Efficacy of ionophores in cattle diets for mitigation of enteric methane   总被引:3,自引:0,他引:3  
Use of ionophores in cattle diets has been proposed as a strategy for mitigation of enteric CH4 emissions. Short- and long-term effects of feeding a single ionophore (monensin) or rotation of 2 ionophores (monensin and lasalocid) on enteric CH4 emissions were evaluated in 36 Angus yearling steers (328 +/- 24.9 kg of BW) over a 16-wk period. Steers were randomly assigned to 6 dietary treatments of 6 steers each. The 6 diets were low-concentrate without ionophore supplementation, low-concentrate with monensin supplementation, low-concentrate with a 2-wk rotation of monensin and lasalocid supplementation, high-concentrate without ionophore supplementation, high-concentrate with monensin supplementation, and high-concentrate with a 2-wk rotation of monensin and lasalocid supplementation. Daily enteric CH4 emissions, as measured using the SF(6) tracer gas technique, ranged from 54.7 to 369.3 L/steer daily. Supplementing ionophores decreased (P < 0.05) enteric CH4 emissions, expressed as liters per kilogram of DMI or percentage of GE intake, by 30% for the first 2 wk and by 27% for the first 4 wk, for cattle receiving the high-concentrate and low-concentrate diets, respectively. Cattle fed a rotation of ionophores did not (P > 0.05) exhibit a greater decrease and did not (P > 0.05) have a longer period of depressed enteric CH4 emissions compared with cattle receiving monensin only. Ionophore supplementation did not (P > 0.05) alter total ruminal fluid VFA concentration; however, the acetate:propionate ratio and ammonia-N concentration in ruminal fluid were decreased (P < 0.001) from the time that ionophores were introduced to the time they were removed from the diets. Both monensin and the rotation of monensin and lasalocid decreased (P < 0.001) total ciliate protozoal populations by 82.5% in the first 2 wk and by 76.8% in the first 4 wk during which they were supplemented in the high-concentrate and low-concentrate diets, respectively. Original ciliate protozoal populations were restored by the fourth and sixth week of supplementation when cattle were fed the high- or low-concentrate diets, respectively. No significant change was observed thereafter. These data suggest that the effects of ionophores on enteric CH(4) production are related to ciliate protozoal populations and that ciliate protozoal populations can adapt to the ionophores present in either low- or high-concentrate diets. Rotation of monensin and lasalocid did not (P > 0.05) prevent ciliate protozoal adaptation to ionophores.  相似文献

Salinomycin, a new ionophore antibiotic, was tested and compared with lasalocid and monensin for preventing experimentally induced lactic acidosis. Five rumen-fistulated adult cattle were used in a 5 X 5 Latin square design, and the treatments were as follows: no treatment (control), 0.11 mg of salinomycin/kg of body weight (S1), 0.22 mg of salinomycin/kg (S2), 0.66 of lasalocid/kg, and 0.66 mg of monensin/kg. Acidosis was induced by intraruminal administration of a ground corn-corn starch mixture (50:50, 12.5 g/kg) once a day for up to 4 days. Antibiotics were administered along with grain-starch mixture. Rumen and blood samples were obtained before and at 6, 12, and 24 hours after each carbohydrate-antibiotic dosing to monitor acid-base status. Control and S1-treated cattle became ruminally acidotic within 54 hours, whereas cattle treated with S2, lasalocid, and monensin resisted acidosis for up to 78 hours after dosing. Cattle treated with S2, lasalocid, or monensin had higher rumen pH and lower L(+)- and D(-)-lactate concentrations than did control or S1-treated cattle. Rumen pH decrease to below 5.0 in S2-, lasalocid-, and monensin-treated cattle was not due to lactic acid, but to increased production of volatile fatty acids. Rumen propionate proportion increased initially in antibiotic-treated cattle, but after 48 hours, butyrate proportion increased significantly. Despite low rumen pH and high lactate concentration, lacticacidemia was not evident, and the systemic acid-base disturbance was mild in control cattle.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Effect of monensin on rumen ciliate protozoa in sheep   总被引:1,自引:0,他引:1  
In a first experiment 4 rams consumed over a period of 70 days 0; 82; 208 and 345 mg of monensin daily, respectively. All doses of monensin caused a marked decrease in total numbers of rumen protozoa in samples taken after this period. More than 90% of total protozoal numbers belonged to the genus Entodinium. The sensitivity of the genus Entodinium to monensin was found to be lower than that of genera Dasytricha and Isotricha. In the second experiment 24 lambs were fed hay and concentrates in the ratio 60:40% (groups 1 and 2) and 40:60% (groups 3 and 4). Groups 2 and 4 received 40 mg monensin per animal daily during 16 weeks. The decrease in protozoal numbers due to monensin in samples taken after this period was significant (P less than 0.001) in lambs fed the concentrate diet (groups 3 and 4) and also in lambs from groups 1 and 2 fed the roughage diet (P less than 0.05). The statistical evaluation of the inhibition of total protozoal numbers by monensin (%) in lambs fed both diets has shown that the antiprotozoal effect of monensin was significantly more intensive with the concentrate diet (P less than 0.025) than with the roughage diet.  相似文献

