Cyprinid herpesvirus 3 (CyHV‐3) or koi herpesvirus (KHV) is a virulent viral infection in common carp and koi. The disease has caused global epizootic and economic loss in fish aquaculture and in the wild. Clinacanthus nutans (Burm. f.) Lindau is a well‐known medicinal plant used in Thai traditional medicine. Virucidal effects of the plant extract against human herpes simplex virus have been reported. In this study, C. nutans crude extract was tested for antiviral activities against CyHV‐3 in koi carp. Results showed effective antiviral activity against CyHV‐3 pre‐ and post‐infection. The 50% lethal concentration (LC50) of extract was higher than 5 mg/ml. The 50% effective dose (ED50) was 0.99 mg/ml, 0.78 mg/ml, 0.75 mg/ml and 0.71 mg/ml at 1, 2, 3 and 4 hr pre‐infection, respectively. The ED50 from post‐infection tests was 2.05 mg/ml and 2.34 mg/ml at 0 and 24 hr, respectively. These results demonstrated that crude extract expressed antiviral activity against CyHV‐3 and can be applied as a therapeutic agent in common carp and koi aquaculture.  相似文献

As a representative acidifier, lactic acid (LA) is widely used in the diets of aquatic animals. LA is the main supporter of energy metabolism and may be associated with drug metabolism. This study was designed to explore the pharmacokinetics of enrofloxacin (ENR) in Eriocheir sinensis (Chinese mitten crab) fed diets containing 0.1%, 0.2% and 0.3% LA (designated groups LA1, LA2 and LA3 respectively). The concentrations of ENR in the haemolymph, hepatopancreas and muscle were determined by HPLC after a single oral dose of 10 mg/kg. Our results showed that LA had a significant effect on the peak ENR concentrations in all the tissues (p < 0.05) by one‐way ANOVA analysis. There was a trend that Cmax (peak concentration) of ENR was elevated with LA levels increasing up to 0.3% in haemolymph, hepatopancreas, muscle and Tmax (time‐point of the peak concentration of the drug), t1/2β (elimination half‐life) and AUC(0‐∞) of ENR were shortened. Taken together, 0.3% LA might be effective in improving ENR pharmacokinetics in E. sinensis. Furthermore, it can be speculated that the enhanced biotransformation of ENR in the hepatopancreas mediated by LA is responsible for the differences in the pharmacokinetics of ENR in E. sinensis.  相似文献

Harmful algal blooms (HABs) are natural phenomena with different effects on the aquatic environment that affect both human economy and health. Several genomic studies have been done to characterize the effects of contaminants on Crassostrea gigas. However, oysters’ molecular response to HABs exposure needs to be studied more. In this study, we challenged C. gigas with Prorocentrum lima, a diarrhoetic toxin producer, under controlled experimental conditions considering dinoflagellate density and exposure time (acute and sub‐chronic) as variables. The expression profile of six stress response genes was analysed by semiquantitative RT‐PCR: Glutamine synthetase (GS), Glutathione S‐transferase (GST), heat shock protein 70 (HSP70), and heat shock protein 90 (HSP90), CuZn superoxide dismutase (SOD) and melanogenic peroxidase (POX). The results revealed that these gene expressions depend on exposure time and cell concentration. In general, an increased expression was observed in all tested cell densities as immediate response to exposure (0–3 h); whereas lower expression levels than control were observed after 6 h. Treatment with 3,000 cells mL‐1 promoted higher expression than control on most genes after 336 h of exposure. This is the first report providing information at molecular level on C. gigas response to dinoflagellate blooms.  相似文献

