Changes of reduced glutathione (GSH) and TBARS (thiobarbituric acid-reactive products of lipid peroxidation) concentrations and activity of gamma-glutamyltransferase (GGT, EC 2.3.2.2) in the blood of Lohman brown cockerels and pullets in response to 48 hour food deprivation and 24 hour refeeding were examined. The experiment was performed on 61-day-old chickens. Blood samples ware collected from the wing vein (v. brachialis) in heparinized tubes for three times: control sampling before fasting, then after 48 hour food deprivation and after refeeding for 24 hours. Blood GSH concentration after refeeding in cockerels was significantly higher compared with prefasting and fasting values. The concentration of GSH in female chickens was significantly lower after fasting as well as after refeeding compared with control values. In addition to that, in pullets GSH concentration in refeeding was higher than in fasting conditions. The level of TBARS in blood in female and male chickens after fasting and refeeding were significantly lower than the prefasting values. The GGT activity on cockerels after 48 hour food deprivation was significantly higher compared with control sampling and in chickens refeed for 24 hours, whereas in pullets significant difference was exhibit compared only with control values. Concentration of GSH in control sampling in cockerels compared with those in pullets was significantly lower. After 48 hours of fasting, the level of GSH was significantly higher in the cockerels than in the pullets. Results of TBARS concentration in the pullets were higher of control and fasting values than in the cockerels. The GGT activity of control sampling was significantly higher in male chicken. Lipid peroxidation in chickens of both sexes decreased with fasting, but prooxidative-antioxidative processes were more intensive in female chickens, probably because they were not reach sexual maturity.  相似文献

Age-related changes of tissue lipid peroxidation (LPO) of liver and brain, as well as plasma antioxidant capacity of broiler chicken cockerels were investigated. Tissue LPO was characterised by the spectrophotometric assessment of thiobarbituric acid reactive substances (TBARS). Plasma antioxidant power was evaluated by the measurement of total antioxidant status (TAS). Newly hatched broiler chicks had similar TAS value (1.19 mmol/l) as newborns of mammalian species. Significant changes (p < 0.05) were observed in the time course of all parameters. Tissue TBARS concentration was higher in the brain than in the liver at hatching, while the latter organ was found to have more effective antioxidant defence during embryonic life. The concentration of TBARS increased up to the 10th day in the liver but only up to the 21st day in the brain, and the former was accompanied by an approximately 50% decrease of plasma antioxidant capacity. This suggests that the liver plays an important role in forming the antioxidant defence mechanisms of the blood plasma in broiler chicks.  相似文献

The influence of fish oil (highly unsaturated) and beef tallow (highly saturated) with vitamin E (100 IU/kg) supplementation on the antioxidant status of broiler chicken cockerels was investigated. Chicks were fed a control diet with no added fat, 40 g/kg each of fish oil and beef tallow diets, respectively, from 11 to 42 days of age. Tocopherol concentration and the rate of lipid peroxidation, thiobarbituric acid reactive substance (TBARS) in liver, fatty acid composition of the liver lipids, blood serum total antioxidant status (TAS), and reduced glutathione (GSH) content were determined. Vitamin E supplementation of the diet increased liver alpha-tocopherol content in chicks regardless of the type of dietary fat. Fish oil diet resulted in higher liver TBARS value while beef tallow diet showed lower values compared to the control diet. Vitamin E supplementation reduced liver TBARS as well as serum GSH, and raised serum TAS for all diets. Serum GSH was the same for vitamin E supplemented diets regardless of the fat supplement. Fish oil diets resulted in a significant increase in hepatic lipid n-3 PUFA content. A significant positive correlation was found between liver TBARS and n-3 PUFA content. No relationships were established, however, between liver TBARS and n-6 PUFA or saturated fatty acids. The results suggest that feeding oils rich in n-3 PUFA increases tissue concentration of these fatty acids, consequently increasing tissue lipid peroxidation and reducing the antioxidative status of broiler chickens. Supplementing high levels of vitamin E with such oils may increase tissue oxidative stability. Serum TAS or GSH may be used as a measure of antioxidative status in chickens.  相似文献

