为探讨季铵盐阳离子表面活性剂在番茄灰霉病防治中的应用潜力,室内采用菌丝生长速率法测定了五种阳离子表面活性剂对番茄灰霉病菌Botrytis cinerea的毒力作用,筛选出具有较高抑菌活性的1227(十二烷基二甲基苄基氯化铵)和C8-10(双八、十烷基季铵盐),并研究了其对该菌菌丝干重、分生孢子萌发、产孢量、菌核形成及细胞膜透性的影响。结果表明,五种阳离子表面活性剂中1227和C8-10对病原菌菌丝生长有较强的抑制作用,EC50值分别为8.39和5.82μg/mL,其毒力高于常规对照药剂腐霉利水悬浮剂,与嘧霉胺水悬浮剂、乙霉威水悬浮剂相当。两者对菌丝干重、孢子萌发均有较强的抑制作用,20μg/mL时抑制率接近或达到100%;对菌核数、菌核形态及分布有一定影响;40μg/mLC8-10对该菌产孢量的抑制率可达90%以上;1227和C8-10的浓度大于20μg/mL时均能明显影响病原菌细胞膜的通透性。  相似文献

中药丁香提取物对番茄灰霉病菌抑制作用及生防效果   总被引:8,自引:0,他引:8  
 试验测定了中药丁香的甲醇提取物对番茄灰霉病菌室内抑菌作用和对番茄灰霉病的温室防治作用。室内抑菌试验采用菌丝干重法测试结果表明,提取物在20 000μg/mL和10 000 μg/mL两个浓度下对灰霉病菌菌丝生长的抑制作用均达到了80%以上,5 000μg/mL浓度下抑制作用迅速下降到35.37%。在抑制分生孢子产生的测试中,浓度为10 000μg/mL的处理完全抑制了分生孢子的产生,浓度为5 000μg/mL的处理也表现出显著的抑制产孢作用,抑制率达到85.40%,而进一步稀释的处理则没有表现出显著抑制作用。采用生长速率法测试了丁香提取物对番茄灰霉病菌菌丝生长的抑制中浓度,结果表明其抑制中浓度为3 292.38μg/mL。在温室盆栽试验中,浓度为20 000μg/mL的处理对番茄灰霉病的保护防治效果达到86.45%,与化学药剂处理达到无显著差异,浓度为10 000μg/mL的处理在保护作用测试中也表现出了显著的防治效果。  相似文献

用含毒介质培养法测定了邻烯丙基苯酚(银果)对番茄灰霉病菌Botrytis cinerea、苹果腐烂病菌Valsa mali和小麦纹枯病菌Rhizoctonia cerealis的影响。通过培养观察发现,邻烯丙基苯酚可抑制番茄灰霉病菌分生孢子的产生、促进灰霉病菌菌核的形成及苹果腐烂病菌子囊壳的形成。光学显微观察表明:用50μg/mL的邻烯丙基苯酚处理番茄灰霉病菌菌丝后,菌丝伸长缓慢,分支间距缩短,分生孢子形成受到抑制。电镜观察表明:经邻烯丙基苯酚处理后,番茄灰霉病菌菌丝内液泡增多,苹果腐烂病菌菌丝细胞壁增厚,小麦纹枯病菌菌丝中内质网处泡囊增多。  相似文献

采用生物测定方法研究了啶菌噁唑对番茄灰霉病菌Botrytis cinerea不同发育阶段的影响。结果表明:其对菌落扩展、芽管伸长、分生孢子产生和菌核形成均具有显著的抑制活性,其中啶菌噁唑对P-9菌株菌丝生长和芽管伸长的EC50值分别为0.193和0.154 μ g/mL,0.5 μ g/mL的啶菌噁唑对菌丝生长和芽管伸长的抑制率大于77%,对分生孢子和菌核萌发的抑制率分别低于10%和40%,表明药剂对分生孢子萌发无明显抑制作用。紫外吸收分析显示,啶菌 NFDA1 唑可破坏细胞的膜结构,促使菌体核酸和蛋白外渗。离体叶片毒力测定结果表明,120 μ g/mL的啶菌 NFDA1 唑可有效抑制番茄灰霉病病斑扩展,抑制率达83.74%。  相似文献

