Staff members of the Department of Botany of Palacký University in Olomouc and Gene Bank Department – Workplace Olomouc, Research Institute of Crop Production in Prague, Czech Republic, conducted an expedition in seven European countries (Austria, Czech Republic, France, Germany, Italy, the Netherlands, Switzerland) in August/September 1999 to collect wild Lactuca spp. germplasm and study its geographic distribution, ecology and biodiversity. During the mission, more than 600 locations were visited resulting in the collection of 602 seed samples (accessions) of wild Lactuca species and 13 seed samples of related genera (Chondrilla and Mycelis). Lactuca serriola f. serriola, L. serriola f. integrifolia, L. saligna and L. viminea subsp. chondrilliflora were prevalent in southern Europe (Italy, France), however, only L. serriola was common in central and western Europe (Austria, Czech Republic, Germany, Netherlands, Switzerland). The greatest diversity of Lactuca species was found in France, where also the most seed samples (165) were collected. The most characteristic habitats with a high density of Lactuca spp. populations were observed along roads and highways, grassy ditches, ruderal communities, and dust-heaps. Natural infections by powdery mildew (Erysiphe cichoracearum) and downy mildew (Bremia lactucae) on some wild Lactuca spp. were observed. Recent observations concerning the geographic distribution, population structure, habitats, and natural occurrence of diseases of Lactuca spp. are discussed. This assemblage of genetic resources of Lactuca spp. can serve as the basis of future studies of species diversification, spatial population structure, plant microevolution, domestication processes, and genetic variability of host-parasite interactions.  相似文献

Lettuce (Lactuca sativa L.) belongs to the genus Lactuca L. and is an important vegetable worldwide. Over the past decades, there have been many controversies about the phylogeny of Lactuca species due to their complex and diverse morphological characters and insufficient molecular sampling. In this study we provide the most extensive molecular phylogenetic reconstruction of Lactuca, including African wild species, using two chloroplast genes (ndhF and trnL-F). The sampling covers nearly 40 % of the total endemic African Lactuca species and 34 % of the total Lactuca species. DNA sequences from all the subfamilies of Asteraceae in Genebank and those generated from Lactuca herbarium samples were used to establish the affiliation of Lactuca within Asteracaeae. Based on the subfamily tree, we selected 33 ndhF sequences from 30 species and 79 trnL-F sequences from 48 species to infer relationships within the genus Lactuca using randomized axelerated maximum likelihood and Bayesian inference analyses. Biogeographical, chromosomal and morphological character states were reconstructed over the Bayesian tree topology. We conclude that Lactuca contains two distinct phylogenetic clades—the crop clade and the Pterocypsela clade. Other North American, Asian and widespread species either form smaller clades or mix with the Melanoseris species. The newly sampled African endemic species probably should be treated as a new genus.  相似文献

During 2002, 2004, 2006 and 2008, trips were undertaken in the United States and Canada by members of the Department of Botany, Palacky University in Olomouc (Czech Republic) to record distributions and sample the diversity of wild and weedy Lactuca species. In that period, 16 states in the USA (Arizona, California, Colorado, Idaho, Iowa, Minnesota, Montana, Nevada, New York, North Carolina, Oregon, South Dakota, Utah, Washington, Wisconsin, Wyoming) and two provinces in Canada (Ontario, Quebec) were visited. Seven wild and weedy Lactuca species (L. serriola, L. saligna, L. virosa, L. canadensis, L. biennis, L. floridana, L. ludoviciana), an interspecific hybrid (L. canadensis?×?L. ludoviciana), and an undetermined Lactuca species were recorded, and 343 seed samples were collected from 200 locations. The largest number of wild Lactuca species that we observed was recorded in Iowa, and the largest number of samples was collected in California. The most common habitats of weedy species (L. serriola, L. saligna and L. virosa) were along transport corridors, such as roadsides, road ditches, parking sites and petrol stations, grassy slopes and ruderal places. The most frequent species was L. serriola, an Old World introduction, which we observed in all states and provinces visited, except for New York, and in broad range of elevations (up to 2,358?m a.s.l.). Lactuca saligna was recorded only one time in Salinas, California. Lactuca virosa was found repeatedly in Washington State along the road to the Mt. St. Helens and in Redwoods, California. Native North American taxa (L. canadensis, L. floridana, L. ludoviciana) were recorded only in Iowa. Lactuca biennis was found only in Canada in the southern part of Quebec. Records of diseases and pests showed only occasional occurrences of downy mildew (Bremia lactucae) and powdery mildew (Golovinomyces cichoracearum). Most plant-disease findings were made in Canada. Results of these efforts enrich our knowledge of the distribution, ecogeography and ecobiology of Lactuca species occurring spontaneously in North America. Seed collections (accessions) will be conserved in the working collection of the genebank at Palacky University to support future research in ecogeography, ecobiology, resistance and genetic polymorphisms, and to enrich of germplasm diversity available for lettuce breeding.  相似文献

