ABSTRACT Two enzyme systems, cellulase (beta-1,4-glucanase) and laminarinase (beta-1,3-glucanase), were added to soil extracts to simulate (in vitro) lytic components found in mulches suppressive to Phytophthora cinnamomi. Concentration ranges of each enzyme were incubated with Phytophthora cinnamomi mycelium, zoospores, zoospores cysts, and zoospore-infected excised roots to evaluate the roles of each enzyme in potential control of avocado root rot disease. Cellulase significantly retarded the development of zoosporangia and chlamydospores when mycelia were incubated in soil extract containing the enzyme at concentrations greater than 10 units/ml. Zoospore production was also reduced by cellulase but not by laminarinase. Laminarinase had little effect on zoosporangia or chlamydospore formation. At high concentrations, laminarinase was consistently more effective at preventing encystment than cellulase. Chlamydospores preformed in root tips were immune to the lytic effects of all treatments except cellulase at 100 units/ml. Zoospores placed in enzyme solutions and plated on a selective medium survived high cellulase concentrations and formed colonies, but there were fewer surviving zoospores when laminarinase was present at greater than 10 units/ml. Low concentrations of cellulase stimulated infection of excised roots, however, low concentrations of laminarinase prevented infection. Cellulase and laminarinase have different effects on the structures of the Phytophthora cinnamomi life history, however, each enzyme may have a role in reduction of inoculum.  相似文献

ABSTRACT A series of samples were taken from mulched and unmulched trees starting at the surface of mulch or soil to a 15 cm soil depth, forming a vertical transect. Saprophytic fungi isolated from the soil samples on rose bengal medium and surveyed visually were most abundant in mulches and at the interface of mulch and soil (P < 0.05). Microbial activity as assayed by the hydrolysis of fluorescein diacetate was significantly greater in mulch layers than in soils. Cellulase and laminarinase enzyme activities were greatest in upper mulch layers and rapidly decreased in soil layers (P < 0.05). Enzyme activities against Phytophthora cinnamomi cell walls were significantly greater in mulch than in soil layers. When Phytophthora cinnamomi was incubated in situ at the various transect depths, it was most frequently lysed at the interface between soil and mulch (P < 0.001). Roots that grew in mulch layers were significantly less infected with Phytophthora cinnamomi than roots formed in soil layers. In mulched soil, roots were commonly formed at the mulch-soil interface where Phytophthora populations were reduced, whereas roots in unmulched soil were numerous at the 7.5 cm depth where Phytophthora cinnamomi was prevalent. Enzyme activities were significantly and positively correlated with each other, microbial activity, and saprophytic fungal populations, but significantly and negatively correlated with Phytophthora recovery.  相似文献

Phytophthora root rot is of paramount importance in avocado orchards of southern Spain. Soil solarization has been demonstrated to control the pathogen in infested areas from which infected trees had been removed. We aimed to determine whether soil solarization in established avocado orchards controls the disease. Soil solarization increased average maximum hourly soil temperatures by 6.5–6.9°C in unshaded areas of avocado orchards in coastal areas of southern Spain, depending on depth and year. The corresponding temperatures in shaded areas were c. 2–3°C lower. P. cinnamomi in soil, on infected avocado rootlets, and in a nutrient substrate buried at 30–60 cm depth was reduced to negligible amounts after 6–8 weeks of solarization in both unshaded and shaded locations of avocado orchards. P. cinnamomi could not be detected in avocado rootlets up to 14 months later, suggesting a long-term effect. Soil solarization did not affect growth of the trees, and fruit yields were increased as compared with control plots. Following soil solarization for 3 weeks from mid-July 1994, when maximum hourly temperatures reached 33–36°C, P. cinnamomi could not be recovered from a depth of up to 45 cm in unshaded areas or from a depth of up to 30 cm in shaded areas after the initial 10-day period. The viability of inoculum of the pathogen buried at depths between 15 and 60 cm in bare soil was determined by sequential sampling in two solarization experiments starting 12 June and 4 July 1995, respectively. In the first experiment, P. cinnamomi could not be detected at any depth after 4–8 weeks of solarization in unshaded areas but could be recovered at all depths except 15 cm in shaded areas. In the second experiment, where temperatures were higher and the soil surface not shaded, P. cinnamomi could not be recovered after 2 weeks at 15 and 30 cm.  相似文献

