利用ISSR标记对32份安徽省地方枣种质资源及山东、河南等邻近省份的42份枣资源进行了遗传多样性分析。26条引物在74份种质中共扩增出241个位点,其中多态性位点214个,多态性比率为88.8%。利用NTSYS软件计算得到DICE相似系数为0.5936 ~ 0.9353;32份安徽地方种质共扩增出219条电泳谱带,其中186条为多态性条带,多态性比率为84.93%,32份样品遗传相似系数在0.6267 ~ 0.9353之间,表明安徽地方枣种质资源遗传多样性较为丰富。采用UPGMA法对74份种质的ISSR数据进行聚类分析,分析结果显示可分为8个类群,种质间亲缘关系与地理来源关系较为密切,来自安徽的地方种质基本上都独立聚成不同的类群,表明安徽地方枣种质与周边其它省份枣种质资源亲缘关系较远;研究结果表明体细胞突变可能是推动安徽地方枣种质进化的重要因素。  相似文献

采用ISSR分子标记对38份红麻品种进行遗传多样性分析。利用自行筛选的引物共扩增出117条带,其中多态性条带98条,多态性条带比率为83.7%。品种之间的遗传相似系数为0.36~0.98,表明部分红麻品种间存在显著的遗传分化。聚类分析表明:红麻栽培品种间基因型差异较小,亲缘关系较近,遗传基础相对狭窄;而野生种、近缘种和栽培种之间存在着较大的遗传差异性。  相似文献

15份葡萄种质亲缘关系的ISSR分析   总被引:10,自引:2,他引:10  
利用ISSR标记对15份葡萄材料进行了基因组多态性分析,从60条引物中筛选出15条用于葡萄的ISSR扩增。共扩增出105条带,其中多态性条带91条,多态性百分率为86.7%。根据ISSR扩增结果,利用NTSYSpc2.10e软件进行Jaccard相似性系数分析,15份葡萄材料的遗传相似系数为0.27~0.75,平均遗传相似系数为0.471。通过UPGMA进行聚类分析,探索亲缘关系。在遗传相似系数0.39处,15份葡萄材料可分为2大类群。第1类包含7份山葡萄资源、2个山欧杂交品种,第2类包含3个欧亚种品种、2个欧美杂种品种和1个美洲杂种品种。由此可见,山葡萄与欧亚种、美洲杂种葡萄的亲缘关系较远,欧亚种与美洲杂种之间亲缘关系较近。  相似文献

通过杏鲍菇多孢分离菌株的ISSR、RAPD分子标记和体细胞不亲和性分析,体细胞不亲和性分析得出子代杏1、杏3、杏4与出发菌株湘杏98有拮抗线,杏1、杏2、杏3、杏4四株子代均有明显的拮抗线。16条ISSR引物和19条RAPD引物分别扩增出99、154个条带,多态性条带分别占48.48%、61.00%;相似系数变异范围分别为0.63~0.86、0.57~0.83。湘杏98、杏2、杏3在2种标记的聚类结果上存在差异。体细胞不亲和性分析从表型上(拮抗线有无)反应了菌株的遗传变异。RAPD标记遗传相似系数小于ISSR标记,表明ISSR标记更适合亲缘关系较近的种群间遗传多样性分析。  相似文献

从分子水平用ISSR标记法对南方长茄资源的遗传多样性进行分析,从100个ISSR引物中共筛选出12个多态性明显、条带清晰、反应稳定的引物,对57个样品DNA共扩增出116条谱带,平均每个引物扩增出9·67条带,其中多态性位点84个(71%)。品种间遗传相似系数在0·51~0·98之间,表明茄子栽培种内品种间的遗传基础相对较狭窄。利用UPGMA聚类分析,能将57个南方长茄品种划分为6个类群,类群的划分与地方来源没有很大的关系。  相似文献

