1. This study was conducted to examine whether oral administration of lysine solution affect food intake and the ventromedial hypothalamic (VMH) monoamines in chickens fed on a lysine-free diet.

2. Chickens were assigned to four treatment groups. Two groups of chickens were given two different doses of lysine solution (0·1?g and 0·07?g in 1?ml of saline) exogenously (orally) while being fed on a lysine-free diet, and these results were compared with a control diet plus saline group. Another group of chickens was fed on a lysine-free diet without lysine supplementation, and their results were compared with the lysine treated groups. The extracellular dopamine (DA), norepinephrine (NE) and serotonin (5-HT) in the VMH of freely moving chicken were measured by in vivo microdialysis.

3. There was no significant difference in food intake between the control diet and the lysine supplemented groups during the time-course of the experiments. Food intake significantly decreased at 4, 5 and 6?h in the lysine-free diet plus saline group compared with the lysine supplemented groups. Of the VMH monoamines, the DA concentration remained close to the baseline in the lysine supplemented groups. This DA concentration was significantly lower than the baseline in the lysine-free diet plus saline group at 3·5?h and thereafter.

4. No significant difference from the baseline was observed for NE in the lysine-free diet plus saline group. The 5-HT concentrations were close to the baseline for all groups throughout the experiments.

5. The findings suggest that oral administration of lysine solution to chickens fed on a lysine-free diet restored food intake which was associated with the variations of VMH DA concentration.  相似文献

Two experiments were conducted in ovariectomized, pituitary stalk-transected ewes to determine if dopamine (DA), norepinephrine (NE) or serotonin (5-HT) alter secretion of luteinizing hormone (LH), follicle-stimulating hormone (FSH) and prolactin (PRL). In experiment 1, ewes were infused (iv) with saline (control), DA (66 micrograms/kg/min), NE (6.6 micrograms/kg/min) or 5-HT (6.6 micrograms/kg/min). Treatments did not alter pulse frequency, but 5-HT increased (P less than .05) amplitude of pulses of LH and mean concentrations of LH, DA and NE were without effect on basal secretion of LH. DA but not NE or 5-HT decreased (P less than .05) the release of LH in response to gonadotropin hormone-releasing hormone (GnRH, 25 micrograms, im). Concentrations of FSH were not affected by treatments. Secretion of PRL was reduced (P less than .05) by treatment with DA and NE but not 5-HT. Each amine reduced (P less than .05) the release of PRL in response to thyrotropin-releasing hormone (TRH; 3 micrograms, im). In experiment 2, ewes were given DA at doses of 0, 0.66, 6.6 or 66.0 micrograms/kg/min, iv. No dose altered basal LH, but each dose reduced (P less than .05) basal and TRH-induced release of PRL. Key findings from these studies include direct pituitary action for: (1) 5-HT enhanced basal secretion of LH, (2) suppression of GnRH-induced secretion of LH by DA. (3) DA and NE inhibition of PRL secretion, and (4) DA, NE and 5-HT inhibition of release of PRL in response to TRH.  相似文献

