Three diploid citrus somatic hybrids (cybrids) were produced by fusions combining nucellar callus-derived protoplasts of Willow Leaf mandarin (Citrus deliciosa Ten.) and Commune clementine (Citrus clementina Hort. ex Tan.) with, respectively, leaf protoplasts of Eureka lemon [Citrus limon (L.) Burm.] and Marumi kumquat [Fortunella japonica (Thunb.) Swing.] and leaf protoplasts of Marumi kumquat. Ploidy and origins of the nuclear, chloroplastic, and mitochondrial genomes were investigated by flow cytometry and nuclear and cytoplasmic simple sequence repeat analyses. Volatile compounds were extracted from the leaves of the three cybrids by a pentane/ether (1:1) mixture, analyzed by GC-MS, and compared to those of their parents. The cybrids were found to be very close to their nucleus-giving parent, suggesting that the main information for volatile compounds biosynthesis is contained in the nucleus. However, nucleo-cytoplasmic interactions occurred: the (mandarin + lemon) cybrid, possessing nucleus and chloroplasts of lemon and mitochondria from mandarin, synthesizes more monoterpene alcohols and esters than its nucleus-giving parent; the (clementine + kumquat) cybrid, possessing nucleus from kumquat and organelles from mandarin, synthesizes more monoterpene and sesquiterpene hydrocarbons and sesquiterpene alcohols than its nucleus-giving parent.  相似文献

In this study, the contamination by chloroparaffin of Sicilian and Calabrian citrus essential oils, produced in the crop years 1994-1996, was investigated. The analyses were carried out on 102 lemon oils, 98 orange oils, and 96 mandarin oils, using a dual-channel GC-ECD. It was found that 53% of lemon oil, 33% of orange oil, and 38% of mandarin oil samples were contaminated. The mean contamination levels were 7.1 ppm (lemon), 2.5 ppm (orange), and 5.3 ppm (mandarin). The highest concentration of chloroparaffin found was 60 ppm in a lemon oil sample.  相似文献

Fruits from seven different varieties of Olea europaea L., grown in the same environmental conditions, were harvested in two succeeding years at the same ripening degree and immediately processed. The oils obtained were submitted to gas chromatographic determination of the volatile compounds extracted by dynamic headspace technique. The results demonstrated that the accumulation of the different metabolites in the oils obtained from the various cultivars were strictly connected with the varietal parameter because of the enzyme differences genetically determined. This feature made possible the differentiation of the examined cultivars on the basis of the percent of each metabolite from the enzymatic transformation of 13-hydroperoxides of linolenic acid. Oils from Picual and Koroneiki varieties cultivated in Spain and Greece, respectively, showed contents of volatiles very similar to those detected in the oils of the same varieties cultivated in Italy, proving that they were not significantly influenced by the environmental conditions.  相似文献

Chlorophyll, lutein, and beta-carotene contents in Actinidia fruits were determined by high-performance liquid chromatography in various genotypes, including five Actinidia deliciosa, seven Actinidia chinensis, two Actinidia rufa, five Actinidia arguta, and three interspecific hybrids. The concentrations of chlorophyll, lutein, and beta-carotene in the fruit of A. deliciosa Hayward were 1.65, 0.418, and 0.088 mg/100 g fresh weight, respectively. Of A. deliciosa cultivars, Koryoku showed significantly higher concentrations in chlorophyll, lutein, and beta-carotene than Hayward. In most cultivars of A. chinensis, although both chlorophyll and lutein contents were significantly lower than in Hayward, the beta-carotene content tended to be slightly higher. In A. rufa, A. arguta, and their interspecific hybrids, the contents of chlorophyll, lutein, and beta-carotene were much higher than in Hayward. In particular, these fruits were found to be the richest dietary source of lutein among commonly consumed fruits.  相似文献

Sugar, organic acid, and carotenoid are the most important indicators of fruit taste and nutritional and organoleptic quality. These components were studied on fruit pulp of the cybrid between Willow leaf mandarin ( Citrus deliciosa Ten.) and Eureka lemon [ Citrus limon (L.) Burm.] and the two parents. The cybrid possessed nuclear and chloroplast genomes of Eureka lemon plus mitochondria from Willow leaf mandarin. The impact of new mitochondria on fruit quality was studied during the mature period. Levels of organic acids were slightly higher in the cybrid fruit pulp than in Eureka lemon. No significant difference in sugar and carotenoid content was observed between the cybrid and the lemon. Results confirm that the main genetic information for the biosynthesis of sugars, organic acids, and carotenoids is contained in the nucleus. In Citrus, cybridization can be used as a strategy to breed specific traits associated with mitochondrial genomes, such as male sterility, without affecting the main organoleptic and nutritional qualities.  相似文献

