SUMMARY: Eight thermotolerant, ethanol-producing yeast strains were isolated from hot spring drainage at Rendaiji in Shizuoka Prefecture. The tolerance to elevated temperatures of three strains (RND13, 14 and 17) was studied in comparison with that of mesophilic yeasts. All three grew on a liquid culture at temperatures up to 42–45°C, with maximum growth rates at 37°C or 35°C, whereas the upper limit temperatures for fermentation were higher, ranging from 48 to 55°C. Above all, RND13 had higher ethanol productivity of 72.1% at 40°C for 24 h than that of 57.0% obtained by brewers' yeast at 25°C for 120 h. Regarding the taxonomy of RND13, 14, and 17, they were found to be Torulaspora delbrueckii , Dekkera sp., and Candida albicans , respectively. This is the first report on yeasts capable of fermentation over 51°C, as well as on the isolation of yeasts from a hydrosphere with elevated temperatures.  相似文献

ABSTRACT:   The synergetic effects of an immobilized cell system in an oleophilic polyurethane foam (PUF) and use of a thermotolerant strain on degradation of hydrocarbons by colorless green microalga Prototheca zopfii are reported. Two strains of P. zopfii , i.e. thermotolerant RND16 and non-thermotolerant ATCC30253, were immobilized in PUF to compare their abilities to biodegrade a mixed hydrocarbon substrate (MHS) containing aliphatic and polycyclic aromatic hydrocarbons (PAHs), as a function of temperature. The thermotolerant strain RND16 degraded MHS at 35°C, while ATCC30253 did not degrade hydrocarbons at temperatures higher than 30°C. Immobilization of P. zopfii in PUF resulted in shortened lag for growth-associated biodegradation of n -alkanes in MHS, the effect of which was most significant in cultures of RND16 at 25°C. Nevertheless, the decrease in the amount of degraded PAHs was caused by PUF immobilization and the level of this decrease was marked in the cultures of RND16, in which rate and extent of n -alkane degradation were higher than for ATCC30253.  相似文献

ABSTRACT:   Thermal tolerance was studied in a rainbow trout strain successively selected through high temperature breeding at 20–27°C since 1966 in Miyazaki Prefecture, Japan. The hatching rate and fry mortality at high temperatures were examined in the selected strain along with normal strains cultured at water temperature of 9–17°C. The hatching rate of embryos fertilized at either 10 or 14°C and subsequently subjected to high temperatures in the blastula or neurula stage of the selected strain, was marginally higher than that of the normal strain counterparts. The upper 50% lethal temperatures (LT50) for embryos in the early segmentation, blastula and neurula stages of the selected strain were also higher than those of the normal strain counterparts. Death temperatures and LT50 of fries acclimated to 20°C of the selected strain were significantly higher than those of the normal strains. However, no difference in the critical thermal maximum was detected between the different strains. These results suggest that the selected strain of rainbow trout established by selecting successively for many generations at high temperatures acquired a degree of thermal tolerance.  相似文献

ABSTRACT:   Feeding activity was examined at high temperatures by using a demand feeder for thermally selected and normal rainbow trout Oncorhynchus mykiss strains. When the water temperature was raised in experiment 1 from 17.5 to 25.7°C at 0.3°C/day in 21 days, the average daily food consumption rate in the thermally selected strain rose to 7.1%, which was significantly higher than that of the normal strain (4.1%, P  < 0.05). The corresponding rate was also significantly higher in the thermally selected (0.8%) than in the normal strain (0.2%, P  < 0.05) of fish in experiment 2 where water temperature was raised to 24.1°C in 0.5°C/day increments. When water temperature was raised rapidly in experiment 3 from 16.7 to 21.7°C in one day and gradually to 24.4°C in 28 days at 0.1°C/day, the average daily food consumption rates were 1.0 and 0.1% for the thermally selected and normal strains, respectively, with significant difference ( P  < 0.01). These results suggest that the thermally selected strain has acquired thermal tolerance as a result of artificial selection.  相似文献

