中华绒螯蟹点状产气单胞菌的分离鉴定与药敏试验   总被引:1,自引:0,他引:1  
为确定诱发中华绒螯蟹肠炎的病原菌,筛选潜在的防控药物,采用传统方法从患肠炎的中华绒螯蟹体内分离到一株病原菌(DD5),通过API 20E细菌鉴定系统和16SrDNA序列分析对菌株DD5进行了鉴定,并采用KB纸片扩散法对菌株DD5的抗菌药物敏感性进行了测定。结果表明,菌株DD5为点状产气单胞菌(GenBank登录号:KP260655),其16SrDNA序列与基因库中气单胞菌属菌株的16SrDNA序列有99%~100%的同源性,而且与点状产气单胞菌136c株(GenBank登录号:EU488688)的亲缘关系最近。此外,菌株DD5对四环素、链霉素、卡那霉素、庆大霉素、恩诺沙星、氧氟沙星、左氟沙星、复方新诺明等抗菌药物高度敏感。本研究证实点状产气单胞菌是中华绒螯蟹肠炎的病原菌,恩诺沙星、庆大霉素等常规渔药可作为点状产气单胞菌引起的中华绒螯蟹肠炎的防控用药。  相似文献

本试验根据菌体对高温和低pH的耐受性、产酶能力及对病原菌的抑菌效果,对中华绒螯蟹肠道中的芽孢杆菌进行了筛选.结果表明,在筛选到的3株芽孢杆菌中,ESB3菌株抑制中华绒螯蟹病原菌嗜水气单胞菌CL99920菌株的能力最强,产蛋白酶和淀粉酶的能力也较好,在肉汤培养基初始pH 3.0的条件下培养2 h时,其存活率为(32.56±2.87)%,90 ℃水浴处理10 min存活率为(68.54±8.28)%;浓度低于1×108CFU/mL对中华绒螯蟹无毒害作用,1×107 CFU/mL的ESB3能提高中华绒螯蟹对嗜水气单胞菌感染的抵抗力;经形态学和生理生化反应鉴定,ESB3为地衣芽孢杆菌.  相似文献

从患病蓝曼龙鱼(Trichogaster trichopterus)体内分离到1株细菌CC0323,经人工感染试验对蓝曼龙鱼有一定致病力。采用形态学观察,生理生化鉴定,16S r RNA序列分析,构建系统发育树等方法进行鉴定。结果分离株CC0323为革兰阴性短杆菌,具多形性,在RS琼脂上可生长,呈中等大小,表面光滑的淡黄色湿润菌落;理化特性和药敏试验结果与维氏气单胞菌基本相同;系统发育树可见CC0323与维氏气单胞菌QXL0711B进化关系最近。试验结果,证明分离株CC0323为维氏气单胞菌(Aeromonas veronii)。  相似文献

对患病黄喉拟水龟(Mauremys mutica)进行细菌学检查和分离,对分离到1株革兰阴性细菌进行形态特征、理化特征、VITEK 2 ZOMPACT 30自动微生物鉴定系统鉴定、结合16S rDNA序列的分子鉴定,并构建系统发育树;进行人工感染试验及药物敏感性试验。结果分离到1株优势菌,根据此株分离菌的形态特征、理化特征、自动微生物鉴定系统鉴定、结合16S rDNA序列测定与系统发育树分析结果,判定其为气单胞菌属的维氏气单胞菌。此菌对供试健康黄喉拟水龟和小鼠有较强的致病作用,对供试20种抗菌药物中的头孢吡肟、阿米卡星、氧氟沙星、米诺环素高度敏感;对洛美沙星、恩诺沙星、氯霉素等3种中度敏感;对头孢噻吩、环丙沙星等13种表现耐药。表明所检出的维氏气单胞菌,是导致黄喉拟水龟以出血和腹水为主要病理特征的致病菌。  相似文献

