The effects of an intraperitoneal hormone injection of gonadotropin‐releasing hormone agonist (D‐Ala6, Pro9‐NEt GnRHa) alone or in combination with a dopamine antagonist, domperidone (DOM), on ovulation induction in yellow catfish Pelteobagrus fulvidraco were tested. The hormone treatments were as follows: 6 mg kg−1 body weight (BW) of carp pituitary extract as a positive control, GnRHa 10, 20, 40 and 80 μg kg−1 BW and a combination of GnRHa and DOM as follows: 10 μg+5 mg, 20 μg+10 mg, 40 μg+20 mg and 80 μg+40 mg kg−1 BW. Physiological saline (0.7% NaCl) was used as a negative control. Significant differences in the ovulation ratio, latency period and ovulation index (OI) were observed among treatments (P<0.05). The combination of GnRHa and DOM at doses of 40 μg+20 mg kg−1 BW had higher values of the ovulation ratio and OI, and a shorter latency period compared with other treatments. The highest OI in GnRHa treatments was only 56.67%, suggesting a dopaminergic tone on gonadotropin secretion in this fish at the pre‐ovulatory stage. Therefore, ovulation can be successfully induced in yellow catfish with 40 μg kg−1 GnRHa+20 mg kg−1 DOM without affecting the egg quality.  相似文献

The effects of a single injection of mammalian superactive analogue [d ‐Ser(tBu)6,Pro9‐NEt]‐LHRHa (20 μg/ kg?1) combined with the dopamine antagonist, haloperidol (HAL, 0.5 mg kg?1), for induction of ovulation in the koi carp broodstocks were determined under routine hatchery conditions. The results were compared with classic carp pituitary extract (CPE, double injection) application (water temperature 22 °C). Physiological saline (0.7% NaCl)‐injected fish were used as a control group and no ovulation occurred in this group. The spawning ratio was high in the LHRHa+HAL treatment group and in the CPE treatment group (6/7 and 5/7 respectively). The latency period was 14–16 h in the LHRHa+HAL treatment group and 12–14 h in the CPE treatment group (after the second injection). There was no difference between the two ovulating groups with respect to the spawning index (the weight of eggs as a percentage of female body weight) and fertilization rate of eggs (P>0.05). As a result, ovulation can be induced successfully in koi carp broodstocks with 20 μg kg?1 [d ‐Ser(tBu)6,Pro9‐NEt]‐LHRHa+0.5 mg kg?1 HAL treatment in a single injection without decreasing the egg quality. Application of this combination can be useful for hatchery and broodstock management in koi carp culture.  相似文献

The traditional GnRH analogue treatments applying the chemicals in pure form proved to be ineffective in inducing ovulation in northern pike (Esox lucius L). Neither mGnRHa ([D‐Ala6, Pro9NEt]‐mGnRH) nor sGnRHa ([D‐Arg6, Pro9‐Net]‐sGnRH) administered alone or together with pimozide (mGnRHa), metoclopramide (mGnRHa) or domperidone (sGnRHa) induced ovulation in females, whereas in groups receiving a carp pituitary injection most females ovulated. Spawning‐inducing agent Dagin did not induce ovulation, whereas all but one female ovulated in the carp pituitary‐treated group. Treatment with another preparation, Ovaprim, resulted in similar or lower ovulation ratio than treatment with carp pituitary. After the Ovaprim treatment, time to ovulation was not as predictable as after the carp pituitary injection. The mean fertilization rate was relatively low and similar in the groups treated with Ovaprim (54.7 ± 12.3% and 58.7 ± 19.1% for the first and second experiment respectively) and with carp pituitary (53.7 ± 10.5% and 58.9 ± 14.9%). The mean pseudogonadosomatic index (PGSI) was also similar between the Ovaprim‐treated group (14.5 ± 6.1%) and the carp pituitary‐treated one (17.9 ± 4.1%). In the present experiments, treatment with Ovaprim was less effective than that with carp pituitary.  相似文献

