Tree peony is well known and sought after for its large, colorful flowers. Its propagation is via vegetative methods. Mech-nisms of the adventitious rooting and the regulation of rooting processes are the principles and techniques of plant propagation and improvement. Microstructures and fluctuations of phytohormones in the adventitious rooting were studied with the etiolated softwood shoots of Paeonia suffruticosa 'Yinfen Jinlin'. There are no pre-primordia in the shoots of the cultivar. Adventitious roots are produced in five stages: shoot selection, primordium initiation, primordium growth, conducting tissue differentiation and root protrusion. Primordia initiated in the cortex. The contents of the endogenous hormones, IAA, ABA and GA, were 5.842, 0.873 and 1.043 nmol·g-1 FW on the bases of shoots, respectively. CTKs which included isopentenyl adenine (iPA), zeatin riboside (ZR) and dihy-drozeatin riboside (DHZR) were 0.949, 0.695 and 2.034 nmol·g-1 FW, respectively. DHZR is active among CTKs. The ratio of IAA to GA, CTK and ABA clearly increased at the stage of primordium initiation, while they showed low levels at the stages of primordium growth. The ratios were restored at the shoot levels at the stage of root protrusion. IBA provoked primordia initiation in the cortex, the vascular cambium, the pith and even in the callus induced on the base of shoots. IAA levels in the treated shoots increased gradually to its highest level (three times of control) at the stage of conducting tissue differentiation. The ratios of IAA to GA, CTK and ABA clearly decreased at the stage of primordium initiation. The ratio of IAA to ABA is regulated at 10:1.  相似文献

‘Zhongqiusucui’ jujube secondary shoots were treated with 3-indolebutyric acid(IBA) at three concentrations, 500, 1000 and 1500 mg/L. Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root growth.Endogenous hormones in leaf tips and bases had an impact on rooting. The activities of endogenous hormones mainly existed in leaf tips because they play a major role in the production and consumption of IAA and its ABA content increased during rooting. The ZR in leaf tips influenced the rooting of cuttings, especially in the callus formation and rooting stage. Leaf tips were the main source of GA.  相似文献

Robinia pseudoacacia ‘Idaho’ is one of several multi-purpose trees used in ornamental, soil and water conservation, fodder and nectar sources. Plant abiotic stress tolerance transformed by genes could meet the requirements for reclamation of arid or alkalid lands and vegetation restoration. For this paper, we studied the effects of auxin and cytokine on Idaho locust in vitro regeneration and the establishment of gene transformation systems for plants mediated by Agrobacterium tumefaciens. Results showed that the ratios of cytokinin and auxin were the major factors affecting adventitious bud differentiation on a MS medium; the concentration of 0.5 mg•L–1 6-BA benefitted callus proliferation and 0.25 mg•L–1 IBA promoted shoot rooting; however, a higher IBA concentration will inhibit rooting. The most effective antitoxin for screening transgenic Idaho locust shoots was G418 and the most sensitive concentra-tion of it was 8 mg•L–1.  相似文献

The objectives of this study were to investigate the effects of successive continuous generation (SCG) cuttings of Buxus sinica var. parvifolia on the rejuvenation of ortets at the hormone level, detect levels of indoleacetic acid (IAA), abscisic acid (ABA), isopentenyladenosine (iPA), zeatin riboside (ZR) and gibberellin4 (GA4) during the rooting process of different generations by means of enzyme-linked immunosorbent assay (ELISA) technology and statistically analyze the rooting characteristics of cuttings. The results show that, the root systems of subsequent generations (generation 1998, 2000) developed satisfactorily, only a few initial gen-eration (generation 1990, 1994) cuttings rooted and the root growth was inhibited. Around the period of callus formation and root projection through the epidermis, there was a marked, regular variation in the levels of IAA, ABA and GA4 between the initial gen-erations and subsequent generations.  相似文献

The differentiation process including somatic embryogenesis in different Ginkgo explants in vitro culture were studied by cytological observation.The results are as follows:1) two complete cotyledons and a embryo bud were observed in mature embryos and several secretory acavitives appeared in maturation region of embryo buds,hypocotyls,cotyledons and radicles after culturing 20 days;two incomplete cotyledons and a embryo bud primordia were found in large cotyledon embryos.The proembryo of two cells,four cells, multi-cellular,and globular embroy were developed from the callus of the small cotyledon embryos.2) The differentiation of cotyledon explants started from epidermal cells,and gradually formed meristematic cell mass in the cortical cells,and eventually adventitious buds were observed.3) The adventitious roots of Ginkgo originated in the cells at the cross of vascular cambium and vascular rays. 4) The type of rooting belongs to induction type by root primordium.The formed adventitious roots were observed after 20 days.  相似文献

