AIM: To explore the effect of Yiqi-Yangyin recipe on myocardial ischemia-reperfusion injury (MIRI) in rats with diabetes mellitus (DM) and the possible mechanism. METHODS: The rats were divided into normal group (control group), DM sham operation (DM-S) group, DM+MIRI group, low-, medium-and high-dose Yiqi-Yang-yin recipe (TL, TM and TH) groups (7.5, 15 and 30 g/kg decoction of Yiqi-Yangyin recipe by gavage), and Nrf2 inhibitor (bardoxolone methyl) group (30 mg/kg bardoxolone methyl by intragastric administration). The gavage volume was 1 mL/kg. There were 15 rats in each group, and they were administered continuously for 7 d. The tail vein blood was collec-ted after the last administration to detect the blood sugar and lipid levels in the rats. The serum levels of cardiac troponin I (cTnI), tumor necrosis factor-α (TNF-α), interleukin (IL)-1β and IL-10 were measured by ELISA. Echocardiography was used to detect the changes of cardiac function in the rats after blood collection. After cardiac function test, the rats were sacrificed to obtain cardiac tissues, and the volume changes of myocardial infarction were assessed by triphenylte-trazole chloride staining. The histopathological changes of myocardium was observed by HE staining. The cardiomyocyte apoptosis was determined by TUNEL assay. The protein levels of phosphorylated extracellular signal-regulated kinase (p-ERK), nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) in the myocardium were determined by Western blot. The myocardial activity of superoxide dismutase (SOD) was measured by nitro blue tetrazolium method, the content of malondialdehyde (MDA) was tested by thiobarbituric acid method, and the production of reactive oxygen species (ROS) was analyzed by iron ion reduction method. RESULTS: Compared with control group, the levels of fasting blood glucose (FBG), total cholesterol (TC) and triglyceride (TG) in DM-S group and DM+MIRI group were significantly elevated, while the level of high-density lipoprotein cholesterol (HDL-C) was significantly lowered (P<0.05). Compared with DM-S group and DM+MIRI group, the levels of FBG, TC, TG in TL, TM, TH and bardoxolone methyl groups were significantly decreased, while HDL-C level was significantly increased (P<0.05). Compared with control group and DM-S group, heart rate (HR) and left ventricular end-diastolic pressure (LVEDP) were increased in DM+MIRI group, mean arterial pressure (MAP), left ventricular systolic pressure (LVSP) and left ventricular ejection fraction (LVEF) were decreased, serum levels of cTnI, TNF-α, IL-1β and IL-10 were increased, the myocardial infarction volume percentage was increased, the myocardial cell breakage and necrosis were increased, the myocardial cell apoptotic rate was increased, the protein levels of p-ERK1/2, Nrf2 and HO-1 were decreased, MDA and ROS levels were increased, and the activity of SOD was decreased (P<0.05). Compared with DM+MIRI group, HR and LVEDP were decreased in TL, TM, TH and bardoxolone methyl groups, MAP, LVSP and LVEF were increased, the serum levels of cTnI, TNF-α, IL-1β and IL-10 were decreased, the myocardial infarction volume percentage was decreased, myocardial cell breakage and necrosis were decreased, myocardial cell apoptotic rate was decreased, the protein levels of p-ERK1/2, Nrf2 and HO-1 were increased, the MDA and ROS levels were decreased, and the activity of SOD was increased (P<0.05). CONCLUSION: Yiqi-Yangyin recipe protects the myocardial tissue of DM+MIRI rats from injury and reduces the oxidative stress level, which may be achieved by activating ERK/Nrf2/HO-1 pathway.  相似文献