Plant protein‐based diets are widely used for common carp (Cyprinus carpio var. Jian). The plant source antagonist, such as phytate, negatively affects the bioavailability of trace minerals, but this aspect was not considered in previous studies on this species. A 10‐week growth trial was conducted to investigate the effects of inorganic Zinc (ZnSO4·7H2O, Zn‐S) and organic Zn (2‐hydroxy‐4‐(methylthio) butanoic Zinc, Zn‐M) on growth performance, Zn retention and antioxidant responses of common carp (initial weight 10.0 ± 0.03 g). Eleven semi‐purified diets containing 7 mg/kg phytic acid were prepared with 0, 5, 20, 35, 50 and 65 mg/kg Zn from Zn‐S and Zn‐M (12.8 mg/kg Zn in basal level), respectively. The results showed that either Zn‐S or Zn‐M inclusion improved growth performance, vertebral Zn deposition and antioxidant responses of fish. The Zn requirement based on WGR, vertebrae Zn and plasma SOD were 47.0, >77.8 and 48.5 mg/kg, and 63.3, 42.2 and 36.8 mg/kg, respectively, with Zn‐S and Zn‐M as Zn sources. The relative bioavailability of Zn‐M was 2.44‐ (vertebrae Zn deposition) and 1.74‐fold (plasma SOD activity) the availability of Zn‐S in the diet of common carp. The lower growth performance and poor bioavailability in vertebrae of Zn‐S group were related to the chelating of zinc with phytic acid.  相似文献

In this study, the testis and semen fatty acid, fat‐soluble vitamin and sterol levels, testis‐somatic index and spermatological values were investigated in the broodstock rainbow trout fed with the different levels of omega‐3 polyunsaturated fatty acid (PUFA) under the regular stripping conditions. For this purpose, a control and three test diets were prepared. The control, D1, D2 and D3 diets were contained the omega‐3 PUFA at 3.14%, 7.84%, 13.63% and 20.54% as a percentage of the total fatty acids respectively. The fish in the experimental groups were fed with these diets for 60 days. The testis‐somatic index (3.81–4.50), semen volume (9.30–18.5 ml) and pH (8.05–8.99), initial sperm motility (68.1%–93.7%), duration of 50% sperm motility (141.3–258.9 s), total duration of sperm motility (268.7–489.6 s) and sperm density (9.95–17.5 cells × 109 ml?1) values of the fish fed the control diet were significantly decrease (p < 0.05) depending on decline of the omega‐3 PUFA, fat‐soluble vitamin and sterol contents in the testis and semen. The omega‐3 PUFA (3.40%–14.76% and 3.96%–16.31%), vitamin A (33.9–61.2 ml/mg and 9.12–26.7 ml/mg), D2 (0.10–0.19 ml/mg and 0.04–0.10 ml/mg), D3 (0.75–1.30 ml/mg and 0.06–0.24 ml/mg), alpha‐tocopherol (30.1–60.4 ml/mg and 8.93–25.3 ml/mg), K1 (0.30–0.94 ml/mg and 0.03–0.08 ml/mg) and K2 (2.0–3.19 ml/mg and 0.25–0.34 ml/mg) vitamins, cholesterol (1,201–1,588 ml/mg and 330–596 ml/mg), ergosterol (3.12–6.40 ml/mg and 2.10–4.08 ml/mg), stigmasterol (5.57–10.4 ml/mg and 10.2–12.6 ml/mg) and beta‐sitosterol (0.20–0.60 ml/mg and 0.14–0.41 ml/mg) contents in the testis and semen of the fish were significantly affected (p < 0.05) by the dietary omega‐3 PUFA supplementation. The highest values for these parameters were provided from the fish fed the D2 and D3 diets (p > 0.05), followed by the fish fed the D1 diet (p < 0.05). These results showed that the dietary omega‐3 PUFA at 13.63% of the total fatty acids could increase the omega‐3 PUFA, fat‐soluble vitamin and sterol contents in the testis and semen, the testis‐somatic index and spermatologic values of the broodstock rainbow trout under the regular stripping conditions.  相似文献