Fifty accessions of 25 Lactuca species,L. serriola ×L. sativa and Mycelismuralis were analyzed for chromosome number and relative DNA amountvariation. In the majority of Lactuca species studiedchromosome counts, as earlier reported (n = 8, 9, 17), were verified; however,for L. dregeana andL. homblei (probablyL. schweinfurthii orL. longespicata) the chromosome number(n = 9) was determined for the first time. Relative nuclear DNA content,estimated by using flow cytometry (DAPI staining), showed that 2C DNA contentranged from 2.02 pg in L.capensis to 17.96 pg inL. canadensis. Statistical and clusteranalysis of data based on relative nuclear DNA contents correspond fairly wellwith recently accepted taxonomic classification of the genusLactuca. However, the position of certain species as wellas clarification of taxonomic determinations of someLactuca accessions needs further examination.  相似文献

Sample populations of 157 Cannabis accessions of diverse geographic origin were surveyed for allozyme variation at 17 gene loci. The frequencies of 52 alleles were subjected to principal components analysis. A scatter plot revealed two major groups of accessions. The sativa gene pool includes fiber/seed landraces from Europe, Asia Minor, and Central Asia, and ruderal populations from Eastern Europe. The indica gene pool includes fiber/seed landraces from eastern Asia, narrow-leafleted drug strains from southern Asia, Africa, and Latin America, wide-leafleted drug strains from Afghanistan and Pakistan, and feral populations from India and Nepal. A third putative gene pool includes ruderal populations from Central Asia. None of the previous taxonomic concepts that were tested adequately circumscribe the sativa and indica gene pools. A polytypic concept of Cannabis is proposed, which recognizes three species, C. sativa, C. indica and C. ruderalis, and seven putative taxa.  相似文献

Patterns of variation for the solanidine-based glycoalkaloids have been determined for tuber material of genebank accessions of landraces of the cultivated potato and three closely-related wild species. Total levels were low in the cultivated taxa investigated,S. phureja, S. stenotomum, and the tetraploidS. tuberosum in its two formsS. tuberosum ssp.tuberosum andS. tuberosum ssp.andigena. The only solanidine-based glycolkaloids found in the tubers investigated in this study were-solanine and-chaconine, with one additional related compound, dehydrocommersonine, found in tubers of an accession ofS. canasense. The ratio of chaconine : solanine differed markedly from a mean of 0.98 inS. phureja to 2.28 in ChileanS. tuberosum ssp.tuberosum. The chaconine : solanine ratio was relatively constant within these two taxa, reflecting their relatively narrow genetic bases, but was much more variable within the taxa regarded as ancestral,S. stenotomum andS. tuberosum ssp.andigena. The wild speciesS. sparsipilum has been implicated in the origin of the tetraploid potato,S. tuberosum, but there was no support for this from the patterns of variation in chaconine : solanine ratio. The cline of chaconine : solanine ratios in cultivated potatoes along the spine of S. America does not appear to relate to the influence of local wild species through introgression, but may instead reflect differing selective pressures acting in different parts of the native range of the cultivated potato.  相似文献

