An infestation of mangrove forests with Neodolichodorus persiangulfus n. sp. in southern Iran is reported. This paper is the first to report a representative species of the genus in the country. The new species was characterized by 2,120 (1795–2400) µm long females, and 1755 (1533–2019) µm long males, a continuous to slightly offset cephalic region separated from the rest of the body by a shallow depression, having 5–7 annuli and a distinct labial plate, a lateral field with four lines, the outer ones crenate, irregularly areolated in anterior and posterior body region, 79 (66–85) μm long female stylet, a didelphic‐amphidelphic female reproductive system with asymmetric sclerotized vaginae, phasmids 5–10 annuli behind the anus, female tail elongate conoid, suddenly narrowing at 5–12 annuli posterior to anus and males with trilobed bursa, 52.5 (47–58) μm long spicules and 21.5 (18–27) μm gubernaculum. With the elongate‐conoid female tail and c? >1.2, the new species is close to five known species of the genus: N. brevistylus, N. leiocephalus, N. paralongicaudatus, N. rageshi and N. sinensis, but is distinguished from them by the morphology of the female tail, areolation type of the lateral field, position of hemizonid and phasmid and morphometric data. Molecular phylogenetic analyses using sequences of the near full‐length fragment of the small and the D2‐D3 expansion segments of the large subunit ribosomal DNA (SSU and LSU rDNA D2‐D3) revealed the new species placed in a clade including one isolate of Neodolichodorus sp. and some species of Globodera, Heterodera and Rotylenchulus with 0.89 Bayesian posterior probability (BPP) in SSU phylogeny. The only available SSU sequence of the genus was the tentative closest relative to the new species in the inferred SSU tree. In the reconstructed LSU phylogenetic tree, N. persiangulfus n. sp. formed a poorly supported clade with Rotylenchulus spp. in Bayesian inference (BI), and the only other available D2‐D3 sequence of the genus occupied a distant placement to the new species.  相似文献

Ektaphelenchoides maafiae n. sp. was isolated during a survey of nematodes associated with bark samples of an oak tree (Quercus castaneifolia) in Gorgan, Golestan Province, northern Iran. The new species has a body length of 480–546 μm (in females) and 431–480 μm (in males). The cuticle is weakly annulated, with three lateral lines. Lip region offset. The stylet is 13–15 μm long without basal swellings. The excretory pore is located at the level of the metacorpus base to slightly posterior, and hemizonid is at 15–17 μm posterior to the excretory pore. The post‐uterine sac is short, 6–8 μm long. Spicules having rounded condylus, rostrum short, conical with bluntly pointed tip, a cucullus (apophysis) presented on the dorsal distal end. Male tail bearing four (2 + 2) caudal papillae, conical, with sharply pointed terminus. The new species is close to four known species of the genus, including E. hunti, E. ruehmi, E. caspiensis and E. poinari, but differs from them by body size, shape of tail terminus, stylet length, shape and size of spicules, length of post‐vulval uterine sac and number of caudal papillae. Phylogenetic analysis based on small subunit (SSU) and partial large subunit (LSU) sequences of rDNA confirmed its status as a new species.  相似文献

Further data on the morphology (the lip region characters) and phylogeny of the genus Sakia are presented. The new observations were based on scanning electron microscopy (SEM) and light microscopy (LM). A new species, Sakia sisanganensis n. sp., was recovered from rotten wood of a dead beech tree (Fagus orientalis) in northern Iran, herein described and illustrated based on an integrative approach, that is morphological, morphometric and molecular characters. The new species is characterized by a combination of the following features: fine transverse striae and vestigial single band in the lateral field in SEM. Labial area dorso‐ventrally flattened. Oral region with two concentric hexagonal plates, the inner one apparently elevated. Amphidial openings short, slit‐like. Stylet delicate. Median bulb fusiform to spindle‐shaped with weak valvular apparatus. Spermatheca functional. Tail filiform with faintly pointed tip and males common. The new species was morphologically compared with four known species of the genus, viz., S. alii, S. arboris, S. castori and S. indica, all having indistinct lateral fields. Molecular phylogenetic analyses were performed based on the small subunit ribosomal RNA gene (SSU rDNA). In the Bayesian inference (BI), S. sisanganensis n. sp. with two isolates was strongly supported as a sister taxon of a clade harbouring S. arboris + Lelenchus species. However, in the maximum likelihood (ML) analysis, the new species formed a clade with S. arboris, thus supporting the reciprocal monophyly of the genera Sakia and Lelenchus. Accordingly, the test of monophyly was performed (using Bayes factor) and the results did not reject the monophyly of sakia (i.e., S. sisanganensis n. sp. and S. arboris as sister taxa) based on the currently available data.  相似文献

