An overview of the genus Bursaphelenchus in the Czech Republic is presented, based on a recent survey for monitoring the presence of the pinewood nematode, Bursaphelenchus xylophilus, as well as on previous reports of this genus in the country. In addition, we provide a morphological and molecular characterization of four Bursaphelenchus species (B. eremus, B. pinophilus, B. vallesianus and B. borealis) found during the monitoring programme for forest pests, conducted during 2006–2010, within the Moravian and Bohemian regions. Nematodes were extracted from over 1917 insects and 1493 wood samples collected from deciduous and coniferous trees exhibiting wilting and declining symptoms. Bursaphelenchus species were found only in 0.73% of insects and 0.47% of the total number of wood samples. Bursaphelenchus borealis and B. pinophilus dauer juveniles were found associated with the insect vectors Dryocetes autographus and Pityogenes bidentatus, respectively. While a total of seven Bursaphelenchus species are now reported from the Czech Republic, the status of B. xylophilus remains as absent.  相似文献

As a result of the detection of the pinewood nematode Bursaphelenchus xylophilus in Portugal, and its subsequent spread to Spain, intense surveys were conducted to screen for the presence of Bursaphelenchus species in Romania. Herein, we report recent surveys of insects potentially vectoring Bursaphelenchus species collected using trap trees or pheromone‐baited traps placed in the forest. Trap felled spruce trees (Picea abies) and pheromone‐baited traps were installed in six different counties in Romania (Bra?ov, Sibiu, Suceava, Hunedoara, Timi? and Dâmbovi?a). Ten different species of insects distributed among Curculionidae and Cerambycidae were obtained. Nematodes were extracted from insects and observed to validate the presence of Bursaphelenchus specimens. One female identified as Monochamus sutor was the only specimen carrying nematodes in the genus Bursaphelenchus. Nematodes were identified as B. mucronatus based on morphological and molecular features. This is the first detection and report of natural spread of B. mucronatus in Romania. The absence of B. xylophilus was confirmed in the areas of Romania surveyed in this work.  相似文献

A survey of the xylophagous insects in the conifer forests, mainly Pinus pinaster woodlands, in Spain was carried out in order to study the phoretic vectors of nematodes of the genus Bursaphelenchus. Fifteen environmental sites were characterized on the bases of altitude, rainfall, temperature and conifer species. Specimens of Orthotomicus erosus (n = 3476) were captured of which 59% carried nematodes under their elytrous, most of them were aphelenchid fungal‐feeding nematodes, but B. fungivorus was found on the beetles from Las Cumbres, Villa Blanca, (Huelva), southern Spain, which represent the first record of O. erosus as vector of a Bursaphelenchus sp. The nematode is characterized by morphometric and biomolecular techniques and distribution and ecology of the insect is discussed.  相似文献

A national research program was initiated in 1988 in Sweden to provide more information on the distribution and bionomics ofBursaphelenchus species. As a result of the intimate association known to occur between the pinewood nematodeB. xylophilus (Steiner andBuhrer) Nickle and its insect vectors a survey was directed to Scots pine wood (Pinus sylvestris L.) in whichMonochamus species were breeding. A total of 59M. sutor L. and twoM. galloprovincialis Ol. emerged from the collected material. Fourteen specimens of theM. sutor and bothM. galloprovincialis specimens containedBursaphelenchus nematodes which resembled the speciesB. mucronatus Mamiya andEnda. To our knowledge this is the first record of theB. mucronatus-type nematodes inM. galloprovincialis and the first finding of such nematodes inM. sutor in Europe.  相似文献

The pinewood nematode (PWN) Bursaphelenchus xylophilus is an invasive pathogen that was introduced from North America to Asian countries and Portugal and is devastating native pine forests. Some native European and Asian Bursaphelenchus nematodes also have weak to moderate pathogenicity to native pine species. To evaluate the potential risk of native Bursaphelenchus species, we inoculated ten Japanese Bursaphelenchus species into native pine species (the dominant forest species) in Japan, and evaluated their pathogenicity using mortality and tracheal tissue damage as indices. Inoculation was conducted on August 3, 2007, and the symptoms were observed every 2 weeks until February 1, 2008. None of the inoculated trees, excluding the pathogenic PWN inoculated control, showed external disease symptoms; however, four species [a less pathogenic PWN isolate, B. luxuriosae, Bursaphelenchus sp. NK215 (undescribed), and NK224 (undescribed)] caused tracheal tissue damage in inoculated seedlings and showed weak pathogenicity. Therefore, we conclude that there are some potentially pathogenic native species of nematodes distributed in Japan. Interestingly, two of these weakly pathogenic species, B. luxuriosae and NK215, are not associated with Pinaceae trees, suggesting that nematode pathogenicity may be a pre-adaptive character. More experimental studies under different conditions are necessary to accurately evaluate the potential risk of these pathogens.  相似文献