Summary Isozyme analyses of genetic diversity in Vigna unguiculata were performed to determine genetic relationships and level of genetic diversity between wild and cultivated cowpea. Thirty-four cultivated accessions of V. unguiculata, 56 wild accessions of V. unguiculata, and six accessions representing five related wild Vigna species were analyzed. Ten enzyme systems were polymorphic within Vigna unguiculata: AAT, ACO, G6PDH, DIAP, LAP, MUE, ME MDH, PRX, and SOD. Fourteen of 24 putative loci (58%) were polymorphic within wild V. unguiculata, but only one locus (4%) was polymorphic within cultivated cowpea; when five related Vigna species were examined, 21 of the 24 bands of activity showed polymorphisms (88%) adding 33 alleles to the 48 identified within V. unguiculata. In one F2 population of 68 plants (UCDVg 36 × UCDVg 21) a loose linkage was indicated between Diap-2 and G6pd-1 (2 = 15.39; p = 0.004) with an estimated distance of 36.0 cM ± 5.02 (recombination (r) = 0.31). Also in another F2 population of 38 plants (CB 88 × UCDVg 21) a loose linkage was indicated between Lap-1 and Prx (\gC2 = 9.62; p = 0.047) with an estimated distance of 39.8 cM ± 7.0 (r = 0.33). Total genetic diversity (HT) was 0.085 over all of the accessions including the one classified as V. nervosa. Within accession diversity (Hs) approached zero and between accession diversity (Dsi) was responsible for all of the genetic diversity present. Therefore the coefficient of gene differentiation (GST = DSTIIT) approached 1. Absolute gene differentiation (Dm) was 0.087. Two of the nine segregations in this study were skewed. Lentil is one of the important cool-season food legumes grown in many countries in the Mediterranean region. But a substantial yield loss is observed every year due to various biotic stresses. The Sitona weevil (Sitona crinitus Herbst) is a major insect pest limiting lentil productivity mainly in the countries of West Asia and North Africa region. The adult insects feed on the leaflets at seedling stage, and the plant suffers due to reduced photosynthesis. The larvae feed on the root systems and on the nodules, thus decreasing the ability of the plant to fix atmospheric nitrogen. Since sources of resistance to this pest in the cultivated lentil Lens culinaris Medikus subsp. culinaris are lacking, we searched for resistant sources in a collection of wild Lens species available in the ICARDA Gene Bank. We screened 315 accessions of wild lentil covering all known species/sub-species based on nodule damage at ICARDA’s main experimental station (Tel Hadya, Aleppo), a hot-spot for the pest in the region. Large variation was observed in the percent nodule damage among accessions across species. Eight accessions, ILWL 110, ILWL 136, ILWL 166, ILWL 203, ILWL 207, ILWL 245, ILWL 254 and ILWL 258 were identified as resistant, with ≤10% nodule damage, compared to >56% damage recorded on the cultivated lentil. This is the first report of resistance against Sitona weevil in lentil. One resistant accession ILWL 245 belongs to the species L. culinaris Medikus subsp. orientalis (Boiss.) Ponert, progenitor of the cultivated lentil, which is crossable with the cultivated lentil. This line is being used to introgress resistance genes to cultivated lentil and to understand the inheritance of Sitona weevil resistance.  相似文献

The management of genebank collections of cultivated potato is costly due to the need for in vitro maintenance and virus eradication. Therefore, it is important to set up conservation strategies, which prevent duplicates entering the collections. In this study, 32 Nordic potato landraces were studied for 57 morphological traits and analysed for amplified fragment length polymorphism (AFLP). Most landraces could be distinguished based on the morphological characters, except five accessions. Using five primer combinations to generate 114 reproducible AFLPs, of which 63 (55%) were polymorphic, the five morphologically indistinguishable accessions were placed into two groups with identical AFLP patterns, suggesting that some of the accessions were redundant for long-term preservation. The AFLP data showed that the Nordic collection of potato landraces is composed of genetically different clones, and morphological analysis revealed a wide range of variability. This variability seems to be distributed randomly over the Nordic region since the cluster analysis based on AFLPs and morphological traits revealed no grouping based on the country of origin. Principal component analysis suggests that fewer morphological traits than used in this study will be sufficient to discriminate between different genotypes of cultivated potato (Solanum tuberosum L.). Future possibilities for rationalising potato collections are discussed.  相似文献

