The frequency and intensity of wildfires are expected to increase in the coming years due to the changing climate, particularly in areas of high net primary production. Wildfires represent severe perturbations to terrestrial ecosystems and may have lasting effects. The objective of this study was to characterize the impacts of wildfire on an ecologically and economically important ecosystem by linking soil properties to shifts in microbial community structure in organic horizon soils. The study was conducted after a severe wildfire burned over 7000 ha of the New Jersey Pinelands, a low nutrient system with a historical incidence of fires. Soil properties in burned and non-burned soils were measured periodically up to two years after the fire occurred, in conjunction with molecular analysis of the soil bacterial, fungal and archaeal communities to determine the extent and duration of the ecosystem responses. The results of our study indicate that the wildfire resulted in significant changes in the soil physical and chemical characteristics in the organic horizon, including declines in soil organic matter, moisture content and total Kjeldahl nitrogen. These changes persisted for up to 25 months post-fire and were linked to shifts in the composition of soil bacterial, fungal and archaeal communities in the organic horizon. Of particular interest is the fact that the bacterial, fungal and archaeal communities in the severely burned soils all changed most dramatically during the first year after fire, changed more slowly during the second year after the fire, and were still distinct from communities in the non-burned soils 25 months post-fire. This slow recovery in soil physical, chemical and biological properties could have long term consequences for the soil ecosystem. These results highlight the importance of relating the response of the soil microbial communities to changing soil properties after a naturally occurring wildfire.  相似文献

Climate warming is projected to increase the frequency and severity of wildfires in boreal forests, and increased wildfire activity may alter the large soil carbon (C) stocks in boreal forests. Changes in boreal soil C stocks that result from increased wildfire activity will be regulated in part by the response of microbial decomposition to fire, but post-fire changes in microbial decomposition are poorly understood. Here, we investigate the response of microbial decomposition to a boreal forest fire in interior Alaska and test the mechanisms that control post-fire changes in microbial decomposition. We used a reciprocal transplant between a recently burned boreal forest stand and a late successional boreal forest stand to test how post-fire changes in abiotic conditions, soil organic matter (SOM) composition, and soil microbial communities influence microbial decomposition. We found that SOM decomposing at the burned site lost 30.9% less mass over two years than SOM decomposing at the unburned site, indicating that post-fire changes in abiotic conditions suppress microbial decomposition. Our results suggest that moisture availability is one abiotic factor that constrains microbial decomposition in recently burned forests. In addition, we observed that burned SOM decomposed more slowly than unburned SOM, but the exact nature of SOM changes in the recently burned stand are unclear. Finally, we found no evidence that post-fire changes in soil microbial community composition significantly affect decomposition. Taken together, our study has demonstrated that boreal forest fires can suppress microbial decomposition due to post-fire changes in abiotic factors and the composition of SOM. Models that predict the consequences of increased wildfires for C storage in boreal forests may increase their predictive power by incorporating the observed negative response of microbial decomposition to boreal wildfires.  相似文献

Plants directly interact with the soil microbial community through litter inputs and root exudates, and these interactions may be particularly important in nutrient poor soils that typically characterize early ecosystem development. However, little is known regarding how plant–microbe interactions may actually drive ecosystem processes in early succession, a perspective this study helps to define. We investigated how soil microbial communities develop and interact with the establishment of the first plants in the recently exposed soils of the Mendenhall Glacier forefield near Juneau, AK, USA. We sampled soils from under two different plant species (alder, Alnus sinuata and spruce, Picea sitchensis) and from unvegetated areas; all samples were collected along a single soil transect that had been exposed for 6 years. The presence or absence of vegetation as well as the type of plant (i.e., alder vs. spruce) structured the soil microbial community. Furthermore, asymbiotic nitrogen (N) fixation rates, which were greater in vegetated soils, correlated with differences in bacterial community composition. Although soil microbial community composition varied with vegetation type, soil nutrient and carbon (C) pools did not correlate with bacterial community composition. Moreover, pH did not significantly vary by vegetation type, yet it was the only soil parameter that correlated with bacterial community composition. Vegetation type explained more of the variation in bacterial community composition than pH, suggesting that plant acidification of soils only partly explains the observed shifts in bacterial communities. Plant specific differences in bacterial community structure may also relate to the chemical composition of litter and root exudates. Our research reveals differences in the bacterial community composition of vegetated soils, and how such differences may promote shifts in fundamental biogeochemical processes, such as rates of asymbiotic N fixation, in early stages of primary succession where low N availability may limit bacterial and plant growth and thus constrain ecosystem development. As such, this suggests that plant–soil microbe interactions in themselves may drive processes that shape the trajectory of primary succession.  相似文献

