Polymorphism of goat milk alphas1-casein was determined and potential relations between genetic variants of this protein fraction and goat performance were evaluated. The investigations were performed on 598 goats assigned to of 4 breed groups (White improved 254 units, Coloured improved--124, White non-improved--146 and Coloured non-improved--74). For each goat, alphas1-casein polymorphism was determined in polyacrylamide gel by the PAGE-SDS method and percentage of milk alphas1-casein and gene frequency established. There was evaluated goat performance at successive lactations. In the goat population investigated, AA, AB, BB, AE, BE and EE alphas1-casein genotypes were identified. In all four breeds, alphas1-casein genotype EE clearly predominated (27.2-39.2%), recognized as "medium" and its share was higher in the groups of non-improved goats. It was conditioned by high frequency of gene E alphas1-casein (0.419-0.622). Generally, EE genotype percentage was higher in the non-improved goat groups. The improved goats, though, obtained higher productivity in each of the lactation studied. Analysis of relationships between alphas1-casein genetic variants and goats performance confirmed a significant influence on milk, protein and fat yields only in the Coloured improved goat group. There was revealed a more general tendency indicating a significant impact of "strong" alphas1-casein genotypes on a concentration of basic milk components, i.e. fat and protein, especially casein. In a group of goats producing milk of the highest casein content (over 2.4%) and protein (over 3.0%), the animals showing "strong" alphas1-casein variants dominated (85 and 70 %).  相似文献

The ϰ-casein content in milk from the Danish cattle breeds SDM, RDM and Jersey was investigated by means of immunoquantitation in antibody containing agarose gels. The method, which determines the total ϰ-casein complex, is suitable for routine work. The reproducibility of the method was found to be ± 4.1 %.The ϰ-casein concentration, expressed as percentage of the total protein content in milk, was significantly lower in milk from SDM (21.5%) and RDM (21.3%) than in Jersey (23.9%). The investigation showed that the ϰ-casein constitutes a greater part of the protein in milk than previously assumed.The amount of ϰ-casein was positively correlated with the protein content in milk, but there was no relation between the relative ϰ-casein content and the percentage of fat, milk yield and the time after calving.Keyword: kappa-casein, milk, cattle, immunoquantitation  相似文献

Effects of DGAT1 variants on milk production traits in German cattle breeds   总被引:15,自引:0,他引:15  
Various QTL mapping experiments led to the detection of a QTL in the centromeric region of cattle chromosome 14 that had a major effect on the fat content of milk. Recently, the gene encoding diacylglycerol O-acyltransferase (DGAT1) was proposed to be a positional and functional candidate for this trait. This study investigated the effects of a nonconservative lysine to alanine (K232A) substitution in DGAT1, which very likely represents the causal mutation, on milk production traits. Existing granddaughter designs for Fleckvieh and German Holstein, the two major dairy/dual-purpose breeds in Germany, were used to estimate allele frequencies and gene substitution effects for milk, fat, and protein yield, as well as fat and protein content. A restriction fragment length polymorphism assay was applied to diagnose the K232A substitution in DGAT1. Estimates of the allele frequencies for the lysine-encoding variant were based on maternally inherited alleles in sons and amounted to 0.072 for Fleckvieh and 0.548 for German Holstein. Effects of DGAT1 variants on content traits were pronounced; estimates of the gene substitution effect for the lysine-encoding variant were 0.35 and 0.28% for fat content and 0.10 and 0.06% for protein content in Fleckvieh and German Holstein, respectively. Conversely, negative effects of the lysine variant of -242 to -180 kg for Fleckvieh and -260 to -320 kg for German Holstein were revealed for milk yield from first to third lactation, resulting in enhanced fat yield of 7.5 to 14.8 kg in Fleckvieh and 7.6 to 10.7 kg in German Holstein. For protein yield, however, mainly negative effects of -3.6 to 0.2 kg in Fleckvieh and -4.8 to -5.2 kg in German Holstein were observed. Pearson correlations between residuals of milk yield and content traits were decreased when omitting DGAT1 effects in the analysis, thereby indicating that DGAT1 contributes to negative correlations between these traits. Molecular tests allow for the direct selection among variants; however, the benefits of the alternative alleles depend on economic weights given to the different milk production traits in the breeding goal.  相似文献

