This study investigated the possible effects of tree species diversity and identity on the soil microbial community in a species-rich temperate broad-leaved forest. For the first time, we separated the effects of tree identity and tree species diversity on the link between above and belowground communities in a near-natural forest. We established 100 tree clusters consisting of each three tree individuals represented by beech (Fagus sylvatica L.), ash (Fraxinus excelsior L.), hornbeam (Carpinus betulus L.), maple (Acer pseudoplatanus L.), or lime (Tilia spec.) at two different sites in the Hainich National Park (Thuringia, Germany). The tree clusters included one, two or three species forming a diversity gradient. We investigated the microbial community structure, using phospholipid fatty acid (PLFA) profiles, in mineral soil samples (0–10 cm) collected in the centre of each cluster.The lowest total PLFA amounts were found in the pure beech clusters (79.0 ± 23.5 nmol g−1 soil dw), the highest PLFA amounts existed in the pure ash clusters (287.3 ± 211.3 nmol g−1 soil dw). Using principle components analyses (PCA) and redundancy analyses (RDA), we found only for the variables ‘relative proportion of beech trees’ and ‘living lime fine root tips associated with ectomycorrhiza’ a significant effect on the PLFA composition. The microbial community structure was mainly determined by abiotic environmental parameters such as soil pH or clay content. The different species richness levels in the clusters did not significantly differ in their total PLFA amounts and their PLFA composition. We observed a tendency that the PLFA profiles of the microbial communities in more tree species-rich clusters were less influenced by individual PLFAs (more homogenous) than those from species-poor clusters.We concluded that tree species identity and site conditions were more important factors determining the soil microbial community structure than tree species diversity per se.  相似文献

Microbial communities exist and are active in a complex 3-D physical framework which can cause a variety of micro-environments to develop that are more or less suitable for microbial growth, activity and survival. If there is a significant microbial biogeography at the pore scale in soil, then the relationship between microbial diversity and ecosystem function is likely to be affected by micro-environmental variations at the pore scale. In this laboratory study we show that there is a significant pore-scale microbial biogeography by labelling microbial communities in different pore size classes of undisturbed soil cores with 13C-labelled fructose (a soluble, labile substrate). This was achieved by adding the substrate solution to the samples at different matric potentials (−100 kPa, −3.15 kPa and −1 kPa; placing the substrate in pores with maximum diameter of 0.97, 9.7 and 97 μm, respectively) and incubating the samples for two weeks. The mineralisation of soil organic carbon and fructose was measured as CO2 and 13C-CO2, respectively, in the jar headspace throughout the incubation. At the end of incubation we analysed the total microbial community structure using PLFA. The structure of microbial communities in different pore size classes was measured by PLFA stable isotope probing. Total PLFA profiles suggested that there was little effect of the incubation conditions on microbial community structure. However, labelled PLFA profiles showed that microbial community structure differed significantly among pore size classes, the differences being due primarily to variations in the abundance of mono-unsaturated lipids (Gram-biomarkers) and of the fungal biomarker (C18:2(9,12)). This is the first evidence for a significant microbial biogeography at the pore scale in undisturbed soil cores.  相似文献

The influence of individual trees in monocrop forests on soil microbial communities is poorly understood. We measured basal respiration, substrate-induced respiration and phospholipid fatty acids (PLFA), bacterial growth rate with the 3H-thymidine incorporation technique and fungal growth rate as 14C-acetate incorporation into ergosterol to investigate whether slow- and fast-growing 12-year-old Norway spruce (Picea abies) clones have affected differently on their associated soil microbial communities. Understorey vegetation, soil chemical properties and elemental concentrations of needles were also determined. The slow- and fast-growing spruce clones differed in PLFA profiles, understorey vegetation and elemental concentrations in needles suggesting that spruce clones have directly or indirectly affected soil microbes.  相似文献