Organochlorine pesticide contamination in 148 lemon essential oils, 123 sweet orange oils, 121 mandarin oils, and 147 bergamot oils produced in Italy in the years 1991-1996 was studied by HRGC-ECD. Confirmation analyses were carried out by GC-MS. Tetradifon, dicofol and its decomposition product 4,4'-dichlorobenzophenone were found. Over the course of the study dicofol and tetradifon residues steadily decreased; the percentage of contaminated samples reflects this course and decreases considerably from 1991 to 1996.  相似文献

Contamination by phthalate esters of Sicilian and Calabrian citrus essential oils, produced in the crop years 1994-1996, was investigated using a GC-MS system with direct injection of the samples. A total of 35 lemon oils, 31 orange oils, and 21 mandarin oils were analyzed. Diisobutyl phthalate and/or bis(2-ethylhexyl) phthalate were found in almost all samples, while di-n-butyl phthalate was present in 8. Concentrations up to a maximum of 62 ppm were found for diisobutyl phthalate and up to a maximum of 29.9 ppm for bis(2-ethylhexyl) phthalate.  相似文献

The composition of the laboratory-prepared essential oils from Uruguayan Nova and Satsuma mandarins has been studied. The volatile fraction was analyzed by HRGC and HRGC/MS (quadrupole); 79 and 73 components were identified in Nova and Satsuma mandarin oils, respectively. The linear retention indices were calculated for almost all identified components on two different stationary phases. The enantiomeric distribution of beta-pinene, sabinene, limonene, linalool, and alpha-terpineol was studied by multidimensional gaschromatography (MDGC). Polymethoxylated flavones, present in the nonvolatile residue, were analyzed by normal-phase HPLC.  相似文献

Fusion of citrus diploid parental protoplasts generates allotetraploid hybrids which do not retain their parental traits with regard to leaf aroma compound biosynthesis. The aim of this study was thus to examine hybrid leaf proteomes in comparison with their parents. Leaf soluble proteins from two citrus allotetraploid hybrids (mandarin + lime and mandarin + kumquat) and their diploid parents (mandarin, lime, and kumquat) were submitted to 2-D gel electrophoresis. Leaf proteome maps of the tetraploid hybrids were compared with those of their parents on the basis of the presence/absence of spots and of their spot relative volumes. The two allotetraploid hybrid maps were found closer to that of their mandarin parent than to those of their nonmandarin parents in terms of the presence/absence of spots as well as at a quantitative level. This approach has to be related to the already observed dominance of mandarin in allotetraploids with regard to volatile compound biosynthesis in leaves.  相似文献

The aim of this research is to find if there is direct evidence relating the fatty acid composition of olive oils to specific cultivars grown within a well-limited geographical region. To group olive oils according to their own cultivars,(13)C high-field nuclear magnetic resonance (NMR) and gas chromatography (GC) were used to analyze 60 extra virgin olive oils from the same Italian region (southwestern Sicily) obtained from four monovarietal cultivars. The (13)C NMR spectrum provides information about glycerol triesters of olive oils, i.e., about the acyl composition of major components and about the fatty acids' positional distribution on the glycerol moiety. GC gives the complete fatty acid profile of olive oil samples. Selection of NMR and GC peaks on the basis of their sensitivity to the different cultivars was performed by using multivariate analysis of variance (MANOVA). Principal component analysis, tree clustering analysis, multidimensional scaling (MDS), and linear discriminant analysis (LDA) were then performed on the MANOVA-selected peaks. Results obtained from (13)C NMR and GC techniques combined with the multivariate statistical procedure are in good agreement and prove the usefulness of fatty acids analysis to group the monovarietal olive oils belonging to the same cultivars. Grouping of olive oils according to their cultivars occurs for particular (13)C resonances all belonging to fatty chains in the sn 1,3 position of the glycerol moiety.  相似文献

