Summary Ascochyta blight induced by Ascochyta fabae f.sp. lentis is a major foliar disease affecting lentil. Screening 248 accessions of the ICARDA wild lentil germplasm collection for resistance to a Syrian isolate of this fungus was conducted under artificial inoculation in a plastic house. The reaction of resistant accessions was confirmed in a second trial. Twenty-four out of 86 accessions of Lens culinaris ssp. orientalis were resistant, as were 12 of 35 accessions of L. culinaris ssp. odemensis, 3 of 35 accessions of L. nigricans ssp. nigricans, 36 of 89 accessions of L. nigricans ssp. ervoides, and all 3 accessions of Vicia montbretii. Sixty-four per cent of resistant sources were from Syria and southeastern Turkey. Disease reaction was uncorrelated both to the altitude of collection and its annual average rainfall. A significant correlation (r = 0.281) between leaflet width and disease reaction was due more to the frequency of the resistant reaction within the narrow-leaved L. nigricans ssp. ervoides than as a function of small leaf area. Disease reaction was uncorrelated with a range of other morphological traits.  相似文献

A collection of 164 Aegilops tauschii accessions, obtained from Gatersleben, Germany, was screened for reaction to leaf rust under controlled greenhouse conditions. We have also evaluated a selection of synthetic hexaploid wheats, produced by hybridizing Ae. tauschii with tetraploid durum wheats, as well as the first and second generation of hybrids between some of these resistant synthetic hexaploid wheats and susceptible Triticum aestivum cultivars. Eighteen (11%) accessions of Ae. tauschii were resistant to leaf rust among which 1 was immune, 13 were highly resistant and 4 were moderately resistant. Six of the synthetic hexaploid wheats expressed a high level of leaf rust resistance while four exhibited either a reduced or complete susceptibility compared to their corresponding diploid parent. This suppression of resistance at the hexaploid level suggests the presence of suppressor genes in the A and/or B genomes of the T. turgidum parent. Inheritance of leaf rust resistance from the intercrosses with susceptible bread wheats revealed that resistance was dominant over susceptibility. Leaf rust resistance from the three synthetics (syn 101, syn 701 and syn 901) was effectively transmitted as a single dominant gene and one synthetic (syn 301) possessed two different dominant genes for resistance.  相似文献

Summary During the winters of 1990/91 and 1991/92, 181 accessions of Triticum dicoccon Schrank from the CIMMYT gene-bank were screened in the field for resistance to Russian wheat aphid, Diuraphis noxia (Kurdjumov). Accessions were sown in hill plots of 10 seeds and artificially infested with D. noxia at the two-leaf growth stage. Hills were visually assessed for damage at tillering, booting and heading. Entries differed significantly in their reaction to D. noxia, and severity of symptoms increased with time. Twenty four of the entries were highly resistant to the aphid. In winter 1991/92, 807 accessions of wild and cultivated wheats (26 species) and synthetic hexaploids were screened similarly for resistance to D. noxia. A large number of A-genome species were resistant, while few D-genome species were identified as resistant. These newly discovered sources of resistance can be used to expand the genetic base of resistance to D. noxia in both bread (T. aestivum L.) and durum wheats (T. turgidum L. convar. durum (Desf.) The genetic diversity of a subset of the Ethiopian genebank collection maintained at the IPK Gatersleben was investigated applying 22 wheat microsatellites (WMS). The material consisted of 135 accessions belonging to the species T. aestivum L. (69 accessions), T. aethiopicum Jacubz. (54 accessions) and T. durum Desf. (12 accessions), obtained from different collection missions. In total 286 alleles were detected, ranging from 4 to 26 per WMS. For the three species T. aestivum, T. aethiopicum and T. durum on average 9.9, 7.9 and 7.9 alleles per locus, respectively, were observed. The average PIC values per locus were highly comparable for the three species analysed. Considering the genomes it was shown that the largest numbers of alleles per locus occurred in the B genome (18.4 alleles per locus) compared to A (10.1 alleles per locus) and D (8.2 alleles per locus) genomes. Genetic dissimilarity values between accessions were used to produce a dendrogram. All accessions could be distinguished, clustering in two large groups. Whereas T. aestivum formed a separate cluster, no clear discrimination between the two tetraploid species T. durum and T. aethiopicum was observed.  相似文献

