The study was aimed to assess the influence that short‐term progesterone treatments have on follicular dynamics, oestrus and ovulation in sheep. The treatment was tested thereafter in a field trial to assess its fertility after AI with fresh semen. In a first experiment, 12 ewes without CL were grouped to receive a new (n = 6) or used CIDR (n = 6) for 7 days and blood samples were obtained to follow plasma progesterone profiles. In a second experiment, 39 cycling ewes were synchronized by a 7‐day P4+PGF2α protocol using a new (n = 20) or a 7‐day used CIDR (n = 19). Half of both groups received 400 IU eCG and half remained untreated as controls. Ultrasound ovarian examination and oestrous detection were used to compare follicular dynamics, oestrus and ovulation in both groups. In a third experiment, 288 ewes in 3 farms were synchronized by the short‐term P4+PGF2α+eCG protocol and ewes were AI with fresh semen 24 h after oestrous detection. Lambing performance was used to test the fertility of the treatment. In Experiment 1, ewes with new inserts presented higher P4 concentration than ewes with used inserts throughout the sampling period (p < 0.05) and exhibited a P4 peak at days 1‐2 of the treatment that was not observed in ewes with used inserts. In Experiment 2, ewes treated with new and used inserts show similar ovarian and behavioral traits (p > 0.10). However, ewes treated with eCG show shorter interval to oestrus (p = 0.004) and tend to have larger mature CL (p = 0.06). In Experiment 3, oestrous presentation and lambing performance after AI with fresh semen was considered normal compared to published results. Results suggest that the oestrous synchronization protocol based on P4+PGF2α allows little control of follicular dynamics without compromising fertility after AI with fresh semen provided that eCG is added at the end of the treatment.  相似文献

Traditional treatments of two prostaglandin F2 alpha (PGF2alpha) doses at 10-day intervals or more did not result in acceptable pregnancy rates in timed artificial insemination (TAI) programmes in ewes. An explanation might be the undefined time-period of the onset of oestrus and ovulation after the treatment. Recently a consistent interval to oestrus and ovulation was obtained by giving PGF2alpha at day 3 post-ovulation, i.e. when the largest follicle of the first follicular wave of the cycle was still growing. This can be achieved when a second dose of PGF2alpha is given 7 days after a first dose. In this work, we evaluated the synchronization of oestrus and determined which of three different moments of TAI was the most successful using a PGF2alpha (PG-7d) protocol in a large flock. A total of 436 nulliparous and multiparous ewes were treated with two doses of a PGF2alpha analogue (delprostenate 160 microg, i.m.) separated by 7 days. Onset of oestrus was recorded twice a day and a single cervical TAI with fresh undiluted semen was performed either at 42 h (n = 152), 48 h (n = 120), or 54 h (n = 164), after the second PGF2alpha dose without taking into account the oestrous response. Pregnancy rate was determined by transrectal ultrasonography 30 days after insemination. Onset of oestrus was detected in 308 of 328 and 89 of 108 multiparous and nulliparous ewes, respectively (p < 0.001), within 72 h after treatment. The distribution of the onset of oestrus did not differ between multiparous and nulliparous ewes and the highest proportion of ewes in oestrus was detected between 25 to 48 h (313/397) from the second PGF2alpha dose. The pregnancy rate in ewes inseminated at 42 h tended to be higher than those inseminated at 48 h (p = 0.09) and was higher than those inseminated at 54 h (p < 0.05) (56/152, 31/120, 37/164; respectively). Therefore, the use of the PG-7d protocol resulted in a very high synchronization of oestrus with the highest concentration (around 80%) between 25 to 48 h from the end of treatment. The best pregnancy rate (37%) was obtained after a single cervical TAI with fresh semen at 42 h.  相似文献

