Armillaria Species Infesting Vineyards in Northwestern Spain   总被引:1,自引:0,他引:1  
Twelve vineyards in northwestern Spain were studied to assess the incidence of white root rot during 1995 and 1997. In both years, diseased plant material was collected and the Armillaria species responsible was identified on the basis of compatibility testing. Subsequently, restriction fragment length polymorphism (RFLP) analysis of the intergenic spacer region of the nuclear ribosomal RNA gene cluster was used to identify Armillaria species in another 45 symptomatic samples submitted for diagnosis from this area. During 1995, 9 of the 12 vineyards showed white root rot, although in eight cases less than 2% of plants were affected; in the remaining vineyard 17% of plants were diseased. During 1997, 10 of the 12 vineyards were affected and three showed a marked increase in the number of plants with white root rot (to 43%, in the vineyard in which 17% were affected in 1995). The compatibility method detected Armillaria mellea in samples from 10 of the 12 vineyards, and Armillaria gallica in samples from two vineyards. RFLP–PCR analysis detected four restriction patterns corresponding to A. mellea, A. gallica and Armillaria cepistipes: patterns mel 1 (A. mellea, 71% of samples), mel 2 (A. mellea, 18%), gal 1 (A. gallica, 9%), and cep 1 (A. cepistipes, 2%, i.e. a single sample). This is the first report of A. gallica and A. cepistipes infecting Vitis spp. The presence of these two Armillaria species may be related to the fact that the vineyards from which they were isolated were located on cleared forestry sites.  相似文献

In 1982, an anamorphic fungus in the genus Monilia was first isolated as the causal agent of brown rot disease of Japanese apricot or mume (Prunus mume) in Oita Prefecture, Kyushu, Japan. Inoculation of flowers, shoots, and fruit of P. mume with the fungus reproduced brown rot disease symptoms similar to those found in nature. The fungus somewhat resembled the colony appearance of Monilinia (anamorph Monilia) laxa, the apricot brown rot fungus, on PSA plates, but it differed from the latter and the other two brown rot fungi, M. fructigena and M. fructicola, in terms of growth rate, temperature optima for mycelial growth and sporulation, morphology and germination pattern of conidia, nuclear number in the conidium, and nucleotide sequences in the ITS region of ribosomal DNA. It is newly described as Monilia mumecola Y. Harada, Y. Sasaki & T. Sano. A key to anamorphic states of four brown rot fungi of fruit trees is provided.  相似文献

Results of a greenhouse Armillaria ostoyae inoculation experiment, designed for screening resistant Pinus pinaster genotypes and for exploring the role of different phenotypic traits in seedling susceptibility, are reported. The experiment included 39 open-pollinated pine families that comprised a random subset of the breeding population of P. pinaster in Galicia (NW Spain). We employed a non-parametric survival-time analysis to analyze patterns of survival times during 14 months after inoculation with a local A. ostoyae strain. Results indicate (i) a significant correlation between seed weight and tree susceptibility, with seedlings originating from large seeds being more susceptible, (ii) a positive family mean correlation between secondary root weight and size and median life expectancy, and (iii) genetic variation of tree tolerance to A. ostoyae, with some families surviving significantly longer than others. Less susceptible families could be used in breeding programmes or directly in forest plantations to reduce the losses caused by A. ostoyae. Large within-family variation in tolerance to the disease was also observed, suggesting that non additive genetic variance was also important. Although being infected, 32 out of the 1200 inoculated trees survived the fungus infection. These tolerant genotypes comprise an attractive collection to further investigate genetic, phenotypic and environmental factors affecting pine susceptibility to Armillaria root rot.  相似文献

