We investigated the effect of polyphenols derived from cacao liquor on the mutagenic action of heterocyclic amines (HCAs) in vitro and ex vivo. In the Ames test, the cacao liquor polyphenols showed antimutagenic effects in bacteria treated with HCA in the presence of an S-9 mixture; however, they showed less efficacy than quercetin. On the other hand, the cacao liquor polyphenols showed potent antimutagenic activity in bacteria treated with activated forms of HCA, compared with quercetin. We also evaluated the effect of these compounds on enzymatic activation of HCA. They weakly suppressed the production of activated HCA. In the host-mediated assay in mice, a method used to estimate the potential carcinogenicity of chemicals ex vivo, oral administration of the cacao liquor polyphenols, reduced the number of colonies of revertant bacteria recovered from the liver. These data suggest that the cacao liquor polyphenols have an antimutagenic effect not only in vitro, but also ex vivo.  相似文献

Abstract

A nutritional survey was conducted by collecting coffee and cacao leaf samples from various ecological zones in Sierra Leone and analysing for various elementst Data obtained were compared with established leaf analysis standards used for assessing nutrient status of coffee and cacao leaves. Varying degrees of deficiencies of N, P, K, Mg, Mn and Cu were observed but excess levels of Fe were also recorded. The soil quality index (SQI) for cacao cropping systems was developed to meet the nutritional criteria of the crop, the environmental safety of the cropping sites and the increasing demand for the production and quality of cocoa. Available water function (AWF), root growth function (RGF), mineral nutrition of plants function (MNF) and environmental safety function (ESF) for potentially toxic elements were included in the additive model of SQI for cacao cropping systems. In this study, over 66% of the cacao fields cropping sites were classified as regular SQI with a range of scores between 0.42 and 0.61. The field cropping site with the highest score (0.73) characterized as typic Alitic Red-Yellow Argisol was rated with high SQI. In contrast, the field cropping sites characterized as abrupt Cohesive Dystrophic Red-Yellow Argisol (0.29), typic Dystrophic Haplic Cambisol (0.39), and latosolic Dystrophic Yellow Argisol (0.40) were rated with low SQI. MNF and AWF were the functions that most contributed to the increase or decrease of SQI scores. The SQI adapted expressed the agro-environmental reality of cacao cropping systems of Bahia, Brazil, and this could be a versatile tool to subsidize the adequate soil management of cacao plantations around the world.  相似文献

A new chromatographic approach for separating cacao procyanidins according to their degree of polymerization has been developed. It utilizes diol stationary phase columns operating in normal phase mode with a binary gradient of acidified acetonitrile and methanol-water. Performance of the diol stationary phase was evaluated on an analytical scale utilizing classical chromatographic conditions for the normal phase separation of procyanidins according to their degree of polymerization. The new separation approach was developed on an analytical scale but further extended to the preparative scale. These newly developed analytical and preparative high-performance liquid chromatography procedures were successfully applied to the separation, as well as isolation, of cacao procyanidins from unfermented cacao seeds. The degree of polymerization associated with each molecular weight fraction was determined by mass spectrometry.  相似文献

Large-scale intensification of smallholder cacao management is currently affecting the agroforestry landscapes of Sulawesi (Indonesia), the world’s third largest cacao producer. Little is known about how this shift from diverse plantations to full-sun cacao will affect functionally important biodiversity within the agroecosystem, and how this is related to landscape-wide patterns in land-use and natural ecosystems. We recorded birds in 43 cacao plots differing in woody and herbaceous vegetation as well as distance to forest in two valleys around the Lore Lindu National Park in Central Sulawesi. Species richness of frugivores and nectarivores decreased with increasing distance to forest, whereas granivorous birds increased in richness. Forest specialists, but not habitat generalists, responded positively to forest edge proximity. Species richness of all functional groups except seed eaters increased at higher density of tall shade trees. Greater species richness of shade trees was associated with higher species richness of frugivores and nectarivores, while herbaceous vegetation did not have a strong impact on the avifauna. The positive effect of shade trees was independent of distance to forest. In conclusion, our study shows the relative importance of local and landscape effects on bird diversity with shade trees being critical for bird conservation in cacao agroforestry landscapes.  相似文献