A number of studies have reported species specific selection of microbial communities in the rhizosphere by plants. It is hypothesised that plants influence microbial community structure in the rhizosphere through rhizodeposition. We examined to what extent the structure of bacterial and fungal communities in the rhizosphere of grasses is determined by the plant species and different soil types. Three grass species were planted in soil from one site, to identify plant-specific influences on rhizosphere microbial communities. To quantify the soil-specific effects on rhizosphere microbial community structure, we planted one grass species (Lolium perenne L.) into soils from three contrasting sites. Rhizosphere, non-rhizosphere (bulk) and control (non-planted) soil samples were collected at regular intervals, to examine the temporal changes in soil microbial communities. Rhizosphere soil samples were collected from both root bases and root tips, to investigate root associated spatial influences. Both fungal and bacterial communities were analysed by terminal restriction fragment length polymorphism (TRFLP). Both bacterial and fungal communities were influenced by the plant growth but there was no evidence for plant species selection of the soil microbial communities in the rhizosphere of the different grass species. For both fungal and bacterial communities, the major determinant of community structure in rhizospheres was soil type. This observation was confirmed by cloning and sequencing analysis of bacterial communities. In control soils, bacterial composition was dominated by Firmicutes and Actinobacteria but in the rhizosphere samples, the majority of bacteria belonged to Proteobacteria and Acidobacteria. Bacterial community compositions of rhizosphere soils from different plants were similar, indicating only a weak influence of plant species on rhizosphere microbial community structure.  相似文献

Plant-soil feedbacks are gaining attention for their ability to determine plant community development. Plant-soil feedback models and research assume that plant-soil interactions occur within days to weeks, yet, little is known about how quickly and to what extent plants change soil community composition. We grew a dominant native plant (Pseudoroegneria spicata) and a dominant non-native plant (Centaurea diffusa) separately in both native- and non-native-cultivated field soils to test if these species could overcome soil legacies and create new soil communities in the short-term. Soil community composition before and after plant growth was assessed in bulk and rhizosphere soils using phospholipid fatty acid analyses. Nematode abundance and mycorrhizal colonization were also measured following plant growth. Field-collected, native-cultivated soils showed greater bacterial, Gram (−), fungal, and arbuscular mycorrhizal PLFA abundance and greater PLFA diversity than field-collected, non-native-cultivated soils. Both plant species grew larger in native- than non-native-cultivated soils, but neither plant affected microbial composition in the bulk or rhizosphere soils after two months. Plants also failed to change nematode abundance or mycorrhizal colonization. Plants, therefore, appear able to create microbial legacies that affect subsequent plant growth, but contrary to common assumptions, the species in this study are likely to require years to create these legacies. Our results are consistent with other studies that demonstrate long-term legacies in soil microbial communities and suggest that the development of plant-soil feedbacks should be viewed in this longer-term context.  相似文献

