The accumulation potential of six, structurally related, dinitroaniline herbicides was investigated in an aquatic ecosystem. The herbicides investigated were trifluralin (α,α,α-trifluoro-2,6-dinitro-N,N-dipropyl-p-toluidine), profluralin [N-(cyclopropylmethyl)-α,α,α-trifluoro-2,6-dinitro-N-propyl-p-toluidine], dinitramine [N3,N3-diethyl-2,4-dinitro-6-(trifluoromethyl)-m-phenylenediamine], chlornidine [N,N-bis(2-chloroethyl)-2,6-dinitro-p-toluidine], fluchloralin [N-(2-chloroethyl)-2,6-dinitro-N-propyl-4-(trifluoromethyl)aniline], and butralin [4-(1,1-dimethylethyl)-N-(1-methylpropyl)-2,6-dinitrobenzenamine]. The herbicide (0.1 mg) plus 1 μCi of 14C-labeled herbicide was adsorbed on 100 g of soil (1 ppm), added to individual aquariums, and flooded with 4 liters of water. Algae, snails, and daphnia were added, and 14C in water was monitored for 30 days. Fish were added on Day 30, and all components were harvested 3 days later. Bioaccumulation ratios (concentration in organism/concentration in water) for fish depended on the amount of their exposure to sunlight: Aquariums held in the dark had higher ratios for fish (235–755) than did those exposed to sunlight (32–83). Bioaccumulation ratios in the dark for fish based on 14C from bound soil residues of butralin and profluralin were 76 and 119, respectively. Direct repeated applications of profluralin (without soil) at 4-day intervals resulted in a rapid increase, then a decrease in bioaccumulation ratios for Gambusia, but a continuous increase for catfish.  相似文献

ABSTRACT A novel sensor for measuring the kinetic energy of impacting raindrops, developed based on a soil-mass erosion sensor, was tested in the laboratory, with a rain simulator, and in the field. Drop impactions on the sensor-consisting of a piezoelectric crystal and associated electronics-produce an electrical charge that equals a fixed amount of energy. Calibration of the sensor was done in the laboratory using water drops of known diameter impacting with known velocity, and thus, with known kinetic energy. The relationship between pulse-count output of the sensor minus the background pulse counts when no drops were impacting (O; per min) and kinetic energy flux density (i.e., power [P; mJ cm(-2) min(-1)]) was found to be described by the formula P; = (0.204 + 0.065 . O)(0.67). The measurement threshold was 0.34 mJ cm(-2) min(-1). Using the sensor, generated rains with intensities of 23 to 48 mm/h were found to have powers of 0.4 to 2.2 mJ cm(-2) min(-1). In 2 years of field testing, 85 individual rain episodes were monitored, with mean intensities ranging from 0.1 to 42 mm/h. These rains had mean powers ranging from 0 to 5 mJ cm(-2) min(-1), and the highest power for a 5-min sampling period was 10 mJ cm(-2) min(-1). Both power and intensity varied greatly over time within rain episodes, and there was considerable variation in power at any given rain intensity, emphasizing the importance of measuring rather than simply predicting power. Although there was no known true power measurements for the generated or natural rains, estimates were realistic based on theoretical calculations, assuming that the gamma distribution represents raindrop sizes. The sensor is important in assessing the risk of rain splash dispersal of plant pathogens.  相似文献

植物病原菌产生蛋白水解酶被认为是病原菌侵染寄主的一个重要策略。作者对水稻纹枯病菌(Rhizoctonia solani Kühn)产生的胞外蛋白酶的特性进行了研究。结果表明,水稻纹枯病菌产生一个分子量约为49.5ku的胞外蛋白酶。该酶对胰蛋白酶专一性底物Benz-Phe-Val-Arg-NA有较高的活性,并能从羧基端降解Arg和Lys。胰蛋白酶抑制剂对该酶活性有较强的抑制作用,表明该蛋白酶是一种类胰蛋白酶。在离体试验下,该蛋白酶可以降解水稻细胞壁蛋白并释放羟脯氨酸。SDS-PAGE电泳分析显示,至少有4个多肽被降解,分子量范围分别在14.4~20.1、20.1~31ku和31~43ku范围内。说明该胞外蛋白酶具有降解水稻细胞壁的能力,因此,该蛋白酶可能在水稻纹枯病菌的致病过程中起重要作用。  相似文献