Sixteen crossbred wethers were distributed among four treatments and fed a control ration based on annual rye-orchardgrass (R-O) for 8 days. Indwelling jugular cannulae were installed and experimental regimes begun the following day (experimental day 1). One-half of the wethers were fed a ration based on endophyte-infected Kentucky-31 fescue while the remainder continued to receive the R-O control diet for 10 days. Spiperone, a dopamine antogonist, was administered to one-half of the wethers receiving each ration on days 8 and 9. Plasma prolactin (PRL), dopamine (DA), norepinephrine (NE) and epinephrine (E) were measured in jugular venous blood on days 1, 3, 5 and 7-10 of the trial. On day 10, the animals were decapitated; and DA, NE, E and 3,4-dihydroxyphenylacetic acid (DOPAC) and monoamine oxidase (MAO) were determined in hypothalamic and pituitary tissue. Plasma DA was elevated (P less than .05) following day 8 in wethers fed infected fescue over those fed (R-O), while plasma PRL was reduced (P = .08). Wethers receiving Spiperone had lowered (P less than .05) plasma DA and elevated (P less than .01) plasma PRL. Plasma DA was negatively correlated (P less than .01) with plasma PRL (r = -0.50) following day 8. Plasma NE and E levels and NE, E, DA and DOPAC tissue concentrations were not affected by Spiperone administration or diet. MAO levels in pituitaries were higher (P less than .01) for Spiperone-treated wethers. Six hours post-hatch, large white turkey poults were inoculated intranasally with 5 X 10(7) organisms of the "W" isolate of Bordetella avium. Three hours after inoculation and subsequently on days 7, 10, 14, 21, and 28 postinoculation, 30 infected and 30 uninfected control poults were injected intramuscularly with alpha methylparatyrosine (AMPT, 250 mg/kg), and an additional 30 of each group received a saline vehicle. Three hours after AMPT injection, whole brains from treated and untreated poults from both infected and control groups were removed, weighed, and frozen until assayed for norepinephrine (NE), dopamine (DA), and serotonin (5-HT). B. avium-infected poults had a significant reduction in steady-state NE and Da and a greater depletion of NE and DA after AMPT treatment compared with control poults. Infected poults had significantly reduced whole brain 5-HT concentrations, which persisted through 21 days postinoculation. Altered brain NE, DA, and 5-HT concentrations suggest that the B. avium-infected poults may be less able to cope with additional stressors.  相似文献

Forty-five Duroc (recognized as not susceptible to stress) and 34 Pietrain (susceptible to stress) pigs were subjected to immobilization stress in a prone position for 5, 15, 30 and 60 min. Plasma concentrations of epinephrine (E), norepinephrine (NE), dopamine (DA) and cortisol (C) were determined in response to restraint stress. The concentrations of E, NE and DA were different between the two strains of pigs (some significant interactions); the highest response was seen after 5 min of stress. The concentration of plasma C increased with duration of stress and there was a significant interaction between strain of animals and the time of stress. Our data substantiate the use of E, NE, DA and C as indicators of stress in swine as early as 5 min after exposure to the stressor. It is also shown that stress-susceptible Pietrain pigs had higher plasma concentrations of E, NE and DA than Duroc pigs.  相似文献

Dopamine (DA) concentrations in the largest ovarian follicle (F1) showed significant decrease at 12 and 8 h prior to ovulation. Nor-epinephrine (NE) and epinephrine (E) concentrations in the F1, sampled at different times of the ovulatory cycle, were found to increase with the approach of ovulation time. However, the concentration of epinephrine in the F1 was low just after oviposition compared with the immediate two previous stages investigated. The concentrations of DA, NE and E in the second largest follicles (F2) did not vary significantly during the ovulatory cycle. Average concentration of DA was low and that of NE and E high in the F1 compared with the respective concentrations in the F2.  相似文献

1. To clarify the difference in behavioural activities and catecholamine metabolism between layer and broiler-type chicks two experiments were conducted. 2. In experiment 1, 1-d-old male layer and broiler chicks were placed in an open-field area and their responses were investigated for 10 min. The responses of the two strains were remarkably different, with broilers being less active than layers. Vocalisations rapidly decreased in broilers whereas those of layers remained elevated during the 10 min. 3. In experiment 2, 1-d-old chicks of both strains were killed and brain catecholamine concentrations were determined in three parts of the brain: telencephalon, optic lobe and brain stem. 4. In the whole brain, dopamine (DA) and dihydroxyphenylacetic acid (DOPAC) were significantly higher in broilers. However, the values for norepinephrine (NE), epinephrine (E) and 4-hydroxy-3-methoxyphenylacetic acid (HVA) were similar between strains. The ratios of metabolite/precursor were also calculated: HVA/DOPAC was higher in layers, while NE/DA, E/NE and DOPAC/DA were not significantly different between strains. 5. These results suggest that behavioural activities differ greatly, while there are some differences in catecholamine metabolism between the two strains.  相似文献

