Summary Genotypic differences for growth stage dependent expression of partial resistance to barley powdery mildew have been identified on the basis of two components of resistance in the glasshouse and the pattern of epidemic development in the field.Differences for infection frequency and proportion of sporulating colonies were highly significant between the 16 genotypes investigated at four stages of plant development in the glasshouse. Both resistance components were significantly correlated mutually (r=0.73 to 0.86) and with the infection level on leaves developed at comparable growth stages in the field (r=0.52 to 0.73). The infection level of seedlings in the glasshouse was significantly correlated with the infection level of the leaves of the first until the third node in the field (r=0.70 to 0.73). Adult plant resistance was predominantly expressed at the uppermost leaf. Summary Ten rice genotypes were inoculated with a virulent isolate of Magnaporthe grisea to study the effect of leaf age on components of partial resistance, and evaluate associations between these components. The number of sporulating lesions per cm2 leaf declined with increase of leaf age in all genotypes. The number of lesions per cm2 leaf area in one week old leaves was about 25% that of the number in very young leaves in the susceptible cultivar CO39, but less than 2% in the more resistant cultivars IR36, IR60 and IR62. Large differences between genotypes were found for the number of sporulating lesions that developed, and this factor was closely related to the period that leaves remained susceptible after appearance. The number of lesions in the most susceptible cultivar CO39 was about 7 times that in the cultivars IR60 and IR64. Differences between genotypes were also found for lesion size. The effect of aging on average lesion size was less pronounced than on lesion density. Lesion size and lesion density were positively correlated, but a rapid decline of density was not necessarily accompanied by a rapid decline of size. No apparent differences between genotypes were observed for latent period. Genotypes with leaves that became highly resistant soon after appearance expressed higher levels of partial resistance in the field.  相似文献

Summary Eight spring barley cultivars, respresenting the known range in partial or slow rusting resistance to leaf rust, Puccinia hordei, were investigated for their effects on the components of partial resistance; infection frequency, latent period, infectious period and spore production per uredosorus per day. Considerable variation was observed among the cultivars for each of the components. The cultivar effects on the components tend to be associated. Cultivar L94 for instance, shows the highest infection frequency, the shortest latent period and a long infectious period. Julia and Vada both have a low infection frequency, a long latent period and a low spore production per sorus per day. This association, though, is only a partial one.The total spore production per unit leaf area (the combined result of the four components) appeared highly correlated with the partial resistance in the field (r=0.85). Only a relatively small portion of the variation in partial resistance cannot be explained by the four components studied. Several other aspects, which might affect the rate of epidemic development, are discussed.Latent period, measuring the onset of the new spore production, estimated partial resistance as well as total spore production did (r=–0.85). In order to evaluate the partial resistance of barley genotypes in the greenhouse the latent period is preferred above total spore production as it is measured more easily, more accurately and sooner after inoculation.  相似文献

Information about slow rusting resistance to leaf rust (Puccinia triticina) in durum wheat (Triticum turgidum var. durum) is limited. Three slow rusting components, latent period, receptivity, and uredinium size, were determined at the adult plant stage for seven durums with slow rusting resistance to leaf rust and two susceptible durums in three greenhouse experiments. Additionally, area under the disease progress curve (AUDPC) and final disease severity (FDS) were determined in three field trials under artificial epidemics with the same P. triticina race BBG/BN. Compared to the most susceptible check, the AUDPC and FDS of slow rusting resistant durums were significantly lower and ranged from 13–47 to 22–59%, respectively. The latent period was significantly longer (8.5–10.3 days) and uredinium size significantly smaller (8.1–14.8 × 10−2 mm2) on slow rusting durums than on the susceptible checks (8.0 days and 17.3–23.8 × 10−2 mm2, respectively). Uredinium size was the most stable slow rusting component across experiments. Correlations between uredinium size versus AUDPC and uredinium size versus FDS for each environment were high (r = 0.86–0.88). Correlations between latent period and field parameters were significant (r = −0.60 to −0.80). Correlations between receptivity and the field parameters were not significant. A multiple regression analysis showed that the variation in AUDPC and FDS was significantly explained only by uredinium size (P < 0.0001). The best slow rusting resistant lines can be used for developing high-yielding durums with more durable resistance to leaf rust.  相似文献