Cacao (Theobroma cacao L.) is the main source for chocolate with an annual production of four million tons worldwide. This Neotropical tree crop was domesticated in Mesoamerica as far back as 3,000 years ago. Knowledge of genetic diversity and population structure in farmer varieties of cacao in the center of domestication is essential for sustainable production of fine-flavored cacao beans and contributes to in situ/on-farm conservation of farmer varieties. Based on 70 single nucleotide polymorphism markers, we analyzed 84 fine-flavored farmer varieties collected from traditional cacao farms in Honduras and Nicaragua. The study also included 31 clones from the international cacao collections to serve as references. The SNP based multilocus matching identified six synonymous groups, including 14 Criollo and two Amelonado varieties. A moderately high level of genetic diversity was observed in these farmer varieties, indicating the possibility to further explore intra-population variation and breed for fine-flavored cocoa. Multivariate analysis showed clustering of the 84 farmer accessions in five genetic groups: ancient Criollo, Amelonado, Trinitario (including Nicaragua Trinitario and Honduras Trinitario) and Upper Amazon Forastero (only one accession). The Honduras Trinitario differed from the Nicaragua Trinitario group. The clustering results largely supported the perceived classification of cacao by local farmers and researchers, which was mainly based on morphological traits. However, the well known traditional variety “Indio” in this region was identified as synonymous with Amelonado. Parentage analysis showed that the variety “Indio” (or Amelonado) contributed more to the Trinitario type farmer varieties, whereas ancient Criollo had less influence. The present study demonstrates the efficacy of using a small set of SNP makers for cacao germplasm characterization, and further depicts the diverse origins and parentage in farmer varieties from Mesoamerica. This information thus will be highly useful for conservation and utilization of cacao germplasm from this region.  相似文献

Forest disturbance and transformations into agricultural land alter tropical landscapes at drastic rates. Here, we investigate bryophyte assemblages on trunk bases in natural forest, selectively logged forest and cacao agroforests that are shaded by remnants of natural forest in Central Sulawesi. Overall, bryophyte richness per site did not differ between forest types. However, mosses and liverworts reacted differently in that moss richness was lowest in cacao agroforests, whereas liverwort communities were equally rich in all forest types. In terms of cover, mosses remained unaffected while liverwort cover decreased significantly in disturbed forest. Species composition of bryophytes clearly changed in cacao agroforests as compared to natural forests and selectively logged forests. In particular some drought-sensitive species were rare or absent in cacao agroforests and were replaced by drought-tolerant ones, thus underlining the importance of microclimatic changes. Moreover, differences in bryophyte species composition between large and small trees were only pronounced in cacao agroforests, presumably due to concomitant changes in stemflow of precipitation water. In conclusion, the bryophyte assemblages of selectively logged forests and cacao agroforests were as rich as in natural forest, but species turn-over was particularly high towards cacao agroforests probably due to microclimatic changes. Maintenance of shade cover is crucial to the conservation of the drought-sensitive forest species.  相似文献

Ammonium and nitrate are the major forms of nitrogen (N) present in tropical soils. An experiment was conducted to assess the influence of nitrate and ammonium forms (NO3?, NH4+, and mix of NO3? + NH4+), and levels (1.5–12.0 mM) of N on the growth and nutrition of cacao (Theobroma cacao L). Growth parameters were significantly influenced by N forms, and nitrogen supplied as NH4+ proved better for the growth of cacao compared with NO3? form and mixtures of these two forms. Irrespective of the forms of N, levels of N had no significant effect on plant growth parameters. Nutrient efficiency ratios (NERs) (shoot dry matter produced per unit of nutrient uptake) for macronutrients were sulfur>phosphorus>calcium>magnesium>nitrogen>potassium (S>P>Ca>Mg>N>K) and for micronutrients NERs were in the order of copper>boron>zinc>iron>manganese (Cu>B>Zn>Fe>Mn).  相似文献