The effects of intraperitoneal injections of [D-Ala6, Pro9-N-ethylaminde]-luteinizing hormone releasing hormone (LHRH-A) and pimozide (PIM), a dopamine receptor antagonist, on ovulation in loach were investigated. LHRH-A and PIM administered separately were ineffective in inducing ovulation. However, injections of PIM and LHRH-A simultaneously or the injection of PIM 2.5–3 h prior to LHRH-A were highly effective means of inducing ovulation. The simultaneous injection of PIM (1.0 μg/g body wt) and LHRH-A (0.05 μg/g body wt) resulted in a greater ovulatory response than injection of carp pituitary extract (1 pituitary/fish).  相似文献

The results of reproduction of females from Lithuanian strain B carp after ovulation stimulation with carp pituitary homogenate (CPH; 0.3+2.7 mg kg?1; group I), Ovopel (1/5+1 pellet kg?1; group II) or [d ‐Tle6, ProNHEt9] GnRH‐a (Lecirelin) with metoclopramide (15 μg kg?1+10 mg kg?1 respectively; group III) were investigated. The lowest percentage of spawning females (71%) was recorded in the group treated with CPH. In case of Ovopel or Lecirelin induced ovulation, 86% of females spawned. No statistically significant effect of the ovulation stimulator (group) on the weight of eggs was found; however, the highest mean weight of eggs (expressed both in grams and in the percentage of female body weight) was recorded for the group treated with Ovopel (1400 g and 13%). After the treatment with CPH or Lecirelin, the weight of eggs was 1140 g (11%) and 1100 g (10%) respectively. The ovulation stimulator significantly affected the percentage of live embryos after 36 and 48 h incubation of eggs (P≤0.05; P≤0.01). After treatment with [d ‐Tle6, ProNHEt9] GnRH‐a, eggs of the best quality were obtained and after 36 and 48 h incubation the mean percentages of live embryos were significantly higher than the means calculated for the remaining two groups. No statistically significant differences were found between the percentage of living embryos after 36 and 48 h incubation of groups I and II.  相似文献

Trials on induced breeding of the native Amazonian fish tambaqui, Colossoma macropomum (Cuvier), using CPE and HCG treatments, were carried out during 1992-93 at the Fish Culture Station in Natal, Brazil. Final maturation, ovulation and spermiation of tambaqui broodstock, reared in earthen ponds, were induced by injecting heteroplastic carp pituitary gland extracts (CPE) and human chorionic gonadotrophin (HCG) separately. The female spawners received double injections and the males received a single dose. Females were tranquillized using quinaldine and their genital openings were sutured before the resolving injection was administered. Latent period between the resolving injection and ovulation varied from 6 to 11 h according to the prevailing water temperature. Milt was obtained by extrusion. Stripping and dry fertilization of the eggs produced viable larvae. Relative fecundity was estimated at 140 000 eggs kg-1. Mean fertilization and hatching rates varied from 50% to 70% and from 60% to 80%, respectively.  相似文献

Repeated injections of salmon pituitary extract (20 mg per fish per week) induced vitellogenesis in feminized, cultivated Japanese eels (Anguilla japonica). Oocytes were attained at the migratory nucleus stage after 11 or 12 injections. Addition of 17,20-dihydroxy-4-pregnen-3-one (DHP) into the incubation medium induced germinal vesicle breakdown (GVBD) in the oocytes at the migratory nucleus stage. An injection of DHP (2 µg g-1 BW), given 24h after an injection of salmon pituitary extract (20 mg fish-1), succeeded in inducing maturation and ovulation in females which contained occytes at the migratory nucleus stage. Most fish ovulated 15–18h following the DHP injection. Eggs that were ovulated within 15h after the DHP injection showed high fertility and hatchability, but eggs ovulated 18 or 21h after the DHP injection, showed considerably lower fertility and hatchability. A delay between ovulation and stripping of the eggs rapidly decreased both the fertility and hatchability within 6–9h after ovulation, indicating that artificial fertilization must be carried out immediately after ovulation. Repeated injections of human chorionic gonadotropin (hCG) at a concentration of 1 IU g-1 BW week-1 induced spermatogenesis, spermiation, and the acquisition of potential for sperm motility in cultivated males. Most males spermiated after the fifth or sixth injection of hCG, and the milt weight gradually increased and remained constant (1–2 g) from the 11th to 31th injection. Sperm motility peaked 24h after each weekly injection, and decreased from the 3rd day after the injection. Potassium ions are an essential constituent for the maintenance of motility in the eel spermatozoa. Artificial seminal plasma containing 15.2 mM KCl is applicable as a milt diluent. Using these techniques developed for female and male eels, we have succeeded in obtaining many fertilized eggs from cultivated eels.  相似文献

