A host resistance factor was integrated into the FAST (Forecaster ofAlternariasolani onTomatoes) system to initiate and to schedule fungicide applications for three levels of potato cultivar susceptibility while maintaining disease control and yield. Four cultivars ranging from susceptible to moderately resistant were each treated with the fungicide chlorothalonil during two field seasons. The five fungicide treatments were a 7-day schedule initiated after 50% flowering; a non-sprayed control; and three FAST application schedules based on combinations of critical levels of cumulative severity (CS, the disease severity values as a function of leaf wetness and temperature) and cumulative rating (CR, the disease rating values as a function of average temperature and hours of relative humidity greater than 90%) values. The FAST system predicted the onset of early blight and subsequent periods conducive for disease development in 1993 and 1994. There were cultivar-by-spray-schedule interactions for both disease severity and yield, suggesting that differences in early blight sus-ceptibility exist between cultivars. Use of the recommended schedules for the cultivar’s targeted level of susceptibility reduced early blight severity and minimized fungicide use while maintaining yield. These results indicate that a host-resistance factor can be incorporated into the FAST system.  相似文献

Three potato cultivars (Russet Burbank, Norchip, and Gemchip) grown with nitrogen applied at three rates were stored at two temperature regimes (Treatment 1: 13 months at 10 CTreatment 2: 1 month at 10 C; followed by a 1 C decrease per week until tubers were 4 C; followed by 6 months at 4 C; followed by a 1 C increase per week until tubers were 10 C; followed by 3 months at 10 C). Tuber chemical components and potato chip appearance were measured at harvest and after 3, 6, 9, 11, 12, and 13 months; these measurements were performed within 24 hours of the time potatoes were removed from storage. Sugar responses (tuber glucose, fructose, sucrose) and potato chip appearance were affected by cultivar over time in both years and storage temperatures. Russet Burbank tubers displayed a significantly higher glucose forming potential and produced darker appearing chips, regardless of storage temperature or time in storage, compared to Norchip and Gemchip. Potatoes receiving a cold-storage treatment contained less sugar and produced lighter appearing chips after 12 months storage compared to tubers stored at a constant 10 C for 12 months. The linear association between tuber chemical components and potato chip appearance varied with storage temperature.  相似文献

Background

The Magel2 gene is most highly expressed in the suprachiasmatic nucleus of the hypothalamus, where its expression cycles in a circadian pattern comparable to that of clock-controlled genes. Mice lacking the Magel2 gene have hypothalamic dysfunction, including circadian defects that include reduced and fragmented total activity, excessive activity during the subjective day, but they have a normal circadian period. Magel2 is a member of the MAGE family of proteins that have various roles in cellular function, but the specific function of Magel2 is unknown.

Methods

We used a variety of cell-based assays to determine whether Magel2 modifies the properties of core circadian rhythm proteins.

Results

Magel2 represses the activity of the Clock:Bmal1 heterodimer in a Per2-luciferase assay. Magel2 interacts with Bmal1 and with Per2 as measured by co-immunoprecipitation in co-transfected cells, and exhibits a subcellular distribution consistent with these interactions when visualized by immunofluorescence. As well, Magel2 induces the redistribution of the subcellular localization of Clock towards the cytoplasm, in contrast to the nucleus-directed effect of Bmal1 on Clock subcellular localization.

Conclusion

Consistent with the blunted circadian rhythm observed in Magel2-null mice, these data suggest that Magel2 normally promotes negative feedback regulation of the cellular circadian cycle, through interactions with key core circadian rhythm proteins.  相似文献

