The purpose of this study was to measure the net absorption of fluid, sodium, potassium and chloride in the small intestine of weaned pigs and of their unweaned littermates and to correlate these values with villus height and crypt depth. Five pairs of segments of the small intestine were prepared in each of 80 pigs; the cranial segment of each pair was injected with an enterotoxigenic strain of Escherichia coli and the caudal segment with a control solution. Net absorption was measured on the day of weaning and four, seven, 11 and 14 days after weaning. In unweaned pigs the net absorption of fluid, potassium and chloride did not vary with time. In weaned pigs the net absorption of fluid in the control segments was significantly less on days 4, 7 and 14 after weaning and of sodium and chloride on days 4 and 7 than in unweaned littermates. In infected segments of weaned pigs the net absorption of fluid was significantly less than in unweaned littermates on day 11 and 14, of sodium and potassium on day 11 and of chloride on days 4 and 11 after weaning. Net absorption was negatively correlated with villus height but only in the infected segments of weaned pigs; no other correlations were found. It was concluded that after weaning the net absorption of fluid and electrolytes in the small intestine of pigs is temporarily decreased, a condition that may initiate diarrhoea.  相似文献

The aim of the study was to measure the effect of creep feeding during lactation on net absorption in the small intestine at 4 days after weaning. Intermittent suckling was used to increase creep feed intake during lactation. Creep feed containing chromic oxide was provided. Based on the colour of the faeces, piglets were classified as ‘eaters’ or ‘non-eaters’, respectively. At day 4 after weaning, an in vivo small intestine segment perfusion test was performed at 5 sites along the small intestine in 24 piglets (12 eaters and 12 non-eaters). At both sides of each intestinal segment a tube was fitted to perfuse and drain fluid in order to assess net absorption. Net absorption was higher in eaters than in non-eaters (P < 0.001). Net absorption varied greatly between and within piglets and was highest in the caudal segments of the small intestine (P < 0.001). These data suggest that creep feeding could be a useful tool in the prevention of post-weaning diarrhoea.  相似文献

Electrolyte concentrations, osmolality and pH were determined in conventionally raised weanling swine fed a liquid diet. Incorporation of a dilution marker into the diet in combination with frequent feeding enabled estimations as to the sites of relative fluid and electrolyte absorption and secretion along the gastrointestinal tract. Unlike many other species the weanling pig depends largely on its large intestine for absorption of fluid and electrolytes with small changes in net fluid movement occurring along the jejunal and ileal segments. Additional observations included the absorption of water by the porcine stomach which increased dilution marker concentration by approximately twofold and the high osmolality values recorded in the small and large intestine. The implications of these observations are discussed with regard to pathogenesis of colibacillary diarrhea in the weanling pig.  相似文献

The objective of the current study was to investigate whether plasma citrulline or intestinal fatty acid-binding protein (I-FABP) concentrations might be used as longitudinal markers for small intestinal function in piglets after weaning. Plasma citrulline and I-FABP concentrations were measured longitudinally in weaned and unweaned piglets, and related to intestinal absorption values (i.e., plasma mannitol and 3-xylose concentrations in a sugar absorption test). Within each litter (n = 10), 2 piglets with a close-to-litter-average BW were selected. At 20.8 +/- 0.4 d of age, the selected piglets per litter were either weaned conventionally (CW) or remained with the sow (UNW). One day before, and 0.5, 2, 4, and 7 d after weaning of the CW piglets, the selected piglets of both groups were subjected to a sugar absorption test. After a 2-h fast, piglets were administered an oral dose of 2 mL/kg of sugar solution, containing 50 mg/kg of mannitol and 100 mg/kg of 3-xylose. One hour after administration, a blood sample was collected from a jugular vein for determination of plasma I-FABP, citrulline, mannitol, and 3-xylose concentrations. Plasma I-FABP concentration showed great variation within treatments, and no difference was observed in plasma I-FABP concentrations between the CW and UNW treatments (P = 0.63). The absorption of 3-xylose was not different between treatments (P = 0.83). Mannitol absorption, however, was less in the weaned CW piglets compared with the UNW piglets (P = 0.003), with the nadir on d 4 postweaning. Weaning also reduced plasma citrulline concentrations in the CW treatment compared with the UNW treatment (P < 0.001). On d 4 and 7 postweaning, plasma citrulline concentrations of CW piglets were less (P < 0.001 and P = 0.0013) than preweaning values. Furthermore, in the CW treatment, plasma citrulline concentrations correlated with plasma mannitol concentrations at d 4 postweaning (r = 0.89, P = 0.008) and overall (r = 0.76, P = 0.001). Based on these results, plasma citrulline concentration seems to be a possible marker for monitoring intestinal function in pigs after weaning.  相似文献