Ovulation was stimulated in carp females of Polish and Hungarian provenance using injections of ( d ‐Ala6) GnRH ProNHEt (Kobarelin) (20 μg kg?1) with metoclopramide (10 mg kg?1) or carp pituitary (3 mg kg?1). For estimating the effects of the substance stimulating ovulation and of the female provenance, analysis of variance was carried out using the least squares method. It was found that the stimulating substance, provenance and interaction of these factors did not significantly affect the weight of eggs obtained. On the other hand, the stimulating substance, provenance and interaction had a highly significant (P ⩽ 0.0001) effect on the percentages of fertilization and live embryos. Females of Hungarian provenance treated with Kobarelin yielded eggs of much better quality but a smaller weight than after the pituitary treatment. In the case of carp females of Polish provenance, a yield of eggs of smaller weight and very poor quality was noted compared with the weight and quality of eggs obtained after hypophysation.  相似文献

Aquaculture of Spinibarbus denticulatus (Oshima, 1926), a fish species indigenous to North Vietnam and Eastern China, is constrained by lack of fingerlings for stocking ponds and cages. As these fish do not naturally breed in captivity, carp pituitary extract (CPE), luteinizing hormone-releasing hormone analogue (LHRHa) with domperidone (DOM) and human chorionic gonadotropin (HCG) were administered at various doses to induce ovulation. A first set of experiments evaluated the response to LHRHa (30, 40 and 50 μg kg−1) with or without DOM (10 mg kg−1), CPE (20, 30 and 40 mg kg−1) and HCG (3000, 4000 and 5000 IU kg−1). A second set of experiments evaluated the dose response to LHRHa (30, 35, 40 and 50 μg kg−1) primed with 6 mg kg−1 of CPE, and HCG (3000, 3500, 4000, 5000 IU kg−1) primed with 6 mg kg−1 of CPE. The treatment groups were compared with each other and the control (injected with 0.9% saline solution). Only 25% and 50% ovulation resulted when treated with LHRHa at 40 and 50 μg kg−1, whereas 100% ovulation was achieved with an addition of DOM to LHRHa. Both 30 and 40 mg kg−1 CPE induced 100% ovulation. However, HCG (4000 and 5000 IU kg−1) induced ovulation in only 33% of females. When primed with CPE, the minimum dose of LHRHa required was 35 μg kg−1 to achieve 70% ovulation. Priming HCG with CPE also resulted in 100% ovulation, and the minimum effective dose of HCG to induce ovulation was 3500 IU kg−1 with 60% ovulation. Fertilization and hatch rates observed in this study with different hormonal stimulation were high (80–93%). The results indicate that while the use of combined hormone strategy has no apparent advantage over a single hormone strategy, LHRHa+DOM (40 μg kg−1+10.0 mg kg−1) and CPE (30 mg kg−1) are most effective in consistently inducing ovulation and thus can be used for commercial hatchery production of S. denticulatus larvae.  相似文献

Wild female catfish Silurus asotus (Linnaeus, 1758) were injected with domperidone (DOM) alone, [d ‐Ala6, Pro9 Net]‐luteinizing hormone‐releasing hormone (LHRH‐A) alone once or twice, LHRH‐A plus DOM once or twice simultaneously at 6‐h intervals, LHRH‐A plus carp pituitary extract (CPE) twice simultaneously at 6‐h intervals and LHRH‐A plus human chorionic gonadotropin (HCG) twice simultaneously 6 h apart respectively. The results indicated that injection of LHRH‐A at a dosage of 0.01–0.02 μg g?1 body weight (BWt) alone induced a low but significant increase in serum gonadotropin (GtH) (P<0.05) and resulted in a very low ovulation rate, while DOM at a dosage of 5 μg g?1 BWt alone did not induce an increase in the serum GtH levels and ovulation; in contrast, LHRH‐A at a dosage of 0.01 μg g?1 BWt plus DOM at a dosage of 5 μg g?1 BWt (termed the Linpe technique) increased the serum GtH (P<0.05) significantly and induced an ovulatory rate of 100%, while LHRH‐A plus CPE or HCG resulted in an increase in the serum GtH (P<0.05) and high ovulatory rate, although the latency period was longer when fish were given LHRH‐A plus HCG or CPE.  相似文献