Bursaphelenchus willibaldi, associated with trees in two separate locations in forests of northern Iran, are characterized and illustrated based on morphological, morphometric and molecular data. The Iranian population of B. willibaldi has a body length of 392–595 μm, stylet length of 12–14 μm, c‐index of 9.0–14.5 and PUS length of 51–82 μm. Males have spicules 15–18 μm long along the arch line. The Iranian population showed morphometric variations compared with the originally described samples. For example, it has a shorter body in females and males, greater c‐index, slightly greater range of V, slightly smaller range of PUS and tail length. Molecular phylogenetic analysis of the recovered populations revealed both sequenced isolates forming a clade with one European isolate of the species using Bayesian inference (BI) analysis with full Bayesian posterior probability (BPP).  相似文献

Abstract

Mercury intrusion porosimetry (MIP) was used to evaluate the impregnation mechanisms of wood by methyl methacrylate (MMA) through examining the changes in porosity, pore volume, pore size distribution and bulk density of solid wood before and after MMA impregnation. Porosities of MMA-impregnated (hardened) wood samples were lower than those of solid wood samples for six studied species, five hardwoods and one softwood. Densities of hardened wood were enhanced from 45 to 130% depending on the species. The pore volume available for mercury intrusion was shifted from pore d>0.1 µm for solid wood to pore d≤0.1 µm for hardened wood. A pore diameter of 0.1 µm was used as the transition point for MMA impregnation and the increased mercury penetration below this point was attributed to the MMA polymer pore structure. Porosity as an intrinsic property of wood appears to be the main determinant of impregnation rate and polymer retention, especially for porosity with pore diameter >0.1 µm. The results indicate that the MIP technique is an effective tool with which to study the impregnation process.  相似文献

Sirococcus is a genus of asexually reproducing fungi that includes important pathogens causing shoot blight and tip dieback of conifers. In this paper, the type species of Sirococcus, S. conigenus, is redescribed and illustrated, and an epitype designated. In addition, two new species are recognized. Sirococcus piceicola sp. nov. is described from species of Picea in Canada and Switzerland. A second new species, S. tsugae sp. nov., is known only from western North America on species of Cedrus and Tsuga. These three species can be distinguished based on morphological differences and molecular sequence data from four genes. The three species of Sirococcus on conifers vary in conidiomatal wall structure, shape of the conidiogenous cells, and shape and size of conidia.  相似文献

Thirty‐two southern Swedish families of Pinus sylvestris L. from a partial 11×11 factorial mating were grown at four night lengths, 3, 5, 7, and 9 h, from the start of their first growth period. At the end of the growth period, the plants were exposed to increasingly longer nights until total growth cessation. During the second and third growth periods, the plants shared the same growing conditions in the growth chambers, starting with two weeks of 6‐h night followed by night prolongation as in the first growth period. The plants from the 9‐h treatment grew more slowly than those from the other treatments in growth periods 2 and 3. The genetic influence of the growing conditions during the first growth period on the height increment during growth periods 2 and 3 was estimated by the female and male by treatment interaction variances. These variance components were relatively small and totally negligible when data from the 9‐h treatment were excluded from the analysis. The data suggest stability in the parents with respect to preconditioning caused by varying photoperiods.  相似文献

For reasons of unequal distribution of more than one nematode species in wood, and limited availability of wood samples required for the PCR‐based method for detecting pinewood nematodes in wood tissue of Pinus massoniana, a rapid staining‐assisted wood sampling method aiding PCR‐based detection of the pine wood nematode Bursaphelenchus xylophilus (Bx) in small wood samples of P. massoniana was developed in this study. This comprised a series of new techniques: sampling, mass estimations of nematodes using staining techniques, and lowest limit Bx nematode mass determination for PCR detection. The procedure was undertaken on three adjoining 5‐mg wood cross‐sections, of 0.5 × 0.5 × 0.015 cm dimension, that were cut from a wood sample of 0.5 × 0.5 × 0.5 cm initially, then the larger wood sample was stained by acid fuchsin, from which two 5‐mg wood cross‐sections (that adjoined the three 5‐mg wood cross‐sections, mentioned above) were cut. Nematode‐staining‐spots (NSSs) in each of the two stained sections were counted under a microscope at 100× magnification. If there were eight or more NSSs present, the adjoining three sections were used for PCR assays. The B. xylophilus– specific amplicon of 403 bp ( DQ855275 ) was generated by PCR assay from 100.00% of 5‐mg wood cross‐sections that contained more than eight Bx NSSs by the PCR assay. The entire sampling procedure took only 10 min indicating that it is suitable for the fast estimation of nematode numbers in the wood of P. massonina as the prelimary sample selections for other more expensive Bx‐detection methods such as PCR assay.  相似文献