Russet Burbank and Shepody potatoes were grown with at-planting nitrogen fertilizer rates varying from 0 to 270 kg ha?1 following small grains and red clover. Petiole samples were collected from the 4th and 5th leaflets at four to six dates each during the 1986 to 1989 growing seasons. The samples were dried and analyzed for NO 3 ? N content. Petiole NO 3 ? N levels were strongly related to nitrogen rate regardless of cultivar, growing season, and cropping system. Differences among treatments and NO 3 ? N content varied substantially as the growing season progressed with petiole NO 3 ? N levels declining rapidly in underfertilized plots as the plants aged. Petiole NO 3 ? N levels were higher at midseason following red clover than following small grains. Sampling 50 to 60 days after planting (DAP) is most appropriate as a tool for scheduling supplemental nitrogen applications. At 50 DAP, critical petiole NO 3 ? N levels were 1.6 and 1.7% for Russet Burbank and Shepody, respectively. Petiole NO 3 ? N levels above 2.2% at 50 DAP resulted in lower yields of Russet Burbank than when petiole NO 3 ? N levels were in the 1.6 to 2.2% range. Petiole NO 3 ? N testing should be particularly useful as a diagnostic tool in management strategies which make maximum use of previous crop residues, organic amendments, and soil reserves as nitrogen sources.  相似文献

Background

An endogenous circadian clock controls locomotor activity in common spiny mice (Acomys cahirinus). However, little is known about the effects of constant light (LL) on this activity or about the existence of an additional food entrainable clock. A series of experiments were performed to investigate the effects of LL and DD on tau and activity levels.

Methods

Spiny mice were housed individually and their running wheel activity monitored. One group of mice was exposed to LD, DD and several intensities of LL. Another group was exposed to a restricted feeding (RF) paradigm in light: dark (LD) during one hour before the L to D transition. Significance of rhythmicity was assessed using Lomb-Scargle periodograms.

Results

In LD all animals exhibited nocturnal activity rhythms that persisted in DD. When animals were exposed to RF (during L), all of these animals (n?=?11) demonstrated significant food anticipatory activity as well as an increase in diurnal activity. This increase in diurnal activity persisted in 4/11 animals during subsequent ad libitum conditions. Under LL conditions, the locomotor rhythms of 2/11 animals appeared to entrain to RF. When animals were exposed to sequentially increasing LL intensities, rhythmicity persisted and, while activity decreased significantly, the free-running period was relatively unaffected. In addition, the period in LL was significantly longer than the period in DD. Exposure to LL also induced long-term changes (after-effects) on period and activity when animals were again exposed to DD.

Conclusions

Overall these studies demonstrate clear and robust circadian rhythms of wheel-running in A. cahirinus. In addition, LL clearly inhibited activity in this species and induced after-effects. The results also confirm the presence of a food entrainable oscillator in this species.  相似文献

Background

Physical activity as measured by activity counts over short time intervals across a 24 h period are often used to assess circadian variation. We are interested in characterizing circadian patterns in activity among adolescents and examining how these patterns vary by obesity status. New statistical approaches are needed to examine how factors affect different features of the circadian pattern and to make appropriate covariate adjustments when the outcomes are longitudinal count data.

Methods

We develop a statistical model for longitudinal or repeated activity count data that is used to examine differences in the overall activity level, amplitude (defined as the difference between the lowest and highest activity level over a 24 hour period), and phase shift. Using seven days of continuous activity monitoring, we characterize the circadian patterns and compare them between obese and non-obese adolescent girls.

Results

We find a statistically significant phase delay in adolescent girls who were obese compared with their non-obese counterparts. After the appropriate adjustment for measured potential confounders, we did not find differences in mean activity level between the two groups.

Conclusion

New statistical methodology was developed to identify a phase delay in obese compared with non-obese adolescents. This new approach for analyzing longitudinal circadian rhythm count data provides a useful statistical technique to add to the repertoire for those analyzing circadian rhythm data.  相似文献

