本文概述了近年鱼类生物技术在中国之发展。在倍数性育种方面,已获得了至少6种同源多倍体和2种异源多倍体;将诱导雌核发育和人工性逆转技术相结合,建立了一套快速获得鱼类自交系的方法。在核移植方面,获得了核、质分别来源于不同种、属和亚科的移核鱼,并成功地以经短期培养的鲫鱼肾细胞作核供体,获得了1尾移核鱼,从而证明了鱼类体细胞仍具发育全能性。用电场诱导鱼类未受精卵和囊胚细胞间,用微束激光诱导受精卵间的融合,均已获得鱼苗。在基因转移方面,围绕外源基因在鱼类的整合、转录、表达及其生物学效应等方面,进行了一系列的研究。所采用的转基因方法包括显微注射、电穿孔及精子载体法等。在分离有价值鱼类基因方面,已获得了3种生长激素基因,2种肌动蛋白基因,1个泌乳素cDNA和1个抗冻蛋白的cDNA。  相似文献

Anterior pituitaries from the dwarf mouse strain "little" did not release growth hormone or accumulate adenosine 3',5'-monophosphate (cyclic AMP) in response to human and rat growth hormone-releasing factor (GRF). Dibutyryl cyclic AMP, as well as the adenylate cyclase stimulators forskolin and cholera toxin, markedly stimulated growth hormone (GH) release. The basis of the GH deficiency in the little mouse may therefore be a defect in an early stage of GRF-stimulated GH release related either to receptor binding or to the function of the hormone-receptor complex.  相似文献

Analogs of adenosine 3',5'-monophosphate (cyclic AMP) inhibit the growth of cultured cell lines. The effects of 8-bromo- and N6-butyryl-substituted analogs of cyclic and noncyclic AMP on six cell lines were examined and were equally inhibitory. Variant cell lines with altered cyclic AMP-dependent protein kinase were more resistant to both cyclic and noncyclic nucleotides. We conclude that growth inhibition by analogs of cyclic AMP (i) does not require a 3',5' phosphodiester bond and (ii) may be mediated by a pathway involving endogenous cyclic AMP.  相似文献

 真核生物基因表达的调控是目前分子生物学研究中重要的前沿领域,形成了多个热点。真核基因表达调控是一个十分复杂而协调有序的调控过程,这一过程不仅与基因本身的功能,也与细胞及机体的功能表现密切相关。而转录水平的调控是基因表达过程中最重要的第一步,由于蛋白质-蛋白质、蛋白质-DNA之间的相互作用,以及一些复杂大分子复合物的形成导致真核生物的转录水平的调控是一个多级的复杂过程。近年来,随着新技术和新方法的出现,发现了许多与基因转录调控有关的DNA顺式作用元件、核蛋白因子及各种因子在核内形成的多种复合物,它们的相互作用使转录调节的效率得到了提高,也使人们更进一步认识了某些生命现象以及细胞行为和疾病的发生机理。本文从顺式作用元件、反式作用因子、转录复合物、激素的调节、协调作用及最新研究siRNA调控6个方面进行了阐述,同时也对目前转录调控存在的问题和前景做了分析。  相似文献

生长调控因子(growth regulation factor,GRF)是植物中特有的一类转录因子,在植物激素调控、生长发育以及胁迫应答等多个领域扮演重要角色。对高粱GRF基因家族成员进行筛选鉴定,得到8个家族基因,分析了这些基因的理化性质、保守蛋白序列、基因功能及其共线性,分析了这些基因在非生物胁迫下转录组表达差异。结果表明,GRF基因家族结构较为保守,启动子元件含有干旱、低温、茉莉酸等响应元件,推测该家族基因参与生物及非生物胁迫应答响应。通过盐胁迫和烯效唑处理下的转录组数据分析,得到一个与耐盐相关的基因(SORBI_3002G297800)和一个与激素相关的基因(SORBI_3006G203400),为进一步探究这些基因在高粱应对非生物胁迫中的作用提供了重要参考。  相似文献

Concentrations of prolactin and growth hormone in the serum of rats were significantly increased by morphine. Dose response studies demonstrated that maximum prolactin release required lower doses of morphine than those needed for the maximum growth hormone response. Selective blockade of mu 1 (high affinity) opiate receptor with the irreversible antagonist naloxazone reduced morphine-induced peak concentrations of prolactin by 80 percent while increasing peak growth hormone levels by 250 percent. These results suggest different receptor mechanisms for the opiate modulation of the two hormones. The mu 1 (high affinity) receptor sites appear to mediate the morphine-induced release of prolactin but not growth hormone.  相似文献

Immunocytochemistry shows that early during phagocytosis of zymosan, adenosine 3',5'-monophosphate (cyclic AMP) appears on the cell surface before the phagosome is internalized. The appearance of cyclic AMP on the cell surface is coincident with that of granule products and regulatory subunit of type I cyclic AMP-dependent protein kinase. Guanosine 3',5'-monophosphate is not associated with the initiation site of phagocytosis, but is observed throughout the granular cytoplasmic region. This sharply localized accumulation of cyclic AMP may serve as a signal for the initiation of phagocytosis.  相似文献

When guanosine 3',5'-monophosphate (cyclic GMP) and adenosine 3',5'-monophosphate (cyclic AMP) are localized in canine thyroid by a flurescence Immunocytochemical procedure, distinct staining patterns for each nucleotide are seen: Cyclic AMP is distributed throughout the follicular cell cytoplasm before and after administration of thyroid-stimulating hormone, while cyclic GMP is localized to the follicular cell mumbrane in the control state, and increased cytoplasmic fluorescence is visualized after acetylcholine. These data provide histological evidence that correlates with cyclic nucleotide tissue measurements, sugesting diverse roles of the two nucleotides in thyroid function.  相似文献

When two different mammalian cell types are fused to generate a stable hybrid cell line, genes that are active in only one of the parents are frequently shut off, a phenomenon called extinction. In this study two distinct, complementary mechanisms for such extinction of growth hormone gene expression were identified. In hybrids formed by fusing fibroblasts to pituitary cells, pituitary-specific proteins that bind to the growth hormone promoter were absent. In addition, a negative regulatory element located near the rat growth hormone promoter was specifically activated.  相似文献

The growth in vitro of human breast cancer cells, line MCF-7, was inhibited by a daily supplement of L-arginine (1 milligram per milliliter). Arginine acted synergistically with dibutyryl adenosine 3',5'-monophosphate (cyclic AMP) (10(-6) molar) to enhance the growth inhibitory effect: the cell replication ceased completely within 2 days after treatment. The growth arrest accompanied a change in cell morphology and was preceded by increases in the cellular concentration of cyclic AMP, adenylate cyclase, and type II cyclic AMP-dependent protein kinase activities as well as a decrease of estrogen binding activity. The results suggest that growth of human breast cancer cells is subject to cyclic AMP-mediated regulation and that arginine may play a specific role in this process.  相似文献