The wild lettuce, Lactuca aculeata Boiss. et Ky., is closely related and fully interfertile with cultivated lettuce, Lactuca sativa L. Altogether, 155 accessions of this wild species, which is rare conserved within the world’s germplasm collections, were collected from 12 localities throughout northeastern Israel. Ten of these localities represent different sites across the Golan Heights, while two, high-density populations were suprisingly found at the Hula Plain (first report of this species in this region). Lactuca aculeata was recorded at various elevations (222–968 m a.s.l.) and habitats. The taxonomic status of 30 L. aculeata accessions was morphologically validated during ex situ seed regeneration of 31 random accessions representing all 12 localities. Characterization of 12 traits showed that the Israeli populations of L. aculeata do not exhibit broad morphological variability, but more great levels of variation were obtained for developmental traits. Relatively little variability was observed in the morphology of cauline leaves or in the distribution of anthocyanin pigmentation in bracteae. In two cases, L. aculeata plants lacked trichomes in the inflorescence, a typical feature in the majority of plants. These unique collections of wild lettuce may carry novel sources of genetic variation for a wide range of traits and, thus, should be of interest for careful evaluation and exploitation in lettuce breeding.  相似文献

Twenty genic- and genomic SSR markers were used to study genetic diversity and geographical differentiation of barley from 29 countries through analysis of a worldwide collection of 304 ICARDA’s barley landraces. Of these, 19 loci were highly polymorphic in the material studied. Based on Nei-distance matrix, Principal Component Analysis (PCoA) and cluster analysis using UPGMA associated with AMOVA the data revealed countries’ grouping within regions. Three distinct germplasm pools were identified in the landraces. The first of these was from Eastern Africa (Eritrea and Ethiopia) and South America (Ecuador, Peru and Chile) suggesting that barley introduced to South America might have originated specifically from East Africa or that they share a common genetic basis for adaptation. The second was the Caucasus (Armenia and Georgia) and the third included the remaining regions of Central Asia, Near East, Northern Africa and Eastern Asia. Genetic diversity of barley subspecies (Six-rowed barley, Two-rowed barley, H. spontaneum C. Koch and H. agriocrithon Åberg) also discriminates them into three groups: cultivated barleys (Six-rowed barley and Two-rowed barley), wild barley H. spontaneum and subspecies H. agriocrithon. These results associated with parsimony analysis demonstrate that H. agriocrithon and H. spontaneum might be distinct and do not support a hybrid origin for H. agriocrithon suggesting further investigation of the basis of more intense sampling of the two subspecies H. spontaneum and H. agriocrithon.  相似文献

An overview of ex situ collections of cultivated flax (Linum usitatissimum L.) and 53 other species of the genus Linum L. is presented. The names of 33 genebanks in 23 countries preserving Linum germplasm and the species preserved at these places are provided. World genebanks engage in ex situ preservation of about 48,000 accessions of cultivated flax (Linum usitatissimum). However, possibly only 10,000 accessions of these are unique. Of the ca. 200 species of the genus Linum, other than L. usitatissimum, 53 are reported to be represented in the world’s ex situ collections with a total of about 900 accessions. Of these, 279 accessions alone refer to the wild progenitor of cultivated flax, Linum bienne Mill. However, L. bienne from the region of origin of cultivated flax is rarely represented in genebank collections. References to publications describing the techniques for ex situ conservation and the infraspecific variation of cultivated flax are made. It is suggested that taxonomic species delimitation within the genus Linum is in need of clarification and that the variation in Linum species for ornamental use or use in breeding is investigated.  相似文献

