The objective of the present study is to examine whether colostrum whey can have an effect on immune function in goats digestive tract. Two milliliters of colostrum whey (colostrum group) or water (control group) were administrated orally to goats every day for 3 weeks. Blood was collected twice a week for 3 weeks to measure immunoglobulin A (IgA), interleukin 8 (IL‐8), and IL‐10. At the end of the experimental period, the parotid glands, oral mucosa, lingua, esophagus, jejunum, and ileum were collected for immunohistochemical detection of IgA, cathelicidin‐7, and S100A8. The ratio of the length of IgA‐positive mucosal surface in the esophagus to the total esophageal length was significantly greater in the colostrum group than in the control group. The number of IgA‐positive cells in the labial gland and ileum in the colostrum group was significantly higher than that in the control group. There were no significant differences between the colostrum and control groups in the number of cathelicidin‐7‐positive cells in the jejunum and ileum and in the number of S100A8‐positive cells in the lingua, jejunum, and ileum. These results suggest that colostrum stimulates the recruitment of plasma cells into the labial gland, which then secrete more IgA into the saliva.  相似文献

Our previous study showed that intrauterine-infused lipopolysaccharide (LPS) can be translocated to the mammary gland to induce weak inflammation. This study aimed to determine whether dexamethasone treatment facilitated the translocation of LPS from the uterus to the mammary gland to induce a heavy inflammatory response. Sixteen goats were divided into control and LPS groups, subjected to daily dexamethasone administration before saline or LPS infusion. Milk and blood samples were collected before and after LPS infusion to determine the milk yield and somatic cell count (SCC) and blood leucocyte count (BLC), cytokines, antimicrobial peptides and serum amyloid A (SAA) concentrations. Mammary gland tissues were collected from two goats before and 24 hr after LPS infusion for immunohistochemical analysis of LPS. The mean SCC in the LPS group was significantly higher, whereas the milk yield was significantly lower than that in the control group after LPS infusion. The mean BLC in the LPS group was significantly lower than in the control group after LPS infusion. Furthermore, milk concentrations of IL-1β, S100A8 and lactoferrin were higher in the LPS group than in the control group after infusion. LPS was detected in the connective tissues and inner alveolar spaces of the mammary glands 24 hr after LPS infusion. We concluded that dexamethasone administration facilitated the translocation of intrauterine-infused LPS to the mammary gland, where it induced an inflammatory response. Therefore, LPS translocated from other organs, such as the uterus, can induce heavy inflammation in the mammary gland under immunosuppressive conditions.  相似文献

In 30 female goats hyperimmunized with bovine insulin, antibody concentrations in serum were determined by radioimmunoassay. Blood samples were taken from the newborn lambs immediately after birth and during the 12 weeks thereafter. Twin lambs were separated and reared in either a colostrum-fed or a colostrum-deprived group. Bovine insulin was added to the milk rations of colostrum-deprived lambs. At the age of 10 weeks, these kids were given a single dose of 1 IU of insulin in combination with complete Freund's adjuvant. Colostrum from nanny goats was also secured for further investigations. Most of the hyperimmunized adult goats responded with the development of insulin antibodies. Their kids were born without these specific immunoglobulins, but several hours after they were allowed to suckle colostrum, the antibody concentration reached values comparable with the values measured in the nanny's blood. Colostrum-deprived kids did not show an increase in binding capacity. The addition of insulin to the milk rations of bottle-fed kids did not act as a primer in antibody production. Immunoglobulins in blood serum of nanny goats and kids and in colostral whey belong mainly to the IgG class. These results indicate that in goats, maternal insulin antibodies are transferred from nanny to offspring only by colostrum.  相似文献

