Summary Cultivars of Cucurbita pepo were analyzed for their isozyme phenotypes by horizontal starch gel electrophoresis. Considerable allozymic variation was observed between cultivars, especially in the aspartate aminotransferase and malate dehydrogenase isozyme systems. Each of the five fruit types represented in the 21 cultivars tested (zucchini, pumpkin, spaghetti, acorn, scallop and yellow straightneck) could be distinguished by specific allozymes or combination of allozymes. Cultivars within a fruit type gave very similar allozyme phenotypes and often could not be distinguished on the basis of the 6 assays used. Despite the outcrossing nature of the species, allozyme polymorphism within most cultivars was low and did not seriously interfere with the analysis. Approximately half of the hybrid lines tested gave heterozygous phenotypes at one or more loci, indicating that such loci could be used to screen for the percentage of hybrid seed obtained from crosses.  相似文献

利用超薄平板微型聚丙烯酰胺凝胶的等电聚焦电泳技术,对286份梨材料进行了等位酶遗传变异分析。在8个酶系中共检测到19个清晰位点和82个等位基因,19个位点均为多态位点,位点最大等位基因数为6,体现出梨丰富的遗传种质多样性;不同的居群具有特有等位基因;通过82个等位基因可以将286份材料完全区分开,表明等位酶基因型指纹可用作梨品种区分与鉴定的依据。  相似文献

Summary Sixty Lolium perenne populations were screened for allozyme diversity at five loci. Objective was to determine whether allozyme diversity could be used as selection criterion for genebank accessions of L. perenne. Subsampling of locations was tested with allozyme analysis to determine whether genetically different populations could be collected at one location. Correlations between allelic frequencies and environmental factors and morphological data were established, to find ecogeographical patterns in the observed variation. Results indicated that with few examples each allelic variant could be observed in each population screened; very few unique alleles were found. Differences between populations were largely due to differences in allelic frequencies. Few correlations were found with environmental factors and morphological data. For some allele frequencies a north-south cline was observed. Generally, allozymic data of the five screened loci did not appear to be useful for the selection of accessions for genebank storage. Significant genotypic differences between populations collected at one location could be established. In general these results agreed with earlier results concerning phenotypic variation in the same populations.  相似文献

Summary Horizontal starch gel electrophoresis was done to assay 10 enzyme systems in root tips of 14 Populus deltoides x P. nigra clones developed by controlled hybridization and selected in North America, and some clones sharing one or both parents. The genotypes of the clones were determined for 31 loci coding for 10 enzyme systems. The interclonal allozyme variability was controlled by 12 loci. Each of the 14 clones had unique 12-locus genotypes, thus could be distinguished from each other. The clones differed from each other on an average of 4.2 loci. The first two Principal Components from Principal Component Analysis of the clonal allozyme genotype data accounted for 48% of the total variation in the 12 variant loci. 6-Pgd-3, Per-2, 6-Pgd-1, Aco-1 and Per-1 were found to be the most discriminating loci for the clones. The ordination of the clones on Principal Component axes 1 and 2 was in general agreement with the origin of the clones.  相似文献

Genetic variability within and among 14 Chinese natural populations of Lespedeza Michx. Used mainly for wildlife food and cover and for erosion control was evaluated on the basis of morphological characteristics, allozyme markers and random amplified polymorphic DNAs (RAPDs). All methods disclosed a high degree of allelic heterogeneity among samples, also revealing a high degree of gene diversity. Genetic variation among populations was higher for RAPD than for enzyme loci. Correlation between morphological, allozyme and RAPD distance matrices were calculated by Mantel’ tests. There was a little disparity between the three level's distances on population relationships. However, there is a weak significant correlation (P < 0.001) between morphology and allozyme distances as well as morphology and RAPD distances, and the test of this correlation is very high.  相似文献