A model stress situation was created by nutrition deprivation. The experiment was performed with 130 cockerels of the initial breed White Leghorn. The experimental cockerels of the age of 18 to 24 days fasted for 24, 48 and 72 hours. Uric acid was determined in the supernatant of liver homogenate by means of a colorimetrically reductometric method with phospho-wolframic agent. A significant increase in the uric acid concentration in the liver of cockerels was found out during the model stress: the concentration of uric acid increased progressively from the initial 70.25 +/- 8.94 mg per 100 g to 93.15 +/- 9.66 mg per 100 g after 24-hour fasting, to 90.23 +/- 7.84 mg per 100 g after 48-hour fasting and even to 137.04 +/- 15.59 mg per 100 g after three-day fasting of 72 hours. After re-alimentation, the concentrations of uric acid were reduced to physiological values already within 24 hours.  相似文献

The relationship between blood plasma level and urinary excretion of allantoin (AN) was examined in sheep and goats during fasting to investigate the possible use of purine derivatives (PD) in urine and/or plasma for estimating the microbial protein production in the rumen, and the further digestion in the lower guts of ruminants. Urinary AN excretion decreased markedly during fasting (0.13 mmol/kgW0.75 per day), although urinary levels of other PD, hypoxanthine + xanthine and uric acid did not differ irrespective of the feeding condition, that is, feeding, fasting and refeeding in both species. The AN concentration in blood plasma also decreased drastically in the starvation period, and was suddenly increased on refeeding in sheep and goats, and these phenomena were very similar to those of urinary AN excretion. Therefore, there was a high positive correlation between plasma AN level and urinary AN excretion, and the coefficient of correlation was statistically significant (P < 0.01). These results clearly indicate that changes in urinary AN reflect change in plasma AN, which is induced by the catabolism of purine base in the body.  相似文献

Fifteen adult female goats were orally exposed to 5.46 mg lead (as lead acetate) per kg body weight daily for 2 weeks to study the antioxidant enzymes of the erythrocyte, lipid peroxide level, total thiol groups and total antioxidant status (TAS) in plasma. Ten goats served as unexposed control. Blood samples were collected before exposure (day 0) and on days 7 and 14. Ten per cent erythrocyte haemolysate was prepared and analysed for glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT), total thiol groups and lipid peroxide. TAS was determined in plasma. There was a significant (P < 0.05) increase of erythrocytic GPx, SOD, CAT, total thiol groups and TAS on day 7 which was followed by a significant (P < 0.05) decrease of all these parameters on day 14. Lipid peroxide level increased significantly (P < 0.05) and the maximum level was attained by day 14. The results obtained indicate a possible role of free radicals in lead poisoning pathogenicity.  相似文献

OBJECTIVE: To determine whether dietary antioxidants would attenuate exercise-induced increases in plasma creatine kinase (CK) activity in sled dogs. ANIMALS: 41 trained adult sled dogs. PROCEDURE: Dogs, randomly assigned to 2 groups, received the same base diet throughout the study. After 8 weeks on that diet, 1 group (21 dogs) received a daily supplement containing vitamins E (457 U) and C (706 mg) and beta-carotene (5.1 mg), and a control group (20 dogs) received a supplement containing minimal amounts of antioxidants. After 3 weeks, both groups performed identical endurance exercise on each of 3 days. Blood samples were collected before and 3 weeks after addition of supplements and after each day of exercise. Plasma was analyzed for vitamins E and C, retinol, uric acid, triglyceride, and cholesterol concentrations, total antioxidant status (TAS), and CK activity. RESULTS: Feeding supplements containing antioxidants caused a significant increase in vitamin E concentration but did not change retinol or vitamin C concentrations orTAS. Exercise caused significantly higher CK activity, but did not cause a significant difference in CK activity between groups. Exercise was associated with significantly lower vitamin E, retinol, and cholesterol concentrations and TAS but significantly higher vitamin C, triglyceride, and uric acid concentrations in both groups. CONCLUSIONS AND CLINICAL RELEVANCE: Use of supplements containing the doses of antioxidants used here failed to attenuate exercise-induced increases in CK activity. Muscle damage in sled dogs, as measured by plasma CK activity, may be caused by a mechanism other than oxidant stress.  相似文献