The identification of seedling resistance to white rust of crucifers was performed in a screening of a B. oleracea core collection with 400 accessions representing the genetic and geographic diversity of the species. Fifty seedlings per accession were tested against the Portuguese isolate Ac502 using the methodology and evaluation procedures developed by and . The percentage of resistant seedlings (%R) and the conventional rating criteria of the mean Disease Index (DI) based on the two different evaluation procedures of disease expression used, were compared and adopted as the criteria to rank the accessions for their interest as sources of resistance. A great variability of reactions was found between and within accessions of the core collection, ranging from complete resistance to full susceptibility. Sources of resistance were found namely among the cauliflowers, broccoli and tronchuda cabbages gene pools. Forty-seven accessions presented at least 20% of resistant seedlings. Nine accessions (the kales INRA18 and INRA62, the cauliflowers HRI4856, HRI4866 and HRI5424, the loose-head cabbage HRI11555, the savoy cabbage BRA848, the black broccoli HRI6318 and the Portuguese tronchuda cabbage ISA207) presented 50–78% of resistant seedlings and so they should be considered as potential and useful sources for direct use in breeding programs for white rust resistance. Fourteen inbred lines, representing the full range of disease expression, derived from resistant accessions of the core collection were also tested for resistance to other two Portuguese isolates (Ac503 and Ac504) and to a UK isolate. The cultivated types of Brassica rapa L. em. Metzg. consist of morphologically distinct subspecies such as turnip, turnip rape, Chinese cabbage, pak choi and pot herb mustard which are classified as ssp. rapa, ssp. oleifera, ssp. pekinensis, ssp. chinensis and ssp. nipposinica (syn. ssp. japonica), respectively. We attempted to elucidate the phylogenetic relationships among the cultivated types of B. rapa. Thirty-two accessions from the Eurasian Continent were analyzed using AFLP markers with a cultivar of B. oleracea as an outgroup. In total, 455 bands were detected in the ingroup and 392 (86.6%) were polymorphic. The Neighbor-Joining tree based on the AFLP markers indicated that the accessions of B. rapa were congregated into two groups according to geographic origin. One group consisted of ssp. rapa and ssp. oleifera of Europe and Central Asia and the other included all the subspecies of East Asia. Our results suggest that cultivars from East Asia were probably derived from a primitive cultivated type, which originated in Europe or in Central Asia and migrated to East Asia. This primitive cultivated type was probably a common ancestor of ssp. rapa and ssp. oleifera. The Neighbor-Joining tree also shows that leafy vegetables in East Asia such as ssp. pekinensis, ssp. chinensis and ssp. nipposinica were differentiated several times from the distinct cultivars of ssp. oleifera in East Asia.  相似文献