Afforestation and deforestation are key land-use changes across the world, and are considered to be dominant factors controlling ecosystem functioning and biodiversity. However, the responses of soil microbial communities to these land-use changes are not well understood. Because changes in soil microbial abundance and community structure have consequences for nutrient cycling, C-sequestration and long-term sustainability, we investigated impacts of land-use change, age of stand and soil physico-chemical properties on fungal and bacterial communities and their metabolic activities. This study was carried out at four sites in two geographical locations that were afforested on long-established pastures with Pinus radiata D. Don (pine). Two of the sites were on volcanic soils and two on non-volcanic soils and stand age ranged from 5 to 20 y. Microbial communities were analysed by biochemical (phospho-lipid fatty acids; PLFA) and molecular (multiplex-terminal restriction fragment length polymorphism; M-TRFLP) approaches. Both site and stand age influenced microbial properties, with changes being least detectable in the 5-y-old stand. Land use was a key factor influencing soil metabolic activities as measured by physiological profiling using MicroResp. Pasture soils had higher microbial biomass (P < 0.001), and metabolic activities (P < 0.001), and basal respiration rates were up to 2.8-times higher than in the pine soils. Microbial abundance analysis by PLFA showed that the fungal to bacterial ratio was higher in the pine soils (P < 0.01). Community analysis suggested that soil bacterial communities were more responsive to site (principal component 1; P < 0.001) than to land use (principal component 5; P < 0.001). In contrast, the fungal community was more affected by land-use change (principal component 1; P < 0.001) than by site, although site still had some influence on fungal community structure (principal component 2; P < 0.001). Redundancy analysis also suggested that bacterial and fungal communities responded differently to various soil abiotic properties, land-use change and location of sites. Overall, our results indicate that the change in land use from pasture to P. radiata stands had a direct impact on soil fungal communities but an indirect effect, through its effects on soil abiotic properties, on bacterial communities. Most of the changes in bacterial communities could be explained by altered soil physico-chemical properties associated with afforestation of pastures.  相似文献

Soil microbial communities were examined in a chronosequence of four different land-use treatments at the Konza Prairie Biological Station, Kansas. The time series comprised a conventionally tilled cropland (CTC) developed on former prairie soils, two restored grasslands that were initiated on former agricultural soils in 1998 (RG98) and 1978 (RG78), and an annually burned native tallgrass prairie (BNP), all on similar soil types. In addition, an unburned native tallgrass prairie (UNP) and another grassland restored in 2000 (RG00) on a different soil type were studied to examine the effect of long-term fire exclusion vs. annual burning in native prairie and the influence of soil type on soil microbial communities in restored grasslands. Both 16S rRNA gene clone libraries and phospholipid fatty acid analyses indicated that the structure and composition of bacterial communities in the CTC soil were significantly different from those in prairie soils. Within the time series, soil physicochemical characteristics changed monotonically. However, changes in the microbial communities were not monotonic, and a transitional bacterial community formed during restoration that differed from communities in either the highly disturbed cropland or the undisturbed original prairie. The microbial communities of RG98 and RG00 grasslands were also significantly different even though they were restored at approximately the same time and were managed similarly; a result attributable to the differences in soil type and associated soil chemistry such as pH and Ca. Burning and seasonal effects on soil microbial communities were small. Similarly, changing plot size from 300 m2 to 150 m2 in area caused small differences in the estimates of microbial community structure. In conclusion, microbial community structure and biochemical properties of soil from the tallgrass prairie were strongly impacted by cultivation, and the microbial community was not fully restored even after 30 years.  相似文献

This study was designed to examine whether or not specific tree species (Picea glauca, Picea mariana, Pinus banksiana, Populus tremuloides), their post-fire stand age, or their position in a successional pathway had any significant effect on the functional diversity of associated soil microbial communities in a typical mixed boreal forest ecosystem (Duck Mountain Provincial Forest, Manitoba, Canada). Multivariate analyses designed to identify significant biotic and/or abiotic variables associated with patterns of organic substrate utilization (assessed using the BIOLOG™ System) revealed the overall similarity in substrate utilization by the soil microbial communities. The five clusters identified differed mainly by their substrate-utilization value rather than by specific substrate utilization. Variability in community functional diversity was not strongly associated to tree species or post-fire stand age; however, redundancy analysis indicated a stronger association between substrate utilization and successional pathway and soil pH. For example, microbial communities associated with the relatively high pH soils of the P. tremuloides-P. glauca successional pathway, exhibited a greater degree of substrate utilization than those associated with the P. banksiana-P. mariana successional pathway and more acidic soils. Differences in functional diversity specific to tree species were not observed and this may have reflected the mixed nature of the forest stands and of their heterogeneous forest floor. In a densely treed, mixed boreal forest ecosystem, great overlap in tree and understory species occur making it difficult to assign a definitive microbial community to any particular tree species. The presence of P. tremuloides in all stand types and post fire stand ages has probably contributed to the large amount of overlap in utilization profiles among soil samples.  相似文献

