A study for about a 30-month period was done to compare strongyle control programs, using per os treatments of ivermectin (IVE) paste exclusively or alternation of 4 antiparasitic paste compounds: IVE, oxfendazole (OFZ), oxibendazole (OBZ), or pyrantel pamoate (PRT). Every 8 weeks, 1 group of horses (barn C; n = 14 to 16) was given IVE paste exclusively, and a second group (barn E; n = 16) was given the 4 antiparasitic pastes on an alternating schedule. Worm eggs and larvae per gram of feces (epg and lpg, respectively) values were determined every 2 weeks during the investigation. This study in grazing horses (mares and fillies), naturally infected with internal parasites, was conducted during the period between Oct 22, 1987 and Feb 8, 1990, with an additional observation on Mar 28, 1990. For barn-C horses, treated exclusively with IVE (200 micrograms/kg of body weight) 14 times, 2-week posttreatment mean strongyle epg and lpg (small strongyle) values were reduced 99 to 100%. Mean strongyle epg and lpg (small strongyle) values for each 2-week sample period remained low (less than 20) throughout the study period, except for 1 moderate transient increase in July 1988. For the entire study period, the aggregate mean strongyle epg value was 12 and the lpg value was 6. Two-week posttreatment mean strongyle epg and lpg (small strongyle) values for barn-E horses, treated alternately with therapeutic (approx) dosage of IVE (200 micrograms/kg; 4 times), OFZ (10 mg/kg; 5 times), OBZ (10 mg/kg; 4 times), or PRT (6.6 mg base/kg; 2 times), varied within and between compounds.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The effect of two treatment programmes on egg shedding in dogs naturally infected with Toxocara canis, one based on a milbemycin oxime-praziquantel-lufenuron combination (SENTINEL) Spectrum; Group 1) and the other based on a febantel-pyrantel embonate-praziquantel combination (DRONTAL) Plus; Group 2), was compared in a study involving 104 suckling pups from three different kennels. The animals in Group 1 were treated at a minimum milbemycin oxime dose of 0.5 mg/kg bw starting at 2 weeks of age and subsequently every 4 weeks until reaching 26 weeks of age. The animals in Group 2 were treated every 2 weeks from week 2 until week 12 of age and then once at week 26 at a minimum febantel and pyrantel embonate dose of 15.0 and 14.4 mg/kg bw, respectively. Toxocara egg counts were determined fortnightly starting at 2 weeks of age and continuing until 26 weeks of age for every pup. Any adverse drug event was recorded during the trial. Both treatment programmes significantly reduced the zoonotic Toxocara egg shedding and were well tolerated by the pups. The pups in Group 1 showed lower average faecal egg counts and were found more frequently shedding no eggs than the pups in Group 2.  相似文献

The outcome of Toxocara canis infections in the canine host depends on the migratory pathway of parasite larvae (somatic or tracheal) which is considered to be related to the host's age and its immune status. However, field studies attest high prevalences of patent T. canis infections in adult animals. The controlled induction of patent infections with low doses of embryonated eggs was investigated in 18 beagles in a 7-month study until their 16th life month. The animals were assigned to three groups, each consisting of three vertically infected dogs (with a short patent infection as pups before anthelmintic treatment) and three helminth-free dogs. At study days 10 and 40, the animals of groups 1 and 3 were given each 100 embryonated T. canis eggs. In each case, group 1 was treated 10 days post-infection with Milbemax, while dogs of group 3 remained untreated. Control group 2 was not experimentally infected but treated as group 1. Two weeks after first egg administration, a sharp increase of specific antibody reactions in ELISA and increased eosinophilic counts indicated larval invasion in all infected dogs. 42-56 days following first infection, patent infections were detected coproscopically in all animals of group 3, but in none of the uninfected dogs (group 2) or the infected and treated dogs (group 1). Following a 3-month observation period, all animals of the three groups were treated with piperazine citrate to eliminate intestinal infections and all were administered 100 embryonated eggs. Subsequently, patent infections developed in animals of all groups: in one of the infected and treated animals of group 1, in five of the so far not infected control group 2 and in four of the dogs with previous patent infections (group 3). Susceptibility to patent infections was not significantly altered in T. canis-free dogs compared to dogs with previous patent infection (vertically acquired or experimentally induced). However, dogs of group 1 treated with Milbemax after repeated egg administration developed a significantly increased resistance to patent infections as compared to control dogs (group 2). Observed prepatency periods were between 40 and 56 days and did not differ in the three groups. Even in urban areas, facing high infection pressure with Toxocara eggs maintained by a high dog and fox population, dogs of all ages are at risk to develop patent T. canis infections.  相似文献