In this study a reliable protocol was developed for the establishment of commercial in vitro cultures of Tripterygium wilfordii Hook f.. Juvenile shoots from one-year-old elite plants were used as the source of explants. New axillary shoots were obtained after 30 days of culture on a MS medium supplemented with BAP (2.0 mg·L–1) and NAA (0.1 mg·L–1). The optimal multiplication medium was a modified MS medium supplemented with BAP (1.0 mg·L–1) and NAA (0.1 mg·L–1). This yielded a multiplication rate of 2.4 for each subculture. Slightly more than 92% of shoots rooted when cultured on a modified MS medium containing IBA (0.2 mg·L–1) and acti-vated charcoal (0.5 mg·L–1). Activated charcoal promoted both a strong and a high rooting rate during the rooting phase. Plantlets were transferred to pots for a short acclimatization stage in a greenhouse where 95% of the plantlets survived. This highly reproduci-ble procedure can be adopted for large-scale propagation of T. wilfordii.  相似文献

Eucalyptus benthamii is a forest species of economic interest that has difficulty with seed production and also is considered to have difficulty with adventitious rooting using propagation techniques,such as cutting or mini-cutting.We aimed to assess the adventitious rooting percentage under different storage times in low temperatures and at various IBA(indole-3-butyric acid) concentrations to determine the optimal time of permanence for rooting Eucalyptus benthamii minicuttings in a greenhouse.Shoots collected from mini-stumps cultivated in a semi-hydroponic system were used to obtain the mini-cuttings.For the first experiment,the mini-cuttings were stored at 4℃ for 0(immediate planting),24,48,72,96 and 120 h.The second experiment evaluated the rooting dynamic to determine the optimal time of permanence for minicuttings in a greenhouse.The basal region of the mini-cutting was treated with various IBA solutions:0(free of IBA),1,000,2,000,3,000 and 4,000 mg·L-1.Every seven days(0(immediate planting),7,14,21 and 28 days),destructive sampling of the mini-cuttings was performed to evaluate the histology of the adventitious rooting.Eucalyptus benthamii minicuttings should be rooted immediately after the collection of the shoots.The 2,000 mg·L-1 IBA concentration induced a greater speed and percentage of adventitious rooting,and an interval of 35 to 42 days was indicated for permanence of the mini-cuttings in the greenhouse.Exposure to low temperature induced adventitious root formation with diffuse vascular connections.  相似文献

The tissue-cultured seedlings of tree peony ‘Wulong Pengsheng’ were used to study the effects of different plant growth regulators, culture methods, and holdfast on rooting. The morphological structure change during rooting was also observed using the method of paraffin section. The result showed that the best combination of plant growth regulators for rooting was IBA 3.0 mg·L-1 + NAA 0.6 mg·L-1. The treatment under the temperature of 4℃ for ten days was benefit to rooting, and the rate could reach 75.67%. It was identified that the adventitious root primordia of shoot in vitro originated from the vascular cambium cells, especially, the cross areas of cambium and pith ray and they started to differentiate at the 5th day and lasted to the 12th day. If the shoots were cultured in the root inducing medium for 12 days, it would lead to not only descend of rooting rate, but also showing callus of stem base, and leaf senescent. However, if they were transferred into the medium without hormone in time, the root primordial protruded the epidermis and developed normally after 5 days’ culture.  相似文献

The calli formation and organ regeneration from different explants in vitro of Betula platyphylla were discribed.The calli of stem and petiole mainly originated from cortex parenchymatous cells in wound,and that of leaf produced from epidermis and the parenchymatous cells under it .The meristematic nodules were formed at high frenquency in developing callus.They further developed into vascular nodules which could grow at single direction and form adventitious roots.The adventitious buds occurred in the meristematic cell mass in the surface area of calli where the suberized periderm was lacking.In axillary bud proliferation way,adventitious buds originate from the epidermis and underepidermis of stem in axillary and nearby areas.The induced adventitious roots were mainly initiated from the ray cell near the cambium.It seemed that callus inhibited rooting.  相似文献

