A polynucleate Rhizoctonia isolate (R3) was analysed for virulence, growth characteristics, enzyme production and presence of dsRNAs. Taxonomic position was assessed morphologically and by anastomosis group (AG) testing and ITS sequence analysis. Results indicated that R3 is a hypovirulent R. solani AG 4. Mechanisms underlying biocontrol towards virulent R. solani and Botrytis cinerea were investigated and plant-mediated resistance was followed using biochemical markers of defence (PR1, laminarinase, chitinase). Control apparently relies on spatial and nutrient competition in soil, and on systemic induced resistance. This is the first report on induction of systemic resistance and of defence markers by a hypovirulent strain of R. solani.  相似文献

A collection of 241 isolates of Rhizoctonia solani obtained from potato plants grown in different areas in France was characterized for anastomosis grouping, symptomatology on tubers of different cultivars and sensitivity to three fungicides. Most isolates collected belonged to (anastomosis groups (AGs)) AG 3, but 2% and 4% of the isolates were AG 5 and AG 2-1. AG 3 and AG 2-1 isolates were mostly obtained from sclerotia on tubers, but all AG 5, some AG 3 and some AG 2-1 isolates were recovered from superficial tuber alterations, like deformations, corky or scabby lesions. Sclerotia were formed on tubers produced by healthy stem cuttings grown in soil artificially infested with AG 3, but not on tubers grown in soil infested with either AG 5 or AG 2-1. No variation in susceptibility to sclerotial formation was observed among five potato cultivars. In all cases, a large proportion of tubers showed superficial corky lesions, often associated with deformations. The proportion of tubers with lesions and deformations was highest in soil infested with AG 2-1 and significantly lower on cv. Samba in all treatments. All isolates were highly sensitive to flutolanil, iprodione and pencycuron, except the AG 5 isolates, moderately sensitive to pencycuron. These results show that, although AG 3 is the most common R. solani group on potato in France, AG 5 and AG 2-1 may be present. Isolates differed for pathogenicity. In vitro sensitivity to fungicides varied among AGs.  相似文献

Tomato and transgenic oilseed rape plants expressing the Cf-9 resistance gene develop a hypersensitive response (HR) after injection of the corresponding Avr9 gene product. It was investigated whether induction of a HR conferred resistance to different fungal pathogens in tomato and oilseed rape. Induction of an AVR9 mediated HR at the pathogen infection site delayed the development of the biotrophs Oidium lycopersicum in tomato and Erysiphe polygoni in oilseed rape, but enhanced the development of the necrotrophs Botrytis cinerea and Alternaria solani in tomato and Sclerotinia sclerotiorum in oilseed rape. Interestingly, delayed fungal disease development was observed in plant tissues surrounding the HR lesion regardless of whether a necrotrophic or biotrophic pathogen was used. In tomato, AVR9 injection induced systemic expression of PR1, PR2 and PR3 defence genes but did not induce systemic resistance to O. lycopersicum, B. cinerea or A. solani. In oilseed rape, AVR9 injection temporarily induced systemic resistance to Leptosphaeria maculans and E. polygoni, but did not induce detectable systemic expression of PR1, PR2 or Cxc750. These results give new insights into the potential uses of an induced HR to engineer disease resistance.  相似文献

The rDNA-ITS sequence of Rhizoctonia solani AG 1-ID was determined and compared to those of R. solani AG 1-IA, AG 1-IB, and AG 1-IC. The similarity of the isolates from each AG 1 subgroup was almost identical (99%–100%), whereas it was lower between subgroups (91%–95%) than within subgroups. Phylogenetic analysis indicated that isolates of AG 1-ID and other subgroups were separately clustered. Isolates of R. solani AG 1 were clearly separated from R. solani AG 2-1, AG 4, and binucleate Rhizoctonia AG-Bb and AG-K. These results showed that analysis of the rDNA-ITS sequence is an optimal criterion for differentiating R. solani AG 1-ID from other subgroups of R. solani AG 1.  相似文献