AIM: To determine if the reproductive performance of dairy cows not previously detected in oestrus but with a detectable corpus luteum before the planned start of mating (PSM), could be improved by treatment with progesterone, oestradiol benzoate (ODB) and prostaglandin F2alpha (PGF). METHODS: Cows in 18 herds which had not been detected in oestrus, but which had a detectable corpus luteum present at veterinary examination 7 days prior to the PSM (Day -7), were allocated to 1 of 2 groups. Treated cows (n=232) received an injection of 2 mg ODB and an intravaginal progesterone releasing device (CIDR insert) on Day -7, and an injection of PGF on the day of insert removal 7 days later (Treated group). The Control group (n=243) remained untreated. Cows were mated to detected oestrus from Day 0, and conception dates confirmed by manual palpation or transrectal ultrasonography. RESULTS: During the first 7 days of mating, 37.4% of Control cows and 65.9% of Treated cows were inseminated on detection of oestrus (p<0.001). Pregnancy rates for this period were 20.4% and 36.3%, respectively (p=0.001). Conception rates to first insemination, pregnancy rates after 21 days of mating and at the end of the mating period were similar between groups (p>0.1). Median interval from the PSM to conception did not differ between treatment groups (24 and 23 days for Control and Treated, respectively, p>0.1). CONCLUSION: Treating postpartum dairy cows which had not previously been detected in oestrus but which had a detectable corpus luteum, with progesterone, ODB and PGF did not significantly improve their reproductive performance compared with no hormonal intervention. A study was undertaken to determine the oestrus response and fertility rates of zebu cows treated with PRID alone or in combination with PGF2alpha. A total of 184 non-suckled cycling Bunaji cows were allotted randomly to four treatment groups of 46 animals per group as follows: group 1 (PRID-12), PRID was inserted for 12 days; group 2 (PRID+7+PGF2alpha-6, PRID was inserted for 7 days and PGF2alpha was administered intramuscularly 1 day prior to PRID withdrawal; group 3 (PRID-7+PGF2alpha-7, PRID was inserted for 7 days and PGF2alpha was administered intramuscularly on the day of PRID withdrawal; group 4 (2 x PGF2alpha-13, two intramuscular injections of PGF2alpha 13 days apart. At the end of each treatment period, the cows were observed for 7 days for behavioural oestrus and were inseminated 12 h following detection of oestrus. Pregnancy was diagnosed by rectal palpation 30-40 days post-insemination. The respective oestrus response rates were 78.3%, 76.1%, 87.0% and 89.1% for groups 1-4. While the corresponding pregnancy rates were 39.1%, 41.3%, 52.2% and 52.2%, the conception rates were 50.0%, 54.3%, 60.3% and 58.6% for groups 1-4. Although individual variations in progesterone levels were observed, the progesterone profiles were generally typical and normal. The results of the study have confirmed the effectiveness of the four regimes in synchronizing and controlling oestrus and ovulation in Bunaji cows. However, groups 3 and 4 showed some superiority over the other treatments. The results of this study provide feasible options from which clinicians involved in intensive breeding programmes and herd health fertility programmes can choose.  相似文献