利用过氧化物同工酶对云南红壤区的几种紫花苜蓿杂交组合进行初步筛选,以期为该地区紫花苜蓿的引种驯化工作提供理论依据,从而获得有价值的杂交后代。对各杂交组合亲本及其后代的过氧化物酶酶谱表现分析结果表明,杂交过程中发生了过氧化物酶位点的遗传重组,从而在F1代产生了酶谱表现与亲本不同和相同的两类个体;亲本及其后代有3条共有酶带;亲本中,爱博、射手2个栽培种比野生种和AC-3的酶谱表现好;过氧化物酶的遗传背景差异较大的杂交组合有可能通过遗传物质的交流产生有利的性状重组,对其进行筛选,可获得有价值的育种材料。  相似文献

Summary Seedling resistance to wheat stem rust was determined in populations of wild emmer wheat, Triticum dicoccoides, and characterized by means of ecological factors and allozyme genotypes. Reactions to wheat stem rust were studied in 102 single plant accessions of T. dicoccoides from ten populations by inoculation with Puccinia graminis tritici race 14, isolate GSR-739. Six populations displayed different degrees of response polymorphism with reactions ranging from high resistance to complete susceptibility, whereas four populations contained only susceptible plants. In some of the accession, unexplained intrasib variation in resistance and intraplant variation of infection-types were found. Resistance to stem rust was negatively correlated with two ecological factors, altitude and number of Sharav (hot-dry) days which are unfavorable to disease development. Variation in stem rust response was shown to exist in ecogeographic regions where climatic variables enhanced the development of the fungus, conceivably maintained by natural selection. Likewise, allozyme genotypes, single or in multiple loci combinations, appeared to be associated with resistance or susceptibility to rust. Such association need to be verified by genetic studies in order to become established as useful markers.  相似文献

Summary Seedlings of date palm (Phoenix dactylifera L.), obtained from seven cultivars crossed with two males, were analyzed by polyacrylamide gel electrophoresis for esterase (EST), glutamate oxaloacetate transaminase (GOT), endopeptidase (ENP) and alcohol dehydrogenase (ADH) polymorphisms. Eleven, eight, five and two phenotypes were revealed for the enzymes tested, respectively. Seedlings of F1 populations derived from Bayoud (Fusarium)-resistant and low fruit quality cultivars were characterized by a high electrophoretic polymorphism, when compared with progenies of Bayoud-susceptible and high fruit quality cultivars. In almost all cases, the most frequent electrophoretic phenotypes scored for each enzyme in different F1 populations, were similar to those of the corresponding parent cultivars. Heterozygous phenotypes have been found for, at least, 3 loci; Got-2, Est-1 and Enp, indicating that such loci could be used to screen for hybrid seedlings. The expected Mendelian segregation of allozymes has been observed for these 3 loci, in many F1 populations. It seems that progenies of Bayoud-resistant cultivars are characterized by a high level of electrophoretic polymorphism. The estimation of this index and the search for genetic linkage with segregating allozymes, may be biochemical criteria useful as an aid in distinguishing date palm seedling populations derived from Bayoud-resistant cultivars and suitable for breeding programs.  相似文献

采用常规聚丙烯酰胺凝胶电泳技术,对海南吊罗山和霸王岭两个国家级自然保护区野生荔枝等位酶遗传多样性进行了研究。13个酶系统、18个酶位点的检测结果表明,两个自然保护区野生荔枝具有较低的居群间遗传变异水平和较高的居群内遗传变异。吊罗山保护区野生荔枝的遗传多样性参数为:P=61.1%,A=2.05,He=0.27。霸王岭居群野生荔枝的遗传多样性参数为:P=61.1%,A=2.05,He=0.28。两个群体的遗传分化极小,GST=0.047.居群间遗传一致度较高I=0.977。共检测到7个稀有等位基因。两个自然保护区的野生荔枝均有各自特有的稀有等位基因,都应采取措施予以保护。  相似文献