Basal rot is a common disease in lettuce greenhouses. A 3-year study on the diversity of pathogens associated with basal rot in Belgium was carried out. A total of 150 isolates were collected originating from 56 greenhouses. Four pathogens appeared to be involved. Rhizoctonia solani was found to be the causal agent at 23 locations, Sclerotinia spp. at 14, Botrytis cinerea at 17 and Pythium spp. at seven. The isolates of R. solani were further characterised to anastomosis groups and subgroups using morphological characteristics, pectic zymogram and PCR-RFLP. Five anastomosis groups could be distinguished: AG1-1B, AG4 HGI, AG10, AG2-1, AG2-1 Nt and AG3, with isolates of AG4 HGI and AG1-1B being the most prevalent and the most aggressive. Sclerotinia sclerotiorum was found at 13 locations, while S. minor was found at only one location. Based on ITS-sequencing Pythium isolates were assigned to three different species. At 20°C, isolates of all pathogens were able to cause lesions on detached lettuce leaves, except isolates of R. solani AG3 and AG2-1 Nt. A correlation could be found between the occurrence of the pathogens and the growing season. Botrytis cinerea was the most common pathogen in winter, whereas R. solani was most frequently isolated in summer. Sclerotinia spp. and Pythium spp. were isolated in spring, summer and autumn. The information obtained in this study will be most useful in the development of an alternative control strategy for causal agents of basal rot.  相似文献

The position of plants withRhizoctonia solani sclerotia (black scurf) on progeny tubers was mapped for an experimental field at Haren where potatoes were grown continuously and in rotation with other crops for five successive years, and for another field at Borgercompagnie with a 12 frequency of potatoes during three potato crops. Initially, the distribution of plants with black scurf on both fields was rather dense and homogeneous. In the following years the distribution became heterogeneous and patchy. The local decline ofR. solani AG 3 (the common potato pathogen) in Haren was apparently caused by an unknown factor selectively suppressingR. solani AG 3, while simultaneouslyR. solani AG 5 increased in mass. This AG 5 type proved to be an inferior competitor of AG 3 on the potato plant in a laboratory experiment. The specificR. solani antagonistVerticillium biguttatum did not play a role. A similar factor could have reduced the formation of black scurf in the experimental field at Borgercompagnie, whereV. biguttatum was also too infrequent to account for the decline.R. solani AG 5 was not present here and could not indicate the presence of a selective factor against AG 3.  相似文献

Findings from 2 years of field experiments investigating the relationship between Globodera rostochiensis and Rhizoctonia solani on unique field sites are reported. In 2000, a field experiment was positioned on land that had previously been used for experimental work investigating integrated potato cyst nematode (PCN) management methods. This study had produced an ‘untypical’ mosaic of PCN population densities ranging from 5 to 221 eggs g−1 soil. In 2001, the field experiment was conducted on a different field site and overlaid on a focus of G. rostochiensis population densities ranging from 11 to 108 eggs g−1 soil. In each experiment, potatoes (cv. Désirée) were grown in plots with similar population densities of G. rostochiensis that were either uninoculated or inoculated with R. solani. A series of potato plant harvests were undertaken to investigate the effects of nematode infestation on the incidence and severity of R. solani diseases and the associated development of plants. In both experiments, a clear relationship was found between the density of G. rostochiensis juveniles present in potato roots and the incidence of stolons infected by R. solani, 6 weeks after planting. For the first time this interaction has been determined under field conditions. The results of the study suggest that the interaction between nematode and fungus is indirect and possible mechanisms are discussed.  相似文献

水稻纹枯病是由死体营养型真菌立枯丝核菌Rhizoctonia solani AG1-IA亚群引起的一种土传病害,在世界水稻种植区均有发生,是限制水稻高产的主要病害之一。虽然国内外学者针对纹枯病菌已开展了大量的研究工作,但由于该菌寄主范围较广、抗性水稻资源缺乏及其田间抗性鉴定的不稳定性等问题,该病害的研究仍没有取得突破性进展。挖掘自然界中存在的纹枯病抗源材料,选育抗病水稻品种是防控该病害、降低水稻产量损失,从而最大限度保障全球水稻产业可持续发展的有效手段。该文对近年来国内外关于水稻纹枯病菌与寄主的互作分子机制、抗性水稻基因资源挖掘及其抗性机制的最新研究进展进行综述,并提出下一步的重点研究方向,以期为推动水稻对纹枯病的抗性机制解析及抗纹枯病水稻育种提供参考。  相似文献