AIM:To investigate the effects of propofol (P) on the inflammatory response of microglia induced by lipopolysaccharide (LPS) and the mechanisms. METHODS:Mouse microglia BV2 cells were treated with LPS at 100 μg/L to establish a neuroinflammatory injury model. The BV2 cells were divided into 4 groups:control group (C group), model group (L group), L+P group and LPS+AMG517 group (L+A group). The level of tumor necrosis factor-α (TNF-α) in the cell culture supernatant was measured by ELISA. The mRNA expression of transient receptor potential cation channel subfamily V member 1 (TRPV1) was detected by real-time PCR. The protein levels of TRPV1, TNF-α, interleukin-1β (IL-1β), interleukin-6 (IL-6) and phosphorylated calcium/calmodulin-dependent protein kinase Ⅱ (p-CaMKⅡ) were determined by Western blot. The content of free Ca2+ in the microglia BV2 cells was detected by Fluo-3 AM assay. RESULTS:Compared with C group, the level of TNF-α was significantly increased in L group (P<0.01), but that in P group was not changed. Compared with L group, the level of TNF-α was significantly lower than that in L+P group within 4 h (P<0.01). Compared with C group, the mRNA expression of TRPV1 was significantly increased in L group (P<0.01). Compared with L group, the mRNA expression of TRPV1 was significantly down-regulated in L+P group (P<0.01).Compared with L group, the protein levels of TNF-α, IL-1β, IL-6 and p-CaMKⅡ and intracellular Ca2+ concentration were significantly lower than those in L+P group and L+A group (P<0.01). CONCLUSION:Propofol inhibits the inflammatory response of microglia by reducing the expression of TNF-α, IL-1 and IL-6, which may be related to the down-regulation of TRPV1 and p-CaMKⅡ and the reduction of intracellular Ca2+ concentration.  相似文献

AIM: To determine the effects of Tongxinluo(TXL) on connexin 43(Cx43) remodeling and ventricular arrhythmia(VA) after myocardial infarction(MI) in rats. METHODS: Male SD rats were randomly divided into sham-operated(sham) group(n=25) and operation group(n=75). The left anterior descending(LAD) was ligated in operated group, while the rats in sham group only underwent pericardiotomy. The rats in operation group which survived for 3 d after operation were randomly assigned to TXL group and MI group. The rats in TXL group was administrated with TXL(2 g·kg-1·d-1, intragastric administration) for 4 weeks, while normal saline was applied to the rats in sham group and MI group. The levels of interleukin-1β(IL-1β) and endothelin-1(ET-1) in the tissue from the border zone were measured by ELISA after treatment. The distribution and the mRNA and protein expression of Cx43 were detected by immunohistochemical staining, RT-PCR and Western blotting, respectively. The burst pacing was used to induce ventricular arrhythmia(VA). RESULTS: Compared with sham group, the levels of IL-1β and ET-1 and the incidence of VA were significantly increased, while the mRNA and protein expression of Cx43 was markedly reduced with irregular distribution in MI group(P<0.05). Compared with MI group, the levels of IL-1β and ET-1 and the incidence of VA were significantly reduced, while the expression of Cx43 at mRNA and protein levels was markedly increased with augmented linear distribution in the myocardial cell intercalated disc in TXL group(P<0.05). CONCLUSION: TXL reduces the incidence of VA after MI via inhibiting the Cx43 remodeling.  相似文献

AIM:To investigate whether hydrogen sulfide (H2S) protects the hearts against inflammatory responses induced by acute myocardial ischemia in isolated rat hearts. METHODS:Rat acute myocardial ischemia injury was induced by ligation of the left anterior descending coronary artery for 4 h, and the normal perfusate was replaced with NaHS (5 μmol/L, 10 μmol/L and 20 μmol/L) perfusate accordingly in NaHS groups 2 h after ischemia. The changes of cardiac function in the myocardial ischemic injury rats were observed. The mRNA expression of TNF-α, IL-1β, IL-6, IL-10 and ICAM-1 was detected by real-time PCR. The protein level of nuclear factor-κB (NF-κB) in the myocardial tissues was detected by Western blotting. RESULTS:The cardiac function in ischemia group was lower than that in sham group (P<0.01). Compared with ischemia group, perfusion of NaHS resulted in the improvement of the cardiac function (P<0.05 or P<0.01). Compared with sham group, the mRNA expression of TNF-α, IL-1β, IL-6 and ICAM-1 in the cardiac tissues was significantly increased, and the mRNA expression of IL-10 in the cardiac tissues was significantly decreased in ischemia group (P<0.01). Compared with ischemia group, the perfusion of NaHS significantly decreased the mRNA expression of TNF-α, IL-1β, IL-6 and ICAM-1 (P<0.05 or P<0.01). The perfusion of NaHS at concentrations of 10 μmol/L and 20 μmol/L significantly increased the mRNA expression of IL-10 (P<0.01). The protein level of NF-κB in ischemia group was markedly higher than that in sham group (P<0.01). Compared with ischemia group, the perfusion of NaHS at concentrations of 10 μmol/L and 20 μmol/L significantly decreased the expression of NF-κB (P<0.05 or P<0.01). CONCLUSION:H2S protects the hearts against acute ischemia injury through inhibition of NF-κB activation and subsequent down-regulation of NF-κB-dependent inflammatory gene expression.  相似文献