Portuguese wheat landraces, ‘Arrancada’ were collected from the Aveiro region, Portugal before the 1950s. We found in eight accessions of `Arrancada' hexaploid wheat with the long glume phenotype. We assessed the comparative genetic diversity among Portuguese `Arrancada' wheat and Triticum petropavlovskyi Udacz. et Migusch. using AFLP assays and discuss the origin of long glumed `Arrancada' wheat. With the four primer pairs a total of 4885 visible bands were scored corresponding to 99 AFLP markers as putative loci, of which 55 markers (54%) were polymorphic. UPGMA clustering and PCO grouping showed that long glumed ‘Arrancada’ wheat and T. petropavlovskyi were genetically diverse. Long glumed ‘Arrancada’ hexaploid wheat separated into two clusters (groups) in both the UPGMA dendrogram and in PCO analysis. Four long glumed accessions fell in the cluster of tetraploid wheat. A similar argument could be made for another four accessions which belong to the cluster of hexaploid wheat. The substantial level of genetic variation indicated that long glumed ‘Arrancada’ wheat and T. petropavlovskyi originated independently. It is most likely that the P-gene of long glumed ‘Arrancada’ hexaploid wheat was introduced from T. turgidum ssp. polonicum (L.) Thell. to T. aestivum via natural introgression or breeding. The International Cocoa Genebank, Trinidad (ICG,T) contains about 2000 cacao (Theobroma cacao L.) accessions. The purpose of the collection is to provide a source of genes for the genetic improvement of cacao through breeding. In fulfilling this objective, it is imperative that this collection be evaluated to identify major sources of genes for the genetic improvement of important agronomic–economic traits. In this study, 816 cacao accessions were evaluated for resistance to Phytophthora pod rot (black pod disease), which causes enormous loss of yield in cocoa production throughout the world.The study identifies promising resistant genotypes that could be exploited in cacao breeding programmes. It further compares the levels of resistance to Phytophthora pod rot among two cacao types (wild and cultivated types), three major groups (Forastero, Trinitario and Refractario) and 11 accession groups (B, AM, CL, ICS, IMC, JA, LP, MOQ, NA, PA and TRD) . The distribution of scores for the 816 accessions showed skewness towards the susceptible end of the disease rating scale, indicating that a large proportion (68.9%) of the sample was susceptible (disease rating 6–8) to Phytophthora pod rot. However, 12.9% of the sample population was found to be resistant (disease rating 1–3) and 18.2% moderately resistant (disease rating 4–5). Significant differences were observed between the wild and cultivated accessions and among the Forastero, Refractario and Trinitario groups. Marked differences were also observed among 11 accession groups, each of which was represented by at least 20 genotypes. Higher proportions of resistant (17.7%) and moderately resistant (22.6%) genotypes were observed in the wild accessions than in the cultivated varieties (9.4% resistant and 14.4% moderately resistant). The Forastero group, consisting of many wild accessions, was found to contain more resistant (18.0%) and moderately resistant (23.1%) genotypes than either the Trinitario (4.8% resistant and 13.6% moderately resistant) or Refractario (11.3% resistant and 15.4% moderately resistant). Among 11 accession groups, the largest percentage of resistant (24.2%) and moderately resistant (28.8%) genotypes were found in the PA (Forastero). Different accession groups had varying proportions of resistant and moderately resistant genotypes. This is not unexpected in an out-breeding crop such as cacao with a high level of heterogeneity. This finding reinforces the idea of a pre-breeding programme (germplasm enhancement) to accumulate resistance genes over several populations as a strategy for improving the genetic base of resistance in national cacao breeding programmes. One hundred and five promising resistant genotypes were identified among the 816 accessions evaluated. These accessions are potential sources of resistance genes for the development of high-yielding resistant varieties in cacao.  相似文献