Bottom rot caused by Rhizoctonia solani is an increasing problem in field-grown lettuce in Germany. During the growing seasons of 1999 and 2000, 95 isolates of R. solani from lettuce plants with bottom rot symptoms were collected from eight locations. The isolates were characterised using hyphal anastomosis, pectic zymograms and morphological characteristics. Ninety-three isolates were identified as anastomosis group (AG) 1-IB, one as AG 1-IC and one as AG 2-1. Optimum hyphal growth was measured over a temperature range of 20–30 °C with an optimum at 25 °C. Aggressiveness of the AG 1-IB isolates varied from weak to strong when tested on detached lettuce leaves. The pathogenic potential of six AG 1-IB isolates was determined on 14 plant species in comparison with lettuce under conditions favourable for the fungus. Radish, broccoli, kohlrabi, spinach and millet seedlings were as severely infected as lettuce seedlings. The same isolates caused little symptoms on maize, tomato and onion. Knowledge about the host range of AGs of R. solani are important for planning an effective crop rotation as part of a control management system.  相似文献

Brachypodium distachyon (Bd) is increasingly being used as a model for cereal diseases and to study cereal root architecture. Rhizoctonia solani AG 8 is a necrotrophic root pathogen that infects wheat soon after germination resulting in reduced plant growth and yield loss. Genetic resistance to R. solani AG 8 is not available in commercial wheat cultivars, although some quantitative levels of resistance have previously been found in mutant lines and grass relatives. Resistance mechanisms in cereals remain unknown. The ability to use Bd as a model to study the wheat–R. solani AG 8 pathosystem was investigated. The results presented show that Bd is susceptible to R. solani AG 8 and that the pathogen infects both species to a similar degree, producing comparable disease symptoms. Root length reduction was the primary indicator of disease, with shoots also affected. The second objective was to develop a repeatable phenotyping method to screen Bd populations for resistance to R. solani AG 8. Results of a preliminary experiment provide evidence for variation in resistance between Bd inbred lines. This is the first report showing the potential of Bd as a model plant for discovery of quantitative genetic variation in resistance to a necrotrophic cereal root pathogen.  相似文献

Rhizoctonia solani AG 2-1 was recorded in Central Italy on kohlrabi plants showing root and stem rot. After artificial inoculation the fungus caused damping-off of 7-day-old seedlings and root and stem rot of 4-month-old plants developed after 15 days of incubation. looseness-1In two observation fields, where six sites were artificially infested with Rhizoctonia solani AG 2-t, bare patches developed. These patches did not re-occur at the site of infestation in three successive years. In fields with and without artificial infestation, natural infection of tulip bulbs by Rhizoctonia spp. occurred. The spatial distribution of infected tulip bulbs was visualised in maps after kriging. The influence of sampling intensity was evaluated by stepwise reduction obtained in the observed data set of the first year. Omnidirectional semivariogram characteristics did not change when sampling intensity was reduced down to 10%. The average maximum prediction error was minimised at sampling intensities varying from 7% to 25%. Naturally occurring bare patches slowly vanished during successive cropping of flower bulbs and did not re-appear in the fourth growing season. A high frequency of isolation of R. solani AG 2-t in one field (Lisse-2) in the fourth consecutive crop did not result in bare patches in that year. It is hypothesised that a reduction in aggressiveness may account for this observation. In contrast, bulb rot due to Rhizoctonia spp. increased during the observation period. R. solani AG 5 isolates were seldom isolated before the bulbs flowered, but were the dominant isolate from bulbs at harvest. In a growth chamber experiment, it was demonstrated that AG 5 did not account for replacement of AG 2-t. However, it was demonstrated that competition may partially explain replacement of AG 2-t isolates during the growing season. At 18 °C, but not at 9 °C, an AG 4 isolate prevented AG 2-t colonising and infecting iris bulbs when both isolates were introduced together to soil. Rhizoctonia populations develop in relation to soil temperature and plant development. It is hypothesised that a temporal niche differentiation may be one of the mechanisms affecting the dynamics of rhizoctonia bare patch of tulips.  相似文献

