This study evaluated the reactions of seven common C4 grasses of the tallgrass prairie of the USA Great Plains to the economically important wheat pathogens Pyrenophora tritici‐repentis and Gaeumannomyces graminis var. tritici (Ggt) isolated from wheat. The P. tritici‐repentis isolates (race 1) were pathogenic on all grasses tested, but symptom severity was markedly low. Three of the grass species inoculated with Ggt were highly susceptible, while four species exhibited no symptoms. Because measures of connectedness can provide a proxy for population processes, connectedness was evaluated within and among the seven grass species in representative tallgrass prairie environments for all potential pathogen‐sharing patterns. Andropogon gerardii was ubiquitous, so all plant species were well connected to it. Andropogon scoparius (= Schizachyrium scoparium), Sorghastrum nutans and Panicum virgatum were fairly common but specialized to particular environments. Bouteloua curtipendula was uncommon but occurred in all environments, while Buchloë dactyloides and Bouteloua gracilis were uncommon and only occurred in upland sites. Co‐occurrence of plant species was generally not reciprocal in that, for many species pairs, species A rarely occurred without potential exposure to inoculum from species B, while species B commonly occurred without species A. The three grass species susceptible to Ggt may act as sources of inoculum for each other within tallgrass prairie, with the potential to influence fitness, and tallgrass prairie and commercial wheat ecosystems in the Great Plains also have the potential to share both pathogens.  相似文献

Grass species were grown in plots, as pure stands or mixed with wheat, after a sequence of wheat crops in which take-all ( Gaeumannomyces graminis var. tritici ) had developed. Annual brome grasses maintained take-all inoculum in the soil as well as wheat (grown as a continuous sequence), and much better than cultivated species with a perennial habit. Take-all developed more in wheat grown after Anisantha sterilis (barren brome) or Bromus secalinus (rye brome), with or without wheat, than in continuous grass-free wheat in the same year, where take-all decline was apparently occurring. It was equally or more severe, however, in wheat grown after Lolium perenne (rye-grass) or Festuca arundinacea (tall fescue), despite these species having left the least inoculum in the soil. It was most severe in plots where these two grasses had been grown as mixtures with wheat. It is postulated that the presence of these grasses inhibited the development of take-all-suppressive microbiota that had developed in the grass-free wheat crops. The effects of the grasses appeared to be temporary, as amounts of take-all in a second subsequent winter wheat test crop were similar after all treatments. These results have important implications for take-all risk in wheat and, perhaps, other cereal crops grown after grass weed-infested cereals or after set-aside or similar 1-year covers containing weeds or sown grasses, especially in combination with cereal volunteers. They also indicate that grasses might be used experimentally in wheat crop sequences for investigating the mechanisms of suppression of, and conduciveness to, take-all.  相似文献

Nucleotide sequence variations were investigated with respect to the geographical distribution patterns of Miscanthus sinensis populations that were sampled from 26 Japanese national parks and three populations of the Ryukyu Islands. Twelve homozygous sequences in the nuclear ribosomal DNA internal transcribed spacer region were detected. The populations of M. sinensis in mainland Japan mainly were composed of a monophyletic group with a symapomorphic character, whereas those in the Ryukyu Islands included a polyphyletic group. Only an internal transcribed spacer haplotype with a plesiomorphic character was found in both mainland Japan and the Ryukyu Islands. Thus, no clear geographical isolation was observed in this species. These facts might be caused by the ability of M. sinensis as a pioneer plant to have a high migration potential and high gene flow by outcrossing. On the basis of the results of this study (nuclear ribosomal DNA) and the previous study (chloroplast DNA), a phylogeographical history of M. sinensis was presumed where the ancestral and derived lineages were distributed in the southern and northern regions of Japan, respectively, without strict geographical isolation.  相似文献

