ABSTRACT:   Lipophilic toxin profiles associated with diarrhetic shellfish poisoning in Korean bivalves were investigated by liquid chromatography-mass spectrometry (LC-MS). Okadaic acid (OA), dinophysistoxins (DTX), pectenotoxins (PTX) and yessotoxin (YTX) in bivalves were quantified by LC-MS. Thirty-eight bivalve samples were collected from five different areas on the south coast of Korea during January to June 2006. Dinophysistoxin-1 (DTX1) and OA were found in some samples of both mussel Mytilus edulis and oyster Crassostrea gigas . The levels of DTX1 and OA found in the hepatopancreas of the samples were 0.04–0.23 µg/g and 0.03–0.08 µg/g, respectively. Dinophysistoxin-3 was found in the hepatopancreas of the short-necked clam Ruditapes philippinarum at a concentration of 0.06 µg/g. PTX and YTX were not detected from all of the samples examined. This is the first report of the unambiguous identification of OA and its derivatives, by LC-MS, in Korean bivalves.  相似文献

笔者初步研究了企鹅珍珠贝(Pteria penguin)组织蛋白酶D(cathepsin D,CTSD)的基因克隆和功能,通过同源克隆方法和cDNA末端快速扩增技术(RACE)获得了企鹅珍珠贝组织蛋白酶D基因(命名为pgCTSD)。该基因cDNA全长1 767 bp,其中5'UTR为38 bp,3'UTR为553 bp,ORF为1 176 bp,编码392个氨基酸,包括信号肽(Met1-Ala18)、前体域(Leu19-Lys47)和成熟域(Tyr48-Ser392)三部分,分子量为42.3 kDa,等电点为8.04。pgCTSD氨基酸序列与大珠母贝(Pinctada maxima)pmCTSD的相似性最高(79%),与其他物种的相似性为59%~75%。荧光定量分析表明,空白对照组中pgCTSD mRNA在闭壳肌、性腺、肝胰脏、外套膜和鳃组织中都有表达,且在闭壳肌中表达量最少,肝胰脏中最高。与试验对照组相比,脂多糖(LPS)刺激6 h后性腺和肝胰脏显著下降,闭壳肌的表达量虽不大但增加显著,外套膜和鳃组织变化不显著;哈维弧菌(Vibrio harveyi)刺激6 h后肝胰腺和外套膜显著下降,闭壳肌和鳃显著上升,性腺无显著变化。肝胰腺中pgCTSD对LPS和弧菌刺激的应答反应表明pgCTSD可能参与了免疫反应。  相似文献

ABSTRACT:   Quantification of lipophilic toxins in bivalves associated with diarrhetic shellfish poisoning was investigated by liquid chromatography–mass spectrometry (LC–MS). Using a C8-silica reversed phase column and a mobile phase of aqueous acetonitrile containing 2 mM ammonium formate and 50 mM formic acid, okadaic acid, dinophysistoxin-1, 7- O -palmitoyldinophysistoxin-1, pectenotoxin-1, pectenotoxin-2, pectenotoxin-6, pectenotoxin-2 seco-acid, yessotoxin, and 45-hydroxyyessotoxin in bivalves were quantified by LC–MS in the negative mode. When the crude 90% methanol extracts were analyzed by LC–MS, there were no significant effects from bivalve matrices on the quantification of toxins. More than 200 bivalve samples collected from various production areas in Japan were analyzed by LC–MS. Pectenotoxin-6 and dinophysistoxin-1 were the dominant toxins in scallops and mussels, respectively. Yessotoxin and 45-hydroxyyessotoxin were also detected in both species. Comparison of the quantitative results obtained for these bivalve samples between LC–MS and mouse bioassay indicates that LC–MS is suitable for routine monitoring of lipophilic toxins in Japanese bivalves.  相似文献