Many biotic and abiotic factors influence recovery of soil communities following prolonged disturbance. We investigated the role of soil texture in the recovery of soil microbial community structure and changes in microbial stress, as indexed by phospholipid fatty acid (PLFA) profiles, using two chronosequences of grasslands restored from 0 to 19 years on silty clay loam and loamy fine sand soils in Nebraska, USA. All restorations were formerly cultivated fields seeded to native warm-season grasses through the USDA’s Conservation Reserve Program. Increases in many PLFA concentrations occurred across the silty clay loam chronosequence including total PLFA biomass, richness, fungi, arbuscular mycorrhizal fungi, Gram-positive bacteria, Gram-negative bacteria, and actinomycetes. Ratios of saturated:monounsaturated and iso:anteiso PLFAs decreased across the silty clay loam chronosequence indicating reduction in nutrient stress of the microbial community as grassland established. Multivariate analysis of entire PLFA profiles across the silty clay loam chronosequence showed recovery of microbial community structure on the trajectory toward native prairie. Conversely, no microbial groups exhibited a directional change across the loamy fine sand chronosequence. Changes in soil structure were also only observed across the silty clay loam chronosequence. Aggregate mean weighted diameter (MWD) exhibited an exponential rise to maximum resulting from an exponential rise to maximum in the proportion of large macroaggregates (>2000 μm) and exponential decay in microaggregates (<250 μm and >53 μm) and the silt and clay fraction (<53 μm). Across both chronosequences, MWD was highly correlated with total PLFA biomass and the biomass of many microbial groups. Strong correlations between many PLFA groups and the MWD of aggregates underscore the interdependence between the recovery of soil microbial communities and soil structure that may explain more variation than time for some soils (i.e., loamy fine sand). This study demonstrates that soil microbial responses to grassland restoration are modulated by soil texture with implications for estimating the true capacity of restoration efforts to rehabilitate ecosystem functions.  相似文献

The extent to which complex interrelationships between plants and microorganisms influence organic matter dynamics is critical to our understanding of global C cycles in changing environments. We examined the hypothesis that patterns of soil microbial activity and functional composition differ among vegetation types in northern peatland ecosystems. Microbial characteristics were compared among peatlands differing in plant growth form (tree, shrub/moss, sedge) in two regions (New York State and West Virginia). Microbial activity (basal respiration) was greater in surface (0-15 cm) than subsurface (15-30 cm) peat and from sites dominated by shrubs and Sphagnum moss (3.9±0.65 μg C g−1 h−1) compared to forested (1.8±0.20 μg C g−1 h−1) or sedge-dominated sites (1.9±0.38 μg C g−1 h−1). Microbial activity was not related to decomposability of peat organic matter among vegetation types, and activity was unexpectedly higher in sites with lower peat pH and higher water table level. Substrate-induced respiration (SIR) did not show a clear pattern among vegetation types, but was greater in surface than subsurface peat. Microbial responsiveness to added glucose was very low. The ratio of basal respiration to SIR varied between 0.39 and 0.72 and, like activity, was highest in shrub/Sphagnum sites. Microbial substrate utilization patterns (assayed with BIOLOG® GN plates) also differed between shrub/Sphagnum sites and forest or sedge sites, suggesting that C fluxes were mediated by different assemblages of microorganisms in shrub/Sphagnum peatlands. Principal component (PC) scores indicated more utilization of N-containing compounds and carboxylic acids, and less utilization of carbohydrates by microbial communities in shrub/Sphagnum sites. PC scores were much more variable both within and among vegetation types for sites in West Virginia than in New York State, and a greater diversity of C sources were utilized in WV (57±3) than NYS (47±2) peat. Our results suggest a link between microbial respiratory activity and microbial functional composition as they vary among these peatland vegetation types.  相似文献