Summary Lesion size and lesion number were measured on cultivars of rice inoculated by clipping or spraying with virulent isolates of Xanthomonas campestris pv. oryzae. Mean percentage diseased leaf area (%DLA) gave a similar ranking for the two inoculation methods but differences in lesion size among cultivars were much more evident after clip than after spray inoculation. Correlation between the methods was high (r=0.82**), but some cultivars responded differently with the two techniques. Cultivars which had low scores following spray inoculation showed low disease progress during the first nine weeks after transplanting into a screen-house experiment. Assessment after clip inoculation measures resistance due to spread of bacteria within the leaf xylem system, an important component of quantitative resistance. Assessment after spray inoculation measures all resistance, including resistance to entrance of bacteria into the leaf. In order to select rice entries with improved quantitative resistance to X. c. oryzae based on both components, a screening based on lesion length after clip inoculation, followed by a test for lesion number after spray inoculation, is advised.  相似文献

Summary Breeding for disease resistance in peppers (Capsicum spp.) to the bacterial spot pathogen (Xanthomonas campestris pv. vesicatoria (Doidge) Dye) has been based on either qualitative or quantitative evaluation methods. Quantitative evaluation of components of resistance, lesion number and lesion diameter, has been useful for determining quantitative resistance, but few breeders have applied these methods in routine selection programs. This study was aimed at determining the heritability and gain from selection for resistance to the bacterial spot pathogen based on three components of resistance. Random selections from a diverse intermated population of Capsicum annuum L. were self-pollinated for two generations to create S1 and S2 families. Thirty S1 families, corresponding S2 families and four homozygous check lines were evaluated. At forty-two days after seeding, two different leaves of each plant were inoculated by leaf infiltration with low concentrations (5×103 colony forming units) of Group 2 (XCV PT, race 1) and 4 (XCV P, race 1) of the bacterium, respectively. After 15 days, lesion number cm-2 and lesion diameter were measured. Total lesion area was calculated. Narrow-sense heritabilities for lesion number, lesion diameter, and total lesion area were 0, 43, and 31%, respectively, with Group 2, and 26, 43, and 33%, respectively, with Group 4. Actual S2 gain from 20% selection pressure in the S1 was approximately 50%, when selection towards resistance was based on total lesion area.  相似文献

Screening of 136 faba bean lines forresistance to chocolate spot caused by thefungus Botrytis fabae was conductedin field conditions with artificialinoculation. Detached leaves of thesegenotypes were also assessed for diseaseseverity after inoculation under controlledconditions with a B. fabaesuspension. The BPL710 and Aguadulcegenotypes were inserted as resistant andsusceptible checks, respectively. Diseasesymptoms were scored visually in bothexperiments. Diameter of lesions andproduction of spores were measured in thedetached leaf assay. The disease scoreindex and diameter of lesions were used tocalculate the area under the diseaseprogress curve (AUDPC) to measure thedisease progress. Significant differenceswere detected among genotypes for reactionto the disease in the field. However, nocomplete resistance was observed. Forhighly susceptible and highly resistantgenotypes, the laboratory assay generallyled to the same result as the field test,nevertheless, for more intermediate levelsthe agreement was weak. On detached leaves,diameter of lesions was significantlycorrelated to the AUDPC of disease scores(r = 0.89) and moderately correlated tosporulation (r = 0.52). The correlationbetween field AUDPC and detached leaf AUDPCof disease scores was low (r = 0.26). A lowcorrelation was also observed between fieldAUDPC and lesion size (r = 0.30) andbetween field AUDPC and sporulation (r =0.32). Among the 136 genotypes evaluated inthis study, nine were convincingly andhighly resistant in both tests. FRYM167 andFRYA58 genotypes were the most resistantwith low AUDPC of disease scores, low AUDPCof lesion diameter and low sporeproduction. These genotypes obviously havepartial resistance.  相似文献

Over 540 accessions of wild Lycopersicon species or their crosses with L. esculentum were screened for resistance in a series of trials. Forty-six accessions were selected for the final screening trial based on lower disease ratings in previous trials. Of these, L. hirsutum had the greatest number of resistant accessions, followed by L. esculentum and L. peruvianum. Twenty accessions were quantified for their levels of resistance based on leaf area infected, area under disease progress curve (AUDPC), and the degree of sporulation. There was a significant positive correlation between the AUDPC calculated from 20 accessions evaluated under growth room and field conditions. Five L. hirsutum accessions had no sporulation associated with leaf lesions, whereas L. esculentum accessions had an average of 1.6×104 conidia/cm2 of leaf tissue. There was significant positive correlation between the AUDPC values and the number of conidia per cm2 of leaf tissue.  相似文献