A pre- and post-monitoring study was conducted of the potential use of capsicum oleoresin as an in-soil repellent for northern pocket gophers (Thomomys talpoides). Pocket gophers were captured in irrigated alfalfa (Medicago sativa L), affixed with radio transmitters, and monitored daily for location. Six plots (4.87 x 4.87 m) each were randomly assigned to capsicum oleoresin and soybean oil treatments; these were set up based upon the centers of initial core areas of gophers. Mean (+/-SD) volumes of capsicum oleoresin and water and soybean oil and water mixtures (10 + 90 by volume) dispensed onto plots equaled 178.5 (+/-4.7) and 175.7 (+/-14.0) liters, respectively. Movements (m) of the radio-transmitted gophers from plot centers were computed for four daily readings (i.e., 0801-1000, 1101-1300, 1501-1700 and 1801-2000 h). Spectrophotometric analysis of soil samples from capsicum oleoresin plots validated the presence of capsicum on plots and the absence of capsicum on placebo- and off-plot locations. Analysis of variance for movement distances of gophers yielded a Date main effect [F(11, 103) = 2.08, P < or = 0.03] and a Date x Reading (time) interaction [F(32, 299) = 3.21, P < or = 0.01]. Results showed that gophers were located farther from plot centers for the 0801-1000 and 1501-1700 h telemetry readings for < or =3 days post-chemical application-a probable 'disturbance' effect rather than a chemical-induced avoidance. In a prior laboratory study, capsicum oleoresin and soil treatments of 1.5% w/w capsicum caused nearly a 50% decrease in soil contact time by gophers relative to placebo-exposed control animals. This implies that procedural variables warrant revision before abandoning this approach. The potential for soil insertion of repellents as a technique for expelling pocket gophers from territories and some methodological changes for future research of the technique are discussed.  相似文献

水稻纹枯病生防内生菌糖蜜草固氮螺菌的分离与鉴定   总被引:1,自引:0,他引:1  
 本文首先对砂仁内生细菌进行分离,以水稻纹枯病菌(Rhizoctonia solani)为靶标菌对获得菌株进行离体拮抗活性、盆栽及田间试验测定。结果表明:获得的27株内生细菌中有4株具有较好的离体抑菌活性,其中SRJ2-4抑菌效果最好,抑菌带达到18 mm;SRJ2-4的盆栽防效及田间防效分别为80.7%与79.4%,与其它菌株相比达极显著水平。SRJ2-4处理的亩产量为488.79 kg,高于其他药剂处理。对该菌株形态、生理生化及16S rDNA序列进行分析,将该内生菌鉴定为糖蜜草固氮螺菌(Azospirillum melinis)。  相似文献

A filamentous virus isolated from a sugar-beet plant showing systemic mosaic collected in South Kazakhstan was identified as an isolate of beet mosaic virus (BMV-K). BMV-K was transmitted by the green peach aphid Myzus persicae in a non-persistent manner, and by sap inoculation to 11 out of 19 species from seven families tested. The virus could not be transmitted to Nicotiana tabacum, N. debneyi, N. glutinosa and N. clevelandii, cither mechanically or with M. persicae. The thermal inactivation point of BMV-K in sugar-beet sap was 55-60 C, dilution end point 1:1000 and longevity in vitro 2 days at 20 C. A purification procedure produced 1-5-3 mg of purified virus from 100 g of infected Stellaria media plants. Purified virus contained a single protein species of molecular weight 34 700 Da. In ELISA tests, BMV-K reacted positively with BMV-specifc antisera obtained from Japan. Germany and Portugal. By competitive DAS- ELISA, the virus isolate was shown to be closely serologically related to all the three isolates of BMV, and very distantly related to bean yellow mosaic and soy bean mosaic viruses.  相似文献

Seeds of Arabidopsis thaliana are hosts for three mycotoxin-producing Aspergillus species, A. flavus , A. nidulans and A. parasiticus, enabling an Aspergillus - Arabidopsis infection (AAI) assay to be developed. The AAI assay involved inoculation of 10- to 12-mg aliquots of uniformly cultivated, surface-sterilized A. thaliana seeds in microcentrifuge tubes. Use of microcentrifuge tubes facilitated qualitative and quantitative analyses of post-infection characteristics such as sporulation and mycotoxin production. Cultivation of A. thaliana seeds under uniform environmental conditions is necessary to limit genotype-independent seed-lot variability. Using the A. nidulans oxylipin mutant, Δ ppoABC , and two well-characterized A. thaliana pathogen-defence mutants, ein2-1 and pad4-1 , the AAI assay permitted genetic analysis of seed infection and mycotoxin production. Sporulation, but not mycotoxin production, was impaired in A. nidulans ΔppoABC , while A. thaliana ein2-1 and pad4-1 had a small but detectable influence on A. nidulans sporulation that appeared to be dependent on seed age.  相似文献