A total of 1,383 accessions of wild barley (Hordeum vulgare ssp. spontaneum) held in the USDA-National Small Grains Collection, Aberdeen were screened for resistance to powdery mildew (Blumeria graminis f. sp. hordei) in the Czech Republic. They were subsequently tested at the seedling stage for reaction to selected powdery mildew pathotypes and simultaneously for their adult plant resistance in the field. One hundred and twenty-three accessions exhibited resistance to 22 pathotypes in the greenhouse tests (reaction type 2–3 or lower) as well as resistance to the natural population of this pathogen in the field in the Czech Republic during at least 2-year testing. Summary The 99 accessions of seven wild Lactuca species (L. serriola, L. saligna, L. virosa, L. viminea, L. perennis, L. tenerrima, L. tatarica) and Mycelis muralis (syn. L. muralis) were tested for natural infection of Erysiphe cichoracearum L. serriola accessions were highly susceptible. L. saligna showed highly variable levels of resistance. An isolate ofPuccinia hordei (ND89-3) originally collected in Morocco is virulent on most barley genotypes reported to possess resistance, except cultivar Estate (CI 3410), which possesses theRph3 gene and exhibits a low to intermediate level of resistance (infection type 12). Isolate ND89-3 possesses one of the widest virulence spectrums reported forP. hordei. Accessions ofHordeum vulgare (1,997 in total) andH. spontaneum (885 in total), mostly originating from the Mediterranean region and parts of North Africa, were evaluated with isolate ND89-3 at the seedling stage to identify new sources of leaf rust resistance. Fifty-eight accessions ofH. vulgare, and 222 accessions ofH. spontaneum exhibited low infection types to this isolate. Further evaluations of these resistant accessions with isolates ofP. hordei virulent forRph3,Rph7, andRph12 suggested that most of the resistantH. vulgare accessions possess theRph3 gene. Data suggested additional sources of effective resistance inH. vulgare are rather limited. FiveH. vulgare accessions and 167H. spontaneum accessions were identified as possible sources of new genes for leaf rust resistance. These accessions likely possess resistance genes that are different fromRph1 toRph12, or gene combinations thereof based on their reaction to four leaf rust isolates. Series of 10,348 accessions belonging to 21 species (hexaploid, tetraploid, diploid) of the genus Triticum and 489 accessions belonging to 20 species of the genus Aegilops were scored for disease resistance during a period of 60 years. Tests were performed at the seedling stage for powdery mildew (Erysiphe graminis DC. f. sp. tritici March.), leaf rust (Puccinia recondita Rob. ex Desm. f. sp. tritici Erikss.), stripe rust (Puccinia striiformis West. f. sp. tritici Erikss.) and eyespot (Pseudocercosporella herpotrichoides (Fron.) Deight.) but also at the adult plant stage considering powdery mildew, leaf rust, stripe rust, eyespot and glume blotch (Septoria nodorum Berk.). About 150,000 disease scores recorded on index cards using different scoring scales were transferred to the computer, converted into a 1–9 scale and used to summarise the results. Within the genus Triticum 20% of the material analysed was classified as heterogeneous. For the accessions without detectable segregation a large variability for resistance/susceptibility was detected. At the adult plant stage resistant accessions without visible infections were identified for all diseases. The percentages of resistant accessions at that growth stage were always higher than the ones found in the material tested at the seedling stage. The probability for finding resistant material was shown to be highest in the diploid species ( > 50%) but decreased with increasing ploidy level to about 10% in the hexaploids. For Aegilops it was shown that most of the accessions were homogeneous and highly resistant against powdery mildew (seedling and adult plant stage), leaf rust (adult plant stage) and eyespot (seedling and adult plant stage/natural infection). Summary A collection of 400 Ae. tauschii (syn. Ae. squarrosa) Coss. accessions were screened for powdery mildew resistance based on the response patterns of 13 wheat cultivars/lines possessing major resistance genes to nine differential mildew isolates. 106 accessions showed complete resistance to all isolates, and 174 accessions revealed isolate-specific resistance, among which were 40 accessions exhibiting an identical response pattern as wheat cultivar Ulka/*8Cc which is known to possess resistance gene Pm2. Expression of both complete and isolate-specific resistance from Ae. tauschii was observed in some synthetic hexaploid wheats derived from four mildew susceptible T. durum Desf. parents, each crossed with five to 38 resistant diploid Ae. tauschii accessions. Synthetic amphiploids involving different combinations of T. durum and Ae. tauschii generally showed a decrease in resistance compared with that expressed by the Ae. tauschii parental lines.  相似文献

Lentil anthracnose (Colletotrichum truncatum (Schwein.) Andrus et W.D. Moore is a potential threat in many lentil (Lens culinaris Medik.) production regions of North America. In the lentil germplasm maintained in Germany and North America, 16 lines were reported to have resistance to race Ct1, but none has resistance reported to race Ct0. The objective of this study was to examine accessions of wild Lens species for their resistance to races Ct1 and Ct0 of lentil anthracnose. Five hundred and seventy-four wild accessions of six species and control lines were screened in two replications under both field and greenhouse conditions using a 1–9 scoring scale (1, highly resistant; 2–3, resistant; 4–5, moderately resistant; 6–7, susceptible; and 8–9, highly susceptible). Indianhead and PI 320937 were resistant while Eston and Pardina were susceptible to race Ct1 as expected. However, none of the check lines were resistant to race Ct0. Among the six Lens wild species tested, accessions of Lens ervoides (Brign.) Grande had the highest level of resistance, 3–5 to race Ct1 and Ct0 followed by L. lamottei Czefr. in the field and greenhouse. Lens orientalis (Boiss.), L. odemensis L., L. nigricans (M. Bieb.) Godron and L. tomentosus L. were highly susceptible, 8–9 to race Ct0 in the greenhouse. The highest frequency of resistance, especially in L. ervoides (Brign.) Grande, was found in accessions originating from Syria and Turkey. The usefulness of these L. ervoides (Brign.) Grande accessions as sources of resistance to the more virulent race of anthracnose in a lentil breeding program is discussed.  相似文献