This experiment was carried out to investigate whether and how much field resistance to late blight, caused by Phytophthora infestans, is present in the local cultivated potato germplasm. In total 36 entries were compared in a field experiment in an area highly conducive to late blight development. Of the 36 cultivars 32 were local cultivars belonging to five Solanum species, S. tuberosum (1 accession), S. andigena (18), S. juzepczukii (2), S. stenotomum (9) and S. ajanhuiri (2). The other four cultivars were derived from breeding programmes, one being the Dutch cultivar Alpha used as a highly susceptible control. The 36 cultivars were planted according to a simple 6 × 6 lattice design with three replicates. Each replicate was divided in six incomplete blocks each with six cultivars. The disease severity was assessed weekly during 9 weeks starting 48 days after planting. The area under the disease progress curve (AUDPC) was used as a measure of the field resistance. Nine isolates from surrounding potato fields were tested for their virulence to the resistance genes R1–R11 using 22 differential cultivars. The components of the field resistance of 19 of these cultivars were compared in the greenhouse using a local isolate with virulence to all known R-genes, except to R9. The nine isolates represented seven races with a race complexity varying from 7 to 10 virulence factors. All isolates carried virulence against R1, R2, R3, R7, R10 and R11, while virulence against R9 was absent. The AUDPC among the 32 local cultivars ranged from very large, significantly larger than that of ‘Alpha’ to very small. The AUDPC from S. stenotomum accessions ranged from very large to intermediate, those from S. andigena accessions from large to very small. Especially among the S. andigena accessions interesting levels of field resistance were found. Four components of field resistance were assessed, latency period (LP), lesion size (LS), lesion growth rate (LGR) and relative sporulation area (RSA). All four showed a considerable variation among the cultivars. The LP ranged from 3½ to 6 days. The LS ranged from 225 mm2 to 20 mm2. The LGR varied about six-fold, the RSA more than 10-fold. The components tended to vary in association with one another. LP and LGR were well associated with each other and had a significant correlation with the AUDPC.  相似文献

Summary Experiments to assess variation in the resistance of winter wheat to infection by Microdochium nivale were conducted over two consecutive years. Resistance was evaluated using an agar disk technique to reproduce stem lesions and by spraying a conidial suspension to reproduce head blight symptoms. Significant variation for stem reaction measured as stem lesion area (SLA), and head reaction measured as disease severity (DS) was found in the 33 winter wheat genotypes tested. Data obtained over two years in controlled environment conditions were significantly correlated (r=0.713 for SLA and r=0.738 for DS), whereas field data showed a significant genotype x year interaction for disease severity. Quantitative variation for susceptibility to stem and head infection by M. nivale was found among the 33 genotypes tested. The majority of genotypes expressed moderate susceptibility, with cultivar Goupil being very susceptible to both stem and head infection, and the remainder, Renan, Arminda, Munstertaler and Saint-Johann were the most resistant. Resistance to stem and head to M. nivale were not correlated (r=0.358).  相似文献

Summary Resistance to gummy stem blight (Didymella bryoniae) was not detected among 1208 cucumber lines of diverse origin when cotyledons of four-day-old seedlings were inoculated with an aqueous suspension of 5000 spores of D. bryoniae delivered to freshly crushed tissue, followed by 48 h incubation at 20°C and 100% RH in the dark. Among 49 lots evaluated for gummy stem blight in the field, cv. Homegreen #2 and Pl 200818 from Burma were resistant. Earliness in fruit maturity was positively correlated with susceptibility to gummy stem blight (r=0.424). Cucumber accessions differed in the incidence of stem and hypocotyl cankers. In greenhouse grown plants at the 2nd–3rd leaf stage, differentiation of resistant lines was obtained by spraying plants with an aqueous suspension of 106/ml D. bryoniae spores followed by 48 h incubation at 20°C and 100% RH in the dark. One cycle of mass selection in Homegreen #2 resulted in improved gummy stem blight resistance.  相似文献

The objective of this study was to identify polymorphic molecular markers associated with partial resistance to coffee leaf rust, Hemileia vastarix. A segregating F 2 population derived from a cross between the susceptible Coffea arabica cv. Caturra and a C. canephora-introgressed Arabica line exhibiting high partial resistance was analyzed. Rust resistance measured as rust incidence (RI) and defoliation (DEF) was evaluated in field conditions in three consecutive years (2003–2005). During the 2003 season, which was characterized by favorable conditions for a rust epidemic, the F 2 plants exhibited different levels of resistance ranging from very susceptible (50.1% for DEF and 49.5% for RI) to highly partial resistance (9.1% for DEF and 3.7% for RI). Molecular analysis enabled identification of seven polymorphic markers (5 AFLP and 2 SSR) exhibiting significant association with partial resistance. Coexistence of resistance homozygous alleles (RR) at codominant SSR loci was correlated with high resistance. This study is the first attempt to develop PCR-based sequence specific markers linked to partial rust resistance in coffee.  相似文献