水稻是我国最主要的粮食作物之一,杂草的危害严重影响了水稻的产量与品质。化学防除仍然是治理水稻田杂草最有效的途径。目前我国水稻田稗属杂草、千金子、马唐、雨久花、野慈姑、异型莎草、耳叶水苋、眼子菜、节节菜、萤蔺等多种杂草对二氯喹啉酸、五氟磺草胺、氰氟草酯、噁唑酰草胺、苄嘧磺隆、吡嘧磺隆、双草醚、噁草酮、乙氧氟草醚等多种常用除草剂产生了抗药性。面对日趋严重的水稻田抗药性杂草的危害,对抗药性杂草进行深入系统的研究以达到科学防治的目的显得尤为重要。本文总结了我国水稻田抗药性杂草的抗药性水平、靶标酶抗药性机理、代谢酶抗药性机理和其他抗药性机理,归纳了抗药性杂草的交互抗性、多抗性发生情况以及抗药性治理现状,分析了我们在杂草抗药性研究与治理方面面临的问题。  相似文献

匍匐翦股颖(Agrostis stolonifera)和细弱翦股颖(A. tenuis)是优良的牧草和草坪草。寄生于翦股颖上的腥黑粉菌共有3种,即小麦网腥黑穗菌(Tilletia caries,TCT)、翦股颖腥黑穗菌(T. sphaerococca)和苍白腥黑粉菌(T. pallida)。在形态、自发荧光和萌发生理3方面比较研究的基础上,选取冬孢子形态非常相似的2种腥黑粉菌TCT和T. sphaerococca为研究对象,依据序列特点设计4套引物,成功建立了TCT和T. sphaerococca菌丝基因组DNA的特异双重PCR检测方法和冬孢子的特异套式双重PCR检测方法。  相似文献

本研究对海南省澄迈县土沉香根结线虫病危害与发生情况进行了调查与研究,并采用比较形态学结合rDNA-ITS序列分析的方法对病原进行了分离鉴定,确定了发生在澄迈县土沉香苗木上的根结线虫为象耳豆根结线虫Meloidogyne enterolobii YangEisenback。这是首次报道海南省澄迈地区土沉香上发生象耳豆根结线虫,为进一步分析土沉香病原根结线虫种群及该病的防治研究奠定了基础。  相似文献

对华东地区松材线虫病疫区和松墨天牛主要发生的13个地区随机抽样调查,共获松墨天牛病原真菌9属12种,病原细菌2属2种。其中,我国首次报道的松墨天牛病原菌新种类有:金龟子绿僵菌小孢变种Metarhizium anisopliae var.anisopliae、粉拟青霉Paecilomyces farinosus、尖镰孢Fusarium oxysporum、蜡蚧轮枝霉Verticillum lecanii、青霉Penicillium spp.、黄曲霉Aspergillus flavus、曲霉Aspergillus spp.、木霉Trichoderma spp.和粘质沙雷氏菌Serratia marcescens。经致病性测定表明,球孢白僵菌、金龟子绿僵菌小孢变种、粉拟青霉对松墨天牛有较强的致病力,处理后16d的致死率分别为100%、100%和70%。  相似文献

Fusarium head blight (FHB) of wheat and other small-grain cereals is a disease complex caused by several fungal species. To monitor and quantify the major species in the FHB complex during the growing season, real-time PCR was developed. TaqMan primers and probes were designed that showed high specificity for Fusarium avenaceum, F. culmorum, F. graminearum, F. poae and Microdochium nivale var. majus. Inclusion of an internal PCR control and serial dilutions of pure genomic DNAs allowed accurate determination of the concentration of fungal DNA for each of these species in leaves, ears as well as harvested grains of winter wheat. The DNA concentration of F. graminearum in grain samples correlated (r 2= 0.7917) with the incidence of this species on the grain as determined by isolation from individual kernels. Application of the TaqMan technology to field samples collected in 40 wheat crops in the Netherlands during the growing season of 2001 revealed that M. nivale var. majus predominated on leaves early in the season (GS 45-65). Ears and harvested grains from the same fields, however, showed F. graminearum as the major species. In 2002, grain samples from 40 Dutch fields showed a much wider range of species, whereas in ears from 29 wheat crops in France, F. graminearum was the predominant species. The concentration of DON correlated equally well with the incidence of the DON-producing species F. culmorum and F. graminearum in the grain samples (r 2= 0.8232) as well as with total DNA of both these species (r 2= 0.8259). The Fusarium TaqMan technology is an important tool to quantify and monitor the dynamics of individual species of the complex causing FHB in cereals during the growing season. This versatile tool has been applied in a comparison of different genotypes, but can also be applied to other disease management systems, e.g. fungicide treatments.  相似文献