Vitamin C content in the fruit of various cultivars of kiwifruit and other Actinidia species was estimated by determination of L-ascorbic acid and L-dehydroascorbic acid using ion-pair reversed-phase high-performance liquid chromatography. Fruit of A. deliciosa cv. Hayward, the most common commercially available cultivar, contained 65.5 mg/100 g fresh weight (FW) vitamin C. Vitamin C content in A. deliciosa fruit varied from 29 mg/100 g FW to 80 mg/100 g FW. In most cultivars of A. chinensis, vitamin C content in fruit was higher than that of Hayward. In particular, vitamin C content in cv. Sanuki Gold fruit reached more than 3-fold that of Hayward on a weight for weight basis. In A. argutafruit, there was wide variation in vitamin C content, with concentrations ranging from 37 to 185 mg/100 g FW. In cv. Gassan, Issai, and Mitsuko, vitamin C content of the fruit was much higher than that of Hayward. In A. arguta fruit, the ratio of L-ascorbic acid to total ascorbic acid tended to be higher than that of other species.  相似文献

Citrus essential oils are widely used in the food, cosmetics, and pharmaceutical industries, so the determination of heavy metals content is of great importance to guarantee their quality. The present work deals with the quantification of Cd(II), Cu(II), Pb(II), and Zn(II) in different varieties of citrus essential oils, using derivative potentiometric stripping analysis. Two different metals extraction procedures, involving concentrated hydrochloric acid treatment and acid-alcoholic dissolution, are tested on lemon, mandarin, sweet orange, and bergamot essential oils, and they give very similar results. Cd(II), Cu(II), Pb(II), and Zn(II) recovery tests spanned from 95 to 100.50%, providing evidence that metals quantification remained unaffected by the cleanup steps of the two procedures. The repeatability of the hydrochloric acid extraction method, applied on different varieties of essential oils, is >95.00% for Cd(II), Cu(II), Pb(II), and Zn(II), whereas the repeatability of the acid-alcoholic dissolution method is >93.00% for Cu and Cd only in lemon oil. Detection limits obtained for the four analytes, using both procedures, ranged from 0.10 to 0.98 ng g(-)(1) in lemon, mandarin, sweet orange, and bergamot essential oils.  相似文献

Kiwifruit cultivars (Actinidia chinensis and A. deliciosa) generally have fruit with yellow or green flesh when ripe. A small number of genotypes also have red pigments, usually restricted to the inner pericarp but varying in intensity and in distribution within the fruit. Carotenoids, chorophylls, and anthocyanins were extracted from the fruit pericarp of such red-fleshed kiwifruit selections. Pigments were analyzed by HPLC and identified by comparison with authentic standards and by liquid chromatography-mass spectroscopy to obtain a tentative identification of the major anthocyanins in red-fleshed kiwifruit. The yellow and green colors of the outer fruit pericarp are due to different concentrations and proportions of carotenoids and chlorophylls. The red color found mainly in the inner pericarp is due to anthocyanins. In the A. chinensis genotypes tested the major anthocyanin was cyanidin 3-O-xylo(1-2)-galactoside, with smaller amounts of cyanidin 3-O-galactoside. In the A. deliciosa genotypes analyzed, cyanidin 3-O-xylo(1-2)-galactoside was not detected; instead, the major anthocyanins identified were cyanidin 3-O-galactoside and cyanidin 3-O-glucoside. However, the two species did not differ consistently in anthocyanin composition.  相似文献

DNA analysis enables genome fingerprinting with consequent identification of different individuals. In the agro-food industry, this can have interesting applications for the identification of species and cultivars of both raw materials and processed food. In this investigation, the efficiency of DNA microsatellite analysis in identifying virgin olive oils from different cultivars was evaluated. Ten virgin oils were obtained in the laboratory from olives of 10 different cultivars and the DNA extracted from the cell residues, recovered by oil centrifugation, was used as a template with seven different primer pairs of microsatellite sequences. The electrophoretic patterns showed an adequate level of amplification and were identical to those obtained from leaves and drupes of the same cultivar. By analyzing all the patterns obtained, the smallest number of microsatellites able to distinguish the examined oils was established and an identification key for the different oils was developed.  相似文献

采用SSR标记分析了43份柑橘(Citrus)不育、低育、含不育胞质及13份可育材料的遗传多样性。结果表明,8对SSR引物扩增得到35个等位基因,平均每个位点有4.4个等位基因。利用NTSYSpc2.10统计分析软件,UPGMA法聚类分析表明,56份柑橘资源可分为3组。宽皮柑橘类品种为第一组,甜橙、葡萄柚、椪柑及杂柑类品种为第二组,包含不育胞质的澳洲指橘为第三组。SSR聚类分析还表明本地广橘与温州蜜柑有着紧密的亲缘关系,而与早橘、槾橘、本地早等的亲缘关系较远?涕?不育与低育资源的遗传多样性分析将为此类资源的收集保存及应用提供依据。  相似文献