稠李链霉菌JAU4234代谢产物对水稻纹枯病菌的作用机制   总被引:1,自引:1,他引:0  
为了明确稠李链霉菌JAU4234菌株在植物病害防治中的实用价值,采用生长速率法、酶活性测定和聚丙烯酰胺凝胶电泳法研究其代谢产物DZP-8对水稻纹枯病菌的作用机制。DZP-8对菌丝生长的抑制作用明显,EC50为1.55μg/mL。随着DZP-8浓度的提高,病菌羧甲基纤维素酶、多聚半乳糖醛酸酶、果胶甲基半乳糖醛酸酶的活性均有不同程度下降,当DZP-8浓度为4.67μg/mL时,3种酶的活性分别为6.76、20.97、21.74 U/mg,较对照显著降低。DZP-8浓度为4.67μg/mL的处理,只可分辨出8条谱带,蛋白质的种类和含量与对照有显著差异。表明DZP-8对纹枯病菌有较强的抑菌活性,其作用机制是抑制菌丝生长、抑制病菌致病因子表达和蛋白质的合成等。  相似文献

采用菌丝生长速率法,测定了雷帕霉素对番茄灰霉病菌、油菜菌核病菌、水稻纹枯病菌和棉花枯萎病菌的抑菌活性,比较了嘧菌酯、丙烷脒及雷帕霉素对番茄灰霉病菌的抑菌效果,并通过电子显微镜观察了雷帕霉素对番茄灰霉病菌菌丝生长的影响。结果表明:雷帕霉素对供试4种植物病原真菌菌丝均表现出了极强的抑制活性,其中对油菜菌核病菌的抑制作用最强,EC50值为2.23×10-4μg/mL,对番茄灰霉病菌、水稻纹枯病菌和棉花枯萎病菌的EC50值分别为1.32×10-3、4.05×10-3及3.82×10-3μg/mL;雷帕霉素对番茄灰霉病菌菌丝的抑制活性显著高于嘧菌酯(EC50值为3.24μg/mL)和丙烷脒(EC50值为3.81μg/mL)。电镜观察发现,经雷帕霉素处理后,番茄灰霉病菌菌丝表现出提前衰老等症状。研究结果可为深入探讨雷帕霉素对植物病原真菌的作用机制奠定基础。  相似文献

采用菌丝生长速率法、孢子萌发法和幼苗法比较了吡唑萘菌胺和氟吡菌酰胺对23种病原菌的毒力。结果表明,吡唑萘菌胺对黄瓜褐斑病菌、黄瓜炭疽病菌、辣椒炭疽病菌、番茄早疫病菌、苹果斑点病菌、辣椒疫霉病菌、马铃薯晚疫病菌、番茄灰霉病菌、番茄菌核病菌、水稻稻瘟病菌和苹果轮纹病菌菌丝生长均有较高的抑制活性,其EC50值为0.10~9.52μg/mL,EC90值为1.87~62.23μg/mL。吡唑萘菌胺对花生白绢病菌、苹果腐烂病菌、棉花黄萎病菌、棉花立枯病菌和镰刀菌属病原菌的抑制活性均高于氟吡菌酰胺,而氟吡菌酰胺却对黄瓜褐斑病菌、番茄早疫病菌、苹果斑点病菌、番茄灰霉病菌、番茄菌核病菌和苹果褐斑病菌具有较高的抑制活性,其EC50值为0.39~3.98μg/mL,与吡唑萘菌胺相当。吡唑萘菌胺对玉米丝黑穗、花生冠腐病菌和番茄灰霉病菌的孢子有较高的毒力,其EC50值分别为0.002 9、0.07和1.38μg/mL。氟吡菌酰胺仅对花生冠腐病菌、番茄灰霉病菌的孢子有较高的活性,且它与吡唑萘菌胺活性相当。吡唑萘菌胺和氟吡菌酰胺对黄瓜白粉病菌均有较高的毒力,EC50值分别为0.04、0.05μg/mL。因此,吡唑萘菌胺比氟吡菌酰胺更加广谱,且抑菌活性相当或更高。  相似文献