Isolates of the take-all fungus, Gaeumannomyces graminis var. avenae , which affects oats, wheat and other grasses, and of G.g. var. tritici , which preferentially affects wheat, rye and barley, contain a high proportion of repeated sequences. Total DNA from 57 fungal isolates collected from many locations and different cereal hosts, and scored for virulence on wheat, rye and oats, revealed many restriction fragment length polymorphisms. These RFLP s were observed either by staining the DNA directly, by hybridization to radioactively labelled total fungal DNA , or by hybridization with labelled wheat ribosomal DNA . With only a few exceptions, the isolates with the same preferred cereal hosts showed more similar patterns of restriction fragments than isolates that had different pathogenicity properties on cereal hosts, irrespective of the geographical origins of the isolates. This was even the case for R isolates of G.g. var. tritici that were virulent on wheat and rye compared with N isolates that were virulent only on wheat. Isolates were identified by hybridizing DNA from infected root samples with 32P-labelled total fungal DNA . The restriction fragment polymorphisms involving families of repeated sequence can therefore be used as a predictive assay for host preference of an isolate, and have probably arisen by host selection of fungal lineages. The variation between isolates in different pathogenicity groups suggests that there is little gene flow between isolates that can infect different hosts, even though they can coexist in the same field.  相似文献

The globally invasive chestnut gall wasp Dryocosmus kuriphilus was recently reported in Italy and threatens European chestnut orchards and native forests. Of Chinese origin, this species has invaded Japan, Korea, the USA, Nepal and Europe and in each region it has been attacked by parasitoids exploiting oak gall wasps. Classical biological control using the parasitoid Torymus sinensis (Hymenoptera: Torymidae) successfully reduced infestation in Japan. Subsequent work in Japan and Korea showed this parasitoid to represent a poorly understood and biologically diverse species complex. Following its success in Japan, T. sinensis was released in Italy in 2005. A growing appreciation of the taxonomic and ecological complexity of the T. sinensis complex in Japan and Korea has stimulated ongoing DNA-based work on relationships and interbreeding of species in this group. Oak cynipid parasitoids provide potential for native augmentative or conservation biological control of D. kuriphilus in Europe. Exploitation of this potential requires improved understanding of the taxonomy of these parasitoids, particularly of morphologically highly conserved lineages such as Eurytoma and Eupelmus in which recognised species may contain many cryptic lineages. There is a growing need for a molecular survey of the communities centered on chestnut and oak gall wasps to: (1) ascertain the identity of T. sinensis lineages released in Italy (2) track the dispersal of T. sinensis in the native community, either in its original form or as hybrids with native species (3) identify possible biological control agents in the native parasitoid community.  相似文献

当归根腐病是当归生产中的重要病害之一,分离纯化后鉴定为尖孢镰刀菌(Fusarium.oxysporum)为了明确中草药党参和黄芪提取液对当归根腐病的抑制作用,分别以两种中药水浸液对小当归根腐病的抑制作用进行测定。结果表明:不同浓度的中草药党参和黄芪提取液对当归根腐病菌丝生长和孢子萌发都有抑制作用,在浓度为0.50 g/mL对当归根腐病菌丝生长的抑制率为25.23%和53.48%,对其孢子萌发抑制率为51.57%和74.56%,且所有处理发生根腐病均比对照低,差异达显著水平。  相似文献