The objective of this study was to analyse the heritabilitiy of the lactational incidence of displaced abomasum (DA) and the relationships of DA with milk production traits in German Holstein cows. Data were recorded between February 1999 and January 2000 in cooperation with five veterinary practitioners. Their veterinary practices were located in the northern part of Lower Saxony. The investigation included 160 dairy farms under the official milk-recording scheme with 9,315 cows. The lactational incidence of the left abomasal displacement amounted to 1.21%, and of the right abomasal displacement to 0.41%, respectively. The linear heritability estimates for the lactational incidences of left and right DA were h(2) = 0.05 +/- 0.012 and h(2) = 0.004 +/- 0.005, respectively. Using the Dempster-Lerner-transformation the corresponding heritabilities were h(2) = 0.53 and 0.09, respectively. Milk losses for the lactation when DA was diagnosed were significant and reached 1016 kg milk, 41 kg fat, 36 kg protein and 0.07% protein. Fat content significantly increased by 0.18%. The analysis could not show significant differences between cows diagnosed with DA and cows not diagnosed with DA in the 305-day milk production traits of the lactation preceding the diagnosis of DA. There was also no indication for an unequal distribution of breeding values for milk performance traits between cows with and without DA. The additive genetic correlations between 305-day milk performance and left DA were low. The results indicated that cows with a high milk production and superior breeding values for milk performance were not exposed to an increased risk for DA.  相似文献

Crossbreeding effects on milk production traits of Czech dual‐purpose and dairy cattle breeds were estimated. Nearly 370 000 cows with known gene proportions from Czech Pied, Ayrshire or Holstein cattle were selected from the national milk recording data base. Single‐trait animal models were calculated for milk, fat and protein yield, fat and protein content. The model of Dickerson including additive, additive maternal, heterotic and recombination effects was used for the part of the animal model describing the crossbreeding effects in all calculations. For milk yield, the additive genetic effect (defined as deviation from Czech Pied cattle) was 850–900 kg for Holstein and 240–480 kg for Ayrshire. The maternal effects were low and negative. Low significant positive heterotic effects were observed being up to approximately 100 kg for Czech Pied × Holstein. The recombination effects were negative and statistically significant for Czech Pied × Holstein. The results for fat and protein yield were similar to the results for milk yield. For fat and protein content, nearly no statistically significant crossbreeding effects were found.  相似文献

Individual mid-lactation milk samples were collected from 116 cows of the Swedish Red and Swedish Holstein breeds with known genotypes of beta-casein, kappa-casein and beta-lactoglobulin. Detailed milk protein composition and allele-specific expression of beta-casein, kappa-casein and beta-lactoglobulin proteins in milk were analysed using reversed-phase high-performance liquid chromatography. Aggregate beta-/kappa-casein genotype was significantly associated with the kappa-casein concentration in milk. The lowest kappa-casein concentration was found in milk of cows with genotypes including kappa-casein E (A(1)A(2)/AE, A(1)A(1)/AE) and also A(2)A(2)/AA milk, whereas highest levels were associated with genotypes including kappa-casein B. Casein number was positively and concentration of beta-lactoglobulin negatively associated with the beta-lactoglobulin BB genotype. In heterozygote cows, beta-casein A(1) and beta-lactoglobulin A proteins were found at higher concentrations in milk compared with the protein variant encoded by the alternative allele at these loci, whereas kappa-casein A and B variants were found at similar concentrations in heterozygote AB cows.  相似文献

Two sets of data with a sum of 6318 lactations for 1391 Spanish Churra ewes, belonging to 10 flocks were used to study phenotypic, genetic and system efficiency parameters of lifetime traits in annual and accelerated lambing systems. The study included four life span traits, three productive traits and two reproductive traits. Trait averages for both milk yield and revenues from sold milk and weaned lambs were calculated per-day of lifetime, productive life and useful life. The animal model included flock and ewe birth year as fixed effects. Both effects contributed significantly to variation in most traits in the annual system while birth year was non-significant for most variables of the accelerated system. Milk production level was included in the model to analyze life span traits. It contributed significantly to variation in all life span traits in both systems. Means for productive and life span traits were significantly higher in the accelerated system compared with the annual system. The average interval between successive lambings was significantly shorter in the accelerated system, while age at first lambing was significantly lower in the annual system. Except for age at first lambing, heritability estimates were clearly lower in the case of the accelerated system (0.02 to 0.20) than in the annual system (0.08 to 0.45). Genetic correlations among traits were also lower in the case of the accelerated system. Failure to lamb three times in two consecutive years and the varying reproductive management among flocks in the accelerated system may be responsible for the differences in genetic results between the two systems. Improving reproductive performance is necessary, especially for the young high yielding ewes in the accelerated system. Management to ensure high fertility, especially among young ewes, would be of more importance than removing non-lambing ewes.  相似文献