Concentrations of noradrenaline (NA), adrenaline (A), dopamine (DA), 5-hydroxytryptamine (5-HT), the DA metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) and the main 5-HT metabolite 5-hydroxyindole-3-acetic acid (5-HIAA) were measured using HPLC in 16 brain areas of control and immobilized Pietrain pigs. The animals were immobilized for 15, 30 and 60 min in the prone position. Control pigs showed patterns of regional distribution of brain monoamines similar to those described for rats, dogs and Duroc pigs. However, the absolute values of NA and A in the hypothalamus and preoptic area were much higher than those in rats and dogs, but similar to those in Duroc pigs. The concentrations of dopamine and its metabolites DOPAC and HVA were highest in the caudate nucleus, the nucleus accumbens, the olfactory tubercle and the ventral tegmental area. The distributions of serotonin and its metabolite 5-HIAA were similar in all examined structures. DOPAC/DA and HVA/DA ratios were higher in the cornu ammonis, the hippocampus posterior and the raphe nuclei than in other structures, which suggests brain structure-related differences in dopamine turnover. The greatest decreases in NA and A content were observed in the hypothalamus, the preoptic area and the olfactory tuberculum during the first 30 min of immobilization stress. 5-HT turnover was increased in the raphe nuclei, while DA turnover was affected in the raphe nuclei, the septum, the substantia nigra and the olfactory tubercle. We suggest that acute immobilization stress caused differences in regional patterns of brain biogenic amines, thereby maintaining adequate transmitter levels during stress in stress-susceptible pigs.  相似文献

研究早熟与晚熟品种母猪下丘脑-垂体-卵巢轴Kiss1和GPR54基因的表达差异。选用8头梅山母猪与12头长大(LY)母猪为研究对象,梅山母猪于70 d和100 d屠宰,LY母猪于70、100 d和199 d屠宰,收集血清及下丘脑、垂体、卵巢组织样品。ELISA检测血清瘦素(Leptin)和雌二醇(E2)水平,PCR克隆梅山与LY母猪Kiss1基因编码序列,实时荧光定量PCR检测母猪下丘脑、垂体、卵巢组织Kiss1和GPR54基因表达水平。结果表明:梅山与LY母猪Kiss1基因编码序列相似性为100%;梅山与LY母猪初情期下丘脑Kiss1基因表达量极显著高于垂体与卵巢(P<0.01),卵巢GPR54基因表达水平显著高于下丘脑与垂体(P<0.05)。梅山母猪下丘脑-垂体-卵巢轴Kiss1基因表达水平都显著高于相同日龄和初情期LY母猪(P<0.05),梅山母猪血清Leptin水平极显著高于相同日龄LY母猪(P<0.01)。而E2水平显著高于100 d与初情期LY母猪(P<0.01)。Leptin与下丘脑Kiss1和GPR54基因表达呈显著正相关(P<0.05),而E2仅与下丘脑Kiss1基因表达有极显著正相关关系(P<0.01);梅山与LY母猪Kiss1基因编码区序列相似性为100%,梅山母猪下丘脑-垂体-卵巢轴上kiss1基因表达量较LY母猪高,主要原因可能是初情日龄早,下丘脑Kiss1基因表达水平的高低与血清Leptin和E2浓度密切相关。  相似文献