对1例鸡鼻炎样病例进行细菌学诊断,从病鸡鼻腔中分离到1株革兰阴性杆菌,编号为ZY17105,对其进行形态学、生理生化、分子生物学鉴定,对其致病性和耐药性进行分析,同时探讨gyrB基因在气单胞菌鉴定中的意义。16S rDNA进化分析显示ZY17105与圣雷利气单胞菌模式菌株LMG24682同源性为99.4%;gyrB基因进化分析显示ZY17105与各圣雷利气单胞菌株形成同一进化支,与圣雷利气单胞菌模式菌株LMG24682和A2-67之间的同源性为分别为97.8%和98.2%。根据形态学、生理生化特性、16S rDNA和gyrB基因进化分析,将分离株ZY17105鉴定为圣雷利气单胞菌,分离株ZY17105对小鼠和鸡有较强致病性;另外,进化分析表明gyrB基因在气单胞菌属各种间的区分能力高于16S rDNA。本研究首次从中国大陆分离到圣雷利气单胞菌,并证实gyrB基因可替代16S rDNA用于气单胞菌属各菌种间的进化分析鉴别。  相似文献

中华鳖致病性嗜水气单胞菌的分离鉴定及药敏试验   总被引:3,自引:0,他引:3  
对江西省进贤县某中华鳖养殖场送检的2只患病中华鳖进行细菌分离,且对分离的细菌进行培养特性观察、生化特性鉴定、毒力因子检测、毒力基因PCR检测、实验动物攻毒及体外药敏试验。结果表明,从2只中华鳖肝脏分离到3株菌,均为革兰阴性短杆菌,其中体型稍大中华鳖2株,体型稍小中华鳖1株;经过实验动物感染试验得出3株菌都为强毒力致病菌;通过生化鉴定、毒力因子及PCR检测确定为嗜水气单胞菌;体外药敏试验结果显示,3株菌对头孢曲松都高度敏感,对利福平、甲氧苄啶、氨苄青霉素都不敏感。  相似文献

为筛选青海地区池塘养殖中华绒螯蟹最佳的水草覆盖度,对中华绒螯蟹在不同水草覆盖度下的体重、成活率及生长发育情况进行了比较。结果表明,当池塘水草覆盖度在60%~80%时,有利于中华绒螯蟹的生长发育,且成活率较高,规格较大。  相似文献

对锦州及周边地区3个水库30尾病鱼进行细菌分离培养、染色镜检、生化试验和PCR扩增气溶素基因aer鉴定,确定6株为嗜水气单胞菌。进一步采用纸片扩散法测定嗜水气单胞菌分离株对多种抗生素的敏感性,各分离菌株均对环丙沙星、氯霉素、强力霉素、妥布霉素敏感;对氨苄青霉素、青霉素不敏感;部分菌株存在不同程度的耐药性。接种试验动物出现典型的嗜水气单胞菌的病理变化和死亡。  相似文献

猪源气单胞菌分离鉴定及耐药谱检测   总被引:1,自引:0,他引:1  
为调查气单胞菌在我国猪粪便中流行情况及耐药谱,2017年3~7月从河南省4个地区,共采集370份猪粪样品,通过缓冲液蛋白胨水增菌培养后,使用添加有氨苄青霉素的气单胞菌基础培养基选择培养,分离气单胞菌。可疑菌株通过形态特征、生化试验和16SrDNA序列比对进一步鉴定。微量稀释法测定分离菌株对17种抗菌药物耐药谱。结果显示,共分离到11株气单胞菌,分离率为2.97%,其中豚鼠气单胞菌5株(占45.45%),杀鲑气单胞菌2株(占18.18%),中间气单胞菌、嗜水气单胞菌、水生气单胞菌和贝斯特润气单胞菌各1株(各占9.09%);阿莫西林/克拉维酸菌耐药率为100%,磺胺甲恶唑、氟苯尼考耐药率81.82%,阿米卡星、美罗培南敏感率均为100%,除了其中1株气单胞菌,其他菌株对头孢噻呋、头孢曲松、庆大霉素、强力霉素、磷霉素、恩诺沙星均敏感,各菌株均呈多重耐药性。表明我国健康猪粪便中携带气单胞菌,如果造成水源或者食物污染,可能有引起人类或者动物感染的风险,应引起重视。  相似文献