The present study investigates the impact of fire (low and high severity) on soil fungal abundance and microbial efficiency in C assimilation and mineralisation in a Mediterranean maquis area of Southern Italy over 2 years after fire. In burned and control soils total and active fungal mycelium, microbial biomass C, percentage of microbial biomass C present as fungal C, metabolic quotient (qCO2) and coefficient of endogenous mineralisation (CEM) were assayed together with several chemical properties of soil (i.e. pH and contents of organic C, total and mineral N, available K, Mg, Mn and water). Fire significantly decreased the fungal mycelium, whereas it stimulated microbial growth probably through the enhancement of bacterial growth because of the increase in organic C and nutrient contents in burned plots. This shift in microbial community composition might explain the observed reduction in soil microbial efficiency of C assimilation (high qCO2) and the increase in C mineralisation rate (CEM) in the first 84 days after fire. Therefore, fire might increase CO2 input to the atmosphere not only during combustion phase but also in the post-fire period.  相似文献

As a result of many decades of fire suppression and atmospheric deposition the deciduous forests of eastern North America have changed significantly in stem density, basal area, tree size-frequency distribution, and community structure. Consequently, soil organic matter quality and quantity, nutrient availability, and microbial activity have likely been altered. This study evaluated the effects of four alternative forest ecosystem restoration strategies on soil microbial activity, microbial functional diversity, soil organic C, and soil N status in two mixed-oak (Quercus spp.) forests in southern Ohio, USA. The soils of these forests were sampled during the fourth growing season after application of (1) prescribed fire, (2) thinning of the understory and midstory to pre-settlement characteristics, (3) the combination of fire and thinning, and (4) an untreated control. Prescribed fire, with or without thinning, resulted in increased bacterial but not fungal activity when assessed using Biolog®. In contrast, assays of acid phosphatase and phenol oxidase activity indicated greater microbial activity in the thinning treatment than in the other three treatments. Functional diversity of both bacteria and fungi was affected by restoration treatment, with the bacterial and fungal assemblages present in the thin + burn sites and the fungal assemblage present in the thinned sites differing significantly from those of the control and burned sites. Treatments did not result in significant differences in soil organic C content among experimental sites; however, the soil C:N ratio was significantly greater in thinned sites than in sites given the other three treatments. Similarly, there were no significant differences in dissolve inorganic N, dissolved organic N, or microbial biomass N among treatments. Bacterial and fungal functional diversity was altered significantly. Based on Biolog® utilization treatments the bacterial assemblage in the thin-only treatment appeared to be relatively N-limited and the fungal assemblage relatively C-limited, whereas in the thin + burn treatment this was reversed. Although effects of restoration treatments on soil organic matter and overall microbial activity may not persist through the fourth post-treatment year, effects on microbial functional diversity are persistent.  相似文献

Most wildfires, even the most severe, burn at mixed intensities across a landscape, depending on local fuel loads, fuel moistures, and wind strength and direction. This heterogeneous patchwork of fire effects can influence the patterns of above- and belowground biotic recovery through altered environmental conditions, nutrient availability, and biotic sources for microbial and vegetative re-colonization. We quantified the effects of low- and high-severity fire 14 months post-burn on key environmental variables typically limiting to microbial activity. We characterized the soil microbial community structure through ester-linked fatty acid analysis (EL-FAME) and identified the soil environmental factors that best explain the pattern of microbial community profiles through canonical correspondence analysis (CCA). Low-severity burning caused no change in soil moisture, pH or temperature while high-severity burning caused an increase in soil moisture, temperature, and a decrease in pH levels, relative to the unburned sites. Soil respiration rates were significantly lower in both the low- and high-severity burn sites, relative to unburned sites, likely due to initial root and microbial death. Overall microbial biomass did not change with either low- or high-severity burning, but the microbial community ordination biplots showed separation of communities by fire, and slight separation by fire severity along three axes. This community separation was driven primarily by a decrease in fungal biomarkers (18:2ω6c, 18:3ω6c) with both low- and high-severity fire. Only 23% of the variation in the microbial community distribution could be explained by three environmental variables: soil pH, temperature, and carbon. These results suggest that the microbial communities in both the low- and high-severity burn sites are structurally different from the populations in the unburned sites.  相似文献