An experiment was carried to determine the plasma fenbendazole (FBZ) concentration and physio‐metabolic responses in juveniles of Labeo rohita (90 ± 4 g) after oral administration of single doses at 10, 20 and 50 mg, 20 mg FBZ/kg b.wt. in multiple times on 1st, 3rd and 7th day. The blood samples were collected at 0.5, 1, 2, 4, 8, 12, 24, 30, 48, 72, 96 and 120 hr, after single‐dose administration, and regularly (upto 15 day) in multiple dose. Plasma FBZ concentration was determined up to the limit of detection (LoD) of 0.09 µg/ml by HPLC. There was no parent drug detected in plasma for administration of 10 mg FBZ/kg b.wt. The drug attained the peak concentration (Cmax) 1.85 and 3.09 µg/ml in plasma at 4 hr (Tmax) after administration of 20 and 50mg FBZ/kg b.wt. respectively. Plasma FBZ was detectable up to 96 and 120 hr with concentration 0.09 ± 0.007 and 0.098 ± 0.006 µg/ml, respectively, after single‐dose administration of 20 and 50mg/kg b.wt. In case of multiple‐dose administration, the maximum concentration of FBZ was 1.01 ± 0.03 µg/ml on 7th day that was less than to the single dose at 50 mg/kg b.wt. However, FBZ was detected up to 11 day after multiple doses. The study revealed that the hepatic antioxidant enzymes activities like superoxide dismutase, catalase and glutathione‐S‐transferase were significantly affected by increasing FBZ in single and multiple doses. The results of the present study could reveal that single‐ or multiple‐oral administration of FBZ at 20 mg/kg b.wt. in feed as antihelminthic drug in L. rohita could be considered as the safe dose.  相似文献

A sixty‐day feeding trial was conducted to determine the ascorbic acid (AA) requirement for growth of striped catfish, Pangasianodon hypophthalmus juveniles. Seven iso‐nitrogenous and iso‐energetic (370 g protein per kg and 19.6 MJ/kg) purified diets were prepared with different levels of ascorbic acid such as control (0), T1 (17.5), T2 (35), T3 (70), T4 (175), T5 (350) and T6 (700) mg ascorbic acid (L‐ascorbyl‐2‐polyphosphate) equivalent per kg diet. Fish with a mean body weight of 3.2–3.4 g were stocked (fifteen fish per tank) in triplicates following a completely randomized design. Each group was fed to satiation twice a day for 60 days. Significant differences were observed in growth, survival, body composition and metabolic enzymes activities with different dietary ascorbic acid levels. Maximum weight gain, specific growth rate (SGR) and protein efficiency ratio (PER) were found in fishes fed with 35 mg AA per kg diet, supported by best feed conversion. Fish fed a diet containing vitamin C had the highest activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) compared to those fed with vitamin C‐depleted diets. In this study, based on using broken‐line regression analysis, the dietary vitamin C requirement for growth of P. hypophthalmus juveniles was estimated to be in the range of 46–76 mg AA per kg, depending on the criterion used, growth and liver storage. Our results will be helpful for the formulation of cost‐effective ascorbic acid incorporated diets for striped catfish, P. hypophthalmus.  相似文献

This study investigated the potential of linalool as an anaesthetic during transportation of common carp, Cyprinus carpio. The fish were transported at a loading density of ~103 g/L for 3 hr in 12 plastic bags (3 L water and 6 L pure oxygen) divided into four triplicated treatments: control (without linalool), L50 (50 mg linalool/L), L100 (100 mg linalool/L) and L200 (200 mg linalool/L). After 3‐hr transportation, serum physiological responses and water physico‐chemical parameters were compared among the treatments. Results showed that water total and un‐ionized ammonia increased and dissolved oxygen decreased in all treatments after transportation. Water total ammonia and dissolved oxygen levels in the linalool‐treated bags were significantly lower than the control. After transportation, the control fish showed significant elevation in serum cortisol and glucose levels; however, the levels were significantly lower than the linalool‐treated fish. All fish showed similar serum lactate levels, significantly lower than the value obtained before transportation. The control fish showed serum urea levels similar to the level obtained before transportation; however, the linalool‐treated fish had significantly higher serum urea levels compared to the control and before transportation levels. After transportation, all fish had decreased serum chloride and sodium levels compared to the before transport level. Overall, the present results showed that linalool is not beneficial for carp transportation in plastic bags, because it reacts with water oxygen, increases stress in fish, interferes with ammonia excretion and has no benefits in preventing ion loss.  相似文献