Among 153 isolates ofRhizoctonia spp. obtained from 95 soil samples collected from different fields in the USA, 42 (27.5%) isolates were hypovirulent or non-pathogenic on cabbage (tested on tap water agar plus 250 μg/ml chloramphenicol plates). Of these, 14 (33.3% of the np-R) isolates protected >60% of the cabbage seedlings againstR. solani, and the best eight isolates protected 73–95% of the cucumber seedlings. The np-R isolates RU56-8 (AG-P) and RU89-1 [AG-B(o)] induced the highest resistance against hypocotyl challenge inoculation with virulentR. solani (38.3–85.7%), whereas most of the challenged control seedlings (85–100%) collapsed. Similarly, isolates RU56-8 and RU89-1 induced the highest resistance (22.2–87.5%) against hypocotyl challenge inoculation withPythium aphanidermatum, whereas most of the challenged control seedlings collapsed (90–100%). Isolates RU56-8 and RU89-1 significantly reduced the lesion numbers and area/leaf (to 8.9–42.0% of the control) caused by challenge inoculation of the first true leaves withPseudomonas syringae pv.lachrymans. No np-R isolate could be recovered from the upper hypocotyls or from the leaves, indicating that there was no contact between the inducer and the pathogen. Root colonization with some np-R increased seedling tolerance to low soil moisture levels.  相似文献

The objective was to identify and characterize the causal agent of foliar necrosis and leaf scorch of Eucalyptus spp. in Brazil. Nineteen putative isolates of Rhizoctonia obtained from Eucalyptus plants during clonal propagation were compared with isolates from other hosts and with tester strains of anastomosis groups of Rhizoctonia solani. Features compared were morphological characteristics of anamorphs and teleomorphs, numbers of nuclei per cell in the vegetative hyphae, anastomosis of hyphae, and ability to produce necrotic lesions on cuttings and damping-off of E. grandis×E. urophylla hybrid seedlings. Rhizoctonia solani AG1 (AG1-IB like) was the most frequent causal agent isolated from Eucalyptus plants and cuttings with symptoms of leaf scorch and foliar necrosis respectively. These isolates were highly virulent on Eucalyptus cuttings and presented naturally epiphytic growth on Eucalyptus shoots. Binucleate isolates and isolates of R. solani AG4 were also virulent on cuttings and were most virulent on Eucalyptus seedlings causing pre- and post-emergence damping-off. Virulence on Eucalyptus cuttings and seedlings was not restricted to a single species or anastomosis group of Rhizoctonia.  相似文献

Rhizoctonia solani root-rot is a major soilborne disease causing growth and yield depression. The ability of Glomus mosseae BEG12 and Pseudomonas fluorescens A6RI to suppress this soilborne disease in tomato was assessed by comparing the shoot and root growth of plants infested with R. solani 1556 when protected or not by these beneficial strains. The epiphytic and parasitic growth of the pathogenic R. solani 1556 was compared in the presence and absence of the biocontrol agents by microscopical observations allowing the quantification of roots with hyphae appressed to epidermal cells (epiphytic growth) and of roots with intraradical infection (parasitic growth). The root architecture of the tomato plants under the different experimental conditions was further characterized by measuring total root length, mean root diameter, number of root tips and by calculating degree of root branching. G. mosseae BEG12 and P. fluorescens A6RI fully overcame the growth depression caused by R. solani 1556. This disease suppression was associated with a significant decrease of the epiphytic and parasitic growth of the pathogen together with an increase of root length and of the number of root tips of inoculated tomato plants. The combined effects of G. mosseae BEG12 and P. fluorescens A6RI on pathogen growth and on root morphogenesis are suggested to be involved in the efficient disease suppression.  相似文献