The present work was an attempt to determine whether the variations in ruminal ammonia concentrations could be directly correlated to corresponding changes in total protozoa numbers of cattle. Four dry Friesian cows fed with hay-based rations were used through several experiments in which the twice daily feeding (6.15-15.30 h) as well as a 30 h-fasting period were studied. Simultaneously to a continuous or a regular collection of rumen liquor (for NH3), samples of rumen contents (for total protozoa numbers) were regularly withdrawn from the ventral sac. Any definite nycthemeral cycle of the protozoa numbers could not be related to feeding time. Moreover, no significant relationship could be found between the ruminal ammonia and the corresponding total protozoa numbers measured in the nocturnal interprandial period or during starvation. The data suggest that the nycthemeral ammonia profiles recorded in cattle are not directly related to protozoal activity.  相似文献

Lasalocid toxicity in cattle: acute clinicopathological changes   总被引:1,自引:0,他引:1  
Thirty-six steers (148 to 500 kg) divided into six equal groups were used in a toxic syndrome study of lasalocid and monensin given as a single oral dose. One group was given a placebo, a second group received monensin (25 mg/kg body weight) and the other four groups received lasalocid at 1, 10, 50 or 100 mg/kg body weight (bw). No toxic signs developed in cattle given placebo or lasalocid at 1 or 10 mg/kg bw dose. The earliest toxic signs were muscle tremors, tachycardia and rumen atony. After 24 h, the cattle were dehydrated, anorectic and had diarrhea. Deaths occurred between d 1 and 22.5 in the groups receiving lasalocid at 50 and 100 mg/kg bw and monensin. Altered values in blood leucocytes, erythrocytes, hemoglobin, hematocrit, total protein, albumin, creatinine, urea nitrogen, total bilirubin, creatine kinase, lactic dehydrogenase, calcium, chloride and inorganic phosphate occurred 1 d after dosing: urine pH and specific gravity also changed 1 d after dosing. Maximum changes occurred at d 3. Most of the changes were indicative of dehydration rather than specific organ damage.  相似文献

Fermentation characteristics were measured and numbers and distribution by genera of ciliate protozoa were determined in ruminal fluid samples collected from 10 ruminally cannulated steers during the first 30 d of their being fed barley-based diets containing 62% (Medium Barley) or 95% (High Barley) barley grain (DM basis). Ruminal samples were collected at 5-d intervals over the 30-d periods beginning after adaptation (i.e., at the first full feeding of each diet). Ruminal pH and ammonia concentrations were lower (P < 0.001) with the High Barley than with the Medium Barley diet. Concentrations of total VFA and propionate and amylase activity of ruminal fluid were higher (P < 0.001) on High Barley than on Medium Barley. Total protozoal numbers in ruminal fluid were 42% lower (P < 0.05) on High Barley (470 x 10(3)/mL) than on Medium Barley (804 x 10(3)/mL). On Medium and High Barley diets, respectively, Entodinium spp. made up 89 and 91% of the ciliate protozoal populations. With the Medium Barley diet, relative proportions of Dasytricha, Ophryoscolex, Ostracodinium, Diplodinium, and Metadinium spp. in the total ciliate population were 4.5, 0.4, 0.5, 0.7, and 0.3%, respectively. When the High Barley diet was fed, these genera were not detected. In a subsequent survey, ruminal samples were collected from 200 finishing cattle at slaughter. Average protozoal population was 328 x 10(3)/mL, and Entodinium spp. constituted 97% of the total. These data demonstrate that a large population of Entodinium spp. can persist in the rumen of cattle fed high barley grain-based finishing diets.  相似文献