SUMMARY: Effects of temperature, irradiation, salinity and inorganic nitrogen concentration on two cultured strains of zooxanthellae isolated from the corals Pocillopora damicornis (strain P) and Montipora verrucosa (strain M) were studied. Each strain showed different growth responses in terms of temperature and light intensity. A maximum growth rate of strain P, 1.2 per day, was observed at 32°C under all light intensities examined (5–40 μEm−2s−1). However, its photosystem 2 activity (FRI) was higher at 28°C than at 32°C under most light intensities. In contrast, the growth rate of strain M was affected more by light intensity and was almost invariably affected at all temperatures examined (24–36°C). Both algal strains had a comparable growth rate and FRI at salinities from 20 to 35 PSU under moderate temperature and irradiant conditions. High temperature and low irradiation reduced the algal tolerance against low salinity. The gross photosynthesis per cell was not affected by the ammonium enrichment more than 5 μM per day although the cellular chlorophyll a content and cell density increased in proportion with the ammonium enrichment up to 20 μM per day. A potential response of zooxanthellae to the multiple environmental stresses was shown from these results.  相似文献

ABSTRACT:   In order to understand the characteristics of burnt meat in cultured yellowtail Seriola quinqueradiata , fish were kept at two different temperatures (13 and 30°C) and slaughtered by either spinal cord destruction (SCD) or suffocation in air (SA). Early postmortem changes during storage at 32°C were analyzed by rheological, biochemical, and histological methods. The burnt meat (with lightness parameter, L* ≥ 55) was observed at 1-h storage in the SA 30°C group, at 2 h in SCD 30°C, and at 4 h in SA 13°C; meat was normal for the SCD 13°C group until 6 h of storage. Breaking strength scores were higher for the normal meat (200 g/cm2) than burnt meat (70 g/cm2) at 4 h of storage. Expressible water content was higher for the burnt meat than for the normal meat. Adenosine triphosphate concentrations for the SCD groups were higher than for the SA counterparts. Moreover, pH decrease was much faster in the 30°C groups, showing pH 5.6 at 2 h of storage. A negative correlation between the pH and lactic acid contents in muscle ( P  < 0.001) was found. Histological analysis evidenced a larger pericellular area (40%) in the burnt samples than in the normal samples (16%). It was confirmed that a higher fish-keeping water temperature and a stressful slaughter method (faster glycolytic process) were determinative factors that influence the occurrence of burnt muscle in yellowtail, and that the effect of the former is larger than the latter.  相似文献

Abstract. Trophonts of Cryptocaryon irritans Brown from infected three-spot damselfish, Dascyllus trimaculatus Ruppell, were kept at temperatures ranging from 7 to 37°C to observe encystment and development of the tomites. At 30, 25 and 20°C, the percentage of trophonts that had encysted in 16 h were 70, 77 and 64% respectively; at 37°C, 44% encysted and at 7°C only 10% had encysted.
The optimum temperature for excystment was 30°C; 50% excysted in 5 days and 100% in 7 days. At 25°C, 60% of the tomites started to excyst on the eighth day, and 70% on the ninth day. At 20°C, 10% started to excyst on the ninth day, reaching 40% on the tenth day. No excystment occurred at 37 and 7°C.
Newly encysted tomonts were placed in various dilutions of sea water (31 %0) and kept at temperatures ranging from 7 to 37°C. Low salinities, i.e. 16%0 and lower caused tomonts to rupture. At 37, 20 and 7°C, 35% of the tomonts started to rupture immediately in 50% sea water, while at 30 and 25 C, 30% of the tomonts ruptured in 25% seawater. However, none of the cysts developed normally at these dilutions. The percentage rupturing increased with decreasing salinity.  相似文献

Copepodite stage V Calanus finmarchicus were collected at locations on the continental shelf north of Scotland, in the Faroe–Shetland Channel and west of Ireland on six occasions covering winter, spring and summer from October 1993 to June 1995. Oxygen consumption by the overwintering and active spring/summer population of animals was determined at temperatures close to in situ temperatures. Laboratory measurements of oxygen consumption were also made at standardized temperatures (0°C, 5°C, 7°C and 12.5°C) to determine the sensitivity of animals to temperature change in the different seasons. Rates of oxygen consumption were very low (7–30 μmol O2 gC−1 h−1) at in situ temperatures during the winter and early spring and significantly higher (105–219 μmol O2 gC−1 h−1) for the active surface population in May and June. Animals collected from the overwintering population showed no significant response to changes in temperature. Due to the low respiration rates, the calculated rate of decrease in carbon content in diapausing copepodite stage CV was very low (approximately 0.250 μgC day−1). The respiration rates were used to construct a model to estimate survival of the animals with an initial carbon content equivalent to that expected of animals in October. The results showed that in order to survive during winter and have enough energy for moulting and migration to the surface in the spring, these animals have to live at temperatures close to 0°C and be in a diapause state.  相似文献