Four antibiotics, kanamycin, geneticin, carbenicillin and cefotaxime,were evaluated for their effect on the regeneration of adventitious buds, shoot differentiation, rooting from regenerated shoots of Populus euphratica as well as on their control of Agrobacterium -mediated transformations. Results showed that the optimum concentration ranges of kanamycin and geneticin were 15-20 and 10-15 mg·L-1 at the stage of transgenic plantlet selection. The inhibitory effects of cefotaxime and carbenicillin varied among different genotypes of Agrobacterium. The inhibition of cefotaxime on Agrobacterium C58 was stronger than that of carbenicillin. The optimum concentration of cefotaxime was 100 mg·L-1. Cefotaxime and carbenicillin had similar effects on Agrobacterium LBA4404 and their optimum concentrations were both 150 mg·L-1.  相似文献

The effect of auxins (IAA or IBA at 100,200,500 mg L 1) on rooting and sprouting parameters differed significantly (p < 0.05).Rooting and sprouting percentages were higher in some treatments while other parameters were significantly higher in other treatments.The results of the present study revealed that rooting was significantly (p < 0.05) affected by the length of cuttings,as well as the position of shoots and type of cuttings.On average,86.0 per cent rooting was observed in the 30-cm long cuttings compared to the other two lengths of cuttings,i.e.,20 and 25 cm,irrespective of any auxin treatment.In a control set,without any auxin treatment,a maximum of 60.0% rooting was recorded in the 30-cm long cuttings and a minimum of 40.0% of the cuttings rooted in the 20-cm long cuttings.Similarly,cuttings collected from the lower and upper portions of shoots resulted in 72.5 and 52.5 per cent rooting respectively,irrespective of auxin treatments.Maximum rooting was observed in the lower portion (40.0%) of cutting compared to the upper portion (30.0%) in the control set without auxin treatment.Leafless branches produced maximum (72.0%) rooting compared to the leafy cuttings (35.0%),irrespective of the auxin treatment.The leafy cuttings produces a minimum of 15.0% sprouting and rooting in the control set.  相似文献

Nothofagus leoni has a restricted distribution in Chilean forests. This work determines suitable culture conditions forin vitro multiplication and rooting through shoots obtained from seedlings. Broadleaved Tree Medium was suitable for shoot multiplication. A medium with a pulse of 0.55 μM BA in the first subculture and two subcultures on BA-free medium resulted in a multiplication rate at day 63 of × 5.7, without callus growth or shoot neoformation. Rooted shoots of good quality (number and length of roots without callus growth) were obtained with 1.23 μM IBA (91.4% of rooting). The first roots appeared at day 11, reaching a higher speed of rooting at day 15.  相似文献

A rapid and efficient method for the regeneration of plantlets from root explants ofRobinia pseudoacacia L. by suspension culture was established. The roots taken from aseptically grown 15-day-old seedlings were used as explants. It was determined that photoperiodicity was necessary for root proliferation, and that the promotive effect of IAA (3-indoleacetic acid) on root proliferation was better than that of IBA (3-indolebutyric acid). The roots cultured in 1/2 MS liquid medium containing 3 μM IAA and 1% sucrose at 25°C under 16-hour photoperiod with 50 μmol m−2s−1 PPFD (photosynthetic photon flux density) shaking at 100 times/min reciprocally showed high efficiency for root proliferation. BAP (6-benzylaminopurine) was found to be essential to induce adventitious shoots from the roots, and the roots cultured in the medium supplied with 3 μM BAP combined with 1–6 μM IAA for 3 weeks under the same conditions as in the root proliferation period were most suitable for adventitious shoot inducement.  相似文献

Cottonhead windhairdaisy (Saussurea laniceps Hand.-Mazz.) is one of the most famous and important medicinal herbs in China. Illegal collection from wild populations is increasingly threatening the present environment of S. laniceps. Establishment of an efficient method for micropropagation is the best way to change its endangered situation. When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supplemented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg&#8226;L–1 BA plus 0.2 mg&#8226;L-1 NAA. In the presence of 0.2 mg&#8226;L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization.  相似文献

Hybrid combinations of Eucalyptus have increased due to expansion of plantations into unconventional areas and to the search for higher quality timber.However,most of these species have difficulties surviving in vitro cultivation.Active chlorine and sealing systems are often used to reduce contamination and increase gas exchange.The aim of the present study is to evaluate the establishment,multiplication,elongation and adventitious rooting of E.grandis × E.urophylla.Two clones(C1 and C2) and four active chlorine concentrations(0.000%,0.001%,0.003%,and 0.005%) were tested in the establishment and multiplication phases.Three sealing forms(W/M,1/M and 3/M) and the same four active chlorine concentrations were applied to the elongation phase.Two luminosities(dark and light)and three sealings(W/M,1/M and 3/M) were tested during adventitious rooting.Active chlorine concentration of0.005% led to the lowest fungal contamination rate and to the highest in vitro establishment.Active chlorine concentration of 0.003% resulted in the greatest length and highest number of shoots per explant in the multiplication phase.There were no phytotoxicity problems and the quality of plants grown in an environment with active chlorine was maintained in comparison with those grown in an autoclave.The increase in gas exchange in ventilation systems had a positive impact on the in vitro growth and development of plants.  相似文献