The need for precise characterisation of crop materials has assumed importance in any established sui generis system for effective protection of plant varieties. Considering this requirement, 37 Indian maize inbreds were analysed for allozyme variations. Seven enzyme systems comprising 16 isozyme loci were analysed using starch gel electrophoresis. The allelic variants were designated using the well-characterised maize inbred Mo 17 as a standard. A total of 29 alleles were scored at the 16-isozyme loci studied. Malate dehydrogenase (Mdh), acid phosphatase (Acp), phosphoglucoisomerase (Pgi), 6-phosphogluconate dehydrogenase (Pad) and isocitrate dehydrogenase (Idh) were the most informative isozyme systems for characterisation of the maize inbreds. These enzyme systems were also useful in detecting and quantifying the extent of `off-types' in each seed lot analysed. The percent off-types (inclusive of the admixtures and segregates) were as high as 47 to 50 percent in seed lots of a few of the inbreds. The results of the study were also useful in studying genetic diversity among the inbreds. An UPGMA based cluster analysis of the allozyme data revealed presence of only moderate genetic diversity among the Indian maize inbreds. This is of significance in crop yield stability as these inbreds have been used extensively in the production of F1 hybrids occupying large area under maize cultivation in India. Summary Grain amaranth populations from their centers of origin in the New World had shown a pattern of allozyme variation that suggested most landraces to be highly homozygous mixtures of genotypes. To compare this pattern of variation with the variation for morphological traits, 15 selfed families from each of six populations were grown in a replicated field experiment. Four pigmentation traits known to be smiply inherited were scored along 18 other morphological traits. Populations varied for the amount of polymorphism for marker loci, and exhibited little heterozygosity. Analysis of variance for the quantitative traits showed significant interpopulation differences for each of the observed characters. Populations differed for the number of metric traits showing significant between-family differences for just one of the metric traits whereas another had between-family differences for all ten. These results suggested high levels of homozygosity within these landraces; thus, variation for quantitative traits conformed well with the allozyme variation patterns.Stepwise multiple regression of all characters on yield as the dependent variable was used to compare the relative contributions of specific characters to yield within individual populations. Plant height, days to flowering, and leaf length were included in the regression equations for 6, 5, and 5, of the populations, respectively. Days to floweing was negatively correlated with yield, while plant height and leaf length were positively correlated. These correlations suggest the potential for developing early flowering, high yielding cultivars having short stature, selected by breaking its correlation with yield. Several breeding strategies based on these findings on genetic resources are discussed briefly.  相似文献

Summary Genetic variation was studied using enzyme polymorphism at 8 loci in ancient olive trees. These were being cultivated in 10 sites along a transect from North (Pre-Rif) to South (Anti-Atlas), involving the main ecological areas where the species is cultivated in Morocco. For the 328 trees studied, 16 alleles and 68 multilocus genotypes, of which one was present in more than half the individuals analysed, were detected. Eighty seven per cent of the total genetic diversity was attributable to within site variation and showed a high proportion of local genotypes. The highest values for both genotype number and heterozygosity were observed in the South of Morocco. Such high variation may be due to the partial domestication of olive material which may be derived from crosses between cultivars or between cultivars and feral or wild olive (oleaster) trees growing frequently in the Southern region. Genetic diversity in Moroccan olive, constitutes an important genetic resource which must be conserved for further breeding.  相似文献

Summary A survey of allozyme polymorphism at 11 loci was carried out on 439 accessions from the genus Lens. This comprised 153 Lens culinaris subsp. orientalis, 35 L. odemensis, 117 L. ervoides, 32 L. nigricans, 2 of a differentiated cytotype of L. nigricans and 100 landrace accessions of the cultivated lentil (L. culinaris subsp. culinaris), from 10 different countries. The aim of the survey was to determine intra-specific genetic diversity and species relationships, based on phylogenetic and phenetic analyses, particularly regarding the position of L. odemensis and the differentiated cytotype of L. nigricans. Diversity was described by three statistics. The level of diversity in the cultivated taxon was lower than in any of the wild species according to two of these statistics, the percentage of polymorphic loci and mean number of alleles per locus. For the third measure (Nei's mean genetic diversity) it was only greater than L. ervoides. Genetic diversity statistics of the wild species indicated differences in the nature of between-population genetic diversity within the different taxa. Phylogenetic analysis suggests that L. odemensis and L. ervoides evolved from a common ancestor, and L. culinaris subsp. orientalis subsequently evolved from L. odemensis. Phenetic analysis, however, places L. odemensis closer to L. culinaris subsp. orientalis than to L. ervoides. Nei's mean genetic distance of L. odemensis from both L. culinaris subsp. culinaris (0.204) and L. culinaris subsp. orientalis (0.110) was greater than the distance between them (0.062). This evidence is not conclusive in determining whether L. odemensis should retain its specific status. Further crossability studies should be carried out on a range of genotypes to assess the potential for gene flow. The evidence presented shows the differentiated cytotype of L. nigricans to be quite distinct from other L. nigricans accessions, both phenetically and phylogenetically. This indicates that the differentiated cytotype of L. nigricans may constitute a new taxon. Discriminant function analysis reveals that isozymes may be useful in validating species classification.  相似文献