A gel formulation containing moxidectin (20 g/kg) and praziquantel (125 g/kg) reduced the geometric mean faecal strongyle egg count in horses to below 100 eggs per gram of faeces (epg) for at least 12 weeks despite their being exposed continuously to reinfection from pasture grazed by treated and untreated horses. The geometric mean egg count of horses treated with a proprietary paste containing abamectin (3.7 g/kg) and praziquantel (46.2 g/kg) increased steadily from six weeks after the treatment, peaking at over 820 epg after 12 weeks. Relative to the efficacy of the abamectin/praziquantel treatment, the reduction in mean faecal egg count compared with the pretreatment counts was significantly (P<0.05) better in the horses treated with moxidectin and praziquantel from eight weeks after the treatment. Both products eliminated tapeworms from horses in a non-invasive modified critical trial.  相似文献

This study was undertaken to determine the prevalence of Toxocara canis eggs on the coats of dogs (a potential etiological factor for human toxocariasis) and to see if there were mainly a dog breed and coat type effects for the presence of eggs on the coat. Hair samples were collected from the different breeds of 51 domestic pet dogs and examined for the presence of T. canis eggs. A total of 62 T. canis eggs (all viable) were found in 21.56% of the dogs. Forty-nine (79.03%) of the eggs recovered were unembryonated, 8 (12.90%) were embryonating, and 5 (8.06%) were embryonated. The maximum densities of the embryonating and embryonated eggs were 93 and 8.45 eggs per gram (epg) of hair, respectively. The number of eggs recovered was much higher than those previously reported for soil samples. Although the statistical analysis for all dogs in this study showed that there were no breed (P>0.4), coat type (P>0.8), sex (P>0.1), age group (P>0.1) and hair length (P>0.3) effects for the presence of T. canis eggs per gram of hair, the majority of dogs (82%) with T. canis eggs in their coats were breeds that had double coats with thick undercoats suggesting that the coat characteristic may play a role for providing a suitable environment for the development of T. canis eggs. Also 82% of infected dogs were under 1 year of age indicating that the age of dog is a very important risk factor. The present study indicates that direct contact with T. canis infected dogs may be a potential etiological factor for human toxocariasis.  相似文献

