Ovine enzootic abortion: experimental studies of immune responses   总被引:3,自引:0,他引:3  
Eight pregnant and four non-pregnant ewes were exposed to experimental oral infection with the chlamydial agent of ovine enzootic abortion. Four non-pregnant ewes received a sham inoculum. Immune responses were investigated using a complement fixation test (CFT), a delayed-type hypersensitivity (DTH) skin test and a lymphocyte transformation assay (LTA). When these responses were correlated with clinical observations it was concluded that fetopathy occurred in those ewes which failed to mount or mounted only weak DTH responses. High antibody titres were not an indication of protective immunity; in pregnant animals there was evidence of an inverse correlation between DTH and humoral responses as detected by the CFT. In the LTAs, chlamydia specific blastogenesis was recorded in all infected sheep but there was little correlation with clinical findings.  相似文献

Ewes at 70 days of gestation were exposed by various routes to a culture of Chlamydia psittaci. Chlamydial infection of fetal tissues, generally accompanied by abortion, was observed only in four of six ewes inoculated via the tonsillar crypts and in five of seven ewes injected subcutaneously; most of these also developed antibodies to C psittaci. Seroconversion after normal lambing was also observed in most ewes inoculated orally or by stomach tube, despite failure to detect chlamydia in them.  相似文献

Invasive and non-invasive strains of Chlamydia psittaci isolated from faeces of clinically healthy ewes and from vaginal swabs of ewes which had aborted were injected intravenously or intradermally into pregnant ewes. The results were studied by recording the ewes' thermal and serological responses, lambing performance and the excretion of chlamydia from the vagina. The differences between the effects of different invasive strains were greater after intradermal inoculation than after intravenous inoculation. After intradermal inoculation non-invasive strains did not disturb pregnancy (11 of 13 ewes lambed normally) whereas invasive strains induced abortion in 23 of 25 ewes, 24 of which excreted chlamydia in vaginal secretions.  相似文献

The protection afforded by an experimental, killed, adjuvanted vaccine derived from the A22 strain of Chlamydia psittaci (ovis) against ovine enzootic abortion was studied. The vaccine was used undiluted (group A), at a dilution of 10(-3) (group B) and at a dilution of 10(-6) (group C). A fourth control group (group D) was inoculated with all components of the vaccine except the chlamydial antigen. A group of rams (group R) was also vaccinated with the chlamydial antigen diluted to 10(-3). Animals were challenged 70 days after mating with the A22 strain of C. psittaci (ovis) and were studied throughout pregnancy and the subsequent lambing period. Their cell-mediated immune responses were examined using a skin test and their humoral immune responses were studied using an ELISA. Tests for excretion of chlamydiae in their faeces and genital tract during pregnancy and after parturition and in the faeces of their lambs were made. The reproductive performance of the ewes was assessed by calculating the average weight of lambs produced per ewe in each group. The experimental vaccine protected the ewes in groups A and B against challenge with C. psittaci (ovis) as none showed clinical signs of OEA or excreted chlamydiae. The average weight of lambs produced per ewe in both groups was greater than 4 kg. Both groups seroconverted after vaccination but not all of them were positive to the skin test. The experimental vaccine at 10(-6) dilution of antigen did not protect the ewes as three of 10 ewes displayed clinical OEA and excreted chlamydiae in the products of parturition.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Nineteen ewes were injected subcutaneously with the agent of enzootic ovine abortion, Chlamydia psittaci serovar 1, at 50 days gestation. Placental and fetal tissues were examined at 15 days postinfection and thereafter at ten day intervals. Placental infection was detected at 15 days postinfection. Only postinoculation sera collected from postinfected ewes contained antibodies reactive to C. psittaci. Five (26%) chlamydial infected ewes experienced inapparent fetal loss before day 105 of gestation. This finding is significant since C. psittaci infection in sheep is commonly associated with abortion and not infertility.  相似文献