Acute, isolated loops of proximal and distal jejunum of weanling swine were exposed to either heat stable porcine Escherichia coli enterotoxin, heat labile porcine Escherichia coli enterotoxin, cholera toxin or theophylline. Unidirectional sodium fluxes in response to heat stable in the proximal jejunum were dependent on the length of time that the intestinal mucosae was exposed to the enterotoxin. Net water, sodium and chloride and unidirectional sodium and chloride flux measurements in the proximal jejunum in response to each agent uniformly indicated that net secretion of fluid and electrolytes was the result of increased unidirectional sodium secretion or blood-to-lumen flux and decreased unidirectional chloride absorption or lumen-to-blood flux. In addition heat stable cholera toxin and theophylline but not heat labile decreased unidirectional chloride secretion a small but significant amount in the proximal jejunum.

Sodium and chloride flux measurements in the distal jejunum demonstrated that all four secretory agents could stimulate net secretion of water, sodium and chloride in that region. The response to these secretory agents as measured by sodium and chloride unidirectional flux rates was not similar to changes observed in the proximal jejunum. In the distal small intestine, whereas heat labile cholera toxin and theophylline induced similar qualitative changes in unidirectional sodium and chloride fluxes, that induced by heat stable differed.

Isosmolar oral rehydrating solutions (ORS) used to treat diarrheic calves are low in energy. In contrast, hyperosmolar ORS may have a dehydrating effect. This study was designed to compare the net water and glucose absorption from the small intestine of normal calves given isosmolar or hyperosmolar ORS. Six calves were used for the experiment. A reentrant ileal cannula was implanted surgically. Each calf was fed with either the isosmolar or the hyperosmolar solution. The next day, the second solution was given. Both contained chromium EDTA as a nonabsorbable marker. Changes in the hematocrit, total plasma protein, plasma osmolality, plasma glucose concentration, and ileal volume, osmolality, glucose content, and chromium concentration were monitored for 10 hours. The only statistically significant difference was in the post-feeding plasma glucose level: it peaked at the higher level (p less than 0.05) in calves fed the hyperosmolar solution. In normal calves, the hyperosmolar ORS had no effect on the hydration or on the small intestinal fluid composition.  相似文献

Enterotoxigenic Escherichia coli (ETEC) infection results in fluid and electrolyte losses in the small intestine. We investigated the effect of non-starch polysaccharides (NSP) hydrolysis products of wheat middlings (WM) and flaxseed (FS) on net absorption of fluid and solutes during ETEC challenge. Products were generated by incubating WM and FS with a blend of carbohydrase enzymes to produce 2 products: 80% ethanol-soluble (ES) and 80% ethanol-insoluble (EI) which were studied in 2 experiments in which 2 factors were investigated: products (EI vs. ES) and time of ETEC challenge (before vs. after perfusion). Pairs of small-intestine segments, one non-challenged and the other ETEC-challenged were perfused with products for 7.5 h. ETEC reduced fluid absorption by more than 380 µL/cm2 in saline (control) perfused segments, whereas this reduction was significantly ( P  < 0.05) less for the WM and FS products. Interaction ( P  > 0.05) between product and time of challenge was not observed. For WM, products effects on ETEC-challenged segments were such that perfusion of ES resulted in higher total solute (measured as osmolality) absorption than EI (138 vs. 103 µOsmol/cm2). In conclusion, hydrolysis products from WM and FS were beneficial in maintaining fluid balance during ETEC challenge, suggesting potential in controlling ETEC induced diarrhea in piglets.  相似文献