The state of microbial communities in gray-humus soils (Eutric Fluvic Arenosols (Ochric)) of pine stands in the city of Tolyatti after forest fires of 2010 is analyzed. It is shown that fires exert negative effects on the structure and metabolic activity of microbial communities in the postpyrogenic soils. The content of the carbon of microbial biomass and the intensity of microbial respiration in the upper organic horizons of the post-fire plots decrease by 6.5 and 3.4 times, respectively, in comparison with those in the soils of background plots. However, the fire has not affected the studied microbiological parameters of the soils at the depths of more than 10 cm. The maximum content of the carbon of microbial biomass carbon and the maximum intensity of microbial respiration have been found in the subsurface AY2 and АС horizons two–three years the fire. An increase in the microbial metabolic quotient (the ratio of soil respiration to microbial biomass) attests to the disturbance of the ecophysiological state of soil microbial communities after the pyrogenic impact.  相似文献

The daily dynamics of the number of copiotrophic and oligotrophic bacteria (in colony-forming units) and CO2 emissions from cultivated soils after short- and long-term disturbances were studied for 25–27 days in a microfield experiment. The relationship of the wavelike fluctuations of the bacterial number and CO2 emission with the succession of the soil microbial community was determined by the polymerase chain reaction method—denaturing gradient gel electrophoresis (PCR-DGGE). Short-term disturbances involved the application of organic or mineral fertilizers, pesticides, and plant residues to the soils of different plots. The long-term effect was a result of using biological and intensive farming systems for three years. The short-term disturbances resulted in increased peaks of the bacterial number, the significance of which was confirmed by harmonics analysis. The daily dynamics of the structure of the soil microbial community, which was studied for 27 days by the DGGE method, also had an oscillatory pattern. Statistical processing of the data (principal components analysis, harmonics and cross-correlation analyses) has revealed significant fluctuations in the structure of microbial communities coinciding with those of the bacterial populations. The structure of the microbial community changed within each peak of the dynamics of the bacterial number (but not from peak to peak), pointing to the cyclical character of the short-term succession. The long-term effects resulted in a less intense response of the microbiota—a lower rate of CO2 emission from the soil cultivated according to the organic farming system.  相似文献

Organism succession during ecosystem development has been researched for aboveground plant communities, however, the associated patterns of change in below-ground microbial communities are less described. In 2008, a study was initiated along a developmental sand-dune soil chronosequence bordering northern Lake Michigan near Wilderness Park (WP). It was hypothesized that soil bacterial communities would follow a pattern of change that is associated with soil, plant, and ecosystem development. This study included 5 replicate sites along 9 soils (n = 45) ranging in age from ∼105 to 4010 years since deposition. Soil bacterial community composition and diversity were studied using bacterial tag-encoded FLX amplicon pyrosequencing of the 16S rRNA gene. Bray–Curtis ordination indicated that bacterial community assembly changed along the developmental soil and plant gradient. The changes were not affected by seasonal differences, despite likely differences in plant root C (e.g. exudates), temperature, and water availability in soil. Soil base cations (Ca, Mg) and pH declined, showing log-linear correlations with soil age (r ∼ 0.83, 0.84 and 0.81; P < 0.01). Bacterial diversity (Simpson's 1/D) declined rapidly during the initial stages of soil development (∼105–450 y) and thereafter (>450 y) did not change. Turnover of plant taxa was also more rapid early during ecosystem development and correlated with bacterial community structural change (P < 0.000001; r = 0.56). It is hypothesized that plants help to drive pedogenic change during early (<450 y) soil development (e.g. pH decline, cation leaching) which drive selection of soil bacterial communities. In mature soils (∼450–4000 y), resilient and stable soil bacterial community structures developed, mimicking steady-state climax communities that were observed during latter stages of primary plant succession. These relationships point to possible feedbacks between plant and bacterial communities during ecosystem development.  相似文献