A fast HPLC method for the determination of the oxygen heterocyclic compounds of citrus essential oils was developed. Five different oils were analyzed under identical conditions, by reversed-phase HPLC with photodiode array detector, for a direct comparison of the composition of their oxygen heterocyclic fraction. Analysis time was 7 min. The oils analyzed were lemon, bergamot, mandarin, sweet orange, and bitter orange. The method developed is good for rapid screening or fingerprinting of these essential oils; a slightly slower method is recommended for higher resolution and better quantitative results.  相似文献

Citrus performance is strongly related with rootstock. This study was conducted to investigate leaf nutrient contents of ‘Okitsu’, ‘Clausellina’ and ‘Silverhill’ mandarin cultivars budded onto sour orange, ‘Carrizo’ and ‘Troyer’ citrange rootstocks in Dörtyol, Turkey in 2004 and 2005. The maximum nitrogen (N), potassium (K), and copper (Cu) contents were determined for ‘Clausellina’; phosphorus (P) for ‘Okitsu’; and sodium (Na) for ‘Silverhill’. Calcium (Ca), magnesium (Mg), iron (Fe), manganese (Mn), and zinc (Zn) uptake were similar for the mandarin cultivars. ‘Carrizo’ citrange at N, K, Mg, Mn, and Cu uptake; ‘Troyer’ citrange at N, P, K, and Fe uptake; and common sour orange at Ca, Zn, and Na uptake was superior on the other rootstocks. It was observed that ‘Carrizo’ and ‘Troyer’ citrange rootstocks had advantages over sour orange in nutrient uptake. Thus, growth performance, yield, and quality parameters considered, ‘Carrizo’ and/or ‘Troyer’ citranges could be suggested as rootstocks for the studied mandarin cultivars under similar ecological conditions.  相似文献

Peel and leaf oils of 43 taxa of lemons and limes were obtained from fruits and leaves collected from trees submitted to the same pedoclimatic and cultural conditions. Their chemical composition was investigated by capillary GC, GC-MS, and (13)C NMR, and the results were submitted to principal component analysis to check for chemical variability. Three major chemotypes were distinguished for lemon peel oils: limonene; limonene/beta-pinene/gamma-terpinene; and limonene/linalyl acetate/linalool. Two chemotypes were identified for lemon leaf oils: limonene/beta-pinene/geranial/neral and linalool/linalyl acetate/alpha-terpineol. In lime peel oils, four chemotypes were distinguished: limonene; limonene/gamma-terpinene; limonene/beta-pinene/gamma-terpinene; and limonene/gamma-terpinene/beta-pinene/oxygenated products. Four others were identified for lime leaf oils: beta-pinene/limonene; limonene/geranial/neral; limonene/linalool/citronellal; and limonene/sabinene/citronellal/linalool. The purpose of the work was to investigate the effect of the maturation process of the olive fruit on the phenolic fraction of drupes and oils from Arbequina, Farga, and Morrut cultivars. The level in the phenolic content of olive drupes declines rapidly during the black maturation phase. A general decreasing trend was observed too in the phenolic content of olive oils during the ripening process in the three varieties studied. Important differences in the high-performance liquid chromatography profile between varieties were observed. These included the presence of very low amounts of lignans in olive oils proceeding from the Morrut cultivar, and the presence of three peaks after elution of 3,4-DHPEA-EDA in the Farga and Morrut cultivars, which could be used as differentiating parameters. Sensory profile differences were observed between olive cultivars and due to the ripening process.  相似文献

Two cultivars of hulled barley (Thoroughbred and Nomini) and two cultivars of hulless barley (Doyce and Merlin) were scarified to abrade the outer layers of hull and pericarp. The resulting scarification fines fractions were evaluated as potential sources of functional lipids (phyto‐sterols, tocopherols, and tocotrienols). The levels of total phytosterols and total tocotrienols in the barley scarification fine fractions were probably not high enough to justify their use as functional foods. However, the levels of total phytosterols and total tocotrienols in the oils extracted from both whole kernels and scarified fines were both sufficiently high to make it reasonable to consider their potential use as new functional oils. Indeed, the levels of total tocotrienols in barley oils (2,911–6,126 mg/kg of oil) are several‐fold higher than those reported in two other oils that are being marketed as high in tocotrienols: palm oil (530 mg/kg) and rice bran oil (770 mg/kg). The levels of total phytosterols in barley oils range from 1.20 to 9.60 g/100 g of oil.  相似文献
