Functional response of Harmonia dimidiata (Fab.) to melon aphid, Aphis gossypii Glover was studied in the laboratory at 25 ± 0.5 °C, 70 ± 5% RH and 12L:12D photoperiod. All the predator stages exhibited Type II functional response to the aphid. Based on Rogers’s random predator equation the predator’s attack rate was lowest (0.128) in the first-instar and highest (0.21) in the adult stage. Handling times of the adult beetle and the fourth-instar (0.148 and 0.164 h, respectively) were much shorter than other stages. The maximum number of aphids that could be consumed over a period of 24 h by first, second, third and fourth instar; and adult of H. dimidiata was estimated to be 47.2, 80.2, 79.7, 146.2 and 161.7 aphids/ predator. Functional response parameters indicate that the adult beetles and the fourth-instar were the most voracious stages and could be important for the biological control of the aphid.  相似文献

An internal fruit rot with a malodor was found in netted melons (Cucumis melo L.) in commercial greenhouses in Kochi Prefecture, Japan, in 1998, despite their healthy appearance and lack of water-soaking or brown spots on the surface. A yellow bacterium was consistently isolated from the affected fruits. To confirm the pathogenicity of eight representative isolates of the yellow bacterium, we stub-inoculated ovaries (immature-fruits) 5–7 days after artificial pollination, with a pin smeared with bacteria. After the melon fruits had grown for 60 more days, an internal fruit rot resembling the natural infection appeared, and the inoculated bacterium was reisolated. The melon isolates had properties identical with Pantoea ananatis, such as gram-negative staining, facultative anaerobic growth, indole production, phenylalanine deaminase absence, and acid production from melibiose, sorbitol, glycerol, and inositol. Phylogenetic analysis based on 16S rDNA sequences showed that the melon bacterium positioned closely with known P. ananatis strains. The melon bacterium had indole acetic acid (IAA) biosynthesis genes (iaaM and iaaH) and a cytokinin biosynthesis gene (etz). The bacterium could be distinguished from the other ‘Pantoea’ group strains by rep-PCR genomic fingerprinting. From these results, the causal agent of internal fruit rot was identified as a strain of P.ananatis [Serrano in (Philipp J Sci 36:271–305, 1928); Mergaert et al. in (Int J Syst Bacteriol 43:162–173, 1993)]. Field studies were conducted to assess the population and the spatial dynamics of the predatory bugMacrolophus costalis Fieber (Hemiptera: Miridae) and of its prey, the aphidMyzus persicae (Sulzer) (Hemiptera: Aphidoidea), on tobacco. From an untreated tobacco field in Tithorea (central Greece), tobacco leaves were collected from the upper and the lower half of the plants from June until September, in 1999 and 2000. The numbers ofM. costalis andM. persicae individuals per leaf were counted. Most aphids were observed during July and August (early and mid season), with densities dropping markedly in September. In contrast,M. costalis population densities increased late in the season (September). Significantly higher numbers of aphids were found on the upper half of the plants than on the lower half. In contrast, significantly moreM. costalis individuals were observed on the lower half. Iwao’s Regression Analysis was used in order to characterize the spatial pattern of the two species. According to this model, in both sampling seasons, aphids andM. costalis nymphs displayed an aggregated spatial pattern, whileM. costalis adults were found to be randomly distributed among sampling units. Although moreM. costalis individuals were recorded on leaves with relatively high aphid densities, this species did not react numerically to changes in prey density. Among the factors affecting the quality and yield of garlic production, blue mold caused by -- Penicillium spp. -- is responsible for economical losses in many countries. Allicin, present in garlic bulbs, has been suggested as having antifungal activity against some Penicillium species. This study was conducted to evaluate the response of garlic accessions against Penicillium hirsutum infection and to compare this response with bulb allicin content. Twelve garlic accessions were inoculated with P. hirsutum, and assayed in greenhouse and growth chamber experiments. Plant growth parameters and the fungal production of conidia were evaluated. Significant differences were found among the accessions. Accessions Castaño and Morado were most resistant whereas AR-I-125 and Fuego were always severely affected by the disease. A low correlation was found (r = 0.17) between allicin content and tolerance, indicating that allicin is not the main factor involved in the resistance against P. hirsutum.  相似文献