The effects of single IV injections of sodium bicarbonate (0.5 mEq/kg of body weight, 1 mEq/kg, 2 mEq/kg, and 4 mEq/kg) on serum osmolality, serum sodium, chloride, and potassium concentrations, and venous blood gas tensions in 6 healthy cats were monitored for 180 minutes. Serum osmolality increased and remained significantly (P less than 0.05) increased for 120 minutes in cats given 4 mEq of sodium bicarbonate/kg. Serum sodium was increased significantly (P less than 0.05) for 30 minutes in cats given 4 mEq of sodium bicarbonate/kg. Serum sodium decreased and remained significantly (P less than 0.05) decreased for 120 minutes in cats given 1 g of 20% mannitol/kg, and serum osmolality was significantly (P less than 0.05) decreased at 30 and 60 minutes. Serum chloride decreased significantly (P less than 0.05) for 10 minutes in cats given 1 mEq of sodium bicarbonate/kg, and was significantly decreased for 30 minutes in cats given 2 mEq and 4 mEq of sodium bicarbonate/kg. Serum chloride decreased and remained significantly (P less than 0.05) decreased for 30 minutes in cats given 1 g of 20% mannitol/kg. Serum sodium and serum osmolality did not change significantly (P less than 0.05) in cats given 4 ml of 0.9% sodium chloride/kg. Serum potassium decreased significantly (P less than 0.05) for 10 minutes in cats given 1 mEq of sodium bicarbonate/kg, and for 120 minutes in cats given 2 mEq/kg or 4 mEq/kg. Intestinal function was assessed in cannulated loops of porcine proximal jejunum during immune-mediated emigration of neutrophils into the intestine. Net water, net sodium, net chloride, undirectional sodium and unidirectional chloride fluxes were measured before and after intestinal exposure to an antigen in both sensitized and nonsensitized pigs. Neutrophil emigration was assessed histologically. The results indicate that fluid and electrolyte movements in the intestine are not significantly altered during immune-mediated enteroluminal neutrophil emigration suggesting that the neutrophil response does not interfere with intestinal function.  相似文献

The effect of an intravenous (i.v.) infusion of hypertonic saline solution (HSS; 7.2%, 2,400 mOsmol/kg.H2O) was evaluated by serum electrolyte concentrations and osmotic pressure in the anesthetized beagles. Sixteen beagles were assigned to 3 experimental groups (2.5, 5 or 15 ml/kg of HSS i.v. infusion) or a control group (5 ml/kg of isotonic saline solution (ISS) i.v. infusion) and were monitored for 120 min after the initiation of fluid infusion. The relative plasma volume (rPV) in the 5 ml/kg and 15 ml/kg HSS groups progressively expanded to 143.1 +/- 7.4% at 3 min and 156.4 +/- 5.9% at 5 min after the initiation of the fluid infusion, respectively. Significant increases were not produced by ISS and 2.5 ml/kg HSS infusion. The serum sodium and chloride concentrations in the ISS group were not altered. The 5 ml/kg HSS infusion induced transient high osmotic and sodium levels, and the serum sodium concentration remained under the 160 mM/l after the completion of the HSS infusion. However, the 15 ml/kg HSS infusion induced a constant high osmotic level (340.5-352.8 mOsmol/kg.H2O) and hypernatremia (161.4-174.5 mM/l) from 10 to 90 min after the initiation of the fluid infusion. The 15 ml/kg HSS infusion induced significant decreases in the partial pressure of oxygen (PaO2), reaching 63.7 +/- 8.0 mmHg at 120 min after the initiation of the fluid infusion compared with an immediately before fluid infusion value. On the basis of these findings, 5 ml/kg HSS infusion can be safely administered to healthy beagles for expanding the plasma volume without inducing hypernatremia. A 5 ml/kg HSS infusion is thus recommended for the initial field resuscitation of dogs.  相似文献

The present study evaluated the biochemical profile of horses that received hypotonic electrolyte solutions associated with energy sources by enteral route in continuous flow using a small caliber tube for naso-esophageal administration. Experimental design was a latin square 3 × 3 with two replicates and three periods. The assays were carried out with “six adult females horses”, divided into three groups submitted to the following treatments each: electrolyte solution containing dextrose (ESDext)—“5 g sodium chloride, 0.5 g potassium chloride”, 1 g of calcium gluconate, 200 mg of magnesium pidolate, and 15 g of dextrose diluted in 1,000 mL of water with measured osmolarity of 264 mOsmol/L; electrolyte solution containing maltodextrine (ESMalt)—5 g of sodium chloride, 0.5 g of potassium chloride, 1 g of calcium gluconate, 200 mg of magnesium pidolate, and 15 g of maltodextrin diluted in 1,000 mL of water with measured osmolarity of 203 mOsmol/L; and electrolyte solution containing sucrose (ESSucr)—“5 g sodium chloride, 0.5 g potassium chloride”, 1 g of calcium gluconate, 200 mg of magnesium pidolate, and 15 g of sugar diluted in 1,000 mL of water with measured osmolarity of 234 mOsmol/L. The electrolyte solutions were administered at the dosage 15 mL/kg/h during 12 hours. Hypotonic enteral electrolyte solutions that contain maltodextrin (ESMalt) and dextrose (ESDext) were effective to increase glycemia in horses without causing any adverse effects, whereas ESSucr presented slight effect on blood glucose, but without causing electrolyte imbalances.  相似文献