Abstract

The milk composition in a Swedish herd, consisting of Swedish Red and White cows (SRB) and Swedish Holstein cows (SLB), and in a Danish herd, consisting of Danish Holstein-Friesian cows (SDM), was evaluated. Concentrations of the major milk proteins (κ-casein, αS1-casein, β-casein, β-lactoglobulin A, β-lactoglobulin B and α-lactalbumin) of 134 individual milk samples were determined by reversed phase (RP) HPLC. Other parameters determined included milk fat, urea, lactose, calcium, lactoferrin, somatic cell count and protein degradation (determined as level of free amino-terminals). Analysis of variance was used to compare concentrations of analysed milk variables between SRB and SLB or between SLB and SDM. Concentration of total protein, total casein, β-casein and κ-casein were significantly higher in SRB milk compared with SLB milk. Concentration of α-lactalbumin and calcium were significantly higher in SDM milk than in SLB milk. The concentration of urea was higher in SLB than in SDM milk and is suggested to reflect differences in feeding regimes between the investigated Swedish and Danish herd.  相似文献

本试验旨在探究母羊出生年度、季节、胎次、泌乳阶段4个固定效应对西农萨能奶山羊产奶性状的影响。以西北农林科技大学萨能羊原种场2006-2018年的645只泌乳母羊为研究对象,每月采集乳样1次,采样日早、晚各采集1次,将两次乳样等比例混合后,采用乳成分分析仪测定乳脂率、乳蛋白率、乳糖率、总脂固形物、非脂固形物、密度、冰点和酸度共8个指标,结合羊场产奶量记录,采用固定效应模型,通过SAS 9.4软件进行表型的描述性统计分析,再采用GLM模型进行固定效应对产奶性状的影响分析。结果表明,西农萨能奶山羊平均300 d产奶量为507.67 kg,乳脂率为3.58%,乳蛋白率为3.20%,乳糖率为4.19%;总固形物含量为12.21%,非脂固形物含量为8.46%;出生年度、胎次对产奶量影响极显著(P<0.01),出生季节对产奶量影响不显著(P>0.05);泌乳阶段、出生年度对乳脂率等8个乳成分性状均有极显著影响(P<0.01),胎次对除乳蛋白率和总固形物的其他6个乳成分指标均存在极显著影响(P<0.01)。综合以上试验结果,胎次及泌乳阶段是影响西农萨能奶山羊产奶性状的两种主要非遗传因素,第3、4胎母羊产奶性能最佳,泌乳早期的乳品质更优,揭示了对种群进行良种选育工作和羊饲养管理的重要性,也为后期进行奶山羊经济性状遗传评估和群体遗传改良提供理论基础。  相似文献

The aim of the study was to determine the changes in composition and physicochemical features (pH, density, thermostability and acidity) of mare colostrum and milk, and of protein fraction contribution (serum albumin, β-casein, γ-casein, α-lactalbumin, G class immunoglobulins) depending on lactation stage. The research material was colostrum and milk samples from 12 Arabian mares. Colostrum samples were collected within 2 h after parturition and milk samples were collected twice, in the 3rd and 6th weeks of lactation. The level of basic milk components decreased significantly (only lactose content increased) as compared to colostrum. Total bacteria count and somatic cell count decreased significantly with an increase in resistance and urea level. The changes observed were connected to differentiated contribution of particular protein fractions and their relative proportions. Lower levels of γ-casein (P ≤ 0.05), β-casein, serum albumin as well as α-lactalbumin were observed in colostrum as compared to those in milk. Any relationship between lactation stage and β-casein content was observed. Serum albumin and α-lactalbumin content increased in subsequent milkings. The level of G class immunoglobulins decreased significantly and its highest level was noted in colostrum. Any significant differences between the 3rd and 6th lactation weeks were obtained.  相似文献

The primary aim of this study was to investigate the quantitative trait loci (QTL) on BTA6 that affect negatively correlated milk traits, using bivariate covariance component analysis of milk yield and fat (or protein) content, protein yield and fat content, and fat yield and protein content. A set of five different genetic models was adapted to differentiate trait‐specific QTL in close linkage from pleiotropy. Using a grand‐daughter design consisting of five half‐sib families from the German Holstein population and 298 sons genotyped for 16 microsatellite markers on BTA6, we found significant trait‐specific QTL for fat content and protein yield, 24 cM apart. Markers BM1329 and FBN12 bracketed the QTL for fat content, and the region between TGLA37 and FBN13 most likely harbours a QTL for protein yield. The analysis based on the close linkage model fully confirmed this result. Despite the pure QTL findings confirming results from the literature, distinguishing pleiotropic and closely linked QTL for competitive traits is a new aspect. Our multivariate analysis results did not suggest a pleiotropic QTL for the investigated negatively correlated traits. The QTL‐based trait correlations were discussed as an important aspect of modelling that needs to be considered in the future.  相似文献