Spatial and seasonal variation in the abundance and composition of phytoplankton and other basal resources (bacteria, terrestrial detritus) influence the availability of essential polyunsaturated fatty acids (PUFA) for upper trophic levels in lake food webs. We studied accumulation, composition and content of fatty acids in planktivorous vendace (Coregonus albula) muscle tissue during the summer feeding period (May to late September) in six boreal lakes. Vendace muscle fatty acid content increased from ~17.7 mg/g DW to ~28.1 mg/g DW from spring to summer, corresponding to a ~59% increase in total fatty acids. PUFA accounted for 45%–65% of all fatty acids, and the most abundant fatty acid was docosahexaenoic acid (DHA, 22:6n‐3). The DHA content remained relatively constant throughout the sampling period (~7–8 mg/g DW), but the proportion of DHA decreased from ~40% in spring to ~30% in summer and autumn. The decrease in DHA proportion was due to accumulation of other fatty acids, primarily C18 PUFA (predominantly synthetised by algae) and monounsaturated fatty acids. A similar fatty acid accumulation pattern was observed in lipid storing copepods, an important prey for planktivorous vendace. The abundance of bacterial and terrestrial biomarker fatty acids in vendace muscle and prey items was low (<5% and <0.1% respectively). The increase in weight and the deposition of internal energy stores in vendace were achieved predominantly by algae‐based food chain. This has ecological implications, as vendace requires sufficient lipid stores for reproduction in autumn and survival over winter.  相似文献

The objective of this study was to investigate the effects of ammonia nitrogen on the embryonic development of the cuttlefish Sepia pharaonis . Embryos were exposed to different concentrations (0, 1, 3, and 5 mg/L) of total ammonia–nitrogen (TAN) during incubation. The developmental rate, malformations, mortality, hatching rate, incubation period, yolk utilization efficiency ratio, and weight of the newly hatched cuttlefish were determined. The results showed that ammonia nitrogen significantly inhibited the development of S. pharaonis embryos and induced malformations and even death. Hatching was delayed, the hatching rate was reduced, mortality and the incubation period increased, and the yolk utilization efficiency ratio and weight of the newly hatched cuttlefish significantly decreased in a dose‐dependent manner after the embryos were exposed to more than 1 mg/L TAN for prolonged period. These variables could be used as an integrative biomarker or indicator of aquatic environmental ammonia contamination. In summary, our results indicated that ammonia caused toxicity in the embryos. When the concentration of TAN is greater than 1 mg/L, ammonia levels should be reduced to prevent toxic effects on embryonic development.  相似文献

The objective of the present study was to estimate the dietary thiamine (vitamin B1) requirement of juvenile soft‐shelled turtles, Pelodiscus sinensis. Eight experimental diets containing 0, 1.90, 3.63, 5.65, 7.51, 9.62, 11.37 and 13.64 mg thiamine/kg diet were fed to 160 soft‐shelled turtles reared individually for 10 weeks. The turtles had an average weight of 5.33 ± 0.21 g. Among all the dietary groups, weight gain, feed utilization and tissue thiamine were the lowest in the turtles fed with a thiamine‐free diet. The variables increased when dietary thiamine increased and then levelled off beyond 3.63 mg/kg diet. In contrast, plasma pyruvate and lactate concentrations in turtles decreased when dietary thiamine increased and then levelled off beyond 3.63 and 5.65 mg/kg diet respectively. Estimation using a broken‐line regression model, the vitamin B1 requirement of soft‐shelled turtles was 3.2 mg thiamine/kg diet based on weight gain and plasma lactate concentration. When tissue thiamine and plasma pyruvate concentrations were used as the indicators, the estimated requirement was 5.4–6.5 mg/kg diet.  相似文献