ABSTRACT Diaporthe phaseolorum and Phomopsis longicolla isolates from soybean were examined using traditional mycological characteristics and molecular methods. Cultural characteristics including types of fruiting bodies and conidia were assessed for isolates collected from soybean stems and seeds. Cultures were identified as P. longicolla, D. phaseolorum var. caulivora, D. phaseolorum var. meridionalis, or D. phaseolorum var. sojae. Molecular markers for these groups were developed and analyzed using polymerase chain reaction restriction fragment length polymorphisms (PCR-RFLP) and DNA sequencing in the internal transcribed spacer (ITS) and the 5.8S ribosomal DNA. The ITS(4) and ITS(5) primers amplified PCR products for all isolates studied. Gel electrophoresis of undigested PCR products and DNA sequencing produced various fragment lengths including 604 bp for P. longicolla, 602 and 603 bp for D. phaseolorum var. caulivora, 603 bp for D. phaseolorum var. meridionalis, and from 597 to 609 bp for D. phaseolorum var. sojae. Digestion of these PCR products with enzymes AluI, HhaI, MseI, RsaI, and ScrFI resulted in distinct bands for identification of P. longicolla and the varieties of D. phaseolorum I. All P. longicolla, D. phaseolorum var. caulivora, and D. phaseolorum var. meridionalis isolates were distinguished using AluI and HhaI with RsaI or ScrFI. The banding patterns of D. phaseolorum var. sojae isolates were complex and were separated into 11 subgroups after digestion with AluI, HhaI, MseI, RsaI, and ScrFI. Phylogenetic analysis of 20 isolates of D. phaseolorum and P. longicolla based on the DNA sequence of the ITS region resolved six clades termed A, B, C, D, E, and F. Clade A included all sequenced D. phaseolorum var. caulivora isolates, two from Italy and one from the United States. Isolates in clade B were exclusively associated with D. phaseolorum var. meridionalis. Clades A and B formed a well-supported monophyletic group. Isolates in clades C, D, E, and F were morphologically defined as isolates of P. longicolla, D. phaseolorum var. sojae, and Diaporthe spp. The ITS sequences similarity of seven geographically diverse P. longi-colla isolates illustrated that P. longicolla isolates have a similar genetic background, with some affiliations to some D. phaseolorum var. sojae isolates. Morphological characteristics of the isolates along with the terminal clades of the ITS phylogeny suggest that P. longicolla is an individual species, D. phaseolorum var. caulivora and D. phaseolorum var. meridionalis are varieties of D. phaseolorum, and D. phaseolorum var. sojae is either several varieties of D. phaseolorum or possibly several distinct species.  相似文献

A region comprising the 5.8S RNA gene and internal transcribed spacers 1 and 2 of the take-all patch fungus, Gaeumannomyces graminis var. avenae , was cloned and sequenced using primers from the flanking 17S and 26S ribosomal RNA genes. The sequenced region showed 99% similarity between the two G. graminis isolates, and 70–80% similarity between these two isolates and several other species of fungi. From the sequence, oligonucleotide primers were selected which permitted specific amplification of DNA from G. graminis vars. avenae and graminis using the polymerase chain reaction (PCR). The assay could detect DNA of G. graminis strains obtained from a wide variety of hosts, but did not amplify DNA from many other fungi, including the important turf-grass root pathogens Magnaporthe poae and Leptosphaeria korrae. The primers also did not amplify DNA from G. graminis var. tritici, M. rhizophila or Phialophora graminicola. The PCR-based assay shows promise as a diagnostic tool for the take-all pathogen in turf-grass pathology.  相似文献

The characteristics of 50 isolates of Mycosphaerella pinodes and 17 isolates of Phoma medicaginis var. pinodella , originating from several regions of France where ascochyta blight is prevalent, were investigated using cultural, physiological, molecular and pathogenicity analyses. M. pinodes was distinguished from P. medicaginis var. pinodella on the basis of presence of pseudothecia, a higher proportion of larger, bicellular conidia, compared with the smaller, predominantly unicellular conidia of P. medicaginis var. pinodella , and a slower linear growth rate on agar under a 12-h light regime. RAPD analysis clearly distinguished the two species, which had low intraspecific variability. Although both species gave identical symptoms, they could be distinguished by their incubation period and aggressiveness, respectively, shorter and higher for M. pinodes . Virulence tests gave no definitive evidence for the existence of pathotypes among the M. pinodes isolates. Two unidentified isolates had similar characters to both M. pinodes and P. medicaginis var. pinodella in some features but were distinguished from them by their RAPD patterns.  相似文献

