A measurement and interpretation on a molecular level of a phase transition in an ordered Langmuir monolayer is reported. The diagram of surface pressure (pi) versus molecular area of a monolayer of chiral (S)-[CF(3)-(CF(2))(9)-(CH(2))(2)-OCO-CH(2)-CH (NH(3)(+))CO(2)(-)] over water shows a change in slope at about pi(s)= 25 millinewtons per meter. Grazing-incidence x-ray diffraction and specular reflectivity measurements indicate a solid-solid phase transition at pi(s). The diffraction pattren at low pressures reveals two diffraction peaks of equal intensities, with lattice spacings d of 5.11 and 5.00 angstroms; these coalesce for pi >/=pi(s). Structural models that fit the diffraction data show that at pi> pi(s) the molecules pack in a two-dimensional crystal with the molecules aligned vertically. At pi < pi(s) there is a molecular tilt of 16 degrees +/- 7 degrees . Independent x-ray reflectivity data yield a tilt of 26 degrees +/- 7 degrees . Concomitant with the tilt, the diffraction data indicate a transition from a hexagonal to a distorted-hexagonal lattice. The hexagonal arrangement is favored because the -(CF(2))(9)CF(3) moiety adopts a helical conformation. Compression to 70 millinewtons per meter yields a unit cell with increased crystallinity and a coherence length exceeding 1000 angstroms.  相似文献

Graphene produced by exfoliation has not been able to provide an ideal graphene with performance comparable to that predicted by theory, and structural and/or electronic defects have been proposed as one cause of reduced performance. We report the observation of domains on exfoliated monolayer graphene that differ by their friction characteristics, as measured by friction force microscopy. Angle-dependent scanning revealed friction anisotropy with a periodicity of 180° on each friction domain. The friction anisotropy decreased as the applied load increased. We propose that the domains arise from ripple distortions that give rise to anisotropic friction in each domain as a result of the anisotropic puckering of the graphene.  相似文献

The L-P ring complex is thought to be a molecular bushing that supports flagellar motor rotation at about 10,000 revolutions per minute with presumably very little friction. Structural studies of this complex have been limited because only very small amount of samples are available. Therefore devising an efficient method of crystallization was essential. The addition of a phospholipid and its subsequent slow depletion by phospholipase A2 have been used to successfully grow well-ordered monolayer crystals that extend up to about 10 micrometers. The interaction of the L-P ring complex with lipid membranes was also visualized during this process.  相似文献

Optical second harmonic generation was used to study the in-plane alignment of self-assembled silane monolayers attached to a glass surface under mechanical loading. The measurements allow correlation of the macroscopic forces acting on the monolayer with the average orientation and the azimuthal molecular alignment of the terminal molecular entity. Compression and shear forces lead to an alignment of the initially randomly oriented molecules on a macroscopic length scale. The change in azimuthal alignment of molecules under mechanical stress was found to be irreversible on the time scale of 12 hours, whereas changes of the molecular tilt angle were reversible.  相似文献

The surface membrane of cells is studded with morphologically distinct regions, or domains, like microvilli, cell-cell junctions, and coated pits. Each of these domains is specialized for a particular function, such as nutrient absorption, cell-cell communication, and endocytosis. Lipid domains, which include caveolae and rafts, are one of the least understood membrane domains. These domains are high in cholesterol and sphingolipids, have a light buoyant density, and function in both endocytosis and cell signaling. A major mystery, however, is how resident molecules are targeted to lipid domains. Here, we propose that the molecular address for proteins targeted to lipid domains is a lipid shell.  相似文献

A smectic liquid-crystal phase made from achiral molecules with bent cores was found to have fluid layers that exhibit two spontaneous symmetry-breaking instabilities: polar molecular orientational ordering about the layer normal and molecular tilt. These instabilities combine to form a chiral layer structure with a handedness that depends on the sign of the tilt. The bulk states are either antiferroelectric-racemic, with the layer polar direction and handedness alternating in sign from layer to layer, or antiferroelectric-chiral, which is of uniform layer handedness. Both states exhibit an electric field-induced transition from antiferroelectric to ferroelectric.  相似文献