The role of the self-sown shrubsDittrichia viscosa (L.) W. Greuter andRubus ulmifolius Schott as reservoirs of aphid parasitoids was investigated. In the field studies conducted,D. viscosa grew adjacent to crops of durum wheat and barley andR. ulmifolius grew adjacent to cotton. The relative abundance of the parasitoids of(a) Capitophorus inulae (Passerini) onD. viscosa, (b) Rhopalosiphum padi (Linnaeus) on durum wheat and barley,(c) Aphis ruborum (Börner) onR. ulmifolius, and(d) Aphis gossypii Glover on cotton in various parts of Greece, was assessed during the years 1996–2000. In 2000, the fluctuation of parasitization of the above four aphid species was recorded and the action of the aphidophagous predators of the family Coccinellidae was studied. It was observed thatAphidius matricariae Haliday predominated onC. inulae andR. padi in all sampling cases. In contrast,Lysiphlebus fabarum (Marshall) was the dominant species parasitizingA. ruborum onR. ulmifolius andA. gossypii on cotton in Thessaly (central Greece) and Macedonia (northern Greece), whereasLysiphlebus confusus Tremblay et Eady andBinodoxys acalephae (Marshall) were the dominant parasitoid species in Thrace (northern Greece).Coccinella septempunctata Linnaeus was the most abundant coccinellid species on durum wheat, whereasAdonia variegata (Goeze) predominated on cotton. However, coccinellid individuals were scarce on bothD. viscosa andR. ulmifolius. The present study indicated that these two shrubs can be regarded as useful reservoirs of aphid parasitoids.  相似文献

Many arthropod agricultural pests have become resistant to pesticides through inappropriate usage. A consequence of this resistance is often cross-resistance with other pesticides with similar or related modes of actions. As a result of increasing pesticide resistance, a movement to alternative, ‘green’ botanical pesticides has been growing. A new formulation of volatile oils, extracted from a variety of plants allows for slow-release and longer duration in the field. In this publication we present results of the efficacy of one of these products against the cotton aphid Aphis gossypii on cucumber plants in replicated choice tests. The slow release Ap2, a Bio-Pel product from Newseal Mineral and Coatings Ltd. (hereafter Newseal), significantly reduced the population of aphids on test plants and prevented their movement upwards on the plant. Additionally, four Newseal products caused no mortality to the aphid parasitoid Aphidius colemani or to the generalist predator, Orius laevigatus.  相似文献