Epidemiology of Fusarium Diseases and their Mycotoxins in Maize Ears   总被引:1,自引:0,他引:1  
Fusarium species cause two distinct diseases on ears of maize, Fusarium ear rot (or pink ear rot) and Gibberella ear rot (or red ear rot), both of which can result in mycotoxin contamination of maize grain. The primary causal agent for Fusarium ear rot is Fusarium verticillioides, but F. subglutinans and F. proliferatum are also important. Gibberella ear rot is caused primarily by F. graminearum, but F. culmorum can also be important, especially in Europe. Aspects of the epidemiology of both diseases have been studied for decades, but only recently have efforts been made to synthesize this information into comprehensive models of disease development. Much of the work on F. graminearum has focused on Fusarium head blight of small-grain crops, but some of the results obtained are also relevant to maize. The primary mycotoxins produced by these fungi, fumonisins and deoxynivalenol, have differing roles in the disease-cycle, and these roles are not completely understood, especially in the case of fumonisins. Progress is being made toward accurate models for risk assessment of both diseases, but key challenges remain in terms of integrating models of pre- and post-infection events, quantifying the roles of insects in these diseases, and characterizing interactions among competing fungi and the environment.  相似文献

黄淮海夏玉米主产区穗腐病病原菌的分离鉴定   总被引:6,自引:0,他引:6  
为明确我国黄淮海夏玉米主产区玉米穗腐病的病原菌种类、优势种群及虫害、年度、省份对病原菌的影响,以形态学为基础,结合分子生物学方法对2013、2015年随机采自河南、河北、山东3省的155份玉米穗腐病样品进行分离鉴定。结果表明,引起黄淮海夏玉米主产区玉米穗腐病的主要致病菌为镰孢菌Fusarium spp.,包括拟轮枝镰孢F.verticillioides、禾谷镰孢F.graminearum、层出镰孢F.proliferatum、木贼镰孢F.equiseti及藤仓镰孢F.fujikuroi,分离频率分别为49.7%、28.4%、12.3%、3.9%和1.3%;其次为木霉菌Trichoderma spp.,包括哈茨木霉T.harzianum、绿色木霉T.viride和棘孢木霉T.asperellum,分离频率分别为8.4%、3.2%和5.2%;青霉菌Penicillium spp.分离频率较低,为14.2%;曲霉菌Aspergillus spp.包括黑曲霉A.niger和黄曲霉A.flavus,分离频率分别为2.6%和1.9%。研究表明,黄淮海主产区玉米穗腐病优势病原菌为拟轮枝镰孢、禾谷镰孢和木霉菌,不同省份不同年度间病原菌种类及优势病原菌存在差异,虫害能加重玉米穗腐病的发生。  相似文献

Scots pine trees (Pinus sylvestris) recovering from a 90–100% defoliation 2–3 years previously were pretreated with small mechanical wounds or inoculations with the blue-stain fungi Leptographium wingfieldii and Ophiostoma canum. Pretreated trees were less susceptible to a subsequent massive inoculation with L. wingfieldii than untreated control trees, which were extensively colonised by the mass-inoculation. A low pretreatment dosage of L. wingfieldii was somewhat more effective in inducing disease resistance than a higher dosage. Pretreatment with L. wingfieldii, O. canum, and mechanical wounding were about equally effective inducers of resistance in Scots pine, even though L. wingfieldii is known to produce much more extensive phloem necrosis than the other pretreatments. Thus, the strength of the induced resistance response did not depend on the amount of host tissues that was destroyed by the pretreatment. Previously, induced disease resistance has been demonstrated in Norway spruce (Picea abies), and the present study shows that similar responses can be activated in Scots pine.  相似文献

The GRAS compounds cinnamaldehyde (at 30 ppm), acetaldehyde (at 70 ppm), benzalde-hyde (at 50 ppm) and potassium metabisulphite (at 250 ppm) either completely inhibited or significantly reduced thein vitro mycelial growth ofGliocephalotrichum microchlamydosponim,Colletotrichum gloeosporioides andBotryodiplodia theobromae, the causative fungi of brown spot, anthracnose and stem end rot, respectively, of rambutan fruits,Nephelium lappaceum. The four compounds also significantly reduced the severity of all three diseases and the activity of pectic lyase and polygalacturonase enzymes secreted by the three fungi.  相似文献