The objective was to test the efficacy of an intravaginal progesterone insert and injection of PGF2alpha for synchronizing estrus and shortening the interval to pregnancy in cattle. Cattle were assigned to one of three treatments before a 31-d breeding period that employed artificial insemination. Control cattle were not treated, and treated cattle were administered PGF2alpha or an intravaginal progesterone-releasing insert (CIDR) for 7 d and treated with PGF2alpha on d 6. The treatments were applied in one of three experiments that involved postpartum beef cows (Exp. 1; n = 851; 56+/-0.6 d postpartum), beef heifers (Exp. 2; n = 724; 442.5+/-2.8 d of age), and dairy heifers (Exp. 3; n = 260; 443.2+/-4.5 d of age). Luteal activity before treatment was determined for individual cattle based on blood progesterone concentrations. In Exp. 1, there was a greater incidence of estrus during the first 3 d of the breeding period in CIDR+PGF2alpha-treated cows compared with PGF2alpha-treated or control cows (15, 33, and 59% for control, PGF2alpha, and CIDR+PGF2alpha, respectively; P < 0.001). The improved estrous response led to an increase in pregnancy rate during the 3-d period (7, 22, and 36% for control, PGF2alpha, and CIDR+PGF2alpha, respectively; P < 0.001) and tended to improve pregnancy rate for the 31-d breeding period for cows treated with CIDR+PGF2alpha, (50, 55, and 58% for control, PGF2alpha, and CIDR+PGF2alpha, respectively, P = 0.10). Improvements in rates of estrus and pregnancy after CIDR+PGF2alpha, were also observed in beef heifers. Presence of luteal activity before the treatment period affected synchronization and pregnancy rates because anestrous cows (Exp. 1) or prepubertal heifers (Exp. 2) had lesser synchronization rates and pregnancy rates during the first 3 d of the breeding period as well as during the entire 31-d breeding period. The PGF2alpha, and CIDR+PGF2alpha but not the control treatments were evaluated in dairy heifers (Exp. 3). The CIDR+PGF2alpha-treated heifers had a greater incidence of estrus (84%) during the first 3 d of the breeding period compared with the PGF2alpha-treated heifers (57%), but pregnancy rates during the first 3 d or during the 31-d breeding period were not improved for CIDR+PGF2alpha compared with PGF2alpha-treated heifers. In summary, the concurrent treatment of CIDR and PGF2alpha improved synchronization rates relative to PGF2alpha alone or control. Improved estrus synchrony led to greater pregnancy rates for beef cows and beef heifers but failed to improve pregnancy rates for dairy heifers.  相似文献

Prostaglandin F2 alpha (PGF2 alpha) was injected on d 5, 8 or 11 postestrus in ewes to determine how stage of the estrous cycle would affect PGF2 alpha-induced changes in concentrations of ovarian and pituitary hormones and intervals to the onset of estrus and the preovulatory surge of luteinizing hormone (LH). Initial concentrations of progesterone and average values during the 12 h after PGF2 alpha were related positively to the day of cycle on which PGF2 alpha was administered. Patterns of decline in progesterone after injection of PGF2 alpha were similar among the 3 d. Concentrations of LH in plasma increased in a similar manner from 0 to 12 h in all ewes. After 12 h LH continued to increase, plateaued or declined in ewes treated on d 5, 8 or 11, respectively. Initial concentrations of follicle stimulating hormone (FSH) in plasma were related positively to day of treatment. After treatment with PGF2 alpha, FSH increased within 2 h on d 5 but declined by that time on d 8 or 11. Concentrations of estradiol following treatment did not vary with day. The onset of estrus and the preovulatory surge of LH occurred at 36 and 35, 40 and 45, and 48 and greater than 48 h in ewes treated on d 5, 8 or 11, respectively. It is concluded that: 1) the initial increase in LH is dependent on a decrease in plasma progesterone and 2) differences in patterns of secretion of gonadotropins before the preovulatory surge of LH might be caused by differences in progesterone or progesterone:-estradiol ratio when luteal regression is induced on different days of the estrous cycle.  相似文献

The study investigated, for cycling sheep, synchronizing protocols simultaneously to the standard “P” protocol using progestogens priming with intravaginal devices and gonadotropin. In November 2014, 90 adult Menz ewes were assigned to either the “P” protocol, “PGF” treatment where oestrus and ovulation were synchronized using two injections of prostaglandin 11 days apart or a “GnRH” treatment where the ewes had their oestrus and ovulation synchronized with GnRH (day 0)–prostaglandin (day 6)–GnRH (day 9) sequence. The ewes were naturally mated at the induced oestrus and the following 36 days. Plasma progesterone revealed that 92% of the ewes were ovulating before synchronization and all, except one, ovulated in response to the applied treatments. All “P” ewes exhibited oestrus during the 96‐hr period after the end of the treatments in comparison with only 79.3% and 73.3% for “PGF” and “GnRH” ewes, respectively (< .05). Onset and duration of oestrus were affected by the hormonal treatment (< .05); “GnRH” ewes showed oestrus earliest and had the shortest oestrous duration. Lambing rate from mating at the induced oestrus was lower for “P” than for “PGF” ewes (55.6% and 79.3%, respectively; < .05). The same trait was also lower for “P” than for “PGF” and “GnRH” ewes (70.4%, 89.7% and 86.7%, respectively; < .05) following the 36‐day mating period. Prostaglandin and GnRH analogue‐based protocols are promising alternatives for both controlled natural mating and fixed insemination of Menz sheep after the rainy season when most animals are spontaneously cycling.  相似文献

