Erythrocytic lipid peroxidation, activities of some antioxidant enzymes and osmotic fragility of red blood cells was studied in adult (>1 year) crossbred cattle naturally infected with Theileria annulata. Twenty clinically healthy animals (group I) and 15 clinical cases (group II) of tropical theileriosis were selected. Cattle suffering from theileriosis had significantly higher (p<0.01) erythrocytic lipid peroxidation and osmotic fragility. Activities of antioxidant enzymes, viz. glucose-6-phosphate dehydrogenase (G6PD) and glutamate peroxidase (GPx) were also significantly increased (p<0.01) in group II. However, superoxide dismutase and catalase did not show significant changes. The results indicated that infection with theileria led to increased oxidative stress to the animals, and even a significant rise in the activities of antioxidant enzymes. G6PD and GPx could not lower this oxidative stress. However, the increase in the activities of antioxidant enzymes pointed towards the body’s defence mechanism against lipid peroxidation during oxidative stress in theileriosis.  相似文献

In order to investigate the alterations of erythrocyte protective antioxidant mechanisms, lipid peroxidation and trace elements associated with anemia in bovine tropical theileriosis, an infected group comprised of 50 crossbred Holstein cattle, about 1-2 years old, naturally infected with Theileria annulata, were divided into 4 subgroups according to their parasitemia rates (<1%, 1-3%, 3-5%, >5%) and also 10 healthy cattle as control were selected. Blood samples were taken and hematological parameters, the activities of antioxidant enzymes including superoxide dismutase (SOD), glutathione peroxidase (GPX) and catalase and serum concentrations of some antioxidant trace elements (copper, iron, zinc, manganese and selenium) were measured. As an index of lipid peroxidation, the level of Malondialdehyde (MDA) was also determined. The results showed a conspicuous decrease in the activities of SOD, GPX and catalase (P<0.01), and a significant decrease in the serum concentrations of Cu, Zn, Mn and Se in cattle with higher than 1% parasitemia (P<0.05) compared to the control. In addition, remarkable elevations in the MDA level (P<0.01) and serum concentration of iron (P<0.05) were observed in the infected animals. These findings pointed to the occurrence of exacerbating oxidative injuries to erythrocytes during parasitemia. Furthermore, it can be concluded that infection with T. annulata can interfere with protective antioxidant mechanisms of RBCs against oxidative damages, which promote the development of anemia.  相似文献

Babesia bigemina infection (also known as Texas fever) is reported as the most prevalent and main causative agent of bovine babesiosis, worldwide. The current study was undertaken to assess indicators of oxidative stress including activities of antioxidant enzymes and total antioxidant capacity (TAC), oxidation status of biomolecules and serum levels of trace elements as well as indicators of inflammation including sialic acid (SA) contents and cholinesterase activity in cattle naturally infected with B. bigemina. An infected group comprised of 20 crossbred Holstein cattle (3–4 years old) were diagnosed to be positive by both microscopy and nested PCR assay. The infected animals were subdivided into two groups according to their parasitemia rates (<20 % and>20 %). Furthermore, 10 healthy cattle were included as the control. The infection caused severe anemia in a parasitemia-burden dependent fashion. The activities of catalase and glucose-6-phosphate dehydrogenase as well as the levels of TAC, zinc, selenium, copper and manganese were significantly decreased as the parasitemia increased, accordingly. However, the activity of superoxide dismutase as well as the levels of malondialdehyde, protein carbonylation, DNA damage and iron, were significantly elevated in a parasitemeia-burden dependent manner. Additionally, glutathione peroxidase activity was significantly elevated with the lower rate of parasitemia, but the higher rate had no significant effect as compared to control. Moreover, total, protein and lipid binding SA contents were significantly increased but the activities of acetylcholinesterase and butyrylcholinesterase were significantly reduced, parasitemia dependently. Conclusively, the infection was remarkably associated with the induction of anemia, oxidative stress and inflammation.  相似文献