We compare forest floor microbial communities in pure plots of four tree species (Thuja plicata, Tsuga heterophylla, Pseudotsuga menziesii, and Picea sitchensis) replicated at three sites on Vancouver Island. Microbial communities were characterised through community level physiological profiles (CLPP), and profiling of phospholipid fatty acids (PLFA).Microbial communities from cedar forest floors had higher potential C utilisation than the other species. The F layer of the forest floor under cedar contained significantly higher bacterial biomass (PLFA) than the F layer under the other three tree species. There were differences in microbial communities among the three sites: Upper Klanawa had the highest bacterial biomass and potential C utilisation; this site also had the highest N availability in the forest floors. Forest floor H layers under hemlock and Douglas-fir contained greater biomass of Gram positive, Gram negative bacteria and actinomycetes than F layers based on PLFA, and H layers under spruce contained greater biomass of Gram negative bacteria than F layers. There were no significant differences in bacterial biomass between forest floor layers under cedar. Fungal biomass displayed opposite trends to bacteria and actinomycetes, being lowest in cedar forest floors, and highest in the F layer and at the site with lowest N availability. There were also differences in community composition among species and sites, with cedar forest floors having a much lower fungal:bacterial ratio than spruce, hemlock and Douglas-fir. The least fertile Sarita Lake site had a much greater fungal:bacterial ratio than the more fertile San Juan and Upper Klanawa sites. Forest floor layer had the greatest effect on microbial community structure and potential function, followed by site, and tree species. The similarity in trends among measures of N availability and microbial communities is further evidence that these techniques provide information on microbial communities that is relevant to N cycling processes in the forest floor.  相似文献

We evaluated changes occurring in the rhizosphere microbial communities of Scots pine (Pinus sylvestris L.) due to tree-felling and decrease of the photosynthetic C flow into the soil under field conditions over one growing season. Samples were taken from tree rhizospheres, freshly felled stump rhizospheres and bulk soil. We used culture dependent (CFU counts, community level physiological profiles, CLPPs) and independent methods (fluorogenic MUF-substrates, PLFA pattern and PCR-DGGE) to monitor the microbial communities in soil samples. The numbers of cultivable bacteria and amounts of phosphatase activity in the rhizosphere of trees were significantly higher compared with those in the bulk soil. The organic C consuming community measured by CLPP was stimulated directly after the tree-felling in stump rhizospheres; utilization of the disintegration components of cellulose, hemicellulose and chitin increased. Furthermore, bacterial and fungal biomass as well as chitin decomposers (CFU) increased in the stump rhizosphere. After 11 weeks of tree-felling the stump rhizosphere soluble PO4-P and NH4-N as well as amounts of total C and N began to resemble the concentrations measured in the bulk soil. However, the stump rhizosphere community structure detected by PLFA and PCR-DGGE still resembled that of the tree rhizosphere.  相似文献

The relationship between organic matter decomposition and changes in microbial community structure were investigated in Antarctic soils using 13C-labelled plant materials. Soils with and without labelled Deschampsia antarctica (a native Antarctic grass) were incubated for 42 days and sampled at 0, 7, 14, 21, 28 and 42 days. Changes in microbial community structure were assessed using phospholipid fatty acid analysis (PLFA) and an analysis of the fatty acids associated with the neutral lipid fraction (NLFA). These studies showed that there were no significant changes in PLFA or NLFA profiles over time suggesting no change in microbial community structure during residue decomposition. There was a marked increase however, in ergosterol levels in these soils indicative of growth of the fungal biomass. Analysis of this ergosterol using gas chromatography-mass spectrometry confirmed the transformation of the plant residue by showing the incorporation of 13C-plant C into the ergosterol. This incorporation of 13C into the ergosterol increased over the incubation period. Importantly, these changes associated with fungal growth were not evident in the analysis of either the PLFA or NLFA fractions thus questioning the reliability of such approaches for studying changes in microbial communities associated with the decomposition of plant residues.  相似文献