Yeast-like fungi were isolated from lesions on azuki bean (cv. Shin-Kyotodainagon) seeds that had been sucked by bean bugs in Kyoto Prefecture, Japan. On the basis of morphological and physiological characteristics and sequence data of the internal transcribed spacer (ITS) regions including the 5.8S rDNA, these yeasts were identified as Eremothecium coryli and E. ashbyi. Pathogenicity of those yeasts was confirmed by a reinoculation test. To our knowledge, this is the first report of the occurrence of yeast spot in azuki bean in Japan. Cucumber mosaic virus (CMV) was isolated from a mosaic diseased plant of Eucharis grandiflora. The virus caused mosaic symptoms on leaves and slight distortion of flower petals in E. grandiflora by either mechanical or aphid inoculation. Thirty-two accessions of wild tomato (Lycopersicon spp.) germplasm were evaluated for resistance to the whiteflyBemisia argentifolii Bellows ⇐p; Perring in a greenhouse choice bioassay. Density data were recorded for the adaxial and abaxial leaf surfaces for (i) all life stages ofB. argentifolii and (ii) types I, IV, V and VI trichomes. Individual plant selections (33 from 22 wild tomato accessions) with high resistance were subsequently tested in the field to verify the resistance found in the greenhouse screening. Resistance was defined by the density of all life stages of the whitefly observed on the eight leaflets sampled at nodes 5 and 7. Only type IV trichomes had a consistent (but low) and significant negative correlation between trichome density and whitefly density for various life stages. The highest whitefly resistance was observed inLycopersicon pennellii accessions LA 716, LA 1340 and LA 2560. Five accessions ofArabidopsis thaliana, Ws-3, Nd-1, Ler, Col-5 and Oy-0, were inoculated withPeronospora parasitica isolate Emoy2 and the accumulation of camalexin within infected tissues was measured. The variations in camalexin accumulation in various accessions ofA. thaliana induced by a specific (P. parasitica isolate Emoy2) and a non-specific elicitor (UV-B irradiation) were investigated. Phenotypic examination of Emoy2/Oy-0 revealed that susceptibility was characterized by extensive asexual sporulation of the pathogen, whereas early restriction of the pathogen in infected plant tissues, accompanied by chlorosis and necrosis — which are associated with the hypersensitive response — was observed in Nd-1, Ws-3 and Ler. Partial resistance detected in Col-0 was characterized by low to medium sporulation of the pathogen. Camalexin was monitored by high pressure liquid chromatography (HPLC) and found to accumulate during both compatible and incompatible interactions and also following treatment with the abiotic inducer, UV-B. Among the accessions tested, Ws-3 yielded significantly more camalexin than the other accessions, regardless of which inducers (biotic or abiotic) were used. There was no significant correlation between resistance and camalexin accumulation in theA. thaliana/P. parasitica interaction. Laboratory and field experiments were conducted to study the effect of UV filtration on the population growth, distribution and flight activity of the green peach aphidMyzus persicae (Sulzer) (Homoptera: Aphididae), and on the fecundity and host-finding behavior of the parasitic waspAphidius matricariae (Haliday) (Hymenoptera: Braconidae). The work was done in the Arava Valley of Israel, in walk-in tunnels covered with polyethylene film, to compare the effects of UV-absorbing filmversus those of regular film. Following artificial aphid infestation on pepper grown under the tested films, aphid population growth and spread under the UV-absorbing films were significantly less than under the regular films. The greatest impact of UV-absorbing film on aphid behavior was observed in winter and early spring, when temperature conditions favor aphid development. Elimination of UV by UV-absorbing film did not affect the parasitic activity ofA. matricariae. Previous results had indicated that covering the greenhouse with UV-absorbing films inhibited the invasion of aphids and other insect pests into it. That effect, in combination with those described in the present paper, makes the use of UV-absorbing films an effective component of IPM that aims to reduce the application of toxic insecticides. Intercellular washing fluid (IWF) obtained from the susceptibleArabidopsis accession Ws-eds1 inoculated withPeronospora parasitica isolate Emoy-2, contained an elicitor of necrosis with ecotype specificity towardsArabidopsis accessions with particular resistance genes. This elicitor caused necrosis on the highly resistant accessions La-er, Nd-1 and partly on Col-5, but not on the susceptible accessions Ws-eds1 and Oy-0. In resistant plants, injection of IWF caused hypersensitive reaction (HR)-like cell collapse which was associated with the accumulation of phenolics and lignin-like material in walls of cells undergoing cell death. The elicitor is sensitive to proteinase K and pronase enzymes, heating and autoclaving but insensitive to periodate oxidation, freezing and thawing, and is not dialyzable. Results suggest that the elicitor is a protein. Muskmelon (Cucumis melo cv. Temprano Rochet) and autumn squash (Cucurbita maxima) seedlings were inoculated either with Acremonium cucurbitacearum or Monosporascus cannonballus, two of the soil-borne fungi implicated in ‘melon collapse’. Inoculation was achieved in two different ways: by growing the plants in pots containing infested soil to study the histological changes produced in the infected tissues using light microscopy and by growing seedlings in Petri dishes together with fungal colonies in order to observe the colonisation of the plant tissues using scanning electron microscopy. Both muskmelon and autumn squash roots infected with A. cucurbitacearum showed a suberised layer in the epidermis and the outermost layers of the parenchymatic cortex, but these symptoms developed earlier in the muskmelon plants. Muskmelon plants infected by this fungus also presented hypertrophy and hyperplasia, which led to a progressive separation of the vascular bundles in the lower stems of the affected plants. This response was not observed in autumn squash during the study. On the other hand, few histological changes were observed in tissues infected with M. cannonballus and only a slight increase in the size of cortical intercellular spaces was noted in the lower stems of muskmelon plants, and infected autumn squash tissues remained free of these symptoms throughout the study. The scanning electron microscope observations revealed that both fungi were able to colonise the tissues of the two host plants which were studied. A. cucurbitacearum colonised the epidermis and cortex of both muskmelon and autumn squash. The hyphae grew both inter- and intracellularly, and the density of the colonisation decreased within the endodermis. The same colonisation of host plants was observed as a result of M. cannonballus infection. The xylem vessel lumina of both muskmelon and autumn squash showed hyphae and tylose formation as a result of both fungal infections. However, non-fungal structures were detected in the hypocotyl vascular tissues. The present study demonstrates that both fungi are capable of infecting the tissues of a species which is resistant (autumn squash) and a species which is susceptible (muskmelon) to melon collapse.  相似文献