Abstract Sea bass head kidney cells were incubated at several temperatures with or without opsonized zymosan, Luminol was added to amplify light emission. When stimulated by zymosan cells emitted higher chemiluminescence (CL) at low temperatures (5–20°C) than at high temperatures (25–40°C). CL reached its peak earlier at high temperatures than at low temperatures. Unstimulated cells showed a low level CL response which rose with increasing temperature. Abstract. The eggs and yolk sac fry of the mouth brooding tilapia. Oreochromis mossambicus (Peters), were artificially reared in an experimental hatchery system at different temperatures. The hatchability, embryonic development of eggs and growth of yolk sac fry were studied under controlled conditions. It was shown that the upper lethal temperature for O. mossambicus eggs was above 40°C and the lower lethal temperature between 11°C and 17°C. For yolk sac fry these values lay between 34 and 40°C and 17 and 20°C respectively. Fry survival between 24·3 and 34·0°C was near to 100% while al 20°C it was less than 60%. The somatic growth rates of fry at temperature between 20 and 34·5°C up to 10 days post hatch are presented and were found to differ significantly. At 34·6°C a negative SGR was recorded during the period 6–9 days post hatching, but, during the first 6 days, the SGR at this temperature was more than four times higher than at 20°C. At the elevated temperatures, the utilization of yolk is faster and the loss of weight observed was due to starvation. This should be taken into account when incubating under artificial rearing condition so that initiation of exogenous feeding is timed most appropriately.  相似文献

ABSTRACT: Heating temperatures of 30–40°C and KCl concentrations of 0.1–0.5 M altered the denaturation mode of carp myofibrils. In 0.1 M KCl medium, heating temperature affected the denaturation of rod more significantly than of subfragment-1 (S-1), and a slow decrease in solubility at 30°C was accompanied by a slow denaturation of rod. KCl concentration at heating altered the denaturation mode differently at 30°C and 40°C. Increased KCl concentrations for heating reduced the rod denaturation rate at 40°C, but it was increased at 30°C. At concentrations above 0.3*Τ*M KCl, the denaturation rate for rod became identical to that for S-1 at both temperatures. Upon heating of chymotryptic digest of myofibrils, S-1 denaturation was similarly detected as in intact myofibrils, whereas practically no rod denaturation was detected. Thus, it was concluded that myosin structure connecting S-1 and rod has an important role in the denaturation process.  相似文献

ABSTRACT:   Fast skeletal muscles of Japanese flounder Paralichthys olivaceus and red sea bream Pagrus major were examined for quantitative and qualitative changes of mitochondrial ATP synthase (FoF1-ATPase) in association with rearing temperatures. The specific activities of FoF1-ATPase from Japanese flounder reared at 10°C, 15°C and 25°C for 4 weeks were determined to be 81 ± 11, 74 ± 13 and 83 ± 11 nmol/min·mg mitochondrial protein, respectively. The corresponding activity from red sea bream reared at 8°C for 5 weeks was determined to be 65 ± 9 nmol/min·mg mitochondrial protein, which was higher than 33 ± 9 nmol/min·mg mitochondrial protein in fish reared at 23°C. The contents of α- and β-F1-ATPase in total mitochondrial proteins were not significantly different between fish reared at different temperatures for the two fish species. However, the contents of β-F1-ATPase in the total fast skeletal muscle extracts, prepared from Japanese flounder reared at 10°C, were 2.1- and 2.9-fold higher than those for fish reared at 15°C and 25°C, respectively. The corresponding content from red seabream reared at 8°C was 2.2-fold higher than that for fish reared at 23°C. Therefore, the changes in FoF1-ATPase depending on rearing temperatures were species-specific.  相似文献