To induce synchronized ovulation, migrating wild Caspian brown trout (Salmo trutta caspius) females were treated with two interperitoneal injections of Des‐Gly10, d ‐Ala6 LHRH (LHRHa), given 3 days apart. Two injections of 100 μg kg?1 body weight of this hormone effectively induced ovulation. Within 27 days from the second injection, all fish injected with 100 μg kg?1 LHRHa had ovulated compared with 54.5% of the controls. The mean time to ovulation was reduced significantly (P<0.05) from 31.67±4.84 days in control fish and 28.83±7.31 days in sham‐treated fish to 16.36±1.61 days in fish injected with 100 μg kg?1 LHRHa. The fertilization rate in 50 and 100 μg kg?1 LHRHa‐injected fish was significantly lower than that in the control fish (P<0.05). In fish injected with 50 and 100 μg kg?1 LHRHa, significant (P<0.05) changes in testosterone (T) and 17α‐hydroxyprogestrone (OHP) levels were observed. After the second LHRHa injection, the fish injected with 100 μg kg?1 showed the highest serum levels of testosterone and OHP. These results demonstrate that the use of LHRHa can effectively reduce the mean time to ovulation and induce synchronized ovulation in Caspian brown trout.  相似文献

Cyprinid herpesvirus 3 (CyHV‐3) or koi herpesvirus (KHV) is a virulent viral infection in common carp and koi. The disease has caused global epizootic and economic loss in fish aquaculture and in the wild. Clinacanthus nutans (Burm. f.) Lindau is a well‐known medicinal plant used in Thai traditional medicine. Virucidal effects of the plant extract against human herpes simplex virus have been reported. In this study, C. nutans crude extract was tested for antiviral activities against CyHV‐3 in koi carp. Results showed effective antiviral activity against CyHV‐3 pre‐ and post‐infection. The 50% lethal concentration (LC50) of extract was higher than 5 mg/ml. The 50% effective dose (ED50) was 0.99 mg/ml, 0.78 mg/ml, 0.75 mg/ml and 0.71 mg/ml at 1, 2, 3 and 4 hr pre‐infection, respectively. The ED50 from post‐infection tests was 2.05 mg/ml and 2.34 mg/ml at 0 and 24 hr, respectively. These results demonstrated that crude extract expressed antiviral activity against CyHV‐3 and can be applied as a therapeutic agent in common carp and koi aquaculture.  相似文献

The study explored the effects of ethyl ether extract, ethyl acetate extract (EAE), acetone extract, ethanol extract and aqueous extract of Angelica sinensis (EAs) and ethoxyquin (EQ) on lipid oxidation in erythrocytes and growth, digestive, absorptive and antioxidant capacity in carp (Cyprinus carpio var. Jian). The results indicated that EAs and EQ respectively at the concentrations of 0.5 and 0.25 mg/ml suppressed lipid oxidation by decreasing the generation of ROS and restoring the activities of antioxidants in hydroxyl radical‐treated erythrocytes. Of all of the examined EAs, EAE showed the strongest protective effects against lipid oxidation in carp erythrocytes. The antioxidant effects of EQ at the concentration of 0.25 mg/ml were stronger than that of EAE at the concentration of 0.5 mg/ml in fish erythrocytes. The antioxidative activities of EAs in carp erythrocytes were positively correlated with their phenols content. Moreover, dietary EAE raised growth performance in fish. The appropriate concentration of EAE for the growth of fish was estimated to be 3.544 g/kg diet. Dietary EAE increased digestive and absorptive capacity in carp and improved antioxidant capacity in copper‐treated carp. So, EAE could be used as a potential natural antioxidant and inhibitor of copper stress in fish.  相似文献