The purpose of this study was to test whether dietary spray-dried porcine plasma (SDPP) in early-weaned piglets prevents small intestinal villus atrophy by trophic or protective activity. Fifty-four weaned, 18-day-old piglets were used to determine the effect of dietary SDPP on small intestinal villus length, crypt depth, enterocyt mitotic activity and brush border enzyme activities during the first week after weaning. The piglets were offered a diet containing either 8% SDPP or 8% casein. At 2 and 7 days after weaning, piglets were anaesthetized to provide samples of the small intestinal wall and killed immediately afterwards. There were no differences in daily gain and daily feed intake between the two dietary treatments. At day 2 after weaning, all piglets showed a marked reduction in villus height when compared with baseline values. In all piglets, small intestinal enterocyte mitotic activity had decreased by day 2 and was increased again on day 7. There were no significant effects of dietary SDPP on small intestinal villus length, crypt depth and enterocyt mitotic activity. This indicates that SDPP has no trophic effect on the small intestinal mucosa and that it does not protect against the damaging effect on the small intestinal villi that is associated with the process of weaning. There was no effect of SDPP on lactase-, sucrase- or maltase-specific activities that are a measure of the digestive function of the small intestine. It can be concluded that SDPP versus casein has no effect on small intestinal morphology and disaccharidase activities in early weaned piglets kept under low infection pressure.  相似文献

Developmental changes of the gastrointestinal tract were probably responsible for the changes in faecal composition during the first week of the foals' life, which resembled small intestinal ingesta of adult horses, suggesting a minimal colonic modification. Faecal composition at the time of foal heat diarrhoea was suggestive of a secretory-type diarrhoea, in that the electrolyte concentration accounted for most of faecal osmolality and the faecal pH was alkaline. After foal heat diarrhoea faecal composition slowly approached that of adult horses. These data suggest that foal heat diarrhoea is most likely caused by hypersecretion in the small intestinal mucosa, which may overwhelm an immature colon that is unable to compensate by increased fluid and electrolyte absorption.  相似文献

Objectives

To establish whether chondrocyte viability, matrix degradation and the induction of proteolytic gene expression in canine cartilage is independent of irrigation fluid osmolality and time following exposure to the irrigation fluid.

Methods

Canine cartilage explants were exposed to one of three different solution types i) Culture medium (270–280 mOsmol/kg) ii) NaCl 0.9% (302 mOsmol/kg) iii) NaCl 0.9% with sucrose (600 mOsmol/kg). Chondrocyte viability and selected proteolytic gene expression were measured at two time points; immediately following exposure and 24 h following exposure. The media samples at 24 h following exposure were assessed for sulphated glycosaminoglycan (sGAG) release.

Results

In all samples, no cell death was observed across the superficial or deeper layers of the cartilage. When adjusting for time, gene expression was not shown to be dependent on solution type. However for all solution types, Matrix Metalloproteinase 13 (MMP13) and A Disintegrin and Metalloproteinase with Thrombospondin Motifs 5 (ADAMTS5) expression was significantly decreased in cartilage samples at 24 h post exposure comparatively to samples tested immediately post exposure. No significant differences were identified in the relative sGAG release between the solution types.

Clinical Signifcance

Arthroscopic solution irrigation of cartilage explants had no effect on cell viability or proteinase production. At present there is no indication to optimise irrigation fluid osmolarity, as conventional arthroscopic solution was not deleterious to healthy cartilage in this model.

Chloride secretion in the intestines of pigs of different age (32 days and 4 months) was examined using the Ussing chamber technique. After stimulating chloride secretion by carbachol and forskolin, alternative chloride channels and finally Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) channels were blocked. While basal short-circuit currents (Isc) were higher in the jejunum than in the colon of the piglets of 32 days, it was inverse for the 4-month-old fatteners. In the jejunum responses to stimulation and final levels of Isc were higher in the piglets than in the fatteners. The decrease in Isc after blocking CFTR channels was smaller in the fatteners than in the piglets in both intestinal segments. In conclusion, 32-day-old piglets show a higher basal chloride secretion as well as a higher response to stimulation in the jejunum than 4-month-old pigs.  相似文献

The effect of heat stable and heat labile Escherichia coli enterotoxins or cholera toxin in combination with theophylline on net water, sodium and chloride and unidirectional sodium and chloride fluxes was examined in acute isolated loops of jejunum of weanling swine. The effect of heat stable enterotoxin in combination with theophylline was determined in loops located in the proximal jejunum, while combinations of theophylline and either heat labile enterotoxin or cholera toxin were studied in the distal jejunum. In each situation the addition of theophylline resulted in an additive rather than a synergistic increment of intestinal secretory activity. This study implies that the intestinal adenyl cyclase system and enterotoxin induced intestinal secretion may not be directly related in the swine small intestine.  相似文献