The extent and patterns of microsatellite diversity in 141 Ethiopian tetraploid wheat landraces consisting of three species Triticum durum Desf., T. dicoccon Schrank and T. turgidum L. were analyzed using 29 microsatellite markers. A high level of polymorphism and a large number of alleles unique for each species were detected. Compared to emmer (T. dicoccon) and poulard (T. turgidum) wheats, a higher genetic diversity was observed in T. durum. The A-genome was more polymorphic than the B-genome in all the three species. Microsatellites with (GA) n -repeats had a higher number of alleles than (GT) n -repeats. A species pairwise comparison was made to determine the percentage of shared alleles and a large number of common alleles among species were observed. Average gene diversity, across the 29 microsatellite loci, was 0.684 for T. durum, 0.616 for T. dicoccon and 0.688 for T. turgidum. Genetic distances were lower between T. durum and T. turgidum (0.26) than between T. durum and T. dicoccon (0.34) or between T. turgidum and T. dicoccon (0.38). A significant correlation (p < 0.01) was found between the number of alleles per locus and the gene diversity in all the three species. Allelic frequency variation was highest between T. turgidum and T. dicoccon (10.62%) and lowest between T. durum and T. turgidum (4.86%). A genetic similarity coefficient of 0.34, 0.46 and 0.37 was found in T. durum, T. dicoccon, and T. turgidum, respectively. The dendogram, which was constructed on the basis of a similarity matrix using the UPGMA algorithm, distinguished all accessions represented in the study.  相似文献

The wide morphological variation of Aegilops tauschii has led to the distinction of different subspecies; a typical ssp. tauschii and a second ssp. strangulata. However some researchers pointed out the existance of the intermediate form among morphologically distinguished subspecies. Distribution, diversity and the relationship between different subspecies and the intermediate form were evaluated in the Iranian Ae. tauschii collection. This collection was classified to 15 different populations according to morphological similarities and the collecting origin of accessions. The highest variation was found in tauschii population of Golestan followed by tauschii populations of Gilan and Ardebill, whereas the lowest variation was observed in tauschii populations of central Iran. Two discriminant functions suggested that the length of rachis node and spikelet glume, particularly, the length/width ratios of these traits had the highest impact on identification of different forms. Mahalanobis distances (D 2 ) between the two subspecies along with intermediate form on the multidimensional scaling plot showed that the intermediate form is more similar to ssp. tauschii than ssp. strangulata. Although, the diversity within the ssp. strangulata was not very high, it widely affected the diversity of Iranian accessions of Ae. tauschii through continues crossing with the more diversed subspecies, tauschii, during thousands of years. This fact had lead to expansion of its distribution from its origin to Northern Khorasan, Northern Semnan and Eastern Ardebill by producing the intermediate form.  相似文献

Seven Brassica species were evaluated for their resistance to the cabbage aphid, Brevicoryne brassicae, in a series of field experiments. Four wild Brassica species, two 8 chromosome species with similarities to the B genome of Brassica nigra (Brassica fruticulosa and Brassica spinescens) and two 9 chromosome species containing the C genome (Brassica incana and Brassica villosa) were identified as possessing consistently high levels of antibiosis mediated resistance to B. brassicae. None of the species were shown to possess consistently high levels of antixenosis resistance. In more detailed glasshouse experiments one B-like genome species, B. fruticulosa, showed considerable variation between accessions collected from different sites for resistance to B. brassicae. In addition, individual accessions of one A genome species (Brassica rapa) and one C genome species (Brassica alboglabra) were shown to be highly variable in their resistance to B. brassicae, some plants of each accession being highly resistant and others very susceptible. The implications of the variability in resistance to B. brassicae within wild Brassica species for exploitation in Brassica breeding programmes are discussed.  相似文献