We isolated eight polymorphic microsatellite markers for the basidiomycete Armillaria cepistipes and characterised them by analysing 50 isolates representing two geographically distinct populations from Switzerland and the Ukraine. The number of alleles per locus and population varied from one to eight, resulting in 43 alleles over the eight loci and two populations. In both populations, no significant linkage disequilibrium was observed between pairs of loci. Significant (P < 0.05) deviations from Hardy-Weinberg equilibrium were observed at one locus in the Swiss population and at three loci in the Ukrainian population. Of the eight loci developed for A. cepistipes, six were also polymorphic in A. gallica, four in A. ostoyae, two in A. mellea, and one in A. borealis. Beside the potential to be used for population genetic studies on A. cepistipes, these microsatellites thus represent additional molecular markers for three of the four annulated Armillaria species occurring in Europe.  相似文献

Collybia fusipes is the cause of a root rot of Quercus petraea (sessile oak), Q. robur (pedunculate oak) and Q. rubra (red oak). This parasite is often reported to cause problems in stands of red oaks and field evidence suggests that this North American oak species is more susceptible than the two European oak species. Young saplings of the three oak species and of chestnut, Castanea sativa, in the glasshouse, and also mature sessile and red oaks growing in the same stand were inoculated with C. fusipes to compare their susceptibility. Red oak, both as young seedlings and mature trees, was more susceptible to C. fusipes than sessile oak. Chestnut seedlings were as susceptible as sessile oak. Susceptibility of pedunculate oak seedlings was intermediate between red oak and sessile oak. In one experiment this species was significantly less susceptible than red oak, and in the other it was as susceptible.  相似文献

A survey was made to identify the most important soilborne fungal pathogens of asparagus crops in the Netherlands. Ten plants were selected from each of five fields with a young (1–4 y) first planting, five fields with an old (6–13 y) first planting and five fields with a young replanting. The analysis included fungi present in the stem base and the roots of plants with symptoms of foot and root rot or showing growth decline without specific disease symptoms. Isolates of each species were tested for pathogenicity to asparagus on aseptically grown plantlets on Knop's agar. Symptoms were caused byFusarium oxysporum, F. culmorum, Botrytis cinerea, Penicillium verrucosum var.cyclopium, Cylindrocarpon didymum, Phialophora malorum, Phoma terrestris andAcremonium strictum. F. oxysporum was by far the most common species and was isolated from 80% of the plants. Not all of its isolates were pathogenic to asparagus. Symptoms were caused by 67%, 78% and 93% of the isolates obtained from young first plantings, old first plantings and replantings, respectively.F. culmorum was isolated from 31% of the plants. Two other notorious pathogens of asparagus,F. moniliforme andF. proliferatum, did not occur in our samples.Species causing symptoms in the vitro test that were found on more than 5% of the plants were additionally tested for their pathogenicity in pot experiments.F. oxysporum f.sp.asparagi caused severe foot and root rot, significantly reduced root weights and killed most of the plants.F. culmorum caused lesions on the stem base often resulting in death of the plant.P. terrestris, a fungus only once reported as a pathogen of asparagus, caused an extensive root rot, mainly of secondary roots that became reddish. The fungus was isolated in only a few samples and is not to be regarded as an important pathogen in Dutch asparagus crops.P. malorum caused many small brown lesions on the stem base and incidentally also on the upper part of small main roots. This is the first report of its pathogenicity to asparagus. The fungus is one of the organisms inciting spear rust and it reduced crop quality rather than crop yield.P. verrucosum var.cyclopium andC. didymum did not cause symptoms in pot experiments.Because of its predominance on plants with foot and root rot and its high virulence,F. oxysporum f.sp.asparagi was considered to be the main soilborne pathogen of asparagus in the Netherlands.  相似文献