Take-all, caused by the soilborne fungus Gaeumannomyces graminis var. tritici, is arguably the most-studied root disease of any crop, yet remains the most important root disease of wheat worldwide. S. D. Garrett launched the study of root diseases and soilborne pathogens as an independent field of science starting in the middle of the 20th century, inspired by and based in large part on his research on take-all during the first half of the 20th century. Because there has been neither a source of host plant resistance nor an effective and economical fungicide for use against this disease, the focus for nearly a century has been on cultural and biological controls. In spite of the intensive and extensive works towards these controls, with mostly site-or soil-specific success, the only broadly and consistently effective controls require either crop rotation (break crops), or the converse, wheat monoculture to induce take-all decline. Take-all decline has become the model system for research on biological control of plant pathogens in the rhizosphere and provided the first proof to the scientific world after decades of debate that antibiotics are both produced in soils and play a role in the ecology of soil microorganisms. On the other hand, even the best yields following take-all decline are rarely equal to those achieved with crop rotation. Because of this, the continuing trends globally to shorten rather than lengthen the rotations in wheat-based cropping systems, and the growing use of direct-seed (no-till) trashy systems to reduce costs and protect soil and water resources, new methods to control take-all are needed more than ever. With high resolution maps of the genomes of cereals and other grasses now available, including a complete sequence of the rice genome, and the interesting differences as well as striking similarities among the genomes of cereals and related grasses, gene transfer to wheat from oats, rice, maize and other grass species resistant to G. graminis var. tritici should be pursued.  相似文献

ABSTRACT This paper reviews the significant advances by the diseases themselves, as well as by the scientists, in the intervening period since the disease trilogy was first delimited in 1989. The impact of these diseases, black pod, witches' broom, and frosty pod rot, has increased dramatically. In addition, there have been radical changes in the taxonomic profiles of these pathogens, which have been based on both traditional (morphological, cytological) and modern (molecular) approaches. Black pod is caused by a complex of Phytophthora species, in which P. palmivora still is the most important worldwide. However, recent invasion of the principal cacao-growing countries of West Africa by the more virulent P. megakarya has been cause for concern. The latter evolved in the ancient forests straddling the Cameroon-Nigerian border as a primary coloniser of fallen fruit. Conversely, frosty pod rot, caused by Moniliophthora roreri, and witches' broom, caused by M. (Crinipellis) perniciosa, both neotropical diseases, are hemibiotrophic, coevolved pathogens. Respectively, M. roreri arose on Theobroma gileri in submontane forests on the north-western slopes of the Andes, whereas M. perniciosa developed as a complex of pathotypes with a considerably wider geographic and host range within South America; the cacao pathotype evolved on that host in the Amazon basin. The inter-relationships of these vicariant species and their recent spread are discussed, together with control strategies.  相似文献

Over 220 crude extracts from repositories generated from plants native to Greece and Kazakhstan were evaluated for termiticidal activity against the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae). Emerging from this screening effort were bioactive extracts from two Greek species (Echinops ritro L. and Echinops spinosissimus Turra subsp. spinosissimus) and extracts from two Kazakhstan species (Echinops albicaulis Kar. & Kir. and Echinops transiliensis Golosh.). Fractionation and isolation of constituents from the most active extracts from each of the four species has been completed, resulting in the isolation of eight thiophenes possessing varying degrees of termiticidal activity. 2,2':5',2"-Terthiophene and 5'-(3-buten-1-ynyl)-2,2'-bithiophene demonstrated 100% mortality against C. formosanus within 9 days at 1 and 2 wt% concentrations respectively. In addition, all but two of the eight compounds tested were significantly different from the solvent controls in the filter paper consumption bioassay.  相似文献