The effects of vitamin B1 on growth, blood metabolites, body composition, intestinal enzyme activities and morphometric parameters were evaluated by a 63‐day feeding trial in Pelteobagrus fulvidraco fed diets containing vitamin B1 with 4.29, 6.02, 7.86, 11.94 and 19.05 mg/kg, respectively. Results showed that 7.86 mg/kg vitamin B1 resulted in significantly higher weight gain (WG), specific growth rate (SGR) and protein efficiency ratio (PER) (p < .05). Serum triglyceride, cholesterol and whole‐body lipid were observed in 11.94 mg/kg vitamin B1 group significantly higher than those in 4.29 mg/kg group, accompanied by the lower serum glucose content in the same group (p < .05). Significantly higher values of whole‐body protein, serum total protein, relative intestinal length, fold height, intestinal trypsin, amylase, alkaline phosphatase, Na+/K+‐ATPase, γ‐glutamyl transpeptidase, creatine kinase, muscular layer thickness and intestosomatic index were observed in 7.86 mg/kg vitamin B1 group compared with those in 4.29 mg/kg group (p < .05). These results suggested that diets supplemented with vitamin B1 improved growth performance, feed utilization, intestinal digestive and absorption capacity of juvenile yellow catfish. The optimal dietary vitamin B1 requirements estimated using a two‐slope broken‐line model based on WG and PER of yellow catfish were 7.42 and 6.01 mg/kg, respectively.  相似文献

This study evaluated efficacy and toxicity of the pyrazinoisoquinoline anthelmintic praziquantel (PZQ) in barbel infected with metacercariae of Diplostomum spathaceum and adult Pomphorhynchus laevis, and assessed antioxidant biomarkers and the lipid peroxidation response in juvenile barbel post‐treatment. The estimated 96‐hr LC50 of PZQ was 28.6 mg/L. For evaluation of efficacy, barbel naturally infected with D. spathaceum were exposed to a 10 and 20 mg/L PZQ 4‐day bath treatment. Both concentrations were 100% effective against D. spathaceum and significantly (< .01) affected the activity of catalase, superoxide dismutase, glutathione reductase and glutathione‐S‐transferase as well as levels of reduced glutathione in liver and muscle. The efficacy of orally administered PZQ was assessed in adult barbel naturally infected with P. laevis. Fish were administered 10, 30 and 50 mg/kg of body weight and examined via gut dissection after 6 days. The 50 mg/kg dose significantly decreased the intensity of infection. Praziquantel is a feasible bath treatment for barbel infected with D. spathaceum and has potential for oral treatment of broodfish infected with P. laevis.  相似文献

Dicentrarchus labrax were fed from 10 to 40 days posthatching with six microdiets differing in the inclusion level (60 g kg‐1 and 120 g kg‐1) and type of protein hydrolysate (PH; yeast, YPH; pig blood, PBPH; pig red blood cells, PRBCPH). A microdiet containing 120 g kg‐1 fish PH (FPH) was used as a control. PH differed in their amino acid (AA) profile and molecular weight distribution and therefore the tested microdiets too. The estimated content in FAA and di‐ and tripeptides in the FPH microdiet was 2 g kg‐1 and 44 g kg‐1, respectively. FAA estimated levels in YPH and PBPH microdiets were 26 g kg‐1 and 53 g kg‐1, whereas levels of di‐ and tripeptides were 30 g kg‐1 and 60 g kg‐1, respectively. The estimated levels of FAA in PRBCPH microdiets were 8 g kg‐1 and 17 g kg‐1, whereas estimated levels of di‐ and tripeptides were 11 g kg‐1 and 22 g kg‐1, respectively. Results revealed that FPH may be replaced by alternative PH from yeast and pig blood products, as fish fed those diets performed, in terms of growth, survival, digestive function and incidence of skeletal deformities, as well as those fed the FPH microdiet. Using YPH, PBPH and PRBCPH, the inclusion level of PH in microdiets might be reduced to a half with respect to current practices using FPH.  相似文献