This study coupled stable isotope probing with phospholipid fatty acid analysis (13C-PLFA) to describe the role of microbial community composition in the short-term processing (i.e., C incorporation into microbial biomass and/or deposition or respiration of C) of root- versus residue-C and, ultimately, in long-term C sequestration in conventional (annual synthetic fertilizer applications), low-input (synthetic fertilizer and cover crop applied in alternating years), and organic (annual composted manure and cover crop additions) maize-tomato (Zea mays - Lycopersicum esculentum) cropping systems. During the maize growing season, we traced 13C-labeled hairy vetch (Vicia dasycarpa) roots and residues into PLFAs extracted from soil microaggregates (53-250 μm) and silt-and-clay (<53 μm) particles. Total PLFA biomass was greatest in the organic (41.4 nmol g−1 soil) and similar between the conventional and low-input systems (31.0 and 30.1 nmol g−1 soil, respectively), with Gram-positive bacterial PLFA dominating the microbial communities in all systems. Although total PLFA-C derived from roots was over four times greater than from residues, relative distributions (mol%) of root- and residue-derived C into the microbial communities were not different among the three cropping systems. Additionally, neither the PLFA profiles nor the amount of root- and residue-C incorporation into the PLFAs of the microaggregates were consistently different when compared with the silt-and-clay particles. More fungal PLFA-C was measured, however, in microaggregates compared with silt-and-clay. The lack of differences between the mol% within the microbial communities of the cropping systems and between the PLFA-C in the microaggregates and the silt-and-clay may have been due to (i) insufficient differences in quality between roots and residues and/or (ii) the high N availability in these N-fertilized cropping systems that augmented the abilities of the microbial communities to process a wide range of substrate qualities. The main implications of this study are that (i) the greater short-term microbial processing of root- than residue-C can be a mechanistic explanation for the higher relative retention of root- over residue-C, but microbial community composition did not influence long-term C sequestration trends in the three cropping systems and (ii) in spite of the similarity between the microbial community profiles of the microaggregates and the silt-and-clay, more C was processed in the microaggregates by fungi, suggesting that the microaggregate is a relatively unique microenvironment for fungal activity.  相似文献

Several biochemical and molecular methods are used to investigate the microbial diversity and changes in microbial community structure in rhizospheres and bulk soils resulting from changes in management. We have compared the effects of plants on the microbial community, using several methods, in three different types of soils. Pots containing soil from three contrasting sites were planted with Lolium perenne (rye grass). Physiological (Biolog), biochemical (PLFA) and molecular (DGGE and TRFLP) fingerprinting methods were employed to study the change in soil microbial communities caused by the growth of rye grass. Different methods of DNA extraction and nested PCR on TRFLP profiles were examined to investigate whether they gave different views of community structure. Molecular methods were used for both fungal and bacterial diversity. Principal component analysis of Biolog data suggested a significant effect of the plants on the microbial community structure. We found significant effects of both soil type and plants on microbial communities in PLFA data. Data from TRFLP of soil bacterial communities showed large effects of soil type and smaller but significant effects of plants. Effects of plant growth on soil fungal communities were measured by TRFLP and DGGE. Multiple Procrustes analysis suggested that both methods gave similar results, with only soil types having a significant effect on fungal communities. However, TRFLP was more discriminatory as it generated more ribotype fragments for each sample than the number of bands detected by DGGE. Neither methods of DNA extraction nor the nested PCR had any effect on the evaluation of soil microbial community structure. In conclusion, the different methods of microbial fingerprinting gave qualitatively similar results when samples were processed consistently and compatible statistical methods used. However, the molecular methods were more discriminatory than the physiological and biochemical approaches. We believe results obtained from this experiment will have a major impact on soil microbial ecology in general and rhizosphere–microbial interaction studies in particular, as we showed that the different fingerprinting methods for microbial communities gave qualitatively similar results.  相似文献

Microbial communities in soil are highly species-rich, recognition of which has led to the view that functional redundancy within communities may buffer many impacts of altered community structure on soil functions. In this study we investigated the impact of long-term (>50 years) exclusion of plant-inputs (bare-fallow treatment) on soil microbial community structure and on the ability of the microbial biomass to mineralise tracer additions of 13C-labelled plant-derived C-substrates. Exclusion of plant-inputs resulted in depletion of soil organic matter (SOM) and a reduction in microbial biomass size. The microbial community structure was also strongly affected, as indicated by the distinct phospholipid fatty acid (PLFA) profiles in bare-fallow and grassland soils. Mineralisation of labile plant-derived substrates was not perturbed by the bare-fallow treatment. The incorporation of labile plant-derived C into PLFA biomarkers was found to differ between soils, reflecting the distinct community structures of the soils and indicating that these substrates were utilised by a broad range of microbial groups. In contrast, the mineralisation of recalcitrant plant-derived substrates was reduced in bare-fallow soil and the fate of substrate-derived C within PLFA biomarkers was, initially, similar between the soils. These results indicate that utilisation of these recalcitrant substrates was a function restricted to specific groups, and that exclusion of plant-derived inputs to soil had reduced the capacity of bare-fallow microbial communities to utilise this substrate type. Therefore, the study suggests that long-term selective pressure on microbial communities, resulting in altered community structure, may also result in altered functional attributes. This structure-function relationship was apparent for utilisation of recalcitrant plant-derived substrates, but not for the more widely distributed attribute of labile C-substrate utilisation.  相似文献