Tree thinning and harvesting produces large amounts of slash material which are typically disposed of by burning, often resulting in severe soil heating. We measured soil chemical properties and microbial community structure and function over time to determine effects of slash pile burning in a ponderosa pine forest soil. Real time data were collected for soil temperature, heat flux, and soil moisture contents in one of two slash piles burned in April 2004. During the burn, soil temperatures reached 300 °C beneath the pile center and 175 °C beneath the pile edge. Slash pile burning increased soil pH, extractable N and P, and decreased total C levels within the first 15 cm of soil. Burning reduced soil bacterial biovolumes within the first 15 cm of soil and fungal biovolumes within the first 5 cm of soil. One month after the burn, soil microbial communities under the pile center were enriched in Gram-positive bacterial fatty acid markers compared to communities from under the pile edge and control (nonburned) soil. Fifteen months later, soil chemical properties had not returned to background levels, and microbial community structure in fire-affected soil, regardless of pile location, was distinct from communities of control soil. In fire-affected soil, concentrations of fungal fatty acid biomarkers were low and arbuscular mycorrhizal fungal biomarkers were absent, regardless of pile location. Slash pile burning also reduced fungal and bacterial respiration and resulted in large fluctuations in microbial potential N mineralization and immobilization activities. By altering soil properties important to soil conservation and plant reestablishment, slash pile burning negatively impacts forest ecosystems at localized scales.  相似文献

Purpose

There have been a number of studies on the succession of vegetation; however, the succession of soil microbes and the collaborative relationships between microbes and vegetation during land restoration remain poorly understood. The objectives of this study were to characterize soil microbial succession and to explore the collaborative mechanisms between microbes and vegetation during the restoration of abandoned land through quantitative ecology methods.

Materials and methods

The present research was carried out in the succession of a 5-year abandoned land and its conversion to Hippophae rhamnoides shrubs, Larix principis-rupprechtii plantation, and a naturally regenerated forest (mixed forest). Soil bacterial, archaeal and fungal characteristics were tested by real-time quantitative PCR assays and terminal restriction fragment length polymorphism. The richness, diversity, and evenness indices were employed to analyze plant and microbial communities’ structure. The stability of plant and microbial communities was tested using Spearman’s rank correlation. The relationships between the regeneration scenarios and environmental factors were determined through canonical correspondence analysis.

Results and discussion

The aboveground biomass was significantly different among the sites. Soil bacterial, archaeal, and fungal rRNA gene abundances did not increase significantly with increasing soil organic carbon content. There were higher correlation coefficients between plant and total microbial communities on the richness, diversity, and evenness indices and ratios of positive to negative association compared to ones between plant and individual bacteria, archaea, and fungi. Soil bulk density, clay, pH, and litter were the primary significant environmental factors affecting the structure of plant and microbial communities. The positive relationships between plant and soil bacteria, fungi, and total microbe communities, as well as the negative relationships between plant and archaea, were demonstrated.

Conclusions

The results suggested that plants promote the growth of soil bacteria and fungi during the process of community succession on a small scale; however, plants inhibit the growth of soil archaea.  相似文献

The ecosystem response to wildfire is often linked to fire severity, with potentially large consequences for belowground biogeochemistry and microbial processes. While the impacts of wildfire on belowground processes are generally well documented, it remains unclear how fire affects the fine-scale composition of microbial communities. Here, we investigate the composition of soil bacterial and fungal communities in burned and unburned forests in an attempt to better understand how these diverse communities respond to wildfire. We explored the belowground responses to three wildfires in Linville Gorge, NC, USA. Wildfires generally increased soil carbon content while simultaneously reducing soil respiration. We employed amplicon sequencing to describe soil microbial communities and found that fires decreased both bacterial and fungal diversity. In addition, wildfires resulted in significant shifts in both bacterial and fungal community composition. Bacterial phylum-level distributions in response to fire were mixed without clear patterns, with members of Acidobacteria being representative of both burned and unburned sites. Fungal communities showed consistent increases in Ascomycota dominance and concurrent decreases in Basidiomycota and Zygomycota dominance in response to burning. Indicator species analysis confirmed shift to Ascomycota in burned sites. These shifts in microbial communities may reflect differences in the quality and quantity of soil organic matter following wildfires.  相似文献