Levamisole, morantel, fenbendazole, or ivermectin was administered at 2-week intervals from May 1 through Sept 14, 1985, to breeding ewes (20 ewes/drug) infected with various gastrointestinal nematodes. All ewes had fewer gastrointestinal nematode eggs per gram of feces (epg) after 2 treatments, compared with pretreatment epg counts. Ewes administered ivermectin continued to have a low mean epg (0 to 3) throughout the study. The mean epg counts of ewes treated with levamisole increased from 3 to 483 during the study. This increase was similar to that of ewes treated with morantel (7 to 485 epg). The mean epg count of fenbendazole-treated ewes increased from 4 to 192 during the study. By the end of the study, the mean epg counts when expressed as a percentage of the pretreatment epg counts were 4% (ivermectin), 249% (fenbendazole), 627% (levamisole), and 630% (morantel). With the exception of the ivermectin-treated ewes, the epg count increased almost linearly in the ewes after the 2nd anthelmintic treatment. These data indicate that the gastrointestinal nematodes (including Haemonchus contortus) may have developed more resistance to levamisole and morantel than to fenbendazole. On the basis of the epg counts, resistance to ivermectin did not develop during the 4.5-month treatment period. The percentage of ewes shedding eggs after 2, 4, and 6 anthelmintic treatments was lowest for ewes treated with ivermectin (20%) and was similar (40%) for ewes treated with 1 of the other 3 anthelmintics. Sub-clinical parasitism in spring-born single suckled beef calves was investigated from the middle of their first grazing season until weaning or housing later the same year. The study was conducted on four beef suckler herds in southern England over a 3-year period and involved a total of 334 spring-born beef suckler calves and their dams. The animals were grazed extensively on pastures naturally infected with nematode larvae. At the start of each period of observation, faecal samples were taken from calves and cows and subjected to routine worm egg counts; calves were re-sampled at the end of the grazing season.In July in each year and at each location the calves were ranked by initial weight within sex, paired according to rank and randomly allocated to either an untreated control group or a group in which the calves were each treated with an ivermectin sustained-release (SR) bolus. The calves in both trial groups, and their dams, were grazed together until weaning or housing. The calves were weighed at the initial allocation and at the end of the study. The adult cows were not treated with any anthelmintic during the study.The faecal nematode egg counts (FECs) conducted in July showed that the suckler cows were excreting worm eggs at low concentrations: range 0-100 eggs per gram (epg), with one individual count of 500epg, 88% of the cows sampled had counts of <50epg. Similarly, the counts from the calf samples were fairly low in July: range 0-250epg, 73% of the calves sampled had counts of <50epg. By the end of the grazing season, the faecal samples from the untreated control calves showed higher values: range 0-650epg, with only 58% having an epg of <50.The average rate of daily liveweight gain in the untreated heifer calves was 0.79kg per day, the corresponding figure for the heifer calves treated with the ivermectin SR bolus in mid-summer was 0.88kg per day; the difference of 90g per day was significantly different (P=0.0118). The average rate of daily liveweight gain in the untreated bull calves was 0.91kg per day, the corresponding figure for the bull calves treated with the ivermectin SR bolus in mid-summer was 1.01kg per day; the difference was significantly different (P=0.0169).  相似文献

To determine the efficacy of a formulation of milbemycins in treating patent infection with Toxocara canis, 8 male and 7 female, 10-week-old, ascarid-free Beagles each were given 125 embryonated eggs of T canis. All dogs developed patent infection within 56 days. On post-infection day 70, the dogs were assigned to 1 of 3 groups of 5 dogs each; members of 1 group were given a placebo, while dogs of the other 2 groups were given either 5.68 or 34.08 mg of the milbemycin formulation, respectively. In both groups of dogs given the drug, the number of eggs passed per gram of feces decreased precipitously. However, a few eggs still were found in the feces of several dogs of each group on the day of necropsy (postinfection day 75). Worms or fragments of worms were passed by the treated dogs from the day of treatment until the day on which necropsy was performed; however, most worms were passed during the first 2 days after treatment. At necropsy, only dogs of the control group were found to harbor adult T canis.  相似文献

The aim of this study was to prove our hypothesis that adult rats with lowering of body weight gain, rats at 12 weeks of age as an example, are suitable for evaluation of myelotoxicity. Age-related differences between young rats (6-week-old study) and adult rats (12-week-old study) were analyzed in hematological examination values. The data of the young rats were reprinted from our previous report (Miyata et al., 2009) since our hypothesis was verified by comparison with that previous report. Several experimental groups were defined for the 12-week-old study as well as for the 6-week-old study; these included 5-fluorouracil (5-FU) treated groups receiving 12, 15 and 18 mg/kg/day (FU12, FU15 and FU18), pair-feeding groups (R12, R15 and R18 receiving the same amount of food as in the FU12, FU15 and FU18 groups, respectively) and a nontreated control group. Numerous hematologic and bone marrow parameters in the 5-FU treated groups were comparable to those in the corresponding pair-feeding groups in both age studies. Generally, the influences of undernutrition were more apparent in the young rats than in the adult rats. Histopathological examinations showed a decrease in hematopoiesis in the bone marrow in the 5-FU treated and pair-feeding groups. No apparent differences were observed in the decreased hematopoiesis between the 5-FU treated and pair-feeding groups in the 6-week-old study, but a difference between these groups was noted in the 12-week-old study; decreased hematopoiesis was more frequently noted in the 5-FU treated groups. These facts suggest that adult rats are more suitable than young rats for evaluation of 5-FU-induced myelotoxicity.  相似文献