Three antibiotics ampicillin, carbenicillin, and cefotaxime were evaluated for their effects on induction, growth, and differentiation of organogenic calli, as well as rooting of regenerated shoots of three Ioblolly pine (Pinus taeda L.) genotypes. Of the antibiotics administered, cefotaxime maximally increased the frequency of callus formation and growth rate of organogeni ccalli, carbenicillin maximally increased the frequency of shoot regeneration and the average number of adventitious shoots per piece of organogenic callus, ampicUlin maximally decreased the rooting frequency of regenerated shoots and mean number of roots per regenerated shoot, in comparison with antibiotic-free media. Compared with the control, ampicillin minimally increased the frequency of callus formation, cefotaxime minimally increased the frequency of shoot regeneration, and carbenicillin minimally decreased the rooting frequency of regenerated shoots in three Ioblolly pine genotypes tested. All three antibiotics increased the frequencies of callus formation and shoot regeneration, and reduced the rooting frequency ot regenerated shoots suggested that the establishment of an efficient Agrobacterium tumefaciens-mediated transformation protocol for stable integration of foreign genes into Ioblolly pine need to select a suitable antibiotic. This investigation could be useful for optimizing genetic transformation of conifers.  相似文献

Softwood shoots were produced from 40 cm long stem segments placed horizontally in flat trays containing sterilized sand under natural light or shade conditions for subsequent rooting and micropropagation studies in teak (Tectona grandis L.). Higher number of shoots (6.17) per log was produced under natural light as compared to shade conditions. Forcing was also better in natural light as compared to shade in terms of shoot length, number of nodes or leaves. For rooting, 2–4 cm long softwood shoots were excised and treated with either indole-3-butyric acid (IBA) or α-naphthyl acetic acid (NAA) at 0, 1000, 2000 or 3000 μmol·L–1 each or with combinations (1000 + 1000, 2000 + 2000 or 3000 + 3000 μmol·L–1) and then placed in flat trays containing autoclaved sand at 25 ± 2ºC in 16 h photoperiod at 35 µmol·m–2·s–1. After 28 days, softwood cuttings treated with IBA + NAA (3000 + 3000 μmol·L–1) had highest rooting percentage (89.3%) with 5.5 mean roots. Shoot apex and nodal explants of softwood cuttings were pretreated with 0.1% (w/v) ascorbic acid, boric acid, activated charcoal, citric acid, glutamine or polyvinylpolypyrollidone (PVP) for 24 h to remove phenolic compounds before surface disinfestation. Glutamine (Gl) and PVP were equally effective resulting in 60% establishment of shoot apices on MS medium supplemented with 10 μmol·L–1 6-benzylaminopurine (BAP) + 5 μmol·L–1 NAA. Using shoot apices, highest (42.80) number of multiple shoots with 54.33 mm shoot length were obtained on MS + BAP (8.8 μmol·L–1) + IBA (2 μmol·L–1) after 45 days. In this paper,a protocol for efficient shoot regeneration was successfully developed from hypocotyl explants of Euonymus fortunei var.radicans.Some factors that influenced shoot regeneration such as different combinations of plant growth regulators,types of medium and inoculation ways were studied in order to establish an efficient plant regeneration for transformation.The results showed that hypocotyl explants wero horizontally cultured on a basic medium composed of MS medium supplemented with 0.5 mg·L-1 BAP and 0.01 mg·L-1 NAA for induction and development of adventidous shoots.Ninety-four percent of regeneration frequency and 5.1 shoots per explants were obtmned after 30 days of culture.Regenerated shootsproliferated efficiently on a shoot multiplication medium consisting of MS medium containing 1.0 mg·L-1 BAP and 0.1 mg·L-1 NAA.Microshoots were rooted on a rooting medium made up of MS medium enriched with O.5 mg·L-1 IBA and O.5 mg·L-1IAA.After hardening,90% of plants were successfully established under greenhouse conditions.Histological observation revealed that shoot primordium originated from subepidermal cells of hypocotyl explants and directly developed into adventitious shoots without caHus formation.  相似文献