Serum and colostrum samples from goats at parturition and serum samples from their kids at 3 days and at 4, 7, 10 and 12 weeks after birth were examined for the presence of antibodies to Corynebacterium pseudotuberculosis hemolysins. The hemolysis inhibition test (HIT) was used. High correlation was found between titre values of antihemolysins in serum and colostrum of goats at parturition (correlation coefficient r = 0.83; P < 0.01). Intermediate correlation was found between antihemolysin titre in colostrum of goats and in the sera of their kids 3 days old (r = 0.56; 0.01 < P < 0.05). Furthermore, titre values for 3 day-old kids showed high correlation with the antihemolysin titres when the kids aged 4 and 7 weeks (r = 0.76 and 0.85, respectively; P < 0.01). Antihemolysin titres decreased linearly in kids from 3 days to 10 weeks old. Calculated half life of antibodies was 12 days. Most of the kids had detectable antibodies up to the age of 5–6 weeks. None of the kids were seropositive at 2½ months of age.Serum samples collected monthly from a group of kids chosen at random, aged 7–10 months, contained antibodies to hemolysins in half of the animals at the first testing. At the age of 10 months 14 out of 15 kids were seropositive. Thus, most of the kids from this herd were exposed to G. pseudotuberculosis antigens during summer and autumn of their first year of life.Prophylactic measures against caseous lymphadenitis is briefly discussed.  相似文献

Thirty new born male kids were allotted into three groups to evaluate the effects of diet and age on complement system activity in serum. After the colostrum feeding period, the control group (C) received a commercial milk replacer; the CLA group received 20 g/kg milk replacer DM of conjugated linoleic acid; and the GMK group was fed with goat milk. The kids were fed colostrum for 2 days and then either milk replacer or goat milk from days 3 to 60. Blood samples were taken at 1, 10, 20, 30, 40, 50 and 60 days of age. The complement system activity was higher (P < 0.05) in the GMK group throughout the whole experiment through alternative pathway. C kids did not present complement activity at any time. CLA group complement activity was statistically higher than that of C kids from 30 to 60 days, through alternative pathways. In conclusion, the milk replacer formula should be reformulated because it did not induce any complement system activity in the first two months of life.  相似文献

Forty female goats in the third parity were randomly divided into four similar groups. The experiment was started 20 days before mating and lasted until the end of the suckling period for 60 days and weaning their kids. The first group were fed diet without supplementation and kept as control while in the second, third and fourth groups, each doe was fed diets with rumen protected choline (RPC) at the rate of 10, 20 and 40 g/day, respectively. Results showed that number of doe kidding twins and triplets and litter weight of kids born per group increased with increasing the level of RPC in the diet of goats and viability rate of born kids during the suckling period improved due to RPC supplementation in the diets of their mothers. Duration of estrous, days from weaning to estrous, days from kidding to estrous and kidding interval decreased significantly, while conception rate increased due to adding RPC. Milk choline concentrations and total choline secretion though milk were progressively increased significantly with increasing the level of RPC supplementation. Live body weight and daily body gain of their suckling male and female kids at weaning increased significantly with increasing RPC levels in the diets of their mothers.  相似文献

The role of opioid peptides in the secretion of oxytocin (OT) and prolactin (PRL) induced by sucking was studied in goats. Seven goats were isolated with their kids (four singletons and three twins) in individual corrals 3–4 weeks after parturition. On day 1 of the experiment, the kids were separated from the does for 7 h and were weighed before and 15 min after being reunited with their mothers to assess the amount of milk obtained by sucking. The does were blood‐sampled 10 min before and at the end of the sucking period. On day 2, a similar protocol was followed, but naloxone was given immediately after the first blood sample. On day 3, the protocol was repeated but saline vehicle was injected instead of naloxone. On day 5, the naloxone experiment was repeated as on day 2. Milk ejection was evaluated as the difference in the weight of the kids before and after sucking for 15 min, and the maternal serum levels of OT and PRL were measured by radioimmunoassay. A significant decrease in the weight gain of the kids was obtained when the mothers were treated with naloxone on day 2. Consistently, serum levels of OT and PRL induced by sucking were significantly reduced; indicating that sucking‐induced OT secretion for milk ejection in lactating goats is facilitated by opioid peptides. In a second experiment performed in the same animals 10 days later, the administration of OT, immediately after naloxone administration, prevented the decrease in the weight gain induced by naloxone, suggesting that the effect of the opioid antagonist on milk ejection in goats is a result of a reduced OT secretion. The results of this study confirm the importance of sucking‐induced OT secretion for milk ejection in lactating goats, and indicate that OT and PRL secretion are regulated by opioid peptides in this species.  相似文献