The β0 and ξ, (Y intercept and error term) and β1, β2 and β3 coefficients are derived from fitting the experimental data. The model was developed by growing ‘Kennebec’ in large containers at sites ranging from 1533 m to 3198 m elevation under shaded (48% insolation reduction) and unshaded conditions at 39° to 41° N latitude. Mean maximum and minimum temperatures and insolation ranged from 29°C to 19°C, 14°C to 6°C, and 200 cal cm?2 day?1 to 530 cal cm2 day?1 respectively. Soil matric potential and soil fertility were not included as variables in the model since they were physically controlled. The model does not include the period from planting to emergence since environment was not permitted to vary differentially. A highly significant multiple linear model with a coefficient of determination of 0.93 was obtained. It is suggested that including the revision $$ - \frac{{min temp \left( { > 4.4^ \circ C} \right) - 4.4^ \circ C}}{2}$$ in the heat input estimation (GDD) as well as air temperature range, emphasizes the influence of respiration on productivity. The model should be tested further and adapted as a practical method for predicting potato yield under “grower” conditions.  相似文献

Nipigon is a high-yielding, early sizing, table potato with good boiling and baking quality and is specifically adapted to northern Ontario. It has some resistance to late blight, rhizoctonia and viruses X and Y. The Nipigon potato was bred and initially selected within the Potato Breeding Project at the Fredericton Rcsearch Station of Agriculture Canada. This variety has been under test as seedling F61051 since 1965 throughout eastern Canada, but mostly in Ontario. Nipigon was selected from the progeny of a cross between two Fredericton seedlings; F53026 an early, scab-resistant seedling with very good tuber type and F51043, which has good processing quality and resistance to the common races of late blight,Phytophthora infestans (Mont.) de Bary. Background

Ataxia with oculomotor apraxia type 1 (AOA1) shows early onset with autosomal recessive inheritance and is caused by a mutation in the aprataxin (APTX) gene encoding for the APTX protein.

Methods

In this study, a 7-year-old girl born of a first-cousin consanguineous marriage was described with early-onset progressive ataxia and AOA, with increased cholesterol concentration and decreased albumin concentration in serum. PCR and direct DNA sequencing was performed after DNA extraction.

Results

Sequencing analysis revealed a novel homozygous deletion in c.643 and A>T single nucleotide polymorphism in c.641 in exon 6 of the APTX gene [ENST00000379825].

Conclusion

Three different fucoidan fractions were isolated and purified from the brown alga, Sargassum mcclurei. The SmF1 and SmF2 fucoidans are sulfated heteropolysaccharides that contain fucose, galactose, mannose, xylose and glucose. The SmF3 fucoidan is highly sulfated (35%) galactofucan, and the main chain of the polysaccharide contains a →3)-α-l-Fucp(2,4SO3)-(1→3)-α-l-Fucp(2,4SO3)-(1→ motif with 1,4-linked 3-sulfated α-l-Fucp inserts and 6-linked galactose on reducing end. Possible branching points include the 1,2,6- or 1,3,6-linked galactose and/or 1,3,4-linked fucose residues that could be glycosylated with terminal β-d-Galp residues or chains of alternating sulfated 1,3-linked α-l-Fucp and 1,4-linked β-d-Galp residues, which have been identified in galactofucans for the first time. Both α-l-Fucp and β-d-Galp residues are sulfated at C-2 and/or C-4 (and some C-6 of β-d-Galp) and potentially the C-3 of terminal β-d-Galp, 1,4-linked β-d-Galp and 1,4-linked α-l-Fucp residues. All fucoidans fractions were less cytotoxic and displayed colony formation inhibition in colon cancer DLD-1 cells. Therefore, these fucoidan fractions are potential antitumor agents.  相似文献