 番茄灰霉病菌在致病过程中能够产生4种细胞壁降解酶,以PMG酶活性最高,其次是β-葡萄糖苷酶和PG酶,Cx最少。灰霉病菌在不同温度下侵染番茄叶片时产生的致病酶活性不同,4种酶在20℃时表现了最高的活性,15℃次之,当温度达到25℃时,各种酶的活性都急剧下降,随着温度的再升高,酶活更低。随着湿度的增高,病菌产生的细胞壁降解酶的活性也增加,当相对湿度达到90%以上时,4种酶的活性也达到最高。温湿度对番茄灰霉病菌产生细胞壁降解酶的影响趋势,与其对发病的影响趋势是一致的。  相似文献

试验测定了石榴皮萃取物对番茄灰霉病菌室内抑菌作用和对番茄灰霉病的温室盆栽防治作用。室内抑菌试验分别采用生长速率法和孢子萌发法测定对番茄灰霉病菌菌丝生长和孢子萌发的抑制作用,结果表明:正丁醇萃取物对病原菌菌丝的抑制活性最高,EC50为50.28 g/L;浓度为200 g/L对番茄灰霉病菌分生孢子的产生具有显著的抑制作用,抑制作用与浓度呈正相关关系。在温室盆栽试验中, 正丁醇萃取物稀释100倍的处理对番茄灰霉病的保护防治效果达到46.41%,治疗防治效果达到48.00%。  相似文献

为了探讨武夷菌素对葡萄灰霉病的防治效果及作用机理,本文通过菌丝生长速率测定、分生孢子萌发抑制测定、胼胝质沉积测定、离体叶片和果实防治等试验,发现武夷菌素可以抑制灰霉病菌的菌丝生长,浓度达到120μg/mL时,对菌丝的抑制作用达到94.3%;当浓度达到100μg/mL时,武夷菌素对葡萄灰霉病菌的孢子萌发抑制率可达到96.0%;通过离体叶片及果实防效试验发现,武夷菌素浓度达到120μg/mL时,防治效果分别为87.5%和96.6%。武夷菌素可以显著降低灰霉病菌的草酸合成能力,导致其致病力减弱,从而减轻灰霉病菌对寄主的侵染。武夷菌素能够通过诱导葡萄叶片的胼胝质沉积,增加葡萄组织的防御能力,从而起到有效防治灰霉病的作用。综上所述,武夷菌素对葡萄灰霉病具有良好的防治效果,可以作为生物防治药剂进行应用推广。  相似文献

丁香酚是从植物中提取的天然化合物。采用菌丝生长速率法测定了其对多种植物病原真菌的抑制作用以及田间分离得到的7株灰葡萄孢Botrytis cinerea对丁香酚的敏感性,同时测定了丁香酚对B.cinerea 03 孢子和菌核的抑制活性。结果显示,丁香酚对多种植物病原真菌均有不同程度的抑制作用;灰葡萄孢不同菌株对丁香酚的敏感性不同,EC50值在29.97 ~83.62 μg/mL之间;丁香酚可抑制灰葡萄孢产孢,但对孢子萌发无影响,可抑制其菌核的产生和萌发。荧光染色结果表明丁香酚能破坏灰葡萄孢菌丝细胞膜。利用原子吸收光谱法和紫外分光光度法分别测定丁香酚处理前后菌丝培养液中K+浓度和OD260值变化,结果显示处理后K+浓度和OD260值均升高。综上所述,细胞膜可能是丁香酚对灰葡萄孢菌丝的作用位点。  相似文献

This paper assessed the antifungal effects of sage oil, lavender oil, mint oil, and tea tree oil on the postharvest fungal pathogen, Botrytis cinerea, which causes gray molds. The change of morphology, physiological, and biochemical characteristics about fungal hyphae and conidia were determined. As results show, all four oils can effectively inhibit the growth of B.cinerea and the antifungal effects are dose dependent. The best antifungal effect has been found from mint oil. According to in vitro studies, volatile vapor worked better than direct contact. With volatile vapor, the growth of B.cinerea was inhibited completely at 2 for all four oils. Mint oil at 500 μL/L and its volatile vapor at 25 could inhibit both conidia germination and disease incidence significantly in vivo. Observations by using scanning electron microscope and transmission electron microscope revealed that, mint oil could destroy the ultrastructure of hyphae and conidia, resulting in markedly shriveling and crinkling of the hyphae and conidia. It could also thicken and disrupt cell wall, causing cellular nucleic acids and proteins divulged with the damage of the cell wall. The chemical composition analysis of mint oil using GC/MS revealed that its main components were cyclohexanol, cyclohexanone, and some alkenes and alkanes. The majority of the components were effective antifungal agents. The content of volatile cyclohexanol and cyclohexanone were found to be 39.79% and 22.24% respectively.  相似文献