Today, emmer wheat, T. turgidum subsp. dicoccon, widely grown in the past is a candidate crop for sustainable agriculture in Italy. As part of a research project aimed at the enhanced use of the hulled wheat germplasm, molecular characterization was carried out to understand the genetic structure of the crop and to identify accessions of interest. A collection of 194 accessions was analyzed with 15 microsatellite loci (SSRs), while only a sample of 38 accessions was tested with 19 RFLP probes. The marker loci were selected on the basis of their independent genomic distribution. Genetic distances and allelic frequencies were calculated for each marker class. The genetic relationships were visualized with dendrograms. RFLP loci were, on average, less polymorphic than SSRs. An average Dice's genetic distance of 0.22 for RFLPs vs 0.38 for SSRs was detected, while an expected average heterozygosity per locus of 0.23 for RFLPs vs 0.26 for SSRs was also estimated. With a least number of 10 loci per marker class it was possible to identify each genotype. The most diverse accessions had different geographic origins. Germplasms from Italy and Ethiopia appear to belong to a more primitive genepool, given that a group of accessions from these countries were genetically differentiated from a Russian-Iranian group.  相似文献

Seedlings of 41 emmer (Triticum dicoccon Schrank) and 56 durum (T. durum Desf.) wheat accessions were evaluated for their response to stem rust (Puccinia graminis f. sp. tritici) infection under greenhouse condition at Kulumsa Agricultural Research Center, Ethiopia. The objectives were to identify tetraploid wheat accessions that could serve as sources of resistance to stem rust, and postulate the stem rust (Sr) resistance genes through multipatotype testing. The test included screening of accessions for stem rust resistance and multipatotype testing. To ensure vigorous screening, a mixture of six isolates (Si-1a, Am-2, Ku-3, Dz-4a, Ro-4 and Na-22) that were collected from severely infected emmer, durum, and bread wheat (Triticum aestivum L.) varieties of major wheat growing areas of Ethiopia was used as inocula. Out of the tested accessions, 18 emmer and 6 durum accessions exhibited low infection types (0–2) response and hence selected as a source of resistance to stem rust infection. Multipatotype testing was done to postulate Sr genes in the selected accessions. In the test, 10 different stem rust races (A2, A9, A11, A14, A16, A17, B3, B7, B15, and B21), 33 stem rust differential lines, and a universal susceptible check variety, Morocco were used, The high (3–4) and low infection type reaction patterns of the tested accessions and differential lines were used to postulate the genes that exhibit gene-for-gene relationship. The presence of Sr 7b, 8b, 9a, 9b, 10, 14, 24, 27, 28, 29, 30, 31, 32 and Tt-3+10 genes were postulated in 16 selected emmer and 5 durum wheat accessions. Efforts to transfer these valuable Sr genes from cultivated tetraploid wheats could be rewarding to get stem rust resistant varieties and boost wheat production.  相似文献

Summary Leaf miner (Liriomyza cicerina Rond.) causes considerable damage to chickpea (Cicer arietinum L.) in West Asia, North Africa and Southern Europe. Use of resistant cultivar is the economical method of control. Screening of over 7000 germplasm accessions did not result in identification of highly resistant accessions to this insect. Therefore, 200 accessions of eight wild Cicer species were evaluated in search of resistance to leaf miner between 1988 and 1991 at ICARDA, Syria. Accessions of Cicer species were screened under natural insect infestation in the field during spring (March–June), along with a susceptible-cum-indicator line. Two accessions of C. cuneatum (ILWC-40 and -187) and 10 accessions of C. judaicum (ILWC-44, -46, -56, -57, -58, -95, -103, -196, -206, and -207) were rated 2 on a 1–9 scale, where 1 = free from any damage and 9 = maximum damage. Another 18 lines of C. judaicum, four lines of C. pinnatidum and one line of C. reticulatum were rated 3 (resistant).