The antibody response of 20 pregnant ewes to oocyst infection with Toxoplasma gondii was determined by the latex agglutination test (LAT) and compared with the indirect fluorescent antibody test (IFAT) and a commercially available indirect haemagglutination test (IHAT). The LAT and IFAT showed a similar rapid response with antibody first appearing by two to three weeks after infection and titres that correlated closely (r = 0.81, P less than 0.001). The IHAT response was slower and less consistent up to seven weeks after infection. The LAT response was biphasic in seven of the sheep. Sera were fractionated using a minicolumn gel filtration technique and specific IgM and IgG titres determined by LAT. IgM titres peaked three weeks after infection and IgG titres exceeded IgM titres at a mean time of 4.7 weeks after infection (range 3 to 7). Eleven sheep exhibited fetopathy with abortion/parturition 12 to 53 days after infection; in nine of them IgG titres exceeded IgM at that time. A non-specific anti-toxoplasma reaction associated with IgM antibody occurred at low titre in one sheep. The results indicate that used from a dilution of 1/64 the LAT is a sensitive, reliable and rapidly responsive serological test for toxoplasma infection in ewes and it may be utilised with sample fractionation techniques to determine IgM titres. It is suggested that the best time to examine ewe sera to assist diagnosis of toxoplasma abortion is one week after abortion. While the determination of specific IgM titres in ewe sera may assist epidemiological studies and, sometimes, diagnosis, in the majority of aborting ewe sera it is unlikely to aid diagnosis.  相似文献

A commercially available enzyme immunoassay (EIA) for the detection of Chlamydia trachomatis in human urogenital and conjunctival specimens was compared with isolation in cell culture for the detection of Chlamydia psittaci in vaginal and placental swabs from aborting ewes and swabs of aborted fetal tissues. The EIA on vaginal swabs collected from 10 ewes experimentally infected with C. psittaci had a sensitivity of 85.7% and a specificity of 85.7%. Vaginal swabs collected at the time of abortion or within 3 days were the best samples for detection of chlamydial infection. The 29 vaginal swabs collected during this period from experimentally infected ewes were all strongly EIA-positive, and chlamydia were isolated from 28. The EIA on vaginal swabs from 78 field cases of abortion had a sensitivity of 78.0% and a specificity of 76.8%. The EIA on swabs of cotyledons from 65 placentas had a sensitivity of 100% and a specificity of 75.0% compared with isolation in cell culture. The EIA on 57 swabs of fetal tissues or body fluids from 10 aborted fetuses or weak lambs from experimentally infected ewes had a sensitivity of 26.6% and a specificity of 88.1% compared with isolation in cell culture. 466 sheep sera out of 19 flocks in Switzerland were examined by a competitive enzyme linked immunosorbent assay (cELISA) for antibodies against Chlamydia psittaci "serotype 1" ("ovine enzootic abortion"). Since numerous positive reactors were found in flocks without abortion history, 30 fecal samples out of two of these flocks were examined by PCR for evidence of chlamydial DNA. One of these samples turned out to contain DNA of Chlamydia psittaci "serotype 1". These results suggest, that in Switzerland "serotype 1" of Chlamydia psittaci is widespread not only as cause of chlamydial abortion but also as latent intestinal infection in sheep. The resulting difficulties for serological diagnosis of chlamydial abortion and possible solutions based on the cELISA are discussed. The complement fixation test (CFT), still considered as standard method for serological examination for Chlamydiae, has additionally been applied.  相似文献

The involvement of prolactin in the periparturient rise in the faecal nematode egg count in sheep was investigated. Ostertagia circumcincta larvae (5000 third stage larvae three times weekly) were administered to adult immune ewes from three weeks before parturition to three weeks afterwards. Ten ewes were injected twice daily with 2-bromo-alpha-ergocryptine (bromocriptine), an antagonist of prolactin secretion, for two weeks starting two days after lambing while 10 ewes remained untreated. Bromocriptine treatment was initiated approximately two weeks pre partum in three other ewes. Plasma pepsinogen concentrations rose significantly by one week after the start of O circumcincta larval challenge in all the ewes but faecal egg counts remained negative until approximately one week post partum. Plasma prolactin concentration was reduced to a very low level in all bromocriptine treated ewes but this did not alter the dynamics of the periparturient rise in faecal egg counts. Neither cell-mediated nor humoral immunity of the ewes, as assessed by their sensitivity to BCG inoculation and by antibody titre raised against horse red blood cells, respectively, were impaired during the rise in faecal egg count, nor were these parameters altered by manipulation of plasma prolactin concentration. Lamb growth rate was not retarded by low plasma prolactin concentration in the bromocriptine treated ewes. These results are not consistent with the generally held hypothesis that elevated plasma prolactin concentration is directly associated with the periparturient rise.  相似文献