The effects of incorporation in the diet of 7% soya oil hydrolysate (SOH) on in vitro incubations of cellobiose + maltose, maize starch and casein by rumen microbes were studied using defaunated and refaunated sheep as rumen fluid donors. Feeding refaunated sheep the SOH supplemented diet lowered the protozoal numbers in the rumen from 1.61 10(6)/ml to 6.1 10(5)/ml. SOH addition reduced in vitro methane production, rather by a depletion of methanogens is than by a simple inhibition of their activity. This reduction seemed to be independent of protozoa depletion. With cellobiose-maltose and maize starch incubations, SOH supplementation increased molar proportion of propionate while acetate decreased. Both variations could be linked to the inhibition of methanogenesis. Volatile fatty acid production from casein was strongly reduced by SOH supplementation with or without protozoa in the rumen of the donors animals.  相似文献

The acute toxicity of lasalocid and monensin was studied in 36 Holstein steers. The cattle were given (orally) a single dosage of lasalocid (1, 10, 50, or 100 mg/kg of body weight) or monensin (25 mg/kg of body weight) or rice hulls. Animals were observed once a day until they died or were euthanatized at 32 days after the dose was given. All cattle were necropsied. Heart, kidney, adrenal gland, liver, spleen, pancreas, lungs, brain, sciatic nerve, skeletal muscle, small intestine, large intestine, and rumen tissue sections, stained with hematoxylin and eosin, were studied microscopically. Lasalocid was lethal at dosages of 50 and 100 mg/kg, and monensin was lethal at the dosage given (25 mg/kg). Cattle dying of lasalocid and monensin toxicoses had gross and microscopic lesions consistent with cardiomyopathy. Dilated heart or petechial and ecchymotic hemorrhages were observed with both drugs. Microscopically, multifocal areas of myocyte necrosis were observed. Those cattle that died within 3 days of dosing with either drug had a marked degranulation of pancreatic acinar cells. Changes were not observed in any other tissues.  相似文献

以4只瘘管山羊为试验动物,采用4×4拉丁方设计,使用克隆测序、遗传指纹以及细胞显微计数等技术,研究以羽毛粉(A)、玉米蛋白粉(B)、豆粕(C)和鱼粉(D)4种蛋白补充料分别配合的混合日粮对瘤胃细菌和原虫群体结构的影响。结果表明:细菌的R.flavefaciens、R.bromii、R-7、Roseburia faecalis、Uncultured T33H60F48、Uncultured GRANT42及1个尚不能定性的类群在组间存在显著差异(P<0.05);原虫的内毛虫、前毛虫、头毛虫、等毛虫4个类群在处理间差异显著(P<0.05)。综上可见,蛋白质源对瘤胃细菌和原虫类群结构有显著影响。  相似文献

Knowledge of the fatty acid profile of microbial lipids is of great nutritional importance to the animals and, subsequently, their products. This study was conducted to examine the fatty acid profiles of mixed rumen bacteria and protozoa. Bacterial and protozoal cells were isolated by differential centrifugation of rumen contents. The main fatty acids were palmitic (16:0) and stearic (18:0) in both the bacterial and protozoal fractions. Palmitic acid was 74% greater in the protozoal fatty acids than in the bacterial fatty acids, whereas bacteria had 2.25-times greater stearic acid (18:0) proportions compared with protozoa. The total odd-chain plus branched-chain fatty acids were 16.5% of bacterial fatty acids and 11.0% of protozoal fatty acids. The anteiso-17:0 proportions in bacterial and protozoal fatty acids were 1.4 and 2.9%, respectively. The most abundant trans-18:1 isomer, vaccenic acid (18:1 trans-11), was 6.6% of total fatty acids in protozoa and 2.0% of total fatty acids in bacteria. The cis-9, trans-11 CLA was 8.6-times greater in the protozoal fraction (1.32% of total fatty acids) than in the bacterial fraction (0.15%). These results suggest that the presence of protozoa in the rumen may increase the supply of CLA and other unsaturated fatty acids for lower gut absorption by ruminants.  相似文献

以莫能霉素、盐霉素添加剂和自制的复合营养舔(饲)块饲喂本地黄牛,研究其对瘤胃原虫(Rumen protozoa)计数厦瘤胃pH值的影响。试验结果表明,黄牛日粮添加莫能霉素、盐霉素使瘤胃原虫数减少,而补喂舔(饲)块使原虫数增加。同时发现,喂稻草型日粮时,黄牛瘤胃原虫计数较放牧时少。研究我国养牛条件下原虫的作用对牛饲料添加剂、瘤胃调控制的利用具有重要意义。  相似文献