AIM:To explore the effects of exercise training on myocardial polyamine metabolism in aged rats. METHODS:Wistar rats were divided into 3 groups:old+exercise group(Old+Ex), old group(Old) and young group(Young). High-performance liquid chromatography was used to test the content of myocardial polyamines(putrescine, spermidine and spermine). The activity of polyamine synthesis and catabolism rate-limiting enzyme ornithine decarboxylase(ODC) and spermidine/spermine acetyltransferase(SSAT) was analyzed by [14C] labeling. The level of malondialdehyde(MDA) and the activity of superoxide dismutase(SOD) in the myocardium were detected by colorimetric method. The levels of TNF-α and IL-1β were determined by enzyme-linked immunosorbent assay. The cardiac functions were measured by echocardiography. The changes of myocardial ultrastructure were observed under transmission electron microscope. RESULTS:(1) Compared with Young group, ODC activity decreased, SSAT activity increased, and the levels of spermidine, spermine and total polyamine were significantly decreased in Old group. SOD activity also decreased significantly, the content of MDA, TNF-α and IL-1β also increased dramatically in Old group. In the old rats, both left ventricular end-systolic diameter(LVESD) and end-diastolic diameter(LVEDD) increased, while left ventricular ejection fraction(LVEF) and fractional shortening(LVFS) decreased.(2) Compared with Old group, ODC activity increased, SSAT activity decreased, the levels of spermidine and total polyamine significantly increased, SOD activity increased significantly, and the content of MDA, TNF-α and IL-1β decreased dramatically in Old+Ex group. The functions of left ventricle also improved significantly in Old+Ex group.(3) Ultrastructural observations indicated that the myofilament and mitochondrial cristae were irregular and mitochondrial matrix density decreased in old rat heart. The cardiac sarcomere structure was clear, the mitochondrial matrix was dense and the mitochondrial cristae arranged in order in Old+Ex group. CONCLUSION:Exercise training promotes polyamine synthesis metabolism and inhibits polyamine catabolism in the heart of aged rats, thus fighting against aging-induced myocardial polyamine decrease. Exercise training also improves the capacity of anti-inflammatory and antioxidant processes of aged heart. Maintaining the appropriate levels of myocardial polyamines may be a new mechanism to delay cardiac aging.  相似文献

AIM:To observed the effect of quercetin on NLRP3 inflammasome activation in the rats with diabetic cardiomyopathy (DCM) and its protective effect on the myocardium. METHODS:Male SD rats (n=40) were randomly divided into normal control group (n=10) and model group (n=30). The rats in model group were intraperitoneally injected with streptozotocin at 60 mg/kg to establish the model of diabetes mellitus (DM). Blood glucose was measured weekly. After 4 weeks, the rats with random blood glucose ≥ 16.6 mmol/L were selected as DM animals. The rats with DM were randomly divided into 3 groups:DM group, DM+vehicle group and DM+quercetin group. The rats in DM+quercetin group were intragastric infusion with quercetin at 100 mg/kg per day. The cardiac function was measured at the end of the 16th week. The methods of Masson staining and HE staining were used to observe the morphological changes of the myocardial tissues. Western blot, ELISA and immunohistochemistry were used to observe the changes of NLRP3, ASC, caspase-1, interleukin (IL)-1β and IL-18. TUNEL staining was used to observe myocardial apoptosis. RESULTS:Quercetin significantly inhibited the activation of NLRP3 inflammasome in the myocardium of the DM rats (P<0.05). The levels of IL-1β and IL-18 in DM+quercetin group were significantly decreased, quercetin reduced cardiac tissue apoptosis, and the cardiac function in DM+quercetin group was significantly improved (P<0.05) compared with DM group and DM+vehicle grpup. CONCLUSION:Quercetin significantly inhibits the activation of NLRP3 inflammasome, and reduces the levels of inflammation and myocardial apoptosis, thus protecting the heart function of DCM rats.  相似文献