This study aimed to determine the effect of feeding rates and water temperatures on the growth, feed utilization and size heterogeneity changes in subadult pikeperch. Fish with an average weight of 84±19 g were fed with a commercial trout diet for 18 weeks at a water temperature of 20 °C and 25 °C. Four feeding rates (PSFR) were predicted at both temperatures, which corresponded to 1.0%, 1.25%, 1.50% and 2.0% of body weight per day (bw day−1) for the first, and to 0.8%, 1.0%, 1.2% and 1.4% of bw day−1 for the second 9 weeks respectively. At the end of the experiment, the individual weight averaged 273±82 g. Pikeperch showed a better growth rate [specific growth rate (SGR)] and apparent feed conversion rates (AFCR) at the higher temperature. Thermal-unit growth coefficient (TGC) was better at the lower temperature. The values of SGR, TGC and AFCR significantly increased with increasing PSFR at both temperatures. The values of TGC plotted against the computed feeding rates allowed estimation of the maximum feeding rates (1.25% and 1.15% of bw day−1 for pikeperch of 150–180 g at 25 and 20 °C respectively). Size heterogeneity changes were affected neither by the feeding rate nor by temperature.  相似文献

Abstract. The study includes mating and spawning data of freshwater crayfish, Austropotamobius pallipes Lereboullet, obtained in four consecutive years (1987–1990). A total number of 586 wild-caught female A. pallipes in the 21·65 mm to 45·60mm carapace length (CL) size were held under laboratory conditions at different dates in September–October. Initial densities at mating time were 21 and 24 crayfish/m2 with a 1:2 male:female ratio. High percentages of mating were obtained (97·4–100%) except in 1989 (57·5%). In all years, most of the matings (90%) were concentrated in a period of 8–12 days with mean water temperatures between 12°C and 13·5°C. Spawning took place a few days after mating (minimum 2 days, maximum 14 days) at mean water temperatures between 8·2°C and 10·8°C. In the first 3 years, almost 100% of mated females spawned. However, in the fourth year (1990), when the mean size of crayfish was smaller, only 72·4% of mated females spawned. The mean number of pleopodal eggs was 64 (maximum 220, minimum 18). Egg diameter ranged between 2·30 and 3·25 mm (mean value 2·78 mm). Pleopodal egg number was positively correlated with carapace length ( r = 0·72).  相似文献

Although water hyacinth is an adventive aquatic plant that threatens the Japanese ecosystem, its high growth rate may be a benefit in terms of its potential as a raw material for the production of bioethanol. In our study, water hyacinth was saccharified with 3?% (v/v) sulfuric acid and subjected to cellulase treatments to determine optimum bioethanol production conditions. The original saccharified solution yielded both glucose at 5.3?±?0.2?g/l and reducing sugars at 9.7?±?0.1?g/l. Concentration of the saccharified solution under vacuum at 70?°C yielded both glucose at 21.5?±?2.9?g/l and reducing sugars at 33.3?±?2.1?g/l. An aquatic yeast Saccharomyces cerevisiae TY2 produced ethanol at 9.6?±?1.1?g/l from the concentrated saccharified solution. In comparison, Ogawa et al. (Afr J Microbiol Res 2:110?C113, 2008) reported that this yeast produced 0.90?g/l and that the production of bioethanol was about 11.2-fold lower than that obtained in our study. These results suggest that refinement of the saccharification process and application of the selected yeast could improve the efficiency of obtaining bioethanol from the water hyacinth.  相似文献

Abstract— Feed intake, specific growth rate and changes in body composition were studied in age 1+ (140-190 g) brown trout ( Salmo trutta L.) reared at three temperatures (2, 4 and 6°C) under continuous light conditions. Feed in take increased from 35.7 kJ-kg−1.day−1 at 2°C to 95.7 kJ-kg−1-day−1 at 6°C, and the growth rate increased from 0.19%-day−1 to 0.42%-day−1 over the same temperature range. The estimated lower temperature limits for feeding and growth were slightly above 0°C. For all groups of fish, the majority (about 75%) of the weight gain comprised water and protein, but lipid deposition tended to increase with increasing temperature. The deposition of lipid accounted for about 50% of the body energy gain at all temperatures.  相似文献