ABSTRACT The exo-beta-1,3-glucanase (EC 3.2.1.58) activity of Pichia anomala strain K, an antagonistic yeast of Botrytis cinerea on postharvest apples, was studied in a synthetic medium supplemented with laminarin, a cell wall preparation (CWP) of B. cinerea, or glucose. The highest enzyme activity was detected in culture media containing a CWP of B. cinerea as the sole carbon source, whereas the lowest activity was observed in culture media supplemented with glucose. Exoglc1, an exo-beta-1,3-glucanase, was purified to homogeneity from culture filtrates of strain K containing a CWP. The molecular mass of exoglc1 was estimated to be under 15 kDa. Optimum activity of exoglc1 was recorded at 50 degrees C and pH 5.5. The exoglc1 K(m) value was estimated at 22.4 mg/ml. Exoglc1 showed in vitro a stronger inhibitory effect on germ tube growth of B. cinerea than on conidia germination and caused morphological changes such as leakage of cytoplasm and cell swelling. Exo-beta-1,3-glucanase activity was detected on apples treated with strain K and was similar to exoglc1 on the basis of activity on native gel. Moreover, the addition of a CWP to a suspension of P. anomala stimulated both in situ exo-beta-1,3-glucanase activity and protective activity against the pathogen, strengthening the hypothesis that exo-beta-1,3-glucanase activity is one of the mechanisms of action involved in the suppression of B. cinerea by P. anomala strain K.  相似文献

 借助光镜和透射电镜观察了武夷菌素对番茄灰霉菌菌丝生长和分生孢子萌发的形态影响,结果表明,武夷菌素处理的菌丝异常生长,分生孢子萌发的芽管膨大缢缩,菌丝体内液泡增多;武夷菌素能引起菌丝细胞膜透性的变化,造成液体培养基电导率增加;同位素标记的14C-谷氨酸、3H-葡糖胺和3H-腺嘌呤掺入实验表明,武夷菌素能抑制菌丝蛋白质的合成,对细胞壁几丁质和核酸的合成没有影响。  相似文献

 产几丁质酶枯草芽孢杆菌G3菌株的固体培养物在黄瓜灰霉病菌和番茄叶霉病菌抑菌试验中证实,抑菌活性物质存在于过滤上清液中,它们是从酸沉淀物中提取出的伊枯草菌素、生物表面活性素和存在于盐析粗蛋白中的几丁质酶。在叶霉孢子萌发试验中,伊枯草菌素微弱地抑制孢子萌发但强烈破坏芽管和新生菌丝;生物表面活性素和几丁质酶则强烈抑制孢子萌发并长久性地抑制芽管伸长。在PDA平板上的灰霉菌丝抑菌试验中,伊枯草菌素抑制菌丝生长,引发菌丝顶端膨大,形成泡囊,泡囊破裂后原生质外泄;几丁质酶抑制菌丝生长,引发产生不规则的菌丝团;生物表面活性素在平皿上对菌丝则不显示出抑菌活性。  相似文献

研究了新型环烷基磺酰胺类化合物N-(2,4,5-三氯苯基)-2-氧代环己烷基磺酰胺(简称化合物108)对灰葡萄孢菌丝生长、孢子形成和萌发以及菌核产生等不同生育阶段的抑制作用及其对菌丝致病力和形态结构的影响。结果表明:化合物108对灰葡萄孢菌丝生长和孢子形成及孢子萌发具有明显的抑制作用,其EC50值分别为6.90、4.70和4.11μg/mL;菌核形成受到明显抑制,当药剂质量浓度达20μg/mL时,无菌核产生。经化合物108处理后的灰葡萄孢菌丝致病力下降,40μg/mL处理的菌丝致病力显著低于对照。超微结构观察结果表明,化合物108能导致灰葡萄孢菌丝萎缩、塌陷和变形,菌体细胞壁增厚、皱缩及分层。  相似文献