以来自不同地理区域的48个灵芝种质资源为试材,采用ITS和ISSR分子标记的方法,研究了灵芝遗传多样性,以期利用遗传信息数据较理想地显示出不同灵芝菌株的遗传多样性。结果表明:通过进行ITS序列扩增测序,将48个灵芝菌株分为赤芝(35个)、无柄灵芝(11个)、四川灵芝(1个)、古巴栓孔菌(1个)。5条ISSR引物共扩增得到53条条带,多态性条带比率为96.23%,Nei′s基因多样性为0.27,Shannon′s信息指数为0.43。ISSR标记遗传相似系数约为0.70,将48个灵芝菌株分为3组,其中第1组为11个无柄灵芝,第2组为菌株29“盆景1”,其他菌株为第3组,说明2种标记结合分析能够更加准确分析不同菌株间的亲缘关系。菌株16和17同为赤芝,且ISSR分子标记遗传相似系数达0.98,推测可能为同一品种,为生产中出现的同物异名现象。该研究选取的用于PCR扩增的ISSR引物,多态性较好、稳定性较高,能有效鉴别出无柄灵芝与其他灵芝,并揭示种质的遗传多样性和群体遗传结构,可为灵芝种质资源保护及优质种质材料选育提供参考依据,以期更好地推动灵芝产业健康发展。  相似文献

通过ISSR技术对19个野生天门冬居群共67个个体进行遗传多样性分析.结果表明:用13个随机引物共扩增出125条清晰条带,其中92条具多态性,平均多态性位点比率为73%,建立了不同居群的ISSR标记;Nei's基因多样性指数H=0.19,Shannon,s多样性指数I=0.30,遗传分化指数Gst=0.8206.聚类分析表明,青海、云南和贵州省的5个居群聚为一大支,其它居群为另一大支.野生天门冬种内具有较高的遗传多样性,遗传变异主要存在于居群间;ISSR标记可以作为研究天门冬遗传多样性及居群鉴定的有效标记.  相似文献

利用ISSR标记对22份观赏贝母材料进行了基因组多态性分析,从100条引物中筛选出13条用于观赏贝母的ISSR扩增。共扩增出178条带,其中多态性条带160条,多态性百分率为90%。根据ISSR扩增结果,利用NTSYSpc2.10e软件进行Jaccard相似性系数分析,22份观赏贝母的遗传相似系数为0.2338~0.8788。通过UPGMA进行聚类分析,探索亲缘关系。在遗传相似系数0.50处,22份观赏贝母材料可分为二大类群。第一大类包括比较原始的贝母组和特化的聚花组和单鳞组的贝母;第二大类包括演化水平较高的多鳞组的3种贝母。由此可见,同一组观赏贝母种质之间表现较近缘的亲缘关系。  相似文献

利用ISSR分子标记对22份南丰蜜橘及其近缘品种的遗传多样性水平进行研究。从31个ISSR引物中筛选出13个引物,通过琼脂糖凝胶电泳共检测到119条稳定的条带,片段大小介于0.5~4 kb之间,条带数在8~13之间;扩增片段中多态性位点98个,平均多态性比率为82.35%。各品种间的Ne i`s基因多样性指数(H)和Shannon信息多样性指数分别为0.289 5和0.433 3,品种间遗传相似指数(GS)范围为0.487 4~0.949 6。通过UPGMA聚类分析,当遗传相似系数为0.72时,可以将22份南丰蜜橘及近缘品种分成3大类。  相似文献

艺性状观测分析。从51 条ISSR 引物中筛选到13 条多态性高、扩增稳定、重复性好的引物,这13 条引
物共扩增出132 个位点,其中109 个多态位点;应用Nei-Li 相似系数法估算了48 份材料间的遗传相似系
数,结果表明,48 份分蘖洋葱遗传相似系数变化范围在0.59 ~ 0.94 之间。对分蘖洋葱的抽薹情况、幼叶
色、鳞茎色和单球质量进行了调查,结果表明,48 份分蘖洋葱遗传相似系数变化范围在0.38 ~ 1.00 之间。
ISSR 标记和农艺性状分析均表明48 份分蘖洋葱的遗传多样性较丰富;两种方法的聚类结果基本一致,聚