A pharmacokinetic study on enrofloxacin (ENR) following different administration regimes (i.e. intramuscular, oral and bath administrations) was carried out for the Chinese mitten-handed crab ( Eriocheir sinensis ) at 17 °C water temperature. Muscle and hepatopancreas samples were collected at each sampling time of 1 and 10 min and 0.5, 1, 2, 8, 16, 24, 48, 72 and 168 h for intramuscular administration and 0.25, 0.5, 1, 2, 4, 6, 8, 12, 24, 48, 72 and 168 h for oral and bath administration. The levels of ENR and ciprofloxacin (CIP) were simultaneously determined ( n =5) by high-performance liquid chromatography. The results showed that similar absorption and elimination patterns of ENR level in muscle and hepatopancreas were observed for oral administration and bath treatment; however, the trend in ENR level changes in muscle and hepatopancreas after intramuscular administration was quite different from oral and bath treatments. Meanwhile, the level of ENR in hepatopancreas was much higher than that in muscle for the same administration regimes. The concentration–time data were analysed with the Practical Pharmacokinetics Program (3P97), and the kinetic profiles of ENR in the hepatopancreas and muscle conformed to two-compartment models. The absorption and elimination of ENR in hepatopancreas for bath treatment were faster than those for oral and intramuscular administrations. However, the pattern was quite different in muscle. The fastest absorption and elimination of ENR in muscle were seen for intramuscular administration.  相似文献

ABSTRACT:     The difference in paralytic shellfish poisoning (PSP) toxin accumulation in the crabs Telmessus acutidens and Charybdis japonica was investigated at Onahama, Fukushima Prefecture, from 2002 to 2005. The level of toxin accumulation in the hepatopancreas of T. acutidens corresponded to that of mussels when examined on a yearly basis. In 2003, some crabs had a high toxicity of approximately 1000 MU, which compares to one-third of the human minimum lethal dose. Therefore T. acutidens should be monitored as a vector species of PSP toxin. The toxin profile of T. acutidens was also investigated. Because an increase in highly toxic species of PSP toxins with a decrease in low toxic species, such as N -sulfocarbamoyl-11-hydroxysulfate toxins, was not clearly observed between consecutive samples, toxin transformation in T. acutidens was considered to have a minimal impact on toxicity. PSP toxins were also detected in several specimens of C. japonica , but the highest toxicity was only 7.4 MU/g in the hepatopancreas. Because C. japonica is widely distributed in the coastal waters of Japan, investigation of toxin accumulation in the crab should be continued, including its investigation in different areas of water from Onahama, where the causative dinoflagellates of PSP occur in different conditions.  相似文献

Pharmacokinetics of oxolinic acid in gilthead sea bream, Sparus aurata L.   总被引:1,自引:0,他引:1  
This is the first study on the pharmacokinetic parameters of oxolinic acid (OA) in gilthead sea bream, Sparus aurata L. The kinetic profile of OA was studied after a single intravascular injection (20 mg kg−1) in 100 g fish at 20 °C. The distribution half-life ( t 1/2α) and the elimination half-life ( t 1/2β) of the drug were found to be short (0.51 and 12.60 h, respectively). The drug penetration from the plasma to the tissues was adequate as the apparent volume of distribution of the drug at steady-state ( V d(ss)) was found to be 2.11 L kg−1. The mean residence time ( MRT ) of OA was short (14.25 h) and the total clearance rate ( Cl T) of the drug was low (0.15 L kg−1 h−1). The bioavailability ( F %) of OA following oral administration (30 mg kg−1) was also low (14%). Maximum values were observed for muscle at 0.5 h after injection, with levels declining as with subsequent sampling. At the first two time points (0.5 and 1 h) plasma levels of OA were higher than muscle, however, the reverse was evident for subsequent samples. Following oral administration, highest muscle levels were found at 16 h and, with the exception of the 24-h sampling, muscle OA concentrations were higher than plasma at all time points. The fast elimination of OA suggests short withdrawal times with reference to human consumption of treated fish.  相似文献