建立了基于多克隆抗体的氯噻啉间接竞争酶联免疫分析(ic-ELISA)方法。设计合成了氯噻啉半抗原,将其分别与牛血清蛋白(BSA)和卵清蛋白(OVA)偶联制备免疫原和包被原。用免疫原免疫新西兰大白兔,制备多克隆抗体,抗体效价为2.56×106。通过对有机溶剂含量、离子强度和pH等影响因素的优化,确定了氯噻啉ic-ELISA的最佳检测条件(含体积分数10%甲醇的磷酸盐缓冲液、Na+浓度为0.14 mol/L、 pH 7.4),并建立了氯噻啉标准竞争曲线,该方法的抑制中浓度(IC50)为0.18 mg/L,最低检测限(IC10)为0.001 8 mg/L。所得多克隆抗体除与吡虫啉有较大的交叉反应外,与其他结构相似的化合物无明显交叉反应。在水、土壤和甘蓝中分别添加0.05~1 mg/kg的氯噻啉标准溶液,平均回收率为91.42% ~113.82%,相对标准偏差(RSD)为0.72% ~8.68%,符合农药残留检测要求。该ELISA方法可用于环境及农产品中氯噻啉残留检测。  相似文献

The method of analysis employed in an in-vitro study on degradation of organochlorine compounds by meat starter micro-organisms was evaluated using the recommendations made by the Food and Drug Administration (FDA). Recoveries of the organochlorine pesticides α-, β- and γ-hexachlorocyclohexane, hexachlorobenzene, pp′DDE, dieldrin and the polychlorinated biphenyl PCB 153 from a mixture added to four different liquid media, tryptic soy broth, brain heart infusion, a commercial broth and a mineral salts medium, were determined. Recoveries were between 80 and 110% with all components, irrespective of the medium used, and there were no analytical interferences due to the reagent blank or to the matrix. The repeatability was very good with relative standard deviations in the range 2·5 to 11·1%. The concentration of each component of the mixture was determined by capillary GC analysis of extracts using an electron capture detector. © 1997 SCI.  相似文献

The sorption of pesticides to soil particles has implications for their distribution and fate in the soil environment. A batch equilibrium technique was used to investigate sorption of the fungicide AEC623892 to intact and hydrogen-peroxide-treated whole soils and several particle-size fractions isolated from them. Sorption of AEC623892 to the soil as a whole was low. K(f oc) values measured in the whole soils were 169.2 and 41.9 ml g(-1) for Soil A and Soil B respectively. The highest values of K(f oc) were measured in soil particle-size fractions <53 microm (266.5 ml g(-1) in the 2-20 microm fraction of Soil A; 471.9 ml g(-1) in the 20-53 microm fraction of Soil B). Sorption was most irreversible in the 2-20 microm fractions. Overall, treatment of soil particle-size fractions with hydrogen peroxide resulted in lower values of K(f oc) (112.3 ml g(-1) in Soil A whole soil and 30.9 ml g(-1) in Soil B whole soil). In both soils, the maximum sorption among hydrogen-peroxide-treated samples was observed in the <2 microm fraction (166.6 and 311.0 ml g(-1) for Soil A and Soil B, respectively). Investigation of the mineralogical composition of the soils suggested that the clay mineralogy (dominated by kaolinite and illite) is less likely to account for the differences in sorption observed than differences in the characteristics of the soil organic matter. Thermal analysis of the different soil fractions indicated that hydrogen peroxide treatment preferentially removed aliphatic fractions of organic matter, but had less effect on lignin-like, aromatic fractions.  相似文献

通过对农药厂周围土壤和农作物中乙草胺含量的测定,研究了乙草胺农药厂对其周边环境的污染情况。从距离农药厂100 m以内15个点共取得73份样品,采用气相色谱法测定了土壤及农作物样品中乙草胺的残留量。结果显示:农药厂南面土壤及农作物中乙草胺的平均检出量分别为38.2~45.3 μg/kg和39.5~46.0 μg/kg,东面分别为15.4~22.9 μg/kg和21.6~30.1 μg/kg;耕作区土壤污染状况在垂直方向及不同距离上均未表现出明显的差异性;东面及南面土壤与农作物中污染情况分别存在对应关系。农作物样品中乙草胺的平均检出量为21.6~46.0 μg/kg,表明该区域生产的农产品存在一定的安全风险。  相似文献

ABSTRACT A simulator for the enlargement of cohorts of circular lesions on cohorts of host tissue was used to examine five epidemiological parameters: radial rate (mm day(-1)) of lesion expansion, k (exp); maximum basic infection rate, R (m); proportion of lesion area as infectious, f; initial lesion size (mm(2)), z; and proportion of susceptible host sites, s. Based on the proportion of disease severity at day 50 and the proportion of the total disease that originated solely from lesion expansion, k(exp) was the most sensitive of the five parameters. A radial rate of only 0.1 mm day(-1) resulted in a proportion of >0.7 of the diseased area that came from lesion expansion. In an extensive survey of phytopathological literature, many plant pathogens had radial rates greater than 0.1 mm day(-1), which would result in a proportion of >0.95 of the total disease that comes from lesion expansion. Susceptible host sites, s, was a sensitive parameter, as this determined the host area into which lesions could expand. Naturally, R(m) was a sensitive parameter for the proportion of disease on day 50, as it controlled the overall speed of the epidemic. Initial lesion size was a relatively insensitive parameter, although z interacted significantly with s. The greatest proportion of disease that originated from lesion expansion occurred with fast k(exp), small z, and low values of s, R(m), and f. The model was validated with lesion numbers and severities obtained in natural epidemics of Cercospora medicaginis on alfalfa and Exserohilum turcicum on maize. We recommend that the 'epidemic quintuplet' used to describe polycyclic epidemics be expanded to the 'epidemic sextuplet' with the inclusion of k(exp), since lesion expansion is a major component of many polycyclic epidemics.  相似文献