In this study, complexation extractants ammonium bicarbonate diethylene triamine pentaacetic acid (AB-DTPA), diethylene triamine pentaacetic acid (DTPA), and ethylene diamine tetraacetic acid (EDTA) and mild cation-exchange extractants calcium chloride (CaCl2) and ammonium nitrate (NH4NO3) were used to evaluate the bioavailability of soil cadmium (Cd) to cacao in the field. Among the five extractants, the extractable Cd generally followed the order EDTA > DTPA > AB-DTPA > CaCl2 > NH4NO3. Correlation analysis was done between the extractable Cd in soil and total Cd content of cacao tissues (nibs, shells, leaves, and pods). The Cd extracted by CaCl2 and NH4NO3 was significantly (P < 0.05) correlated with some of the tissues but their Pearson correlation coefficients were weak. In contrast, extractants AB-DTPA, DTPA, and EDTA showed stronger, significant correlations to the Cd concentration in all four tissues. Overall, regression analysis demonstrated that AB-DTPA, DTPA, or EDTA can be used to predict bioavailable Cd in soils for cacao. Of these, AB-DTPA and DTPA both showed the strongest correlations compared to EDTA. However, the ease of preparation and the superior shelf-life of DTPA over AB-DPTA make it the preferred reagent for Cd bioavailability extractions from cacao soils and is currently being used to develop cost-effective soil treatments to reduce bioavailable Cd to cacao plants.  相似文献

The present work analyzes the lipid fraction from seeds of wild Ecuadorian Theobroma subincanum and selected commercial varieties of Theobroma cacao from Mexico (var. Criollo) and Ecuador (var. Arriba). The lipid fraction was obtained from the seeds through supercritical fluid extraction and analysis performed by preparatory thin-layer chromatography followed by gas chromatography. The results revealed that in T. subincanum the triglycerides contain fatty acids with longer chains. The melting point and peroxide and saponifiable numbers were determined for each Theobroma sample. The results lead to the conclusion that T. subincanum would produce a poorer quality butter than T. cacao. Nevertheless, the results do point toward a significant commercial use of T. subincanum for low-profile products.  相似文献

The ex situ conservation of cacao genetic resources is in the form of field genebanks. Misidentified trees though, represent a serious problem to (i) curators, who need to capture genetic diversity, and (ii) users of germplasm material, who must be certain of the identities of the accessions for proper evaluation of their results. The presence of mislabelled trees was assessed from data compiled in the International Cocoa Germplasm Database (ICGD) 2000 v4.1CD-ROM, which contains the published records of cacao accessions in global holdings. Circumstantial evidence revealed that many germplasm collections, especially those in Brazil (CEPEC), Colombia (ICA), Costa Rica (CATIE), Côte d'Ivoire, Mexico, Malaysia, Togo, Trinidad (ICG,T) and Venezuela (EEC) probably possessed misidentified material. The severity of the problem ranged from 15–44 % and is suggested as an area for immediate resolution. Based upon the morphological data provided by the ICGD CD-ROM, several accessions were highly suspect, including 'EET 19 [ECU]', 'EET 48 [ECU]', 'EET 162 [ECU]', 'ICS 1', 'ICS 6', 'ICS 43', 'ICS 89', 'ICS 100', 'IMC 67', 'PA 121 [PER]', 'POUND 7 [POU]', 'SCA 6', 'SPA 9 [COL]', 'UF 29', 'UF 613' and 'UF 667'. However, the identification of mislabelled trees should depend ultimately on molecular analysis. Hence, the use of voucher specimens for verification is forwarded and the concept of a public domain database for microsatellite analyses of germplasm holdings is raised.  相似文献