We determined the incidence of Bursaphelenchus species in declining Pinus sylvestris stands in the Swiss Alps. A total of 217 trees showing different degree of decline were sampled from various locations in Valais between 2001 and 2004. Bursaphelenchus species were only found in recently dead or dying trees with an overall incidence of 40%. There was a positive relationship between Bursaphelenchus infestation and blue stain in these trees. Five Bursaphelenchus species were identified: Bursaphelenchus vallesianus, Bursaphelenchus mucronatus, Bursaphelenchus sexdentati, Bursaphelenchus leoni and Bursaphelenchus silvestris. The most frequent species were B. vallesianus detected in 75% and B. mucronatus in 20% of all Bursaphelenchus‐positive trees. Bursaphelenchus vallesianus is a recently described species within the B. sexdentati group, and could be a contributing factor in the observed pine decline in Valais.  相似文献

The pine wood nematode, Bursaphelenchus xylophilus, causes severe damage to pines in Eastern Asia. Bursaphelenchus mucronatus and B. doui resemble closely B. xylophilus morphologically, moreover they were found frequently in this area recently. It is necessary to identify the three species precisely and rapidly. In this study, we report the results of a multiplex one‐step polymerase chain reaction (PCR) utilizing five primers to identify and discriminate the three Bursaphelenchus species simultaneously. The multiplex one‐step PCR yielded one fragment of about 1000 bp for all Bursaphelenchus populations tested. Futhermore, B. xylophilus, B. mucronatus and B. doui produced another fragment of about 100, 350 and 600 bp respectively. This approach is simple and reliable to simultaneously identify the above three species within the xylophilus group usually encountered together in a nematode assay.  相似文献

Bursaphelenchus tusciae is reported for the first time in Tunisia and North Africa, associated with the insect Hylurgus ligniperda Fabricius (Coleoptera: Curculionidae: Scolytinae). Nematode identification was based on restriction fragment length polymorphism analysis and sequencing of the internal transcribed spacer (ITS) regions of ribosomal DNA. Phylogenetic analysis revealed that Tunisian B. tusciae clusters together with two other B. tusciae isolates forming a separate group close to B. hildegardae and B. eggersi. As H. ligniperda is among maritime pine scolytids pests in Tunisia and is widely distributed in North Africa, this study is an important contribution to the knowledge of Bursaphelenchus species associated with bark beetles of pine forests in Tunisia and North Africa.  相似文献

The genus Ganoderma Karst. has broad‐spectrum usage in biotechnology, medicine and the food industry. The complexity of the morphology within species has led to uncertain identification in the past, but recent advancements in molecular identification methods have provided scientists with the opportunity to better understand the taxonomy of the species. The present study attempts for the first time to elucidate the distinctiveness of the Ganoderma species growing in Iran concerning those elsewhere in the world based on mitochondrial small subunit ribosomal DNA (mtSSU rDNA) and internal transcribed spacer (ITS) sequence data. The results disclosed that the G. lucidum Karst. samples collected in Iran are more similar to the European Ganoderma species than to the Asian (Chinese) one used in this study.  相似文献

Coniferous wood imported from the Asian part of Russia was surveyed in Germany (Mukran ferry terminal, highway and railway border station in Frankfurt/Oder) and Austria (railway in Marchegg, Retz and Wr. Neustadt, Lower Austria). The consignments consisted of mixed timber of Pinus/Picea or Pinus/Larix. Out of 625 samples investigated, 51 samples (8.5%) yielded eight Bursaphelenchus species. Bursaphelenchus mucronatus was found 42 times in Pinus, Picea and Larix wood, Bursaphelenchus hylobianum and Bursaphelenchus fraudulentus were detected twice, in Pinus/Larix and in Larix, respectively. The following species were each found once: Bursaphelenchus leoni in mixed timber of Pinus/Picea, Bursaphelenchus ‘borealis’ in Pinus, Bursaphelenchus hellenicus and Bursaphelenchus paracorneolus in Larix. One sample of Larix wood from Krasnoyarsk region contained a few specimens corresponding to Bursaphelenchus xylophilus. Bursaphelenchus mucronatus was present in about 30% of the samples showing signs of insect attack. A Monochamus species was found in a sample from Irkutsk. Most of the B. mucronatus isolates found belonged to the European genotype, whereas the East Asian genotype was found in three instances. This is the first report of B. fraudulentus, B. hellenicus, B. leoni, B. paracorneolus and the East Asian genotype of B. mucronatus in Russia. Bursaphelenchus hylobianum is the only species found in Russian wood and not in Europe so far. The species were identified morphologically and by internal transcribed spacer (ITS)‐restriction fragment length polymorphism (RFLP) technique. Species‐specific ITS‐RFLP patterns were established for B. hylobianum. In the case of the isolate morphologically corresponding to B. xylophilus, DNA extraction from the available low number of specimens failed to yield sufficient rDNA for ITS‐RFLP analysis.  相似文献