ABSTRACT:   Yessotoxin (YTX) is a shellfish toxin and its contamination in bivalves has seriously damaged shellfish industries. The biogenetic origin of YTX was identified as the dinoflagellate Protoceratium reticulatum (Claparède et Lachmann) Bütschli collected in New Zealand and Yamada Bay, Iwate in Japan. Scallops cultured in Mutsu Bay, Japan, were frequently contaminated with YTX, however, occurrence of P. reticulatum in this bay and YTX production by the local strains have not been investigated. Eight strains of P. reticulatum , isolated from the bay, were cultured in the laboratory, and analyzed by fluorometric high-pressure liquid chromatography and liquid chromatography/mass spectrometry methods for YTX production and composition. All strains tested were confirmed to produce YTX, and none of them produced known YTX analogs. Toxin amount and composition differed from strain to strain. This result is also confirmation of one of the biogenetic origins of YTX in Mutsu Bay.  相似文献

Six diets were formulated with vitamin B6 levels (2.6, 32.7, 54.8, 90.7, 119.6 and 247.4 mg kg−1, dry diet) to determine the requirement for juvenile Pacific white shrimp, Litopenaeus vannamei. Triplicate groups of 40 juvenile shrimp (approximately 1.0 g) were provided four times each day to apparent satiation (8 weeks). Weight gain (WG), specific growth rate, feeding efficiency, protein efficiency ratio (PER) and protein productive value of the shrimp were significantly influenced by the vitamin B6 levels. No significant differences in whole‐body and muscle composition, except for dry matter and protein contents in whole body. Vitamin B6 concentration in the hepatopancreas significantly increased with the dietary vitamin B6 level increasing from 2.6 to 32.7 mg kg−1. High‐density lipoprotein cholesterol in the haemolymph improved with the dietary vitamin B6 levels increasing from 2.6 to 90.7 mg kg−1 diet and no significant differences in low‐density lipoprotein cholesterol, cholesterol, glucose and total protein concentrations. Aspartate aminotransferase, alanine aminotransferase, superoxide dismutase, catalase and lysozyme in the haemolymph were significantly influenced by dietary vitamin B6 levels. The optimal dietary vitamin B6 requirements estimated using a two‐slope broken‐line model based on WG and SGR and an exponential model based on the vitamin B6 concentration in the hepatopancreas were 110.39, 110.08 and 167.5 mg kg−1, respectively.  相似文献

The fatty acid compositions of the tail muscle and hepatopancreas of captive noble crayfish Astacus astactus and of signal crayfish pacfastacus leniusculus were investigated. The major fatty acids in the tail muscle of A. astacus were 16:0. 18:ln-7 + 9, 20:4n–6 and 20:5n–3 and the fatty acid composition of hepatopancreas was principally 16:0, 18:In-7 + 9 and 20: ln-9+ 11. These animals had been fed marine fish (sprat and herring) for 2 mo. They were kept in a flowthrough system with gravity-fed fresh water from a nearby lake with a high concentration of calcium and pH above 8. The major fatty acids in the tail muscle of P. leniusculus were 16:0. 18:ln-7+9, 18:2n–6 and 20:5n–3, and in the hepatopancreas 16:0, 18:1n–7 + 9 and 18:2n–6. These animals had been fed mainly plant material (green peas, maize and alder leaves) in a different laboratory for a period of about 2 yr and kept in a flow-through system with ambient water of pH around 7. In A. astacus the total fatty acid content was nearly 13 times higher in the hepatopancreas than in the tail muscle, however, three fatty acids 18:0, 20:4n–6 and 20:Sn-3 were, respectively, 289, 403, and 320% more concentrated in the fatty acids of tail muscle than in the hepatopancreas. Total fatty acids of the hepatopancreas of male P. Ieniusculus was 42 times higher than in the tail muscle and was three times greater than in the hepatopancreas of A. astacus .  相似文献