The changes of mono- and oligosaccharides, carboxylic acids, purine alkaloids, and fatty acid composition, and of aspartic endoproteinase activity, were analyzed during seed development in two varieties of cacao (Theobroma cacao). The majority of the components examined either decreased or accumulated steadily in concentration during the second half of bean development. Sucrose is the major sugar in the mature embryo, whereas fructose and glucose are at higher concentrations in the endosperm tissue. Considerable amounts of malate are found in the endosperm, whereas citrate is the dominant carboxylic acid in the embryo. A major change in the fatty acid composition occurs in the young embryo when the proportion of stearic acid increases rapidly at the expense of linoleic acid, which is reduced from about 18 to 3%. Theobromine is the dominant purine alkaloid (ca. 80%), and caffeine appears only toward the end of seed maturity. Aspartic endoproteinase activity increases rapidly during embryo expansion, reaching a maximal activity before final maturity. The results are discussed in conjunction with physiological changes in developing seeds, and the potential contributions of the compounds analyzed for cocoa quality.  相似文献

Cacao (Theobroma cacao L.) is a significant agricultural commodity in the Dominican Republic, which ranks 11th in the world for cacao exports. To estimate genetic diversity, determine genetic identity, and identify any labeling errors, 14 SSR markers were employed to fingerprint 955 trees among cacao germplasm accessions and local farmer selections (LFS). Comparisons of homonymous plants across plots revealed a significant misidentification rate estimated to be 40.9 % for germplasm accessions and 17.4 % for LFS. The 14 SSRs amplified a total of 117 alleles with a mean allelic richness of 8.36 alleles per locus and average polymorphism information content (PIC) value of 0.67 for the germplasm collection. Similar levels of variation were detected among the LFS where a total of 113 alleles were amplified with a mean of 8.07 alleles per locus and PIC of 0.57. The observed heterozygosity (Hobs) was 0.67 for the germplasm collection and 0.60 for LFS. Based on population structure analysis 43.9 % of the germplasm accessions and 72.1 % of the LFS are predominantly of the Amelonado ancestry. Among these Amelonado, 51.7 % for the germplasm collection and 50.6 % for LFS corresponded to Trinitario hybrid lineage. Criollo ancestry was found in 7.6 and 9.5 % of the germplasm accessions and LFS, respectively. The Contamana, Nacional, and Iquitos backgrounds were also observed in both populations, but the Curaray background was only detected in the germplasm accessions. No Purús or Guiana ancestry was found in either of the populations. Overall, significant genetic diversity, which could be exploited in the Dominican Republic breeding and selection programs, was identified among the germplasm accessions and LFS.  相似文献

In commercial terms, Criollo cacao trees (Theobroma cacao L.) are reputed to be the source of the commercial product (fermented and dried cocoa beans), which sells for the best price on the market. Nevertheless, the term “Criollo” has numerous meanings and interpretations depending on if it is used by commercial users or botanists, growers or breeders. Our review aims to specify which cocoas can justifiably carry the Criollo name. “Criollo” is a botanical subspecies of Theobroma cacao, i.e. Theobroma cacao subsp. cacao; however, the true Criollos form just one of the ten currently accepted genetic groups in the species. We thus provide an overview of genetic studies on the subject (published or not), along with what is currently known about “True Criollo” or “Ancient Criollo” cacao trees. In fact, there are few representatives in collections that are duly acknowledged to be true Criollos, particularly in the two International Cocoa Genebanks, where only seven clones are available. It is nonetheless certain that some true Criollos do exist in other collections but have not been formally identified (by genetic studies) as members of the Criollo genetic group. Likewise, some true Criollos, be they cultivated or subspontaneous, exist in Mexico and Central and South America (Venezuela and Colombia). However, certain clones called “modern Criollos”, which are closely related to the true Criollos but arise from hybridization with other genetic groups, are more common.  相似文献