The influence of dietary palm olein, in comparison to fish oil and lard, on lipid levels in egg yolk and blood plasma, the fatty acid composition of egg yolk, and various production parameters were studied. Brown Lohman laying hens (n=45) were randomly assigned into three groups of 15 birds, and treated with experimental diets with either 3% palm olein (PO), fish oil (FO) or lard (L) for 6 weeks. At the end of the experiment, feed consumption was significantly lower for hens fed the PO diet, except week 6 of the experiment. The concentration of plasma triglycerides was increased by all experimental diets, whereas there were no significant increases of plasma total lipid and total cholesterol concentrations only in the PO group. For yolk lipids a decrease in triglycerides in the FO and L groups was observed, while total cholesterol and total lipid were significantly decreased in the PO group. Feeding with the PO diet resulted in the lowest concentrations of palmitic, stearic and linoleic acid, as well as in the highest concentration of monounsaturated oleic acid in the yolk total lipid. It was concluded that the composition of yolk lipids did not closely match the concentrations of lipids observed in experimental diets or plasma. Based on the current work it seems that the PO diet modulates egg yolk lipid content best.  相似文献

Selenium, as part of selenocysteine, is the active centre of selenoenzymes. Excess amount of selenium generates oxygen free radicals and reacts with thiol compounds such as reduced glutathione, composing seleno-diglutathione. These reactions impair the amount and/or activity of the biological antioxidant defence system. In the present experiment the effects of two inorganic selenium sources (Na2SeO3, Na2SeO4) on lipid peroxidation and on the content and activity of some components of the antioxidant defence mechanism were studied in Ross 308 hybrid cockerels. In the tissues examined, the glutathione redox system was altered in different ways as an effect of excess selenium. The amount of glutathione and, consequently, glutathione peroxidase activity declined in the blood plasma and in the red blood cell haemolysate, while in the liver malondialdehyde concentration increased only at the end of the experimental period as a result of the boosted enzyme activity. The authors suppose that the cause of increased lipid peroxidation was the potential toxic effect of selenium accumulation above the actual demand.  相似文献

1. Heterozygous cockerels which inherited the sex‐linked recessive dwarfing gene (dw) from a normal dam had significantly different contents of sodium and potassium in the blood serum compared with heterozygous cockerels from dwarf dams.

2. Hemizygous dwarf pullets had a significantly lower packed cell volume than normal pullets, while mean corpuscular haemoglobin concentration was significantly lower in the normal pullets.

3. Total serum calcium values were significantly higher in normal pullets. Sodium concentration was higher in dwarf pullets. It is suggested that the better adaptation of dwarf pullets to higher temperatures might be due to their capacity to retain more water in body tissue under a stressful environment.  相似文献

1. We examined the changes in plasma IGF-I concentration and tissue IGFBP-2 gene expression of young fasted chickens refed a commercial diet or administered bovine insulin intravenously. 2. Plasma IGF-I concentration was decreased by fasting for 2 d. Although plasma IGF-I concentration was increased by refeeding, it didn't recover to the level of chickens fed a commercial diet ad libitum. 3. Insulin administration lowered plasma IGF-I concentration compared to other groups. 4. Hepatic IGFBP-2 mRNA was increased by fasting for 2 d and decreased by refeeding for 6 h. Insulin administration also decreased hepatic IGFBP-2 gene expression stimulated by fasting to the level of refed chickens. 5. IGFBP-2 mRNA in the gizzard was increased by fasting for 2 d and tended to decrease after refeeding for 6 h. Insulin administration decreased gizzard IGFBP-2 gene expression to less than that in refed chickens. 6. There was no between-treatment difference in IGFBP-2 mRNA in the brain and kidney. 7. These results suggest that the changes in IGFBP-2 gene expression in the liver and gizzard by fasting and refeeding might be partly regulated by the alteration in plasma insulin concentration.  相似文献