Musa L. commonly known as the banana group is one of the most important and oldest food crops of humankind. Among the wild relatives with ornamental interest in the genus, Musa ornata Roxb. shows a disjunct distribution between Asia and North America (Mexico). The wild occurrence of this species in Mexico has led to speculation about the evolutionary relationships with its Asian relatives. This study examined the phylogenetic relationships between intercontinental specimens of this species and, based on registered evidence, explored the more likely hypothesis about the origins of its distribution. The phylogeny of intercontinental specimens, along with other representatives of the same genus, was carried out using three molecular markers (ITS, trnL-F, and atpB-rbcL) and applying three phylogenetic reconstruction methods: maximum parsimony, maximum likelihood, and Bayesian inference. The genetic analysis of the combined dataset grouped together all the Mexican and most Asian specimens, but the monophyly of the species was not supported. The relationships suggest that Mexican populations may have originated from an Asian invasion. However, several studies and historical documents suggest the presence of Musa in America long before the arrival of Europeans. Based on its current distribution, phylogenetic evidence, and fossil record, this species’ disjunct distribution could be explained in terms of an ancestral distribution range that encompassed America and Asia, followed by its subsequent restriction to the Old World and a secondary dispersal by humans. However, further studies are necessary to shed more light on the origins of this disjunct distribution.  相似文献

Oryza officinalis complex is the largest and the most complicated group in the genus Oryza L., consisting of about ten species with the B, C, BC, CD, and E genomes. Taxonomy and identification of the species, particularly those with the B, C and BC genomes, are difficult due to the similar morphology and overlapping distribution of some species. The difference in ploidy levels of some species adds more complexity. In the present study, we surveyed 64 accessions of rice germplasm in the O. officinalis complex using RFLP analysis of PCR-amplified Adh genes in addition to chromosome counting. The results confirmed that all O. rhizomatis accessions are diploids with the C genome, whereas all O. minuta accessions are tetraploids having the BC genome. However, both diploid and tetraploid forms were found for the accessions identified in the genebank as O. officinalis, O. punctata and O. eichingeri. The tetraploid form of O. officinalis with the BC genome is exclusively distributed in India and has been described as O. malampuzhaensis. The tetraploid form of O. punctata which has been called O. schweinfurthiana by some workers was found to be as widely distributed as its diploid form in Africa. It is noteworthy that two accessions that had been determined as tetraploid O. officinalis were actually belonging to a species with the CD genome (O. latifolia). Our results promote a better understanding of the genomic constitutions of many accessions in the O. officinalis complex and correct determination of the genebank material, which serves as an important basis of germplasm cataloguing for further research and utilization.  相似文献

Lettuce, spinach and chicory are generally considered the main leafy vegetables, while a fourth group denoted by ‘minor leafy vegetables’ includes, amongst others, rocket salad, lamb’s lettuce, asparagus, artichoke and rhubarb. Except in the case of lettuce, central crop databases of leafy vegetables were lacking until recently. Here we report on the update of the international Lactuca database and the development of three new central crop databases for each of the other leafy vegetable crop groups. Requests for passport data of accessions available to the user community were addressed to all known European collection holders and to the main collection holders located outside Europe. Altogether, passport data of 17,530 accessions from a total of 129 collections were collected. The four separate databases were made available on line via a common entry page accessible at http://documents.plant.wur.nl/cgn/pgr/LVintro/. Based on a literature study, an analysis of the gene pool structure of the crops was performed and an inventory was made of the distribution areas of the species involved. The results of these surveys were related to the contents of the newly established databases in order to identify the main collection gaps. Priorities are presented for future germplasm acquisition aimed at improving the coverage of the crop gene pools in ex situ collections.  相似文献