本文选择在我国分离得到的一株能典型产生麻痹性贝毒(PSP,Paralytic Shellfish Poison)的有毒赤潮甲藻塔玛亚历山大藻(Alexandrium tamarense,ATHK株),研究其是否能通过引发中国明对虾的脂质过氧化作用, 使机体产生过多活性氧自由基, 导致氧化损伤而发挥其毒性作用。塔玛亚历山大藻粗提液经肌肉注射方式染毒中国明对虾,于染毒后1、3、6、12、24和48 h 测定肝胰腺和鳃SOD,GST活力和MDA含量的影响。结果显示,染毒后 1-6 h, 对虾肝胰腺和鳃组织SOD,GST酶活力均增加,12和48h鳃组织的上述指标受到抑制。对虾肝胰腺MDA含量除1h外未见明显改变,鳃中MDA含量随时间增加呈升高趋势。研究表明:塔玛亚历山大藻粗提液在中国明对虾体内代谢过程中能诱导对虾产生活性氧自由基,塔玛亚历山大藻粗提液能诱导MDA含量增加,降低SOD和GST活力,能够引发鳃的脂质过氧化,对鳃造成氧化损伤。  相似文献

Quantitative real time PCR, recently developed in molecular biology, is applied in this paper to quantify the white spot syndrome virus (WSSV) in infected shrimp tissue. The WSSV content in moribund shrimp of all species tested ( Penaeus stylirostris, P. monodon, P. vannamei ) ranged from 2.0 × 104 to 9.0 × 1010 WSSV copies μg–1 of total DNA ( n =26). In whole moribund post-larvae, 4.3 × 109 WSSV copies μg–1 of DNA were detected which is equivalent to 5.7 × 1010 WSSV copies g–1 of post-larvae. The comparison of WSSV content between different tissues showed that muscle and hepatopancreas tissues contained 10 times less virus than gills, pleopods and haemolymph. With inocula of known virus content, bioassays by immersion challenge showed that a minimum of five logs of WSSV copies was necessary to establish disease in the challenged shrimp. In contrast, five logs of WSSV copies injected into shrimp muscle produced a LT-50 of 52 h. This real time polymerase chain reaction (PCR) technique is sensitive (four copies), specific (negative with DNA from shrimp baculoviruses and parvoviruses), dynamic (seven logs) and easy to perform (96 tests in <4 h).  相似文献

This study was designed to assess the protective effects of dietary selenium (Se) on abalone Haliotis discus hannai Ino against the toxicity of waterborne copper (Cu). A 60‐day feeding trial was conducted in a static water system for abalone (initial weight: 3.17 ± 0.01 g) exposed to 0.02 mg L?1 of waterborne Cu. The animals were fed one of the three experimental diets with 0.10, 1.31 and 4.20 mg kg?1 of Se from Na2SeO3·5H2O respectively. Results showed that the abalone fed 1.31 mg kg?1 of dietary Se had the lowest Cu concentration in shell, muscle, mantle, gill, hepatopancreas and serum. Meanwhile, the significant lowest contents of malondiadehyde and protein carbonyl in hepatopancreas were also found in the treatment with 1.31 mg kg?1 of dietary Se (P < 0.05). In addition, this treatment had significant higher glutathione content and thioredoxin reductase activity in hepatopancreas (P < 0.05). However, the activity of Se‐dependent glutathione peroxidase (Se‐GPx) was significantly decreased in the treatment with 4.20 mg kg?1 of dietary Se (P < 0.05). In this treatment, the protein carbonyl content in hepatopancreas was significantly higher than that in the group with 1.31 mg kg?1 of dietary Se (P < 0.05). In conclusion, in terms of anti‐oxidation and Cu accumulation, the protective effects of dietary Se on abalone against waterborne Cu were dose‐dependent. The 1.31 mg kg?1 of dietary Se had this effect, but not 4.20 mg kg?1 of dietary Se. Moreover, the latter increased the oxidative stress in abalone exposed to the waterborne Cu.  相似文献