Genetic diversity within Zinnia elegans is key to the genetic improvement of this important ornamental species. Here, morphological traits and RAPD and ISSR molecular markers were used to assess levels of polymorphism across 20 inbred lines. Thirty-four morphological traits were scored and also 147 RAPD marker-fragments, as amplified by 12 arbitrary primers, and 128 ISSR marker-fragments as generated by 9 primers. The number of polymorphic loci, the percentage of polymorphic loci, Shannon's Information index (I) and the effective number of alleles (Ne) were calculated from the RAPD data as 100, 68.03%, 0.3559 and 1.4169, respectively. From the ISSR data, these respective statistics were calculated as 97, 76.38%, 0.4013 and 1.4728. Thus, ISSR markers were considered slightly superior to RAPD markers for assessing genetic diversity between the accessions; however, Mantel's test indicated significant correlation (R = 0.733) of the RAPD and ISSR results. By contrast, the morphological matrix showed low correlation with both RAPD and ISSR data matrices (R = 0.3814 and 0.3765, respectively). Cluster analysis showed that groupings of the accessions according to all three methods correlated well with their geographic region of origin, but flower color was not strongly associated with the genetic classification of these inbred lines of Z. elegans.  相似文献

以保定野菊(Chrysanthemum indicum)、菊花脑(C.nankingense)、元氏甘菊(C.lavandulifolium)、龙脑菊(C.japonicum)、若狭滨菊(C.japonicumvar.wakasaense)、淄博野菊(C.indi-cum)6种菊属植物为试材进行细胞学研究。结果表明:保定野菊的核型公式为2n=2x=18=16m+2st,核不对称系数为56.58%;菊花脑的核型公式为2n=2x=18=12m+6sm,核不对称系数为60.47%;元氏甘菊的核型公式为2n=2x=18=12m+4sm+2st,核不对称系数为62.12%;龙脑菊的核型公式为2n=2x=18=14m+4sm,核不对称系数为66.46%;若狭滨菊的核型公式为2n=4x=36=28m+6sm+2st,核不对称系数为59.92%。淄博野菊的核型公式为2n=4x=36=26m+10sm,核不对称系数为65.88%。  相似文献

 通过ISSR标记结合形态学性状,分析了茶树‘黄山种’自然杂交后代的遗传多样性。所调查形态学性状的平均变异系数为20.5%(12.5% ~ 33.2%),Shannon-weave指数平均为1.89(1.60 ~ 2.07)。22条ISSR引物在供试样品中共扩增出402条带,其中多态性条带396条,多态性比率为98.5%,引物的多态性信息指数(PIC)平均达0.90,供试样品的基因多样性(H)和Shannon信息指数(I)分别为0.32和0.49。充分表明‘黄山种’通过自然杂交,发生了基因的分离和重组,产生了丰富的遗传变异。Mantel检测基于形态学与基因型估算的遗传距离矩阵间,存在极显著正相关(r = 0.61,P = 0.01)。基于形态学性状和ISSR标记的聚类结果显示,大多数‘黄山种’自然杂交后代的单株可聚成一大类群,只有少数单株遗传距离较远,单独聚为一类。这些单株一芽三叶长、百芽质量较大,可为今后‘黄山种’系统选种提供优异种质材料。  相似文献

In this study, the genetic diversity of 50 individuals of rocket, Eruca vesicaria, from five accessions, four of them wild type collected from different parts of Spain and one commercial, were evaluated using morphological, agronomical and inter simple sequence repeat DNA (ISSR) data. Molecular analysis was carried out using the ISSR technique with 20 primers. Out of these 20 primers, nine were polymorphic, producing a total of 395 DNA bands, 247 of which were polymorphic among the accessions. A dendrogram drawn on the basis of a similarity matrix using the UPGMA algorithm revealed that the 50 samples of rocket plants could be classified into three major clusters at a Nei's genetic distance of 0.36. The experiment shows that molecular markers such as ISSR are a good instrument for distinguishing and selecting rocket accessions to group different wild populations. In general, a high variation was observed for most of the 16 morphological and 6 agronomical traits showing significant differences. Some morphological traits such as leaf length, petiole length and lamina width explained 69.1% of the whole variation observed in the populations, and some agronomical traits such as leaf area, nitrate and chlorophyll contents accounted for 65.7%, but the clusters generated by means of agronomical and morphological variables were less evident than when ISSR markers used. Some accessions showed good qualities, such as small leaves, high chlorophyll content, late-flowering or low nitrate content. All these parameters, together with the high degree of genetic homogeneity found, could make the local accessions good candidates for a future breeding programme.  相似文献