We tested the hypothesis that treatment of growing, susceptible (to hip dysplasia) pups by IM administration of glycosaminoglycan polysulfates would mitigate the signs of incipient hip dysplasia. In 1 experiment, 7 pups, selected at random from 2 litters, were administered glycosaminoglycan polysulfates (2.5 mg/kg of body weight, IM) twice weekly, and 7 control pups from the same litters were given sterile buffered 0.9% saline solution from the age of 6 weeks to 8 months. Hip joints were examined by radiography, with pups in the standard, limbs-extended position. At 8 months of age, all pups in this experiment did not manifest femoral head subluxation radiographically. The Norberg angle, a measure of coxofemoral congruity, improved from a mean +/- SEM value of 102 degrees +/- 1 degrees in controls to 106 degrees +/- 1 degrees in treated pups (P = 0.008). Pups were not subjected to necropsy. In the second experiment, 8 pups were selected at random from 2 litters and were administered 5 mg of glycosaminoglycan polysulfates/kg, IM, twice weekly from 6 weeks to 8 months of age. Similarly, 8 control pups were administered saline solution. At 8 months of age, hip joints were examined by radiography with pups in the standard position; at necropsy, intra-articular tissues were evaluated macroscopically and biochemically. Of 8 treated pups, none had subluxation radiographically, whereas 4 of 8 control dogs had femoral head subluxation. Mean Norberg angle on the radiographs was 109.7 degrees +/- 1.6 degrees for the treated group and was 101.5 degrees +/- 1.6 degrees for controls, representing a mean improvement in coxofemoral congruity of 8.2 degrees in the treated pups.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

CASE DESCRIPTIONS: 16 horses treated daily with pyrantel tartrate (2.64 mg/kg [1.2 mg/lb], PO) as part of a prophylactic anthelmintic program. CLINICAL FINDINGS: Fecal worm egg counts (FWECs) were obtained on all 16 horses. Mean FWEC was 478 eggs/g (epg; range, 0 to 4,075 epg). Three of the 16 horses were responsible for 85% of the total fecal egg output for the herd on the day of sampling. Six horses had FWECs < 200 epg. Three horses that had arrived within 4 months of the sampling date had FWECs < 100 epg. TREATMENT AND OUTCOME: An FWEC reduction test was initiated the day after FWECs were obtained; all horses with FWECs > 100 epg (9 horses) were treated with pyrantel pamoate (6.6 mg/kg [3 mg/lb], PO), and 14 days later, the FWEC was repeated. During the 14-day period, all horses received pyrantel tartrate (2.64 mg/kg, PO) daily. Fecal worm egg count reduction was calculated for each horse. Mean FWEC reduction for the group was 28.5% (range, increase of 21% in FWECs 14 days after treatment to a decrease of 100% in FWEC 14 days after treatment). CLINICAL RELEVANCE: Farms should be monitored for cyathostomes resistant to pyrantel pamoate prior to use of pyrantel tartrate. Fecal worm egg counts should be monitored routinely in horses before and after treatment to ensure efficacy of cyathostome control measures.  相似文献