Erythrocytic lipid peroxidation has been implicated as a cause of anemia in Theileria annulata infection in cattle. The present study aimed to evaluate oxidative damage of membrane lipids and proteins in addition to hemoglobin (Hb) as three criterions of erythrocyte oxidation and their relation to erythrocyte deformability and anemia of newborn crossbred calves (Friesian × Egyptian Balady breed) naturally infected with T. annulata. Twenty-five T. annulata-infected calves (aged 20-30 days) along with 15 age matched healthy controls were used. Percentage of parasitemia varied from 12% to 63% (34.76 ± 3.05%). In comparison to controls, infected calves showed increased levels (P<0.001) of lipid peroxidation (malondialdehyde; MDA, 52%) and protein oxidation (protein carbonyls; PCs, 132%) in erythrocyte membrane as well as increased values of Hb oxidation (methemoglobin; MetHb, 186%), corpuscular osmotic fragility (15.1%) and hemolysis (free Hb; 195.5%). Parasitemia was positively correlated with MDA (r=0.41, P=0.039), PCs (r=0.45, P=0.023) and MetHb (r=0.40, P=0.042). Also, percent of erythrocytic deformability (echinocytosis) was positively correlated with MDA (r=0.49, P=0.013) and PCs (r=0.63, P<0.001). On the other hand, erythrocytic packed cell volume was negatively correlated with MDA (r=-0.44, P=0.028), PCs (r=-0.72, P<0.001) and MetHb (r=-0.42, P=0.037). In conclusion, T. annulata infection is associated with a parasitic burden-dependant oxidative damage to the erythrocyte membrane protein and lipid contents in addition to Hb. This oxidative damage is linked to the morphological changes of the erythrocyte and may act as mechanisms contribute to pathogenesis of anemia in T. annulata infection in newborn calves.  相似文献

Background

Theileria annulata is a blood parasite affecting ruminants. Hemolytic anemia, secondary hypoxia, and vasculitis are the most important features of tropical theileriosis.

Objectives

Evaluation of electrocardiographic findings and changes in cardiovascular biochemical markers including cTnI concentrations in cattle naturally infected with theileriosis in the absence of acute cardiac failure.

Animals

Ninety adult Holstein cattle (>1 year) with clinical and laboratory evidence of theileriosis and 30 healthy cattle served as controls.

Methods

Case‐control study in which blood samples were collected and randomized after clinical, hematologic, parasitologic examination and laboratory confirmation and electrocardiographic recording on all animals, serum cardiac troponin I (cTnI), aspartate aminotransferase (AST), and creatine kinase‐MB (CK‐MB) were evaluated.

Results

Serum concentration of cTnI was significantly higher in cattle with theileriosis (mean: 0.028 ng/mL; range: 0.005–0.21 ng/mL; control mean: 0.011; range: <0.005–0.09 ng/mL; P = .003). There was significant correlation between serum level of cTnI and PCV (r = ?0.257; P < .001) and also between cTnI and parasitemia (r = 0.515; < .001). Mean serum activities of AST and CK‐MB were 107 ± 46 and 301 ± 103 U/L in sick animals, which were significantly higher than healthy cattle (P = .002 and P = .041, respectively). There were no pathologic arrhythmias detected in sick animals.

Conclusions and Clinical Importance

Theileriosis is a risk factor for elevation of cardiac biomarkers in naturally infected Holstein cattle. Severity of anemia and parasitemia might contribute to the pathophysiology of myocardial damage. The prognostic significance of increased serum cardiac troponin I concentrations in cattle with hemolytic anemia merits further investigation.

To investigate the mechanism of anemia accompanying Japanese bovine theileriosis, we examined whether production of methemoglobin (MetHB), an indicator of erythrocyte oxidation, was associated with anemia in cattle experimentally infected with Theileria sergenti. The percentage of MetHB, which is an oxidized form of hemoglobin, increased according to the onset of anemia. During severe anemia, high levels of acquired methemoglobinemia were observed in all infected cattle. A significant correlation (r=-0.649; P<0.01) between an increase in MetHB concentration and a decrease in packed cell volume (PCV) was observed. It was considered that hemoglobin oxidation may be one of the aggravating factors of anemia in T. sergenti infection.  相似文献