Several biochemical and molecular methods are used to investigate the microbial diversity and changes in microbial community structure in rhizospheres and bulk soils resulting from changes in management. We have compared the effects of plants on the microbial community, using several methods, in three different types of soils. Pots containing soil from three contrasting sites were planted with Lolium perenne (rye grass). Physiological (Biolog), biochemical (PLFA) and molecular (DGGE and TRFLP) fingerprinting methods were employed to study the change in soil microbial communities caused by the growth of rye grass. Different methods of DNA extraction and nested PCR on TRFLP profiles were examined to investigate whether they gave different views of community structure. Molecular methods were used for both fungal and bacterial diversity. Principal component analysis of Biolog data suggested a significant effect of the plants on the microbial community structure. We found significant effects of both soil type and plants on microbial communities in PLFA data. Data from TRFLP of soil bacterial communities showed large effects of soil type and smaller but significant effects of plants. Effects of plant growth on soil fungal communities were measured by TRFLP and DGGE. Multiple Procrustes analysis suggested that both methods gave similar results, with only soil types having a significant effect on fungal communities. However, TRFLP was more discriminatory as it generated more ribotype fragments for each sample than the number of bands detected by DGGE. Neither methods of DNA extraction nor the nested PCR had any effect on the evaluation of soil microbial community structure. In conclusion, the different methods of microbial fingerprinting gave qualitatively similar results when samples were processed consistently and compatible statistical methods used. However, the molecular methods were more discriminatory than the physiological and biochemical approaches. We believe results obtained from this experiment will have a major impact on soil microbial ecology in general and rhizosphere–microbial interaction studies in particular, as we showed that the different fingerprinting methods for microbial communities gave qualitatively similar results.  相似文献

There is an increasing concern that the continuous use of chemical fertilizers might lead to harmful effects on soil ecosystem. Accordingly, a biocompatible approach involving inoculation of beneficial microorganisms is presented to promote plant growth and simultaneously minimize the negative effect of chemical fertilizers. In this study, Rhodopseudomonas palustris, a plant growth-promoting rhizobacterium (PGPR), was inoculated into both fertilized and unfertilized soils to assess its influence on Stevia rebaudiana plant growth and microbial community in rhizosphere soils in a 122-d field experiment. Soil enzyme assays (dehydrogenase, urease, invertase, and phosphomonoesterase), real-time quantitative polymerase chain reaction (RT-qPCR), and a high-throughput sequencing technique were employed to determine the microbial activity and characterize the bacterial community. Results showed that the R. palustris inoculation did not significantly influence Stevia yields and root biomass in either the fertilized or unfertilized soil. Chemical fertilization had strong negative effects on soil bacterial community properties, especially on dehydrogenase and urease activities. However, R. palustris inoculation counteracted the effect of chemical fertilizer on dehydrogenase and urease activities, and increased the abundances of some bacterial lineages (including Bacteroidia, Nitrospirae, Planctomycetacia, Myxococcales, and Legionellales). In contrast, inoculation into the unfertilized soil did not significantly change the soil enzyme activities or the soil bacterial community structure. For both the fertilized and unfertilized soils, R. palustris inoculation decreased the relative abundances of some bacterial lineages possessing photosynthetic ability, such as Cyanobacteria, Rhodobacter, Sphingomonadales, and Burkholderiales. Taken together, our observations stress the potential utilization of R. palustris as PGPR in agriculture, which might further ameliorate the soil microbial properties in the long run.  相似文献

A field study was carried out to analyze the short-term (2 years) effect of tillage and crop rotation on microbial community structure and enzyme activities of a clay loam soil. The experimental design was a split-plot arrangement of treatments, consisting of two tillage treatments—ridge tillage (RT) and no-tillage (NT)—in combination with two crop rotation treatments—corn (Zea mays L.) monoculture and a 2-year corn-soybean (Glycine max L.) rotation. Phospholipid fatty acid (PLFA) profiles were used to assess soil microbial community structure. No-tillage resulted in significantly higher total PLFAs compared to the RT treatment, which was accompanied by higher activities of protease, β-glucosaminidase, and β-glucosidase. This suggests a close link between soil microbial communities and enzyme activities in response to tillage. The increase of total microbial lipid biomass in the NT soils was due to the increase in both fungal and bacterial PLFAs. Crop rotation had little effect on soil bacterial communities and enzyme activities, but it significantly influenced soil fungal communities, particularly arbuscular mycorrhizal fungi. Soils under monoculture corn had higher fungal biomass than soils under corn-soybean rotation regardless of tillage treatment.  相似文献