To define the optimal combination time and LRH-A (luteinizing releasing hormone analogue) administration route for inducing spermiation in carp, two experiments were carried out. In the first experiment, carp kept in raceways in running recycled water in August (at a water temperature of 15–20°C) were injected intraperitoneally with pimozide (PIM) at 1 or 10 mg/kg body weight as well as with Des-Gly10 [D-Tryp6] LH-RH-ethylamide (LRH-A) at 0.1, 1 or 10 μg/kg; these products were given 6 h apart. The response was evaluated by the volume of milt and the number of spermatozoa emitted and by the gonadotropin (GTH) concentration in the plasma. At doses of 0.1, 1 and 10 μg/kg LRH-A, the total milt production was increased 2, 4 and 6-fold respectively when these treatments were combined with 1 mg PIM. In the second experiment, adult male carp, held in cages in a pond in a natural environment (at a water temperature of 11–14°C) at the beginning of May, received an intraperitoneal injection of PIM (1 mg/kg body weight) and a silicone rubber implant of LRH-A (Des-Gly10 [D-Ala6] - LH-RH-ethylamide) at dose of 10 μg/kg body weight simultaneously. Other groups of fish received PIM plus a blank implant. All of the treatments were given at either 6 a.m. or 8 p.m. When given at 6 a.m., the treatment induced a similar rise in plasma GTH (12–15 ng/ml) 24 h later in the groups treated with PIM and LRH-A implant alone or in combination. When the same treatment was given at 8 p.m., no response was observed 24 or 48 h later. Spermiation was not stimulated in any of the groups.  相似文献

Stimulation of ovulation was carried out in female carp (Cyprinus carpio L.) of Polish line 3, with carp pituitary homogenate, Ovopel (the preparation contains d ‐Ala6, Pro9NEt‐mGnRH and metoclopramide), and Ovopel and carp pituitary homogenate. The effect of the ovulation stimulators on the weight of eggs expressed in grams and as a percentage of female body weight was determined. The effect of the treatments on the quality of eggs expressed as the percentage of living embryos after 24‐ and 36‐h incubation was also determined. The best results were obtained after stimulation with Ovopel (the priming dose) and pituitary homogenate (the resolving dose). In this case all the females spawned, and the highest yield and quality of eggs were recorded. The poorest results were noted if the ovulation was stimulated with repeated injection of carp pituitary homogenate. The females treated with carp pituitary homogenate gave eggs of poorer quality than those stimulated with two doses of Ovopel or with Ovopel and pituitary homogenate. The percentage of living embryos after 24‐ and 36‐h incubation of eggs was significantly higher for the group stimulated with Ovopel and pituitary homogenate than for the other treatments.  相似文献

In order to investigate the effects of lysine and dissolved oxygen on grass carp, the grass carp were fed with 13, 15 and 17 g kg −1 lysine diet at about 6 mg L−1 (high dissolved oxygen, HO group) or 3.5 mg L−1 (low dissolved oxygen, LO group) dissolved oxygen level, for 8 weeks. The fish were fed to apparent satiation by hand. The results showed that apparent digestibility of protein, energy and dry matter were decreased significantly when grass carp were fed at 3.5 mg L−1 dissolved oxygen, and feed intake (FI) was also inhibited by low dissolved oxygen (< 0.05). Weight gain, protein retention, protein efficiency, feed conversion ratio and amino acid retention of fish at 6 mg L−1 dissolved oxygen level were significantly improved at 3.5 mg L−1 dissolved oxygen level (< 0.05). Weight gain, protein and amino acid retention, and feed efficiency of grass carp at the two dissolved oxygen levels were significantly improved by lysine supplementation (< 0.05). The dietary lysine level and dissolved oxygen of water had an interaction effect on feed conversion ratios (< 0.05). Grass carp fed at low dissolved oxygen level showed lower liver protein and fat contents. Plasma aspartate aminotransferase (AST) activity of grass carp fed at 3.5 mg L−1 dissolved oxygen level was significantly increased compared to 6 mg L−1 dissolved oxygen level (< 0.05). Our results show that low dissolved oxygen level of water is harmful to the liver of grass carp.  相似文献

Induced spawning in Eurasian perch, Perca fluviatilis L., was studied using human chorionic gonadotrophs (hCG) or acetone-dried common carp pituitary with or without hCG. The dose administered to fish was 5700 IU hCG kg-1 or 4.0 mg kg-1 carp pituitary with or without 500-700 IU hCG kg-1 for females and 2850 IU hCG kg-1 or 2.0 mg kg-1 carp pituitary with or without 250-350 IU hCG kg-1 for males. There were no statistically significant differences in quantity of milt in treated and control groups, although the best result was observed when males were treated with a triple injection of hCG and carp pituitary extract. Male spermiating success, expressed as a quantity of milt, was negatively correlated with fish weight. All females treated in this experiment had oocytes at the same division, so time of ovulation was highly synchronous. Spawning success, expressed as a spawning effectivity coefficient (Sc), was highest in fish treated with the triple injection. Spawning methods described in this paper were successful, even though hormones from another order of fish were used.  相似文献