The osmotic fragility and the surface structure of erythrocytes obtained from 3 calves infected with Theileria sergenti and from 3 phlebotomized ones were compared. As the parasitemia progressed, the osmotic fragility of the erythrocytes significantly increased in the infected calves. Particularly the hemolysis ratio in the isotonic area (21.5-94.1%) obviously increased. On the other hand, the percentage of parasitized cells in the erythrocytes did not show so much high values (16.1-21.3%). Similar phenomenon was found in each different percentage of erythrocytes suspension which was separated from density gradient centrifugation. No significant difference in the serum osmotic pressure between the infected calves and the phlebotomized calves was found. By scanning microscopy, the erythrocytes of infected calves, which were collected at the crisis period of parasitemia, were almost completely deformed and showed echinocyte form. Moreover, the appearance ratio of echinocyte form in the erythrocytes population was superior to the percentage of parasitized erythrocytes. Similar membranous alterations were also observed in the erythrocytes of grazing cattle in the crisis period of the theileriosis. It was proven that abnormality of osmotic fragility and morphological disorders of erythrocytes occurred not only in parasitized erythrocytes but also in non-parasitized ones in T. sergenti parasitemia.  相似文献

Theileria sergenti is the causative agent of persistent theileriosis in cattle. The ubiquitous infection of theileiriosis causes chronic anemia and fever in cattle, especially in exogenous cattle. In this study, we applied real-time polymerase chain reaction (PCR) for the diagnosis and quantification of parasite using specific primers for 33 kDa gene fragment of T. sergenti. Comparison of TaqMan PCR with traditional microscopic method, Giemsa's staining, on blood collected from cattle revealed the specificity up to 0.00005% of parasitemia to traditional diagnosis. In addition, it was found that this method can estimate the relative status of infection among herds. The results of present study showed that this method is not only applicable to detect the chronic infection of Theileria, but also effective in evaluation on parasitemia status of cattle, thus it can be used in monitoring the health status in field.  相似文献

The purpose of this study was to determine serum ADA activity in cattle naturally infected with Theileria annulata. In this study, a total of 37 cross-bred cattle which 27 of it showing clinical signs of theileriosis constituted infected group and 10 healthy cattle as control group were used as animal materials. Infected group divided into three groups according to their PCV values. Cattle with PCV > or = 25 were put on group I (n = 9), those with PCV 13-24 were put on group II (n = 11) and those with PCV < or = 12 were put on group III (n = 7). Microscopical diagnosis of the disease was also made. Hematological parameters, serum enzyme activities (ADA, AST, ALT and ALP) were determined in all cattle. Hematological results revealed that significant progressive decreases in HGB, PLT, PBML counts and ratios from group I onwards to group III, whereas the WBC, PBPL counts and ratios showed an increase from group I onwards to group III. The serum ADA, AST, ALT and ALP activity increased significantly in all infected groups compared to control group. However, these parameters were also observed to decrease progressively from group I to group III. Furthermore, the highest increase in enzyme activities observed in the infected group I. But, these enzyme's activities started to decrease in infected group II and III in parallel with PBML and PLT counts. Eventhough, this decrease did not reach to the values obtained from control group. On the contrary, PBPL counts and ratios increased in infected group II and III in contrast to decrease in PCV. As a result, increased serum ADA activity in tropical theileriosis may reflect the involvement of the cellular immune responses.  相似文献