1. Uterovaginal junction (UVJ) tissues were collected 24 h after artificial insemination (AI) from 85- and 125-week-old broiler breeder hens in three physiological states: laying hard shell eggs (HS), laying shell-less eggs (SL) and non-laying (NL). This was confirmed by egg production records and visual appraisal of the oviduct at the time of necropsy. 2. Three longitudinal sections of each UVJ were evaluated microscopically and sperm host glands (SHG) were scored in 5 categories: glandular morphology evident but not lumen present, basophilic stained epithelium and no spermatoza present, glands that contained one to five spermatozoa; glands that contained 6 to 20 spermatozoa and glands that contained more than 20 spermatozoa. 3. Laying hens (HS and SL) at 85 weeks of age had significantly more sperm host glands (SHG) containing spermatozoa than NL hens. At 125 weeks of age HS and SL hens had significantly more unscorable glands. 4. The only category that showed no difference between age and physiological status group was the empty category. No significant differences were observed for any gland category in 82- and 125-week-old NL hens. 5. A greater proportion of the 85-week-old group of HS and SL hens had more SHG containing spermatozoa and fewer unscorable glands that the 125-week-old birds. Haemonchus contortus eggs were extracted from sheep feces and known numbers were added to helminthologically sterile bovine feces to provide samples with seven, 30 and 60 eggs per gram (epg). At 60 epg, dilution techniques (modified Cornell-McMaster and modified McMaster) tended to overestimate the number of eggs and more eggs were recovered (mean of 121 and 88% respectively) with these techniques than with centrifugal concentration procedures (modified Cornell—63% and Wisconsin— 69%). At 30 epg, all techniques were comparable (modified Cornell-McMaster 67%, modified McMaster 63%, modified Cornell and Wisconsin 64%). At 7 epg, the Wisconsin (61%), modified Cornell (60%) and Cornell-McMaster (94%) techniques were comparable and better than the modified McMaster technique (16%). At all levels of epg, the modified Cornell and Wisconsin techniques recovered eggs from 100% of the samples. The Cornell-McMaster and modified McMaster techniques recovered eggs from 90 and 100% of samples at 60 epg; 40 and 100% at 30 epg; and 21 and 11% at 7 epg. With a gravitational concentration procedure, the Standard Vial, no more than 16% of the eggs at any level of epg were recovered and at 7 epg eggs were recovered from only one-half of the samples. Five gravitational concentration techniques were assessed over 66 to 490 epg. The Ovassay, Fecalyzer and modified Standard Vial techniques were comparable in efficiency (28%, 25% and 24% respectively), but the Standard Vial technique was less efficient (11%).

Introduced into diagnostic parasitology was the concept of predictive values which is the proportion of samples that a technique correctly identifies as being negative for parasite eggs. At 7 epg this was calculated to be zero for the modified Cornell-McMaster, modified McMaster and Standard Vial techniques and 100 for the Wisconsin and modified Cornell techniques.

Equine helminth infections: control by selective chemotherapy   总被引:3,自引:0,他引:3  
A programme of selective anthelmintic therapy was used in a herd of 31 horses. Faecal egg counts were done during the months of September, November, January, March, May and the following September. Horses with greater than or equal to 100 eggs per gram (epg) were treated with ivermectin, and those with less than 100 epg were not treated. The criteria for adequate internal parasite control in the herd was a median herd faecal egg count of less than or equal to 100 epg. Effectiveness of selective therapy was assessed by faecal egg count after nine months of treatment and was determined to be adequate when a median herd egg count of 0 epg was obtained. However, on returning from pasture the following September, median herd egg count had risen to 325 epg. A statistically significant correlation was seen in the paired September faecal egg counts of the horses in that initial September faecal egg count was predictive for the following September. Initial September faecal egg count was related to the number of anthelmintic treatments required during the period of selective therapy, whereas age of horse was not. We propose that faecal egg counts be incorporated into strategic anthelmintic programmes as an economical tool for identifying and targeting herd members predisposed to shedding elevated numbers of helminth eggs.  相似文献

Babesia canis infection was diagnosed in a litter of seven 3-week-old Mastiff pups kept in a north Florida kennel. The pups were evaluated because of poor weight gain; the smallest pup also was markedly lethargic. Six of the pups were anemic and thrombocytopenic. A positive linear correlation between PCV and absolute reticulocyte count suggested that the variation in PCV may have been related more to the ability of a pup to increase erythrocyte production than to a difference in magnitude of erythrocyte destruction. All pups recovered from clinical signs and hematologic abnormalities attributable to babesiosis within 2.5 weeks after treatment with diminazene aceturate. Transient neurologic signs observed in 1 pup 3 days after treatment were believed to represent an adverse drug reaction. The dam of the litter had a serum titer of 1:640 for B canis, but appeared healthy, as did approximately 30 other adult dog in the kennel. The strain of B canis infecting dogs in the kennel caused severe illness and death in some pups, but clinically inapparent disease in adult dogs.  相似文献