With the growing interest in silvicultural techniques that more closely emulate natural disturbance regimes, there is a need to better understand how partial harvesting affects the soil microbial community in stands with varying ecological characteristics, e.g., tree species composition. Four and a half and 5.5 years post-harvest, we used phospholipid fatty acid (PLFA) and substrate-induced respiration (SIR) analyses to compare the microbial biomass and microbial community structure of forest floors from stands dominated by white spruce (Picea glauca; SPRUCE) or by trembling aspen (Populus tremuloides; ASPEN) and from mixed-species (MIXED) stands in northern Alberta, Canada, that had been clearcut, partial-cut with 20% retention, partial-cut with 50% retention or left uncut (controls). PLFA and SIR analyses revealed that ASPEN forest floors supported a larger microbial biomass with a very different community structure than MIXED or SPRUCE forest floors. The microbial community structure of these soils appeared to be strongly affected by the presence of white spruce and the composition of the understory vegetation. There were no effects of timber harvesting detected within or across stand types on any of the variables measured, with the exception of the PLFA 16:1ω5, which was relatively more abundant in the clearcuts and 50% retention treatments than in the uncut controls, perhaps in response to an increased forest floor pH and grass cover in the disturbed areas. The resilience to timber harvesting of the forest floors from these stands may be the result of efforts to minimize soil disturbance during harvesting and to allow vegetation to regenerate naturally. From the perspective of the forest floor microbial community, partial harvesting does not appear to have any benefit over clearcut harvesting at these boreal forest sites.  相似文献

Re-vegetation of eroded soil restores organic carbon concentrations and improves the physical stability of the soil, which may then extend the range of microhabitats and influence soil microbial activity and functional stability through its effects on soil bacterial community structure. The objectives of this study were (i) to evaluate the restorative effect of re-vegetation on soil physical stability, microbial activity and bacterial community structure; (ii) to examine the effects of soil physical microhabitats on bacterial community structure and diversity and on soil microbial functional stability. Soil samples were collected from an 18-year-old eroded bare soil restored with either Cinnamomum camphora (“Eroded Cc”) or Lespedeza bicolour (“Eroded Lb”). An uneroded soil planted with Pinus massoniana (“Uneroded Pm”) and an eroded bare soil served as references. The effect of microhabitats was assessed by physical destruction with a wet shaking treatment. Soil bacterial community structure and diversity were measured using a terminal restriction fragment length polymorphism (T-RFLP) approach, while soil microbiological stability (resistance and resilience) was determined by measuring short-term (28 days) decomposition rate of added barley (Hordeum vulgare) powder following copper and heat perturbations. The results demonstrated that re-vegetation treatment affected the recovery of physical and biological stability, microbial decomposition and the bacterial community structure. Although the restored soils overshot the Uneroded Pm sample in physical stability, they had lower microbial decomposition and less resilience to copper and heat perturbations than the Uneroded Pm samples. Soil physical destruction by shaking had the same effect on soil physical stability, but different effects on soil microbial functional stability. There were significant effects of vegetation treatment and perturbation type, and interactive effects among vegetation treatment, shaking and perturbation type on bacterial community structure. The destruction of aggregate structure increased resilience of the Eroded Lb sample and also altered its bacterial community structure. Both copper and heat perturbations resulted in significantly different community structure from the unperturbed controls, with a larger effect of copper than heat perturbation. Bacterial diversity (Shannon index) increased following the perturbations, with a more profound effect in the Uneroded Pm sample than in the restored soils. The interactive effects of vegetation treatment and shaking on microbial community and stability suggest that soil aggregation may contribute to the generation of bacterial community structure and mediation of biological stability via the protection afforded by soil organic carbon. Differential effects of re-vegetation treatment suggest that the long-term effects are mediated through changes in the quality and quantity of C inputs to soil.  相似文献