A cross-sectional study based on clinical examination, inspection of herd health records and a questionnaire was designed to determine the epidemiology, economics and potential impact of immunisation against theileriosis in Tanzania. The results showed annual theileriosis costs to be US$ 205.40 per head, whereas the introduction of immunisation reduced this by 40-68% depending on the post immunisation dipping strategy adopted. Morbidity risk due to theileriosis was 0.048 in immunised and 0.235 in non-immunised cattle, and the difference was significant (chi(2)=66.7; P=0.000). The questionnaire results indicated that immunised cattle had a significantly (chi(2)=6; P=0.015) higher risk of anaplasmosis compared with non-immunised cattle, whereas the risk of bovine babesiosis did not differ significantly (chi(2)=0.06; P=0.807) between the two groups. Mortality risk due to anaplasmosis was 0.046 in immunised and 0.018 in non-immunised cattle and this difference was statistically significant (chi(2)=4.48; P=0.043). The theileriosis mortality risk was 0.203 in the non-immunised cattle, while the risk was 0.009 in the immunised cattle and these differences were also significant (chi(2)=103; P=0.000). It was concluded that farmers who have immunised their cattle may cautiously cut down acaricide application by 50% for extensively grazed herds and by 75% for zero grazed animals depending on the level of tick challenge at the herd level.  相似文献

对分离自我甘肃中部地区土种黄牛体的泰勒虫未定种作了鉴定。与环形泰勒虫(T.annulata)、瑟氏泰勒虫(T.sergenti)、小泰勒虫(T.parva)、突变泰勒虫(T.mutans)、斑羚泰勒虫(T.tautotragi)、附膜泰勒虫(T.velifera)、水牛泰勒虫(T.buffeli)作了形态学上的比较研究,观察到该未定种除了泰勒虫所共有形态外,还有其它泰勒虫所没有的且难以描述的特异形态,尤其对染虫率高峰期,特异形态占虫体总数的20%左右。该种还具有出芽增殖的特性,可在除脾牛体内大量繁殖,染虫率可达52.69%的高峰,试验感染牛出现高烧、极度贫血、精神沉郁、食欲不振等临床症状,导致个别牛只死亡,剖检观察到某些脏器有严重病变,表明具有一定的致病力。媒介蜱尚不清楚,但已证实,在我国传播家畜泰勒虫和巴贝斯虫的7种媒介蜱对该种无传播能力。传统分类学研究结果表明,该种不同于已知牛泰勒虫有效种,系一新种。由于这一新种首次在中国分离到,因而被命名为中华泰勒虫(Theileria sinensis sp.nov.)(梨形虫亚目:泰勒虫科)。  相似文献

河南省桐柏县是牛环形泰勒虫病流行的多发区,对养牛业危害十分严重,1985年该县从宁夏农林科学院畜牧兽医研究所引进“虫苗”2万头份,在区域试验的基础上在全县16个乡(镇)进行了大面积防疫注射,经年终统计表明:虫苗安全性100%,注苗15000头(其中有水牛45000头),发病3头,发病率0.02%;未注苗36000头,发病2060头,发病率5.7%,实际有效保护率达99.98%,取得了明显的社会经济效益。  相似文献

Insulin-dependent diabetes mellitus (IDDM) is a common metabolic disease often complicated by a number of pathological conditions among which are haematological changes and alterations in blood cell function. Human and feline diabetes mellitus patients have been reported to be associated with oxidative stress that can lead to membrane alterations and to reduced erythrocyte life-span. Erythrocyte function in dogs affected by IDDM has been investigated during insulin therapy, paying attention to antioxidant status, membrane resistance, enzyme activities and 2,3-diphosphoglycerate (2,3DPG) concentration. Thirteen diabetic and 36 healthy dogs were bled and haematology and blood chemistry assays were performed to evaluate the degree of compensation. Osmotic fragility, the activities of the enzymes glucose-6-phosphate dehydrogenase (G6PD) and pyruvate-kinase (PK) and the concentrations of reduced glutathione (GSH) and 2,3DPG were evaluated in the erythrocytes. Diabetic dogs did not differ from controls in terms of haematological parameters, except for higher numbers of platelets. Higher values of fructosamine, glucose, protein, plasma potassium and calculated osmolality were detected in the plasma from diabetic dogs. No differences were detected in osmotic fragility, GSH concentration and PK activity between the two groups but 2,3DPG concentration and G6PD activity were statistically significantly higher in the diabetic group. The results indicate minimal alterations in erythrocyte functions occur in insulin-treated diabetic dogs. This contrasts with what has been reported for IDDM humans and cats.  相似文献