Ostertagia circumcincta larvae were administered to adult immune ewes either during the breeding period or around parturition to investigate whether reproductive performance was affected. Two groups of 25 ewes were given water or 12,000 ostertagia larvae three times weekly for five weeks before and five weeks after breeding to synchronised oestrus. Plasma pepsinogen levels were raised in the larval-challenged ewes during the treatment period but no significant effect of treatment was observed on ovulation rate or plasma progesterone concentration. Neither ewe live-weight nor body condition score differed between groups throughout gestation or for four weeks post partum and there was no effect from larval challenge during the breeding period on lambing rate, lamb birthweight or liveweight at four weeks old. A second experiment involved 20 twin-bearing immune ewes which were administered 17,000 ostertagia larvae three times weekly for three weeks before and three weeks after parturition. Plasma pepsinogen concentration was elevated during the larval challenge period and nematode eggs were detected in the faeces after parturition but there was no effect from treatment either on ewe liveweight and body condition score or on lamb birthweight and lamb liveweight at four weeks old. These results indicate that larval challenge with ostertagia larvae at dose rates comparable to larval intake from heavily contaminated pasture did not affect the general well being and reproductive performance of immune Greyface ewes.  相似文献

This study evaluated the carry‐over effects of ewe body reserves during early pregnancy on peri‐partum adipose tissue metabolism. Forty‐nine multiparous ewes were divided in three categories according to their body condition score (BCS) at day 30 of pregnancy (BCS < 3, 2.5–2.75; BCS = 3; BCS > 3, 3.25–3.5). Live‐weight (LW) and BCS gains from 1st to 4th month of pregnancy were greater in ewes with BCS < 3 and 3 than in >3 animals. In contrast, in the last month of pregnancy, there was BCS decrease in all groups, although LW continued increasing. There were no differences in LW or BCS across ewe categories during this period. Peripheral leptin levels throughout the three last weeks of pregnancy were greater in ewes with BCS > 3 than in the rest, but this difference did not persist after lambing. Plasma metabolites related to energy metabolism, milk yield and lamb growth were not affected by ewe BCS in early pregnancy. Long‐chain saturated milk fatty acids (FA) (C16–C24) were greater in ewes with lowest BCS (<3 and 3). Ewes with greater BCS showed greater monounsaturated and lowest polyunsaturated milk FA content. Ewe post‐mating body reserves affect both pre‐partum leptinaemia and post‐partum milk polyunsaturated fatty acids content, but it had little effect on lamb performance.  相似文献

The hypocalcaemic response to protamine, as a measure of bone resorption rate, was studied in cattle and sheep. Three groups of calves were studied (1–3 days, 2–3 weeks and 4 months old). The oldest calves showed the greatest response, indicating a more rapid skeletal turnover in these animals, probably related to a higher growing rate. A group of cows was tested at 2 occasions, near parturition and 2–3 weeks after parturition. The decrease in plasma calcium was small near parturition, while a profound drop was obtained 2–3 weeks post partum, which supports earlier findings that bone resorption is more or less blocked in parturient cows. Young pregnant ewes showed the same degree of hypocalcaemia as adult pregnant ewes but the recovery was more rapid in the young animals. Young rams were given protamine before and after diethylstilboestrol treatment. No differences in calcium response were obtained. Varying plasma phosphate responses were obtained. A pronounced hyperphosphataemia was found in the adult pregnant ewes. Diethylstilboestrol caused hypermagnesaemia in the rams.  相似文献