Peatlands still suffer from the scarcity of available data about the characterization and the response to climate forcing of the main oxidative enzymes that occur over the seasons. In the present study, phenoloxidase and peroxidase activities were examined in Sphagnum lawns along a narrow fen-bog gradient under experimental elevated temperatures. We showed that peroxidase activities from Sphagnum mosses were 1000-fold higher than those of phenoloxidases irrespective of seasons and sampling areas. Peroxidase activities increased (+30%) with the rise of air temperatures (an average of 1 °C), while warming did not alter phenoloxidase activities. These results suggest that the monitoring of peroxidase activities in peatlands may represent a suitable and forward indicator of the impact of climate warming on carbon cycle in peatlands.  相似文献

Biological communities differ over time and in space, and in the forest these communities often vary according to trees and tree gaps, mediated by mechanisms that are likely to change over time and as a tree are removed. In this paper we ask the questions: What is the influence of individual trees on soil microbial community structure? Does the soil microbial community change in the short-term when a tree is removed, and does this change depend on the initial influence of the tree? We use phospholipid fatty acid (PLFA) analysis and a geostatistical approach to study effects of trees and tree removal (thinning) on soil microbial community structure in a young boreal Norway spruce (Picea abies) forest. An experiment was setup where half (four) of the included trees were cut and soil was collected prior to (t0) and one month after (t1) tree felling. The samples were collected along two perpendicular transects originating from each of the eight study trees. A tree influence index was calculated for each sample point from the distances to neighbouring trees, weighted by tree diameter. We found that individual trees are important in structuring the soil microbial community as microbial community structure responded to the gradient in tree influence. Also strong spatial structure was found corresponding to the patch structure induced by trees. Changes in microbial community structure before and after tree felling (t0 and t1) was found to differ significantly between felled and non-felled trees: samples from felled trees came to resemble samples with a low value of tree influence and samples from below non-felled trees came to resemble samples with a high value for tree influence. We thus found that soil microbial community structure in a boreal forest is spatially structured by the distribution of single trees, and that soil microbial community structure varies seasonally and is affected by tree removal, in an intricate manner that reflects the initial influence of trees.  相似文献

The effects of coloniser plant species on microbial community growth and composition were investigated on recently deglaciated terrain at Glacier Bay, south-east Alaska. Analysis of microbial communities using phospholipids fatty acid analysis (PLFA) revealed that Alnus and Rhacomitrium had the greatest impact on microbial growth, increasing total PLFA by some 6-7 fold relative to bare soil, whereas Equisetum led to a 5.5 fold increase in total PLFA relative to bare soil. These coloniser species also had significant effects on the composition of their associated microbial communities. Rhacomitrium, Alnus, and Equisetum increased bacterial PLFA, a measure of bacterial biomass, relative to bare soil. Rhacomitrium and Alnus also dramatically increased the concentration of the fungal fatty 18:2ω6 in soil relative to bare soil, by 12-fold and 8-fold, respectively. The net effect of the above changes was a significant increase in the ratio of fungal: bacterial fatty acids in soil associated with Alnus and Rhacomitrium, but not Equisetum. Possible reasons for these effects of particular plants on microbial communities are discussed, as is their significance in relation to the development of microbial communities in relatively sterile, recently deglaciated ground.  相似文献