ABSTRACT A mechanism that could contribute to the suppression of Botrytis cinerea during pathogen sporulation was examined in this study. Yeasts capable of binding to B. cinerea were formulated with a cellulose carrier and applied to sporulating colonies of the pathogen. The particles from this yeast/cellulose product attached to B. cinerea conidia in the sporulating colony. Inoculum from treated colonies was harvested and applied to tomato stem tissue to test for subsequent pathogenicity. Disease development from inoculum obtained from cultures that had been treated with Trichosporon pullulans was significantly retarded (P = 0.0001) compared with cellulose-only controls. However, between 5 and 11% of conidia applied were attached to yeast cells. The removal of conidia not attached to yeast resulted in inoculum composed of >90% of conidia attached to yeast, and from this inoculum, disease development was significantly retarded (P < 0.05). When inoculum from treated B. cinerea colonies was applied to nutrient limiting agar and then incubated, the B. cinerea conidia germinated, and yeast cells infested the new hyphal growth. Constraints of the formulation of the yeast used in this study, and the implications of this vectoring approach for the suppression of B. cinerea during pathogen sporulation are discussed.  相似文献

ABSTRACT The effect of treatments with conidial suspensions of Ulocladium atrum and Gliocladium roseum on leaf rot of cyclamen caused by Botrytis cinerea was investigated under commercial greenhouse conditions. Spraying U. atrum (1 x 10(6) conidia per ml) or G. roseum (2 x 10(6) conidia per ml and 1 x 10(7) conidia per ml) at intervals of 2 to 3 weeks during the production period and spraying U. atrum (1 x 10(6) conidia per ml) at intervals of 4 to 6 weeks resulted in a significant reduction of natural infections of petioles by B. cinerea. U. atrum or G. roseum (1 x 10(7)conidia per ml) was as effective as the standard fungicide program. B. cinerea colonized senesced leaves within the plant canopy and infected adjacent petioles and leaves later. The antagonists colonized senesced leaves and reduced B. cinerea development on these leaves. Thus, the inoculum potential on petioles adjacent to necrotic leaf tissues was reduced. The fate of U. atrum conidia on surfaces of green cyclamen leaves during a 70-day period after application was studied. The number of conidia per square centimeter of leaf surface remained relatively constant during the entire experiment. Sixty percent of the conidia sampled during the experiments retained the ability to germinate. When green leaves were removed from the plants to induce senescence and subsequently were incubated in a moist chamber, U. atrum colonized the dead leaves. Senesced leaves also were colonized by other naturally occurring fungi including B. cinerea. On leaves treated with U. atrum from all sampling dates, sporulation of B. cinerea was significantly less as compared with the untreated control. Our results indicate that early applications of U. atrum before canopy closure may be sufficient to achieve commercially satisfactory control of Botrytis leaf rot in cyclamen.  相似文献

为验证哈茨木霉菌与啶酰菌胺联用对番茄灰霉病菌的增效作用,采用显微镜观察法观察了菌药联用后哈茨木霉菌和番茄灰霉病菌的菌丝形态,用菌丝生长速率法和圆盘滤膜法测定了菌药联用后哈茨木霉菌挥发性物质、非挥发性物质和粗酶液对番茄灰霉病菌的抑菌活性.结果显示:啶酰菌胺对哈茨木霉菌的菌丝基本无影响,但能使灰霉病菌的菌丝发生间断性溶解;菌药联用后哈茨木霉菌挥发性物质对灰霉病菌的最高抑菌活性为61.17%,而哈茨木霉菌和啶酰菌胺单独对灰霉病菌的抑菌活性分别为48.79%、0.97%,表现增效作用;菌药联用后哈茨木霉菌非挥发性物质和粗酶液对番茄灰霉病菌的抑菌活性未明显增强.研究表明,啶酰菌胺与哈茨木霉菌联用的增效机制是啶酰菌胺导致灰霉病菌的菌丝部分溶解,同时增强了哈茨木霉菌挥发性物质对灰霉病菌的抑菌活性.  相似文献