Recent publications indicate genetic variation in milk production traits on proximal BTA14, which cannot be explained solely with genetic variation in the DGAT1 gene. To elucidate these QTL effects, animals from a German Holstein granddaughter design (18 families, 1,291 sons) were genotyped for CYP11B1 (V30A) and DGAT1 (K232A) polymorphisms. Frequencies of alleles of maternal descent were estimated for CYP11B1(V) (0.776) and DGAT1(K) (0.549). Allele substitution effects (alpha/2) were first calculated for both alleles in separate models and then in a joint model. From the joint analysis, CYP11B1(V) effects on fat content (+0.04%) and protein content (+0.01%) were positive. Effects on milk yield (-82 kg), fat yield (-0.5 kg), and protein yield (-1.9 kg) were negative. Compared with the individual analysis, DGAT1(K) effects on fat content (+0.28%), protein content (+0.06%), and milk yield (-258 kg) were reduced; fat yield (+10.8 kg) was enhanced; and protein yield (-3.8 kg) was reduced. In the joint analysis, allele substitution effects of CYP11B1(V) and DGAT1(K) together explained more of the variation in milk production traits than DGAT1(K) alone. Further significant effects were found for CYP11B1(V) and DGAT1(K) among 6 reproduction traits and 14 conformational traits. These observations indicate a possible negative influence of DGAT1(K) on maternal nonreturn rate, and thus, on length of productive life.  相似文献

旨在开展奶牛群体高乳成分功能基因的验证与筛选,利用微流控芯片自主选育技术分析北京地区母牛群体高乳蛋白、高乳脂基因频率,同时分析基因多态性及其与产奶性状的相关性。本研究对北京地区8个大型奶牛场1 596头中国荷斯坦奶牛RPL23A、ACACB基因的多态性进行了检测,所有个体均为3胎以内的泌乳牛,收集每头牛所有测定日的产奶量、乳脂率、乳蛋白率等数据,同时对3个多态位点不同基因型与产奶性状进行了关联分析。RPL23A基因的SNP位点g.20146771G>A,在第一泌乳期,与产奶量、乳脂量、乳脂率和乳蛋白量均达到极显著关联(P<0.01),第二泌乳期,g.20146771G>A与5个产奶性状均呈极显著关联(P<0.01)。ACACB基因的SNP位点g.63962768C>T,在第一泌乳期,与产奶量、乳脂量、乳脂率和乳蛋白量均呈极显著关联(P<0.01),在第二泌乳期,与产奶量、乳脂量、乳脂率和乳蛋白率呈极显著关联(P<0.01)。结果表明,RPL23A、ACACB基因可以作为影响中国荷斯坦奶牛产奶性状的候选基因用于标记辅助选择,以上基因位点可能通过直接或间接的途径影响奶牛的乳脂或乳蛋白性状,对产奶性状起到重要调控作用。本研究为荷斯坦奶牛后续的标记辅助选择奠定了良好基础。  相似文献

本研究选取北京市 4个牛场共 73 1头健康荷斯坦奶牛 ,利用聚丙烯酰胺凝胶垂直板状电泳(PAGE)对其乳蛋白进行分型 ,采用基因替代模型来分析乳蛋白多态性与奶牛产奶性能间的关系。结果表明 :αsl CN座位B基因对产奶量的替代效应最大 ,为 62 4kg ;β CN座位A1基因对A2 基因的替代使乳脂率提高 0 .1 2 % ,但产奶量降低 70kg ;κ CN座位和 β Lg座位A基因对B基因的替代效应分别对乳蛋白率、乳脂率有显著效应 ,使二者分别降低 0 .2 0 %。  相似文献

Leptin gene has been found to be associated with various economic traits including milk production and fat quality in dairy animals. In the present study, we investigated genetic variations in intron 1 region of leptin gene in riverine buffaloes (Bubalus bubalis) using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing methods and associated them with milk traits. The study revealed three SSCP variants A, B and C among a total of 301 buffaloes from nine breeds. The frequency of variant C was found invariably high among all the breeds except in Marathwada buffalo. Variant A was found to be absent in Chilika, Nili-Ravi, Nagpuri and Pandharpuri breeds and also had the lowest frequencies in Mehsana, Jaffarabadi, Murrah and Toda breeds. Sequencing of SSCP variants revealed a total of five polymorphic sites, with three haplotypes. Statistical analysis revealed significantly high fat percentage at 150?days in SSCP variant B in Mehsana buffaloes. However, the associations of SSCP variants of leptin gene with total milk yield, 305?days milk yield and total fat yield were found to be non-significant. The present study is the first report on association analysis of leptin gene polymorphisms with milk production and milk quality traits in river buffalo.  相似文献