To combat the problem of residual film pollution and ensure the sustainable development of agriculture in oasis areas, a field experiment was carried out in 2019 at the Wuyi Farm Corps Irrigation Center Test Station in Urumqi, Northwest China. Four types of biodegradable mulches, traditional plastic mulchs and a control group (bare land; referred to as CK) were compared, including a total of six different treatments. Effects of mulching on soil water and heat conditions as well as the yield and quality of processing tomatoes under drip irrigation were examined. In addition, a comparative analysis of economic benefits of biodegradable mulches was performed. Principal component analysis and gray correlation analysis were used to evaluate suitable mulching varieties for planting processing tomatoes under drip irrigation. Our results show that, compared with CK, biodegradable mulches and traditional plastic mulch have a similar effect on retaining soil moisture at the seedling stage but significantly increase soil moisture by 0.5%-1.5% and 1.5%-3.0% in the middle and late growth periods (P<0.050), respectively. The difference in the thermal insulation effect between biodegradable mulch and plastic mulch gradually reduces as the crop grows. Compared with plastic mulch, the average soil temperature at 5-20 cm depth under biodegradable mulches is significantly lowered by 2.04°C-3.52°C and 0.52°C-0.88°C (P<0.050) at the seedling stage and the full growth period, respectively, and the water use efficiency, average fruit yield, and production-investment ratio under biodegradable mulches were reduced by 0.89%-6.63%, 3.39%-8.69%, and 0.51%-6.33% (P<0.050), respectively. The comprehensive evaluation analysis suggests that the black oxidized biological double-degradation ecological mulch made from eco-benign plastic is the optimal film type under the study condition. Therefore, from the perspective of sustainable development, biodegradable mulch is a competitive alternative to plastic mulch for large-scale tomato production under drip irrigation in the oasis.  相似文献

ABSTRACT In New Jersey, Phytophthora cinnamomi is the pathogen most commonly isolated from diseased roots and runners of the cultivated cranberry (Vaccinium macrocarpon). A second distinct species of Phytophthora has been isolated from dying cranberry plants and surface irrigation water. This species is homothallic with paragynous antheridia and ellipsoid-limoniform, nonpapillate sporangia. It was tentatively identified as P. megasperma in an earlier report. Laboratory experiments demonstrate that the cardinal temperatures for vegetative growth are between 5 and 30 degrees C with an optimum near 25 degrees C. Sporangia are produced at temperatures between 10 and 20 degrees C with the majority of sporangia produced at 10 and 15 degrees C. In pathogenicity tests, no growth effect was observed on cranberry plants (cv. Early Black) when tests were conducted at 25 degrees C; however, significant reductions in plant growth occurred when tests were conducted at 15 degrees C. This species was insensitive to metalaxyl but was sensitive to buffered phosphorous acid. Sequence analysis of the internal transcribed spacer 1 (ITS1), 5.8S rDNA, and ITS2 regions place these isolates in Phytophthora clade 6 with greatest similarity to Phytophthora taxon raspberry. To our knowledge, this is the first report of isolates of this affiliation in North America. However, the observation of low temperature preferences makes this species unique in an otherwise high temperature clade. The isolates described in this study are tentatively classified as Phytophthora taxon cranberry.  相似文献

Colony growth and germ tube emergence of sporangia and encysted zoospores of Phytophthora infestans were highly sensitive to cymoxanil (ED50 0.5–1.5 μg/ml), whereas differentiation of sporangia and zoospore release were insensitive at concentrations up to 100 μg/ml. Treated sporangia did not show distorted germ tubes. Oxygen consumption for glucose oxidation by germinating sporangia and zoospore motility were not inhibited at concentrations up to 100 μg/ml. Cymoxanil hardly affected the uptake of radiolabeled precursors of DNA, RNA, and protein at concentrations up to 100 μg/ml. Incorporation of [14C]phenylalanine into protein was completely insensitive. RNA synthesis as measured by [3H]uridine incorporation was differentially inhibited in the various developmental stages of the fungus. Inhibition did not occur at differentiation of sporangia, whereas at cyst and sporangial germination and mycelial growth this process was inhibited 20–45% at a concentration of 100 μg cymoxanil/ml. Endogenous RNA polymerase activity of isolated nuclei was not inhibited by cymoxanil. DNA synthesis as measured by [methyl-3H]thymidine incorporation was inhibited 20–80% at the various stages of development at cymoxanil concentrations higher than 10 μg/ml. Metalaxyl, a specific inhibitor of ribosomal RNA synthesis, inhibited [3H]uridine incorporation 40–60% at all developmental stages. The data suggest that although DNA synthesis is affected more than RNA synthesis, inhibition of both biosynthetic processes is a secondary effect. The primary mode of action of cymoxanil thus remains unknown.  相似文献