A new species of the hyphomycete genus Chalara is described. It has been frequently isolated from stems and branches and sometimes also from roots of wilting and dying Fraxinus excelsior in Poland. It differs from previously described species of Chalara by its small, short cylindrical phialoconidia extruded in chains or in slimy droplets, morphological features of the phialophores and by colony characteristics. Two popular indigenous browse, shade and poles trees, Acacia galpinii and Faidherbia albida, were evaluated for survival and growth on-farm in Malotwana, Botswana, with a mean annual rainfall of 450 mm. The trial was a two species × three spacing factorial experiment in a randomised complete block design replicated five times. The three spacings were 5 m × 5 m, 6.3 m × 5 m and 8.3 m × 5 m. The study was conducted over 6.5 years. The results showed that A. galpinii is a promising poles, browse and shade species, while F. albida exhibited high mortality (67%) at 4.5 years (after below-average rainfall). The crown width of A. galpinii ranged from 5.86 ± 0.24 m at high density to 6.56 ± 0.25 m in low-density plots. The difference in crown width among densities was significant (p = 0.0406). Diameter at breast height ranged between 10.4 ± 0.68 cm at high density to 12.5 ± 0.64 cm in low-density plantings and was significantly different among spacings (p = 0.0003).  相似文献

This paper reports three newly recorded species, Diphascon modestum Binda, Pilato and Dastych, 1984, Diphascon nobilei Binda, 1969, and Diphascon triodon Maucci, 1996, of the genus Diphascon (Tardigrada; Hypsibiidae) from China. Five specimens of Diphascon modestum were collected from Taibai Mt. (34°18′N, 107°42tpE) at 2,500 m above the sea level (a.s.l.), two specimens of Diphascon nobilei were collected from Tonggu County, Jiangxi Province (28°31’N, 114°26′E) at 900 m a.s.l, and one specimen of Diphascon triodon was collected from Linzhi County, Tibet (29°40′N, 94°23′E) at 3,500 m a.s.l. All specimens are deposited at the College of Life Sciences, Shaanxi Normal University, China. A key to the Chinese species of Diphascon was also given. Cherry leaf roll virus (CLRV) infection is common in Betula pendula and B. pubescens in Middle and North Europe, easily observable by chlorotic leaf vein banding, mottling and leaf roll, partially adherent with progressive loss of vitality or death of twigs and branches. In Fennoscandia, a severe viral epidemic in various birch species in forest stands, public greens and roadsides is associated with CLRV. In Corsica, CLRV‐typical symptoms were observed on birch trees (Betula sp.) in a montane stand (1470 m) at Col de Vergio. CLRV was detected by RT‐nested PCR in all leaf samples from 11 randomly selected birch trees exhibiting characteristic symptoms. Along with the fact that this is the first report of CLRV in Betula sp. of both montane and Mediterranean origins, remarkably high genetic variation and a new distinct phylogenetic cluster are comprised by a small randomly sampled CLRV population that has evolved in one of the few scattered birch stands in Corsica.  相似文献

Bursaphelenchus mucronatus is closely related to the pinewood nematode Bursaphelenchus xylophilus, the causative agent of pine wilt disease. B. xylophilus became a devastating pest when it was introduced in the Far East; however, B. mucronatus is considered to have low virulence. Morphological similarities between Bxylophilus and Bmucronatus make the accurate morphological identification of both species difficult. Thus, it has become important to pay attention towards B. mucronatus impact and the need of discrimination of these two species. To distinguish among the two species, a B. mucronatus‐specific sequence‐characterized amplified region (SCAR) marker has been developed. The specific Random amplified polymorphic DNA (RAPD) fragment of B. mucronatus, OPY01‐M850 was excised from agarose gels and purified. The gel‐purified fragment was cloned into the pGEM®‐T Vector and subjected to sequencing. Based on the sequenced RAPD fragments, a number of SCAR primers were designed. It is demonstrated that OPY01‐M850 through primers Y01F/R can be transformed into a B. mucronatus‐specific SCAR‐Y01‐M609 marker. Primers set Y01F/R had high specificity that could be used for the discriminative identification of B. mucronatus versus B. xylophilus.  相似文献