The clinical pathology of Schistosoma curassoni infection in sheep and goats was studied for 22 weeks following experimental infection with 7000 and 4000 cercariae, respectively. Excretion of eggs began at week 7 after infection: in goats the numbers increased to 30 to 50 eggs per gram faeces (epg) at weeks 8 to 18, followed by a reduction. In a pregnant goat, epg values increased markedly before and after parturition. The mean faecal egg counts in sheep were lower than in goats, increasing to a maximum level of 30 epg at weeks 16 and 17 after infection. Infected sheep maintained growth rates roughly comparable with controls, whereas infected goats failed to gain as much weight as the controls. Infected goats and sheep produced eosinophil counts of about 3 x 10(3) mm-3, five and eight weeks after infection, respectively. Sheep developed a progressive anaemia from week 11 after infection, in goats blood values remained within normal limits. Differences in serum protein concentration were observed between infected and uninfected goats about nine weeks after infection, but not in sheep. Increased total protein values, hyperglobulinaemia and lowered albumin to globulin ratios were features of infected goats. Serum glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, gamma-glutamyl transferase, total lactate dehydrogenase and bilirubin were not significantly changed. The mean recovery in sheep was 608 worms, in goats 428 worms, but the total tissue egg counts were higher in the latter. Of the total eggs deposited in the goats 92 per cent were found in the liver with 51.5 per cent in the ovine liver. The histopathological changes were studied.  相似文献

Haemonchosis is considered to be the most economically important gastrointestinal disease of small ruminants in the tropics and subtropics. However, chemical anthelmintics, which were the mainstay of control, have been compromised by a high prevalence of resistance worldwide. Copper oxide wire particles (COWP) have been shown to have anthelmintic effects, but few studies have examined their use under field conditions. The use of COWP was therefore evaluated as a tactical anthelmintic treatment in indigenous goats raised under communal farming conditions in Bergville, KwaZulu-Natal Province, South Africa. At the beginning of the summer rainfall season (October 2007), the faecal egg counts of 172 female goats belonging to 15 farmers were determined and this sampling continued every four weeks until the second week of January 2008. The goats within each of the 15 herds were ranked according to their faecal egg counts for this week. The goats were sequentially paired off within each ranking starting with those goats with the highest counts. One goat from each pair was randomly allocated to a treated or control group. Two weeks later, a 4 g COWP bolus was randomly administered to each goat in the treated group. Faecal egg counts were carried out on the goats two weeks following treatment, and the sampling of the goats then proceeded every four weeks until October 2008. Except for the six-week period prior to the administration of the COWP, the goats were examined according to the FAMACHA© system and symptomatically treated with 12 mg/kg levamisole when anaemic. The percentage reduction in faecal egg count due to the COWP treatment was 89.0%. Mean pre- and post-treatment faecal egg counts for the COWP-treated group (n = 73) were 2347 eggs per gram of faeces (epg) and 264 epg, respectively. The corresponding values for the untreated controls (n = 66) were 2652 epg and 2709 epg. The prevalence of Haemonchus spp. larvae in pre- and post-treatment faecal cultures was 72% and 46%, respectively. Symptomatic anthelmintic treatments in combination with mid-summer tactical treatments with COWP appear to be useful strategies for the control of Haemonchus contortus in indigenous goats in this farming system and this approach could have application in other similar agro-ecological zones.  相似文献

Previous studies have indicated that host genetics significantly affects the number of gastrointestinal nematode eggs per gram (epg) in the feces of calves during their first grazing season. An entire calf crop of approximately 190 animals was monitored monthly until weaning to verify these earlier results, and to begin to discern the basis for this phenomenon. A significant genetic effect on fecal epg values was not observed until calves had been on pasture for 2-3 months, and was demonstrable until late in the grazing season when the effect was lost. The loss of a genetic effect coincided with the appearance of significant numbers of the more highly fecund nematode species Haemonchus placei and Oesophagostomum radiatum, and with an apparent increase in Ostertagia ostertagi transmission, indicating that the observed genetic control of epg values may be species specific, dose dependent or both. Calves were selected from the population, and grouped according to their epg phenotype over the grazing season as either high or low epg calves. Postmortem examination of some of these calves indicated that worm burdens in the low epg calves were 60% of those of the high epg calves. Experimental challenge inoculation of the remaining calves indicated that: (1) challenge with Cooperia oncophora resulted in low epg calves harboring worm numbers that were 65% of those of high epg calves; (2) challenge with O. ostertagi resulted in similar numbers of worms in both groups, but the fecundity of worms in the low epg groups was significantly lower (P less than 0.05) than in the high epg group. Analysis of serum anti-Ostertagia antibody levels in the grazing calf population showed rises in serum IgG1, IgG2, IgM and IgA antibody levels during the grazing season. Peak serum IgG2 and IgG1 anti-Ostertagia antibody levels were found to be significantly affected by host genetic factors while IgA and IgM levels were not under such control.  相似文献