Many species in genebank collections are maintained vegetatively either as in vitro cultures or as plants in the field or greenhouse. In these cases, individual genotypes (cultivars) are the focus of conservation. In crop wild relatives, where the value of an accession is in the heterogeneity of segregating genotypes, seeds may be the most representative form of conservation. We used two sets of field-planted Malus sieversii apple trees that originated from seeds collected from two wild populations in Kazakhstan as a model system to determine if seeds from hand-pollinated crosses could be used to represent the allelic diversity of the parent trees in a genebank orchard. A crossing design was developed that used each of the selected trees from the two Kazakhstan collection sites as mothers and also as fathers in the form of bulked pollen pools. The sets of offspring from these crosses were genotyped using microsatellite markers. Paternity analyses revealed the percentage of offspring that was derived from each of the parent trees. We also determined the contribution of each parent to the fertilization events from the pollen pools. Although there were some unintended pollination events (fertilization by trees in unexpected pollen pools) and unequal contribution of pollen parents within pollen pools, we demonstrated that the seedlings effectively captured the allelic frequency and diversity of the parents. A seed collection strategy is the most efficient way to conserve the diversity of crop wild relatives even in collections typically conserved as individual clones.  相似文献

Thailand is a center of diversity for section Angulares of the Asian Vigna (genus Vigna subgenus Ceratotropis) and 4 Vigna species are cultivated in Thailand. Using newly collected wild and cultivated germplasm of Vigna from Thailand and outgroup accessions AFLP analysis was conducted to clarify genetic diversity and relationships. The results suggest that cultivated V. umbellata and V. mungo evolved from wild relatives in a single domestication event. Vigna umbellata is poorly differentiated from its wild and weedy relatives compared to V. mungo. Results suggest northern Thailand and the neighboring Shan state, Myanmar, is the probable center of domestication for V. umbellata as wild accessions from this area and cultivated rice bean from a wide area in Asia are not greatly diverged. Vigna minima, V. tenuicaulis and V. exilis accessions in Thailand are well differentiated with considerable intra-specific variation. Vigna hirtella consists of two well differentiated subgroups, suggesting taxonomic revision may be necessary. Close genetic relationships between V. radiata and V. grandiflora, and between V. mungo and V. trinervia are confirmed. Naturally growing V. mungo populations in northern Thailand appear to be true wild species as they are well differentiated from Indian wild and Thai cultivated populations. The origin of naturally growing cowpea in Thailand needs to be further studied using a more comprehensive set of materials. This study clarifies inter and intra-specific genetic diversity and inter species relationships of Thai Vigna species.  相似文献

Lentil anthracnose (Colletotrichum truncatum (Schwein.) Andrus et W.D. Moore is a potential threat in many lentil (Lens culinaris Medik.) production regions of North America. In the lentil germplasm maintained in Germany and North America, 16 lines were reported to have resistance to race Ct1, but none has resistance reported to race Ct0. The objective of this study was to examine accessions of wild Lens species for their resistance to races Ct1 and Ct0 of lentil anthracnose. Five hundred and seventy-four wild accessions of six species and control lines were screened in two replications under both field and greenhouse conditions using a 1–9 scoring scale (1, highly resistant; 2–3, resistant; 4–5, moderately resistant; 6–7, susceptible; and 8–9, highly susceptible). Indianhead and PI 320937 were resistant while Eston and Pardina were susceptible to race Ct1 as expected. However, none of the check lines were resistant to race Ct0. Among the six Lens wild species tested, accessions of Lens ervoides (Brign.) Grande had the highest level of resistance, 3–5 to race Ct1 and Ct0 followed by L. lamottei Czefr. in the field and greenhouse. Lens orientalis (Boiss.), L. odemensis L., L. nigricans (M. Bieb.) Godron and L. tomentosus L. were highly susceptible, 8–9 to race Ct0 in the greenhouse. The highest frequency of resistance, especially in L. ervoides (Brign.) Grande, was found in accessions originating from Syria and Turkey. The usefulness of these L. ervoides (Brign.) Grande accessions as sources of resistance to the more virulent race of anthracnose in a lentil breeding program is discussed.  相似文献