The most abundant albumin present in seeds of Theobroma cacao was purified to apparent homogeneity as judged by high-performance liquid chromatography/electrospray ionization mass spectrometry (HPLC/ESI-MS), sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and NH(2)-terminal sequence analysis. Tryptic peptide mass fingerprinting of the purified protein by HPLC/ESI-MS showed the presence of 16 masses that matched the expected tryptic peptides corresponding to 95% of the translated amino acid sequence from the cDNA of the 21 kDa cocoa albumin. Collision-induced dissociation MS/MS analysis of the C-terminal peptide isolated from the CNBr cleavage products provided unequivocal evidence that the mature cocoa albumin protein is nine amino acid residues shorter than expected from the reported cDNA of its corresponding gene. The experimentally determined M(r) value of 20234 was in excellent agreement with the truncated version of the amino acid sequence. The purified cocoa albumin inhibited the catalytic activities of bovine trypsin and chymotrypsin. The inhibition was stoichiometric with 1 mol of trypsin or chymotrypsin being inhibited by 1 mol of inhibitor with apparent dissociation constants (K(i)) of 9.5 x 10(-8) and 2. 3 x 10(-6) M, respectively, for inhibitor binding at pH 8.5 and 37 degrees C. No inhibition of the catalytic activities of subtilisin, papain, pepsin, and cocoa endoproteases was detected under their optimal reaction conditions.  相似文献

In cacao crop the understanding of the multiple relationships between bean and pod husk dry biomass and mineral nutrition in the soil-plant system is scarce. The main objective of this work is to verify the linear, bivariate and multivariate correlations between the mineral nutrient concentrations in the soil, leaves and beans of 53 cacao trees with known dry biomass of cacao beans and pod husk, and further graphically identifying the consistency of these information. For this study, soil samples, leaves and fruits (pod husk and beans) were collected from each cacao trees, for the analysis of nitrogen-N, phosphorous-P, potassium-K, calcium-Ca, magnesium-Mg, iron-Fe, manganese-Mn, zinc-Zn and cooper-Cu mineral nutrient concentrations. The interpretation of the results by the exploratory analysis technique linked to the linear correlation analysis proved to be an essential tool to support studies related to mineral nutrition in the soil-plant system. Foliar N concentration were directly related to the high and very high dry biomass classes of cacao beans. Levels of exchangeable Ca and Mg in the soil were inversely correlated with the dry biomass of cacao pod husk (DBCH), in contrast to results with the positive correlations of these nutrients in the leaves and cacao beans data sets. The foliar concentrations of Zn, Mn, Fe and Cu are shown to be more positively associated with DBCH than dry biomass classes of cacao beans. The concentrations of Fe, Mn and Cu in dry cacao beans show such similar pattern of association with dry biomass of pod husk and beans.  相似文献

Productivity and sustainability of cacao (Theobroma cacao L.) in tropical soils are affected by levels of iron. Information is lacking on the cacao response to various sources of iron (Fe). A greenhouse experiment was conducted to evaluate the effects of five iron sources iron sulfate heptahydrate, ferric ethylenediamine-N,N’-bis(2-hydroxyphenylacetic acid), ferric diethylenetriaminepentaacetic acid, ferric ethylenediaminetetraacetic acid, fiesta herbicide (FeSO4 · 7H2O, FeEDDHA, FeDTPA, FeEDTA,) at 10 mg Fe kg?1 soil on growth, photosynthesis, content of photosynthetic pigments and starch and macro- and micronutrient nutrition of cacao. The various iron sources had significant effects on shoot and root dry biomass accumulation, leaf chlorophyll a and b content, carotenoid levels, SPAD index and PN. These parameters were significantly correlated with concentration, uptake, influx, and transport and use efficiency of Fe. In cacao net photosynthesis, stomatal conductance, internal carbon dioxide (CO2), and transpiration in leaf level responded differently to the sources of Fe. Invariably, macro and micronutrient uptake, influx, transport, and use efficiency showed differential responses to sources of iron but significant effects were only observed for copper (Cu), Fe, manganese (Mn), and zinc (Zn). Overall, FeDTPA, FeEDTA and FeHEDTA could be the best sources of Fe in improving, growth, photosynthesis and macro and micro nutrition of cacao.  相似文献