Early and accurate identification of almond [Prunus dulcis (Mill.) D.A. Webb] cultivars is critical to commercial growers and nurseries. Previously published simple sequence repeat loci were examined for their ability to distinguish commonly grown almond cultivars. Twelve highly polymorphic loci were selected for their ability to uniquely identify a set of 18 almond cultivars commonly grown in California, many of which are closely related. These markers also allow an accurate assessment of parent/progeny relationships among cultivars. This system can reliably identify at an early stage of development all major California almond cultivars in current production.  相似文献

Summary Morphological, isozyme and random amplified polymorphic DNA (RAPD) markers were used to estimate genetic variation within and between cultivars of red clover (Trifolium pratense L.), an important temperate forage legume. Two cultivars of red clover, Essi from Europe and Ottawa from Canada, were evaluated. Six monogenic morphological characters were observed for 80 plants from each of these two cultivars. All six morphological loci were polymorphic in the cultivar Essi whereas only four loci were polymorphic in the cultivar Ottawa. Forty plants from each cultivar were assayed for isozyme markers. A total of 21 enzyme-coding loci with 43 alleles was detected using twelve enzyme systems. Thirteen and nine of these loci were polymorphic in Essi and Ottawa, respectively. The mean number of alleles per locus was 1.81 in Essi and 1.67 in Ottawa. Seventeen random 10-mer primers were screened for RAPD markers. Nine primers which gave clear and consistent amplified products were used to assay 20 individuals from each cultivar. Each primer gave from 7 to 20 amplified bands with an average of 14.8 bands per primer. One hundred and eight of 116 putative loci were polymorphic in Essi and 90 of 98 loci were polymorphic in Ottawa. High within-cultivar variation was observed in both cultivars using both isozyme and RAPD markers. This high polymorphism makes these markers useful for germplasm characterization and genetic studies in red clover.  相似文献

以新疆截止2012年审定的23份新彩棉品种为材料,利用SSR标记进行DNA指纹图谱的构建和遗传多样性分析。从5000对SSR引物中,挑选出多态性高、稳定性好、均匀分布在棉花26条染色体上的52对引物,在23份新彩棉品种中筛选出核心引物47对,SSR扩增检测到多态性基因型位点数共计162个,每个标记检测到的基因型位点数在2~7之间,平均为3.45个;引物多态信息量(PIC)值介于0.4537~0.8686之间,平均值为0.7096。结果显示:在23份新彩棉品种中,14份品种采用特异或特征引物可以一次性区分开,其余9份品种需要采用引物组合来实现区别该品种与其他品种。最少选用18对特异引物及组合引物就可以完全区分开新彩棉1~23号品种。利用18对SSR标记构建了新彩棉1号至23号品种的指纹图谱。利用NTSYS-pcV2.10软件聚类分析表明:23个新彩棉品种遗传相似系数变化范围是0.3781~0.9298,平均为0.5511,表明新彩棉品种之间存在着丰富的遗传多样性。  相似文献