Goat kids from a herd endemically infected with caprine arthritis encephalitis (CAE) virus were raised according to 3 methods. One group of ten goat kids was removed from infected does at birth before suckling or licking by the doe could occur (snatch birth technique). Kids were fed on goat colostrum, which had been heated to 57 degrees C for ten minutes and then held in a thermos flask for one hour. Subsequently the kids were fed reconstituted spray dried cows' milk powder. They were raised apart from infected goats with separation maintained by a wire fence. Contact occurred across-the-fence. Passively acquired serum antibody to CAE virus was detected in some kids at two to three months of age. Nine of the ten goats were negative for serum antibody to CAE virus when tested at 5-6, 9 and 12 months of age. One goat was positive at three and nine months of age but was negative when tested at 12 months of age. A second group of four kids was removed at birth and fed heat-treated goat colostrum, followed by milk from CAE virus-infected does. All four kids became infected with CAE virus; they developed serum antibody to CAE virus between 5-6 and 9 months of age. A third group of two kids was not removed from their infected dams. Both kids were infected at 5-6 and 9 months of age.  相似文献

The objective of this study was to compare serum IgG concentrations, blood metabolites indicative of nutritional status, weight gain and mortality rate in goat kids fed a commercial colostral supplement containing immunoglobulins against several pathogen microorganisms, prior to the ingestion of the mother colostrum, and goat kids ingesting natural colostrum only. There was no difference in serum IgG concentrations between 27 kids fed a colostrum supplement (20 g, derived from cow lacteal secretions) prior to the kids’ first meal (658 ± 703 mg dl−1) and 21 kids ingesting maternal colostrum freely (1011 ± 1140 mg dl−1) at 24 hours of birth. Hematocrit values, serum glucose and urea concentrations at 24 hours and 5 days of age were unaffected by treatment. Serum total proteins were 14% higher (P < 0.05) in the unsuplemented group than in the supplemented group at 5 d of age. There was no significant difference between the supplemented and unsupplemented kids in daily weight gain from birth to 70 days of age (92 ± 4.8 vs 102 ± 5.1 g day−1). Mortality was 4% for kids receiving the colostrum supplement as compared with 0.0% for kids ingesting maternal colostrum only. Results suggest that, in intensively managed non-dairy goats with kiddings in summer, the supplementation of this commercial colostrum derived from cow lacteal secretions and containing antibodies against diverse pathogens organisms did not enhanced growth, survival or immunity under the farming conditions of this study.  相似文献

Caprine arthritis-encephalitis virus was isolated from goat milk and transmitted most efficiently to kids through both milk and colostrum. In addition, it appeared that transmissions through other secretions of the doe occurred, but were less important than transmission in milk and colostrum. Intrauterine infection may have occurred in 2 of 32 cesarean-derived goats, but postpartum horizontal transmission could not be ruled out. Transmission by the aerosol route was not demonstrated, and even short-term direct contact between virus-infected bucks and virus-free does during breeding did not result in transmission. Prolonged direct contact for over 12 months between weaned cesarean-derived goats and virus-infected goats was necessary before horizontal transmission could be demonstrated under nondairy conditions. However, when uninfected does were milked with infected does, a high percentage became infected in less than 10 months. Heat inactivation (56 C) reduced approximately 10(5) median tissue culture infective doses of caprine arthritis-encephalitis virus to below titratable levels, and virus was not transmitted to kids fed virus-infected colostrum that had been heated at 56 C for 1 hour. A program of eradication is discussed in which kids are removed from their dams at birth, fed safe sources of colostrum and milk, and isolated from other goats until weaning.  相似文献