Saline or glucose solution was infused for approximately 4 hours into six healthy mares in two separate experiments to determine the effect of infusion of crystalloid solutions on fractional excretion (FE) of sodium (Na), chloride (Cl), potassium (K), and phosphorus (P), ratio of urinary creatinine to serum creatinine (UCr/SCr), and ratio of urinary osmolality to serum osmolality (Uosm/Sosm). After intravenous infusion of either saline or glucose solution, FENa, FECl and FEP were significantly increased, whereas UCr/SCr and Uosm/Sosm were significantly decreased. In addition, FEK was significantly increased after infusion of glucose solution. It was concluded that urinary indices were altered by intravenous infusion of crystalloid solutions in healthy mares and that fluid therapy may interfere with the use of these indices for diagnostic purposes. Faecal samples of different consistency from 15 calves were subjected to two methods to separate faecal fluid. Osmolality as well as the concentrations of electrolytes and short chain fatty acids were determined for each of the fluid samples obtained and the results compared. For dialysis the following procedure was used: small dialysis bags filled with dextran 10% were placed in the cooled (4 degrees C) faecal sample to which gentamycin was added to inhibit fermentation. Equilibrium between dialysate and surrounding fluid was reached after 24 hours. However, depending on the consistency of the faeces, after 6 hours 90 to 95% of the final osmolality at 24 hours was reached. In regard to osmolality, sodium, potassium and chloride concentrations as well as to concentrations of short chain fatty acids no significant differences could be observed between the faecal fluid obtained by dialysis or by centrifugation (3500 g, 20 min, 4 degrees C) and subsequent filtration. Distinctly higher concentrations of calcium and inorganic phosphate were measured in the filtrates; the probable explanation is that these ions in faeces are largely present as poorly diffusible compounds (in particular with faeces of a more solid consistency) or bound to the cell membranes.  相似文献

125I-labeled immunoglobulin (Ig)G1 in colostral whey was used to determine the region of maximum absorption of Ig from the small intestine of the neonatal calf and the variation in Ig absorption among calves at the intestinal level. In experiment 1, 5 segments (approx 5%, 35%, 60%, 80%, and 95% of the duodenocecal length) were formed in the small intestine of 9 colostrum-deprived calves shortly after birth. These segments were injected with colostral whey containing 125I-IgG1 4 hours after birth, and uptake, transfer, and absorption (defined as uptake plus transfer) were determined for each segment 2 hours later. Raw data were adjusted for the milligrams of IgG1 injected per gram of intestinal tissue to obtain the least squares mean (LSM) value. The LSM values for absorption of IgG1 from distal segments 3, 4, and 5 were significantly greater (P less than 0.05) than those values for proximal segments 1 and 2. The region of the maximum IgG1 absorption was the lower small intestine, 60% to 80% of the duodenocecal length. There was also an indication of independence between uptake and transfer in each of the segments. Significant differences (P less than 0.05) were present among calves in the LSM values for uptake and absorption, but not for transfer. In experiment 2, thoracic ducts of 8 newborn calves were cannulated 4 to 5 hours after birth. Responses of jejunal transcapillary and transmucosal fluid fluxes to IV infusion of crystalloid or colloid solutions were evaluated in 12 dogs. One isolated intestinal segment in each dog was used as the control segment, and 2 segments were distended to a intraluminal hydrostatic pressure of 10 cm of H2O. The artery supplying 1 of the 2 distended (autoperfused) segments was cannulated and perfused with blood from the femoral artery. One of the 2 distended segments was autoperfused from the femoral artery. Intraluminal pressure was increased in the autoperfused segment and in 1 other segment for three, 20-minute periods after administration of the crystalloid or colloid solution. Net transmucosal fluid flux was estimated, using a volume recovery method. In each autoperfused segment, blood flow, capillary pressure, lymph flow, and plasma protein and lymph protein concentrations were measured during each 20-minute distention period. Systemic arterial pressure was monitored throughout the procedure. Plasma and tissue oncotic pressures were calculated from the plasma protein and lymph protein concentrations. Total vascular resistance and precapillary and postcapillary resistances were determined. Capillary pressure increased after infusion with colloids and crystalloids, with the effects being more prolonged in the colloid group. Plasma oncotic pressure transiently increased after infusion with colloids and decreased after infusion with crystalloids. Lymph flow increased only in crystalloid-treated dogs. Due to alterations in transcapillary fluid filtration, crystalloids induced a net loss of fluid into the intestinal lumen, whereas the fluid absorptive capacity of the jejunum was unaltered by colloid treatment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