AIM: To investigate the protective effect of heme oxygenase-1 (HO-1) on the kidney of septic rats and the influence of HO-1 on the expression of thrombomodulin (TM) in the kidney. METHODS: Sepsis in rats was developed with cecal ligation and puncture (CLP). The septic rats were randomly divided into sham group, CLP group, CLP+HO-1 inducer group and CLP+HO-1 inhibitor group (n=18). The plasma levels of creatinine (Cr), cystatin-C (Cys-C), carboxyhemoglobin (COHb), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and TM, and the changes of prothrombin time (PT) and activated partial thromboplastin time (APTT) in each group were measured. Histopathological examination was performed in the kidney. The expression of TM in the kidney tissue was detected by Western blot. RESULTS: Compared with sham group, significantly elevated plasma levels of Cr, Cys-C, TNF-α, IL-1β and TM (P<0.05), shortened PT and APTT (P<0.05), significantly increased microthrombus formation, and lowered TM expression in the kidney (P<0.05) of CLP group were observed. The administration of hemin lowered the plasma levels of Cr, Cys-C, TNF-α, IL-1β and TM (P<0.05), prolonged PT and APTT (P<0.05), attenuated microthrombus formation, and up-regulated the expression of TM in the kidney (P<0.05). In contrast, ZnPP had the opposite effects. CONCLUSION: HO-1 increases the expression of TM in the kidney and exerts anticoagulatory and antiinflammatory effects, thereby improving renal function in the septic rats.  相似文献

AIM: To investigate the effect of microRNA (miR)-451 by targeting proteasome subunit β type 8 (Psmb8) on the inflammatory responses in mouse glomerular mesangial cells (MCs) under high-and low-glucose conditions. METHODS: The expression levels of miR-451, IL-18 mRNA and TNF-α mRNA were detected by qPCR. The protein expression levels of IL-18, TNF-α and Psmb8 were determined by Western blot when miR-451 was over-expressed and down-expressed in the MCs. Moreover, the expression of IL-18 and TNF-α was detected when Psmb8 was silenced by si-Psmb8 in MCs. RESULTS: The expression of miR-451 was significantly decreased in the MCs treated with high glucose compared with low glucose group (P<0.01). However, the expression of Psmb8 was increased in high glucose group as compared with low glucose group (P<0.01). Moreover, the expression levels of Psmb8, IL-18 and TNF-α were significantly decreased when miR-451 was over-expressed in high glucose group (P<0.01). Additionally, the expression levels of IL-18 and TNF-α were significantly reduced when Psmb8 was silenced in the MCs under high glucose condition. CONCLUSION: miR-451 reduces the expression of inflammatory factors via targeting Psmb8 in the MCs under high glucose condition. Therefore, miR-451 may play a role in inflammation of diabetic nephropathy.  相似文献

AIM:To observe the effect of pretreatment with external trigeminal nerve electrostimulation (eTNS) on behavioral changes and the expression of interleukin-1β (IL-1β) and  tumor necrosis factor α (TNF-α) in hippocampus of pentylenetetrazol (PTZ)-treated rats. METHODS:The rats were randomly divided into control group, PTZ group and eTNS group, and kindled by PTZ after administered 7 d, 14 d and 28 d of consecutive fake electrostimulation or eTNS. Subsequently, the severity and duration of seizure were quantitatively evaluated. The concentrations of IL-1β and TNF-α in hippocampus were detected by the methods of ELISA and immunohistochemisty. RESULTS:Compared with PTZ group, treatment with eTNS significantly inhibited the severity and duration of seizure (P<0.05), and significantly reduced the content of IL-1β and TNF-α in the hippocampus after status epilepticus (P<0.05 or P<0.01). CONCLUSION:Pretreatment with eTNS may provide a new approach for prevention and treatment of epileptogenesis.  相似文献