Isolated sheep lung parenchymal strips responded to histamine > carbachol > PGF2a > 5-HT with contractions, and to isoproterenol (Isop), and to large doses of epinephrine (E), norepinephrine (NE) and phenylephrine (PE) with relaxations. PGF2a-contracted lung strip responded to PGE1 and PGE2 with relaxation. The strips which were partially contracted to histamine, PGF2a, 5-HT and carbachol also responded to isop, E and NE with relaxations. Histamine responses were not modified by metiamide (an H2-receptor antagonist). Mepyramine and atropine selectively antagonized contractions to histamine and carbachol, respectively. After β-blockade with propranolol, lung strips responded to NE > E > PE > isop with contractions, which were inhibited or reversed by phentolamine and dibenzyline. It is concluded that H1 receptors are present in sheep peripheral airway smooth muscles, and that a- and β-adrenoceptors mediate contraction and relaxation, respectively, in sheep lung strips.  相似文献

为探讨蛋鸡攻击行为与五羟色胺、皮质酮的相关性,选用1日龄京白939雏鸡96只,随机分为6个重复,每重复16只,采用瞬时观察法和连续观察法筛选不同周龄攻击行为有差异的蛋鸡,结合ELISA和qRT-PCR方法测定蛋鸡血清激素水平和5-HT1A受体mRNA表达量的变化。结果显示:蛋鸡的攻击行为随周龄的增加呈现逐渐上升的趋势(P<0.05);整体看,血清五羟色胺(5-HT)含量随周龄的增加呈下降趋势,皮质酮(CORT)呈现上升趋势(P<0.05),而下丘脑5-HT1A受体mRNA表达量随周龄增加有上升的趋势(P<0.05或P<0.01)。Pearson相关分析结果显示,蛋鸡攻击行为与血清五羟色胺(5-HT)呈极显著负相关(R=-0.741,P<0.01),与皮质酮(CORT)呈显著正相关(R=0.751,P<0.01)。综上所述,血清激素水平与蛋鸡攻击行为有一定联系,5-HT1A受体基因可能参与调控蛋鸡的攻击行为。  相似文献

本试验旨在研究妊娠后期营养限制对蒙古绵羊胎儿生物原胺类神经递质的影响。选择健康、体况相近的蒙古绵羊18只,对其进行同期发情、配种后,从妊娠第90天开始,随机分为3个组:营养限制1组[NG1组,n=6,代谢能(ME)=0.175 MJ/(kg BW 0.75·d)]、营养限制2组[NG2组,n=6,ME=0.330 MJ/(kg BW 0.75·d)]和对照组[CG组,n=6,ME=0.670 MJ/(kg BW 0.75·d)],在妊娠第140天时屠宰,取各组胎儿及其脑组织和血液。结果显示:NG1组胎儿的脑重较CG组显著增高(P<0.05),并且NG1组胎儿脑中5-羟色胺(5-HT)(P<0.01)、肾上腺素(EPI)(P<0.05)含量显著或极显著高于CG组,去甲肾上腺素(NA)的含量极显著低于CG组(P<0.01)。NG2组胎儿脑中5-HT含量极显著高于CG组(P<0.01),而NA(P<0.01)、多巴胺(DA)(P<0.05)含量显著或极显著低于CG组。另外,NG1组胎儿血浆中5-HT(P<0.01)、EPI(P<0.01)、NA(P<0.05)、DA(P<0.01)含量显著或极显著低于CG组;NG2组胎儿血浆中5-HT(P<0.01)、NA(P<0.01)、DA(P<0.05)含量显著或极显著低于CG组。由此得出,妊娠后期营养限制使得蒙古绵羊胎儿脑中5-HT、EPI含量升高和NA含量降低,血浆中5-HT、EPI、NA、DA含量降低。  相似文献