Insect-borne viruses promote several changes in plant phenotype, which can modify plant-vector interactions in favor of virus survival and dissemination. Although co-infections commonly occur in the field, little is known about their effects on interactions with the vector. The ecological interactions between Barley Yellow Dwarf Virus (BYDV) and its aphid vector, Rhopalosiphum padi, have been investigated extensively, but the vector’s behavior in more complex scenarios has yet to be examined. We assessed olfactory response and performance of R. padi to wheat singly and doubly infected by the pathogenic fungus Giberella zeae and BYDV. Non-viruliferous aphids preferred odors of BYDV-infected wheat over healthy wheat, as previously reported in the literature, and they were still preferentially attracted to BYDV-infected plant during co-infection. However, around 35% more non-viruliferous aphids chose healthy wheat over G. zeae-infected wheat. Viruliferous aphids did not show any preference to the treatments. BYDV-infected wheat was a superior host than healthy wheat for the aphids whose population increased in 25%. We observed a synergistic effect of the co-infected wheat, which was the best host for aphids, and promoted an elevation of 42% on population growth. Our results indicate that co-infection might be beneficial for virus spread as does not interfere with aphid olfactory preference and provides greater colony growth than in singly infected plants.  相似文献

Sixty-nine tomato genotypes representing nine Solanum species were evaluated for resistance to Cucumber mosaic virus (CMV) subgroup IA and its aphid vector Myzus persicae. Resistance was assessed by visual scoring of symptoms in the field under natural conditions, and in the greenhouse by artificial inoculations through aphid M. persicae and mechanical transmissions in the year 2007 and 2009. Considerable variation in responses was observed among the evaluation methods used. Field evaluations were found liable to errors as different levels were observed for the same genotypes in the different years, however mechanical inoculation was found to be the most useful in identifying CMV subgroup IA resistance, in contrast aphid transmission was most useful in identifying insect transmission resistance. All genotypes observed as highly resistant to CMV subgroup IA in the field or through vector transmission became systemically infected through mechanical inoculations. Using mechanical inoculation, six genotypes (TMS-1 of S. lycopersicum, LA1963 and L06049 of S. chilense, LA1353, L06145 and L06223 of S. habrochaites) were found resistant and another six (L06188 and L06238 of S. neorickii, L06219 of S. habrochaites, L05763, L05776 and L06240 of S. pennellii) were found tolerant showing mild symptoms with severity index (SI) ranging 1-2 and with delayed disease development after a latent period (LP) of 18–30 days. However, these genotypes were found to be resistant to highly resistant in the field and through inoculation by M. persicae; and they also supported low population levels of M. persicae except TMS-1. Another nine genotypes (LA2184 of S. pimpinellifolium L., LA2727 of S. neorickii, LA0111, L06221, L06127 and L06231 of S. peruvianum L., LA1306, L06057 and L06208 of S. chmielewskii) showing a susceptible response after mechanical inoculation were highly resistant, resistant and tolerant after M. persicae transmission. The resistant genotypes, identified in the present study can be exploited in the breeding programmes aimed at developing tomato varieties resistant to CMV subgroup IA and broadening the genetic base of CMV-resistant germplasm. The differences observed between mechanical and aphid transmission suggests that one should consider both evaluation methods for tomato germplasm screening against CMV subgroup IA.  相似文献