In China, pinewood nematode (PWN), Bursaphelenchus xylophilus, was first discovered from Pinus thunbergii in 1982. Thus far, 14 species in the genus Pinus have been reported to be infected by PWN under natural conditions. Pinus yunnanensis, a pine species native to south‐western China, is considered a pioneer tree for barren hill afforestation in areas undergoing rocky desertification. In this study, we detected PWN in dead P. yunnanensis trees in Anlong County, Guizhou Province, China, using both morphological and molecular methods. To our knowledge, this is the first report of PWN from P. yunnanensis in China. To investigate the possible origin of this new outbreak, mitochondrial cytochrome oxidase gene subunit I and cellulase gene sequences were used to evaluate genetic relationships among worldwide PWN isolates. Phylogenetic tree and haplotype networks revealed that the Anlong isolate (BxChQAL008) sequence was identical to those of seven Chinese isolates collected from Sichuan, Chongqing, Zhejiang, Anhui and Shandong (372–1500 km from Anlong County), but different from the isolate BxChQZY030 collected from the same province (330 km from Anlong County). It is suggested, therefore, that more than one introduction of PWN into Guizhou Province has taken place. The Anlong isolate was likely introduced from neighbouring or more distant provinces rather than from outside China. Moreover, the absence of a correlation between geographic and genetic distance was observed using Mantel test analysis, providing evidence that human‐induced dispersal plays a fundamental role in the spread of the PWN in this region.  相似文献

The pinewood nematode, Bursaphelenchus xylophilus (Steiner and Buhrer, J. Agric Res. 48, 1934, 949), Nickle (J. Nematol. 2, 1970, 375), is the causative agent of the pine wilt disease and causes serious damage to pine forests around the world. During a survey for the pinewood nematode, four other Bursaphelenchus species (Bursaphelenchus mucronatus, B. sexdentati, B. anamurius and B. vallesianus) were isolated from wilted pine trees in Turkey. To understand the effects of these Bursaphelenchus species on wilting of pine trees, a study was conducted under greenhouse conditions. Two‐year‐old seedlings of three pine species (Pinus nigra, P. brutia and P. pinea) and one cedar species (Cedrus libani) were used. Fifteen seedlings of each species were inoculated with nematodes and 10 seedlings of each species served as controls. The inoculum densities used for each seedling contained approximately 1000 (±100) nematodes of all life stages in 0.25 ml of distilled water. The first wilting symptoms were observed in the fifth week in all pine species but not in the cedar seedlings. All seeding mortality occurred between the 5th and 13th weeks of the study; no mortality was observed outside of this period. The most pathogenic nematode species was B. mucronatus, closely followed by the other species. The most susceptible seedling species was P. nigra, and C. libani was the most resistant species.  相似文献

A fungus forming black stromata with several immersed ascomata was found associated with living branches of Protium warmingianum (Burseraceae) in the Mata da Biologia, city of Viçosa, Minas Gerais, Brazil. The new species differs from all five species found on literature and is fully described, illustrated and compared with the other species described in the genus. This is the first species of Rosenscheldia found on burseraceae hosts in Brazil and the first species on the host genus Protium.  相似文献

Bursaphelenchus fungivorus is reported for the first time in Portugal, identified as associated with Pinus pinaster bark and characterized on the basis of morphological and morphometrical characters for this species. Species identification was confirmed using restriction fragment length polymorphism analysis and sequencing of the internal transcribed spacer (ITS) regions of ribosomal DNA. Intraisolate genetic variability was detected among ITS sequences of the Portuguese B. fungivorus isolate. Phylogenetic analysis, obtained from multiple sequence alignment between ITS sequences of Bursaphelenchus species, revealed that the Portuguese B. fungivorus isolate clusters with other B. fungivorus isolates, forming a separate group close to B. seani highlighting a molecular proximity of these two species.  相似文献