Severe rot was found at the base of leaves and stems of chingensai (Brassica campestris L. chinensis group) in Okayama Prefecture in 2000. The causal fungi were morphologically identified as Pythium ultimum Trow var. ultimum and P. aphanidermatum (Edson) Fitzpatrick. This is the first report of rot caused by Pythium species on chingensai. We named this disease Pythium rot of chingensai.  相似文献

Trichoderma harzianum (TrH 40) isolated from soil samples from rambutan orchards (Nephelium lappaceum) had antagonistic effects against three postharvest pathogens of rambutan:Botryodiplodia theobromae, Colletotrichum gloeosporioides andGliocephalotrichum microchlamydosporum, causative fungi of stem end rot, anthracnose and brown spot, respectively. The effects were due to both antibiosis and mycoparasitism.T. harzianum (TrH 40) treatment, while reducing the occurrence of the three postharvest diseases, also retained the overall quality and color of the fruits.  相似文献

为明确不同梨和苹果种质对阿太菌果腐病菌Athelia bombacina的抗性以及筛选防治其有效杀菌剂,采用离体菌丝块有伤接种方法对40份梨种质和154份苹果种质进行病斑直径测定,通过聚类分析法和平均病斑直径法对不同种质进行了抗病性分级,并利用菌丝生长速率法测定了13种常用杀菌剂对阿太菌果腐病菌的毒力。结果表明,根据聚类分析法和平均病斑直径法均可将梨和苹果种质划分为高抗、抗、中抗、感和高感5类。与平均病斑直径法相比,梨和苹果种质分别以欧式距离为14和10作为最佳聚类分割点时进行聚类分析能够更加科学地划分类别,从40份梨种质中筛选出金锤子梨1个高抗种质,仅占总数的2.50%,从154份苹果种质中筛选出垂丝海棠、莫斯科透明、新疆苹果、路边石、伏帅等32个高抗种质,占总数的22.73%。药剂试验结果表明,戊唑醇、腈菌唑、咯菌腈和噻呋酰胺对阿太菌果腐病菌菌丝生长的抑制效果较好,抑制中浓度EC_(50)分别为0.027、0.048、0.054和0.095 mg/L。表明梨和苹果种质均可被阿太菌果腐病菌侵染,但不同种质间抗病性差异明显,戊唑醇是采前防治阿太菌果腐病菌菌丝生长的最佳药剂。  相似文献

Fifteen species of dermestid beetles were recorded at ‘Evolution Canyon’ (EC), Lower Nahal Oren, Mt. Carmel, Israel. They represent ~35% of known Israeli dermestid species. The following three species were recorded for the first time in Israel:Trogoderma svriaca Dalla Torre, 1911;Ctesias svriaca Ganglbauer, 1904; andAnthrenus (s.str.) jordaniens Pic, 1934. Adults of 13 species were collected on the more solar radiated, warmer and climatically more fluctuating south-facing slope (SFS); ten species were collected on the opposite, north-facing slope (NFS), which was cooler and climatically more stable. The abundance of adult dermestid beetles was 1.9 times higher on the SFS than on the NFS (86 and 47, respectively). Species richness and abundance distribution at EC (three collecting stations on each slope and one at the valley bottom) were significantly negatively correlated with the plant cover that consisted of trees and bushes (Spearmanr s ,P=0.007 and 0.039, respectively) and perennials (Spearmanr s ,P=0.039 and 0.077, respectively), indicating that non-woody plants were preferred by adult dermestid beetles.  相似文献