AIM: To investigate the effects and molecular mechanisms of poly(adenosine diphosphate ribose) glycohydrolase (PARG) on rat hippocampus neurons after seizures and to study the effects of gallotannin on the expression of apoptosis-inducing factor (AIF), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in rat hippocampus after seizures. METHODS: Seizures were induced by kainic acid (KA). The damage of hippocampus neurons was evaluated by Nissl staining. The protein expression levels of poly(adenosine diphosphate ribose) (PAR), AIF, IL-1β and TNF-α were detected by Western blotting analysis. RESULTS: The number of damaged hippocampal pyramidal neurons in gallotannin-treated group was significantly lower than that in KA-treated group (P<0.05). The expression of PAR was increased in gallotannin-treated group compared with KA-treated group(P<0.05).AIF in mitochondrial fraction increased and its accumulation in nucleus fraction decreased in gallotannin-treated group compared with KA-treated group (P<0.05). In addition, gallotannin significantly decreased the protein expression of IL-1β and TNF-α, which were obviously increased in hippocampus after seizures (P<0.05). CONCLUSION: PARG inhibition by gallotannin has the neuroprotective effect on the damage of hippocampal neurons induced by seizures. In addition, gallotannin suppresses the translocation of AIF from mitochondria to nucleus and the expression of IL-1β and TNF-α in rat hippocampus after seizures.  相似文献

AIM:To explore the effects of different β-adrenergic receptors (β-AR) on the left and right ventricular systolic and diastolic functions in rats under acute hypoxic stress. METHODS:The healthy male SD rats were randomly divided into 4 groups (n=7):control group, non-selected β-AR blocker propranolol group, selected β1-AR blocker atenolol group and selected β2-AR blocker ICI 118,551 group, and then the rats were exposed to normoxia (20.9% O2, 79.1% N2) and hypoxia (15.0% O2, 85.0% N2) condition respectively at the altitude of 2 260 m (Xining, China). The heart rate (HR), the left ventricular systolic pressure (LVSP), the right ventricular systolic pressure (RVSP), and the maximum raise/decline rate of left and right ventricular pressure (±dp/dtmax) were monitored, and the arterial blood gas in normoxia and hypoxia condition were compared to explore the effect of β-AR on the left and right ventricular systolic and diastolic functions in acute hypoxic stress rats. RESULTS:Under normoxia condition, the LVSP, ±dp/dtmax of left ventricular were decreased in propranolol group, atenolol group and ICI 118,551 group, the RVSP and ±dp/dtmax of right ventricle were decreased in propranolol group and atenolol group (P<0.05). Under hypoxia condition, the PaO2, LVSP, ±dp/dtmax of left ventricle were decreased in all groups compared with the normoxia group, and the ±dp/dtmax of right ventricle was increased in all groups (P<0.05), also the degree of index change in control group was more obvious than that in propranolol group and atenolol group. CONCLUSION:The activation of β1-AR is an important compensatory regulation for heart function during hypoxic stress. However, the compensatory enhancement of right heart function under acute hypoxia condition which through tonogenic dilation is more significant for maintaining the normal circulating blood flow.  相似文献

AIM: To observe the effects of adiponectin(APN) on the expression of connexin 43 (Cx43) in rat myocardium during ischemia-induced arrhythmias. METHODS: The SD rats were randomly divided into 4 groups (n=12): sham operation group (SM group), ischemia and reperfusion group (I/R group), I/R+adiponectin(APN1) group: pre-ischemia with 3.5 μg/kg of APN; I/R+APN2 group: post-ischemia with 3.5 μg/kg of APN. The incidence of ventricular arrhythmias and ventricular arrhythmia score (VAS) were determined. The expression of Cx43 in the ischemic myocardium was studied by the techniques of immunohistochemistry and RT-PCR. The levels of malondialdehyde(MDA) and superoxide dismutase(SOD) were measured by the methods of xanthine oxidase and thiobarbituric acid. The expression of endothelial nitric oxide synthase (eNOS) at mRNA and protein levels was determined by RT-PCR and Western blotting,respectively.The morphological changes of the myocardial tissues were observed under electronic microscope. RESULTS: The VAS and concentration of MDA increased obviously and the activity of SOD was decreased in I/R group as compared with SM group (P<0.01). The expression of Cx43 was evidently decreased and the distribution of Cx43 in the myocardium was disturbed. The expression of eNOS at mRNA and protein levels was decreased in I/R group (P<0.05). The ultrastructure of ventricular myocardium was abnormal in I/R group. Compared with I/R group, APN obviously decreased the VAS caused by ischemia and reperfusion (P<0.01) no matter the drug was given before or after ischemia. APN increased the activity of SOD, inhibited the MDA content in serum, and resulted in normal distribution of Cx43 and increased the expression of Cx43 and eNOS. Compared with I/R group, the changes of heart ultrastructure attenuated greatly in APN group, but didn't recover to normal state. CONCLUSION: Adiponectin antagonizes the arrhythmias during myocardial ischemia and reperfusion via inhibiting oxidative stress and regulating Cx43.  相似文献