Microcystins accumulate in aquatic organisms and can be transferred to higher trophic levels, eventually affecting vector animals and consumers. We examined three levels of an aquatic food chain (Microcystis aeruginosa, Daphnia magna and Macrobrachium rosenbergii) to identify the transfer efficiency and risk of microcystin on prawns. Samples were analysed using ultra performance liquid chromatography‐mass spectrometry (MS)/MS and microcystin‐LR (MC‐LR) distributions in prawn tissues were studied. The results showed that prawns accumulate MC‐LR both directly from M. aeruginosa and indirectly through D. magna which was pre‐exposed to M. aeruginosa. MC‐LR was detected in the gills, digestive tracts and hepatopancreas of the prawns 2 h after exposure. MC‐LR accumulated in prawns to 0.49 ± 0.04 μg g?1 dry weight in hepatopancreas within 24 h, while it was not detected in muscle samples, and rarely appeared in blood samples in such a short period. Although MC‐LR was not detected in muscle, the head including hepatopancreas of the prawns accumulated troublesome amounts of MC‐LR. These results demonstrate that microcystis blooms in prawn farming potentially pose a risk to human consumers, although prawns may be exposed to the bloom for a very short time, hence regular monitoring of blue green algae population is recommended.  相似文献

通过对日本蟳(Charybdis japonica)分别注射生理盐水(对照组)、溶藻弧菌(攻毒组)、免疫多糖(免疫组)、免疫多糖+溶藻弧菌(免疫攻毒组)后,再测定4组肝胰腺及肌肉中溶菌酶(LZM)和超氧化物岐化酶(SOD)的活性变化,探讨免疫多糖对日本蟳抗溶藻弧菌能力的影响。结果表明:日本蟳肝胰腺中LZM和SOD的酶活均高于肌肉中的酶活,且免疫多糖对肝胰腺的免疫增强效应显著高于肌肉;免疫组肝胰腺和肌肉中LZM酶活,在注射后24h时显著升高至峰值(P<0.05),分别为(11.2±2.78)U/mg和(3.75±1.05)U/mg,肝胰腺和肌肉中的SOD酶活也在24h达到最高值,分别是对照组的2.2倍和1.4倍;免疫攻毒组在72h时,肝胰腺和肌肉中的LZM酶活分别高出攻毒组62.8%和75.2%,两组织中SOD酶活同样显著高于攻毒组(P<0.05),是攻毒组的3.5倍和4.2倍。由此可见,免疫多糖能提高日本蟳抗溶藻弧菌能力,且在72h达到最佳免疫保护效果。  相似文献

The pearl oyster Pinctada fucata is cultured for pearl production in China, but its germplasm is degenerated. Thus, lipid and fatty acids were investigated in P. fucata based on different tissues, body sizes and condition indexes using GC–MS to provide a lipidomic baseline for evaluation of breeding variety in terms of physiology. The major fatty acids over all samples included saturated (C16:0 and C18:0), monounsaturated [C16:1(n-7), C18:1(n-7) and C18:1(n-9)], and polyunsaturated fatty acids [arachidonic acid: C20:4(n-6), EPA: C20:5(n-3) and DHA: C22:6(n-3)]. Relative percentages of lipid contents of the mantle and visceral mass were, respectively, 40.75 and 42.03 %, significantly higher than adductor muscle (14.48 %) and gill (17.15 %). Fatty acid analysis showed that there were 15 types of fatty acids in adductor muscle, 17 types in mantle and gill, respectively, and 19 types in visceral mass. Meanwhile, total fatty acids, SFA, MUFA and PUFA were the highest in visceral mass (P < 0.05). Yet the lipid contents and fatty acid compositions showed no significant differences between body sizes or condition indexes based on visceral mass. The observations above suggested that visceral mass and mantle are the major tissues for P. fucata to store lipids and thus are important candidate tissues for lipidomic study.  相似文献