Two cell-wall-associated polysaccharides were isolated and purified from the deep-sea marine bacterium Devosia submarina KMM 9415T, purified by ultracentrifugation and enzymatic treatment, separated by chromatographic techniques, and studied by sugar analyses and NMR spectroscopy. A new ultrasound-mediated approach has been developed to introduce neomycin-resistance to activate silent pathways for secondary metabolite production in a bio-inactive, deep-sea fungus, Aspergillus versicolor ZBY-3. Upon treatment of the ZBY-3 spores with a high concentration of neomycin by proper ultrasound irradiation, a total of 30 mutants were obtained by single colony isolation. The acquired resistance of the mutants to neomycin was confirmed by a resistance test. In contrast to the ZBY-3 strain, the EtOAc extracts of 22 of the 30 mutants inhibited the human cancer K562 cells, indicating that these mutants acquired a capability to produce antitumor metabolites. HPLC-photodiode array detector (PDAD)-UV and HPLC-electron spray ionization (ESI)-MS analyses of the EtOAc extracts of seven bioactive mutants and the ZBY-3 strain indicated that diverse secondary metabolites have been newly produced in the mutant extracts in contrast to the ZBY-3 extract. The followed isolation and characterization demonstrated that six metabolites, cyclo(d-Pro-d-Phe) (1), cyclo(d-Tyr-d-Pro) (2), phenethyl 5-oxo-l-prolinate (3), cyclo(l-Ile-l-Pro) (4), cyclo(l-Leu-l-Pro) (5) and 3β,5α,9α-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one (6), were newly produced by the mutant u2n2h3-3 compared to the parent ZBY-3 strain. Compound 3 was a new compound; 2 was isolated from a natural source for the first time, and all of these compounds were also not yet found in the metabolites of other A. versicolor strains. Compounds 1–6 inhibited the K562 cells, with inhibition rates of 54.6% (1), 72.9% (2), 23.5% (3), 29.6% (4), 30.9% (5) and 51.1% (6) at 100 μg/mL, and inhibited also other human cancer HL-60, BGC-823 and HeLa cells, to some extent. The present study demonstrated the effectiveness of the ultrasound-mediated approach to activate silent metabolite production in fungi by introducing acquired resistance to aminoglycosides and its potential for discovering new compounds from silent fungal metabolic pathways. This approach could be applied to elicit the metabolic potentials of other fungal isolates to discover new compounds from cryptic secondary metabolites.  相似文献

Jemseg is an early sizing variety of early maturity. It has outyielded the variety Warba for total and marketable yield when dug at 80 days from planting. It has moderate resistance to common scab, is immune to virus X and has high resistance to virus Y, plus resistance to virus S. Jemseg was originated at Fredericton, New Brunswick, and is a product of the potato breeding program of Agriculture Canada. The variety has been under test in New Brunswick for 10 years as seedling F67072. It has also been tested extensively in the Maritime Provinces and throughout Canada through the facilities of regional and national evaluation trials. Jemseg was selected from a progeny of a cross between the variety Sable and a Fredericton seedling F55069. Sable is an early variety developed at Fredericton. The use of fluoride enriched groundwater for irrigating the paddy crops is prevalent in many parts of the world. The fluoride in the irrigated water not only affects the growth and productivity of the crops, but is also accumulated in the plant tissues. We studied the accumulation of fluoride in roots, leaves and seeds two paddy varieties (i.e. Oryza sativa L. var. IR-36 and Oryza sativa L. var. Swarno) when treated with different concentrations of fluoridated water. The translocation pattern of fluoride was also observed. The water soluble fluoride ( $ {\text{F}}_{{{\text{H}}_{ 2} {\text{O}}}} $ ) in the soil and plant parts increased substantially with increasing fluoride concentration in treated water. The Swarno variety showed a slight but consistent higher uptake of fluoride than the IR-36 variety. Fluoride uptake by the germinated seedlings was many folds higher than the uptake by mature plants. At 30 mg l?1 fluoride exposure, the mean $ {\text{F}}_{{{\text{H}}_{ 2} {\text{O}}}} $ accumulation (mg kg?1 dry weight) in root, leaves, and seeds of Swarno was 54.1, 51.4 and 42.3, whereas the corresponding values in IR-36 were 50.9, 48.5 and 39.2. For the same exposure, the fluoride accumulation in root and shoot of Swarno and IR-36 seedlings were 3,480, 3,463 and 3,386, 3,360 mg kg?1, respectively. Normally, the fluoride accumulation follows the order of soil > root > shoot > grain. However, at early stage of fluoride contamination (5 mg l?1 NaF) roots tended to hyper accumulate fluoride from the soil.  相似文献