AIM: To investigate the role of TLRs/NF-κB pathway in experimental allergic encephalomyelitis (EAE) rats treated with tripterygium glycosides (TG) + dexamethasone (DX). METHODS: Lewis rats were used in the study and divided into control group, EAE model group, therapy 1 group (EAE rats treated with DX) and therapy 2 group (EAE rats treated with DX+TG). The mean clinical score of the rats was determined. The expression of TLR4 and TLR9 at mRNA and protein levels was detected by the methods of real-time quantitative RT-PCR and immunohistochemistry. The protein level of NF-κB p65 was also measured. The levels of TNF-α, IL-1β and IL-6 were assayed by ELISA. RESULTS: The mean clinical scores at 5th, 16th and 20th day were lower in therapy 1 group and therapy 2 group than that in EAE model group. The mean clinical score in therapy 2 group was even lower than that in therapy 1 group. At the 16th day (the peaking period), the mRNA expression of TLR4 and TLR9 in therapy 1 group and therapy 2 group were obviously lower than that in EAE model group. The protein levels of TLR4, TLR9 and NF-κB p65 were also significantly lower in therapy 1 group and therapy 2 group than those in EAE model group at peak stage of EAE. The levels of TNF-α, IL-1β and IL-6 were lower in therapy1 group and therapy2 group than those in EAE model group. The significant differences of the mean clinical score, the mRNA expression of TLR4 and TLR9, the positive ratio of NF-κB p65 and the levels of TNF-α, IL-1β and IL-6 between therapy 1 group and therapy 2 group were found. The result of orthogonal factorial analysis of variance indicated that the difference of therapeutic effect between DX and DX+TG was significant (F=75.749, P<0.01). CONCLUSION: The TLRs/NF-κB pathway takes part in the pathological process of EAE. TG combined with DX alleviates the symptoms of EAE by suppressing inflammatory and immunological reactions of EAE.  相似文献

AIM: To study the effect of G-protein-coupled receptor kinase 5 (GRK5) on the activation of astrocytes in the brain cortex of newborn Wistar rats. METHODS: GRK5 gene was silenced in the model of rat brain cortex astrocytes in vitro for 24 h. N-acetylcysteine (NAC), which is a known inhibitor of NF-κB, was added into the culture medium according to gene silencing for 24 h. The expression levels of GFAP and caspase-3 were detected by the method of immunofluorescence, and the mRNA levels of NF-κB, TNF-α, IL-1β and iNOS were determined by real-time PCR. Moreover, the activity of SOD and concentrations of TNF-α and NO were measured. RESULTS: GRK5 gene silencing increased the expression of NF-κB at mRNA and protein levels obviously (P<0.01), and the mRNA levels of IL-1β and iNOS increased synchronously (P<0.01). Furthermore, caspase-3-positive cells in GRK5 siRNA group were increased compared with control siRNA group (P<0.01). Treatment with NAC obviously reduced the activity of NF-κB and weakened the effects induced by GRK5 siRNA (P<0.05). CONCLUSION: GRK5 siRNA increases NF-κB activity and induces the activation of astrocytes.  相似文献