1. The effects of feeding three types of cereal grain (wheat, triticale or rye) and soyabean oil (0 or 20 g/kg) over a 12-week period on the production, yolk cholesterol and yolk fatty acid concentrations of three strains of laying pullets were studied. 2. Pullets fed on wheat- or triticale-based diets had higher body weight gains, egg productions, egg weights, egg mass and lower yolk cholesterol concentrations than pullets fed on rye-based diets. However, there were no significant differences between the cereals in yolk cholesterol content. 3. There were no significant differences between the three cereals in total food consumption of pullets nor of yolk weight nor yolk concentration of palmitic, stearic and oleic acids. 4. Pullets fed on triticale-based diets had higher yolk linoleic acid concentrations and lower yolk oleic acid: linoleic acid ratios than pullets fed on rye- or wheat-based diets. 5. Soyabean oil supplementation increased egg production, egg mass, yolk linoleic concentration and yolk unsaturated to saturated fatty acid ratio, but reduced yolk oleic acid: linoleic acid ratio. 6. There were differences between strains of pullets in weight gain, food consumption, rate of lay, egg weight and yolk cholesterol, but not in yolk fatty acid concentrations. 7. It was concluded that wheat- or triticale-based diets gave good production of eggs of lower cholesterol content, that soyabean oil supplementation gave eggs with a high unsaturated to saturated fatty acid ratio and that two strains of layers produced eggs with lower yolk cholesterol concentrations than a third strain.  相似文献

1. We examined the influence of refeeding after 2 d of fasting on plasma insulin-like growth factor-I (IGF-I) concentration and hepatic IGF-I gene expression in chickens at 6 weeks of age. 2. Hepatic IGF-I mRNA was measured by ribonuclease protection assay and plasma IGF-I concentration was determined by radioimmunoassay. 3. Plasma IGF-I concentration decreased following fasting, increased to the level of fed controls after 2 h of refeeding but then fell back to the level of fasted chickens after 6 h of refeeding. 4. Fasting reduced hepatic IGF-I mRNA concentrations to less than half of those in the fed controls. Refeeding increased IGF-I mRNA sharply at 2 h after refeeding, but by 6 h after refeeding they had taller back again to levels significantly lower than at 2 h. 5. A significant correlation between plasma IGF-I concentration and hepatic IGF-I gene expression was found, suggesting that when chicks are refed after 2 d of fasting, the short-term increase in plasma IGF-I concentration may be partly regulated by the alteration in hepatic IGF-I mRNA.  相似文献

1. Free amino acid concentrations were measured in the blood plasma of immature pullets receiving a folate-deficient diet and injected with folate and/or oestradiol, with appropriate controls. 2. Folate deficiency caused a decrease and oestradiol administration an increase in the plasma concentration of most of the amino acids studied. 3. Plasma glycine concentration increased in foliate deficiency and decreased with oestradiol treatment. 4. All the observed effects could not be readily explained in terms of the known metabolic role of folate in amino acid metabolism. 5. The results did not indicate that the role of folate in amino acid metabolism was significant in the inhibition of oestradiol-stimulated oviduct growth.  相似文献

The effect exerted by overdosage of monensin, an ionophore antibiotic, on the lipid peroxide status of broiler chickens was studied. Three-week-old broiler cockerels were given 150 mg monensin/kg body mass through a tube, and the malondialdehyde (MDA) concentration, glutathione peroxidase (GSH-Px) and catalase activity of the liver and breast muscle, and MDA concentration and GSH-Px activity of the blood plasma were determined. Liver MDA and catalase values rose rapidly and significantly during the experimental period. GSH-Px activity initially decreased, then tended to rise. Blood plasma and breast muscle variables did not change during the experiment. Acute monensin poisoning induced substantial enhancement of lipid peroxidation processes in the liver, while it did not appreciably affect the lipid peroxide status of the blood plasma and breast muscle. The role of the observed phenomenon in the rather complex pathogenesis of monensin poisoning is not known sufficiently. Further studies are needed to elucidate the problem.  相似文献

The main objective of the present study was to investigate the effects of short-term fasting in gilts on endocrinological and blood biochemical parameters and, further, the effects of subsequent oral endotoxin (ET) administration. Group 1 was fasted for 30 h and then received feed with ET added. Group 2 was fasted for 30 h but received standard feed at refeeding. In group 3, gilts were fed every 6 h for 30 h. The major effects of fasting were: gradually increased concentration of plasma prostaglandin F metabolite, serum total bilirubin, serum free fatty acids, and decreased serum glucose. The values were normalized within 1–4 h of refeeding. Twelve hours after refeeding, the ET-refed gilts showed higher levels of serum total bile acids and polymorphonuclear leukocytes than those in group 2. It is possible that the observed changes during fasting reflect either an increased intestinal uptake of naturally present endotoxin or a reduced endotoxin detoxifying capacity of the liver. The increased bile acid concentration and polymorphonuclear leukocyte count following refeeding with ET-feed may indicate that orally administered ET is to some extent absorbed from the gut.  相似文献