Ground green waste is used as mulch in ornamental landscapes and for tree crops such as avocados. Survival of Armillaria mellea, Phytophthora cinnamomi, Sclerotinia sclerotiorum, and Tylenchulus semipenetrans was assessed for 8 weeks within unturned piles of either recently ground or partially composted green waste. S. sclerotiorum survived at the pile surface and at 10, 30, and 100 cm within the pile for the entire 8 weeks in both fresh green waste (FGW) and aged green waste (AGW). A. mellea and T. semipenetrans did not survive more than 2 days in FGW, while P. cinnamomi persisted for over 21 days in FGW. AGW was less effective in reducing pathogen viability than FGW, most likely because temperatures in AGW peaked at 45 degrees C compared with 70 degrees C in FGW. Survival modeling curves based on pile temperatures indicate the time to inactivate 10 propagules of pathogens was 11, 30, 363, and 50 days for A. mellea, P. cinnamomi, S. sclerotiorum, and T. semipenetrans, respectively. Sclerotia-forming pathogens pose the greatest risk for escape; to ensure eradication of persistent fungi, green waste stockpiles should be turned intermittently to mix pile contents and move pathogen propagules to a location within the pile where they are more likely to be killed by heat, microbial attack, or chemical degradation.  相似文献

ABSTRACT Three carboxylic acid amide (CAA) fungicides, mandipropamid (MPD), dimethomorph (DMM) and iprovalicarb (IPRO) were examined for their effects on various asexual developmental stages of Phytophthora infestans in vitro and in planta. Germination of cystospores and direct germination of sporangia were inhibited with nanomole concentrations of MPD (0.005 mug/ml) and micromole concentrations of DMM (0.05 mug/ml) or IPRO (0.5 mug/ml). A temporary exposure of 1 h to CAAs was not detrimental to germination and infectivity of sporangia or cystospores. CAAs applied to cystospores at 1 h after the onset of germination did not prevent the emergence of germ tubes, but inhibited their further growth and deformed their shape. None of the fungicides affected discharge of zoospores from sporangia or the encystment (cell wall formation/assembly) of the zoospores. Mycelium growth in solid or liquid media was inhibited with micromole concentrations. CAAs mixed with sporangia and drop inoculated onto detached leaves strongly suppressed infection. Curative application at 1 day postinoculation (dpi) required higher concentrations of CAAs than preventive application to inhibit infection and lost its effectiveness at 2 dpi. When sprayed on established late blight lesions 4 days after inoculation, CAAs reduced sporangial production in a dose-dependent manner. Trans-laminar protection of potato or tomato leaves, although achieved with higher doses, was more effective with MPD than with DMM or IPRO. Shade house studies demonstrated superior control of late blight epidemics by MPD compared with the other molecules. The data suggest that germ tube formation by cystospores or sporangia is the most sensitive stage in the life cycle of P. infestans to CAAs. Of the three CAAs, MPD had the highest intrinsic activity against spore germination. This property, together with its better trans-laminar activity, makes MPD more effective than DMM or IPRO in controlling epidemics caused by P. infestans.  相似文献

In 1981 metalaxyl-resistant Phytophthora infestans was found in 44% of the 63 crops examined in south-west Scotland. In 1982, when fungicides containing acylalanines (metalaxyl or ofurace) were not used in the area, it was detected in 15%. Metalaxyl-resistant strains were found in several crops which had been sprayed only with non-acylalanine fungicides or had received no fungicidal treatment. In neither year was resistance associated with a significant breakdown of disease control.
Metalaxyl-resistant isolates of the blight fungus were obtained from tubers from crops in which resistance was detected on the foliage.
In detached leaf experiments, resistant isolates retained their ability to grow on leaves treated with 100 μg/ml of metalaxyl after 52 weekly transfers on untreated leaves, and produced sporangia on leaves treated with up to 2000 μg/ml of metalaxyl or ofurace. In mixtures with sensitive sporangia the proportions remained stable after nine weekly transfers on untreated leaves. However, at concentrations as low as 50 μg/ml, metalaxyl and ofurace reduced the extent of colonization and the numbers of sporangia produced by resistant isolates by an average of 50%.  相似文献