AIM:To investigate the effects of pioglitazone, a peroxisome proliferator-activated receptor γ (PPARγ) agonist, on the cognitive impairments and inflammatory cytokine production induced by isoflurane in aged mice. METHODS:Male C57BL/6J mice (11-month-old, n=136) were assigned randomly into 5 groups: control group (Con), isoflurane group (Iso), 10 mg/kg pioglitazone + isoflurane group (Pi10+Iso), 20 mg/kg pioglitazone + isoflurane group (Pi20+Iso) and 20 mg/kg pioglitazone alone group (Pi20). The mice in all isoflurane-treated groups were exposed to oxygen mixed with 1.4% isoflurane for 2 h. The mice in Con group and in Pi20 group were exposed to oxygen only for 2 h. Pioglitazone was suspended in 1% carboxymethyl cellulose (CMC). Pioglitazone (10 mg/kg or 20 mg/kg) was gavaged 2 h prior to the exposure of isoflurane or oxygen alone. The same volume of 1% CMC was gavaged in Con group and in Iso group. Fear conditioning tests were performed to determine the learning and memory abilities 48 h after isoflurane exposure. Fresh cerebral cortice and hippocampi were dissected to measure the protein expression of PPARγ by Western blotting, and the contents of IL-1β and TNF-α were analyzed by ELISA 6 h after isoflurane exposure. RESULTS:Compared with Con group, the response of freezing behavior decreased (P<0.05) and IL-1β content in the hippocampus increased (P<0.05) in Iso group. Compared with Iso group, the response of freezing behavior and PPARγ protein expression level had no significant change (P>0.05) in Pi10+Iso group, but the response of freezing behavior and PPARγ protein expression level increased significantly (P<0.05) and IL-1β content in the hippocampus decreased (P<0.05) in Pi20+Iso group. IL-1β content in the cerebral cortex and TNF-α levels both in the cerebral cortex and the hippocampus showed no significant difference among all groups (P>0.05). CONCLUSION:Pioglitazone attenuates cognitive impairments and the elevates the level of IL-1β in the hippocampus induced by isoflurane in aged mice.  相似文献

AIM: To observe the clinical therapeutic effect of Jiawei Danzhi Xiaoyao powder combined with transplantation of human umbilical cord mesenchymal stem cells (HUC-MSCs) in the treatment of post-stroke depression (PSD) patients and to detect the changes of the serum cytokine levels of the tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, norepinephrine (NE), 5-hydroxytry-ptamine (5-HT) and brain derived neurotrophic factor (BDNF). METHODS: The patients with PSD (n=60) were randomly divided into observation group and control group. The patients in observation group was given Jiawei Danzhi Xiaoyao powder combined with HUC-MSCs, and the patients in control group was given Jiawei Danzhi Xiaoyao powder combined with fluoxetine. The course of treatment was 8 weeks. The effects of the treatments on the patients were evaluated with Hamilton Depression Scale (HAMD). The serum levels of IL-1β, IL-6, TNF-α, 5-HT, NE and BDNF were measured by ELISA. RESULTS: After 8 weeks of treatment, the total effective rate in observation group was significantly higher than that in control group (P<0.05). Compared with the same group before treatment, the HAMD scores and the serum levels of TNF-α, IL-1β, IL-6 of the 2 groups were significantly decreased after treatment (P<0.05), while 5-HT, NE and BDNF were significantly increased (P<0.05). After 8 weeks of treatment, the HAMD scores in observation group was significantly lower than that in control group (P<0.05), and the serum levels of TNF-α, IL-1β, IL-6 were significantly decreased in observation group (P<0.05), while 5-HT, NE and BDNF were significantly increased (P<0.05). CONCLUSION: Jiawei Danzhi Xiaoyao powder combined with human umbilical cord mesenchymal stem cells is effective in the treatment of post-stroke depression. The mechanism may be related to the effects of HUCMSCs such as anti-inflammatory effect, increased release of monoamine neurotransmitters, and stimulation of secretion of neurotrophic factors in the brain.  相似文献

AIM: To investigate the effects of curcumin (Cur) on the expression of High mobility group box 1 protein (HMGB1), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) in amyloid-β (Aβ)-induced primary rat microglial cells. METHODS: Microglia were derived from the cerebral cortices of postnatal rat brains. The cells were identified by immunocytochemistry using mouse anti rat Iba-1 monoclonal antibody. A cell model using primary rat microglial cells incubated with Aβ25-35 as an inflammation model of Alzheimer's disease (AD) was set up. The morphological characters of primary rat microglial cells were observed. The concentration of Aβ25-35 and the treatment concentration of curcumin were selected by CCK-8 assay. Cultured primary rat microglial cells were divided into 5 groups:normal cell group, Aβ25-35 group, Cur group, Aβ25-35+Cur group and Aβ25-35+DMSO group. The expression of HMGB1, NF-κB, and receptor for advanced glycation end products (RAGE) was detected by Western blot. The levels of HMGB1, IL-1β, and TNF-α in the culture supernatant were measured by ELISA. RESULTS: The purity of primary microglias determined by Iba-1 immunofluorescence was more than 95%. The protein levels of HMGB1, RAGE and NF-κB were significantly increased after Aβ25-35 stimulation. After treatment with Cur, the protein levels of HMGB1, RAGE and NF-κB were significantly decreased (P<0.05). The levels of HMGB1, IL-1β and TNF-α in the supernatant were significantly increased after Aβ25-35 stimulation. Cur significantly decreased the level of HMGB1, IL-1β and TNF-α in the supernatant. CONCLUSION: Curcumin significantly inhibits neuroinflammation stimulated by Aβ25-35 in primary rat microglial cells.  相似文献