对引起丁鲑鱼发病的菌株——XN602进行鉴定和生物学特性研究,经形态特征与生理生化鉴定,结果表明:该菌为革兰氏阴性杆菌,其16SrDNA序列与GenBank中多株维氏气单胞菌相似性高达99%,系统发育树显示该菌与维氏气单胞菌聚为一簇。因此,分离菌株XN602在分类学上应属于维氏气单胞菌。  相似文献

In this study we describe biochemical, toxigenic and surface characteristics of 33 motile Aeromonas isolated from diseased mammals, 3 from moribund marine mammals, 24 from healthy fish and 4 from moribund fish. Aeromonas hydrophila, A. caviae and A. sobria were isolated from both mammals and fish but at a different incidence. Aeromonas hydrophila was the predominant species isolated from clinical specimens; it was isolated from pneumonia, wound infections, septicemia and abortion in horses, cattle and pigs. Aeromonas sobria was isolated from one mammal and 11 healthy fish. Aeromonas caviae was isolated in 2 cases from healthy fish and in 9 cases from diseased mammals. Variations in some biochemical tests including sorbitol, amylase and citrate, were observed between isolates from different sources. However, these differences did not allow the differentiation of isolates from diseased mammals and healthy fish. The majority of A. hydrophila isolates produced different extracellular products; A. sobria isolates produced less exotoxin. With A. caviae isolates no hemolysin, protease, enterotoxin or elastase were detected. There was no quantitative difference in hemolysin, protease, enterotoxin or elastase production between isolates from mammals and fish. It is suggested that A. hydrophila could be a potential pathogen for domestic animals, and fish may represent a potential reservoir of infection.  相似文献

对江苏省吴县、吴江等地区养殖的患“腐皮病”的中华鳖的病原进行了分离,从病鳖的肝脏分离得菌株B-1,用菌株B-1进行人工感染,100%的鳖患病,出现与自然病鳖相同的症状。对B-1株进行菌体形态特征、培养特性、生理生化反应及致病因子检测,证明该菌为致病性嗜水气单胞菌。  相似文献

In November 1987 high losses of carp (Cyprinus carpio) with the main symptom of skin ulcera were observed in a farm in northern Greece. Sixty-six isolates of bacteria, characterized mainly as Aeromonas hydrophila or Pseudomonas spp. could be isolated from lesions of diseased fish. Transmission experiments with these isolates using mirror carp showed that Aeromonas hydrophila strains induced identical clinical and pathological pictures after intra- or subcutaneous injection. Extracts of these Aeromonas hydrophila isolates, as well as a supernatant of culture bouillon were toxic for carp and mice, indicating the presence of endo- and exotoxins. The results prove that carp erythrodermatitis (CE) may be caused by different bacteria, mainly including A. hydrophila.  相似文献