Simulations and perturbation theory are used to study the molecular origins of friction in an ideal model system, a layer of adsorbed molecules sliding over a substrate. These calculations reproduce several surprising features of experimental results. In most cases, the frictional force on a solid monolayer has a different form from that observed between macroscopic solids. No threshold force or static friction is needed to initiate sliding; instead, the velocity is proportional to the force. As in experiments, incommensurate solid layers actually slide more readily than fluid layers. A comparison of experiment, simulation, and analytic results shows that dissipation arises from anharmonic coupling between phonon modes and substrate-induced deformations in the adsorbate.  相似文献

Lateral variations in membrane composition are postulated to play a central role in many cellular events, but it has been difficult to probe membrane composition and organization on length scales of tens to hundreds of nanometers. We present a high-resolution imaging secondary ion mass spectrometry technique to reveal the lipid distribution within a phase-separated membrane with a lateral resolution of approximately 100 nanometers. Quantitative information about the chemical composition within small lipid domains was obtained with the use of isotopic labels to identify each molecular species. Ratios of the lipid monolayer area to the erythrocyte surface area are 2:1 at low surface pressures and approach 1: 1 at collapse pressures. Un saturated phospholipids in cholesterol-phospholipid complexes of membrane ex tracts resemble their saturated derivatives at collapse pressures. Domain I of the activated Crystal protein from Bacillus thuringiensis has a seven α-helix bundle structure, which is responsible for membrane channel formation in its insecticidal mechanism. Cry1Ie is toxic to Asian corn borer, Ostrinia furnacalis (Guenée), and plays important roles in insect biological control. The domain I from Cry1Ie has been expressed and purified in its normal conformation, as embedded in the full length homologous toxin structure. The membrane insertion ability of this single domain was compared with the full length homologous toxin using a monolayer insertion experiment. The results indicated that the Cry1Ie-domain I had the ability to insert into the lipid monolayer, and this ability is greater than that of the IE648 toxin. However, the state of insertion is not stable and remains for only a short period of time. The Cry1Ie-domain I plays no role in receptor binding as it had a nonspecific binding with the brush border membrane vesicles of the Asian corn borer.  相似文献

The self-assembly mechanism of alkanethiol monolayers on the (111) surface of gold was discovered with the use of an ultrahigh-vacuum scanning tunneling microscope. Monolayer formation follows a two-step process that begins with condensation of low-density crystalline islands, characterized by surface-aligned molecular axes, from a lower density lattice-gas phase. At saturation coverage of this phase, the monolayer undergoes a phase transition to a denser phase by realignment of the molecular axes with the surface normal. These studies reveal the important role of molecule-substrate and molecule-molecule interactions in the self-assembly of these technologically important material systems.  相似文献

Adaptor protein 180 (AP180) and its homolog, clathrin assembly lymphoid myeloid leukemia protein (CALM), are closely related proteins that play important roles in clathrin-mediated endocytosis. Here, we present the structure of the NH2-terminal domain of CALM bound to phosphatidylinositol-4,5- bisphosphate [PtdIns(4,5)P2] via a lysine-rich motif. This motif is found in other proteins predicted to have domains of similar structure (for example, Huntingtin interacting protein 1). The structure is in part similar to the epsin NH2-terminal (ENTH) domain, but epsin lacks the PtdIns(4,5)P2-binding site. Because AP180 could bind to PtdIns(4,5)P2 and clathrin simultaneously, it may serve to tether clathrin to the membrane. This was shown by using purified components and a budding assay on preformed lipid monolayers. In the presence of AP180, clathrin lattices formed on the monolayer. When AP2 was also present, coated pits were formed.  相似文献

The ion distribution in an electrolyte solution in contact with a charged polymerized phospholipid membrane was directly measured with long-period x-ray standing waves. The 27-angstrom-thick lipid monolayer was supported on a tungsten/silicon mirror. X-ray standing waves were generated above the mirror surface by total external reflection of a 9.8-kiloelectron volt x-ray beam from a synchrotron undulator. The membrane surface, which contained negatively charged phosphate headgroups, was bathed in a dilute ZnCl2 solution. The concentration of Zn2+ in the condensed layer at the membrane surface and the Zn2+ distribution in the diffuse layer were measured as a function of headgroup charge. The Debye length of the diffuse layer varied between 3 and 58 angstroms. The results qualitatively agree with the Gouy-Chapman-Stern model.  相似文献