Milk performance traits are likely influenced by both additive and non‐additive (e.g. dominance) genetic effects. Genetic variation can be partitioned using genomic information. The objective of this study was to estimate genetic variance components of production and milk component traits (e.g. acetone, fatty acids), which are particularly important for milk processing or which can provide information on the health status of cows. A genomic relationship approach was applied to phenotypic and genetic information of 1295 Holstein cows for estimating additive genetic and dominance variance components. Most of the 17 investigated traits were mainly affected by additive genetic effects, but protein content and casein content also showed a significant contribution of dominance. The ratio of dominance to additive variance was estimated as 0.64 for protein content and 0.56 for casein content. This ratio was highest for SCS (1.36) although dominance was not significant. Dominance effects were negligible in other moderately heritable milk traits.  相似文献

A study was carried out to establish the factors affecting milk, butterfat and protein yields in Zimbabwean Jersey cattle. A total of 10,986 unedited 305-day lactation records were obtained from Zimbabwe Livestock Identification Trust containing 25 herds, with cows calving in the period 1996–2008. The general linear model procedure of the Statistical Analysis Systems version 9.1.3 was used to determine the environmental factors. Herd-year-season, calving interval, days dry and both the linear and quadratic effects of age at calving fitted as covariates significantly (P < 0.0001) affected the milk, fat and protein yields. Herd-year-season accounted for 36%, 36% and 35% to the total variation in milk, fat and protein yields, respectively. Milk, fat and protein yields increased with an increase in calving interval. The optimum dry period for milk, fat and protein yields obtained was 60–75 days dry. The relationship between the production traits and age at calving was both linear and quadratic, implying that milk, fat and protein yields increase with the age of the animal. It is thus necessary to preadjust data for these environmental factors when carrying out genetic evaluations of production traits in dairy cattle.  相似文献

The somatotrophic axis consisting of pituitary-derived growth hormone and circulating insulin-like growth factor 1 has been well established as key regulators of animal health, metabolism, lactation, fertility, body composition and growth rate. The aim of this study was to simultaneously quantify the associations between SNPs in candidate genes of the somatotrophic axis (i.e., IGF-1, GH1 and GHR) with performance traits in Holstein-Friesian (HF) dairy cattle. Both novel SNPs identified previously by this group alongside other published SNPs within these genes were analysed for associations with performance in dairy cattle. Multiple regression analyses regressing genetic merit of up to 848 HF sires on novel SNPs (n = 76) and published SNPs (n = 33) were undertaken using weighted animal mixed linear models. Twenty-three SNPs were significantly associated with at least one of 18 traits analysed and involved in milk production, udder health, fertility and growth. Eight traits including milk fat composition, carcass conformation, stature, chest width, body depth, rump width, carcass and cull cow weight were independently associated with SNPs in two genes. Furthermore, for several traits including milk fat yield, somatic cell count, survival and carcass fat, SNPs in all three genes were independently associated with performance. Milk fat yield and carcass fat showed the highest number of independent associations across all three genes with five SNPs associated with both traits. The cumulative effects of the favourable alleles of all five SNPs across GH1, GHR and IGF-1 result in an increase of 5.9 kg and 28.6 units of milk fat and carcass fat, respectively. Cow survival was associated with a single SNP in each of the three genes with a cumulative allele effect of 1.5%. Independent effects of polymorphisms in GH1, GHR and IGF-1 reinforce the central role of the somatotrophic axis on animal development and performance.  相似文献

为分析GH基因SNPs与中国荷斯坦牛泌乳性状的相关性。以Z32头中国荷斯坦牛为研究材料,采用PCRSSCP技术检测GH基因第5外显子及其相邻区域的多态性,并进行相关性分析。结果发现GH基因第4内舍子(2017bp处)存在C—T的碱基转换,该群体在此位点处于Hardy-Weinberg平衡状态,为中度多态。与泌乳性状的相关性分析表明,该多态位点与泌乳性状显著相关(P〈0.05),TT型305d产奶量、305d乳脂量、305d乳蛋白量和305d乳糖量显著高于CC型(P〈0.05);TC型与TT型、CC型之间305d产奶量、305d乳脂量、305d乳蛋白量差异均不显著(P〉0.05),但TC型在数值上有优于CC型的趋势。建议将等位基因T作为提高奶牛产奶量、乳脂量和乳蛋白量的候选分子标记。  相似文献