An experiment was conducted to examine the effect of progesterone prior to a GnRH‐PGF2α treatment on oestrus and pregnancy in seasonally anoestrous Awassi ewes. Twenty‐four ewes were randomly assigned to three groups to be pre‐treated with 60 mg medroxyprogesterone acetate sponges (group A), 600 mg progesterone sponges (group B) or blank sponges (group C) for 4 days. All ewes were injected with 100 μg of GnRH 24 h after sponge removal followed, 5 days later, by 20 mg PGF2α injection. Ewes were exposed to three fertile rams at the time of PGF2α injection (day 0, 0 h) and were checked for breeding marks at 6‐h intervals for 5 days. Blood samples were collected from all ewes 1 day (day ?10) prior to sponge insertion, at the time of sponge removal (day ?6), 1 day following sponge removal (day ?5, at the time of GnRH injection) and at the time of PGF2α injection (day 0) for analysis of progesterone. Progesterone concentrations on days ?10 and ?5 were basal and averaged 0.2 ± 0.04 and 0.2 ± 0.2 ng/ml, respectively. Progesterone concentrations on day ?6 were elevated only in group B ewes and were higher (p < 0.0001) than those of groups A and C. Progesterone concentrations on day 0 were higher (p = 0.002) in groups A and B than group C. Oestrous responses occurred only in ewes of groups A and B (p > 0.05). Induced oestrus conception rate was greater (p < 0.01) in group A than groups B and C. Ewes returned to oestrus 17–20 days following day 0 were two of eight, six of eight and three of eight of groups A, B and C, respectively, all of which eventually lambed. The overall lambing rate was 82% in progesterone‐primed ewes compared with only 38% non‐progesterone‐primed ewes (p < 0.05). Progesterone priming apparently sensitizes GnRH‐PGF2α‐treated seasonally anoestrous ewes and increases their response in oestrus and pregnancy rates.  相似文献

The effect of two insemination policies after synchronization of oestrus on reproductive performance in two groups of cows was investigated. Oestrus was synchronized by two treatments with prostaglandin F2 alpha (PGF2 alpha) at a 14-day interval. Cows in Group 1 (n = 234) were inseminated twice on appointment 66 and 90 h after the second treatment. Cows in Group 2 (n = 222) were intensively watched for signs of oestrus after the synchronization protocol and inseminated on observed oestrus. Cows with abnormal discharge after synchronization were excluded from breeding and inseminated later during the study period. Service rate within 1 week after synchronization was higher in Group 1 than in Group 2 (89.3 versus 67.1%). Conception rates on first service did not differ between groups (33.2 versus 30.0%). Days to first service and days open were shorter in Group 1 (P < 0.05). The number of cows culled for infertility did not differ between the groups. Endometritis 14-20 days post-partum decreased the percentage of cows pregnant at the end of the study period in both groups but did not have a significant effect on conception rates and days open. It is concluded that additional inseminations required in the timed artificial insemination protocol were economically justified by the reduction in days open in comparison with insemination on observed oestrus after synchronization.  相似文献

A total of 137 cycling zebu cows, each receiving a single dose of prostaglandin PGF(2alpha)were used in an oestrus synchronization programme on three different farms. Of the cows on the three farms, 60.6 and 90.5% showed overt oestrus and luteolysis, respectively. Pregnancy rate to fixed time inseminations following single injection of PGF(2alpha)was 61.4% for farm 1, significantly higher than the values of 45.7 and 46.9% for farms 2 and 3, respectively. The pregnancy rates to second service of rebred cows were 53.3, 50.0 and 50.0% for the three farms, respectively, with no significant differences between each.Fertility classification of the cows based on progesterone (P(4)) concentration showed that 6.6% of cows on the three farms were incorrectly diagnosed as having corpora lutea; 2.9% of them had incomplete luteolysis and 5.1% may have lost their embryos between days 21 and 45 post-insemination. The pregnancy rate was 10% higher in the rainy season than in the dry season. Cows with body condition scores of 3 and 4 had a higher overall pregnancy rates than those with a body condition score of 2. The findings of this study further confirm the luteolytic efficacy of prostaglandin in inducing oestrus in zebu cattle and indicate that the nutritional status of the cows must be satisfactory before embarking on oestrus synchronization programmes.  相似文献