为探讨大豆疫霉Phytophthora sojae细胞凋亡潜在的调控机制,根据已知的细胞凋亡蛋白,利用在线工具BLASTP、pFAM和SMART在大豆疫霉蛋白组数据库中鉴定细胞凋亡同源蛋白并构建其进化树,通过转录组数据和实时荧光定量PCR技术分析细胞凋亡相关基因在大豆疫霉生长、发育及侵染不同时期的表达情况。结果显示:在大豆疫霉中共鉴定到13个细胞凋亡同源蛋白,包括核酸内切酶G(PsNUC1)、细胞色素c(PsCYCS)、凋亡诱导因子(PsAIF)、丝氨酸蛋白酶(PsHtrA-1、PsHtrA-2和PsHtrA-3)、多聚ADP核糖聚合酶(PsPARP-1、PsPARP-2和PsPARP-3)和TatD核酸酶(PsTatD1、PsTatD2、PsTatD3和PsTatD4)。在进化上,PsNUC1、PsCYCS、PsAIF、PsHtrA-1、PsPARP-1、PsPARP-2、PsPARP-3、PsTatD1和PsTatD2与人及秀丽隐杆线虫Caenorhabditis elegans的同源蛋白亲缘关系较近,而与真菌相关蛋白亲缘关系较远,PsHtrA-2、PsHtrA-3、PsTatD3和PsTatD4与酿酒酵母Saccharomyces cerevisiae相关蛋白相似度更高,说明大豆疫霉细胞凋亡蛋白在进化中发生了较大变异。大豆疫霉细胞凋亡相关基因PsHtrA-1和PsRARP-1在孢子囊阶段诱导表达,PsHtrA-2和PsRARP-2在游动孢子阶段上调表达,PsAIF、PsHtrA-3、PsRARP-1和PsRARP-2在侵染阶段明显诱导表达,PsCYCS在侵染阶段下调表达。细胞凋亡相关基因在大豆疫霉不同阶段的表达模式有较大差异,说明细胞凋亡在大豆疫霉生长、发育及致病过程中具有重要作用。  相似文献

Recent data on the epidemiology of the common mycotoxigenic species of Fusarium, Alternaria, Aspergillus and Penicillium in infected or colonized plants, and in stored or processed plant products from the Mediterranean area are reviewed. Emphasis is placed on the toxigenicity of the causal fungal species and the natural occurrence of well known mycotoxins (aflatoxins, ochratoxins, fumonisins, trichothecenes, zearalenone, patulin, Alternaria-toxins and moniliformin), as well as some more recently described compounds (fusaproliferin, beauvericin) whose toxigenic potential is not yet well understood. Several Fusarium species reported from throughout the Mediterranean area are responsible of the formation of mycotoxins in infected plants and in plant products, including: Fusarium graminearum, F. culmorum, F. cerealis, F. avenaceum, F. sporotrichioides and F. poae, which produce deoxynivalenol, nivalenol, fusarenone, zearalenone, moniliformin, and T-2 toxin derivatives in wheat and other small grains affected by head blight or scab, and in maize affected by red ear rot. Moreover, strains of F. verticillioides, F. proliferatum, and F. subglutinans, that form fumonisins, beauvericin, fusaproliferin, and moniliformin, are commonly associated with maize affected by ear rot. Fumonisins, were also associated with Fusarium crown and root rot of asparagus and Fusarium endosepsis of figs, caused primarily by F. proliferatum. Toxigenic A. alternata strains and associated tenuazonic acid and alternariols were commonly found in black mould of tomato, black rot of olive and citrus, black point of small cereals, and black mould of several vegetables. Toxigenic strains of A. carbonarius and ochratoxin A were often found associated with black rot of grapes, whereas toxigenic strains of A. flavus and/or P. verrucosum, forming aflatoxins and ochratoxin A, respectively, were found in moulded plant products from small cereals, peanuts, figs, pea, oilseed rape, sunflower seeds, sesame seeds, pistachios, and almonds. Finally, toxigenic strains of P. expansum and patulin were frequently found in apple, pear and other fresh fruits affected by blue mould rot, as well as in derived juices and jams.  相似文献