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for cyhalofop-butyl was developed with a polyclonal antibody produced against a hapten (cyhalofop acid) conjugated with bovine serum albumin (BSA). The ELISA of cyhalofop-butyl showed an IC50 value of 0.067 ± 0.004 mg/l and the limit of detection (LOD, IC10) of 0.0029 ± 0.0001 mg/l at the optimal conditions. No significant cross-reaction to other structure-related compounds suggested high specificity for cyhalofop-butyl of the method. The average recoveries of cyhalofop-butyl from fortified water and soil were in the range of 83.2-119.7% and 80.1-104.0%, respectively. These data indicate that this method is a convenient analytical technique for monitoring cyhalofop-butyl in water and soil without purification steps.  相似文献

A trihaloimidazole derivative (S-377; 1-(4-chlorobutoxymethyl)-2,4,5-trichloroimidazole) caused giant excitatory junctional potentials by a single electrical stimulus to the excitatory nerves. Repetitive discharges were simultaneously induced in the nerve fibres. S-377 also caused repetitive firing in the inhibitory nerve fibres. Permethrin showed quite similar actions on both the junction and nerve fibres. It was supposed that S-377 inhibited the inactivating system of the sodium channel, like pyrethroids. However, S-377 reduced the number of spikes of the permethrin-induced repetitive discharges on the excitatory nerve fibres. These observations suggest that binding sites of S-377 on sodium channels are different from those of pyrethroids.  相似文献

The behaviour of imazapyr (2-(4-isopropyl-4-methyl-5-oxo-2-imidazolin-2-yl)nicotinic acid), a broad spectrum herbicide of the imidazolinone family, has been studied under UV radiation in the presence of metal salts. Complexation interactions between imazapyr and metal ions decreased imazapyr photolysis. A chemometric study compared the photodegradation of imazapyr in aqueous solutions in the presence of Na+, Ca2+ or Cu2+ and their concomitant anionic species (Cl-, NO3-) at various pesticide/metal ion molar ratios. The study showed the major role of metal ions in the degradation of imazapyr and its main photoproducts. The molecules were strongly stabilised on complexation with metal ions, leading to an increase in persistence of the pesticide.  相似文献

对山东省侵染马铃薯的一个马铃薯X病毒(PVX)分离物PVX—SD1的外壳蛋白(CP)基因进行了克隆和序列分析。以提纯的病毒RNA为模板,应用RT-PCR扩增目的基因,通过常规的基因克隆法将扩增的CP基因导入pUC19载体,测序。结果表明,PVX—SD1的CP基因长719bp,可编码248个氨基酸;与Gen—Bank中报道的15个有代表性的株系或分离物相比较,核苷酸同源性在80.1%-99.7%,氨基酸同源性在89.8%-100%;与欧洲株系UK3仅1个核苷酸不同,同源性为99.7%,氨基酸同源性达100%,表明它们可能为同一株系,属于X^3组.  相似文献

An improved simulation model (PCPF-1) has been evaluated for the prediction of the fate of mefenacet in an experimental paddy field. This model simulates the fate and transport of pesticide in paddy water and the top 1 cm of paddy soil. Observed concentrations of mefenacet in the paddy water and the surface soil exponentially decreased from their maximum concentrations of 0.70 mg litre(-1) and 11.3 mg kg(-1), respectively. Predicted mefenacet concentrations both in the water and surface soil were in excellent agreement with those measured during the first 2 weeks after herbicide application, but concentrations in paddy water were appreciably overestimated thereafter. The model simulated mefenacet losses through runoff, percolation and degradation to be respectively 41.9%, 6.4% and 57.3% of applied, and the mass balance error was about -6%. The model simulation implied that drainage and seepage control, especially shortly after application when herbicide concentrations are high, is essential for preventing pesticide losses from paddy fields. In focusing on pesticide concentrations in this early period the PCPF-1 model can be a beneficial tool for risk assessment of pesticide losses and in the evaluation of agricultural management for reducing pesticide pollution associated with paddy rice production.  相似文献