OBJECTIVE: To compare the use of gonadotrophin releasing hormone (GnRH) and oestradiol benzoate (ODB) administered following a synchronised pro-oestrus on reproductive performance of lactating dairy cows and the submission rates of non-pregnant cows following resynchronisation. DESIGN: Cohort study. PROCEDURE: Lactating Holstein cows enrolled in a controlled breeding program were first treated with an intravaginal progesterone releasing insert (IVP4) for 8 days, 2.0 mg of ODB intramuscular (i.m.) at device insertion (Day 0), an analogue of PGF2alpha at device removal and either 1.0 mg of ODB i.m., 24 h after device removal (ODB group, n = 242), or 0.25 mg of a GnRH agonist (GnRH group, n = 152) injected i.m. approximately 34 h after device removal. Every cow was artificially inseminated between 49 and 56 h after removal of its insert (Day 10). Cows detected in oestrus 1 day after artificial insemination (AI) that were not detected in oestrus on the previous day were re-inseminated on that day. All cows treated on Day 0 were resynchronised for reinsemination by insertion of a used IVP4 device on Day 23. Oestradiol benzoate at a dose of 1.0 mg was administered i.m. at the time of device insertion. Inserts were removed 8 days later (Day 31) and 1.0 mg of ODB was injected i.m. 24 h later. Those cows detected in oestrus between Days 31 and 35 were artificially inseminated. On Day 46 these cows were resynchronised for a third round of AI by insertion of an IVP4 device, used previously to synchronise cows for the first and second rounds of AI, and administration of 1.0 mg of ODB i.m.. Eight days later inserts were removed. Cows detected in oestrus between Days 54 and 58 were artificially inseminated. Bulls were run with the herd between rounds of AI and removed after 21 weeks of mating. RESULTS: Treatment with ODB or GnRH at the first synchronised pro-oestrus did not significantly alter the reproductive performance over three rounds of AI or over a 21-week breeding period. Treatment also did not alter submission rates at the second round of AI or the proportion of non-pregnant and non-return cows ('phantom' cows) detected and did not result in significant differences in concentrations of progesterone in plasma 10 and 18 days after removal of inserts at the first round of AI. Treatment with GnRH reduced the proportion of cows detected in oestrus at the first round of AI (36.2 vs 97.5%; P < 0.001). CONCLUSION: Administration of GnRH compared to ODB at a synchronised pro-oestrus results in similar reproductive performance. Treatment with GnRH reduced the proportion of cows detected in oestrus following treatment. This may offer advantages to the way AI is managed by enabling insemination at a fixed-time and removing the need for the detection of oestrus.  相似文献