Rosellinia necatrix , the cause of white root rot in fruit trees, was efficiently controlled in an infested avocado orchard soil, by applying methyl bromide by deep injection (up to 90 cm) of cold gas and by surface application of hot gas, at a dosage (for heavy soil) of 1500 kg/ha and 1000 kg/ha, respectively. Efficiency of control was evaluated: 1) by testing pieces of naturally diseased avocado roots buried in the soil before treatment at different depths up to 100 cm; 2) by planting avocado and almonds in the treated field and in soil samples; 3) by burying avocado leaves in the soil samples. All the evaluaton tests showed that both treatments of methyl bromide were effective in controlling R. necatrix completely up to the depth of 100 cm, immediately and 9 months after treatment.  相似文献

Agrochemicals and plastics represent a burden on natural ecosystems and there is an urgent need to introduce alternative plant protection measures that have fewer negative impacts on the environment. Replacement of plastic mulches and synthetic pesticides with their biodegradable alternatives offers a way to decrease chemical residues. Pyrolysis liquids (PL) have been suggested as easily degradable and residue-free herbicides for agriculture. We tested the efficiency of PL (1%–10% of volume) containing peat mulch (PLM) in weed control under glasshouse and field conditions. We also estimated the length of the withdrawal period needed between PLM spreading and sowing/planting for crops and examined the light permeability and effects of PLM on soil temperature. In the glasshouse, the mulch amended with 5%–10% PL inhibited weed germination entirely. In the agricultural field, the number of weeds remained 44%–60% lower under PLM than under the control mulch. In a study performed in a city park, weed cover around the base of trees remained 64%–85% lower under PLM than without a mulch. However, a 7–21 d withdrawal period, depending on crop plant species, is needed to avoid injuring the crops. PLM inhibited sunlight effectively and the effects on soil heat sum across the growing season remained small. The weed-inhibiting effect of PLM is probably a result of both the PL compounds and the solid cover, formed by the sticky PL and peat fibres, which acts as a mechanical barrier. We conclude that the PLM is a promising alternative to plastic mulches.  相似文献

Weed suppression in sugar beets (Beta vulgaris.) is commonly achieved with two to three post-emergent herbicide applications across the entire field. Field studies were performed, in order to investigate the weed suppressing ability of Medicago lupulina, Trifolium subterraneum and a mixture of Lolium perenne and Festuca pratensis as living mulches in sugar beet at four locations in South Germany during 2014 and 2015. Living mulches were sown 2 and 30 days after sowing (DAS) of sugar beet. Weed densities ranged from 0 to 143 plants m?2 with Chenopodium album, Polygonum convolvulus and Polygonum aviculare being the most abundant weed species. It has been found that living mulches could reduce herbicide input up to 65?%. Weed suppression of living mulch was highest with Trifolium subterraneum (71?%). The early sown living mulches (2 DAS) revealed a 28 g m?2 higher biomass compared to late sowing (30 DAS). However, no any linear correlation was found between living mulch biomass and weed suppression. White sugar yield (WSY) was highest in the herbicide treatments (12.6 t ha?1). Trifolium subterraneum yielded the highest WSY of the living mulches with 11.1 t ha?1 across all locations. Our work reveals that living mulch can play a major role in integrated plant protection by reducing herbicides in sugar beet production.  相似文献

Phytophthora cinnamomi is an ecologically and economically important pathogen. In this study, PCR assays were developed with primer pair LPV2 or LPV3 for rapid detection and identification of this organism. Both primer pairs were selected from putative storage protein genes. The specificity of these primer pairs was evaluated against 49 isolates of P. cinnamomi , 102 isolates from 30 other Phytophthora spp., 17 isolates from nine Pythium spp. and 43 isolates of other water moulds, bacteria and true fungi. PCR with both primer pairs amplified the DNA from all isolates of P. cinnamomi regardless of origin. The LPV3 primers showed adequate specificity among all other species tested. The LPV2 primers cross-reacted with some species of Pythium and true fungi, but not with any other Phytophthora species. PCR with the LPV3 primers detected the pathogen at levels of a single chlamydospore or 10 zoospores in repeated tests. The PCR assay was at least 10 times more sensitive than the plating method for detection of the pathogen from artificially infested soilless medium, and, to a lesser extent, from naturally infected plants. PCR with LPV3 primers can be a useful tool for detecting P. cinnamomi from soilless media and plant tissues at ornamental nurseries, whereas the LPV2 primers can be an effective alternative for identification of this species from pure culture. Applications of these assays for detection of P. cinnamomi in other environments were also discussed.  相似文献