The study was performed on 16 Holstein‐Friesian calves divided into two groups of eight animals each. The first group was composed of calves whose mothers did not receive selenium supplements (Se0). The second group consisted of calves whose mothers were administered intramuscular injections of a selenium and vitamin E supplement containing 0.5 of sodium selenite/ml and 50 mg of tocopherol acetate/ml in a single dose of 30 ml (Se30) ml, 10 days before the expected parturition date (10 ± 2 days). The calves were fed 2.5 L of the mother's colostrum administered by stomach tube 2 hr after birth and another 2 L 6 hr after birth. Blood from all calves was collected 7 times from external jugular vein (day 0–before colostrum administration and on the 2nd, 3rd, 4th, 7th, 14th and 21st days of life) for analyses of selenium, ceruloplasmin, transferrin, lactoferrin, immunoglobulin G (IgG) concentrations and gamma‐glutamyl transferase (GGT) and lysozyme activity. Selenium concentration was significantly higher in calves whose mothers received selenium supplements than in the offspring of non‐supplemented cows until 72 hr after birth (p ≤ .05). Lysozyme and GGTP activity and IgG concentration were significantly higher in the S30 group during the entire experiment (p ≤ .05). Supplementation of selenium to the mothers did not influence the ceruloplasmin, lactoferrin and transferrin levels in calves. A single injection of a selenium supplement administered to cows during late pregnancy increases selenium levels in calves and enhances passive transfer from the mother to the offspring.  相似文献

An investigation was conducted to determine if depriving goat kids of colostrum and rearing them away from the herd would prevent transmission of caprine retrovirus infection. Twenty-four newborn goat kids were deprived of colostrum and reared on cow's milk away from their dams from an endemically infected goat herd. Twenty-three colostrum-deprived kids had no evidence of retrovirus infection at birth. One kid had sucked briefly and obtained some colostrum resulting in passive transfer of antibody but it did not develop evidence of infection. Nineteen showed no serological evidence of infection over the 370 days of the study. One colostrum-deprived, segregated goat was subsequently found to be infected and developed arthritis-synovitis. Three had doubtful positive response in one or 2 serological tests during the period but no evidence of infection in leucocyte co-cultures. Cells centrifuged from colostrum of infected goats were co-cultivated with foetal goat synovial membrane cultures. Caprine retrovirus was isolated from cells in the colostrum from the 3 goats examined.  相似文献

Effects of supplementing the basal diets with Mn, Zn and Cu, as sulphate, glycine or methionine salts, on colostrum and milk performance, some blood immunity indices and blood minerals of pre- and post-partum Holstein cows were accessed. Forty cows in different groups received: 1) a diet without supplementary Mn, Zn and Cu (control), 2) a diet containing Mn, Zn and Cu sulphates, 3) a diet containing Mn, Zn and Cu glycine, or 4) a diet containing Mn, Zn and Cu methionine with 10 cows per group from d 60 before calving (dry period) to d 100 of lactation. Dry matter intake (DMI), dry matter digestibility (DMD), colostrum and milk performance, milk somatic cell count (SCC), blood and milk total antioxidant capacity (TAC), immunoglobulin M (IgM) and immunoglobulin A (IgA), and blood Mn, Zn and Cu were determined. Dietary supplementation with Mn, Zn and Cu as methionine, glycine or sulphate salts had positive effects on DMD, DMI, colostrum and milk performance, milk SCC, and blood Mn and Zn. Addition of Mn, Zn and Cu in diets could increase (P < 0.05) blood and milk TAC and blood IgA and IgM in the cows and their new-born calves. There were no differences in DMI, DMD, colostrum and milk yields, milk SCC, blood Mn (except d 50 postpartum), Zn and Cu and TAC (except d 50 postpartum) among the organic and inorganic minerals-supplemented groups (P > 0.05), however, the blood concentrations of IgA (except d 1 postpartum) and IgM in the cows supplemented with organic Mn, Zn and Cu were higher (P < 0.05) than those in the cows receiving the sulphate sources of minerals. Overall, dietary supplementation of Mn, Zn and Cu as methionine, glycine or sulphate salts can improve colostrum and milk performance, blood Zn and Mn and immunity indices in Holstein cows and their new-born calves. Moreover, the organic sources of Mn, Zn and Cu have advantage over the sulphate forms in terms of the blood immunoglobulins.  相似文献