Abstrast  Three-hundred-twenty-five isolates ofRhizoctonia (215R. solani and 110 binucleateRhizoctonia) were obtained from roots and crowns of alfalfa, sainfoin and common vetch grown in Erzurum, Turkey. The isolates were assigned to five anastomosis groups (AG) ofR. solani (AG-2-1, AG-3, AG-4, AG-5, and AG-10) and two anastomosis groups of binucleateRhizoctonia (AG-I and AG-K). In pathogenicity tests on alfalfa, sainfoin and common vetch, the highest disease severities were caused by isolates of AG-4 and AG-5. Isolates of AG-10 and AG-I were not pathogenic on the three tested forage legumes, whereas isolates of AG-K on alfalfa and sainfoin, and of AG-2-1 on sainfoin, were moderately virulent. Alfalfa isolate AG-3 was moderately virulent on sainfoin. This is the first report ofR. solani AG-3, AG-5, AG-10 and binucleateRhizoctonia AG-I on alfalfa. Phosphites (Phi) are alkali metal salts of phosphorous acid, with the ability to protect plants against different pathogens. In this research, the effect of Phi applied to potato plants on severity of three important potato diseases in Argentina was assessed. Seed tubers and foliage of potato cvs Shepody and Kennebec were treated with Phi to assess effects on resistance against Phytophthora infestans, Fusarium solani and Rhizoctonia solani. Protection resulting from Phi treatment in seed tubers was high against P. infestans, intermediate against F. solani, and low against R. solani. In addition, seed tubers treated with calcium or potassium phosphites (CaPhi and KPhi, respectively) at 1% of commercial product emerged earlier than untreated ones. When Phi were foliarly applied two or four times at different doses, high levels of protection against P. infestans were achieved in both cultivars. Higher protection was observed in Kennebec when CaPhi was applied, while in Shepody this was true for KPhi. Expression of β-1,3-glucanases was induced at different times after treatment but no correlation between β-1,3-glucanases expression and foliar protection level was found. On the other hand, Phi positive protection effects did not produce negative effects in plant growth. Leaves from CaPhi-treated plants showed a darker green colour than leaves from control plants; also an increase in Rubisco protein and a delay in crop senescence was observed.  相似文献

The population structure of Rhizoctonia solani AG-1 IA causing rice sheath blight from India was evaluated for 96 isolates using seven RFLP loci. Nineteen of the isolates did not hybridise to R. solani AG-1 IA RFLP probes and rDNA analyses subsequently confirmed that they were either Ceratobasidium oryzae-sativae isolates or another Rhizoctonia sp. The population structure of the remaining 77 R. solani AG-1 IA Indian isolates was similar to that of a previously characterized Texas population. Clonal dispersal of R. solani AG-1 IA in India was moderate within fields and no clones were shared among field populations. Low levels of population subdivision and small genetic distances among populations were consistent with high levels of gene flow. Frequent sexual reproduction was indicated by the fact that most populations were in Hardy–Weinberg equilibrium (HWE). The two loci (R68 and R111) that deviated significantly from HWE showed an excess of heterozygosity. Although Texas and Indian populations were geographically very distant, they exhibited only moderate population subdivision, with an FST value of 0.193.  相似文献

Water agar, artificially infested soil and leaf sheath inoculation methods were used to assess the suitable time of application, varietal host response and persistence of Rhv7, a hypovirulent, binucleate Rhizoctonia collected from soil in the Philippines, to effectively control virulent Rhizoctonia solani AG1-IA isolate RS35 on corn. With the water agar method, prior inoculation (2 to 3 days) with the biocontrol agent is essential to achieve maximum control of the pathogen. Disease protection was increased from 23 to 70% as the pre-incubation time of Rhv7 prior to the challenge inoculation with virulent isolate was lengthened from 0 to 3 days. Disease severity and incidence of banded leaf and sheath blight (BLSB) on the three corn hybrids were also suppressed in artificially infested soil. In leaf sheath inoculation, the suppressive ability of Rhv7 against BLSB on corn persisted during lesion expansion. This suppression was expressed as slower disease progress in plants with Rhv7 than in plants without Rhv7. Early and timely brace root formation that detached infected sheaths, also aided in reducing the number of diseased plants in Rhv7-treated plots at the final observation. Mycelial growth activity of RS35 was reduced when corn plants were pre-inoculated with Rhv7 before challenge even if there was no contact detected between Rhv7 and RS35, which suggests that Rhv7 protects corn against BLSB by induced resistance. The hypovirulent, binucleate Rhizoctonia Rhv7 strain effectively controlled R. solani AG1-IA isolate RS-35 in corn. Received 30 August 1990/ Accepted in revised form 25 October 1999  相似文献