Plants of the eucalypt. Eucalyptus marginata. selected through a glasshouse screening procedure for resistance or susceptibility to Phytophthora cinnamomi , were established in tissue culture and micropropagated. After inoculation with P. cinnamomi , root lesions in clonal lines selected as resistant (RR) to P. cinnamomi were restricted and became contained within four days after inoculation while lesions in roots of those lines susceptible (SS) to P. cinnamomi continued to extend rapidly. Activity of phenylalanine ammonialyase (PAL) was increased above controls in root segments of the RR lines 48 h after inoculation with P. cinnamomi while activity in unselected seedlings and the SS lines was reduced or unchanged. After inoculation, lignin concentration was increased and reached high levels compared with uninoculated control levels in roots of the two RR lines tested. Constitutive levels of phenolics in roots of the RR lines were up to 94% higher than in seedling roots and levels were further increased after inoculation. Levels of phenolics in the other lines and seedlings were unaltered by inoculation. A line derived from resistant seedlings from a susceptible family (RS) had the highest constitutive levels of lignin, which were further increased after inoculation. Resistance to P. cinnamomi in clonally propagated E. marginata seedlings is based on similar mechanisms to those of field resistant species.  相似文献

测定了致病杆菌CB43菌株代谢物对灰葡萄病菌菌丝生长和孢子萌发的影响。结果表明:CB43代谢物对灰葡萄孢菌丝生长有较强的抑制作用,500、250、125ml/L代谢物能完全抑制菌丝的生长,并具有杀死作用。62.5ml/L的发酵液处理灰葡萄病菌菌丝72h的抑制率仍达83.35%。20ml/L的发酵液处理灰葡萄孢菌丝能引起菌丝畸形,异常分支、粗短。CB43代谢物能杀死灰葡萄孢分生孢子或抑制其萌发,500~62.5ml/L的发酵液处理16h后,孢子萌发抑制率达88.1%~97.91%。温室试验表明,CB43代谢物对黄瓜灰霉病的控制效果达84.09%,化学农药40%施佳乐的效果为72.94%。  相似文献

From 1999 to 2001, a survey on the occurrence of Phytophthora spp. in the rhizosphere soil of healthy and declining oak trees was conducted in 51 oak stands in Turkey. Seven Phytophthora spp. were recovered from six out of the nine oak species sampled: P .  cinnamomi , P .  citricola , P .  cryptogea , P .  gonapodyides , P .  quercina , Phytophthora sp. 1 and Phytophthora sp. 2. The most frequently isolated species, P .  quercina , was very common on slopes susceptible to drought. It occurred in four different climatic zones and on six Quercus spp., suggesting that it is native to oaks. The second most common species, P .  citricola , was separated into three subgroups: type C was recovered only in Anatolia, whereas A and B occurred only in the European part of Turkey. Phytophthora cinnamomi was recovered at one site only, and may not be involved in oak decline in Turkey. The other four species were recovered sporadically. On affected sites there was a significant association between deteriorating crown status and the presence of Phytophthora spp., particularly P .  quercina . The occurrence of Phytophthora species was significantly influenced by soil pH. Stem inoculation tests on oak seedlings revealed that Q .  petraea was the most susceptible species.  相似文献

Each of six pairs of metalaxyl-sensitive (MS) and metalaxyl-resistant (MR) field isolates of Phytophthora infestans was inoculated as a mixture of 90% MS sporangia with 10% MR sporangia onto fungicide-free potato plants (cv. Alpha) in six walk-in plastic tunnels in each of 2 years. In both experiments, late blight killed the crops in 26-31 days. In all tunnels, the frequency of MR sporangia increased during the logarithmic phase of the epidemic to 70-85% of the population. Later, however, the frequency of M R sporangia decreased to 7-38% in six tunnels but continued to increase to 94-100% in the other six. The increased frequency of MR during the logarithmic phase was attributed to the shorter latent period and larger lesion size of MR compared to MS isolates and to the unlimited availability of uninfected host tissue. The later decrease in the frequency of MR in six tunnels was attributed to the lower sporulation capacity and the shorter infectious period of MR compared to MS isolates, and to the limitation of uninfected host tissue. The data showed that the population dynamics of P. infestans was dependent not only on fitness components of isolates, but also on the availability of uninfected host tissue.  相似文献