AIMS: To use multilocus enzyme electrophoresis to determine the genetic structure of Staphylococcus intermedius from normal skin of dogs and those isolated from a variety of disease conditions and to distinguish clinically important strains in dogs. METHODOLOGY: The diversity amongst 129 isolates of S intermedius from the skin and mucosa of 32 healthy dogs and 120 isolates from diseased sites in 120 individual dogs was examined using multilocus enzyme electrophoresis. Associations among ETs were examined to determine the diversity of isolates. RESULTS: Twenty two ETs were distinguished comprising 21 containing isolates from diseased sites and 11 containing isolates from normal dogs. The majority of isolates (171 of 249; 69% were located in two ETs (ET1 and ET 4), that were not distinguishable phenotypically. ET 1 contained 94 isolates (54 isolates from healthy dogs and 40 isolates from diseased sites) and ET 4 contained 77 isolates (46 from healthy dogs and 31 isolates from diseased sites). Further, 77.5% of isolates from healthy dogs were present in ET 1 and ET 4 and 59% of isolates from diseased dogs belonged to the same two ETs. There was only a small difference in genetic diversity among isolates taken from healthy dogs (11 ETs; H = 0.182) and those isolates taken from clinical specimens from diseased dogs (21 ETs; H = 0.218). Of the 21 ETs from diseased sites, ET 16 contained all six isolates from Staphylococcal Scalded Skin Syndrome in racing Greyhounds. CONCLUSIONS: The small difference in genetic diversity between isolates from the skin and mucosa of healthy dogs and isolates from various diseases, as well as the presence of the majority of isolates in two ETs, is consistent with the role of S intermedius as an opportunistic pathogen. The confinement of all Staphylococcal Scalded Skin Syndrome isolates within one ET is confirmation of this entity as a distinct disease of dogs.  相似文献

江苏某养殖场种鸭突然发生散发性死亡,为了探究其死亡原因,本研究通过临床解剖观察、病原分离、生化试验、药敏试验、雏鸭人工感染试验和致病性试验对分离菌株进行检测。结果显示该分离菌为革兰阴性短杆菌,在麦康凯培养基和LB培养基中呈现圆形灰白色菌落,在血琼脂培养基中呈现出β溶血;16S rRNA基因序列分析结果显示该菌为维氏气单胞菌(A.veronii);药敏结果显示该菌对头孢氨苄、氨苄西林等10种抗生素敏感;人工感染试验显示该菌可造成雏鸭的死亡;小鼠的致病性试验显示该菌对小鼠的LD50为5.0×10^6.5cfu。总之,本实验从病鸭中分离到1株A.veronii,并筛选出敏感药物,为A.veronii病的诊断和治疗提供了参考依据。  相似文献

从上海某水族馆因血痢、厌食、消瘦而死亡的秘鲁帝企鹅的心脏、肝脏中分离到2株细菌SH051237和SH060104,其培养性状及生化特性基本一致,经生化试验、VITEK试验等鉴定为嗜水气单胞菌.动物试验表明,分离株对小鼠均具有较强的致病性,且从死亡鼠回收到相应细菌.结果表明,分离到的菌株为致病性嗜水气单胞菌.  相似文献

Escherichia coli isolates from calves were investigated by multiplex PCR assays for the presence of genes encoding K99, F41, F17-related fimbriae, heat-stabile enterotoxin a (STa), intimin (eae) and Shiga toxins (stx1 and stx2). A total of 120 E. coli isolates, 75 isolated from diarrhoeic or septicemic calves and 45 from clinically healthy calves aged between 1 day and 2 months were tested. Each isolate was obtained from different calves in different herds. Among the isolates from diseased animals, 12 (16%) isolates from 1- to 7-day-old diarrhoeic calves were detected as enterotoxigenic E. coli which possessed K99, F41 and STa in combination; F17-related fimbriae genes were detected in 33 (44%) isolates and they were found in combination with K99 + F41 + STa in two isolates. Of 120 isolates, 16 carried eae, eight stx1 and five stx2 genes alone or in combination. None of the eae- or stx-positive strains was identified as O157:H7. However, results indicate that calves may be carrier of Shiga toxin-producing E. coli which have potential as a human pathogen. Antimicrobial susceptibility of 75 isolates from diseased calves was determined by agar disk diffusion method for 14 antimicrobial agents. In 77.3% of the isolates, multiresistance was detected. Higher resistance rates were detected for cephalothin (72%), tetracycline (69.3%), kanamycin (69.3%), ampicillin (65.3%), nalidixic acid (53.3%), trimethoprim-sulphamethoxazole (52%) and enrofloxacin (41.3%), respectively. No resistance was found for ceftiofur and cefoxitin.  相似文献