The effects of dietary administration of inorganic zinc (zinc sulphate, ZnSO4) and nano zinc (zinc oxide nanoparticles, ZnO‐NP) were evaluated in rohu, Labeo rohita fingerlings. Fish were fed with a basal diet (Control) supplemented with ZnSO4 (T1, T2 and T3) and ZnO‐NP (T4, T5 and T6) at 10, 20 and 30 mg/kg, respectively, for a duration of 45 days. The results revealed that fish fed diet containing 20 mg ZnO‐NP per kg (T5) had the highest weight gain and specific growth rate (SGR, % per day), which was significantly different (p < .05) from the other experimental diets. Significantly (p < .05), higher activities of the digestive and metabolic enzymes were recorded in the fish fed ZnO‐NP containing diets as compared to the diets containing inorganic Zn or control diet. The maximum serum glucose and protein levels were noted in fish reared on diet T5. Both SGOT and SGPT activities were significantly increased in fish fed Zn‐supplemented diets (T1 to T6), as compared to the control group. Similarly, innate immune parameters were improved with feeding Zn incorporated diets. The highest phagocytic (40.74 ± 0.65%) and respiratory burst (0.33 ± 0.001, OD 630nm) activities were recorded in the fish fed diet containing ZnO‐NPs at 20 mg/kg (T5). The maximum superoxide production and serum peroxidase activity were detected in the fish fed T5 and T6 diets. Overall, results indicated that short‐duration feeding (≤45 days) of dietary ZnO‐NP (20 mg/kg) improved growth, enzyme activity, serum biochemical parameters and immune function in rohu fingerlings.  相似文献

Roasted coffee powder (RCP; Coffea arabica) is usually used as a beverage for human but there are few attempts to use it as a natural feed supplement in fish diets. In this study, common carp, Cyprinus carpio L., (11.8 ± 0.09 g) were reared in zinc (Zn)‐containing water at concentrations of 0.0 or 5.0 mg/L and cosupplemented with 0.0 or 1.0 g RCP/kg diet for 6 weeks to investigate effects of RCP supplementation, Zn exposure and their interaction on fish performance, biochemical variables, antioxidant activity and Zn bioaccumulation in whole fish body. Fish growth and feed intake were significantly affected by RCP supplementation, Zn toxicity and their interaction. However, fish fed a RCP‐supplemented diet did not exhibit better performance than those fed the RCP‐free diet and both diets produced higher fish performance than the Zn‐toxicated fish. It is noticed that RCP supplementation to Zn‐toxicated fish enhanced their growth, and feed utilization as compared to Zn‐toxicated fish alone. Fish fed control and RCP‐enriched diets showed no significant differences in biochemical variables, which were significantly altered due to waterborne Zn toxicity. Moreover, Zn reduced significantly; meanwhile, RCP supplementation increased significantly superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities. Notably, Zn exposure could reduce fish growth and antioxidant activity and increase Zn deposition in whole fish body. And RCP intake could enhance the antioxidant activity exerting a protective effect against Zn toxicity, thereby reducing Zn bioaccumulation in whole fish body.  相似文献

We studied the simultaneous effect of sex and dose on anaesthesia efficacy to estimate, if possible, the lowest effective dose (LED) for clove oil, tricaine methanesulphonate (MS‐222), 2‐phenoxyethanol (2‐PE) and propofol in mature guppies. LED is the lowest dose needed to reach A5 stage in a mean time of 3 min, with mean recovery (R5) time of 5 min. We used four doses/anaesthetic: 25, 50, 75 and 100 mg/L for clove oil; 120,140,160 and 180 mg/L for MS‐222; 800, 1,000, 1,200 and 1,400 mg/L for 2‐PE, and 7.50, 8.25, 10.00 and 11.25 mg/L for propofol. Each dose was tested on 10 females and 10 males. Morbidity, mortality and behavioural changes were checked on two control groups (10 males and 10 females/group). Sedation (A3), A5 and R5 times were recorded. Significant interactive effect dose*sex on A5 time was found for each anaesthetic agent (pdose*sex < .05). Except for MS‐222 (pdose*sex = .284), significant interactive effect dose * sex on R5 time was found (pdose*sex < .05). A5 time in females tended to be greater than in males, but, in general, R5 times were longer in males. Body size differences between males and females could explain these differences in MS‐222 on A5 time and for clove oil, 2‐PE and propofol on R5 time. No dose simultaneous meet LED′s conditions relating to both A5 and R5 times; therefore the lowest doses inducing A5 in a mean time of 3 min could be a safe guideline for anaesthetic procedure in both male and females.  相似文献