Background

Variation in circadian rhythms and nocturnality may, hypothetically, be related to or independent of genetic variation in photoperiodic mediation of seasonal changes in physiology and behavior. We hypothesized that strain variation in photoperiodism between photoperiodic F344 rats and nonphotoperiodic Harlan Sprague Dawley (HSD) rats might be caused by underlying variation in clock function. We predicted that HSD rats would have more activity during the day or subjective day, longer free-running rhythms, poor entrainment to short day length, and shorter duration of activity, traits that have been associated with nonphotoperiodism in other laboratory rodent species, relative to F344 rats. An alternative hypothesis, that differences are due to variation in melatonin secretion or responses to melatonin, predicts either no such differences or inconsistent combinations of differences.

Methods

We tested these predictions by examining activity rhythms of young male F344 and HSD rats given access to running wheels in constant dark (DD), short day length (L8:D16; SD), and long day length (L16:D8; LD). We compared nocturnality (the proportion of activity during night or subjective night), duration of activity (alpha), activity onset and offset, phase angle of entrainment, and free running rhythms (tau) of F344 and HSD rats.

Results

HSD rats had significantly greater activity during the day, were sometimes arrhythmic in DD, and had significantly longer tau than F344 rats, consistent with predictions. However, HSD rats had significantly longer alpha than F344 rats and both strains entrained to SD, inconsistent with predictions.

Conclusion

The ability of HSD rats to entrain to SD, combined with longer alpha than F344 rats, suggests that the circadian system of HSD rats responds correctly to SD. These data offer best support for the alternative hypothesis, that differences in photoresponsiveness between F344 and HSD rats are caused by non-circadian differences in melatonin secretion or the response to melatonin.  相似文献

The single groups are discussed with regard to their contents of amino acids. They are compared with the pattern of amino acids found in mothers' milk, so allowing to deduce typical patterns for each group. In this connection it is of great interest that legumes alter the pattern with the stage of ripening. With a decrease in contents of lysine a tendency of increase in methionine and tryptophane is observed. This is distinctly marked in comparing proteins of legumes with those of cereals. The composition of a ‘typical plant protein’ has been calculated considering the whole analytical material. This in turn has been related to the use of consumption in the BRD considering also the share of proteins delivered from the different plant groups. This protein shows in comparison with that of mothers' milk a very favorable composition (EAA-Index (9 EAA) according to Oser = 81) and confirmed the statement being of good use for human nutrition.  相似文献

Background

Recent studies on humans and rodents have suggested that the timing of food intake plays an important role in circadian regulation and metabolic health. Consumption of high-fat foods during the inactive period or at the end of the awake period results in weight gain and metabolic syndrome in rodents. However, the distinct effects of breakfast size and the breakfast/dinner size ratio on metabolic health have not yet been fully examined in mice.

Methods

We examined whether the parameters of metabolic syndrome were differentially affected in mice that consumed a large meal at the beginning of the awake period (breakfast; one meal group) and a relatively smaller meal at end of the awake period (dinner; two meals group). The mice of each group were provided equal food volume per day.

Results

Mice on one meal exhibited an increase in body weight gain, hyperinsulinemia, hyperleptinemia, and a decrease of gene expression associated with ??-oxidation in adipose tissue and liver compared with those on two meals. The circadian expression pattern of the Clock gene in mice on one meal was disturbed compared with those on two meals.

Conclusions

In conclusion, a bigger breakfast with a smaller dinner (two meals per day) but not breakfast only (one meal per day) helps control body weight and fat accumulation in mice on a high-fat meals schedule. The findings of this study suggest that dietary recommendations for weight reduction and/or maintenance should include information on the timing and quantity of dietary intake.  相似文献