Ovulation and spermiation of maturing Pacific herring were accelerated using single injections of acetone-dried salmon pituitary extract (10 mg/kg), partially purified salmon gonadotropin (0.1 mg/kg and 1.0 mg/kg) and the mammalian gonadotropin-releasing hormone analogue des-Gly10 [D-Ala6] GnRH-ethylamide (0.02 mg/kg and 0.2 mg/kg). Studies in mammals have shown that synthetic Met-enkephalin derivatives, called growth hormone-releasing peptides (GHRPs), stimulate growth hormone (GH) release. In the present study, GHRP-6 action on GH secretion was examined in vivo and in vitro in sexually immature grass carp. GHRP-6 injected intraperitoneally had no influences on serum GH levels in juvenile grass carp. Following intraperitonal injection of GHRP-6 and dopamine (DA) or cysteamine hydrochloride (CSH), alone and in combination into juvenile grass carp, DA and CSH were effective in elevating serum GH levels, but GHRP-6 was not effective in this respect; in addition, the synergistic action of GHRP-6 and DA or CSH on GH secretion was not seen. In this work, we had adapted and validated a perifusion system and a culture system for GH regulation studies. In a perifusion system, GHRP-6 (1000 to 0.1 nM), GHRP-6 (0.1 to 1000 nM), GHRP-6 (1 μM), and Hexarelin (an analog of GHRP, 1 μM) had no action on GH release from juvenile grass carp pituitary fragments or cells. Under static incubation conditions, GHRP-6 was inactive on GH release from juvenile grass carp pituitary fragments after 1 h and 6 h incubation, but human growth hormone-releasing hormone (hGHRH; 1 to 100 nM) as positive control could stimulate GH release in a dose-dependent manner. Furthermore, when GHRP-6 (100 nM) in combination static incubation with neuropeptides [e.g., hGHRH (100 nM), salmon gonadotropin-releasing hormone analogue (sGnRH-A) (100 nM), or D-Ala6,Pro9-NEt-luteinizing hormone-releasing hormone (D-Ala6,Pro9-NEt-LHRH, LHRH-A) (100nM)], GHRP-6 did not strengthen GH secretion actions of neuropeptides, and at the same time neuropeptides also did not modify the effects of GHRP-6 on GH secretion. The present results obtained using in vivo and in vitro techniques adapted for GH regulation studies show that GHRP-6 does not function as a GH-releasing factor in juvenile grass carp as it does in tilapia, amphibians, chickens, and mammals.  相似文献

A 60‐day feeding trial was conducted to estimate the effects of dietary iron (Fe) levels on growth, Fe concentration in the liver, spleen, and blood, and transferrin and hepcidin concentrations in the blood of bighead carp (Aristichthys nobilis). The six experimental diets were formulated to contain different Fe levels (0, 43.1, 84.2, 123.3, 162.2 and 203.1 mg/kg of dry diet) using ferrous sulphate (FeSO4) as the source. The weight gain (WG) and the specific growth ratio (SGR) of A. nobilis fed with a dietary Fe level of 123.3 mg/kg were significantly higher than that of the 0 mg/kg Fe group (p < .05). The results indicated that the growth was affected by dietary Fe levels. Regression analysis of WG and SGR at different levels of dietary Fe suggests that the appropriate dietary requirement of Fe for the bighead carp larvae is 120–134.36 mg/kg. The Fe contents in different tissues were as follows: spleen > liver > whole body. When the Fe dietary content increases to 162.2 mg/kg, the blood concentrations of Fe significantly decreased and thereafter increased, hepcidin significantly decreased and thereafter decreased, and transferrin significantly increased and thereafter decreased. The results indicate that the transferrin blood content significantly increased with decreasing hepcidin of up to 264.63 μg/ml content and thereafter decreased. It could be concluded that after transferrin saturation, hepcidin functions to maintain iron balance in the blood of A. nobilis by decreasing transferrin content.  相似文献
