphidius gifuensis, a dominant endoparasitoid of Myzus persicae on tobacco in Southwest China. [Method] The tobacco cultivar Honghuadajinyuan(Nicotiana tabacum) was used as the host plant and M. persicae was the host insect. In a greenhouse, tobacco seedlings were reared in plastic trays. The seedlings at three-true-leaf stage were inoculated with two to three aphids per plant using aphid source with a parasitism rate of 47% ±3.9%. [Result] By this inoculation method, the aphids and parasites were simultaneously inoculated on host plants. After approximately 25 d of rearing, we were able to produce 82.5±5.17 aphid mummies per tobacco seedling. A total of 445 500 aphid mummies were produced in one greenhouse(36 rearing trays per greenhouse) during an approximately 50-day rearing period. The emergence rate was 93.4% ±2.76%, and 54% of the mummies were females. [Conclusion] The demonstrated technological feasibility of using tobacco seedlings for the mass rearing of A. gifuensis increases the potential for the biological control of M. persicae.  相似文献