Improvement of fruit traits is an important objective in current mulberry breeding programs. In this study, 93 mulberry accessions of diverse origin were genotyped using 15 ISSR markers to identify marker–trait associations with fruit traits. Fifteen ISSR primers generated a total of 104 amplification products, of which 94 were polymorphic, revealing 90.38% polymorphism; the mean PIC value was 0.2698. UPGMA cluster analysis showed clear genetic relationships between the 93 mulberry cultivars, and the major clusters were related to known pedigree relationships and their ecotype. The mean r2 value for all intra-chromosomal loci pairs was 0.0210. Marker–trait associations were investigated using the unified mixed-model approach, considering both population structure (Q) and kinship (K). In total, 24 marker–trait associations (< 0.01) were identified using different ISSR markers. The results suggest that association mapping in mulberry is a viable alternative to quantitative trait loci mapping, and detection of associations between markers and mulberry fruit traits will also provide important information for marker-assisted breeding.  相似文献

In this study, RAPD and ISSR markers were used to investigate the genetic diversity and genetic relationships among different germplasm of Nelumbo including 70 Chinese ornamental cultivars, 7 wild Thai genotypes, 2 Nelumbo lutea genotypes and 8 hybrids of Nelumbo nucifera and N. lutea. High genetic diversities of 96.4% and 91.2% respectively were detected in the Nelumbo accessions using RAPD and ISSR markers. A dendrogram based on both RAPD and ISSR clustering data indicated that: (1) the genotypes of N. nucifera and N. lutea from different geographical origins were clustered into different groups. This indicated significant genetic differentiation attributed to extensive periods of geographical isolation and lack of gene exchange; (2) the Thai wild genotypes were separated from Chinese genotypes. This indicated genetic divergence between germplasm from Southeast Asia and that from China. Geographical location appears to have affected genetic diversity due to adaptation of the plants to the different environments. A new Southeastern Asia Lotus category is suggested as an addition to the current lotus cultivars classification system; (3) data on three morphological traits (namely: plant size, petal shape and flower color), showed that only the data on plant size was consistent with the dendrogram constructed from molecular data. This finding suggests that using data on genetic relationships in combination with morphological characteristics would serve to improve the classification system of lotus cultivars currently in use. The finding of previously unknown germplasm in this study indicated the potential of RAPD and ISSR techniques in identifying and managing lotus resources. Both marker techniques are potentially useful in improving the current strategies in breeding and germplasm conservation to enhance the ornamental and economic value of lotus.  相似文献

利用由野生种番茄(Solanum galapagense)‘LA0317’和普通栽培种番茄(Solanum lycopersicon)‘9706’构建包含130个株系的BC2S8群体,对控制果实质量和果实形状的QTL进行分析,共检测到18个QTL,LOD值介于2.40~5.17之间,可解释8.0%~32.4%的表型变异,其中LOD值大于3的QTL有5个,贡献率超过10%的QTL有8个。与果实质量相关的QTL有5个,加性效应值均为负值。与果形指数相关的QTL有6个,与果实纵径相关的QTL有2个,与果实横径相关的QTL有5个。在分子标记SSR135、In Del_FT-77、TMS66和In Del_FT-143处均聚集了多个控制单果质量和果形的QTL,说明这些位点可能同时对多个不同的果实表型性状起作用,属于多效位点。  相似文献

利用组织培养的方法,首次以野菊花蕾作为外植体,借鉴前人组织培养观赏菊花的经验,找到一种使野菊能够快速增殖的方法.以MS为基本培养基,添加BA 1.0 mg/L和NAA0.2 mg/L作为外植体的分化与继代培养基,得到愈伤组织和大量的丛生芽;用MS培养基进行增殖培养;用1/2MS+BA 1.0 mg/L+NAA 0.2 mg/L进行生根培养,生根率在98%以上.最后通过驯化练苗得到大量的野菊再生植株.  相似文献