The objective of the study was to develop an in vitro shoot regeneration protocol by utilising shoot tips explant from Vitex trifolia L. Shoot tip explants obtained from a 3-year old plant was cultured on Murashige and Skoog (MS) medium supplemented with various concentrations (1.0, 2.5, 5.0, 7.5 or 10.0 µM) of thidiazuron (TDZ). The optimal level of TDZ supplementation to the culture medium was 5.0 μM for 15 d induction period. The highest number of shoots (22.2 ± 0.1) and shoot length (5.1 ± 0.1 cm) were achieved when TDZ-exposed explants were sub-cultured on MS medium containing 6-benzyladenine (1.0 µM) and 0.5 µM α-naphthalene acetic acid (NAA) after 8 weeks of culture. In vitro rooting of isolated shoots was achieved best in half-strength MS medium containing 0.5 µM NAA. During the acclimatization period, changes in activities of antioxidant enzymes were observed. Superoxide dismutase activity increased reaching maximum at 28th day after transplantation. Likewise, an upregulation of the catalase, ascorbate peroxidase and glutathione reductase enzyme activities were also observed. These observed changes reflected the ability of plants in developing an antioxidant enzymatic defence system aiding in survival against oxidative stress and in reducing release of free radicals. Plantlets were successfully hardened off and acclimatized in earthen pots containing garden soil with a survival rate of 90 %.  相似文献

The effects of thinning on plant species diversity and composition of understory herbs in a larch plantation were investigated. The relationships between plant species diversity and composition of understory herbs and light conditions were established. Twenty-five 1 m × 1 m plots and fifteen 13 m × 1 m transects were set up in unthinned and thinned stands, respectively. All the transects in the thinned stands were set across the thinned rows and unthinned rows, and each of them was divided into nine 1 m × 1 m sub-plots. The herb diversity and light conditions were observed in each plot and sub-plot. The results show that there was a significant difference in herb diversity between the thinned and unthinned stands. All biodiversity indices except for evenness index in the thinned stand were higher than those of the unthinned stand, i.e., the herb diversity increased after thinning. According to the changes in herb densities and whether one species could be found in a stand or not before and after thinning, all herb species were classed into three types: positive, neutral and negative species, which referred to a species newly appeared and having an obviously increased density after thinning, with no obvious changes in its appearance and density after thinning, and disappeared and having an obviously decreased density after thinning, respectively. Many new species were found in the thinned stand like Corydalis pallida, Prenanthes tatarinowii, Vicia unijuga and Sonchus brachyotus etc. However, most species found in both the thinned and unthinned stands were negative species. In all nine subplots, only 11 and 10 species were found in spring and in autumn respectively, accounting for 17.74% and 15.15% of all the species in the thinned stand, respectively. All biodiversity indices were the highest in the center sub-plots and most of them tended to reduce from middle to side sub-plots. Diplodia tip blight is a serious disease of >30 conifer species worldwide. Symptoms are particularly severe on non‐native, two‐needled Pinus species, and typically include stunted, necrotic needles and shoots and a general decline of the tree. Latent Diplodia pinea infections occur in current‐year shoots of some symptomless pines, and in some apparently healthy current‐year shoots of diseased pines. Latent infections also occur in symptomless terminal buds. A histological approach was used to investigate the nature of latent infections in shoot stems and terminal buds of landscape Pinus nigra. Fungal colonization was compared in healthy, diseased and latently infected tissues. A nested‐PCR technique that is specific for D. pinea was used to differentiate latently infected tissues from uninfected ones. Latent D. pinea infections were localized in the outer stem cortex, usually in the vicinity of needle scales at leaf axes. In contrast, pathogenic D. pinea infections were characterized by fungal colonization throughout the shoot stem tissues, even very early in symptom development. The presence of necrophylatic periderms in two of the latently infected samples suggests that host defences play a role in the production and maintenance of latent stem infections. Latent infections of terminal buds appeared to originate from the distal bud scales of axillary buds in the terminal bud cluster, and not from the subtending shoot. Fungal tissues were never observed inside asymptomatic, PCR‐negative shoots.  相似文献