This study was designed to assess the effects of a moderate increase in dietary sulphur (S) in cattle. Twelve animals were initially fed a basal concentrate (S = 0.2%) and then divided into two groups; one fed basal and the other high S (S = 0.75%) concentrates. Health, body weight gains, and activities of erythrocyte enzymes-glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), glucose-6-phosphate dehydrogenase (G6PD), acetylcholinesterase (AChE), plasma- asparate aminotransferase (AST), and whole blood concentrations of selenium (Se) were monitored at various stages of the study. Marked increases in the activities of GSH-Px, SOD and G6PD from the pretrial values were observed upon initial feeding of basal concentrate diet. Sex related differences were not evident in enzyme activities and Se concentrations of the blood. A high linear correlation (r = 0.92) between averages of GSH-Px activity and Se concentration of blood was observed in both sexes. Increasing the amount of S in the concentrate diet (from 0.2 to 0.75%) did not produce any statistically significant change in enzyme activities and Se concentrations, body weight gains, and health of the cattle during the 85 days feeding period. The results indicate that a moderate increase in the dietary S would not impair Se and copper status or cause related disorders in cattle.  相似文献

To study the mechanisms by which certain West African taurine cattle are able to resist trypanosomiasis, the disease course and several immune parameters were examined in eleven Baoulé and five Zebu cattle after infection with tsetse-transmitted T. congolense (clone 1180 of stock Serengeti/71/STIB/212). All animals showed a similar onset of parasitemia but subsequently a continuum of disease was observed ranging from four Baoulé which were mildly susceptible (controlled parasitemia by week 10 post-infection and had little anemia) to two Baoulé and two Zebu which were highly susceptible (unable to control parasitemia, severe anemia leading to death or drug treatment in extremis). The remaining five Baoulé and three Zebu showed intermediate susceptibility. Although the most resistant animals were of the Baoulé breed, there was a spectrum of susceptibility which crossed the two breeds. Neutralizing antibody to metacyclic trypanosome antigens was detectable by day 18 in four of the mildly susceptible and three of the highly susceptible individuals but such antibodies were delayed in the remaining one severely susceptible animal. Neutralizing antibodies to antigenic variants of the first peak of blood trypanosomes were of significantly higher titre and appeared earlier in the four mildly susceptible as opposed to the highly susceptible animals. No differences in lymphocyte in vitro mitogen responses were observed in these animals except shortly before death in those severely affected. In vitro lymphocyte responses to allogeneic cells did appear to correlate with disease severity, in that animals with mild disease showed little immunosuppression of this response whilst in severely affected individuals the response was almost ablated.  相似文献

The biochemical parameters were studied in adult and young Friesian cattle naturally infected with Theileria annulata in the Qassim Region, Saudi Arabia. Forty-three clinical cases of tropical theileriosis were studied, together with 40 clinically healthy Friesian cattle. Cattle clinically infected with T. annulata had significantly lower serum total protein, albumin, globulin, creatinine, calcium, phosphorus, magnesium, potassium, iron and copper concentrations and significantly higher AST activity and bilirubin concentration than the healthy cattle.  相似文献

Oxidative stress parameters and erythrocyte characteristics were studied in 15 normal healthy dogs and 33 renal azotaemic dogs from Small Animal Hospital, Faculty of Veterinary Science, Chulalongkorn University. Dogs with renal azotaemia had reduced mean corpuscular volume (MCV) (P < 0.01), packed cell volume (PCV) (P < 0.001) and increased mean corpuscular hemoglobin concentration (MCHC) (P < 0.001). The relationship was found between degree of azotaemia and MCV, PCV and MCHC. Dogs with severe renal azotaemia had higher intraerythrocytic sodium contents (RBC-Na) (P < 0.05). The red blood cell catalase activity and glutathione and plasma malondialdehyde were unaltered while urinary malondialdehyde-creatinine ratio (U-MDA/Cr) increased significantly (P < 0.001). The U-MDA/Cr was correlated significantly with plasma creatinine concentration (P < 0.05), urinary protein-creatinine ratio (P < 0.05) and fractional excretion of sodium (P < 0.001). The results suggest some changes in RBC characteristics and urine oxidative stress marker in renal azotaemic dogs. Moreover, the U-MDA/Cr is a sensitive biochemical parameter which increased along with degree of renal dysfunction.  相似文献