Altitudinal gradients are useful to study the potential effects of climate change on ecosystems. Historically, studies on elevation gradients have primarily focused on macro-organisms and ecosystem processes, while microorganisms have been mostly ignored despite their ubiquity and functional importance. We studied the temporal (about every two months from June 2008 until May 2009) variation of testate amoeba communities in four Sphagnum-dominated peatlands along a 1300 m elevation gradient in the Swiss Mountains (580–1880 m) in relation to water table depth and hydrochemistry with special focus on dissolved organic carbon (DOC), a useful proxy for changes in C-cycling in peatlands.The lowest site had significantly (P < 0.01) lowest testate amoeba density, species richness. The highest site had highest testate amoeba density (38 ind mg−1 dry mass of Sphagnum). Seasonal fluctuations in testate amoeba species richness and diversity were not consistent among sites but density tended to peak in spring at all sites, autumn in the three highest sites and mid-winter in the upper two sites. In a redundancy analysis (RDA) community structure was more strongly correlated to altitude (33.8% of variance explained in living community) than to soil hydrological and hydro-chemical variables (together explaining 16.2% of variance). In a partial RDA with altitude used as covariable, the four sites were separated by DOP, DOC, DON, pH and average depth to water table.The abundance of high trophic level testate amoeba species (shell-aperture over their body size >0.20; i.e. primarily predators of protists and micro-metazoa) as well as the community size structure increased from lowest to highest elevation (respectively by 3.7× and 6×) and followed the seasonal patterns of total density, while DOC, DON, and DOC/DON decreased with elevation.These results agree with previously reported alteration of peatland microbial food chains in response to experimental warming, suggesting that climate-induced changes in microbial community structure (here a shortening of microbial food chains) represent a mechanism controlling the carbon balance of peatlands.  相似文献

Land usage is a strong determinant of soil microbial community composition and activity, which in turn determine organic matter decomposition rates and decomposition products in soils. Microbial communities in permanently flooded wetlands, such as those created by wetland restoration on Sacramento-San Joaquin Delta islands in California, function under restricted aeration conditions that result in increasing anaerobiosis with depth. It was hypothesized that the change from agricultural management to permanently flooded wetland would alter microbial community composition, increase the amount and reactivity of dissolved organic carbon (DOC) compounds in Delta waters; and have a predominant impact on microbial communities as compared with the effects of other environmental factors including soil type and agricultural management. Based on phospholipid fatty acid (PLFA) analysis, active microbial communities of the restored wetlands were changed significantly from those of the agricultural fields, and wetland microbial communities varied widely with soil depth. The relative abundance of monounsaturated fatty acids decreased with increasing soil depth in both wetland and agricultural profiles, whereas branched fatty acids were relatively more abundant at all soil depths in wetlands as compared to agricultural fields. Decomposition conditions were linked to DOC quantity and quality using fatty acid functional groups to conclude that restricted aeration conditions found in the wetlands were strongly related to production of reactive carbon compounds. But current vegetation may have had an equally important role in determining DOC quality in restored wetlands. In a larger scale analysis, that included data from wetland and agricultural sites on Delta islands and data from two previous studies from the Sacramento Valley, an aeration gradient was defined as the predominant determinant of active microbial communities across soil types and land usage.  相似文献

Soil management practices that result in increased soil C also impact soil microbial biomass and community structure. In this study, the effects of dairy manure applications and inorganic N fertilizer on microbial biomass and microbial community composition were determined. Treatments examined were a control with no nutrient additions (CT), ammonium nitrate at 218 kg N ha−1 (AN), and manure N rates of 252 kg manure-N ha−1 (LM) and 504 kg manure-N ha−1 (HM). All plots were no-till cropped to silage corn (Zea mays, L. Merr) followed by a Crimson clover (Trifolium incarnatum, L.)/annual ryegrass (Lolium multiflorum, Lam.) winter cover crop. Treatments were applied yearly, with two-thirds of the N applied in late April or early May, and the remainder applied in September. Soil samples (0–5, 5–10, and 10–15 cm) were taken in March 1996, prior to the spring nutrient application. Polar lipid fatty acid (PLFA) analysis was used to assess changes in microbial biomass and community structure. Significantly greater soil C, N and microbial biomass in the 0–5 cm depth were observed under both manure treatments than in the CT and AN treatments. There was also a definable shift in the microbial community composition of the surface soils (0–5cm). Typical Gram-negative bacteria PLFA biomarkers were 15 and 27% higher in the LM and HM treatments than in the control. The AN treatment resulted in a 15% decrease in these PLFA compared with the control. Factor analysis of the polar lipid fatty acid profiles from all treatments revealed that the two manure amendments were correlated and could be described by a single factor comprised of typical Gram-negative bacterial biomarkers. The AN treatments from all three depths were also correlated and were described by a second factor comprised of typical Gram-positive bacterial biomarkers. These results demonstrate that soil management practices, such as manuring, that result in accumulations of organic carbon will result in increased microbial biomass and changes in community structure.  相似文献