This study was designed to determine whether any relationship exists between exposure to artificial long days, milk yield, maternal plasma insulin‐like growth factor 1 (IGF‐1) levels, and kid growth rate in goats. One group of lactating goats was maintained under naturally decreasing day length (control group; n = 19), while in another one, they were kept under artificial long days (LD group; n = 19). Milk yield was higher in goats from the LD group than that in the control group (P < 0.05). Maternal IGF‐1 levels at day 57 of lactation were higher (P < 0.05) in goats from the LD group than the levels in the control group and were positively correlated with the total milk yields per goat at days 43 and 57 of lactation (r = 0.77 and r = 0.84, respectively; P < 0.01). Daily weight gain at week 4 was higher (P < 0.01) in kids from the LD group than that in kids from the control group and was correlated with total and average IGF‐1 maternal levels (r = 0.60 and r = 0.60, P < 0.05). It was concluded that submitting lactating goats to artificial long days increases milk yield, plasma IGF‐1 maternal levels and the growth rate of the kids.  相似文献

Antibacterial factors act as innate immune components, which respond as soon as bacteria enter a living organism. To prevent and treat mastitis in cattle, understanding the concentrations of these substances inside the udder is important; however, they remain to be studied. In this investigation, the concentration of lingual antimicrobial peptide (LAP), S100 protein (S100A7), lactoferrin (LF), and immunoglobulin antibody were measured in the different fractions of foremilk. Lactating Holstein cows were examined, and 10 foremilk fractions were obtained from sequential samples up to 150 ml. The LAP concentrations in milk samples increased until 25 ml. The LF concentrations increased up to the 10 ml fraction, then stabilized at low level after the 50 ml fraction. For S100A7, some fractions had significantly higher (p < .05) concentrations than the 5 or 10 ml fractions. The IgA antibody concentration increased up to the 5 ml fraction, then after 50 ml fraction showed relatively low concentrations. This investigation determined the concentration patterns of LAP, LF, S100A7, and IgA antibody secreted in milk inside the udders of healthy lactating cows as baseline data. These distinct concentration patterns might indicate various protective responses.  相似文献

Two groups of 6 newborn goat kids were artificially fed colostrum containing antibody to caprine arthritis-encephalitis (CAE) virus, obtained from clinically affected does. Kids in group A were fed the colostrum from birth until 7 days of age, while kids in group B were fed colostrum from 1 to 3 days after birth for 7 days. Kids were fed cow's milk at all other times. Serum antibody resulting from the consumption of colostrum, detected by agar gel immunodiffusion (AGID) tests, lasted for up to 8 weeks in group A, but none was detected in group B. Four kids from each group became infected with CAE virus as demonstrated by the emergence of active immunity and by virus isolation procedures. It appeared that uptake of colostral antibody by group A did not prevent viral transmission, interfere with development of active immunity, or modify the outcome of the CAE virus infection.  相似文献

Three consecutive years of monitoring 248 goats in the same flock, found that the first lactation milk yield was significantly higher in seronegative (578 L) than in seropositive (447 L) animals but this difference disappeared in the subsequent second to fourth lactations. No significant differences were found in the proportions of seronegative and seropositive does in the flock, the percentage of animals culled, the number of offspring, or in the number of cases of udder bacterial infection, irrespective of age.Removal of kids from their dams before suckling and the feeding of pasteurised colostrum resulted in reduced numbers of seropositive animals. Nevertheless, by approximately 24 months of age, 76.9% of these initially seronegative animals were seropositive, a factor that significantly contributed to flock seropositivity. This finding could be attributed to lateral virus transmission from seropositive to seronegative kids because of lack of segregation within the flock.  相似文献

Closantel (CLS) is currently used in programs for the strategic control of gastrointestinal nematodes. CLS is extralabel used in different dairy goat production systems. From available data in dairy cows, it can be concluded that residues of CLS persist in milk. The current work evaluated the concentration profiles of CLS in plasma and milk from lactating orally treated dairy goats to assess the residues pattern in dairy products such as cheese and ricotta. Six (6) female Saanen dairy goats were treated orally with CLS administered at 10 mg/kg. Blood and milk samples were collected between 0 and 36 days post‐treatment. The whole milk production was collected at 1, 4, 7, and 10 days post‐treatment to produce soft cheese and ricotta. CLS concentrations in plasma, milk, cheese, whey, and ricotta were determined by HPLC. The concentrations of CLS measured in plasma were higher than those measured in milk at all sampling times. However, the calculated withdrawal time for CLS in milk was between 39 and 43 days postadministration to dairy goats. CLS residual concentrations in cheese (between 0.93 and 1.8 μg/g) were higher than those measured in the milk used for its production. CLS concentrations in ricotta were sixfold higher than those in the milk and 20‐fold higher than those in the whey used for its production. The persistent and high residual concentrations of CLS in the milk and in the cheese and ricotta should be seriously considered before issuing any recommendation on the extralabel use of CLS in dairy goat farms.  相似文献