Summary The partial resistance to leaf rust (Puccinia hordei) of 40 West-European spring barley cultivars was measured in plots isolated from one another to reduce inter plot interference. The leaf area affected by leaf rust was also measured in small plots of 0.5 m2 adjacent to each other, and on individual plants. The latent period was measured in the seedling stage and the adult plant stage, the infection frequency in the seedling stage only. The cultivars varied widely for partial resistance, many cultivars carrying a considerable level. Both the small adjacent plots and the single plants showed a marked inter plot interference strongly reducing the difference between cultivars. H wever, the ranking order of the cultivars was hardly, if at all, affected. Both latent period and the infection frequency showed large differences between cultivars, the latent period in the adult plant stage being highly correlated (r=0.82) with partial resistance, infection frequency in the seedling stage only rather weakly (r=–0.33).Selection for partial resistance appeared very effective in all stages tested; the seedling, the single adult plant, and the small plot stage. Selection in the small plot stage was the most effective followed by selection in the seedling stage. Selection for partial resistance therefore appears very well possible at all stages of the selection program.  相似文献

Summary Phenotypic stability of bacterial leaf spot resistance in peach (Prunus persica (L.) Batsch) regenerants, either selected at the cellular level for insensitivity to a toxic culture filtrate of Xanthomonas campestris pv. pruni or screened at the whole plant level for resistance to X. campestris pv. pruni, was investigated. A detached-leaf bioassay was used to evaluate the original regenerants again after three years in the greenhouse and also after a two to three year cycle of tissue culture propagation. Peach trees derived through micropropagation from the original regenerants were also evaluated after one to three years growth in the field. Although leaf spot resistance was retained in some regenerants over time in the greenhouse, following in vitro propagation, and under field conditions, resistance was either lost or not expressed in others. Regenerants # 19-1 and #156-6, derived from embryo callus of bacterial spot susceptible Sunhigh, were significantly more resistant than Sunhigh. High levels of resistance were exhibited in greenhouse plants and field-grown trees of regenerant #122-1, derived from embryo callus of moderately resistant Redhaven. This research provides additional evidence that selecting or screening for somaclonal variants with disease resistance is a feasible approach to obtaining peach trees with increased levels of bacterial spot resistance.Abbreviations TC Tissue-Cultured - TF Toxic culture Filtrate  相似文献

Late leaf spot (LLS) and rust cause substantial yield losses and reduce the fodder and seed quality in groundnut (Arachis hypogaea L.). Adoption of resistant cultivars by the semi-arid tropic farmers is the best option to overcome yield losses. Knowledge on components of resistance to these diseases should facilitate the development of groundnut cultivars with enhanced resistance to LLS and rust. The objectives of the experiments were to study the genetic variability and relationships among components of resistance to LLS and rust, and assess their significance in disease resistance breeding. Fifteen interspecific derivatives for LLS and 14 for rust and a susceptible control, TMV 2, were evaluated in a randomised complete block design with two or three replications under greenhouse conditions. The experiments were repeated twice. Genotypic differences were highly significant for all the traits studied. Resistance to LLS is due to longer incubation and latent periods, lesser lesions per leaf, smaller lesion diameter, lower sporulation index, and lesser leaf area damage and disease score. Selection based on components of resistance to LLS may not lead to plants with higher retained green leaf area. The remaining green leaf area on the plant should, therefore, be the major selection criteria for resistance to LLS in breeding programs. Resistance to rust is due to longer incubation and latent periods, fewer pustules per leaf, smaller pustule diameter, lower sporulation index, and lesser leaf area damage and disease score. Rust resistant components appear to work additively, therefore, selection based on resistance components together with green leaf area retained on the plant should be the basis of selecting for resistance to rust in breeding programs. ICGV 99005, 99003, 99012, and 99015 for rust and ICGV 99006, 99013, 99004, 99003, and 99001 for LLS are the better parents for use in resistance breeding programs. Summary Breeding of Phaseolus vulgaris L. for resistance to common bacterial blight (CBB) can be done with visual evaluations of symptoms to distinguish broad resistance classes, but a more quantitative measure was needed for genetic studies of resistance. A novel method of evaluation was developed by quantifying Xanthomonas campestris pv. phaseoli (XCP) in bean leaf tissue infected with CBB using a 32P-labeled probe and densitometric analysis of hybridization signals. Quantification of bacterial populations using the probe was highly correlated (r=0.98) with the number of colony forming units (CFU) from plate counts of the same leaf samples. The probe was used to follow XCP population dynamics on susceptible (BAT 41) and resistant (OAC 88-1) bean genotypes. OAC 88-1 supported a maximum XCP population which was approximately tenfold less than BAT 41. The probe was also used to study an F2/F3 population segregating for resistance. Narrow sense heritability estimates were less for resistance measured on the basis of bacterial populations (0.18–0.26) than on visual scores of symptoms (0.29–0.38). The anticipated response to selection for CBB resistance would be less based on bacterial numbers than based on symptom expression in this population. Summary F2-plant progenies, derived from seven crosses between susceptible erectoides breeding lines and conventional, nutans breeding lines with partial resistance to leaf rust incited by Puccinia hordei Otth., derived from Vada x Cebada Capa, were examined in a field and a glasshouse experiment. In the field experiment, the nutans plant progenies generally were more resistant to leaf rust and powdery mildew than the erectoides progenies. In the glasshouse experiment, the components of partial resistance to leaf rust of six erectoides and six nutans progenies derived from one cross were studied. The nutans progenies generally had lower infection frequencies than erectoides progenies with a similar infection level in the field experiment. The variation for leaf rust and powdery mildew infection in the field was similar for the nutans and erectoides progenies. This suggests that erectoides lines with a fairly high level of partial resistance to leaf rust and powdery mildew can be selected from these populations.  相似文献