Inocula containing 75, 250 or 1000 Toxoplasma gondii tissue cysts were used to infect seronegative gimmers and seropositive ewes in the fourth month of pregnancy. The seronegative gimmers developed typical toxoplasma infections at all dose levels. Four of them aborted and the surviving lambs showed rising indirect haemagglutination titres in the first two to three months of life indicating congenital infection. The seropositive ewes showed no response to challenge, all their lambs survived and there was no serological evidence of congenital infection. Indirect haemagglutination titres in the seropositive ewes remained unchanged throughout the experiment, titres in the gimmers rose sharply from the 10th day after infection and by three months were the same as those in the ewes.  相似文献

The IDEIA ELISA was used to detect Chlamydia psittaci (ovis) antigen in ewes' milk to which were added serial dilutions of chlamydiae titrated as inclusion forming units (ifus) in McCoy cell tissue culture. The test was able to detect as few as 35 ifus/ml of the organism. The ELISA was then used to detect chlamydial antigen in fetal membranes and milk from ewes clinically affected with ovine enzootic abortion (OEA). The results were compared with results of isolation of chlamydiae in McCoy cell tissue culture from the same material. The fetal membranes of 17 of 19 ewes were positive for chlamydia when tested with the ELISA but chlamydia could be cultured from only 15 of them. Milk samples from 26 ewes which had aborted between 1 and 34 days previously were tested: chlamydiae could not be cultured from any of them and only one was positive when tested by the ELISA. The results show that the IDEIA ELISA is a sensitive test for the detection of C. psittaci (ovis) antigens. The positive results to this test for the three samples from which chlamydiae could not be cultured suggest that the test is not as specific as culture or that it detected dead organisms. Chlamydiae do not appear to be excreted in the milk of ewes affected with OEA.  相似文献

Live attenuated vaccines against Chlamydia psittaci var ovis, Brucella melitensis and Salmonella abortus ovis have previously been shown to be compatible in mice by subcutaneous administration. Immunity against challenge with virulent chlamydia was, however, slightly decreased in associations including the B melitensis Rev 1 vaccine. The chlamydia strain 1B vaccine was administered to four- to five-month-old female lambs, either alone or in combination with the B melitensis Rev1 and the S abortus ovis Rv6 vaccines. Clinical, serological and bacteriological observations demonstrated the compatibility of the three vaccines. Control, singly and triply vaccinated ewes were challenged with a virulent strain of chlamydia during their second pregnancy, 15 months after vaccination. Five of the 12 control ewes lambed normally and 10 of them were infected, as shown by the excretion of the challenge chlamydia in genital secretions. Sixteen of the 17 ewes in the triple vaccine group lambed normally and none was infected. All 12 in the single vaccine group lambed normally and three of the 12 were infected. In spite of this unusually poor protection by the single vaccine, antichlamydial immunity was clearly not decreased by the association with the two other vaccines.  相似文献

Faeces samples were taken per rectum from sheep on 26 farms in England and Wales and examined for thepresence of chlamydia by culture McCoy cell monolayers. Thirteen of the fars were known to have had abortion outbreaks associated with Chlamydia psittaci (enzootic abortion) and 13 were free of this infection. The chlamydia isolated were characterized by cultural techniques. Chlamydia were isolated from the faeces of lambs on all 26 farms and the prevalence of infection varied from 5–50% on individual farms. There was no significant difference between the proportion of infected lambs on farms where enzootic abortion was present or absent. Lambs first showed infection when they were 3 months old and the prevalence rate of infection increased up to 9 months old. No chlamydia were isolated from the faeces of 316 adult ewes. The chlamydia were identified as enteric rather than abortion type and thus were C. pecorum rather than C. psittaci.  相似文献