Severe spotting, blight and drop of leaves caused by Colletotrichum dematium were found on potted plants of Polygonatum falcatum, a liliaceous ornamental, in open fields in Kagawa Prefecture, Japan, in May 2001. This new disease was named anthracnose of P. falcatum. Keisuke Tomioka, Jouji Moriwaki, Toyozo Sato contributed equally to this work. The fungal isolate and its nucleotide sequence data obtained in this study were deposited in Genebank, National Institute of Agrobiological Sciences and the DDBJ/EMBL/GenBank databases under accessions MAFF239500 and AB334523, respectively.  相似文献

The study deals with the tachinid parasitoids ofAncyrosoma leucogrammes (Gmelin) (Heteroptera: Pentatomidae):Clytiomya dupuisi Kugler,Clytiomya sola (Rondani) andGymnosoma clavatum (Rohdendorf) (Diptera: Tachinidae). All species are recorded for the first time as parasitoids ofA. leucogrammes, andC. dupuisi was reared for the first time from a host. Solanum nigrum, black nightshade, is a wild non-tuber bearing hexaploid species with a high level of resistance to Phytophthora infestans (Colon et al. 1993), the causal agent of potato late blight, the most devastating disease in potato production. However, the genetic mode of resistance in S. nigrum is still poorly understood. In the present study, two S. nigrum accessions, 984750019 (N19) and #13, resistant (R) and susceptible (S), respectively, to three different isolates of P. infestans, were sexually crossed. The various kinds of progeny including F1, F2, F3, and backcross populations (BC1; F1 × S), as well as two populations produced by self-pollinating the R parent and S parent, were each screened for susceptibility to P. infestans isolate MP 324 using detached leaf assays. Fifty seedling plant individuals of the F1 progeny were each resistant to this specific isolate, similarly to the seedling plants resulting from self-pollination of the resistant R parent. Thirty seedling plants obtained from self-pollination of the S parent were susceptible. Among a total of 180 F2 plants, the segregation ratio between resistant and susceptible plants was approximately 3: 1. Among the 66 seedling plants of the BC1 progeny originating from crossing an F1 plant with the susceptible S parent, there were 26 susceptible and 40 resistant plants to P. infestans. The segregation patterns obtained indicated monogenic dominant inheritance of resistance to P. infestans isolate MP 324 in S. nigrum acc. 984750019. This gene, conferring resistance to P. infestans, may be useful for the transformation of potato cultivars susceptible to late blight.  相似文献

Downy mildew, incited by the comycetePseudoperonospora cubensis (Berk.et Curt.) Rost., was recorded in Israel during the years 1979–2001 on cucumber (Cucumis sativus) and melon (Cucumis melo) and classified as pathotype 3. In July 2002 severe outbreaks of downy mildew occurred on pumpkin (Cucurbita moschata) and summer squash (Cucurbita pepo subsp.pepo). Host range pathogenicity studies revealed high compatibility of the new population with cucumber, melon, pumpkin and summer squash but low compatibility with watermelon.