The effects of nutritional status, such as fasting and refeeding, on leptin and ghrelin secretion in swine were examined. The swine (n = 4) were fasted for 54 h and plasma hormone levels were measured before, during and after fasting. Plasma leptin and insulin concentrations began to decrease 12 and 6 h into the fasting period, respectively (P < 0.05), and maintained a low level for the remaining period of fasting. Plasma leptin and insulin returned to the pre‐fasting value 6 and 12 h after refeeding, respectively. Plasma ghrelin concentrations showed a nocturnal periodicity during the fasting period; it increased nocturnally at 36 and 42 h into the fasting period (P < 0.05). Plasma growth hormone levels did not show any remarkable changes during the fasting. Plasma glucose levels showed a modest fall during fasting and significantly decreased (P < 0.05) at 24 h into the fasting period, returning to pre‐fasting levels after refeeding. Plasma nonesterified fatty acid levels increased (P < 0.05) at 12 h into the fasting period and returned to the pre‐fasting level 6 h after refeeding. These results indicate that plasma leptin, insulin and ghrelin play an important role in maintaining energy homeostasis in swine. The plasma ghrelin did not continuously increase, but showed nocturnal periodicity during fasting. This may suggest that ghrelin is also involved in physiological processes other than energy homeostasis.  相似文献

We studied the influence of an embryonal application of antiestrogen tamoxifen on gonad differentiation and sex dimorphism during the growth of Slovgal chickens. A single application of 500 micrograms tamoxifen to the incubated eggs before the onset of gonad differentiation induced in pullets the growth of the right gonad, which usually develops only in cockerels. Comparing the size of both gonads it is clear that the above dose of antiestrogen had the 22.31% masculinizing effect. Morphological changes observed just after hatching correlated with the testosterone concentration in plasma. The pullets masculinized by tamoxifen had a significantly higher level of male hormone. The changes in sex differentiation were reflected in the altered growth of pullet bodies. With respect to the higher growth rate and more effective feed conversion, their growth can be appreciated as masculinized. The higher body weight at the end of seven-week fattening (4.25% in comparison with the control group of pullets) was due to the higher trunk weight. The growth was stimulated by tamoxifen application also in cockerels (5.57% increase in body weight) but it was not accompanied by any changes in feed conversion.  相似文献

An experiment was conducted with commercial White Leghorn type chickens to determine the effect of gender on tissue ascorbic acid concentration, antioxidant capacity and ascorbic acid synthesis. 2. Birds reared and maintained on litter were given a standard layer diet, without supplemental ascorbic acid, from 18 weeks of age. Tissue ascorbic acid concentration, plasma total antioxidant capacity and renal L-gulonolactone oxidase activity were measured at 30 weeks of age. 3. Females and males differed in ascorbic acid synthesis, as measured by renal L-gulonolactone oxidase activity, and tissue ascorbic acid concentration. 4. Plasma total antioxidant capacity and adrenal, gonadal, plasma and pituitary ascorbic acid concentrations were significantly higher in males, whereas ascorbic acid synthesis and splenic and thymic ascorbic acid concentrations were significantly higher in females. 5. L-Gulonolactone oxidase activity was not detected in the comb of cockerels.  相似文献

1. The effects of daily injections of corticosterone (1 or 5 mg/bird) on growth, fat deposition, liver lipid and plasma concentrations of uric acid, glucose, insulin and growth hormone were studied using genetically selected lines of fat (FL) and lean (LL) chickens. 2. Both doses of corticosterone depressed body weight gain and increased the liver lipid and the abdominal fat to the same extent in both lines. 3. In both lines, corticosterone caused a dose-dependent increase in the plasma concentrations of uric acid, glucose and insulin in the fasted and refed states. 4. In untreated birds, plasma concentrations of growth hormone (GH) were slightly higher in FL than in LL chickens and slightly decreased during refeeding. The response was not modified by injection of 1 mg corticosterone. Injections of 5 mg decreased plasma GH in both lines in the fasting state and in LL chickens during refeeding. In contrast, the same dose increased GH in FL chickens during refeeding. This contradiction remains unexplained. 5. The results suggest that corticosterone sensitivity is not involved in difference of fattening between FL and LL chickens.  相似文献