The aim of this study was to evaluate lipid peroxidation, protein oxidation and activity of enzymes that are indicators of oxidative stress in Rangelia vitalii infection in dogs. Animals were divided into two groups: negative control (n=5) and infected with R. vitalii (n=7). After inoculation, the parasitemia was estimated daily by microscopic examination of smears. Lipid peroxidation (TBARS) and advanced oxidation protein products (AOPP); and delta-aminolevulinate dehydratase (δ-ALA-D), superoxide dismutase (SOD) and catalase (CAT) activities in blood were evaluated. The samples were collected at days 10 and 20 post-inoculation (PI). TBARS and AOPP levels were higher in the infected group in both analyzed periods (P<0.01). The δ-ALA-D activity was reduced in blood of dogs infected with R. vitalii on days 10 and 20 PI. SOD activity was significantly increased (P<0.01) in the blood of dogs infected with R. vitalii at days 10 and 20 PI, while CAT activity was significantly increased (P<0.01) only at day 20 PI when compared to non-infected animals. A positive correlation was observed between the degree of parasitemia and TBARS and AOPP levels and activity of antioxidant enzymes. The δ-ALA-D activity was negatively correlated with the degree of parasitemia. Based on the increased levels of TBARS, AOPP, SOD and CAT activities, and inhibition δ-ALA-D activity, we concluded that dogs experimentally infected with R. vitalii develop a state of redox unbalance and that these changes might be involved in the pathophysiology of disease.  相似文献

为了评价施肥处理对牧草矿物质元素含量和牦牛抗氧化酶活性的影响,在黄河首曲草地进行草地施肥和草地放牧试验。结果表明,硫酸氨施肥组超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH—Px)、过氧化氢酶(CAT)极显著低于(P〈0.01)Ⅱ、Ⅲ组,Ⅱ、Ⅲ组之间无显著差异;铜蓝蛋白(Cp)的含量,硫酸氨施肥组极显著低于Ⅱ、Ⅲ组(P〈0.01),Ⅱ、Ⅲ组之间无显著差异;血液MDA的含量,硫酸氨施肥组极显著高于Ⅱ、Ⅲ组(P〈0.05),Ⅱ、Ⅲ组之间无显著差异。在试验结束时硫酸氨施肥组Hb、MCV、MCH都极显著低于Ⅱ、Ⅲ组(P〈0.01),放牧牦牛出现小红细胞贫血。  相似文献

The present study aimed to assess antioxidant status and oxidative stress in sheep naturally infected with Babesia ovis. Red blood cell (RBC) count, hemoglobin (Hb) concentration, packed cell volume (PCV), piroplasm parasitemia percentage, malondialdehyde (MDA) concentration, erythrocyte superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) activities and total antioxidant capacity (TAC) were determined in 52 sheep naturally infected with B. ovis as well as same number of healthy sheep in West-Azerbaijan province, Iran. Microscopic examination of Giemsa-stained peripheral blood smears revealed B. ovis infection. The parasitological diagnosis was confirmed using polymerase chain reaction (PCR) analysis by amplifying a partial small subunit ribosomal RNA (ssu rRNA) gene sequence of B. ovis of 52 diseased sheep, 18 (34.61%), 11 (21.15%), 16 (30.76%) and 7 (13.48%) had <1%, 1-2%, 2-3% and >3% parasitemia, respectively. Compared to controls, the activities of erythrocyte GSH-Px, SOD, TAC and CAT showed a significant decrease, whereas the concentration of MDA in erythrocytes of infected sheep increased significantly. Parasitemia rate was positively correlated with MDA and negatively correlated with PCV, SOD, CAT, GSH-Px and TAC. Also, MDA was negatively correlated with PCV, SOD, catalase, GSH-Px and TAC. The study demonstrated that B. ovis plays an important role in the occurrence of oxidative damage to RBCs and anemia in ovine babesiosis.  相似文献