Mixed ruminal bacteria (n = 4) were incubated in anaerobic media for 24 h in vitro with either hay, corn meal, protein hydrolyzate or hydrogen gas as the substrate. The ionophore monensin and the polypeptide antibiotic bacitracin were added to the incubation flasks at concentrations ranging from 0 to 10 or 40 mg/liter. As was expected, monensin decreased methane production, increased the ratio of propionate to acetate and decreased the deamination of amino acids. Monensin had little effect on methane production, however, if hydrogen gas was the fermentation substrate. Bacitracin, another gram-positive antibiotic with a distinctly different cellular target, was somewhat less potent than monensin, but it produced strikingly similar responses. This similarity of fermentation patterns suggested that monensin action in the rumen is probably due to its activity as a gram-positive antibiotic, and that any gram-positive antibiotic not suppressed by resistance may produce fermentation effects similar to those of monensin. The cellular action of monensin as an ionophore in membranes is probably little more than a means of inhibiting sensitive species. Many gram-positive antibiotics have little affect on ciliate protozoa or coccidia.  相似文献

Nine steers (300 to 350 kg body weight) were divided into three groups and received either no ionophore, monensin or lasalocid in a 70% cracked corn basal diet for the entire experiment. Within each group, a 3 X 3 Latin-square design was used to evaluate the effect of a high cation concentration added to the diets on in vivo methanogenesis. The three cation treatments consisted of NaCl or KCl addition (Na or K increased to 2.5% of diet dry matter) or no cation addition. Sodium addition decreased methane production in the monensin group 19% when compared with control (P less than .05). Sodium also tended to decrease methane in the lasalocid group (P less than .1) but had no effect on the no-ionophore group. There was no significant effect of K addition on methane production when compared to no added cation. However, K tended to increase methane in both the lasalocid and no-ionophore groups and decrease methane in the monensin group. Results of this experiment support in vitro evidence that cation levels may modulate ionophore effects on methane production.  相似文献

The activities of five anticoccidials were compared against Eimeria species in/of chickens, in controlled in vivo and in vitro laboratory studies. Two more recent and potent market entries (maduramicin and halofuginone) were compared with three older polyether antibiotic anticoccidials (monensin, lasalocid and salinomycin). Halofuginone, lasalocid, maduramicin, monensin and salinomycin were evaluated at 3, 125, 5, 120 and 66 ppm, respectively, of active drug in the diets. At these levels, all five drugs demonstrated significant activity against Eimeria tenella, E. maxima, E. necatrix, E. brunetti and E. acervulina (in vivo). Monensin was least effective against E. tenella, and one of the lesser efficacious drugs against E. necatrix, maduramicin, was least effective against E. maxima. In studies of single Eimeria species infections, comparable weight gains were noted for the drugs. In the mixed Eimeria species infections, however, birds treated with maduramicin had significantly higher weight gains than did birds medicated with monensin. Unlike in vivo potencies, titration in vitro indicated that monensin was most potent (active at 10(-6) mcg ml-1), and maduramicin and lasalocid least potent (inactive at less than or equal to 10(-3) mcg ml-1).  相似文献

The effects of adding crude glycerine with sodium monensin or essential oils to beef cattle diets on the intake, degradability of DM and nutrients, rumen concentration of volatile fatty acids (VFA) and in vitro gas production were evaluated. Five ruminally cannulated Nellore steers were randomly assigned to a 5 × 5 Latin square design. The treatments were as follows: CONT, without crude glycerine and additives; EO, with essential oils and without crude glycerine; MON, with sodium monensin and without crude glycerine; EOG, with essential oils and crude glycerine; MONG, with sodium monensin and crude glycerine. Treatments with essential oil and sodium monensin increased the NDF and STC intake and the DM degradability. When crude glycerine was combined with either sodium monensin or essential oil, there was a reduction in DM, NDF and STC intake and an increase in DM and CP degradability of the diets. The adding crude glycerine to essential oil diets reduced the CH4 production. Sodium monensin treatments reduced DM and NDF intake and the production of total gas, CH4, total VFA and acetic acid concentration. In conclusion, the adding crude glycerine (200 g/kg DM) with either sodium monensin (0.03 g/kg DM) or essential oil (0.5 g/kg DM) can be utilized in diets for Nellore cattle without causing detrimental effects on feed intake and improving the DM degradability.  相似文献