AIMS: To determine if the frequency distribution of time to oestrus observed after the introduction of rams, and additional ewes in oestrus, is affected by the administration of oestradiol- 17beta. To determine if the fertility following mating at the first induced oestrus in progesterone-primed ewes may be increased with administration of oestradiol-17beta. METHODS: In Experiment 1, ewes received 40 mug oestradiol- 17beta 3 (E3; n=61) or 5 (E5; n=56) days before the introduction of rams; 61 ewes were controls (C1). In Experiment 2, a controlled internal drug-releasing (CIDR) device was inserted in 95 ewes on Day -5 (Day 0 = introduction of rams). In addition, 47 ewes received oestradiol-17beta on Day 0 (CE), and the remaining 48 were controls (C2). In both experiments, additional ewes, brought into oestrus between Days 0 and 2 by hormonal treatments, were introduced with the rams. Onset of oestrus was estimated by visual observation of ewes marked by rams. Pregnancy status of ewes in Experiment 2 was determined using ultrasound, 30 days after oestrus. RESULTS: The total number of ewes detected in oestrus in Experiment 1 was similar between the three groups. The frequency distribution of ewes in oestrus in the E5 group differed from that of ewes in both C1 (p=0.05) and E3 (p<0.001). A similar number of ewes were detected in oestrus between Days 16-20, but the proportion of ewes in oestrus between Days 21-26 was greater for ewes in C1 than E5 or E3. Mean interval to the onset of oestrus was shorter in C1 than E3 which was shorter than E5 ewes (p<0.001). In Experiment 2, the total number of ewes in oestrus were similar between groups. Ewes in oestrus between Days 1-4 (65% and 64% for C2 and CE, respectively) and conception rates over the same period (81% and 80% for C2 and CE, respectively) were also similar between groups. CONCLUSIONS: Administration of oestradiol-17beta to anoestrous ewes altered the pattern of onset of oestrus, but it did not affect submission or conception rates, regardless of whether or not a short period of progesterone priming with a new CIDR device was included prior to the introduction of rams.  相似文献

A study was undertaken to determine the effectiveness of a progesterone-releasing intravaginal device (PRID) and prostaglandin F2 alpha (PGF2alpha) in synchronizing oestrus in N'dama and Bunaji cows and heifers and the fertility following artificial insemination at the synchronized oestrus. A total of 116 cows and heifers (58 N'dama and 58 Bunaji) were used in two separate trials. In the first trial, oestrus was synchronized using a PRID, which was inserted for 12 days; in the second trial, oestrus was synchronized by giving two injections of PGF2alpha 13 days apart. Only animals that did not respond to the first injection were given the second injection. At the end of each treatment period, the animals were observed for oestrus for 7 days and inseminated approximately 12 h following detection of oestrus. Standing to be mounted was the single criterion used to judge an animal to have been in oestrus. PGF2alpha and PRID were both effective in synchronizing oestrus in N'dama and Bunaji cows and heifers. The respective oestrus response rates, pregnancy rate and conception rates for PRID and PGF2alpha were 85.7%, 53.6% and 62.5% for PRID, and 91.7%, 68.3% and 74.6% for PGF2alpha. N'dama cattle showed significantly (p<0.05) better oestrus response rate, pregnancy rate and conception rate than Bunaji cattle following both PRID and PGF2alpha treatments. The pregnancy rate and conception rate following PGF2alpha treatment were better (p < 0.05) than for PRID, although the oestrus response rate did not differ. It is concluded that both PRID and PGF2alpha are effective in synchronizing oestrus in N'dama and Bunaji cattle in the hot humid zone of Nigeria and the fertility to artificial insemination at the synchronized oestrus was normal and acceptable. Thus, PRID and PGF2alpha can effectively be used in intensive breeding programmes for the rapid multiplication and distribution of both cattle breeds, especially the N'dama, which is a unique and beneficial animal genetic resource for the tsetse infested hot humid zone of Nigeria.  相似文献