Summary Winter wheat genotypes were tested for resistance in the field by assessing the percentage sporulating leaf area after infection with wheat leaf rust. The disease level in the first field trial was too low for selection. In the second field trial a low sporulating leaf area was found on several genotypes showing a susceptible infection type. These genotypes possibly have partial resistance. Six genotypes possibly possess adult plant resistance, as they showed a resistant infection type and a low sporulating leaf area.The latency period, infection frequency and uredosorus size of sixteen genotypes were determined in the greenhouse after infection with two races of leaf rust at two temperature regimes. The temperature × genotype interaction, found for latency period and infection frequency, was mostly influenced by the cultivars Cerco, Tundra and Miller. Adult plant resistance was postulated for four genotypes whereas another four appeared to have partial resistance.Only one of the sixteen genotypes (Apexal) possessed adult plant resistance and two genotypes (Arminda and Cappelle Desprez) showed partial resistance in the field as well as in the greenhouse.  相似文献

Mist-chamber, field, and detached leaf inoculation procedures identified plants resistant to bacterial soft rot [Erwinia carotovora subsp. carotovora, (Ecc)] in Brassica rapa and related species. The mist-chamber seedling inoculation provided the best correlation of mean disease severity ratings with the field plant inoculation (r = 0.67**) and was used to identify resistant materials. The optimum mist-chamber incubation conditions to distinguish the resistance of accessions were 23 °C and 100% RH and were used for primary screening. A total of 752 accessions of B. rapa and related Cruciferae were screened. In general, accessions of B. oleracea were more resistant than accessions of B. rapa. Within B. rapa, subspecies pekinensis and chinensis were more susceptible than other subspecies. No completely resistant material was found. In species of B. rapa, only 7% of accessions showed some degree of resistance with plant-to-plant variation within the accessions. G30444, G30449, and AVRDC2837 were identified as the most resistant materials in B. rapa by both mist-chamber and field inoculations. The resistance was correlated between an USA isolate (Geneva-1) and two Chinese isolates (RL4-1 and RL-19) of Ecc. Summary Lesion length, leaf length, and leaf width were measured on infected leaves two weeks after clip inoculation of 64 rice cultivars with two virulent isolates of Xanthomonas campestris pv. oryzae (X.c. pv. oryzae). No significant correlation was found between the lesion length and the leaf dimensions, indicating that physical leaf size does not affect the spread of the bacteria once these have entered the leaf. Lesion length is therefore an acceptable parameter for assessing resistance to (X.c. pv. oryzae), and is to be preferred above the parameter percentage diseased leaf area (% DLA), especially when small differences between genotypes are to be assessed. The confounding influence of differences in leaf length can cause large changes in the ranking order of cultivars when assessed by the % DLA. For this reason lesion length is a better assessor of the value of a quantitative resistance for breeding and research purposes than % DLA.  相似文献