Genetic variation among five elite winter barley cultivars (H. vulgare L.) currently grown in Bulgaria was assessed at the molecular level using restriction fragment length polymorphism (RFLP) and randomly amplified polymorphic DNA (RAPD) markers. The present study sampled RFLPs in four well characterized multigene families in barley: the seed storage protein loci; the 18S, 5.8S and 26S ribosomal DNA loci; the loci coding for 5S ribosomal RNA and the loci coding subunit α of ATP-A complex in the mitochondrial genome. RFLPs were detected in three out of five investigated chromosomal loci in the barley cultivars studied. RAPD assay using arbitrary 10-base primers was applied to generate amplified length polymorphic markers in barley. Overall a total of 15 polymorphic phenotypes were found among the studied barley cultivars by using 11 out of 25 tested primers. All RAPDs were considered as dominant genetic markers except for two, where PCR and Southern blot analysis indicated the presence of codominant amplification products. Five RAPD polymorphisms in F1 and F2 progenies of the cross between Alpha and Obzor were inherited in Mendelian fashion. The determined values for the genetic variation proved a high genetic similarity among the tested cultivars. Genetic similarity (GS) calculated from RFLP and RAPD data ranged from 0.888 to 0.997 with a mean GS – 0.933. This paper describes the relative efficiency of three marker systems, RAPD, ISSR, and AFLP, in terms of fingerprinting 14 rice genotypes consisting of seven temperatejaponica rice cultivars, three indica near-isogenic lines, three indica introgression lines, and one breeding line of japonica type adapted to high-altitude areas of the tropics with cold tolerance genes. Fourteen RAPD, 21 ISSR, and 8 AFLP primers could produce 970 loci, with the highest average number of loci (92.5) generated by AFLP. Although polymorphic bands in the genotypes were detected by all marker assays, the AFLP assay discriminated the genotypes effectively with a robust discriminating power (0.99), followed by ISSR (0.76) and RAPD (0.61). While significant polymorphism was detected among the genotypes of japonica and indica through analysis of molecular variance (AMOVA), relatively low polymorphism was detected within the genotypes of japonica rice cultivars. The correlation coefficients of similarity were significant for the three marker systems used, but only the AFLP assay effectively differentiated all tested rice lines. Fingerprinting of backcross-derived resistant progenies using ISSR and AFLP markers easily detected progenies having a maximum rate of recovery for the recurrent parent genome and suggested that our fingerprinting approach adopting the ‘undefined-element-amplifying’ DNA marker system is suitable for incorporating useful alleles from the indica donor genome into the genome of temperate japonica rice cultivars with the least impact of deleterious linkage drag.  相似文献

Summary The morphological and isozyme variation was studied in 22 accessions of Solanum chacoense from Paraguay and Argentina. Clear geographic groups were identified through the use of multivariate analyses. S. chacoense from mountain sites in Argentina could be readily distinguished from plains forms from Paraguay, on the basis of several correlated morphological characters. Three isozyme systems, namely phosphoglucose isomerase (PGI), glutamate-oxaloacetate transaminase (GOT) and peroxidase (PRX) were studied using starch gel electrophoresis. The banding patterns indicated that for each isozyme there were several loci, which were polymorphic. A genetic interpretation of one of the PGI loci was made, and indices of genetic diversity and genetic identity calculated. Principal components analysis, cluster analysis and genetic diversity indicated a close relationship between the geographical groups. These results are discussed in the context of in situ genetic conservation.  相似文献

Summary Isozyme patterns were used to characterize ten commercial rape seed (Brassica napus) cultivars. Extracts of cotyledons were made 4–7 days after germination and separated by electrophoresis on starch gels. A sample of more than 100 plants of each variety was analysed and stained for 4 polymorphic enzyme loci (Lap, Gpi, Acon, and Sdh). Therewith, it was possible to distinguish all ten rape varieties qualitatively by at least one typical enzyme pattern. Further, it could be demonstrated that all pairs of varieties showed clear qualitative differences in isozyme patterns, when only tow loci were screened (Acon, Sdh). Using 2 test of homogeneity all pairs of varieties differ significantly in their frequencies of isozyme patterns for Acon and in all but one for Sdh.  相似文献