An 8‐week experiment was designed to determine the optimum dietary iron requirement of juvenile cobia Rachycentron canadum (mean initial weight, 15.89 ± 0.84 g) with iron sulphate (FeSO4•7H2O) and iron methionine (FeMet) as iron sources, using a semi‐purified diet based on casein and white fish meal as the protein sources. The basal diet was supplemented with 0, 30, 60, 120, 240 and 480 mg iron kg−1 dry diet from either FeSO4 or FeMet, respectively. Survival was not significantly affected by the all dietary treatment. Weight gain (WG), feed efficiency (FE), serum catalase activity (SCAT), and haemoglobin were significantly affected by any of the dietary treatments from both of two iron sources. Based on broken‐line regression analysis of WG, FE and SCAT, a minimum requirement for dietary iron was recommended to be 80.5–94.7 mg kg−1 from FeSO4 and 71.3–75.1 mg kg−1 from FeMet. Iron supplement to the basal diet had no significant effect on haematocrit, erythrocyte count, iron concentration in whole body and fillet. Our experiment also showed that the bioavailability of FeMet and FeSO4 to juvenile cobia was similar for WG and FE, and the relative bioavailability of FeMet and FeSO4 to juvenile cobia was 275% for maximum SCAT.  相似文献

Aeromonas veronii is one of the main pathogens causing freshwater fish sepsis and ulcer syndrome. More and more cases have shown that it has become an important zoonotic and aquatic agent. In this study, a A. veronii TH0426 mutant strain (ΔlamB) with an in‐frame deletion removed nucleotides 10–1,296 of the lamB gene was firstly constructed to investigate its functions. The results showed that the LD50 value of the mutant ΔlamB to zebrafish and mice was 13.7‐fold and 5.6‐fold higher than those of the wild‐type strain, respectively. The toxicity of wild‐type strain to EPC cells was 2.1‐fold and threefold higher than those of ?lamB when infected for 1 and 2 hr. Furthermore, the ability of biofilm formation and the adhesion and invasion to EPC cells of ?lamB significantly decreased for 5.6‐fold and 1.8‐fold separately. In addition, motility detection result indicated that ?lamB lost the swimming ability. The results of flagellar staining and TEM demonstrated that the flagella of ?lamB were shed. In general, the deletion of lamB gene caused a significant decrease in the virulence and adhesion of A. veronii TH0426, and it can be known that the lamB gene of A. veronii plays a crucial role in the pathogenesis.  相似文献

The binding of carotenoids to the myofibrillar protein F‐actin purified from the white muscle of Atlantic salmon (Salmo salar L.) was studied using in vitro reconstitution. The binding of astaxanthin and canthaxanthin was saturable, and analysis revealed the presence of a single carotenoid‐binding site. The dissociation constants (Kd) for actin prepared from 2.5 kg FW (Fresh Weight) fish were 1.04 ± 0.13 μg carotenoid per milligram of actin and 0.54 ± 0.11 μg/mg for astaxanthin and canthaxanthin, respectively. The saturation binding level (Bmax) for astaxanthin was 1.39 ± 0.07 μg/mg and 1.04 ± 0.08 μg/mg for canthaxanthin. These values were higher for F‐actin prepared from organic and small (~0.5 kg FW) salmon than for non‐organic and larger, mature fish. The structural specificity of carotenoid binding revealed a preference for carotenoids that possess a keto group at C‐4 on the β end group of the molecule, but the presence of hydroxyl groups at C‐3 or C‐4 reduced overall binding efficiency. The study suggests that the ability of myofibrillar proteins to bind carotenoids is not a limiting factor governing the deposition of carotenoids in the muscle of salmonids.  相似文献