1. Saline (10 microliters), norepinephrine (NE) and Serotonin (5-HT), 500 nmol each, were injected into the anterior third ventricle (A3V; n = 7) or the posterior third ventricle (P3V; n = 11) of ananesthetised, unrestrained White Leghorn cockerels. Plasma arginine vasotocin (AVT) and mesotocin (MT) were measured 20, 60 and 120 min after injection. 2. Injection of NE into both the A3V and P3V had no significant effect on either plasma AVT or plasma MT at any of the sampling times. 3. Administration of 5-HT into the A3V significantly increased plasma MT about two-fold 20 min following injection. At 120 min time, plasma MT returned to normal. 4. In P3V birds, 5-HT had no effect on plasma MT in the first 20 min, but a significant increase in plasma MT occurred 60 to 120 min after injection. The magnitude of the response was lower than in the A3V cockerels. 5. Plasma AVT was not affected by 5-HT administration into the A3V at any of the sampling times, but 5-HT administration into the P3V caused significant rises in plasma AVT at 120 min. 6. Serotonergic, but not noradrenergic, induction of neurohypophysial peptide secretion was demonstrated.  相似文献

Glucagon-like peptide-1 (7-36) amide (GLP-1), secreted by the small intestine, has insulinotropic and glucose-lowering action. Basal plasma GLP-1 concentrations were measured in calves around the weaning period, the effect of short-chain fatty acids (SCFA) on plasma GLP-1 concentrations was examined, and the effects of GLP-1 administration on plasma insulin, glucagon, and glucose concentrations were measured. Thirteen Holstein bull calves were fed whole milk and solid feed and weaned at 7 wk of age. Preprandial plasma samples were obtained from 5 calves once a week from week 0 to 13 to measure basal concentrations of plasma GLP-1 and insulin (experiment 1). Four calves were intravenously administered with a mixed solution of SCFA (2.4 mmol/kg body weight [BW]) in week 2 and 11 to measure plasma GLP-1 concentrations (experiment 2). Another 4 calves were intravenously injected with GLP-1 (1.0 μg/kg BW) to elucidate the response of plasma insulin, glucagon, and glucose concentrations in week 1, 2, 4, 6, 7, 9, 11, and 13 (experiment 3). In experiment 1, age and weaning did not affect preprandial basal concentrations of plasma GLP-1 throughout the experimental period. Preprandial insulin concentrations increased after weaning (P < 0.05), and GLP-1 and insulin were more strongly correlated postweaning than preweaning. In experiment 2, intravenous treatment with SCFA increased plasma GLP-1 concentrations in both week 2 and 11 (P < 0.05.) In experiment 3, intravenous GLP-1 treatment decreased plasma glucose concentrations throughout the experiment (P < 0.05), but increased plasma insulin concentrations only after weaning (P < 0.05). Treatment with GLP-1 did not affect plasma glucagon concentrations, regardless of age. These results indicate that preprandial basal concentrations of plasma GLP-1 in calves are not changed by weaning, but SCFA stimulate GLP-1 secretion. The insulinotropic action of GLP-1 is detected only after weaning, but the glucose-lowering action of GLP-1 is not affected by weaning.  相似文献

In order to clarify the role of serotonin (5-HT) in the regulation of pituitary hormones, the effects of 5-HT injected into the third ventricle (3V) on prolactin (PRL) and growth hormone (GH) release were investigated in Holstein steers. A chronic cannula was implanted in 3V by stereotaxic surgery under general anesthesia. After sufficient recovery from surgery, 5-HT (0, 0.1, 1.0, 2.0 mg) was injected into via the cannula and blood samples were collected over 4 h. Plasma PRL and GH concentrations were determined by radioimmunoassay. PRL release was significantly stimulated by the injection of 5-HT. The increase in PRL was observed at 20 min after the injection at three doses and the highest dose (2.0 mg) was the most effective in stimulating PRL release. The injection of 5-HT into 3V, at all doses tested, did not alter GH release significantly. Our results suggest that 5-HT is involved in the regulation of PRL release partly through the hypothalamus in cattle.  相似文献