An infestation of mangrove forests with Neodolichodorus persiangulfus n. sp. in southern Iran is reported. This paper is the first to report a representative species of the genus in the country. The new species was characterized by 2,120 (1795–2400) µm long females, and 1755 (1533–2019) µm long males, a continuous to slightly offset cephalic region separated from the rest of the body by a shallow depression, having 5–7 annuli and a distinct labial plate, a lateral field with four lines, the outer ones crenate, irregularly areolated in anterior and posterior body region, 79 (66–85) μm long female stylet, a didelphic‐amphidelphic female reproductive system with asymmetric sclerotized vaginae, phasmids 5–10 annuli behind the anus, female tail elongate conoid, suddenly narrowing at 5–12 annuli posterior to anus and males with trilobed bursa, 52.5 (47–58) μm long spicules and 21.5 (18–27) μm gubernaculum. With the elongate‐conoid female tail and c? >1.2, the new species is close to five known species of the genus: N. brevistylus, N. leiocephalus, N. paralongicaudatus, N. rageshi and N. sinensis, but is distinguished from them by the morphology of the female tail, areolation type of the lateral field, position of hemizonid and phasmid and morphometric data. Molecular phylogenetic analyses using sequences of the near full‐length fragment of the small and the D2‐D3 expansion segments of the large subunit ribosomal DNA (SSU and LSU rDNA D2‐D3) revealed the new species placed in a clade including one isolate of Neodolichodorus sp. and some species of Globodera, Heterodera and Rotylenchulus with 0.89 Bayesian posterior probability (BPP) in SSU phylogeny. The only available SSU sequence of the genus was the tentative closest relative to the new species in the inferred SSU tree. In the reconstructed LSU phylogenetic tree, N. persiangulfus n. sp. formed a poorly supported clade with Rotylenchulus spp. in Bayesian inference (BI), and the only other available D2‐D3 sequence of the genus occupied a distant placement to the new species.  相似文献

The spectrum and dynamics of xylophagous insects infesting Pinus pinaster after a fire event were studied from November 2007 to December 2008 at Sierra de Outes (Galicia, NW Spain). Insects were collected within and outside the fire area. Relationships among species status, species abundance, tree parameters and tree injury were assessed. Mortality of injured trees was also recorded. Insect colonization built up consistently during the first year after the fire, although the pests did not colonize unburned trees in nearby areas. Thirteen insect species from six xylophagous families were collected. Tomicus spp., Buprestis novenmaculata, Anobium punctatum and Pissodes castaneus were the first species detected. Tomicus spp. (29%) and Ips sexdentatus (23%) were the dominant taxa. Bole char height and soil burn severity were positively correlated with insect presence. Trees with short stem diameter and thinner bark were also preferred. B. novenmaculata presence was positively correlated with crown scorch. Loss of cambium hydration was negatively related to the occurrence of pests, likely because sapwood desiccation and loss of nutrient impede brood development. About 33.3% of the fire-scorched pines died. The results recommend the removal of injured and dying pines after fire, to avoid the rise of pest population threatening recovering trees in the burned areas.  相似文献

[目的]为探讨兼性互利关系在生物多样性保护方面的作用,[方法]于2015年5月和10月,采用陷阱法对云南墨江县雅邑镇4种类型样地地表蚂蚁群落进行调查。[结果]显示:(1)物种组成及多度:纯砂仁地(样地I)采集蚂蚁400头,23种;从未放养紫胶虫的紫胶-砂仁混农林样地(样地II)采集蚂蚁382头,15种;曾经放养过紫胶虫的紫胶-砂仁混农林样地(样地III)采集蚂蚁406头,18种;放养紫胶虫的紫胶-砂仁混农林样地(样地IV)采集蚂蚁655头,23种。(2)多样性:4种类型样地地表蚂蚁群落物种丰富度和相对多度存在显著差异,兼性互利关系可以提高地表蚂蚁的物种丰富度和相对多度,物种丰富度大小为IVIIIIII,相对多度排序为IIIIVIII。(3)群落结构相似性:样地III、IV和样地II、I不相似。(4)指示物种:样地I指示物种为毛发铺道蚁,样地II指示物种为卡泼林大头蚁和茸毛铺道蚁,样地III、IV中指示物种为巴瑞弓背蚁。[结论]互利关系对蚂蚁群落的物种丰富度、相对多度和群落结构具有明显的积极影响,并且这种兼性互利关系对生物多样性保护有一定的时空效应。  相似文献