ABSTRACT:   Accumulation profiles of 22 trace elements in abdominal muscle, abdominal exoskeleton and the hepatopancreas of the giant river prawn Macrobrachium rosenbergii were analyzed. The giant river prawn is an indigenous freshwater species from South Vietnam, and is cultured commercially and fished in the wild. Samples were collected from Ho Chi Minh City and the surrounding area (SKEZ, South Key Economic Zone), and from the Mekong River Delta between 2003 and 2005. Highest accumulations of essential (Cu, Se and Mo) and toxic (As, Ag, Cd and Hg) elements were observed in hepatopancreatic tissue, except for Mn, Sr, Sn, Ba and V in the exoskeleton and Rb and Cs in muscle tissue. Spatial differences showed concentrations of Cs and Pb in muscle and Sr in exoskeletons from the SKEZ were higher than those from the Mekong River Delta. The opposite trend was observed for Cr, Se and Sb in muscle, Mo, Sb and Tl in exoskeleton, and Se, Hg, Mo, Cd, Sb, Tl and Bi in the hepatopancreas. These differences in trace element concentrations in prawns likely reflect differences in industrialization and human activities between the two regions of South Vietnam.  相似文献

ABSTRACT:   Paralytic shellfish poisoning (PSP) toxins produced by Alexandrium isolates from Korea were analyzed by high-pressure liquid chromography. Species designation of the regional isolates was determined by morphological criteria and ribotyping inferred from sequences of the 28S rDNA D1-D2 region. Toxin analysis performed at the exponential growth phase, revealed that the two strains of A. fraterculus were non-toxic, while the strains of A. tamarense and A. catenella were toxic. Toxic isolates DPC7 and DPC8 of A. catenella produced GTX1, 2, 3, 4, 5, dcGTX2, 3, C1, 2, neoSTX and STX with trace or non-detectable levels of C3 and C4, while isolates UL7, KDW981, SJW97043, SJW97046, KJC97111 and KJC97112 of A. tamarense produced GTX1, 2, 3, 4, dcGTX3, C1, 2, neoSTX with trace or non-detectable levels of C3, 4, dcSTX and STX, and no GTX5 and dcGTX2. The major toxins produced by A. catenella were C1 +2, and those of A. tamarense were C1 +2 and GTX4 in most of the isolates. A. tamarense strains other than SJW97046 produced a relatively high proportion of carbamate toxins, reflecting the high toxicity scores of shellfish intoxication in sampled coastal areas. Two representative toxic isolates, A. tamarense SJW97043 and A. catenella DPC7, were cultured for 30 days in batch mode and subjected to toxin analysis at 5-day intervals. Comparison of toxin productivity in terms of total toxin content, toxin components, and their variations with culture age revealed marked differences between the two strains.  相似文献

The freshwater pearl mussel Hyriopsis cumingii is the most important mussel species for freshwater pearl production in China. Mussel shell color is an important indicator of pearl quality. The objective of this study was to assess whether total carotenoid content (TCC) in H. cumingii is related to shell color. TCC of different tissues (gonad, gill, hepatopancreas, kidney, axe foot, mantle, and adductor muscle) of purple and white inner-shell H. cumingii was determined by UV–Vis spectrophotometry. The results revealed that TCC ranged from 12.91 ± 0.78 to 56.30 ± 0.74 μg/g. In general, TCC was higher in the hepatopancreas, followed by the mantle, gonad, gill, kidney, axe foot, and adductor muscle. TCC in gonad, gill, hepatopancreas, kidney, and mantle of purple mussels was significantly higher than that of white mussels. TCC in mussel tissues of H. cumingii was significantly different (P < 0.001) with respect to shell color. There were significant positive correlations between TCC in mussel mantle and shell color intensity. Future studies will assess the biological roles of carotenoids in shell color formation.  相似文献