ABSTRACT: Transglutaminase seems to be related to the setting phenomenon of fish meat paste that occurs at temperatures below 40°C. In many reports on the relationship between transglutaminase and setting phenomenon, the enzyme activity has been measured at 25°C. However, it is known that the setting phenomenon is complicated and the effect of calcium and the 3-D structure of myofibrils, which are sensitive to temperature, play important roles in the reaction. In the present study, total activities of transglutaminase of threadfin bream, white croaker, red sea bream and carp meats were measured at various temperatures. Total transglutaminase activities at 25°C of tested fish meats are arranged in order as follows; white croaker, red sea bream, carp, threadfin bream. In contrast, optimal temperature of carp meat is 30°C, red sea bream 40°C, threadfin bream 50°C and white croaker 50–55°C. In carp meat, the enzyme activity at optimal temperature (30°C) became 8.5-fold higher than at 25°C. The data of the total activity at various temperatures are useful in order to comprehend the details of the reaction.  相似文献

Abstract. The present report deals with experiments concerning the theoretical basis for development of a live, attenuated vaccine against viral haemorrhagic septicaemia (VHS) of rainbow trout, Salmo gairdneri Richardson, on the basis of a virus strain derived from the Egtved virus reference strain F, the pathogenicity of which has been strongly reduced by in vitro passage. It is shown that the low pathogenicity of the virus (designated the Reva strain) is a genetically stable feature and that the protection against VHS induced by infection with the virus is due to an immune response. Following immunological priming at 10°C partial protection developed at 5° 10° and 15° but not at 20°C, whereas neutralizing antibody was produced at all four temperatures by some of the fish. The duration of virus persistence in trout was found to be inversely proportional to the water temperature.  相似文献

Abstract. Kurasan and BHT were tested at doses of 100, 200 and 400mg kg-1 incorporated in dry pellets. The administration of these antioxidants did not influence the red blood count of rainbow trout, Salmo gairdneri Richardson, at body weights of 100 to 400g, reared in flow-through tanks at water temperatures of 7 to 13°C and in cage culture at water temperatures of 15 to 2O°C. In laboratory experiments at a water temperature of 14°C, the highest Kurasan dose increased the red blood cell (RBC) count insignificantly by 14% (1.06 vs 0.93 10121-1) after 80 days, haematocrit (PCV) was increased by 27% (0.410 vs 0.32511-1), and haemoglobin (Hb) by 16% (72 vs 62gl-1), This was seen in the cage culture experiment, but not in the experiments in the flow-through tanks. A trend of diminishing haematological parameters of the red blood count and total blood serum protein (TP) of the fish fed with BHT-stabilized diet was recorded only under laboratory conditions at the water temperature 9°C, The decline of RBC count in the experimental group with 0.04% BHT represented 14% (0.90 vs 1.05 10121-1), the decline in PCV amounted to 18% (0.328 vs 0.39811-1), Hb to 17% (57 vs 69g 1-1), and TP to 11% (39 vs 44g 1-1).  相似文献

Abstract. Chinese catfish, Clarias fuscus (Lacepède), within three size ranges were evaluated for growth, survival and feed conversion ratios (FCR). A 4 × 3 factorial design was used during year-long trials which included four temperatures (20,25,30 and 35 °C) and three feeding rates with commercially formulated salmon/trout feed. Specific growth rates of juvenile fish (1–60g) were highest, ranging from 1–64% body weight/day at 20°C (2%/day feed rate), to 4–94%/day at 25°C (6%/day feed rate). Specific growth rates of sub-adult fish (60–150g) averaged 0–65%/day, with no significant difference between temperature-feed rate treatments. Specific growth rates of adult fish (>150g) were lowest, averaging 0–30%/day for grouped feed rate treatments at 25°C, and 0–16%/day at 30°C. Most mortalities occurred during the juvenile stage, with survival ranging from 0% at 35°C(2% feed rate) to 96% at 30°C(6% feed rate). Survival was 95–100% with sub-adults and adults. FCR were 1 to 4 with juveniles, 2 to 12 with sub-adults, and 3 to 22 with adults. These trials suggest that optimal growth and FCRs were obtained at 25°C; with feed rates of 6%/day with juveniles, 2%/day with sub-adults, and 1%/day with adult fish. Final yields at 25 and 30°C indicate that even though growth and FCR are optimal at 25°C, survival was greater at 30°C, which compensated for reduced growth performance at this temperature. Yields at 25 and 30°C were almost double those at 20°C.  相似文献