Fifty ewes were randomly divided into four groups. Groups A and B were vaccinated with an experimental vaccine derived from the A22 isolate of Chlamydia psittaci (ovis), an isolate known to cause ovine enzootic abortion (OEA). Groups C and D were unvaccinated controls. In mid-pregnancy, animals in group A and C were challenged with live A22 C. psittaci (ovis) and those in B and D were challenged with a field isolate of the organism (BS) against which the commercially available A22 vaccine appeared to offer poor protection. In group A, three animals showed clinical signs of OEA and six excreted chlamydiae. In group B, five ewes showed clinical signs of OEA and excreted chlamydiae. In group C, three ewes had clinical signs of OEA but seven excreted chlamydiae. In group D, all 11 ewes showed clinical signs of OEA and excreted chlamydiae in the products of parturition. This group produced only four live lambs with an average weight of viable lamb per ewe of 1.4 kg, whereas the other groups each produced 12 or 13 lambs with an average weight of viable lamb per ewe of more than 4 kg. The BS isolate was much more virulent than the A22 isolate for unvaccinated, pregnant ewes. However, the A22 vaccine offered significant protection against the heterologous BS isolate although on this occasion it did not appear to alter the course of disease produced by the less pathogenic A22 isolate.  相似文献

A Chlamydophila abortus-induced abortion model was carried out on the basis of the experimental infection of ewes at day 75 of gestation. The infection induced abortions and the birth of weak lambs during the last 3 weeks of pregnancy. To study the kinetics of the infection in the placenta and in other organs, infected ewes were killed at 105, 120, and 130 days of gestation and also several days after abortion or parturition. Infected ewes developed a systemic infection that caused a mild and transient pneumonia and focal hepatitis. Pathologic changes were observed in placentas at 120 day of gestation, although the lesions varied between animals and even between placentomes of the same placenta. The first placental area infected was the maternal stroma and epithelium next to the intercaruncular areas, where neutrophilic response seemed to control the infection. A substantial degree of multiplication of C. abortus was then observed in the trophoblast cells of the placentome, periplacentomal choriallantoic membranes, and hilius, with an inflammatory exudate composed mainly of neutrophils, some macrophages, and very scarce lymphocytes. After abortion, the lesions affected the intercotyledonary areas of the aborted placentas, whereas in the uterus significant lymphocyte infiltration was observed, together with a rapid decrease of the C. abortus antigen in the degenerated caruncular tissues.  相似文献

Cloprostenol, 100 micrograms, given intramuscularly to the nanny, with 50 micrograms 10 hours later, precipitated parturition in goats after 36 +/- 1 hours (mean +/- SEM), when administered at 137 +/- 0.5 days gestation. All kids were born alive and survived to weaning. Milk yield over 40 weeks post partum was not significantly different from that after spontaneous parturition. Three hundred micrograms cloprostenol (200 micrograms with 100 micrograms 10 hours later) also initiated parturition at 137 +/- 0.5 days gestation but caused a significant (P less than 0.01) suppression of lactation. Cloprostenol-induced parturition in more than one pregnancy had no adverse effects except for an increased incidence of placental retention, which was treated successfully with intrauterine pessaries containing oestrogen. During the first eight days after spontaneous parturition efficiency of milk secretion was inversely related to udder mass, suggesting a gradual maturation of the secretory alveolar epithelium over this time. When parturition was induced by cloprostenol there was a four to eight day delay before the establishment of this relationship which appeared essential for a successful lactation. Cloprostenol proved to be a useful tool for the control of parturition in goats, having applications to both general animal husbandry and for the study of mammary development and secretory competence.  相似文献

The objective of this study was to record how the antibody levels change over time during pregnancy in dairy cows naturally infected with the protozoan parasite Neospora caninum, and relate this to the reproductive performance. Eighteen cows with antibodies to N. caninum were serum sampled monthly during their first pregnancy and 13 of them were also followed for a second pregnancy. In all, five pregnancies ended in abortion and two in stillbirth. Antibodies to N. caninum in serum were demonstrated by immune stimulating complex enzyme-linked immunosorbent assay (iscom ELISA). The N. caninum antibody titres remained well above the 1:100 cut-off limit for the test used during 2 years in all cows. In the non-aborting cows, mean values of antibody titres to N. caninum rose 1.5-2.5 dilution steps to reach a plateau 4-5 months before parturition, and thereafter decreased from 2 months before parturition. These changes were statistically significant (p < 0.001). The same pattern was seen in the aborting cows. The consistent pattern of rise in antibody titres observed during both pregnancies in all cows indicated a reactivation rather than a reinfection of the parasite at mid-gestation.  相似文献