A prospective observational cohort study of 361 dairy goat kids was conducted to compare 2 methods of controlling caprine arthritis-encephalitis virus infection under commercial dairy conditions. To compare effectiveness of feeding kids pasteurized milk vs serologic testing and segregation in addition to pasteurized milk feeding, goats were monitored up to the age of 30 months by use of monthly agar gel immunodiffusion testing. Survival analysis methods were used to determine whether age at seroconversion differed between the 2 groups. Significantly lower rates of seroconversion were observed in the segregated group (P < 0.001), compared with the nonsegregated group. Of 193 goats in the pasteurized milk-only group, 146 (75.6%) seroconverted within the 30-month study period, whereas infection was detected in 39 (23.2%) of 168 goats in the test/segregated group. Nonsegregated goats were 3.37 times more likely to seroconvert by 24 months of age, and 70.3% of seroconversions by 24 months of age could be attributed to nonsegregation. For age-specific intervals beyond 180 days of age, 70 to 100% of seroconversions could be attributed to lack of segregation. Cohort life tables for age at seroconversion were reported for each group. Type of colostrum fed, sex, and weaning group (season) were not significantly associated with age at seroconversion. Saanen goats had lower age-specific risk of seroconversion in the nonsegregated group alone and overall. Non-Saanen goats wee 1.5 times more likely to seroconvert than were Saanen goats, when adjusted for a possible confounding effect of weaning group. Results indicate that pasteurized milk feeding and routine test and segregation would be a substantially more effective means of control of the disease in dairy goat herds than would pasteurized milk feeding alone.  相似文献

The objectives were to investigate if partial substitution of a grazing diet with a supplemental high-starch feed during the last 12 d of gestation improved colostrum yield and neonatal activity in goat does grazing semi-arid rangeland. For the first objective, 25 pregnant does were randomly assigned to 1 of 2 treatments: 1) grazing only (control; n = 11), and 2) grazing plus 0.6 kg/d of supplemental corn (as fed), a high starch feed, during the last 12 ± 1.0 d before parturition (SC; n = 14). Colostrum was collected at parturition and again at 1, 3, 6, and 10 h postpartum for yield and composition. In all goats, blood concentrations of glucose and plasma progesterone were determined. Total colostrum yield/10 h was greater (P = 0.002) in the SC does (1,102 ± 144 g) than in control does (405 ± 50 g). From 6 to 10 h postpartum, contents of colostrum protein and solids not fat were less (P ≤ 0.012) whereas lactose contents were greater (P = 0.035) in the SC does than in control does. Concentration of glucose in the blood at parturition was greater (P = 0.037) in the SC does (160 ± 13 mg/dL) than in control does (115 ± 12 mg/dL). A significant decrease (P = 0.001) in plasma progesterone concentrations occurred 1 d before parturition in control does, whereas the decrease (P = 0.008) occurred 3 d before parturition in SC does. For the second objective, 20 does (10 per group) and their single kids were randomly assigned to the same treatments as above for evaluation of dietary treatment on neonatal activity. Kid activity was assessed using videos recorded during the first 90 min after birth. Frequency of low-pitched bleats was greater (P < 0.001) in kids of SC does than in kids of controls. Frequencies and durations of teat seeking from 30 to 90 min after birth and of suckling activity were greater (P ≤ 0.015) in kids of SC does than in kids of controls. Duration of parturition was longer (P = 0.001) in control does (58 ± 10.2 min) than in SC does (21 ± 2.8 min). Finally, percentage of fetal malpresentation was greater (P = 0.036) in control does (33.3%) than in SC does (9.0%). We conclude that partial substitution of the grazing diet with a high-starch feed, corn, during the last 12 d of gestation improved colostrum yield and neonatal activity in goats grazing subtropical semi-arid rangeland.  相似文献