小麦茎基腐病生防菌株YB-161的分离鉴定及防效测定   总被引:4,自引:0,他引:4  
为筛选防治小麦茎基腐病的高效生防菌株,采用稀释涂布法从小麦茎基腐病病土中分离细菌,以假禾谷镰孢Fusarium pseudograminearum为靶标菌,通过平板对峙培养法和室内盆栽试验筛选优良菌株并测定其田间防治效果,分析优良菌株发酵滤液对假禾谷镰孢的抑菌能力,并结合形态特征、Biolog微生物自动鉴定系统和gyrA序列分析对其进行种类鉴定,以及测定该菌株的抑菌谱。结果表明,10份供试土样中共分离到98株细菌,对假禾谷镰孢的菌丝生长抑制率超过40.00%的菌株有14株,其中菌株YB-161对假禾谷镰孢的菌丝生长抑制率最高,达63.50%。室内盆栽试验结果显示,菌株YB-161处理小麦的病情指数最低,为31.40,对小麦茎基腐病的防治效果最好,为68.15%,其对小麦也表现出较好的促生作用。田间调查结果显示,菌株YB-161菌液拌种处理对小麦茎基腐病的田间防治效果高于52.35%,并对小麦具有一定的增产作用。菌株YB-161的发酵滤液5倍稀释液和10倍稀释液对假禾谷镰孢的菌落抑制率分别达66.67%和44.33%,该菌株被鉴定为解淀粉芽胞杆菌Bacillus amyloliquefaciens。此外,菌株YB-161对辣椒炭疽病菌Colletotrichum capisci、烟草疫霉病菌Phytophthora parasitica等5种植物病原真菌均有较好的拮抗作用。表明生防菌株YB-161对绿色防控小麦茎基腐病具有良好的开发利用潜能。  相似文献

为明确铁皮石斛根腐病病原真菌,于其主产地浙江省金华市武义县收集铁皮石斛根腐病病株,采用平板分离方法对病原真菌进行分离,使用镰刀菌种特异性引物并结合ITS和TEF序列分析及形态学鉴定确定该镰刀菌的分类地位。结果表明,共分离纯获得真菌117株,其中有105株镰刀菌;经分子生物学分析及形态学鉴定结果显示,分离出的镰刀菌为层出镰刀菌Fusarium prolife‐mum、茄病镰刀菌F. solani、尖孢镰刀菌F. oxysporum和厚垣镰刀菌F. chlamydosporum四个种,其中层出镰刀菌在数量上具有优势地位,占总镰刀菌数的44.8%;茄病镰刀菌、尖孢镰刀菌、厚垣镰刀菌分别占总镰刀菌数的21.0%、15.2%和19.0%。在致病性测定中发现层出镰刀菌和茄病镰刀菌并不具备致病性,尖孢镰刀菌的致病性明显弱于厚垣镰刀菌,表明厚垣镰刀菌为浙江省金华市武义县铁皮石斛根腐病的主要致病菌。  相似文献

The huanglongbing (HLB) disease of citrus trees, caused by Candidatus Liberibacter asiaticus and Ca. Liberibacter americanus, was first reported in Brazil in March, 2004. The presence of the disease has caused serious concerns among growers. Pruning experiments were conducted to determine if removal of symptomatic branches or the entire canopy (decapitation) would eliminate infected tissues and save HLB-affected trees. Pruning was done in five blocks on a total of 592 3- to 16 year-old ‘Valência’, ‘Hamlin’ or ‘Pêra’ sweet orange trees showing no symptoms or with two levels of symptom severity. Ten decapitated trees per block were caged and all trees were treated with insecticides to control the psyllid vector, Diaphorina citri. Mottled leaves reappeared on most symptomatic (69.2%) as well on some asymptomatic (7.6%) pruned trees, regardless of age, variety, and pruning procedure. Presence of the pathogen (Ca. Liberibacter americanus) in all symptomatic trees was confirmed by PCR. In general, the greater the symptom severity before pruning the lower the percentage of trees that remained asymptomatic after pruning.  相似文献