A lactic-acid producing bacterium was isolated from the rumen of lambs with rumen acidosis. The cells were Gram-positive, nonmotile, nonsporing, catalase negative spherical, 1.5-2.0 μm in diameter, and occur in pairs and tetrads. Analysis of 16S ribosomal RNA indicated that the rumen bacterium was a strain of Pediococcus acidilactici with 99% of nucleotide homology. This bacterium was sensible to monensin and lasalocid at the unique dose tested of 300 ppm. The concentration of lactic acid and DM degradation decreased (P < 0.05) when monensin or lasalocid were added to the culture media after 24, 48 and 72 h of incubation. In contrast, total VFA concentration and pH were higher (P < 0.05) in the culture media added with the ionophores. Up to now S. bovis is considered the main ruminal bacterium related with rumen acidosis, but the importance of P. acidilactici should be also reconsidered in experimental studies focused on the control rumen acidosis.  相似文献

Scientific literature on the use of antibiotic growth promoters (AGP) in beef cattle consuming forage-diets was reviewed. Database summarizes 136 comparisons between untreated and AGP-treated cattle from 48 bibliographic references. Performance data of cattle receiving AGP either alone or in combination with 17 β-estradiol implants was statistically analyzed. Forage quality, in terms of average daily gain (ADG), differentially influenced (P = 0.1) the effect of AGP on beef cattle performance. As the quality of forage increased, the estimated net ADG response to monensin decreased and that to lasalocid increased. ADG increased quadratically (P < 0.01) with increasing doses of monensin (R2 = 0.71) or lasalocid (R2 = 0.63). Ionophore-dosage level quadratically improved (P = 0.01; R2 = 0.52) feed conversion (FCONV) of cattle without affecting their dry matter intake (DMI; > 0.1). A linear relationship (P < 0.01) between ADG and dose of tetronasin (R2 = 0.64) or lysocellin (R2 = 0.52) was also observed. The combination of monensin and 17β-estradiol implants resulted in an additive effect on ADG of grazing cattle. The experimental results reviewed show that, in beef cattle consuming forage-based diets, ionophores improve ADG and FCONV in a dose-dependent manner, with little or no effect on DMI. In addition, results suggest that forage quality influences the direction of the ADG response to AGP supplementation in cattle. Summarized data from beef cattle implanted with 17β-estradiol and/or supplemented with monensin indicate that the combined response of these two compounds on cattle grazing high-quality pastures has no effect on ADG over that obtained by 17β-estradiol alone.  相似文献

The effect of monensin on the in vitro utilization of lactate in the rumen contents of a steer, cow and two wethers was investigated. The rate of the utilization of lactate in monensin-treated cultures was on average only two thirds of that found in cultures without monensin. Monensin tended to increase acetate and to decrease butyrate in cultures with lactate. After 11-week adaptation of a wether to monensin the inhibitory effect of ionophore persisted, being, however, less pronounced and without influence on the molar percentages of VFA. Effects of monensin reported here seem to be caused by its inhibiting action against rumen lactilytic protozoa. There were no significant differences between monensin from Elanco and the Czechoslovak analogue.  相似文献

Objective To determine the prevalence of subacute rumen acidosis (SARA) in beef cattle grazing lush pasture and the effect of monensin on reducing SARA and improving animal performance. Design Commercial Angus and Murray Grey steers received a monensin slow‐release capsule (n = 19) or remained untreated (n = 19). Cattle grazed an oats crop or tetraploid ryegrass pasture for a total of 91 days. Rumen fluid pH, volatile fatty acids (VFA) and lactic acid concentrations and body weight data were collected prior to treatment and again 28, 56 and 91 days after treatment. Changes in measures over time were analysed using mixed model repeated measures analysis. Differences in average daily gain between treatment groups were determined. Results The prevalence of SARA was low during the study, with only one animal satisfying criteria for SARA at one time point. Cattle treated with monensin capsules were 11.9 kg heavier at the completion of the study compared with untreated controls (414.5 ± 3.88 kg vs 402.6 ± 4.03 kg, P = 0.04). Rumen VFA and L‐ and D‐lactate levels did not differ between cattle treated with monensin and untreated cattle. However, the ratio of propionate to acetate plus two times butyrate was higher (P < 0.001) when cattle were treated with monensin. Conclusions Subacute rumen acidosis was not consistently detected under the conditions of the study. The higher body weight of cattle treated with monensin may have been due to improved energy utilisation of the pasture, indicated by increased propionate proportions in the rumen, rather than prevention of SARA.  相似文献