AIM: To evaluate the effects of renal denervation (RDN) on the expression of tumor necrosis factor-α (TNF-α), interleukin-1α (IL-1α) and interleukin-6 (IL-6) in a rabbit model of early atherosclerosis. METHODS: New Zealand male rabbits were divided into control group, RDN+ high-fat diet (HFD) group (RDN group), sham+HFD group (sham group) and HFD group. The rabbits in later 3 groups were fed with 2% cholesterol for 8 weeks to establish an early atherosclerosis model. The blood samples were collected to test the levels of lipids, norepinephrine (NE), TNF-α, IL-1α and IL-6. The protein expression of angiotensin Ⅱ (Ang Ⅱ) was detected by the method of immunohistochemistry. The levels of nuclear factor-κB (NF-κB) and Ang II 1 type receptor (AT1R) were evaluated by Western blot. The mRNA expression of TNF-α, IL-1α and IL-6 was determined by real-time PCR. RESULTS: After 1 d of RDN procedure, the NE level was lower in RDN group than that in sham group (P<0.01). After 8 weeks, the NE level was lower in RDN group than that in sham group and HFD group (P<0.05), and triglyceride (TG) was lower in RDN group than that in HFD group (P<0.05). The protein expression of Ang II was decreased in RDN group compared with sham group and HFD group (P<0.01). The protein expression of NF-κB was lower in RDN group than that in sham group (P<0.05). The plasma levels of TNF-α and IL-1α were reduced in RDN group compared with sham group and HFD group (P<0.05). The mRNA expression of TNF-α, IL-1α and IL-6 was reduced in RDN group compared with sham group (P<0.05). CONCLUSION: RDN inhibits sympathetic activity, decreases the plasma level of TG, and alleviates inflammatory reactions in the rabbits with atherosclerosis.  相似文献

AIM:To explore the therapeutic effect of Qishen-Yiqi dripping pills (QS) on atherosclerosis (AS) and the mechanism. METHODS:AS rat model was established by high-fat diet, and SD rats were randomly divided into normal control group, AS model group, low-dose, middle-dose and high-dose QS groups, and positive group (n=6 each). After administration for 12 weeks, serum samples were collected to detect the serum lipid and Ca2+ levels. HE staining was used evaluated the histopathological changes of arterial tissue. The serum levels of interleukin-1β (IL-1β), IL-6 and tumor necrosis factor-α (TNF-α) were measured by ELISA. The nitric oxide (NO) level was detected by nitrate reductase method. The protein levels of transient receptor potential channel protein 1 (TRPC1), stromal interaction molecule 1 (STIM1) and endothelial NO synthase (eNOS) were determined by Western blot. RESULTS:QS significantly reduced the arterial damage via inhibiting the formation of atherosclerotic plaque and attenuated intimal thickening and vascular stenosis. Compared with AS group, the serum levels of total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) were decreased significantly and the levels of high-density lipoprotein cholesterol (HDL-C) were increased significantly in high-dose QS group (P<0.05). The serum levels of IL-1β, IL-6 and TNF-α in high-dose QS group were lower than those in AS group (P<0.05). Compared with AS group, the serum Ca2+ level was lowered and the arterial tissue NO level was elevated in QS groups (P<0.05). Compared with AS rats, the protein levels of TRPC1 and STIM1 were decreased significantly and the protein level of eNOS was increased significantly in the rats treated with QS (P<0.05). CONCLUSION:QS regulate calcium homeostasis via TRPC1/STIM1 pathway, increase the production of NO and inhibit the inflammatory responses, thus exerting anti-AS effect.  相似文献