AIM: To determine whether conception rates of anoestrous dairy cows treated with progesterone and oestradiol benzoate (ODB) could be increased by treating them with additional progesterone following insemination at the induced oestrus. METHODS: Cows which had not been detected in oestrus for at least 21 days after calving in 18 herds were confirmed anovulatory anoestrus (AA) by veterinary examination, due to the absence of a detectable corpus luteum in the ovaries. All cows were treated with intra-vaginal progesterone (CIDR insert) for 6 days and injected with 1 mg ODB 24 h after insert removal (Day 0). Only cows which were seen in oestrus on Days 0, 1 or 2 were enrolled in the trial. These cows were either treated with a second CIDR insert on Day 8, for 7 days (P4+; n=422), or remained untreated (Control; n=756). Milk progesterone concentrations were measured in a subset of enrolled cows (n=669) on Day 8 to determine the proportion of cows that ovulated following the induced oestrus. RESULTS: Conception rates to first insemination were similar in P4+ and Control cows (40.3% and 37.2%, p=0.59). Of cows which had milk progesterone concentrations measured on Day 8, 78.6% displayed oestrus and ovulated, (range: 53.8% to 94.6% among herds). Of the cows that ovulated, conception rate to first insemination was 46.8% and 43.5% in P4+ and Control cows, respectively (p=0.86). CONCLUSION: Conception rates to first insemination in AA cows treated with progesterone and ODB were not increased by progesterone supplementation using CIDR inserts following insemination. Twenty-five 2-3-year-old cycling does weighing 17-25 kg were obtained from semi-nomadic farmers and managed under controlled conditions while simulating the traditional management system. Oestrus was synchronized using progestogen impregnated vaginal pessaries. Blood samples were collected daily for progesterone assay from the day of pessary withdrawal up to one complete oestrous cycle. Oestrus was checked twice daily using vasectomized bucks. Ovulation rate was determined by direct observation of the ovaries following laparotomy on day 5-7 of the oestrous cycle. Following oestrus synchronization, mean ovulation rate was 1.68 +/- 0.13. Mean oestrous cycle length and duration of oestrus were 21.30 +/- 0.28 days and 21.37 +/- 0.24 hours respectively. Plasma progesterone concentrations ranged from non-detectable levels on the day of oestrus to 5.2 +/- 0.28 ng ml at mid-cycle. The duration of elevated progesterone level (greater than 2 ng/ml) was about 12 days. The peak progesterone values did not differ between animals with different ovulation rates. However, the plasma progesterone concentration during the early cycle (days 0-6) was significantly lower in the single ovulators compared with others. There were no major differences in plasma progesterone levels during the oestrous cycle of Red Sokoto does with different ovulation rates.  相似文献

An experiment was conducted to determine whether controlled internal drug release devices-type G (CIDR-G) could be used in conjunction with the ram effect to advance the breeding season and lambing and thereafter could be reapplied to stimulate breeding of ewes to produce a second lambing in late summer. Finn crossbred and Columbia ewes were treated with CIDR-G from July 6 to 18. Rams, which had been isolated from ewes, were joined with the flock upon CIDR-G removal. All 59 CIDR-G-treated ewes exhibited estrus, and 55 lambed from breeding within a 30-d period after CIDR-G withdrawal. Eighteen of 19 contemporary control ewes (no CIDR-G) were bred, and 16 lambed. Ewes that had lambed from the CIDR-G and control groups were retreated as before, with CIDR_G inserted March 1 and withdrawn March 13, concomitant with sudden exposure to rams. Of 55 CIDR-G-treated sheep, 53 were bred and 45 lambed. All 16 control ewes were bred and lambed. A second experiment was conducted to determine whether treatment with CIDR-G and the ram effect was effective in mid-spring. The CIDR-G were applied for 12 d and removed on May 1 or 6 from Finn crossbred, Columbia, and Polypay ewes. Rams were introduced upon CIDR-G withdrawal. In the autumn oestrus season, 20 Slovak Merino ewes were exposed to synchronization of oestrus, treated with the PGF2alfa at doses 125 micrograms (Oestrophan, inj. Spofa). followed by an injection of PMSG at doses 1000 IU (Antex Leo Denmark) and 50,000 IU of Vitamin A (Axerophtol Spofa). 23 anoestrus ewes were synchronized with an intravaginal sponges containing 20 mg of chlorsuperlutine (Agelin, Spofa) for 12 days and after sponge withdrawal, the ewes were injected with 750 and 1000 IU of PMSG (Antex Leo Denmark). Ovulatory response was observed and the possibility of ova recovered from the genital organs in ewes after synchronization of oestrus and superovulation in oestrus season. Higher values of the total follicular response (CFO), and the average number of ovulation (PO) after administering equal doses of PMSG were found out both in anoestrus ewes (CFO 6.62 +/- 4.24; PO 4.25 +/- 4.52) and in oestrus ewes (CFO 2.70 +/- 2.10; PO 2.60 +/- 1.74; resp. CFO 2.80 +/- 1.83; PO 3.4 +/- 3.0), if the ewes were treated with PMSG together with vitamin A. The average number of ova flushed was higher in anoestrus ewes (3.0-0.5) than in oestrus ewes (1.67-3.75). In both trials the equal ratio in the number of released ova was gained from ewes of experimental groups (83-88% of the total number). After ova flushing from the genital organs in ewes of the experimental groups most ovas were found in the isthmatal part of the uterine tube (36-60%). On the basis of gained results it was concluded, that synchronized oestrus ewes on receiving PMSG in anoestrus season the ovarial response was more significant than in autumn breeding season.  相似文献