Two experiments were conducted to compare the effects of feeding a blend of grains naturally contaminated with Fusarium mycotoxins on brain regional neurochemistry of starter pigs and broiler chickens. A polymeric glucomannan mycotoxin adsorbent (GM polymer) was also tested for its efficacy in preventing Fusarium mycotoxicoses. In Exp. 1, a total of 150 starter pigs (initial weight = 9.3+/-1.1 kg) were fed five diets (six pens of five pigs per diet) for 21 d. Diets (as-fed basis) included control, 17% contaminated grains, 24.5% contaminated grains, 24.5% contaminated grains + 0.2% GM polymer, and a pair-fed control for comparison with pigs receiving 24.5% contaminated grains. In Exp. 2,360 1-d-old male broiler chicks were fed for 56 d one of four diets containing the same source of contaminated grains as was fed to pigs. The diets included control, 37% contaminated grains, 58% contaminated grains, and 58% contaminated grains + 0.2% GM polymer (as fed). Neurotransmitter concentrations in the cortex, hypothalamus, and pons were analyzed by HPLC. The following brain neurotransmitter alterations (P < or = 0.05) were observed. In pigs, inclusion of contaminated grains in the diet 1) linearly increased cortex 5-hydroxytryptamine (5HT, serotonin) concentrations, while linearly decreasing hypothalamic tryptophan concentrations; 2) quadratically increased hypothalamic and pons 5-hydroxyindoleacetic acid (5HIAA):5HT ratios, whereas the ratio decreased linearly in the cortex; and 3) linearly increased the ratio of hypothalamic 3,4-dihydroxyphenylacetic acid:dopamine (DA) concentrations, whereas hypothalamic norepinephrine (NRE) and pons DA and homovanillic acid (HVA) concentrations linearly decreased. In broiler chickens, inclusion of contaminated grains in the diet 1) linearly increased concentrations of 5HT and 5HIAA in the pons and 5HT concentrations in the cortex; 2) linearly decreased 5HIAA:5HT ratio; and 3) linearly increased pons NRE, 3-methoxy-4-hydroxyphenylethylene glycol, DA, and HVA concentrations. Supplementation of GM polymer to the contaminated diet decreased (P < 0.05) 5HT and 5HIAA concentrations in the cortex of pigs. It was concluded that the differences in alterations of brain neurochemistry might explain the species differences in the severity of Fusarium mycotoxin-induced feed refusal.  相似文献

Incretin hormones are secreted from the intestines in response to specific nutrients. They potentiate insulin secretion and have other beneficial effects in glucose homeostasis. We aimed to study the incretin effect in cats and to compare the effect of oral glucose, lipids, or amino acids on serum concentrations of insulin, total glucose-dependent insulinotropic peptide (GIP) and total glucagon-like peptide 1 (GLP-1). Ten healthy cats were used in a repeated measures design. Glucose, lipid, or amino acids were administered through nasoesophageal tubes on separate days. Blood glucose (BG) concentrations were matched between experiments by measuring BG every 5 min and infusing glucose intravenously at a changing rate. Intravenous glucose infusion with no prior treatment served as control. The incretin effect was estimated as the difference in insulin area under the curve (AUC) after oral compared with intravenous glucose. Temporal changes and total amount of hormone secretions were compared between treatment groups with the use of mixed models. Total glucose infused (TGI) at a mean dose of 0.49 g/kg resulted in slightly higher BG compared with 1 g/kg oral glucose (P = 0.038), but insulin concentrations were not significantly different (P = 0.367). BG and the TGI were not significantly different after the 3 oral challenges. Total GIP AUC was larger after lipids compared with amino acids (P = 0.0012) but GIP concentrations did not increase after oral glucose. Insulin and GIP concentrations were positively correlated after lipid (P < 0.001) and amino acids (P < 0.001) stimulations, respectively, but not after oral glucose stimulation. Total GLP-1 AUC was similar after all three oral stimulations. Insulin and GLP-1 concentrations were positively correlated after glucose (P = 0.001), amino acids (P < 0.001), or lipids (P = 0.001) stimulations. Our data indirectly support an insulinotropic effect of GIP and GLP-1. Potentiation of insulin secretion after oral glucose is minimal in cats and is mediated by GLP-1 but not GIP.  相似文献