The efficacy of selamectin in the treatment and prevention of naturally acquired Toxocara canis infections and experimentally induced flea (Ctenocephalides felis felis) infestations in dams and their suckling pups was evaluated by administering selamectin to the adult females only, approximately 40 and 10 days before parturition and 10 and 40 days after parturition. Unit doses of the commercial formulation of selamectin were administered to the dams to provide at least the minimum recommended dosage of 6mgkg(-1) (range, 6-12mgkg(-1)). Dams and their pups were housed in carpeted environments able to support the flea life cycle. Flea infestations were established initially by experimental infestation before treatment administration and by repeated re-infestation of dams at approximately weekly intervals throughout the study, which was completed 45 days after parturition. There were no adverse drug experiences related to treatment with selamectin and no treatment-related mortalities. Percentage reductions in geometric mean T. canis faecal egg counts for the selamectin-treated dams, compared with those receiving the negative-control treatment (vehicle only) were 99.7% at the end of the study (P=0.0001). Geometric mean faecal egg counts in pups from selamectin-treated females were reduced by > or =96% on the 24th and 34th days after birth (P=0.0001), and the number of adult worms recovered from the gastrointestinal tract of pups from selamectin-treated dams was reduced by 98.2% (P=0.0001), compared with that for pups from dams treated with the vehicle only. Percentage reductions in geometric mean flea counts for selamectin-treated dams and their pups, compared with vehicle-treated dams and their pups, were > or =99.8% (P=0.0001) and 100% (P=0.0001), respectively, throughout the study. Thus, selamectin administered topically at a minimum unit dosage of 6mgkg(-1) to dams with naturally acquired T. canis infections and experimentally induced C. felis infestations was safe and highly effective in the treatment, control, and prevention of adult T. canis infection and C. felis infestation affecting both the dams and their pups.  相似文献

The current level of anthelmintic resistance in the horse-breeding industry is extremely high and therefore more emphasis is being placed on studies that focus on the judicious use of anthelmintic products. The aims of the study were to: 1) establish if there is variation in the egg excretion pattern of strongyles between the different age classes of Thoroughbred horses in the Western Cape Province (WCP), 2) test if a selective treatment approach successfully reduces the number of anthelmintic treatments and maintains acceptably low helminth burdens in adult Thoroughbred horses, and 3) evaluate the efficacy of subsampling large horse herds for faecal egg counts (FECs) to monitor the strongyle burden. In 2001 the FECs of 4 adult mare, 5 yearling and 3 weanling herds from 8 different farms were compared in the WCP Within the mare herds there were generally fewer egg-excreting individuals with lower mean FECs compared with the younger age classes. Individual faecal samples were collected every 3-4 weeks from 52 adult Thoroughbred mares from 1 farm in the WCP during a 12-month period (2002/2003). Animals with strongyle FECs > or =100 eggs per gram (epg ) were treated with an ivermectin-praziquantel combination drug (Equimax oral paste, Virbac). The mean monthly strongyle FEC for the entire group was <300 epg throughout the study and the number of treatments was reduced by 50 %. Resampling methods showed that an asymptote to mean FEC was reached at 55 animals for each of the pooled weanling, yearling and mare egg counts. Resampling within 4 different mare herds recorded asymptotes of between 24 and 28 animals. Subsampling entire herds for FECs therefore provided an effective approach to treatment management. This study demonstrates that selective treatment is both a practical and an effective approach to the management of anthelmintic resistance.  相似文献