AIM: To compare the reproductive performance of heifers after oestrus synchronisation and fixed-time artificial insemination with non-synchronised heifers bred by herd sires. METHODS: Heifers from 10 spring-calving herds were randomly divided into two groups by herd, breed and age. Heifers in one group (the synchronised group, n = 478) were synchronised with a combination of progesterone, oestradiol benzoate and PGF2alpha, and inseminated 50-54 hours after progesterone treatment. Returns to first service were resynchronised with progesterone treatment 16-21 days after the fixed-time artificial insemination. Heifers in the other group (the control group, n = 470) did not receive any treatment and were bred by herd sires. RESULTS: The conception rate of synchronised heifers to the fixed-time artificial insemination was 51.2% and to the artificial insemination after resynchronisation 40.4%. The pregnancy rate at the end of the breeding season was lower (p<0.001) for the synchronised (92.9%) than for the control (97.2%) group. The interval from start of breeding to calving was earlier for synchronised (295.9 +/- 22.5 days, mean +/- s.d.) than for control (298.5 +/- 17.3 days) heifers. CONCLUSION: Results from this study indicate that the oestrus synchronisation programme used in the present study can reduce reproductive performance by increasing the empty rate compared with natural mating.  相似文献

The fertility of ten West African Dwarf goats in which oestrus was induced with PGF2 alpha was comparable to that of the five West African Dwarf goats in the control group. Performance indices as determined by duration of oestrus and length of gestation were not affected. Similarly the birth weight of kids was not significantly influenced. However, treated goats were superior to controls when the interval from commencement of experiment to oestrus is considered, although there was no difference between the two treated groups of goats (5 mg vs 10 mg PGF2 alpha). Results showed that PGF2 alpha could be an important tool in oestrus synchronization and thus in controls of the reproductive performance of the West African Dwarf goats under humid lowland tropical environmental conditions.  相似文献

The objective of the present study was to determine the temporal relationships among luteal adenylate cyclase activity, luteal phosphodiesterase activity, luteal progesterone concentration and plasma progesterone concentration during prostaglandin F2 alpha (PGF2 alpha)-induced luteolysis in ewes. Corpora lutea (CL) were removed from cycling ewes on d 9 (d 0 = first day of estrus) at 0, 2, 4, 6, 12 and 24 h (seven to eight ewes/group) after PGF2 alpha administration (im). Jugular blood samples were collected at the time of enucleation of CL and analyzed for progesterone. Plasma and luteal progesterone concentrations were decreased (P less than .05) by 4 and 12 h after PGF2 alpha injection, respectively. Basal adenylate cyclase, luteinizing hormone (LH)-activated adenylate cyclase, guanylylimidodiphosphate [Gpp(NH)p]-activated adenylate cyclase and LH plus Gpp(NH)p-activated adenylate cyclase activities were decreased (P less than .05) by 2 h after PGF2 alpha injection. The decrease in adenylate cyclase activity paralleled the decrease in plasma progesterone concentration over time. Luteinizing hormone stimulated (P less than .05) adenylate cyclase activity relative to basal activity at 0, 2, 12 and 24 h post-PGF2 alpha; whereas, Gpp(NH)p stimulated (P less than .01) adenylate cyclase activity relative to basal activity at each time point. In contrast to the decrease in adenylate cyclase activity, phosphodiesterase activity was increased (P less than .05) at 2 and 4 h post-PGF2 alpha. These results suggest that a decrease in adenylate cyclase activity coupled with an increase in phosphodiesterase activity may decrease the intracellular adenosine 3',5' cyclic monophosphate (cAMP) concentration.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