Two years of local earthquake, temperature, and rainfall data taken near a tiltmeter site were used in a study of the numerical relation between these phenomena and the recorded tilt response. A least-squares shaping and predictive error filter approach was used. The relations were ranked in part according to the root mean square (r.m.s.) error of fit across the entire sample space. The tilt data with an annual range of tilt of approximately 10 microradians were fitted to the combined weather data of temperature and rainfall with a 0.75-microradian r.m.s. error. The best fit of earthquakes to these same tilt data is the subclass of events with magnitude (M) > 2.5 within 30 kilometers of the tilt site. The filter that mapped earthquakes to tilt yielded a 1.03-microradian r.m.s. error. The most unusual tilt anomaly over the entire 2-year period has the best fit of rainfall to the data for any single month of the entire data set. This unusual anomaly was the basis of an erroneously predicted earthquake (M approximately 5). These data indicate that if there are premonitory earthquake signals, they are buried in local meteorlogical noise. Separating an earthquake anomaly from the response to surface phenomena becomes more difficult as the earthquake anomaly lead time approaches the rise time of the soil to weather and seasonal variations.  相似文献

Prolonged inspection of convex lines of one color and concave lines of another color can cause the appearance of oppositely colored aftereffects in curved, achromatic test lines. These effects, strongly dependent on radius of curvature, cannot be attributed to tilt or orientation. It is concluded that these results are not encompassed by models of the visual system that are based on rectilinear arrays and that curvature is a specific feature of human visual perception.  相似文献

The use of molecular self-assembly to fabricate microstructures suitable for advanced material development is described. Templating techniques that transform biomolecular self-assemblies into rugged and stable nano- and microstructures are described. By using a lipid-based microcylinder (tubule) as a paradigm, the path followed from research and development to emerging technological applications is detailed. This process includes modification of the lipid molecular structure, the formation and subsequent characterization of cylindrical microstructures, the use of these structures as templates for metallization, and the characterization and assessment of these hollow metal microcylinders for several potential applications.  相似文献

All previous experimental and theoretical studies of molecular interactions at metal surfaces show that electronically nonadiabatic influences increase with molecular velocity. We report the observation of a nonadiabatic electronic effect that follows the opposite trend: The probability of electron emission from a low-work function surface--Au(111) capped by half a monolayer of Cs--increases as the velocity of the incident NO molecule decreases during collisions with highly vibrationally excited NO(X(2)pi((1/2)), V = 18; V is the vibrational quantum number of NO), reaching 0.1 at the lowest velocity studied. We show that these results are consistent with a vibrational autodetachment mechanism, whereby electron emission is possible only beyond a certain critical distance from the surface. This outcome implies that important energy-dissipation pathways involving nonadiabatic electronic excitations and, furthermore, not captured by present theoretical methods may influence reaction rates at surfaces.  相似文献

Highly localized chemical catalysis was carried out on the surface groups of a self-assembled monolayer with a scanning probe device. With the use of a platinum-coated atomic force microscope tip, the terminal azide groups of the monolayer were catalytically hydrogenated with high spatial resolution. The newly created amino groups were then covalently modified to generate new surface structures. By varying the nature of the catalyst and the chemical composition of the surface, it may be possible to synthesize molecular assemblies not readily produced by existing microfabrication techniques.  相似文献

The nuclear pore complex regulates cargo transport between the cytoplasm and the nucleus. We set out to correlate the governing biochemical interactions to the nanoscopic responses of the phenylalanineglycine (FG)-rich nucleoporin domains, which are involved in attenuating or promoting cargo translocation. We found that binding interactions with the transport receptor karyopherin-beta1 caused the FG domains of the human nucleoporin Nup153 to collapse into compact molecular conformations. This effect was reversed by the action of Ran guanosine triphosphate, which returned the FG domains into a polymer brush-like, entropic barrier conformation. Similar effects were observed in Xenopus oocyte nuclei in situ. Thus, the reversible collapse of the FG domains may play an important role in regulating nucleocytoplasmic transport.  相似文献
